PATENTANSPRÜCHE
1. Verfahren zur Herstellung des l-(2-Allyloxyphenoxy)3-isopropylamino-2-propanol und Salzen davon, dadurch gekennzeichnet, dass man in einer Verbindung der Formel

worin A die Gruppe der Formel -CHOH-CO (IIa) oder Formel -CO-CO- (IIb) darstellt, die Gruppe der Formel A zur Gruppe der Formel -CHOH-CH2 (IIc) reduziert und, wenn erwünscht, die erhaltene freie Verbindung in ein Salz oder ein erhaltenes Salz in die freie Verbindung oder ein anderes Salz überführt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man mittels eines Metallborhydrids reduziert.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man mittels eines Dileichtmetallhydrids reduziert.
4. Verfahren nach einem der Ansprüche 1 oder 3, dadurch gekennzeichnet, dass man mittels Lithiumaluminiumhydrid reduziert.
5. Verfahren nach einem der Ansprüche 1 oder 3, dadurch gekennzeichnet, dass man mittels Natrium-bis-(methoxy-äthoxy)-aluminiumhydrid reduziert.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass man die Reduktion in einem inerten Lösungsmittel vornimmt.
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass man ätherartige Lösungsmittel verwendet.
8. Verfahren nach einem der Ansprüche 6 bis 7, dadurch gekennzeichnet, dass man Tetrahydrofuran als Lösungsmittel verwendet.

Salze der Verbindung sind in erster Linie Säureadditionssalze und insbesondere pharmazeutisch verwendbare, nichttoxische Säureadditionssalze mit geeigneten organischen oder anorganischen Säuren. Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Die Reduktion der Gruppe A wird mittels eines geeigneten Reduktionsmittels, z.B. eines Metallborhydrids, z.B.
Natriumborhydrid, oder eines Dileichtmetallhydrids wie Natrium-bis-(methoxyäthoxy)-aluminiumhydrid oder insbesondere Lithiumaluminiumhydrid durchgeführt. Zweckmässigerweise wird die Reduktion in einem geeigneten Lösungsmittel, z. B. einem solchen inerten Charakters, vorgenommen. Hierzu eignen sich z. B. ätherartige Lösungsmittel wie solche aliphatischen Charakters, z. B. Diäthyläther oder Din-buthyläther, oder auch cyclische Verbindungen, wie z. B.
Dioxan oder insbesondere Tetrahydrofuran. Auf diese Weise werden z.B. in der Gruppe IIb beide Carbonylgruppen gleichzeitig unter Bildung der Gruppe IIc reduziert.
Ein Ausgangsstoff der Formel II kann, sofern er neu ist, auf übliche Weise erhalten werden. So kann man z. B.
2-Allyloxyphenol mit einer Verbindung der Formel X-CH2-CHOH-COOH (III), worin X eine geeignete Abgangsgruppe, z. B. eine reaktionsfähige veresterte Hydroxygruppe, etwa Halogen, z.B. Chlor oder eine mit einer Sulfonsäure veresterte Hydroxygruppe, z.B. die Methansulfonyloxy- oder die p-Toluolsulfonyloxygruppe, darstellt, zur entsprechenden 3 -(2-Allyloxyphenoxy)-2-hydroxy-propionsäure umsetzen und diese, etwa in Form eines reaktionsfähi- gen Derivats, z. B. ihres Anhydrids oder gemischten Anhydrids, etwa mit Pivalinsäure, mit Isopropylamin in die entsprechende Verbindung der Formel II mit der Gruppe IIa überführen.
Der Ausgangsstoff der Formel II mit der Gruppe IIb kann z.B. durch Umsetzung von 2-Allyloxyphenol mit einer Verbindung der Formel X-CH2-CO-CO-NH-CH(CH3)2 (IV), worin X obige Bedeutung hat und z. B. für Halogen, etwa Brom, steht, erhalten werden. Diese Umsetzungen werden in an sich bekannter Weise durchgeführt.
Die Erfindung wird durch die folgenden Beispiele erläutert. Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Eine Lösung von 6 g (0,02 M) rohem 3-(2-Allyloxy-phen oxy)-N-isopropylmilchsäureamid in 30 ml wasserfreiem Tetrahydrofuran wird bei 20-40" unter Rühren zu einer Suspension von 1,5 g (0,04 M) Lithiumaluminiumhydrid in 50 ml Tetrahydrofuran zugetropft. Das Reaktionsgemisch wird über Nacht unter Rückfluss gerührt, anschliessend unter Eiskühlung mit 6 ml Wasser und 6 ml konzentrierter Natronlauge zersetzt und filtriert. Das Filtrat wird eingedampft und der Rückstand in 100 ml Diäthyläther gelöst.
Die Lösung wird mit 20 ml 2n-Salzsäure extrahiert und aus dem sauren Extrakt die Base mittels Natronlauge freigesetzt. Üb- liche Aufarbeitung ergibt rohes 1 -(2-AlIyloxy-phenoxy)-3- isopropylamino-2-propanol; Smp. 77-79 ; das Hydrochlorid schmilzt bei 107-109 . Ausbeute an Base: 3,1 g, 57% d.Th.
Der Ausgangsstoff kann wie folgt hergestellt werden: a) Analog zu dem von G. Coppi et al., Arzneimittelforschung
16, 601 (1966), beschriebenen Verfahren erhält man aus 2 Allyloxyphenol und 3-Chlormilchsäure die 3-(2-Allyloxyphenoxy)-2-hydroxy-propionsäure vom Smp. 74-77" (aus Methanol).
b) Eine Lösung von 4,8 g 3-(2-Allyloxyphenoxy)-2-hy droxy-propionsäure in je 30 ml Dioxan und Dichlormethan
wird bei einer Temperatur von 0-10" mit 3,1 g Triäthylamin und 3,2 g Pivalinsäurechlorid versetzt und das Reaktionsgemisch 1 Stunde bei dieser Temperatur nachgerührt. Durch Eintropfen von 5 g Isopropylamin in das das gemischte Anhydrid enthaltende Reaktionsgemisch und dessen nach 6 Stunden erfolgender üblicher Aufarbeitung erhält man 5,3 g rohes 3-(2-Allyloxyphenoxy)-2-hydroxy-propionsäure-Nisopropylamid, welches als solches weiterverarbeitet wird.
Beispiel 2
Eine Lösung von 5 g rohem 3-(2-Allyloxyphenoxy)brenztraubensäure-N-isopropylamid in 30 ml wasserfreiem Tetrahydrofuran wird zu einer gerührten Suspension von 2,3 g Lithiumaluminiumhydrid in 50 ml Tetrahydrofuran zugetropft, wobei die Temperatur, wenn nötig durch Kühlen, bei 20-40" gehalten wird. Nach dem Kochen über Nacht unter Rückfluss wird das Reaktionsgemisch analog Beispiel 1 aufgearbeitet, wonach man 1 -(2-Allyloxy-phenoxy)-3-iso- propylamino-2-propanol als langsam kristallisierendes Öl erhält; Smp.77-79 ; das Hydrochlorid schmilzt bei 107-109 .
Ausbeute an Base: 2,7 g, 61% d.Th.
Der Ausgangsstoff kann auf folgende Weise hergestellt werden: Eine Suspension von 15,0 g 2-Allyloxy-phenol, 22,7 g 3-Brombrenztraubensäure-N-isopropylamid und 20 g Kaliumcarbonat in 300 ml Dioxan wird über Nacht unter Rückfluss gekocht. Das Reaktionsgemisch wird abgekühlt, filtriert und das Filtrat unter reduziertem Druck eingedampft. Der Eindampfrückstand wird in 200 ml Äthylacetat gelöst, die organische Phase mit 50 ml 2n-Natronlauge und anschliessend mit 50 ml Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Das so erhaltene rohe 3 (2-Allyloxy-phenoxy)-brenztraubensäure-N-isopropylamid wird ohne weitere Reinigung weiterverarbeitet.
