Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 2-(2-Aminothiazol-4-yl)-glyoxylsäure oder eines Derivats oder eines Salzes derselben.

Als Verfahren zur Herstellung dieser Ausgangsverbindungen sind bekannt: (1) ein Verfahren, bei dem ein Ester von 2-[2-(geschützt oder ungeschützt)-Aminothiazol-4-yl]-essigsäure mit Selendioxid oder Kaliumpermanganat oxidiert wird (JA-OS 125 190/77 oder 5 193/78), und (2) ein Verfahren, bei dem ein Ester der Acetylglyoxylsäure halogeniert wird, das resultierende Halogenierungsprodukt mit Thioharnstoff umgesetzt wird und anschliessend das Reaktionsprodukt hydrolysiert wird (JA-OSen 112 895/78 und 154 785/79).
Von den Erfindern wurden umfangreiche Untersuchungen durchgeführt mit dem Ziel, ein neues Verfahren zur Herstellung einer Verbindung der allgemeinen Formel (I) oder eines Salzes derselben zu schaffen. Dabei wurde von den Erfindern ein neues Herstellungsverfahren entwickelt, welches im folgenden beschrieben wird.
Es ist somit Aufgabe der vorliegenden Erfindung, ein neues Verfahren zur Herstellung von 2-(2-Aminothiazol-4-yl) -glyoxylsäure oder eines Derivats der allgemeinen Formel (1) oder eines Salzes desselben zu schaffen.
Die Erfindung wird im folgenden im Detail erläutert. Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 2-(2-Aminothiazol-4-yl)-glyoxylsäure oder eines Derivates der allgemeinen Formel (I) oder eines Salzes derselben auf folgendem Reaktionsweg:

Schutzgruppen für die Aminogruppe R' umfassen alle Gruppen, welche herkömmlicherweise als Amino-Schutzgruppen verwendet werden können. Als Halogenatom für Xl können beispielsweise verwendet werden: ein Fluoratom, ein Chloratom, ein Bromatom, ein Jodatom und dgl.
Als Alkylgruppe für R2 kommen beispielsweise in Frage: niedere Alkylgruppen, wie Methyl, Äthyl, n-Propyl und dgl., und als die Aralkylgruppe für R2 können beispielsweise Arniederalkylgruppen, wie eine Benzylgruppe und dgl., verwendet werden.
Das Salz der Verbindung gemäss der allgemeinen Formel (I) umfasst Salze an der Aminogruppe oder Salze an der Carboxylgruppe. Als Salze an der Aminogruppe können beispielsweise verwendet werden: Salze mit einer Mineralsäure, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Fluorwasserstoffsäure, Schwefelsäure oder dgl.; Salze mit einer organischen Carbonsäure, wie Oxalsäure, Ameisensäure Trichloressigsäure, Trifluoressigsäure oder dgl.; oder Salze mit einer Sulfonsäure, wie Methansulfonsäure, p-Toluolsulfonsäkre, 1oder 2-Naphthalinsulfonsäure oder dgl. Als Salze an der Carboxylgruppe kommen beispielsweise Salze mit einem Alkalimetallatom, wie Natrium, Kalium oder dgl. oder Salze mit einem Erdalkalimetallatom, wie Calcium, Magnesium oder dgl. in Frage.
Der Begriff Salz der Verbindung gemäss der allgemeinen Formeln (II) oder (III) bedeutet ein Salz an der Aminogruppe in den Formeln (II) oder (III) und umfasst speziell die gleichen Salze, wie diejenigen, die als Salze an der Aminogruppe der Verbindung gemäss der allgemeinen Formel (1) erwähnt wurden.
Die Umsetzung zur Herstellung einer Verbindung gemäss der allgemeinen Formel (III) oder eines Salzes derselben aus einer Verbindung gemäss der allgemeinen Formel (II) oder eines Salzes derselben wird durchgeführt, indem man die Verbindung der allgemeinen Formel (II) oder das Salz derselben mit einem Dialkylsulfoxid, wie Dimethylsulfoxid, Diäthylsulfoxid, Di-n-propylsulfoxid oder dgl., oder mit einem Diaralkylsulfoxid, wie Dibenzylsulfoxid oder dgl.. in einem Lösungsmittel, welches unter den Reaktionsbedingungen inert ist, z.B. ein Alkohol, wie Methanol, Äthanol, Isopropanol oder dgl., ein Äther, wie Tetrahydrofuran. Dioxan oder dgl., ein Amid, wie N,N-Dimethylformamid. N,N-Dimethylacetamid, Hexamethylphosphoramid oder dgl., oder ein Lösungsmittelgemisch derselben, umsetzt.
Das Dialkylsulfoxid oder das Diaralkylsulfoxid wird vorzugsweise in einer Menge von 2,0 Mol oder mehr und besonders bevorzugt 3,0 bis 4,0 Mol/Mol der Verbindung der allgemeinen Formel (11) oder des Salzes derselben eingesetzt. Gegebenenfalls kann das Sulfoxid als Lösungsmittel verwendet werden.
Falls eine Verbindung der allgemeinen Formel (11) eingesetzt wird, bei der X' ein Chloratom bedeutet, oder falls ein Salz derselben eingesetzt wird, so wird die Umsetzung vorzugsweise in Gegenwart eines Bromids, wie Bromwasserstoff, Kaliumbromid, Ammoniumbromid, Triäthylammoniumbromid oder dgl., durchgeführt. Die Menge des bei dieser Umsetzung eingesetzten Bromids beträgt vorzugsweise 0,5 Mol oder mehr und besonders bevorzugt 0,5 bis 1,0 Mol/Mol der Verbindung der allgemeinen Formel (II) oder des Salzes derselben. Die Umsetzung ist gewöhnlich innerhalb von 5 min bis 20 h bei einer Reaktionstemperatur von 10 bis 80 C vollständig abgelaufen.
Die Umsetzung wird beschleunigt duch Zusatz eines Dialkylsulfids, wie Dimethylsulfid, Diäthylsulfid oder dgl., eines Dialkyldisulfids, wie Dimethyldisulfid, Diäthyldisulfid oder dgl., eines Diaralkylsulfids, wie Dibenzylsulfid oder dgl., eines Diaralkyldisulfids, wie Dibenzyldisulfid oder dgl., eines Alkylmercaptans, wie Methylmercaptan, Äthylmercaptan oder dgl., oder eines Aralkylmercaptans, wie Benzylmercaptan oder dgl., in einer Menge von 1,0 Mol oder mehr/Mol der Verbindung gemäss der allgemeinen Formel (II) oder des Salzes derselben.
Dadurch, dass man die auf diese Weise erhaltene Verbindung der allgemeinen Formel (III) oder ein Salz derselben einer gewöhnlichen Hydrolyse unterwirft, lässt sich die Verbindung in eine Verbindung der allgemeinen Formel (I) oder ein Salz derselben umwandeln, welche brauchbar sind zur Herstellung einer Cephalosporin-Verbindung. Die Hydrolyse wird in diesem Fall in Wasser oder einem Alkohol, wie Methanol, Äthanol oder dgl., durchgeführt, vorzugsweise in Gegenwart einer Base. Als Base können beispielsweise verwendet werden: anorganische Basen, wie Natriumhydroxid, Kaliumhydroxid, Bariumhydroxid, Calciumhydroxid, Natriumcarbonat, Kaliumcarbonat und dgl., oder organische Basen, wie Triäthylamin, Pyridin und dgl. Diese Basen werden in einer Menge von 2,0 Mol oder mehr/Mol der Verbin dung der allgemeinen Formel (III) oder des Salzes derselben eingesetzt.
Die Verbindungen der allgemeinen Formeln (I), (II) und (III) oder die Salze derselben bilden Addukte mit verschiede nen Lösungsmitteln. Alle diese Addukte sind von der vorliegenden Erfindung umfasst.
Eine Verbindung der allgemeinen Formel (II) oder ein Salz derselben kann auf folgende Weise hergestellt werden:

X2a in der allgemeinen Fomel (VIa) bedeutet ein Halogenatom, wie ein Fluoratom, ein Chloratom, ein Bromatom, ein Jodatom oder dgl.
Die Halogenierung zur Herstellung eines l-Halogenbutan-2,3-dions gemäss der allgemeinen Formel (V) aus Butan2,3-dion gemäss der Formel (IV) und die Halogenierung zur Herstellung eines 1,4-Dihalogenbutan-2,3-dions gemäss der allgemeinen Formel (VIa) aus einem l-Halogenbutan- 2,3-dion gemäss der allgemeinen Formel (V) werden unter gleichen Bedingungen durchgeführt.
Beispielsweise können diese Halogenierungen in Abwesenheit eines Lösungsmittels oder in Gegenwart eines Lösungsmittels erfolgen, welches gegenüber der Reaktion inert ist, z.B. ein aromatischer Kohlenwasserstoff, wie Benzol, Toluol, Xylol oder dgl., ein Äther, wie Diäthyläther, Diisopropyläther, Tetrahydrofuran, Dioxan oder dgl., ein halogenierter Kohlenwasserstoff, wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Dichloräthan oder dgl., eine Carbonsäure, wie Essigsäure oder dgl., oder eine Lösungsmittelmischung derselben. Als Halogenierungsmittel kann ein Halogenierungsmittel eingesetzt werden, das üblicherweise für die Halogenierung eines Paraffins verwendet wird.
Beispielsweise können als Chlorierungsmittel Chlor, Sulfurylchlorid, N-Chlorsuccinimid, N-Chlorphthalimid und dgl. und als Bromierungsmittel Brom, Sulfurylbromid, N Bromsuccinimid, N-Bromphthalimid und dgl. verwendet werden. Die Menge des eingesetzten Halogenierungsmittels ist vorzugsweise etwa äquimolar, bezogen auf die Verbindung der allgemeinen Formel (IV) oder der allgemeinen Formel (V). Wenn auch die Reaktionsbedingungen abhängig von der Art des eingesetzten Halogenierungsmittels oder dgl. variieren können, so wird doch die Umsetzung allgemein durchgeführt bei einer Temperatur von 10 "C bis zur Rückflusstemperatur des Lösungsmittels während eines Zeitraums von 30 min bis 10 h.
Falls eine Verbindung der allgemeinen Formel (VIa) hergestellt wird, bei der X' und X2a die gleichen Halogenatome bedeuten, kann die Dihalogenierung in einem Schritt durchgeführt werden, indem man ein Halogenierungsmittel direkt mit einer Verbindung der Formel (IV) umsetzt, und zwar in einer Menge von etwa 2 Mol/Mol der Verbindung. In diesem Fall sind die Reaktionsbedingungen die gleichen wie oben erwähnt.
Bevorzugte Halogenierungen stellen Umsetzungen dar, bei denen die Verbindung der Formel (IV) zunächst mit Sulfurylchlorid chloriert wird, um eine Verbindung der allgemeinen Formel (V) zu erhalten, wobei X1 ein Chloratom bedeutet, und welche anschliessend mit Brom bromiert wird, um eine Verbindung der allgemeinen Formel (VIa) zu erhalten, bei der X2a für ein Bromatom steht.
Anschliessend wird, um eine Verbindung der allgemeinen Formel (II) oder ein Salz derselben durch Umsetzung eines 1,4-Dihalogenbutan-2,3-dions, z.B. I-Brom-4-chlorbutan2,3-dion, mit einem Thioharnstoff der allgemeinen Formel (VII) zu erhalten, die Umsetzung in Gegenwart eines Lösungsmittels, das gegenüber der Reaktion inert ist, z.B. ein Alkohol, wie Methanol, Äthanol, Isopropanol oder dgl., ein Äther, wie Tetrahydrofuran, Dioxan oder dgl., ein Amid, wie N,N-Dimethylformamid, N,N-Dimethylacetamid, Hexamethylphosphoramid oder dgl., oder ein Lösungsmittelgemisch derselben oder ein gemischtes Lösungsmittel aus einem oder mehreren derselben und Wasser, durchgeführt. Die eingesetzte Menge des Thioharnstoffs der allgemeinen-Formel (VII) kann 0,90 Mol oder mehr/Mol der Verbindung der allgemeinen Formel (VIa) betragen, und besonders bevorzugt sind 0,95 bis 1,00 Mol/Mol der Verbindung.
Diese Ringschlussreaktion verläuft gewöhnlich innerhalb von 5 min bis 20 h bei einer Reaktionstemperatur von - 50 bis 10 "C vollständig ab.
Die vorliegende Erfindung betrifft ferner eine Verbindung der folgenden allgemeinen Formeln (VI) oder (VIII) oder ein Salz der Verbindung der allgemeinen Formel (VIII), wobei es sich bei der genannten Verbindung oder dem Salz um ein neues und brauchbares Zwischenprodukt handelt, und betrifft ferner ein Verfahren zur Herstellung desselben. Es handelt sich dabei um folgende Verbindungen: ein l,4-Diha- logenbutan-2,3-dion gemäss der allgemeinen Formel (VI)

Als Monohalogenmethylgruppe für R3 können beispielsweise eine Chlormethylgruppe, eine Brommethylgruppe, eine Jodmethylgruppe und dgl. verwendet werden. Als Alkylthiocarbonylgruppe kommen beispielsweise eine Methylthiocarbonylgruppe, eine Äthylthiocarbonylgruppe, eine n- Propyl- thiocarbonylgruppe und dgl. und als Aralkylthiocarbonylgruppe beispielsweise eine Benzylthiocarbonylgruppe und dgl. in Betracht.
Unter den Verbindungen der allgemeinen Formel (VIII) sind besonders bevorzugte Verbindungen solche, bei denen R' für eine Aminogruppe oder eine Formylaminogruppe steht und R3 eine Chlormethylgruppe oder eine Methylthiocarbonylgruppe bedeutet.

Im folgenden wird die Erfindung anhand von Beispielen erläutert, ohne dass dadurch eine Beschränkung der Erfindung beabsichtigt ist.
Beispiel 1
Eine gemischte Lösung von 30,4 g des 1:1 Solvatkomplexes von Äthanol und Bromwasserstoffsalz von 2-Amino4-chloracetylthiazol, 91 ml Dimethylsulfoxid und 11,9 g Kaliumbromid wird auf 30 "C erhitzt und 8,9 ml Dimethyldisulfid werden zugesetzt. Die resultierende Reaktionsmischung wird 2 h bei 30 bis 35 "C gerührt und anschliessend in 300 ml Eis Wasser gegossen.
Nachfolgend wird das resultierende Gemisch mit Natriumhydrogencarbonat auf pH 5,5 eingestellt. Der ausgefallene Feststoff wird durch Filtration gesammelt und in 80 ml IN Chlorwasserstoffsäure aufgelöst. Eine geringe Menge unlöslichen Materials wird durch Filtration entfernt, woraufhin das Filtrat mit Natriumhydrogencarbonat auf pH 5,5 eingestellt wird. Die ausgefallenen Kristalle werden durch Filtration gesammelt, mit Wasser gewaschen und anschliessend getrocknet. Man erhält 11,7 g (61,4 'o Ausbeute) 2-(2-Aminothiazol4-yl)-thioglyoxyl-S-säure-methylester, Fp. 130 C (Zers.).

Beispiel 2
Zu 10,1 g 2-(2-Aminothiazol-4-yl)-thioglyoxyl-S-s ureme- thylester und 80 ml Wasser gibt man unter Kühlen mit Eis 10,6 g Natriumcarbonat und rührt die resultierende Mischung 1 h bei der gleichen Temperatur. Anschliessend wird die so erhaltene Reaktionsmischung mit 6N Chlorwasserstoffsäure bei der gleichen Temperatur auf pH 2,5 eingestellt. Die ausgefallenen Kristalle werden durch Filtration gesammelt, mit Wasser gewaschen und dann getrocknet. Man erhält 6,2 g (67,8% Ausbeute) 2-(2-Aminothiazol-4-yl)-glyoxylsäure, Fp.

Beispiel 3
Eine Mischung von 40,8 g Essigsäureanhydrid und 18,4 g Ameisensäure wird 1 h bei 40 bis 45 "C gerührt. Die resultierende Mischung wird unter Kühlen mit Wasser mit 20,2 g 2-(2-Aminothiazol-4-yl)-thioglyoxyl S-säuremethylester versetzt. woraufhin die resultierende Mischung 1 h bei 25 "C gerührt wird. Anschliessend werden 160 ml Wasser tropfenweise und Eiskühlung zu der so erhaltenen Reaktionsmischung zugesetzt. Dann wird die resultierende Mischung 30 min unter Kühlen mit Wasser gerührt, und die ausgefallenen Kristalle werden durch Filtration gesammelt.
Die Kristalle werden nacheinander mit Wasser und Aceton gewaschen und dann getrocknet. Man erhält 21,9 g (94,4% Ausbeute) 2-(2-Formylaminothiazol-4-yl)-thioglyoxyl S-säure-methylester, Fp. > 230 C.

Beispiel 4
In 50 ml Wasser werden 7,8 g des 1:1 Solsatkomplexes von Äthanol und Bromwasserstoffsalz von 2-Amino-4-chloracetylthiazol suspendiert. Zu der Suspension gibt man allmählich 2,3 g Natriumhydrogencarbonat bei 20 C unter Rühren während eines Zeitraums von 15 min. Die ausgefallenen Kristalle werden durch Filtration gesammelt, mit 10 ml Wasser gewaschen und dann getrocknet. Man erhält 4,5 g (98,8% Ausbeute) 2-Amino-4-chloracetylthiazol, Fp. 147 "C (Zers.).

Beispiel 5 (1) In 200 ml Wasser werden 23 g 2-(2-Formylaminothia zol-4-yl)-thioglyoxyl S-säure-methylester suspendiert. Dazu gibt man tropfenweise unter Kühlen mit Wasser während 30 min 125 ml einer 2N wässrigen Natriumhydroxidlösung und rührt die resultierende Mischung anschliessend 1 h bei Zimmertemperatur. Nach Beendigung der Reaktion wird der pH der so erhaltenen Reaktionsmischung mit 6N Chlorwasserstoffsäure auf 2,5 eingestellt.
