Es ist aus der österreichischen Patentschrift 241 436 beziehungsweise der deutschen Patentschrift 1 215 729 bekannt, dass bestimmte N,N'-Bis-[2-(3',4'-dihydroxyphenyl)-2- hydroxyäthyl]-polymethylendiamine, darunter vor allem das N,N'-B is- [2 -(3' ,4'-dihydroxyphenyl)-2-hydroxyäthyl]- hexamethylendiamin sehr gute broncholytische Eigenschaften besitzt. Zur Herstellung dieser Verbindung wurde das N,N' Bis-[2-(3',4'-dihydroxyphenyl)-2 -oxoäthyl]-hexamethylendiamin katalytisch hydriert.
Das N ,N' -Bi-[2-(3 ' ,4' -dihydroxyphenyl)-2-hydroxy- äthyl]-hexamethylendiamin besitzt zwei gleichartige asymmetrische C-Atome, so dass es neben der Racemform, eine Mesoform sowie die D- und L-Form dieser Verbindung gibt.
Bei dem Verfahren der österreichischen Patentschrift 241 436 werden durch die Reduktion der beiden Ketogruppen die asymmetrischen C-Atome erst gebildet und da die Bildung einer bestimmten Stereoform dabei nicht begünstigt ist, erhält man bei diesem Verfahren ein Gemisch der Racemform und der Mesoform. Die einzelnen stereoisomeren Formen müssten daher erst aus dem Gemisch durch Isomerentrennung hergestellt werden, was äusserst schwierig ist. Die Herstellung der optisch aktiven Formen, insbesondere der L Form, ist aber interessant, da diese nicht nur stärker, sondern auch länger wirksam ist als die D-Form.
Ferner zeigte sich, dass das Verfahren der österreichischen Patentschrift 241 436 zu chromatographisch nicht völlig einheitlichen Produkten führt, da zum Teil die Reduktion nicht vollständig ist, zum Teil aber weiterreduzierte Produkte als Nebenprodukte anfallen, bei denen ein Sauerstoffatom völlig abreduziert ist. Diese Verunreinigungen, die für die Verwendung als Pharmazeutikum störend sind, können auch durch weitere Reinigung nicht beseitigt werden.
Da für die Herstellung der optisch aktiven Formen ein Verfahren gewählt werden muss, bei dem die aktiven Kohlenstoffatome schon vor der Kondensation mit der Hexamethylenkette vorgebildet sind, bietet sich dafür vor allem eine Umsetzung von 2 Mol des entsprechenden Phenyläthanolaminderivates mit Dihalogenhexan als möglicher Weg an. Bei Durchführung dieser Umsetzung, beispielsweise von 1,6-Dibromhexan mit 1-(3',4'-Dibenzyloxyphenyl)-2-benzylamino äthanol-(1) im Molverhältnis 1:4 in der üblichen Weise in verschiedenen inerten Lösungsmitteln wie Benzol, Toluol, Xylol, Decan, Dimethylformamid oder Acetonitril, wobei 2 Mol der Aminkomponente als säurebindendes Mittel dienen, werden aber sehr niedrige Ausbeuten und obendrein unreine Produkte erhalten.
Lediglich in aliphatischen Alkoholen findet bei erhöhter Temperatur, beispielsweise von 65-120 C, ein befriedigender Umsatz statt, doch sind die Produkte bezüglich Reinheit sehr unbefriedigend.
Überraschenderweise zeigte sich, dass die Umsetzung von 1,6-Dihalogenhexanen mit dem Phenyläthanolaminderivat doch in guter Ausbeute zu Produkten mit befriedigender Reinheit, auch der optischen Reinheit, bei der Herstellung von optisch aktiven Formen, führen kann, wenn man die Umsetzung in Abwesenheit von Lösungsmitteln in der Schmelze durchführt. Dies ist sehr überraschend, da gerade bei einer solchen Umsetzung mit den bekannten Schwierigkeiten der Homogenisierung des Reaktionsgemisches und der höheren Temperaturbelastung (es werden Temperaturen von 120160 C benötigt) eine niedrige Ausbeute und ein verunreinigtes Endprodukt zu erwarten gewesen wäre.
Gegenstand der vorliegenden Erfindung ist demnach ein Verfahren zur Herstellung des optisch inaktiven oder optisch aktiven N,N'-Bis-[2-(3',4'-dihydroxyphenyl)-2-hydroxy- äthyl]-hexamethylen-diamins und der Salze desselben, das dadurch gekennzeichnet ist, dass mindestens 4 Mol der Racemform, der L-Form oder der D-Form eines Phenyl äthanolaminderivates der allgemeinen Formel

in der R einen hydrogenolytisch abspaltbaren Rest, vorzugsweise den Benzyl- oder Benzhydrylrest, und R' Wasserstoff oder einen hydrogenolytisch abspaltbaren Rest, vorzugsweise den Benzyl- oder Benzhydrylrest, bedeuten, in der Schmelze mit 1 Mol eines 1,6-Dihalogenhexans umgesetzt, das Reaktionsprodukt zur Abspaltung der Schutzgruppen katalytisch hydriert und die Verfahrensprodukte gewünschtenfalls in Salze übergeführt werden.
Die erfindungsgemässe Reaktion geht sehr rasch vor sich.
Während die Umsetzungen in Lösungsmitteln mindestens 8-10 Stunden benötigten, um zu halbwegs tragbaren Ausbeuten zu führen, ist die erfindungsgemässe Umsetzung in der Schmelze nach stündiger Reaktionszeit oder sogar noch weniger bereits vollständig. Die Reaktionstemperatur beträgt dabei zweckmässig etwa 120-160" C. Eine Schutzgasatmosphäre ist nicht erforderlich.
Auch die Aufarbeitung gestaltet sich sehr einfach, was ein weiterer Vorteil des erfindungsgemässen Verfahrens ist.
Nach Erkalten der Schmelze wird diese z. B. mit einem geeigneten Lösungsmittel, z. B. Diäthyläther digeriert, wobei die gewünschte Base in Lösung geht, während das gebildete Hydrohalogenid des Phenyläthanolaminderivates der Formel I in kristalliner Form zurückbleibt und von der Lösung abgetrennt wird. Aus diesem Hydrohalogenid kann wieder die reine Einsatzbase der Formel I zurückgewonnen werden, so dass der zur Säurebindung nötige Überschuss an der Verbindung der Formel I nicht verlorengeht, was besonders bei der Herstellung der optisch aktiven Formen wichtig ist, da die optisch aktive Einsatzbase möglichst vollständig verwertet werden muss, soll das Verfahren wirtschaftlich tragbar sein.
Die das Reaktionsprodukt enthaltende Lösung wird eingeengt, wobei das N,N'-Bis-[2-(3',4'-Dihydroxyphenyl)- 2-hydroxyäthyl]-hexamethylendiamin, dessen phenolische OH-Gruppen und ev. das Stickstoffatom noch geschützt sind, so rein gewonnen wird, dass es direkt der katalytischen Hy drierung zur Abspaltung dieser Schutzgruppen unterworfen werden kann. Für diese Abspaltung ist jede übliche Methode geeignet, insbesondere die Hydrierung in Gegenwart von Palladiumkohle in Eisessig. Aus der Hydrierlösung kann das erwünschte Endprodukt entweder als Base oder als Salz gewonnen werden.
Die Herstellung der als Ausgangsmaterial dienenden optisch aktiven Formen des substituierten Phenyläthanolamins der Formel I gelingt auf übliche Weise durch fraktionierte Kristallisation von deren Salzen mit einer optisch aktiven Säure. Als solche kann z. B. die Weinsäure dienen, als Lösungsmittel gibt Dimethylformamid oder Gemische von Dimethylformamid und Wasser günstige Ergebnisse bei der Trennung. Diese Trennung bewährt sich in erster Linie an Verbindungen der Formel I, in der R' ein Wasserstoffatom darstellt. Die entsprechende N-Benzylverbindung ist dann aus solchen am N-Atom nicht benzylierten, optisch aktiven Basen leicht zugänglich. So ist es z. B. möglich, diese mit Benzaldehyd zur entsprechenden Benzalverbindung umzu setzen und anschliessend zu hydrieren, ohne dass die optische Konfiguration geändert würde.
Die in den nachfolgenden Beispielen angegebenen Teile sind Gewichtsteile.
Beispiel 1
7,03 Teile 1 -(3' ,4'-Dibenzyloxyphenyl)-2-benzylamino- äthanol-(1) und 0,98 Teile 1,6-Dibromhexan werden ge schmolzen und 1 Stunde bei 135 0C B adtemperatur gerührt.
Nach dem Erkalten wird das Reaktionsgemisch dreimal mit je 100 Volumteilen Äther ausgerührt. Dabei bleibt das gebildete Hydrobromid des 1-(3',4' -Dibenzyloxyphenyl) -2benzylamino-äthanols-(1) in kristallisierter Form ungelöst und wird von der Lösung abgetrennt. Es sind 4,05 Gewichtsteile (theor. 4,09), Fo.: 139-142 C. Die Ätherlösung wird auf 40 Volumteile eingeengt, wobei die Base auskristallisiert.
Sie wird isoliert, mit Äther gewaschen und getrocknet. Man erhält 2,80 Teile N,N'-Dibenzyl-N,N' -bis-[2-(3',4'-di- benzyloxyphenyl)-2-hydroxyäthyl]-hexamethylendiamin, das sind 73 % der Theorie. Fp: 73-750 C.
5,0 Teile N,N' -Dibenzyl-N,N'-bis-[2 -(3',4' -dibenzyl- oxyphenyl)-2-hydroxyäthyl]-hexamethylendiamin werden in 70 Volumteilen Eisessig gelöst und in Gegenwart von 0,5 Gewichtsteilen Palladiumkohle (10%ig) bei Raumtemperatur und Normaldruck hydriert. Nach beendeter Wasserstoffaufnahme wird der Katalysator abgetrennt und die Eis essiglösung mit der berechneten Menge alkoholischer Salzsäure versetzt und im Vakuum zur Trockene eingedampft.
Der Abdampfrückstand wird in 25 Volumteilen Äthanol suspendiert und das Salz isoliert. Man erhält 2,26 Teile N,N' Bis- [2-(3' ,4' -dihydroxyphenyl)-2-hydroxyäthyl]-hexame- thylendiamin-Dihydrochlorid vom Fp: 197-198 C.
Das als Ausgangsmaterial verwendete 1-(3',4'-Dibenzyloxyphenyl)-2-benzylaminoäthanol-(1) wird aus Dibenzyloxyphenyl)-2-aminoäthanol durch Umsetzung mit Benzaldehyd zur Schiffschen Base und anschliessende Reduktion mit Lithiumaluminiumhydrid erhalten.
Beispiel 2
17,56 Teile L-(-)-1-(3',4'-Dibenzyloxyphenyl)-2- benzylamino-äthanol-(1) ([a]5456-19,5 ) werden geschmolzen und mit 2,44 Teilen 1,6-Dibromhexan versetzt.
Das Gemisch wird unter Rühren auf 1200 C erhitzt. Es tritt eine Eigenerwärmung von etwa 8" C ein, die etwa 5 Minuten anhält. Die Temperatur wird weitere 30 Minuten bei 125 bis 1300 C gehalten. Nach Aufarbeitung analog zu Beispiel 1 erhält man 6,10 Teile (63,5% der Theorie) reines L-(-)-N,N' Dib enzyl -N, N' -bis- [2-(3',4' -dib enzyloxyphenyl)-2 -hy- droxyäthyl] -hexamethylendiamin .
MFp.: 98-1000 C [α]524162,25 , [a]5%18-27,250, [aJn21 35,60 (Eisessig).
Bei der Aufarbeitung werden 10,6 Teile Hydrobromid des L-(-)-1-(3' ,4'-dibenzyloxyphenyl)-2-benzylamino-äthanol (1) rückgewonnen, das entspricht rund 50% der eingesetzten Basenmenge; MFp.: 162-166 C [a]t54 6-16,0 , [α]5-14,0 , [a]20 -13,4
Aus diesem Salz kann die Ausgangsbase mit einem praktisch unveränderten spezifischen Drehwert (R = 546) von -19,3 C gewonnen werden.
Die Abspaltung der Benzylreste erfolgt wie in Beispiel 1 beschrieben. Man erhält dabei das rohe Dihydrochlorid in einer Ausbeute von 89% der Theorie. Nach Lösen in 2,5 Volumteilen heissem Wasser und Fällen mit Isopropanol erhält man das reine L-(-)-N,N'-Bis-[2-(3',4'-Dihydroxyphenyl)-2-hydroxyäthyl]-hexamethylendiamin-Dihydrochlo- rid mit dem MFp: 197-200"C (Zersetzung) und den Drehwerten [α]22-38,8 , [α]5272-34,10, [a]D233,1 (C = 1, Wasser).
Wird die Hydrierung auf gleiche Weise durchgeführt, aber nach Abtrennung des Katalysators die Eisessiglösung mit einer Lösung von 2,5 Teilen Natriumsulfat in 35 Volumteilen Wasser versetzt, so kristallisiert aus der Lösung das Sulfat aus. Die Ausbeute liegt bei 75% der Theorie.

Beispiel 3
5,85 Teile 1-(3',4'-Dibenzyloxyphenyl)-2-benzylamino- äthanol-(1) und 0,52 Teile 1,6-Dichlorhexan werden geschmolzen und 1 Stunde bei 1600 C gerührt. Wird die Schmelze wie in Beispiel 1 der Patentanmeldung aufgearbeitet, werden 2,7 Teile Hydrochlorid des 1-(3',4'-Dibenzyloxyphenyl)-2-benzylaminoäthanol-1 und eine entsprechende Menge N,N' -Dibenzyl-N,N' -bis-[2-(3',4' -dibenzyloxy- phenyl)-2-hydroxyäthylj-hexanaethylendiamin erhalten.

It was also shown that the process of Austrian patent specification 241 436 leads to chromatographically not completely uniform products, since the reduction is partly incomplete, but partly further reduced products are obtained as by-products in which one oxygen atom is completely reduced. These impurities, which are disruptive for use as a pharmaceutical, cannot be removed even by further cleaning.
Since a process has to be chosen for the production of the optically active forms, in which the active carbon atoms are already formed before the condensation with the hexamethylene chain, a reaction of 2 moles of the corresponding phenylethanolamine derivative with dihalohexane is a possible route. When carrying out this reaction, for example, of 1,6-dibromohexane with 1- (3 ', 4'-dibenzyloxyphenyl) -2-benzylamino ethanol (1) in a molar ratio of 1: 4 in the usual manner in various inert solvents such as benzene, toluene , Xylene, decane, dimethylformamide or acetonitrile, 2 mol of the amine component serving as an acid-binding agent, but very low yields and impure products on top of that are obtained.
Only in aliphatic alcohols does a satisfactory conversion take place at elevated temperature, for example from 65-120 ° C., but the products are very unsatisfactory in terms of purity.
Surprisingly, it was found that the reaction of 1,6-dihalohexanes with the phenylethanolamine derivative can lead in good yield to products with satisfactory purity, including optical purity, in the production of optically active forms, if the reaction is carried out in the absence of solvents performs in the melt. This is very surprising, since a low yield and a contaminated end product would have been expected precisely in such a reaction with the known difficulties of homogenizing the reaction mixture and the higher temperature load (temperatures of 120-160 ° C. are required).

The pure starting base of the formula I can be recovered from this hydrohalide, so that the excess of the compound of the formula I necessary for acid binding is not lost, which is particularly important in the production of the optically active forms, since the optically active starting base is utilized as completely as possible must be, the process should be economically viable.
The solution containing the reaction product is concentrated, the N, N'-bis [2- (3 ', 4'-dihydroxyphenyl) -2-hydroxyethyl] hexamethylene diamine, its phenolic OH groups and possibly the nitrogen atom still being protected , is obtained so pure that it can be directly subjected to catalytic hydrogenation to split off these protective groups. Any customary method is suitable for this elimination, in particular hydrogenation in the presence of palladium carbon in glacial acetic acid. The desired end product can be obtained from the hydrogenation solution either as a base or as a salt.
The optically active forms of the substituted phenylethanolamine of the formula I, which are used as starting material, are prepared in the customary manner by fractional crystallization of their salts with an optically active acid. As such, e.g. B. the tartaric acid are used, as a solvent, dimethylformamide or mixtures of dimethylformamide and water give favorable results in the separation. This separation has proven its worth primarily on compounds of the formula I in which R 'represents a hydrogen atom. The corresponding N-benzyl compound is then easily accessible from optically active bases which are not benzylated on the N atom. So it is e.g. B. possible to implement this with benzaldehyde to the corresponding benzal compound and then to hydrogenate without changing the optical configuration.
The parts given in the following examples are parts by weight.
example 1
