Die Erfindung betrifft ein Verfahren zur Herstellung von Benzylpyrimidinen der Formel

R1 und R2 unabhängig voneinander C13-Alkyl und
R3 Halogen bedeuten, bzw. von deren Salzen.
Der Ausdruck C1,-Alkyl beinhaltet Methyl, Äthyl, Propyl und Isopropyl.
Eine bevorzugte Gruppe von Verbindungen der Formel I sind diejenigen, in denen R3 Brom oder Fluor ist. Von besonderem Interesse ist das 2,4-Diamino-5-(4-brom- oder 4 fluor) -3,5 -dimethoxybenzyl)-pyrimidin.
Die Verbindungen der Formel I bzw. ihre Salze können erfindungsgemäss dadurch erhalten werden, dass man eine Verbindung der Formel

worin
R4 Niederalkyl oder beide R4 zusammen Niederalkylen;
R5 eine Abgangsgruppe und Rl, R2 und R3 dasselbe wie oben bedeuten, mit Guanidin umsetzt und eine so erhaltene Verbindung der Formel I gegebenenfalls in ein Salz überführt.
In der Verbindung der Formel IIb stellt das Symbol R5 eine Abgangsgruppe dar. Repräsentative Beispiele für solche Abgangsgruppen sind Ätherreste, z. B. niedere Alkoxygruppen, wie Methoxy, Äthoxy, Thioätherreste, wie niedere Alkylthiogruppen, oder von primären oder sekundären Aminen abgeleitete Aminogruppen. Beispiele solcher Aminogruppen sind a) von primären aliphatischen, aryl-aliphatischen oder aromatischen Aminen abgeleitete Gruppen, wie Niederalkylamino, Benzylamino, Arylamino, wie Naphthylamino, insbesondere aber Phenylamino (Anilino), dessen Phenylring gegebenenfalls ein- oder mehrfach halogen-, niederalkyl oder niederalkoxysubstituiert sein kann, ober b) von sekundären aliphatischen, aromatischen oder heterocyclischen Aminen abgeleitete Gruppen, wie N,N-Di-niederalkylamino, N-niederalkyl-N-arylamino, z. B.
N-Methyl-N-phenylamino (N-Methylanilino), dessen Phenylring gegebenenfalls ein- oder mehrfach halogen-, niederalkyl- oder niederalkoxysubstituiert sein kann, Pyrrolidino, Piperidino, Piperazino, Morpholino. Eine speziell bevorzugte Amino-Abgangsgruppe ist die Anilinogruppe.
Die Reaktion der Ausgangsverbindungen IIa bzw. llb mit Guanidin kann nach an sich bekannten Methoden durchgeführt werden (siehe z. B. die belgischen Patentschriften Nrn.
594 131, 671 982 und 746 846), z. B. in einem Lösungsmittel wie einem Alkanol, z. B. Methanol, Äthanol, oder in Dimethylformamid, Dimethylsulfoxid, N-Methylpyrazolon, bei ungefähr 25 bis 200 , vorzugsweise 50 bis 170"C.
Die Verbindung der Formel lib kann sich unter diesen Reaktionsbedingungen aus der tautomeren Verbindung der Formel

worin R1, R2, R3 und R5 obige Bedeutung haben, in situ bilden. Die Verbindungen der Formeln IIb und IIc können als cis- oder trans-Isomere oder als Gemische derselben vorliegen.
Für die Herstellung von Säureadditionssalzen, insbesondere von in pharmazeutischen Präparaten brauchbaren Salzen, kommen die üblicherweise für diesen Zweck verwendeten anorganischen Säuren, wie Salzsäure, Schwefelsäure, Phosphorsäure, usw. oder organischen Säuren, wie Ameisensäure, Essigsäure, Bernsteinsäure, Milchsäure, Citronensäure, Maleinsäure, Fumarsäure, Weinsäure, Methansulfonsäure, p-Toluolsulfonsäure usw. in Betracht.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I und ihre Salze sind antibakteriell wirksam. Sie hemmen die bakterielle Dihydrofolat-Reduktase und potenzieren die antibakterielle Wirkung von Sulfonamiden, wie z. B. Sulfisoxazol, Sulfadimethoxin, Sulfamethoxazol, 4-Sulfanilamido5,6-dimethoxy-pyrimidin, 2-Sulfanilamido-4,5-dimethyl-pyrimidin oder Sulfachinoxalin, Sulfadiazin, Sulfamonomethoxin, 2-Sulfanilamido-4,5-dimethyl-isoxazol und anderer Inhibitoren für Enzyme, die an der Folsäurebiosynthese beteiligt sind, wie z. B. Pteridinderivate.
Für solche Kombinationen einer oder mehrerer der ge nannten Verbindungen I mit Sulfonamiden kommt in der Humanmedizin orale, rectale und parenterale Applikation in Frage. Das Verhältnis von Verbindung I zu Sulfonamid kann innerhalb eines weiten Bereiches variieren; es beträgt z. B.
zwischen 1:40 (Gewichtsteile) und 1: 1 (Gewichtsteile); bevorzugte Verhältnisse sind 1:2 bis 1: 10.
So kann z. B. eine Tablette 80 mg einer erfindungsgemässen Verbindung I und 400 mg Sulfamethoxazol, eine Kindertablette 20 mg einer erfindungsgemässen Verbindung I und 100 mg Sulfamethoxazol; Sirup (pro 5 ml) 40 mg Verbindung I und 200 mg Sulfamethoxazol enthalten.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I zeichnen sich durch eine hohe antibakterielle Wirksamkeit bzw. einen ausgeprägten synergistischen Effekt in Kombination mit Sulfonamiden und gute Verträglichkeit aus.
In den nachstehenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Die Ausgangsstoffe können, soweit ihre Herstellung nicht bekannt oder im folgenden beschrieben ist, in Analogie zu den in den Beispielen beschriebenen Verfahrensweisen hergestellt werden.
Beispiel 1
Eine Lösung von 16,7 g Natrium in 220 ml abs. Methanol wurde mit 200 g 4-Brom-3,5-dimethoxy-a-(methoxy-methyl)zimtsäurenitril gelöst, in 110 ml Dimethylsulfoxid (DMSO) versetzt und in einem 70" warmen Ölbad erwärmt, bis die Innentemperatur 70" erreicht hatte. Das Reaktionsgemisch wurde bei dieser Temperatur eine Stunde weiter gerührt. Danach wurden 63,5 g Guanidincarbonat unter Rühren zugegeben und die Mischung langsam erwärmt bis die Innentemperatur 1100 erreichte, wobei im aufgesetzten Wasserabscheider ca. 120 ml Methanol kondensierten. Die Mischung wurde noch eine Stunde bei 110 gerührt, auf 5 abgekühlt, mit 250 ml Wasser versetzt und 0,5 Stunden unter Kühlen gerührt.
Das ausgefallene Produkt wurde abgenutscht und gut mit kaltem Wasser und Benzol gewaschen. Die Kristalle wurden mit ca. 1000 ml in HCI verrieben, abgesaugt und das entstandene 2,4-Diamino-5-(4-brom-3 ,5-dimethoxybenzyl)-py rimidin-hydrochlorid aus heissem Wasser (Kohle) umkristallisiert. Smp. 280-285 .
Das Hydrochlorid wurde in 400 ml 3n NH40H 30 Minuten bei Raumtemperatur gerührt. Das auskristallisierte 2,4 Diamino-5-(4-brom-3 ,5-dimethoxybenzyl)-pyrimidin wurde abgesaugt, gut mit Wasser gewaschen und 15 Stunden im Vakuum bei 70" getrocknet. Smp. 234-235 .
Das Ausgangsmaterial wurde wie folgt hergestellt:
Zu einer Suspension von 460 g 4-Bromo-3,5-dimethoxybenzoesäure in 1200 ml abs. Benzol und 25 ml Dimethylformamid wurden 238 g Thionylchlorid zugetropft und die Mischung unter Rühren 3 Stunden am Rückfluss erwärmt. Die entstandene Lösung wurde im Vakuum zur Trockene eingedampft. Der Rückstand wurde 2mal in je 100 ml abs. Benzol gelöst und das Lösungsmittel wieder entfernt. Der Rückstand wurde in wenig Benzol gelöst, die Lösung filtriert und mit tiefsiedendem Petroläther versetzt. Das ausgefallene Säurechlorid wurde abgenutscht und getrocknet. Smp. 128-130 .
100 g 4-Bromo-3,5-dimethoxybenzoylchlorid wurden in 1000 ml abs. Xylol gelöst. Unter Begasung mit N2 wurden 10 g 5% Pd/BaSO4 zugegeben, weitere 10 Minuten mit N2 ausgeblasen und danach wurde unter Rühren bei 121 Wasserstoff durchgeleitet. Der Verlauf der Reaktion wurde durch Titrieren des entstandenen HC1 verfolgt. Nach ca. 6 Stunden (90% der theoretischen Menge HCI freigesetzt) wurde die Reaktion abgebrochen, die Suspension unter N2 abgekühlt und der Katalysator abgenutscht. Das Filtrat wurde auf die Hälfte eingeengt, mit 1,5 Liter 40% NaHSO3-Lösung 2 Stunden kräftig gerührt und über Nacht stehengelassen. Die dicke Suspension wurde mit Wasser verdünnt, die wässerige Phase mit Benzol gewaschen und unter Rühren und Kühlen mit 50% NaOH auf pH 10 gestellt.
Der Aldehyd wurde abgenutscht und in Benzol gelöst. Die Lösung wurde getrocknet (MgSO4), filtriert und eingedampft. Nach Umkristallisation aus Heptan schmilzt der 4-Bromo-3,5-dimethoxybenzaldehyd bei 112 bis 113".
Zu einer Lösung von 11,0 g Natrium in 400 ml abs. Methanol wurden 160 g dest. ss-Methoxypropionitril und 233 g 4-Brom-3,5-dimethoxybenzaldehyd gelöst in 500 ml abs. Methanol gegeben. Nach 3 Stunden Kochen unter Rückfluss wurde die Lösung (kein Aldehyd war mehr nachweisbar) abgekühlt und über Nacht im Kühlschrank stehengelassen.
Das auskristallisierte Produkt wurde abgesaugt, mit wenig kaltem Methanol gewaschen und getrocknet. Das 4-Brom-3,5 dimethoxy-a-(methoxymethyl)-zimtsäurenitril schmilzt nach Umkristallisieren aus Methanol bei 113-114".
Beispiel 2
21 g 4-Chlor-3,5-dimethoxy-a-cyano-hydrozimtaldehyddimethylacetal wurden mit 140 ml einer in methanolischen Guanidinlösung 11/2 Stunden zum Rückfluss erhitzt. Das Lösungsmittel wurde darauf durch Destillation in einem Ölbad von 1600 entfernt und der Rückstand 15-20 Minuten bei der gleichen Temperatur erhitzt, bis er sich vollständig zu einer kristallinen Masse verfestigt hatte. Die Masse wurde mit 80 bis 100 ml Wasser aufgeschlämmt und abgesaugt. Man erhielt 2,4-Diamino-5 (4-chlor-3,5-dimethoxybenzyl)-pyrimidin vorh Schmelzpunkt 218".
Zur Reinigung wurde das Produkt in 150 ml Essigsäure gelöst, die Lösung gekühlt, das kristallisierte Acetat abgesaugt und in 150 ml heissem Wasser gelöst. Die Lösung wurde heiss mit Kohle behandelt und die Base aus dem Filtrat durch Zusatz von Ammoniak gefällt. Schmelzpunkt 222-223".
Das Ausgangsmaterial wurde wie folgt hergestellt:
Zu einer Lösung von 45 g Natrium in 670 ml Methanol wurden 227 g Acetessigsäuremethylester und 196 g Crotonsäuremethylester gegeben. Das Gemisch wurde unter Rühren 5 Stunden zum Rückfluss erhitzt. Nach Abkühlen des Reaktionsgemisches wurde das Natriumsalz abgesaugt. Eine zweite und dritte Fraktion des Kristallsalzes wurden durch Einengung der Mutterlauge erhalten. Durch Ansäuern einer konzentrierten wässerigen Lösung des so erhaltenen Produktes mit Essigsäure wurde 6-Methylcyclohexadion-2,4-carbonsäuremethylester vom Schmelzpunkt 126 erhalten.
121 g 6-Methyl-cyclohexadion-2,4-carbonsäuremethylester wurde in 650 ml Dimethylformamid gelöst. Das Gemisch wurde unter Eis-Kochsalzkühlung im Verlauf von 2 Stunden tropfenweise unter Rühren bei 0-5" mit 196 g Sulfurylchlorid versetzt. Man rührte dann noch eine weitere Stunde bei 0-5" und mehrere Stunden bei Raumtemperatur. Darauf wurde das Reaktionsgemisch 21/2 Stunden auf 80" erwärmt, das Lösungsmittel unter vermindertem Druck auf dem Wasserbad entfernt und der Rückstand in 700 ml Äthylacetat gelöst, 2mal mit 300 ml Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der kristalline Rückstand wurde aus 300 bis 400 ml Toluol umkristallisiert und lieferte 3-Chlor-2,4-dihydroxy-6-methylbenzoesäuremethylester vom Schmelzpunkt.
122-125 . Ein Analysenpräparat wurde durch wiederholte Umkristallisation aus Toluol erhalten und schmolz bei 129".
143 g 3-Chlor-2,4-dihydroxy-6-methylbenzoesäuremethylester wurden mit einem Liter 3n Natronlauge 30 Minuten zum Rückfluss erhitzt. Das Reaktionsgemisch wurde vorsichtig mit 400 ml 10n Schwefelsäure angesäuert und das 4-Chlor-3,5dihydroxy-toluol durch Zusatz von 300-400 g Ammoniumsulfat ausgesalzen. Nach Extraktion mit Äthylacetat und Abdampfen des Lösungsmittels unter vermindertem Druck wurde der Rückstand aus Wasser kristallisiert. Man erhielt 4-Chlor3,5-dihydroxy-toluol vom Schmelzpunkt 138-139 .
126,5 g 4-Chlor-3,5-dihydroxy-toluol wurden in 150 ml Methanol gelöst und mit 182 ml Dimethylsulfat versetzt. Danach wurden eine Lösung von 92 g Natriumhydroxid in 180 ml Wasser tropfenweise im Verlauf von 45-60 Minuten bei 40 bis 45" unter Rühren zugesetzt. Danach wurde das Reaktionsgemisch 20 Minuten zum Rückfluss erhitzt. Das Reaktionsgemisch wurde mit Wasser verdünnt und mit Äther extrahiert und lieferte 4-Chlor-3,5-dimethoxy-toluol vom Schmelzpunkt 72".
77,5 g 4-Chlor-3,5-dimethoxy-toluol wurden in 1030 ml Pyridin gelöst. Die Lösung wurde zum Rückfluss erhitzt und im Verlauf von einer Stunde tropfenweise mit einer Lösung von 197 g Kaliumpermanganat in 3100 ml heissem Wasser versetzt. Das Reaktionsgemisch wurde weitere 30 Minuten zum Rückfluss erhitzt. Sodann wurden 1700 ml eines Pyridin Wasser-Gemisches abdestilliert. Der Rückstand wurde heiss abgesaugt und das Filtrat nach Abkühlen mit 600 ml 10n Schwefelsäure angesäuert. Der Niederschlag wurde abfiltriert und zwecks Rückgewinnung von nicht umgesetztem Ausgangsmaterial mit Äther extrahiert. Man erhielt 4-Chlor-3,5-dimethoxybenzoesäure vom Schmelzpunkt 247".
97 g 4-Chlor-3,5-dimethoxybenzoesäure wurden 21/2 Stunden mit 64 g Thionylchlorid und 3 ml Dimethylformamid zum Rückfluss erhitzt. Das erhaltene Säurechlorid wurde aus 500 ml Ligroin (Siedepunkt 60-120") kristallisiert und über Paraffin und Natronkalk getrocknet. Schmelzpunkt 102-103".
28 g Säurechlorid wurden in 200 ml Methylenchlorid ge löst und die Lösung wurde auf 0-5" abgekühlt. Danach wur- den 12 g Triäthylamin und anschliessend 5,6 g Äthylenimin unter Rühren im Verlauf von 20 Minuten bei 05 eingetropft. Das Gemisch wurde mit Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wurde mit 300 ml Tetrahydrofuran gelöst und bei - 5-0" mit 1,8 g Lithiumaluminiumhydrid reduziert. Nach 45 Minuten wurden 130 ml 5n Schwefelsäure in das Reaktionsgemisch eingetropft und das Gemisch sodann 30 Minuten zum Rückfluss erhitzt.
Darauf wurden 75 g Ammoniumsulfat zugesetzt, die organische Schicht wurde abgetrennt und lieferte 4-Chlor-3,5-dimethoxybenzaldehyd vom Schmelzpunkt 119".
Ein Gemisch einer methanolischen Natriummethylatlösung (hergestellt durch Auflösen von 1,15 g Natrium in 100 ml Methanol), 17 gss-Methoxypropionitril und 20 g 3,5-Dimethoxy-4-chlor-benzaldehyd wurde 31/2 Stunden zum Rückfluss erhitzt. Das 3,-5 -Dimethoxy-4-chlor-c(- (methoxymethyl)-zimt- säurenitril kristallisierte beim Kühlen. Schmelzpunkt 112 bis 113".
Eine Lösung von 4,6 g Natrium in 70 ml Methanol wurde mit 3 ,5-Dimethoxy-4-chlor-a-(methoxymethyl)-zimtsäureni tril versetzt. Das Gemisch wurde 24 Stunden zum Rückfluss erhitzt, die Reaktion durch Zusatz von Wasser abgestoppt, das Reaktionsgemisch mit Methylenchlorid extrahiert und das Lösungsmittel unter vermindertem Druck abgedampft. Der Rückstand wurde aus 2-Propanol kristallisiert und lieferte 4
Chlor-3 ,5-dimethoxy-a-cyano-hydrozimtaldehyd-dimethyl- acetal vom Schmelzpunkt 84 .
Beispiel 3
Eine Lösung von 0,5 g Natrium in 32 ml absolutem Ätha nol wurde mit 2,11 g Guanidin-hydrochlorid und 2,4 g 3-Ani- lino-2- (4-chloro-3 , 5-dimethoxybenzyl)-acrylonitril versetzt und 20 Stunden unter Stickstoff und unter Rühren gekocht.
Das Äthanol wurde unter vermindertem Druck entfernt, der
Rückstand in Wasser aufgenommen, abenutscht, mit Wasser gewaschen und aus Dimethylformamid-Methanol umkristalli siert. Das 2,4-Diamino-5-(4-chlor-3 5-dimethoxybenzyl)- pyrimidin schmolz bei 232".
Das Ausgangsmaterial wurde wie folgt hergestellt:
Ein Gemisch von 25 g 4-Chlor-3,5-dimethoxybenzoesäure und 50 ml Thionylchlorid wurde eine Stunde am Rückfluss gekocht, wobei eine klare Lösung entstand. Das Reaktionsgemisch wurde unter vermindertem Druck zur Trockene eingedampft, der Rückstand in Benzol gelöst, das Benzol wegdestilliert, der Rückstand nochmals in Benzol gelöst und diese Lösung zu 300 ml Äthanol zugetropft. Das Ganze wurde 30 Minuten am Rückflusskühler gekocht, zur Trockene eingedampft, der Rückstand in Benzol gelöst, die benzolische Lösung mit Wasser, Natriumbicarbonat und Wasser gewaschen, getrocknet und eingedampft. Durch Umkristallisieren des Rückstandes aus Essigester-Petroläther wurde der 4-Chlor3,5-dimethoxybenzoesäureäthylester vom Smp. 100-102" erhalten.
Eine Suspension von 9,4 g Dimethylsulfon und 3,6 g Na triunthydrid (50%ige Dispersion in Ö1) in 35 ml absolutem Dimethylsulfoxid wurde unter Feuchtigkeitsausschluss und Stickstoff 2 Stunden bei 50 gerührt. Die Heizung wurde abgestellt und 12,2 g 4-Chlor-3,5-dimethoxybenzoesäureäthylester zugegeben, worauf die Temperatur auf 63" anstieg. Es wurde noch 11/2 Stunden bei Zimmertemperatur gerührt, mit 200 ml Wasser verdünnt, die wässerige Lösung mit 100 ml Essigester geschüttelt, über Kohle filtriert und über Nacht im Kühlschrank stehengelassen. Das ausgefallene 4'-Chlor-3', 5'-dimethoxy-2-(methylsulfonyl)-acetophenon schmolz nach Umkristallisieren aus Essigester-Petroläther bei 189-190".
Eine Suspension von 4,7 g 4'-Chlor-3',5'-dimethoxy-2 (methylsulfonyl)-acetophenon in 3 ml Äthanol und 10 ml Wasser wurde mit einer Lösung von 0,2 g Natriumborhydrid in 4 ml Wasser (unter Zusatz von 0,1 g Natriumhydroxid) versetzt. Es wurde noch 2 Stunden bei Raumtemperatur gerührt, mit Eis gekühlt, abgenutscht, mit Eiswasser gewaschen und aus Äthanol umkristallisiert. Der 4-Chlor-3,5-dimethoxy a-[(methylsulfonyl)-methyl]-benzylalkohol schmolz bei 171".
Eine Mischung von 0,6 g Natriummethylat, 2,9 g 4-Chlor 3 ,5-dimethoxy-z-{(methylsulfonyl)-methylj-benzylalkohol und 1,6 g Anilinopropionitril in 10 ml absolutem Dimethylsulfoxid wurde unter Stickstoff und Feuchtigkeitsausschluss 5 Stunden bei 40 gerührt, in 100 ml Wasser gegossen, mit 100 ml Essigester extrahiert, die Essigesterlösung getrocknet und eingedampft. Nach Reinigung über Aluminiumoxid und Umkristallisation aus Essigester schmolz das 3-Anilino-2-(4chlor-3,5-dimethoxybenzyl)-acrylnitril bei 192-194".
Beispiel 4
In einer Lösung von 0,11 g Natrium-Metall in 15 ml absolutem Methanol wurden 1,6 g ss-Methoxypropionitril und 1,65 g 4-Fluor-3,5-dimethoxybenzaldehyd gelöst und 3 Stunden am Rückfluss gekocht. Die Mischung wurde eingedampft und der Rückstand in 50 ml Benzol und 10 ml Wasser aufgenommen. Die wässerige Phase wurde 2maI mit je 20 ml Benzol gewaschen. Die vereinigten Benzolauszüge wurden getrocknet und eingedampft. Das so erhaltene 4-Fluor-3,5-di methoxy-o - (methoxymethylen) -hydrozimtsäurenitril wurde in der nächsten Reaktion eingesetzt.
Eine Lösung von 0,17 g Natrium in 4 ml absolutem Methanol wurde mit 1,50 g 4-Fluor-3,5-dimethoxy-a-(methoxy- methylen)-hydrozimtsäurenitril gelöst in 2 ml DMSO versetzt und in einem 70" warmen Ölbad erwärmt, bis die Innentemperatur 70" erreicht hatte. Das Reaktionsgemisch wurde bei dieser Temperatur eine Stunde weitergerührt. Danach wurden 0,64 g Guanidincarbonat zugegeben und die Mischung langsam erwärmt, bis die Innentemperatur 110 erreichte, wobei im aufgesetzten Wasserabscheider Methanol kondensierte.
Nach einer Stunde wurde die Mischung abgekühlt, mit Wasser versetzt und 30 Minuten gerührt. Das ausgefallene Produkt wurde abgenutscht, mit kaltem Wasser und Benzol gewaschen.
Nach Umkristallisation aus Methanol erhielt man 2,4-Di amino-5-(4-fluor-3,5-dimethoxybenzyl)-pyrimidin vom vom Smp.
216-220 .
Herstellung des Ausgangsmaterials:
13,8 g Natrium werden in 900 ml CH3oH gelöst. Zu dieser Lösung wurden 46,8 g 3-Hydroxy-5-keto-3-cyclohexencarbonsäure zugegeben. Dieses Gemisch wurde gerührt, mit einem kalten Bad zwischen - 4 und - 8 gehalten und mit einer aus 27,9 g Anilin, 450 ml Wasser, 72 ml HC1 konz. und 21,0 g Natriumnitrit in 90 ml Wasser hergestellten Phenyldiazoniumchloridlösung im Laufe von 30 Minuten versetzt.
Das Ganze wurde noch eine Stunde bei - 5 bis - 100 gerührt.
Das ausgefallene, rote Reaktionsprodukt wurde abgenutscht und mit ca. 1000 ml Wasser gewaschen, Smp. 218".
60 g so erhaltene 3-Hydroxy-5-keto-4-phenylazo-3-cyclohexen-carbonsäure, 200 ml Methanol, 1200 ml Benzol und 5 g p-Toluolsulfonsäure wurden zusammen am Wasserabscheider 18 Stunden unter Rückfluss gekocht. Nach dem Erkalten wurde die Lösung mit 500 ml 5 %iger Natriumbicarbonatlösung, dann mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wurde in Essigester gelöst und durch eine Aluminiumoxid-Säule, 500 g, Aktivitätsstufe I, gereinigt. Nach Eindampfen des Essigesters hinterblieb ein festes Produkt vom Smp. 144" (aus Benzol-Petroläther).
54,8 g so erhaltener 3 -Hydroxy-5 -keto-4-phenylazo-3- cyclohexen-carbonsäuremethylester, 12,0 g Acetamid und 2,0 g Bromsuccinimid wurden in 600 ml Chloroform gerührt und tropfenweise mit 32,0 g Brom (gelöst in 400 ml Chloroform) versetzt. Die Reaktionstemperatur wurde unter 35 gehalten. Bald begann die Ausscheidung von Acetamid HBr.
Man rührte noch 30 Minuten bei Zimmertemperatur weiter, filtrierte das Acetamid HBr ab und dampfte zur Trockene ein.
Der Rückstand wurde in wenig Äthanol aufgenommen abgenutscht und mit Äthanol gewaschen. Smp. 216-218 .
Eine Mischung von 27,2 g des wie vorstehend beschrieben erhaltenen 3 ,5-Dihydroxy-4-phenylazo-benzoesäuremethyl- esters, 150 ml Methanol und 64 g Dimethylsulfat wurde unter Rühren im Laufe von 45 Minuten mit einer Lösung von 23 g NaOH in 50 ml Wasser versetzt. Mit einem kalten Bad sorgte man dafür, dass die Temperatur 55" nicht überstieg. Es wurde noch eine Stunde bei Zimmertemperatur gerührt, mit Eiswasser gekühlt, abgenutscht und aus 400 ml Äthanol umkristallisiert. Rote Kristalle vom Smp. 130-132".
12 g so erhaltener 3,5-Dimethoxy-4-phenylazo-benzoesäuremethylester wurden in 400 ml Äthanol gelöst und nach Zugabe von 0,80 g Pd/Kohle, unter normalem Druck und bei Zimmertemperatur hydriert. Unter leichter Erwärmung wurden in 11/2 Stunden 2 Mole Wasserstoff aufgenommen. Der Katalysator wurde abgetrennt und das Filtrat im Vakuum konzentriert. Das gebildete Anilin wurde mit Wasserdampf abdestilliert. Nach Kühlung wurde der als wässerige Suspension zurückgebliebene 4-Amino-3,5-dimethoxy-benzoesäureme- thylester abgenutscht, getrocknet und aus Cyclohexan umkristallisiert. Smp. 115-116".
Eine Suspension von 214 g Dimethylsulfon und 78,2 g Natriumhydrid (50 %ige Dispersion in Ö1) in 400 ml absolutem Dimethylsulfoxid wurde unter Feuchtigkeitsausschluss und Stickstoff 3 Stunden bei 50 gerührt. In die auf 30 abgekühlte Mischung wurden 137 g 4-Amino-3,5-dimethoxybenzoesäuremethylester gegeben, worauf die Temperatur auf 50 anstieg. Nach ca. einer Stunde Rühren unter Stickstoff bei Raumtemperatur entstand ein Brei, der nach 3 Stunden Stehen unter Zugabe von Eis in 2 Liter Wasser gelöst wurde. Die Lösung wurde mit Eisessig auf pH 6-7 eingestellt.
Nach einer Stunde Rühren unter Eiskühlung wurde das auskristallisierte 4'-Amino-3',5'-dimethoxy-2-(methylsulfonyl)-Åacetophenon abgesaugt, mit Wasser gewaschen, getrocknet und aus Essigester umkristallisiert. Smp. 166-167 .
Eine Suspension von 123 g 4'-Amino-3',5'-dimethoxy2-(methylsulfonyl)-acetophenon und 68 g Natriumborhydrid in 1,5 Liter Alkohol wurde 20 Stunden bei Raumtemperatur gerührt. Die Suspension wurde mit 1,5 Liter Wasser verdünnt.
Der Alkohol wurde im Vakuum abgedampft und der 4 Amino- 3 ,5-dimethoxy-a- [(methylsulfonyl) -methyl]-benzyl alkohol abgesaugt, mit Wasser gewaschen und getrocknet.
Smp. 178-179 .
138 g 4-Amino-3 ,5-dimethoxy-a -[(methylsulfonyl)-methyl]-benzylalkohol in 250 ml Dimethylsulfoxid wurden mit
9,75 g Natriumamid versetzt. Das Reaktionsgemisch wurde
11/4 Stunden bei Raumtemperatur gerührt und darauf in 2 Liter Wasser gegossen. Die entstandene Fällung wurde mit 2 Liter Essigester extrahiert, die Wasserphase mit 2 Liter Essigester extrahiert, die Essigesterphasen zweimal mit je 1 Liter Wasser ionenfrei gewaschen, über Magnesiumsulfat getrocknet, filtriert und im Vakuum bei 40 zur Trockene eingedampft. Der kristalline Rückstand wurde in 250 ml Methyl alkohol heiss gelöst, die Lösung mit 150 ml Wasser versetzt und 18 Stunden bei 4" stehengelassen.
Der auskristallisierte 4-Amino-3,5-dimethoxybenzaldehyd wurde abgesaugt, mit einer Mischung von 40 ml Methylalkohol und 20 ml Wasser ionenfrei gewaschen und im Vakuum bei 50 getrocknet.
Smp. 90930.
Eine Lösung von 1,4 g 4-Amino-3,5-dimethoxybenzalde- hyd in 20 ml 50% HBF4 wurde bei 0 mit einer Lösung von
0,59 g NaNO2 in ca. 2 ml Wasser diazotiert. Nach 6stündiger Bestrahlung mit einer Niederdruck-Hg-Lampe (Hanau NK
6620) bei Zimmertemperatur, wurde die Lösung mit Äther extrahiert, die Äther-Auszüge mit MgSO4 getrocknet und eingeengt. Der Rückstand ergab, nach Umkristallisation aus Heptan, 3,5 -Dimethoxy-4-fluorbenzaldehyd, Smp. 97-98 .
Beispiel 5
Aus 0,48 g Guanidin HC1 wurde mit einer Lösung von
110 mg Na-Metall in 10 ml Methanol Guanidin freigesetzt.
Nach Abnutschen des NaCl wurde das Filtrat mit 0,9 g 4-Jod-
3,5 -dimethoxy-a -(methoxymethylen) -hydrozimtsäurenitril versetzt und die Mischung 5 Stunden am Rückfluss gekocht.
Nach dem Abkühlen wurde das Produkt abgetrennt und aus Methanol umkristallisiert. Es wurden 0,7 g 2,4-Diamino-5 (4-jod-3,5-dimethoxybenzyl)-pyrimidin vom Smp. 238-240 erhalten.
Herstellung des Ausgangsmaterials:
Zu einer Lösung von 23,9 g 4-Amino-3,5-dimethoxybenzaldehyd in 315 ml In HCI wurde bei 0-5" unter Rühren eine Lösung von 9 g NaN02 in 30 ml Wasser zugetropft (ne gativer KJ-Test) und 10 Minuten weitergerührt. Das entstandene Diazoniumsalz wurde mit einer Lösung von 26,1 g KJ in 15 ml Wasser versetzt, 15 Minuten bei Raumtemperatur und 45 Minuten bei 50 bis zur Beendigung der Stickstoff entwicklung gerührt. Nach dem Abkühlen wurde der gebil dete Niederschlag abgenutscht und in ca. 1,5 Liter Äther auf genommen. Die Ätherphase wurde 2mal mit je 200 ml 2n NaOH und 3mal mit je 200 ml Wasser gewaschen, über MgS04 getrocknet und eingedampft.
Der Rückstand, aus heissem n-Heptan umkristallisiert, ergab 19,2 g 3,5-Dimeth oxy-4-jodbenzaldehyd. Smp. 127-128 .
Zu einer Lösung von 70 mg Natrium-Metall in 10 ml absolutem Methanol wurden 0,84 g ss-Methoxypropionitril ge geben, die Mischung mit 2,9 g 3,5-Dimethoxy-4-jodbenzalde hyd versetzt und 5 Stunden am Rückfluss gekocht. Danach wurde das Reaktionsgemisch abgekühlt. Das ausgefallene 4 Jod-3,5 -dimethoxy-a - (methoxymethylen)-hydrozimtsäurenitril schmolz nach Umkristallisation aus Methanol bei 93-94 .
Beispiel 6
Zu einer Lösung von 2,6 g Na-Metall in 250 ml absolutem Methanol wurden 20,5 g Guanidincarbonat zugegeben und die Suspension wurde 15 Minuten am Rückfluss erwärmt. Nach der Zugabe von 16 g a-(Anilinomethylen)-4-jod-3,5-dimethoxy-hydrozimtsäurenitril wurde das Reaktionsgemisch 18 Stunden unter Rückfluss gekocht. Nach dem Abkühlen wurden die festen Produkte abgenutscht, in 100 ml Wasser aufgenommen und 2 Stunden bei Raumtemperatur gerührt. Das auskristallisierte 2,4-Diamino-5- (4-jod-3,5 -dimethoxyben- zyl)-pyrimidin wurde abgenutscht und aus heissem Methanol umkristallisiert. Smp. 238-240 .
Herstellung des Ausgangsmaterials:
Zu einer Suspension von Natriummethylat (hergestellt aus 0,56 g Na-Metall und 50 ml absolutem Methanol und anschliessendem Eindampfen) in 20 ml Dimethylsulfoxid wurden unter Rühren 15,3 gss-Morpholinopropionitril gegeben.
Nach Zugabe von 24 g 3,5-Dimethoxy-4-jodbenzaldehyd (wobei die Temperatur auf 40 stieg) wurde die Lösung 30 Minuten bei 40 weitergerührt. Nach dem Abkühlen wurden der Mischung 15 ml Methanol und Wasser bis zur bleibenden Trübung zugesetzt. Das kristallisierende Reaktionsgemisch wurde weitere 15 Minuten gerührt und das Produkt abgenutscht. Nach Umkristallisation aus Methanol.wurden 24,5 g 4-Jod-3 , 5-dimethoxy-a-(morpholinomethylen)-hydro- zimtsäurenitril vom Smp. 121-125" erhalten. Eine Probe, mehrmals aus Methanol umkristallisiert, schmolz bei 136 bis 138".
Ein Gemisch von 6,8 g Anilin, 7,9 ml konz. HCI und 23,5 g 4-Jod-3 ,5-dimethoxy-a- (morpholinomethylen) -hydro- zimtsäurenitril in 50 ml Isopropanol wurde unter Rühren am Rückfluss eine Stunde erwärmt. Nach dem Abkühlen wurde das a -(Anilinomethylen)-4-jod-3 ,5-dimethoxy-hydrozimt- säurenitril abgenutscht und aus Methanol umkristallisiert.

If their preparation is not known or is described below, the starting materials can be prepared analogously to the procedures described in the examples.
example 1
A solution of 16.7 g sodium in 220 ml abs. Methanol was dissolved with 200 g of 4-bromo-3,5-dimethoxy-a- (methoxy-methyl) cinnamonitrile, added in 110 ml of dimethyl sulfoxide (DMSO) and heated in a 70 "warm oil bath until the internal temperature had reached 70". The reaction mixture was further stirred at this temperature for one hour. Then 63.5 g of guanidine carbonate were added with stirring and the mixture was warmed slowly until the internal temperature reached 1100, about 120 ml of methanol condensing in the attached water separator. The mixture was stirred for a further hour at 110, cooled to 5, mixed with 250 ml of water and stirred for 0.5 hours with cooling.
The precipitated product was filtered off with suction and washed well with cold water and benzene. The crystals were triturated with about 1000 ml in HCl, suction filtered and the resulting 2,4-diamino-5- (4-bromo-3,5-dimethoxybenzyl) -py rimidine hydrochloride was recrystallized from hot water (coal). M.p. 280-285.
The hydrochloride was stirred in 400 ml of 3N NH40H for 30 minutes at room temperature. The 2.4 diamino-5- (4-bromo-3,5-dimethoxybenzyl) pyrimidine which had crystallized out was filtered off with suction, washed well with water and dried in vacuo at 70 "for 15 hours. Mp. 234-235.
The starting material was produced as follows:
To a suspension of 460 g of 4-bromo-3,5-dimethoxybenzoic acid in 1200 ml of abs. Benzene and 25 ml of dimethylformamide were added dropwise to 238 g of thionyl chloride and the mixture was heated under reflux for 3 hours with stirring. The resulting solution was evaporated to dryness in vacuo. The residue was 2 times in 100 ml of abs. Dissolved benzene and the solvent removed. The residue was dissolved in a little benzene, the solution was filtered and low-boiling petroleum ether was added. The precipitated acid chloride was suction filtered and dried. M.p. 128-130.
100 g of 4-bromo-3,5-dimethoxybenzoyl chloride were dissolved in 1000 ml of abs. Xylene dissolved. 10 g of 5% Pd / BaSO4 were added while gassing with N2, the mixture was blown out with N2 for a further 10 minutes and then hydrogen was passed through with stirring at 121. The course of the reaction was followed by titrating the HC1 formed. After about 6 hours (90% of the theoretical amount of HCl released), the reaction was stopped, the suspension was cooled under N2 and the catalyst was filtered off with suction. The filtrate was concentrated in half, stirred vigorously with 1.5 liters of 40% NaHSO3 solution for 2 hours and left to stand overnight. The thick suspension was diluted with water, the aqueous phase washed with benzene and adjusted to pH 10 with stirring and cooling with 50% NaOH.
The aldehyde was filtered off and dissolved in benzene. The solution was dried (MgSO4), filtered and evaporated. After recrystallization from heptane, the 4-bromo-3,5-dimethoxybenzaldehyde melts at 112 to 113 ".
To a solution of 11.0 g sodium in 400 ml abs. Methanol was distilled 160 g. ss-methoxypropionitrile and 233 g of 4-bromo-3,5-dimethoxybenzaldehyde dissolved in 500 ml of abs. Given methanol. After 3 hours of refluxing, the solution (no more aldehyde was detectable) was cooled and left in the refrigerator overnight.
The crystallized product was filtered off, washed with a little cold methanol and dried. The 4-bromo-3,5-dimethoxy-a- (methoxymethyl) -cinnamonitrile melts after recrystallization from methanol at 113-114 ".
Example 2
21 g of 4-chloro-3,5-dimethoxy-a-cyano-hydrocinnamaldehyde dimethyl acetal were heated under reflux with 140 ml of a solution in methanolic guanidine solution for 11/2 hours. The solvent was then removed by distillation in a 1600 oil bath and the residue heated at the same temperature for 15-20 minutes until it solidified completely to a crystalline mass. The mass was slurried with 80 to 100 ml of water and suction filtered. 2,4-Diamino-5 (4-chloro-3,5-dimethoxybenzyl) pyrimidine with melting point 218 "was obtained.
For purification, the product was dissolved in 150 ml of acetic acid, the solution was cooled, the crystallized acetate was filtered off with suction and dissolved in 150 ml of hot water. The solution was treated with hot coal and the base was precipitated from the filtrate by adding ammonia. Melting point 222-223 ".
The starting material was produced as follows:
227 g of methyl acetoacetate and 196 g of methyl crotonic acid were added to a solution of 45 g of sodium in 670 ml of methanol. The mixture was heated to reflux with stirring for 5 hours. After the reaction mixture had cooled, the sodium salt was filtered off with suction. A second and third fraction of the crystal salt were obtained by concentrating the mother liquor. Acidification of a concentrated aqueous solution of the product thus obtained with acetic acid gave methyl 6-methylcyclohexadione-2,4-carboxylic acid, melting point 126.
121 g of 6-methyl-cyclohexadione-2,4-carboxylic acid methyl ester was dissolved in 650 ml of dimethylformamide. The mixture was added dropwise with ice-sodium chloride cooling over the course of 2 hours with stirring at 0-5 "with 196 g of sulfuryl chloride. The mixture was then stirred for a further hour at 0-5" and for several hours at room temperature. The reaction mixture was then heated to 80 "for 21/2 hours, the solvent was removed on the water bath under reduced pressure and the residue was dissolved in 700 ml of ethyl acetate, washed twice with 300 ml of water, dried over sodium sulfate and evaporated. The crystalline residue was 300 up to 400 ml of toluene recrystallized and gave 3-chloro-2,4-dihydroxy-6-methylbenzoic acid methyl ester from the melting point.
122-125. An analytical preparation was obtained by repeated recrystallization from toluene and melted at 129 ".
143 g of 3-chloro-2,4-dihydroxy-6-methylbenzoic acid methyl ester were heated under reflux with a liter of 3N sodium hydroxide solution for 30 minutes. The reaction mixture was carefully acidified with 400 ml of 10N sulfuric acid and the 4-chloro-3,5dihydroxy-toluene was salted out by adding 300-400 g of ammonium sulfate. After extraction with ethyl acetate and evaporation of the solvent under reduced pressure, the residue was crystallized from water. 4-Chloro3,5-dihydroxy-toluene with a melting point of 138-139 was obtained.
126.5 g of 4-chloro-3,5-dihydroxy-toluene were dissolved in 150 ml of methanol and 182 ml of dimethyl sulfate were added. A solution of 92 g of sodium hydroxide in 180 ml of water was then added dropwise over the course of 45-60 minutes at 40 to 45 "with stirring. The reaction mixture was then refluxed for 20 minutes. The reaction mixture was diluted with water and extracted with ether and provided 4-chloro-3,5-dimethoxy-toluene with melting point 72 ".
77.5 g of 4-chloro-3,5-dimethoxy-toluene were dissolved in 1030 ml of pyridine. The solution was heated to reflux and a solution of 197 g of potassium permanganate in 3100 ml of hot water was added dropwise over the course of one hour. The reaction mixture was refluxed for a further 30 minutes. Then 1700 ml of a pyridine-water mixture were distilled off. The residue was suctioned off hot and, after cooling, the filtrate was acidified with 600 ml of 10N sulfuric acid. The precipitate was filtered off and extracted with ether to recover unreacted starting material. 4-Chloro-3,5-dimethoxybenzoic acid with a melting point of 247 "was obtained.
97 g of 4-chloro-3,5-dimethoxybenzoic acid were heated under reflux for 21/2 hours with 64 g of thionyl chloride and 3 ml of dimethylformamide. The acid chloride obtained was crystallized from 500 ml of ligroin (boiling point 60-120 ") and dried over paraffin and soda lime. Melting point 102-103".
28 g of acid chloride were dissolved in 200 ml of methylene chloride and the solution was cooled to 0-5 ". 12 g of triethylamine and then 5.6 g of ethylene imine were then added dropwise at 05 with stirring over the course of 20 minutes. The mixture was mixed with Shaken water, dried over magnesium sulfate and evaporated under reduced pressure. The residue was dissolved in 300 ml of tetrahydrofuran and reduced at - 5-0 "with 1.8 g of lithium aluminum hydride. After 45 minutes, 130 ml of 5N sulfuric acid was dropped into the reaction mixture and the mixture was then heated to reflux for 30 minutes.
75 g of ammonium sulfate were then added, the organic layer was separated off and gave 4-chloro-3,5-dimethoxybenzaldehyde with a melting point of 119 ".
A mixture of a methanolic sodium methylate solution (prepared by dissolving 1.15 g sodium in 100 ml methanol), 17 gss-methoxypropionitrile and 20 g 3,5-dimethoxy-4-chloro-benzaldehyde was heated to reflux for 31/2 hours. The 3, -5-dimethoxy-4-chloro-c (- (methoxymethyl) -cinnamic acid nitrile crystallized on cooling. Melting point 112 to 113 ".
A solution of 4.6 g of sodium in 70 ml of methanol was treated with 3,5-dimethoxy-4-chloro-a- (methoxymethyl) cinnamic acid. The mixture was heated to reflux for 24 hours, the reaction was stopped by adding water, the reaction mixture was extracted with methylene chloride, and the solvent was evaporated under reduced pressure. Example 3
A solution of 0.5 g sodium in 32 ml absolute ethanol was mixed with 2.11 g guanidine hydrochloride and 2.4 g 3-anilino-2- (4-chloro-3, 5-dimethoxybenzyl) acrylonitrile and cooked for 20 hours under nitrogen and with stirring.
The ethanol was removed under reduced pressure
Residue taken up in water, suction filtered, washed with water and recrystallized from dimethylformamide-methanol. The 2,4-diamino-5- (4-chloro-3 5-dimethoxybenzyl) pyrimidine melted at 232 ".
The starting material was produced as follows:
A mixture of 25 g of 4-chloro-3,5-dimethoxybenzoic acid and 50 ml of thionyl chloride was refluxed for one hour to give a clear solution. The reaction mixture was evaporated to dryness under reduced pressure, the residue was dissolved in benzene, the benzene was distilled off, the residue was again dissolved in benzene and this solution was added dropwise to 300 ml of ethanol. The whole was boiled for 30 minutes on a reflux condenser, evaporated to dryness, the residue dissolved in benzene, the benzene solution washed with water, sodium bicarbonate and water, dried and evaporated. The 4-chloro-3,5-dimethoxybenzoic acid ethyl ester of mp 100-102 "was obtained by recrystallization of the residue from ethyl acetate-petroleum ether.
A suspension of 9.4 g of dimethyl sulfone and 3.6 g of sodium trihydride (50% dispersion in oil) in 35 ml of absolute dimethyl sulfoxide was stirred at 50 for 2 hours with the exclusion of moisture and nitrogen. The heating was switched off and 12.2 g of ethyl 4-chloro-3,5-dimethoxybenzoate were added, whereupon the temperature rose to 63 ". The mixture was stirred for a further 11/2 hours at room temperature, diluted with 200 ml of water and the aqueous solution with 100 ml of ethyl acetate was shaken, filtered through charcoal and left in the refrigerator overnight. The precipitated 4'-chloro-3 ', 5'-dimethoxy-2- (methylsulfonyl) acetophenone melted after recrystallization from ethyl acetate-petroleum ether at 189-190 ".
A suspension of 4.7 g of 4'-chloro-3 ', 5'-dimethoxy-2 (methylsulfonyl) acetophenone in 3 ml of ethanol and 10 ml of water was washed with a solution of 0.2 g of sodium borohydride in 4 ml of water (under Addition of 0.1 g of sodium hydroxide). The mixture was stirred at room temperature for 2 hours, cooled with ice, suction filtered, washed with ice water and recrystallized from ethanol. The 4-chloro-3,5-dimethoxy a - [(methylsulfonyl) methyl] benzyl alcohol melted at 171 ".
A mixture of 0.6 g of sodium methylate, 2.9 g of 4-chloro 3, 5-dimethoxy-z - {(methylsulfonyl) methylj-benzyl alcohol and 1.6 g of anilinopropionitrile in 10 ml of absolute dimethyl sulfoxide was left under nitrogen and with exclusion of moisture for 5 hours stirred at 40, poured into 100 ml of water, extracted with 100 ml of ethyl acetate, the ethyl acetate solution dried and evaporated. After purification over aluminum oxide and recrystallization from ethyl acetate, the 3-anilino-2- (4chlor-3,5-dimethoxybenzyl) acrylonitrile melted at 192-194 ".
Example 4
1.6 g of ss-methoxypropionitrile and 1.65 g of 4-fluoro-3,5-dimethoxybenzaldehyde were dissolved in a solution of 0.11 g of sodium metal in 15 ml of absolute methanol and refluxed for 3 hours. The mixture was evaporated and the residue taken up in 50 ml of benzene and 10 ml of water. The aqueous phase was washed twice with 20 ml of benzene. The combined benzene extracts were dried and evaporated. The 4-fluoro-3,5-di methoxy-o - (methoxymethylene) hydrocinnamonitrile thus obtained was used in the next reaction.
A solution of 0.17 g of sodium in 4 ml of absolute methanol was mixed with 1.50 g of 4-fluoro-3,5-dimethoxy-a- (methoxy-methylene) -hydrocinnamonitrile dissolved in 2 ml of DMSO and in a 70 "warm Oil bath heated until the internal temperature reached 70 ". The reaction mixture was further stirred at this temperature for one hour. Then 0.64 g of guanidine carbonate was added and the mixture was slowly warmed until the internal temperature reached 110, whereupon methanol condensed in the attached water separator.
After one hour the mixture was cooled, water was added and the mixture was stirred for 30 minutes. The precipitated product was filtered off with suction, washed with cold water and benzene.
After recrystallization from methanol, 2,4-di amino-5- (4-fluoro-3,5-dimethoxybenzyl) pyrimidine of mp.
216-220.
Production of the starting material:
13.8 g sodium are dissolved in 900 ml CH3oH. 46.8 g of 3-hydroxy-5-keto-3-cyclohexenecarboxylic acid were added to this solution. This mixture was stirred, kept between -4 and -8 with a cold bath and concentrated with a mixture of 27.9 g aniline, 450 ml water, 72 ml HCl. and 21.0 g of sodium nitrite in 90 ml of water prepared phenyldiazonium chloride solution were added over the course of 30 minutes.
The whole was stirred for an hour at -5 to -100.
The precipitated, red reaction product was filtered off and washed with about 1000 ml of water, mp. 218 ".
60 g of 3-hydroxy-5-keto-4-phenylazo-3-cyclohexene-carboxylic acid thus obtained, 200 ml of methanol, 1200 ml of benzene and 5 g of p-toluenesulfonic acid were boiled together under reflux for 18 hours on a water separator. After cooling, the solution was washed with 500 ml of 5% sodium bicarbonate solution, then with water, dried and evaporated. The residue was dissolved in ethyl acetate and purified through an alumina column, 500 g, activity level I. After evaporating the ethyl acetate, a solid product of mp 144 "(from benzene / petroleum ether) remained.
54.8 g of 3-hydroxy-5-keto-4-phenylazo-3-cyclohexene-carboxylic acid methyl ester thus obtained, 12.0 g of acetamide and 2.0 g of bromosuccinimide were stirred in 600 ml of chloroform and added dropwise with 32.0 g of bromine ( dissolved in 400 ml chloroform). The reaction temperature was kept below 35. The excretion of acetamide HBr soon began.
The mixture was stirred for a further 30 minutes at room temperature, the acetamide HBr was filtered off and evaporated to dryness.
The residue was sucked off in a little ethanol and washed with ethanol. M.p. 216-218.
A mixture of 27.2 g of the 3, 5-dihydroxy-4-phenylazo-benzoic acid methyl ester obtained as described above, 150 ml of methanol and 64 g of dimethyl sulfate was stirred with a solution of 23 g of NaOH in 50 over the course of 45 minutes ml of water are added. A cold bath was used to ensure that the temperature did not exceed 55 ". The mixture was stirred for another hour at room temperature, cooled with ice water, suction filtered and recrystallized from 400 ml of ethanol. Red crystals of mp 130-132".
12 g of 3,5-dimethoxy-4-phenylazo-benzoic acid methyl ester thus obtained were dissolved in 400 ml of ethanol and hydrogenated after addition of 0.80 g of Pd / carbon, under normal pressure and at room temperature. 2 moles of hydrogen were taken up in 11/2 hours with gentle heating. The catalyst was separated and the filtrate was concentrated in vacuo. The aniline formed was distilled off with steam. After cooling, the 4-amino-3,5-dimethoxy-benzoic acid methyl ester which remained as an aqueous suspension was filtered off with suction, dried and recrystallized from cyclohexane. M.p. 115-116 ".
A suspension of 214 g of dimethyl sulfone and 78.2 g of sodium hydride (50% dispersion in oil) in 400 ml of absolute dimethyl sulfoxide was stirred at 50 for 3 hours with the exclusion of moisture and nitrogen. 137 g of methyl 4-amino-3,5-dimethoxybenzoate were added to the mixture, which had cooled to 30, and the temperature rose to 50. After stirring under nitrogen at room temperature for about an hour, a slurry formed which, after standing for 3 hours, was dissolved in 2 liters of water with the addition of ice. The solution was adjusted to pH 6-7 with glacial acetic acid.
After stirring for one hour while cooling with ice, the crystallized 4'-amino-3 ', 5'-dimethoxy-2- (methylsulfonyl) -acetophenone was filtered off with suction, washed with water, dried and recrystallized from ethyl acetate. M.p. 166-167.
A suspension of 123 g of 4'-amino-3 ', 5'-dimethoxy2- (methylsulfonyl) acetophenone and 68 g of sodium borohydride in 1.5 liters of alcohol was stirred at room temperature for 20 hours. The suspension was diluted with 1.5 liters of water.
The alcohol was evaporated in vacuo and the 4 amino- 3, 5-dimethoxy-a- [(methylsulfonyl) methyl] benzyl alcohol was suctioned off, washed with water and dried.

138 g of 4-amino-3,5-dimethoxy-a - [(methylsulfonyl) methyl] benzyl alcohol in 250 ml of dimethyl sulfoxide were mixed with
9.75 g of sodium amide are added. The reaction mixture was
Stirred for 11/4 hours at room temperature and then poured into 2 liters of water. The resulting precipitate was extracted with 2 liters of ethyl acetate, the water phase was extracted with 2 liters of ethyl acetate, the ethyl acetate phases were washed ion-free twice with 1 liter of water each time, dried over magnesium sulfate, filtered and evaporated to dryness in vacuo at 40. The crystalline residue was dissolved in hot 250 ml of methyl alcohol, the solution was mixed with 150 ml of water and left at 4 "for 18 hours.
The crystallized 4-amino-3,5-dimethoxybenzaldehyde was suction filtered, washed ion-free with a mixture of 40 ml of methyl alcohol and 20 ml of water and dried in vacuo at 50.
M.p. 90930.
A solution of 1.4 g of 4-amino-3,5-dimethoxybenzaldehyde in 20 ml of 50% HBF4 was mixed with a solution of
0.59 g NaNO2 diazotized in approx. 2 ml water. After 6 hours of irradiation with a low-pressure mercury lamp (Hanau NK
6620) at room temperature, the solution was extracted with ether, the ether extracts dried with MgSO4 and concentrated. The residue gave, after recrystallization from heptane, 3,5-dimethoxy-4-fluorobenzaldehyde, mp 97-98.
Example 5
0.48 g of guanidine HC1 was mixed with a solution of
110 mg of Na metal are released in 10 ml of methanol guanidine.
After the NaCl had been filtered off, the filtrate was treated with 0.9 g of 4-iodine
3,5-dimethoxy-a - (methoxymethylene) hydrocinnamonitrile were added and the mixture was refluxed for 5 hours.
After cooling, the product was separated off and recrystallized from methanol. 0.7 g of 2,4-diamino-5 (4-iodo-3,5-dimethoxybenzyl) pyrimidine of mp 238-240 was obtained.
Production of the starting material:
To a solution of 23.9 g of 4-amino-3,5-dimethoxybenzaldehyde in 315 ml. In HCl, a solution of 9 g of NaNO 2 in 30 ml of water was added dropwise at 0-5 "with stirring (negative KJ test) and 10 A solution of 26.1 g of KJ in 15 ml of water was added to the resulting diazonium salt, the mixture was stirred for 15 minutes at room temperature and for 45 minutes at 50 until the evolution of nitrogen had ended, after which the precipitate formed was filtered off with suction and dissolved in About 1.5 liters of ether were taken in. The ether phase was washed twice with 200 ml of 2N NaOH and three times with 200 ml of water, dried over MgSO4 and evaporated.
The residue, recrystallized from hot n-heptane, gave 19.2 g of 3,5-dimethoxy-4-iodobenzaldehyde. M.p. 127-128.
0.84 g of ss-methoxypropionitrile was added to a solution of 70 mg of sodium metal in 10 ml of absolute methanol, 2.9 g of 3,5-dimethoxy-4-iodobenzaldehyde were added to the mixture and the mixture was refluxed for 5 hours. The reaction mixture was then cooled. The precipitated 4 iodine-3,5-dimethoxy-a - (methoxymethylene) hydrocinnamonitrile melted after recrystallization from methanol at 93-94.
Example 6
20.5 g of guanidine carbonate were added to a solution of 2.6 g of Na metal in 250 ml of absolute methanol and the suspension was heated under reflux for 15 minutes. After the addition of 16 g of a- (anilinomethylene) -4-iodo-3,5-dimethoxy-hydrocinnamonitrile, the reaction mixture was boiled under reflux for 18 hours. After cooling, the solid products were filtered off with suction, taken up in 100 ml of water and stirred at room temperature for 2 hours. The 2,4-diamino-5- (4-iodo-3,5-dimethoxybenzyl) pyrimidine which had crystallized out was filtered off with suction and recrystallized from hot methanol. M.p. 238-240.
Production of the starting material:
15.3 gss-morpholinopropionitrile was added with stirring to a suspension of sodium methylate (prepared from 0.56 g of Na metal and 50 ml of absolute methanol and subsequent evaporation) in 20 ml of dimethyl sulfoxide.
After adding 24 g of 3,5-dimethoxy-4-iodobenzaldehyde (the temperature rising to 40), the solution was stirred at 40 for a further 30 minutes. After cooling, 15 ml of methanol and water were added to the mixture until it became cloudy. The crystallizing reaction mixture was stirred for a further 15 minutes and the product was filtered off with suction. After recrystallization from methanol. 24.5 g of 4-iodo-3, 5-dimethoxy-a- (morpholinomethylene) -hydrocinnamonitrile of mp 121-125 "were obtained. A sample, recrystallized several times from methanol, melted at 136 to 138 ".
A mixture of 6.8 g of aniline, 7.9 ml of conc. HCl and 23.5 g of 4-iodo-3,5-dimethoxy-a- (morpholinomethylene) hydrocinnamonitrile in 50 ml of isopropanol were heated under reflux for one hour with stirring. After cooling, the a - (anilinomethylene) -4-iodo-3, 5-dimethoxy-hydrocinnamic acid nitrile was filtered off with suction and recrystallized from methanol.
