Die Erfindung betrifft ein Verfahren zur Herstellung neuer Benzocycloheptenderivate der Formel I, worin
R1 für Wasserstoff, Alkyl mit 1-4 Kohlenstoffatomen, Alkoxy mit 1-4 Kohlenstoffatomen oder Halogen mit einer Atomzahl von 9 bis 35 steht und
R2 für Wasserstoff oder, falls R1 Alkoxy mit 1-4 Kohlenstoffatomen bedeutet, auch für Alkoxy mit 1-4 Kohlenstoffatomen steht oder
R1 und R2, falls sie an benachbarte Kohlenstoffatome gebunden sind, auch gemeinsam eine Methylendioxygruppe bilden können,
R3 Alkyl mit 1-4 Kohlenstoffatomen, Cycloalkylalkyl mit 410 Kohlenstoffatomen, Cycloalkyl mit 3-7 Kohlenstoffatomen oder eine gegebenenfalls durch Chlor,

Das strukturell naheliegende 7-Dimethylamino-5,6,7,8tetrahydro-7-phenyl-7H-benzocyclohepten ist aus Current Science, 46, Nr. 6, 174-176 (1977) bekannt (20. März 1977).
Die in den Verbindungen der Formel I vorstehend definierten Alkyl-, Alkoxy- oder Alkylthiogruppen enthalten vorzugsweise 1 oder 2 Kohlenstoffatome und stellen insbesondere Methyl, Methoxy oder Methylthio dar. Sofern ein Substituent für vorstehend definiertes Halogen steht, stellt er vorzugsweise Chlor dar.
Die Substituenten R1 und R2 stehen vorzugsweise für Wasserstoff. Sonstige Substituenten R1 oder R2 sind vorzugsweise in 2- oder 3-Stellung des Benzocycloheptangerüstes angeordnet.
Sofern R3 eine vorstehend definierte Cycloalkylgruppe
darstellt, enthält diese vorzugsweise 5 oder 6 Kohlenstoffatome. Durch R3 symbolisierte vorstehend definierte Cycloalkylalkylgruppen enthalten vorzugsweise 3 bis 6 Kohlenstoffatome im Cycloalkylrest und 1 oder 2 Kohlenstoffatome in der Alkylenkette.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man in Verbindungen der Formel V, worin R1, R2, R3 und R4 obige Bedeutung besitzen, durch Umsetzung mit Grignard-Verbindungen der Formel VI, worin Z obige Bedeutung besitzt und X für Chlor, Brom oder Jod steht, die Cyangruppe in eine Gruppe Z überführt und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säu- readditionssalze überführt.
Die Umsetzung der Aminonitrilverbindungen der Formel V mit Grignard-Verbindungen der Formel VI kann auf an sich bekannte Weise durchgeführt werden. Beispielsweise kann die Umsetzung in einem für Grignard-Reaktionen geeigneten Lösungsmittel, z. B. einem offenkettigen oder cyclischen Äther wie Diäthyläther oder Tetrahydrofuran durchgeführt werden.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I können in Form der freien Basen oder ihrer Säureadditionssalze vorliegen. Die freien Basen können auf an sich bekannte Weise in ihre Säureadditionssalze überführt werden und umgekehrt. Zur Säureadditionssalzbildung können z.B.
folgende Säuren verwendet werden: Chlorwasserstoff-, Schwefel-, Fumar-, Malein- oder Naphthalin-1,5-disulfonsäuren.
Die Ausgangsverbindungen der Formel V können ausgehend von Verbindungen der Formel VII, worin R, und R2 obige Bedeutung besitzen, erhalten werden, indem man diese mit Acetondicarbonsäureäthylester umsetzt, die erhaltenen Reaktionsprodukte der Formel VIII, worin Rl und R2 obige Bedeutung besitzen, anschliessend nach an sich bekannten Methoden reduziert und decarboxyliert und die so erhaltenen Verbindungen der Formel IX, worin Rl und R2 obige Bedeutung besitzen, durch Umsetzung mit einem Alkalimetallcyanid und einem Säureadditionssalz einer Verbindung der Formel X, worin R3 und R4 obige Bedeutung besitzen, in Verbindungen der Formel V überführt.
Die Umsetzung der Verbindungen der Formel IX mit dem Alkalimetallcyanid und dem Salz der Verbindung der Formel X kann in einem protischen Lösungsmittel, vorzugsweise einem Alkohol/ Wassergemisch, vorzugsweise bei Raumtemperatur durchgeführt werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre physiologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakologische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Die Verbindungen sind brauchbar, da sie pharmakologische Aktivität in Tieren zeigen. Insbesondere besitzen sie für Antidepressiva typische Eigenschaften und können daher als Antidepressiva zur Prophylaxe und Therapie von Depressionen in der Psychiatrie Verwendung finden. An Tetrabenazin-behandelten Ratten bewirken die Substanzen z. B. in Dosen von 0,5-5 mg/kg eine für Antidepressiva typische Hemmung der durch Tetrabenazin hervorgerufenen Symptome.
Für die vorgenannte Anwendung variieren die zu verwendenden Dosen naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Jedoch werden im allgemeinen befriedigende Resultate mit einer Tagesdosis von ca. 0,0015 bis 40 mg/kg Körpergewicht Tier erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere kann die Tagesdosis z. B. zwischen 0,1 und 100 mg/kg betragen. So enthalten z. B. für orale Applikationen die Teildosen etwa 0,025 bis 50 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Ausserdem eignen sich die Verbindungen der Formel I als Psychostimulantien zur Stimulierung des Zentralnervensystems, zur Antriebssteigerung sowie zur Behandlung von Störungen der Vigilanz, z. B. zur Behandlung der Cerebralinsuffizienz, da sie auf das Zentralnervensystem stimulierend und vigilanzerhöhend wirkende Eigenschaften besitzen, wie aus pharmakologischen Standardversuchen hervorgeht. Beispielsweise zeigt sich bei der Beobachtung der Wirkungen dieser Verbindungen auf das spontane Verhalten von Mäusen, dass die Verbindungen in Dosen von ca. 15 bis ca.
100 mg/kg Körpergewicht i. p. eine Steigerung der Erregbarkeit der Mäuse bewirken. In Ratten führen die Verbindungen in Dosen zwischen ca. 50 und 200 mg/kg Körpergewicht s. c. zu einer Erhöhung der motorischen Aktivität.
Für die vorgenannte Anwendung variieren die zu verwendenden Dosen naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Jedoch werden im allgemeinen befriedigende Resultate mit einer Tagesdosis von ca. 0,01 bis 200 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 1 bis 500 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 0,25 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I und ihre physiologisch verträglichen Säureadditionssalze zusammen mit üblichen pharmazeutischen Hilfsstoffen in galenischen Zubereitungen wie z. B. Tabletten, Kapseln oder Lösungen enthalten sein. Diese galenischen Zubereitungen können nach an sich bekannten Methoden dargestellt werden.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1: 7-Dimethylamino-5,6,8,9-tetrahydro-7-(2-thienyl)-7H-ben- zocyclohepten
Zu einer Lösung von 2-Thienylmagnesiumbromid (hergestellt aus 39,2 g 2-Bromthiophen und 5,6 g Magnesiumspäne) in 100 ml wasserfreiem Äther lässt man eine Lösung von 10,0 g 7-Dimethylamino-5,6,8,9-tetrahydro-7H-ben- zocyclohepten-7-carbonitril in 150 ml wasserfreiem Äther innerhalb von 1 Stunde bei Raumtemperatur zutropfen. Das Reaktionsgemisch wird noch 30 Minuten weitergerührt, dann unter starkem Kühlen tropfenweise mit 90 ml 20%iger Ammoniumchloridlösung versetzt. Nach Abtrennen der organischen Phase wird die wässrige Lösung nochmals mit Ather ausgeschüttelt, die vereinigten organischen Lösungen werden mit gesättigter Kochsalzlösung gewaschen, über Kaliumcarbonat getrocknet und eingedampft.
Die als öliger Rückstand verbleibende Titelverbindung wird aus Äther/Pe troläther kristallisiert. Smp.: 98-100 .
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Zu einer Lösung von 14,2 g Kaliumcyanid in 80 ml Äthanol und 20 ml Wasser wird zuerst 17,8 g Dimethylaminhydrochlorid und dann eine Lösung von 35,0 g 5,6,8,9-Tetrahydro-7H-benzocyclohepten-7-on in 50 ml Äthanol innert 1,5 Stunden zugegeben. Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt, stark eingeengt, mit Eiswasser verdünnt und durch Zugabe von Kalilauge leicht alkalisch gestellt. Die ausgefallene Substanz wird mit Äther extrahiert, die ätherischen Extrakte werden mit Wasser gewaschen, über Kaliumkarbonat getrocknet und eingedampft und das als Rückstand verbleibende 7-Dimethylamino5,6,8,9-tetrahydro-7H-benzocyclohepten-7-carbonitril wird aus Äther/Petroläther umkristallisiert. Smp.: 1 lO-112 .
Analog Beispiel 1 werden auch die folgenden Verbindungen der Formel I durch Umsetzung entsprechender Verbindungen der Formel V mit entsprechenden Grignard-Verbindungen der Formel VI erhalten:

The substituents R1 and R2 are preferably hydrogen. Other substituents R1 or R2 are preferably arranged in the 2- or 3-position of the benzocycloheptane skeleton.
If R3 is a cycloalkyl group as defined above
represents, preferably contains 5 or 6 carbon atoms. Cycloalkylalkyl groups as defined above by R3 preferably contain 3 to 6 carbon atoms in the cycloalkyl radical and 1 or 2 carbon atoms in the alkylene chain.
According to the invention, the new compounds of the formula I and their acid addition salts are obtained by reacting in compounds of the formula V in which R1, R2, R3 and R4 have the above meaning, by reaction with Grignard compounds of the formula VI in which Z has the above meaning and X represents chlorine, bromine or iodine, the cyano group is converted into a group Z and, if desired, the compounds of the formula I are converted into their acid addition salts.
The reaction of the aminonitrile compounds of the formula V with Grignard compounds of the formula VI can be carried out in a manner known per se. For example, the reaction in a solvent suitable for Grignard reactions, e.g. B. an open chain or cyclic ether such as diethyl ether or tetrahydrofuran.
The compounds of the formula I obtained according to the invention may be in the form of the free bases or their acid addition salts. The free bases can be converted into their acid addition salts in a manner known per se and vice versa. For acid addition salt formation e.g.
the following acids are used: hydrochloric, sulfuric, fumaric, maleic or naphthalene-1,5-disulfonic acids.

The compounds of the formula I and their physiologically tolerable acid addition salts have not hitherto been described in the literature. They are characterized by interesting pharmacological properties and can therefore be used as a remedy.
The compounds are useful because they show pharmacological activity in animals. In particular, they have properties typical of antidepressants and can therefore be used as antidepressants for the prophylaxis and therapy of depression in psychiatry. On tetrabenazine-treated rats, the substances cause e.g. B. in doses of 0.5-5 mg / kg an antidepressant typical inhibition of the symptoms caused by tetrabenazine.
For the aforementioned application, the doses to be used naturally vary depending on the type of substance, the administration and the condition to be treated. However, generally satisfactory results are obtained with a daily dose of approximately 0.0015 to 40 mg / kg of animal body weight. If necessary, this dose can be administered in 2 to 4 portions or as a slow-release form. For larger mammals, the daily dose can be e.g. B. between 0.1 and 100 mg / kg. So contain z. B. for oral applications, the partial doses of about 0.025 to 50 mg of the compounds of the formula I in addition to solid or liquid carriers.
In addition, the compounds of formula I are suitable as psychostimulants for stimulating the central nervous system, for increasing drive and for treating disorders of vigilance, e.g. B. for the treatment of cerebral insufficiency, since they have stimulating and vigilance-increasing properties on the central nervous system, as can be seen from standard pharmacological experiments. For example, observing the effects of these compounds on the spontaneous behavior of mice shows that the compounds in doses from about 15 to about
100 mg / kg body weight i. p. increase the excitability of the mice. In rats, the compounds lead in doses between approx. 50 and 200 mg / kg body weight s. c. to an increase in motor activity.
For the aforementioned application, the doses to be used naturally vary depending on the type of substance, the administration and the condition to be treated. However, generally satisfactory results are obtained with a daily dose of approximately 0.01 to 200 mg / kg body weight. If necessary, this dose can be administered in 2 to 4 portions or as a slow-release form. For larger mammals, the daily dose is around 1 to 500 mg. So contain z. B. for oral applications, the partial doses about 0.25 to 250 mg of the compounds of formula I in addition to solid or liquid carriers.
As a remedy, the compounds of formula I and their physiologically acceptable acid addition salts together with conventional pharmaceutical auxiliaries in pharmaceutical preparations such as. B. tablets, capsules or solutions may be included. These pharmaceutical preparations can be prepared by methods known per se.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all the temperatures are given in degrees Celsius.
Example 1: 7-Dimethylamino-5,6,8,9-tetrahydro-7- (2-thienyl) -7H-benzocycloheptene
A solution of 10.0 g of 7-dimethylamino-5,6,8,9- is added to a solution of 2-thienylmagnesium bromide (made from 39.2 g of 2-bromothiophene and 5.6 g of magnesium shavings) in 100 ml of anhydrous ether. Add tetrahydro-7H-benzocyclohepten-7-carbonitrile in 150 ml of anhydrous ether within 1 hour at room temperature. The reaction mixture is stirred for a further 30 minutes, then 90 ml of 20% ammonium chloride solution are added dropwise with vigorous cooling. After the organic phase has been separated off, the aqueous solution is shaken out again with ether, the combined organic solutions are washed with saturated sodium chloride solution, dried over potassium carbonate and evaporated.
The title compound remaining as an oily residue is crystallized from ether / petroleum ether. M.p .: 98-100.
The starting material can be produced as follows:
To a solution of 14.2 g of potassium cyanide in 80 ml of ethanol and 20 ml of water is first 17.8 g of dimethylamine hydrochloride and then a solution of 35.0 g of 5,6,8,9-tetrahydro-7H-benzocyclohepten-7-one added in 50 ml of ethanol within 1.5 hours. The reaction mixture is stirred for 24 hours at room temperature, strongly concentrated, diluted with ice water and made slightly alkaline by adding potassium hydroxide solution. The precipitated substance is extracted with ether, the ethereal extracts are washed with water, dried over potassium carbonate and evaporated, and the 7-dimethylamino5,6,8,9-tetrahydro-7H-benzocyclohepten-7-carbonitrile remaining as residue is converted from ether / petroleum ether recrystallized. M.p .: 1 10-112.
