Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen D-Homosteroiden aus der Pregnanreihe der Formel

Salzes und Rt7a Hydroxy oder Acyloxy darstellen und die gestrichelte
1,2-Bindung fakultativ ist.
Der Ausdruck Halogen umfasst Fluor, Chlor, Brom und Jod. Eine Acyloxygruppe kann sich von einer gesättigten oder ungesättigten aliphatischen Monocarbonsäure, einer cycloaliphatischen, araliphatischen oder einer aromatischen Monocarbonsäure mit vorzugsweise bis zu 15 C-Atomen ableiten. Beispiele solcher Säuren sind Ameisen-, Essig-, Pivalin-, Propion-, Butter-, Capron-, Oenanth-, Undecylen-, Ö1-, Cyclopentylpropion-, Cyclohexylpropion-, Phenylessigund Benzoesäure. Besonders bevorzugt sind Ci 7-Alkanoyl- oxygruppen. Ein Rest einer Di- oder Tricarbonsäure kann sich z. B. ableiten von der Oxal-, Malon-, Bernstein-, Fumar-, Äpfel-, Wein- oder Zitronensäure, vorzugsweise von der Bernsteinsäure. Als wasserlösliche Salze solcher Säurereste kommen vor allem die Alkalimetallsalze wie Na-, K- oder Ammoniumsalze in Frage.
Von den in 6-Stellung substituierten Verbindungen sind die 6a-Isomeren bevorzugt.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man an die 9,11-Doppelbindung eines D Homosteroids der Formel

worin R2i2 Wasserstoff oder R21 darstellt und R2i, RO, Rl7a sowie die gestrichelte 1,2-Bindung die oben angegebenen Bedeutungen haben, entsprechendes Halogen anlagert.
Zwecks Durchführung des erfindungsgemässen Verfahrens löst man das Ausgangssteroid III zweckmässigerweise in einem geeigneten Lösungsmittel, z. B. einem Äther, wie Tetrahydrofuran, oder Dioxan, einem chlorierten Kohlenwasserstoff, wie Methylenchlorid oder Chloroform, oder einem Keton, wie Aceton, und Iässt Halogen einwirken. Als Halogen kann Cl2, Br2, BrF, ClF oder ClBr eingesetzt werden.
Die Ausgangsstoffe können, soweit sie nicht bekannt oder nachstehend beschrieben sind, in Analogie zu bekannten oder in den Beispielen beschriebenen Methoden dargestellt werden.
Die D-Homosteroide der Formel I sind endokrin, insbesondere antiinflammatorisch wirksam. Sie sind durch Selektivität der Wirkung gekennzeichnet, z. B. stark topisch wirksam bei schwächerer systemischer Wirkung.
Die Verfahrensprodukte können als Heilmittel z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline usw., enthalten. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees, Suppositorien, Kapseln; in halbfester Form, z. B. als Salben; oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw.
oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Als Richtlinie für den Dosisbereich topischer Präparate kann etwa 0,01-1 /0, bei systemischen Präparaten etwa 0,1 bis 10 mg pro Applikationseinheit gelten.
Die Herstellung der Arzneimittel kann in an sich bekannter Weise erfolgen, indem man die Verbindungen der Formel I mit zur therapeutischen Verabreichung geeigneten, nicht-toxischen, an sich in solchen Präparaten üblichen festen und/oder flüssigen Trägermaterialien, wie z. B. den vorstehend genannten, vermischt und gegebenenfalls in die gewünschte Form bringt.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
400 mg 21-Acetoxy-17a-hydroxy-D-homopregna-4,9(1 1)- dien-3,20-dion werden in 5 ml Chloroform tropfenweise mit 1,2 ml chlorgesättigtem Tetrachlorkohlenstoff versetzt.
Nach 1 Stunde wird die Chloroformlösung mit Wasser gewaschen, getrocknet und eingedampft. Nach Chromatographie an Kieselgel erhält man 180 mg 21-Acetoxy-9a,11ss-dichlor17a-hydroxy-D-homopregn-4-en-3,20-dion. Smp. 225-2280, UV: E240 = 15800, [alD = + 1500, (c = 0,103 O/o in Dioxan).
Das Ausgangsmaterial kann wie folgt hergestellt werden:
D-Homohydrocortisonacetat wird in Dimethylformamid in Gegenwart von Pyridin bei erhöhter Temperatur mit Methansulfonylchlorid dehydratisiert. Man erhält 21-Acetoxy 17a-hydroxy-D-homopregna-4,9(11)-dien-3,20-dion, Smp.
238-2400 C, [a]D = + 710 (c = 0,104 in Dioxan); e239 = 16750.
Beispiel 2 -Auf gleiche Weise wie in Beispiel 1 beschrieben, erhält man aus 400 mg 21-Acetoxy- 17a-hydroxy-D-homopregna Acetoxy-9a, 1 1fl-dichlor-17a-hydroxy-D-homopregna-1 4- dien-3,20-dion. Smp. 222-2240, UV: E237 = 15300, [alD = + 1420 (c = 0,100 0/0 in Dioxan).
Das Ausgangsmaterial kann durch 1,2-Dehydrierung von 21-Acetoxy-17a-hydroxy-D-homopregna-4,9(11)-dien-3,20- dion mittels 2,3-Dichlor-5,6-dicyanobenzochinon erhalten werden.
Beispiel 3
400 mg 21-Acetoxy-17a-hydroxy-D-homopregna-4,9(1 1)- dien-3,20-dion und 200 mg N-Chlorsuccinimid werden in 4 ml einer Lösung aus Fluorwasserstoff und Harnstoff im Gewichtsverhältnis von 1:1,25 während 1 Stunde bei Raumtemperatur gerührt. Das Reaktionsgemisch wird auf eine wässrige Lösung von Natriumhydrogencarbonat und Natriumsulfit gegossen und mit Methylenchlorid extrahiert. Die Methylenchloridlösungen werden mit Wasser gewaschen, getrocknet und eingedampft. Nach Chromatographie erhält man aus Methylenchlorid-Aceton 130 mg 21-Acetoxy-17a-hydroxy 9a-chlor-11fl-fluor-D-homopregn-4-en-3,20-dion. Smp. 238 bis 2390, UV: e238 = 16350, [a]D = + 131" (c = 0,102 0/o in Dioxan).
Beispiel 4
Auf gleiche Weise wie in Beispiel 3 beschrieben, erhält man aus 400 mg 1-Acetoxy- 17a-hydroxy-D-homopregna- 1,4,9(1 1)-trien-3,20-dion 270 mg 21-Acetoxy-9a-chlor-1 1ss- fluor-17a-hydroxy-D-homo-pregna-1,4-dien-3,20-dion. Smp.

Beispiel 5
In zu Beispiel 4 analoger Weise erhält man 9a-Chlor-llss- nuor- 17a,21-dihydroxy-D-homopregna-1,4-dien-3 ,20-dion, UV: e237 = 15150, 21-Acetoxy-6a,9a-dichlor-llss-fluor-17a- hydroxy-D-homopregn-4-on-3,20-dion, UV: e236 = 14500, unc 21-Acetoxy-6ss,9a-dichlor-11ss-fluor-17a-hydroxy-D-homopregn-4-en-3,20-dion, UV: e239 = 14800.

The term halogen includes fluorine, chlorine, bromine and iodine. An acyloxy group can be derived from a saturated or unsaturated aliphatic monocarboxylic acid, a cycloaliphatic, araliphatic or an aromatic monocarboxylic acid with preferably up to 15 carbon atoms.
