La présente invention a pour objet un dérivé hydrosoluble du triméthoprim (diamino-2,4-[(triméthoxy-3,4,5-phényl)méthyl]-5-pyr- imidine). L'invention eoncerne également des procédés de fabrication de ce dérivé hydrosoluble.
Le triméthoprim est un composé bien connu, employé en chimiothérapie pour sa grande activité contre les germes Gram positifs et
Gram négatifs, ce cornposé étant employé seul ou, de préférence, associé à au moins une autre substance active en tant que produit pharmaceutique, telle qu'un sulfamide ou un antibiotique. L'utilisation en combinaison de triméthoprim et d'un sulfamide ou d'un antibiotique permet, par effet de synergie, de renforcer l'activité des substances actives individuelles, tant en intensité qu'en ce qui concerne l'étendue du spectre antimicrobien. Le triméthoprim est donc très utilisé en tant qu'agent potentiateur des sulfamides et des antibiotiques.
Il est souvent nécessaire d'administrer le produit pharmaceutique renfermant le triméthoprim, associé à des sulfamides et/ou des antibiotiques, sous la forme d'une solution aqueuse, soit par voie orale, en dilution dans l'eau de boisson, soit par voie parentérale. Dans certains cas, il est souhaitable que ces praduits pharmaceutiques ren ferment en outre des acides polycarboxyliques, tels que l'acide glutamique, I'acide aspartique ou l'acide lactique, qui forment des sels par réaction avec le triméthoprim en solution aqueuse. En présence de sels alcalins solubles de sulfamide ou d'antibiotique, les sels ainsi formés donnent lieu à une réaction de double décomposition ayant pour effet de former un précipité, ce qui empêche l'administration de telles associations médicamenteuses sous forme de solution aqueuse.
L'invention a pour but d'éliminer cet inconvénient en fournissant des dérivés de triméthoprim solubles en milieu aqueux, sous forme neutre et stable permettant l'administration par voie orale ou parentérale de solutions aqueuses renfermant ces dérivés hydrosolubles de triméthoprim associés à des sels alcalins de sulfamide ou d'antibiotique.

Conformément à un premier procédé de fabrication du dérivé hydrosoluble de triméthoprim répondant à la formule a) indiquée ci-dessus, on fait réagir l'aldéhyde cinnamique avec un sulfite acide de métal alcalin, de façon à former un composé de formule:

et l'on fait ensuite réagir le composé de formule (II) ainsi formé avec la diamîno-2,4-[(triméthoxy-3 ,4,5-phényl)méthyl]-5-pyrimidine, de façon à former le composé de formule (I).
Selon un autre procédé de fabrication du composé de formule (I), on fait réagir l'aldéhyde cinnamique avec la diamino-2,4-[(trimé- thoxy-3,4,5-phényl)méthyl]-5-pyrimidine, de façon à former un composé de formule:

et l'on fait ensuite réagir le composé de formule (III) ainsi formé avec un sulfite acide de métal alcalin, de façon à former le composé de formule (I).
Lors de la mise en oeuvre de ces deux procédés, on effectue de préférence les réactions par chauffage d'un mélange des composés de départ, en proportion stoechiométrique ou en proportion voisine des proportions stoechiométriques, sous la forme d'une suspension ou d'une solution de ces composés en milieu aqueux.
Le composé final est obtenu à l'état cristallisé, sous la forme d'une poudre blanche ou légèrement crème, soluble dans l'eau à pH 7, insoluble dans l'éthanol, I'éther et le chloroforme.
Exemple 1:
On met en suspension 40 g d'aldéhyde cinnamique et 58 g de sulfide acide de sodium dans 250 ml d'eau, à la température ambiante, et l'on porte ensuite progressivement la suspension ainsi obtenue à la température de reflux, tout en agitant, puis l'on continue à chauffer à reflux, en maintenant l'agitation du milieu, jusqu'à obtention d'une solution limpide. On refroidit alors la solution ainsi obtenue et on y ajoute 250 ml d'alcool éthylique, de façon à provoquer la précipitation de disulfonate sodique de cinnamylidène. On filtre ce précipité et on le lave au moyen d'un mélange eau/éthanol (50/50, en volumes). On dissout ensuite le précipité purifié ainsi obtenu dans 150 ml d'eau, on ajoute 43,94 g de triméthoprim à la solution ainsi obtenue et on chauffe le mélange jusqu'à complète dis
solution du triméthoprim. On obtient ainsi une solution légèrement opaline que l'on clarifie par filtration et, finalement, on soumet la solution ainsi obtenue à une atomisation (séchage par pulvérisation) sous atmosphère protectrice d'azote.
On obtient ainsi 127 à 131 g (rendement: 92 à 95%) de dérivé sodique soluble de triméthoprim, ayant la formule (I) dans laquelle R=Na(bis-cinnamylidènedisulfonate sodique de triméthoprim), ce composé étant obtenu sous la forme d'une poudre blanche, cristalline, soluble dans l'eau à pH 7 et insoluble dans l'éthanol, I'éther et le chloroforme (teneur du composé en triméthoprim: 31,7% en poids; point de fusion: 246-248"C, avec décomposition).
La solubilité dans l'eau à pH7 du composé ainsi obtenu est supérieure à 50% en poids, la solution ainsi obtenue restant parfaitement neutre.
Exemple 2:
On procède comme dans l'exemple 1 mais, au lieu d'effectuer l'atomisation (séchage par pulvérisation) de la solution contenant le produit final, on sépare ce dernier par précipitation en ajoutant 250 ml d'alcool à 00C à la solution.
On obtient ainsi 128 à 132 g de dérivé sodique soluble de triméthoprim de formule (I) ayant le même degré de pureté que le produit obtenu de la manière indiquée dans l'exemple 1.
Exemple 3:
Dans 250 ml d'eau, on introduit successivement 43,94 g de triméthoprim, puis, en agitant, 20 g d'aldéhyde cinnamique. On porte ensuite à 80-90 C le milieu réactionnel ainsi formé et on le maintient à cette température pendant 30 min. Il se forme ainsi une base de
Schiff. On ajoute ensuite au milieu aqueux contenant cette base de
Schiff 58 g de bisulfite sodique et on chauffe jusqu'à obtention d'une solution parfaitement transparente. Finalement, on soumet la solution ainsi obtenue à une atomisation (séchage par pulvérisation) sous atmosphère protectrice d'azote. On obtient ainsi 128 à 132 g (rendement: 92-95%) de dérivé sodique soluble de triméthoprim, de formule (I), sous la forme d'une poudre blanche cristalline.

The present invention relates to a water-soluble derivative of trimethoprim (diamino-2,4 - [(trimethoxy-3,4,5-phenyl) methyl] -5-pyr-imidine). The invention also relates to methods of manufacturing this water-soluble derivative.
Trimethoprim is a well-known compound used in chemotherapy for its great activity against Gram-positive and
Gram negative, this compound being used alone or, preferably, associated with at least one other active substance as a pharmaceutical product, such as a sulfonamide or an antibiotic. The use in combination of trimethoprim and a sulfonamide or an antibiotic makes it possible, by synergistic effect, to strengthen the activity of the individual active substances, both in intensity and with regard to the extent of the antimicrobial spectrum. Trimethoprim is therefore widely used as a potential agent for sulfonamides and antibiotics.
It is often necessary to administer the pharmaceutical product containing trimethoprim, combined with sulfonamides and / or antibiotics, in the form of an aqueous solution, either orally, diluted in drinking water, or parenteral. In certain cases, it is desirable that these pharmaceutical praducts also contain polycarboxylic acids, such as glutamic acid, aspartic acid or lactic acid, which form salts by reaction with trimethoprim in aqueous solution. In the presence of soluble alkali salts of sulfonamide or antibiotic, the salts thus formed give rise to a double decomposition reaction having the effect of forming a precipitate, which prevents the administration of such drug combinations in the form of an aqueous solution.

During the implementation of these two processes, the reactions are preferably carried out by heating a mixture of the starting compounds, in stoichiometric proportion or in proportion close to the stoichiometric proportions, in the form of a suspension or of a solution of these compounds in an aqueous medium.
The final compound is obtained in the crystallized state, in the form of a white or slightly creamy powder, soluble in water at pH 7, insoluble in ethanol, ether and chloroform.
Example 1:
40 g of cinnamic aldehyde and 58 g of sodium acid sulfide are suspended in 250 ml of water at room temperature, and the suspension thus obtained is then gradually brought to reflux temperature, while stirring , then heating is continued at reflux, while stirring the medium, until a clear solution is obtained. The solution thus obtained is then cooled and 250 ml of ethyl alcohol are added thereto, so as to cause the precipitation of cinnamylidene sodium disulfonate. This precipitate is filtered and washed with a water / ethanol mixture (50/50, by volume). The purified precipitate thus obtained is then dissolved in 150 ml of water, 43.94 g of trimethoprim are added to the solution thus obtained and the mixture is heated until complete dis
trimethoprim solution. A slightly opaline solution is thus obtained which is clarified by filtration and, finally, the solution thus obtained is subjected to atomization (spray drying) under a protective nitrogen atmosphere.
127 to 131 g are thus obtained (yield: 92 to 95%) of soluble sodium derivative of trimethoprim, having the formula (I) in which R = Na (bis-cinnamylidenedisulfonate sodium of trimethoprim), this compound being obtained in the form d 'a white, crystalline powder, soluble in water at pH 7 and insoluble in ethanol, ether and chloroform (content of trimethoprim compound: 31.7% by weight; melting point: 246-248 " C, with decomposition).
The solubility in water at pH 7 of the compound thus obtained is greater than 50% by weight, the solution thus obtained remaining perfectly neutral.
Example 2:
The procedure is as in Example 1 but, instead of carrying out atomization (spray drying) of the solution containing the final product, the latter is separated by precipitation by adding 250 ml of alcohol at 00C to the solution.
128 to 132 g of soluble sodium derivative of trimethoprim of formula (I) are thus obtained having the same degree of purity as the product obtained in the manner indicated in Example 1.
Example 3:
43.94 g of trimethoprim are successively introduced into 250 ml of water, then, with stirring, 20 g of cinnamic aldehyde. The reaction medium thus formed is then brought to 80-90 C and maintained at this temperature for 30 min. This forms a basis for
Schiff. Then added to the aqueous medium containing this base of
Schiff 58 g of sodium bisulfite and the mixture is heated until a perfectly transparent solution is obtained. Finally, the solution thus obtained is subjected to atomization (spray drying) under a protective nitrogen atmosphere. 128 to 132 g are thus obtained (yield: 92-95%) of soluble sodium derivative of trimethoprim, of formula (I), in the form of a white crystalline powder.
