Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer 1,2,4-Benztriazin-1,4-di-N-oxidderivate.

Die Oxydation kann in vorteilhafter Weise mittels H20 in Eisessig unter Zusatz von Essigsäureanhydrid, vorzugsweise in Trifluoressigsäure gegebenenfalls unter Zusatz von Trifluoressigsäureanhydrid bei einer Temperatur zwiç .cn 30 und 80"C durchgeführt werden.
Die Ausgangsverbindungen der Formel III werden gemäss US-Patent 2489 555 oder den Vorschriften von Arndt, Chem. Ber. 46 (1913) 3522 hergestellt.
Die für X und Y stehenden Alkyl- oder Alkoxygruppen besitzen vorzugsweise 1 bis 4 Kohlenstoffatome. Bei den entsprechenden Phenyl oder Phenoxygruppen kommen als Substituenten insbesondere Alkyl oder Alkoxy mit 1 bis 4 C-Atomen oder Halogen in Betracht.
Bevorzugte Alkylgruppen in der Bedeutung von R1 sind solche mit 1 bis 6 Kohlenstoffatomen. Ein Alkoxysubstituent dieser Alkylgruppe weist vorzugsweise 1 bis 2 Kohlenstoffatome auf.
Als Cycloalkylgruppen R1 kommen insbesondere solche mit 3 bis 8 vorzugsweise 3 bis 6 Kohlenstoffatomen in Betracht.
Rl, als Aralkylgruppe bedeutet insbesondere die Benzyloder die Phenäthylgruppe, die im Phenykest durch Alkyl oder Alkoxy mit 1 bis 4 C-Atomen, Halogen, Nitro, Halogenalkyl mit 1 bis 4 C-Atomen substituiert sein können, wobei als Halogen, Fluor, Chlor, Brom oder Jod und als Halogenalkyl insbesondere die Trifluormethylgruppe in Betracht kommt.
Die neuen 1 ,2,4-benztriazin- 1 ,4-di-N-oxidderivate der Formel I zeichnen sich durch gute mikrobizide Wirkung aus und können dementsprechend allgemein und in breitem Umfang zur Bekämpfung von Mikroorganismen verwendet werden. Sie zeigen insbesondere eine ausgezeichnete Wirkung gegen durch E. coli airsacculitis hervorgerufene Luftwegerkrankungen (CRD) beim Geflügel. Ferner zeigen sie eine gute hemmende und abtötende Wirkung gegenüber grampositiven und gramnegativen Bakterien sowie gegen Pilze.
Weiterhin sind sie gut wirksam gegen tiervirulente Keime.
Die Verbindungen der Formel I bzw. die Mittel, die sie enthalten, können augrund ihrer Wirkung zur Bekämpfung von schädlichen Mikroorganismen dienen. Dabei können die Verbindungen in Form von Lösungen, Emulsionen, Suspensionen, Streupudern, Salben und Cremes zur Konservierung von organischen Materialien, wie Holz, Papier, Kunststoffen, Anstrichmitteln usw. sowie als Desinfektionsmittel z.B. in Seifen, kosmetischen Artikeln, Waschmitteln und Spülbädern verwendet werden.
Die Verwendung der neuen antimikrobiellen Verbindungen ist auf sehr breiter Basis möglich, insbesondere zum Schutz von organischen Substraten gegen den Befall durch schädigende und pathogene Mikroorganismen. Die erwähnten Antimikrobika eignen sich demnach als Konservierungs- und Desinfektionsmittel für technische Produkte aller Art.
Unter den technischen Produkten, welche mit Hilfe der erfindungsgemäss hergestellten Verbindungen der Formel I konservie t werden können, seien die folgenden als Beispiele genannt: Leime, Bindemittel, Anstrichmittel.
Farb- bzw. Druckpasten und ähnliche Zubereitungen auf der Basis von organischen und anorganischen Farbstoffen bzw. Pigmenten, auch solche, welche als Beimischungen Casein oder andere organische Verbindungen enthalten. Auch Wand- und Deckenanstriche, z.B. solche, die ein eiweisshaltiges Farbbindemittel enthalten, werden durch einen Zusatz der erfindungsgemäss hergestellten Verbindungen vor dem Befall durch Schädlinge geschützt.
Die Wirkung der erfindungsgemäss hergestellten Verbindungen kann auch in konservierenden und desinfizierenden Ausrüstungen von Kunststoffen ausgenützt werden. Bei Verwendung von Weichmachern ist es vorteilhaft, den antimikrobiellen Zusatz dem Kunststoff im Weichmacher gelöst bzw.
dispergiert zuzusetzen.
Der Gehalt an gemäss vorliegender Erfindung hergestellten Wirkstoffen kann je nach Anwendungszweck zwischen 0,1 und 50 g, vorzugsweise zwischen 1 und 30 g Wirksubstanz pro Liter Behandlungsflüssigkeit liegen.
Die neuen Wirkstoffe können allein oder zusammen mit anderen bekannten antimikrobiellen Materialschutzmitteln angewendet werden.
Durch Kombination der erfindungsgemäss hergestellten Verbindungen mit grenzflächenaktiven, insbesondere waschaktiven Stoffen gelangt man zu Wasch- und Reinigungsmitteln mit ausgezeichneter antibakterieller Wirkung.
Die Wasch- und Reinigungsmittel können in beliebiger, z.B. flüssiger, breiartiger, fester, flockiger oder körniger Form vorliegen. Die erfindungsgemäss hergestellten Verbindungen können sowohl in anionaktive Verbindungen, wie Seifen und andere Carboxylate (z.B. Alkalisalze höhere Fettsäuren), Abkömmlinge von Schwefel-Sauerstoffsäuren (z.B.
Natriumsalz der Dodecylbenzolsulfonsäure, wasserlösliche Salze von Schwefelsäuremonoestern höhermolekularer Alkohole oder ihrer Polyglykoläther, wie etwa lösliche Salze von Dodecylalkohol-sulfat oder von Dodecylalkoholpolyglykoläthersulfat), Abkömmlinge von Phosphor-Sauerstoffsäuren (z.B. Phosphate), Abkömmlinge mit saurem (elektrophilem) Stickstoff in der hydrophilen Gruppe (z.B. Disulfinsalze), als auch in kationaktive Tenside, wie Amine und ihre Salze (z.B. Lauryldiäthylentriamin), Oniumverbindungen, Aminoxide oder nichtionogene Tenside, wie Polyhydroxyverbindungen, Tenside auf Mono- oder Polysaccharidbasis, höhermolekulare Acetylenglykole, Polyglykoläther (z.B. Polyglyläther, höherer Fettalkohole, Polyglykoläther höhermolekular alkylierter Phenole), bzw. Gemische aus verschiedenartigen Tensiden eingearbeitet werden.
Dabei bleibt ihre antimikrobielle Wirksamkeit in vollem Umfang erhalten. Der Wirkstoffgehalt der Wasch- und Reinigungsmittel, bezogen auf das Gewicht dieses Mittels, beträgt im allgemeinen 0,01 bis 5%, meistens 0,1 bis 3%. Wässrige Zubereitungen solcher Wasch- und Reinigungsmittel, welche erfindungsgemäss hergestellte Verbindungen enthalten, eignen sich als antimikrobielle Reinigungsmittel in der Lebensmittel- und Getränkeindustrie, z.B. Brauereien, Molkereien, Käsereien und Schlachthöfen.
Im weiteren lassen sich die erfindungsgemäss hergestellten Verbindungen auch in kosmetische Präparate, einarbeiten, womit diesen Mitteln zusätzlich antimikrobielle Wirkung verliehen wird. Dabei genügt im allgemeinen, bezogen auf das Gesamtgewicht des Mittels, ein Wirkstoffgehalt von 0,01 bis 5%, vorzugsweise von 0,1 bis 3%.
Für Desinfektions- und Konservierungszwecke können die Verbindungen der Formel I auch in Kombination mit bekannten antimikrobiellen Mitteln verwendet werden. Hierzu gehören z.B.: Halogene und Halogenverbindungen mit aktivem Halogen z.B. Nakiumhypochlorit, Calciumhypochlorit, Chlorkalk, Na trium-p-toluolsulfochloramid, p-Toluolsulfodichloramid, N-Chlorsuccinimid, 1,3-Dichlor-5,5-dimethyl-hydantoin, Trichlorisocyanursäure, Kaliumdichlorisocyanurat, Jod, Jodtrichlorid, Komplexverbindungen von Jod und Jodtrichlorid mit oberflächenaktiven Mitteln wie Polyvinylpyrrolidon, Alkylphenoxypolyglykolen, Polyoxypropylenglykolen, Alkyl- aminoäthansulfonsäuren und -sulfonaten, Alkylarylsulfonaten, quaternären Ammoniumverbindungen.

Die neuen Verbindungen zeigen auch eine ausgezeichnete wachstumsfördernde Wirkung für Nutztiere, z.B. Schweine, Geflügel, sowie für Wiederkäuer wie Rinder oder Schafe.
Die Wirkstoffe können in Form von Lösungen, Emulsionen, Suspensionen, Pulvern, Tabletten, Bolussen und Kapseln peroral, abomasal oder via Injektion den Tieren direkt und zwar als Einzeldosis wie auch wiederholt verabreicht werden. Die Wirkstoffe, bzw. sie enthaltende Gemische kön- nen auch dem Futter oder den Tränken zugesetzt werden oder in sogenannten Futtervormischungen enthalten sein.
Infolge ihres breiten mikrobiziden Wirkungsspektrums können die neuen Verbindungen auch in der Veterinärmedizin zur Bekämpfung von pathogenen Mikroorganismen am und im Tier, insbesondere auf der Haut, im Intestinal- und Urogenitalsystem verwendet werden. Zur Bekämpfung von pathogenen Mikroorganismen in der Veterinärmedizin und/ oder zur Erzielung einer wachstumsfördernden Wirkung bei Nutztieren können die Verbindungen der vorliegenden Erfindung mit folgenden Stoffen kombiniert werden:

Bestimmung der minimalen Hemmkonzentrationen (MIX) gegen Bakterien und Pilze:
Mit den Verbindungen der Formel 1 werden 1 ,5%ige Stammlösungen in Methylcellosolve hergestellt und diese anschliessend derart verdünnt, dass die Inkorporation von je 0,3 ml der Stammlösungen und deren Verdünnungen in je 15 ml warmen Nutrient-Agar eine Konzentrationsreihe von 300, 100,30, 10, 3, 1 usf. ppm Wirksubstanz im Agar ergibt.
Die noch warmen Mischungen werden in Platten gegossen und nach dem Estarren mit folgenden Testorganismen beimpft.

Nach einer Bebrütung von 48 Stunden bei 37"C (Bakterien) resp. 5 Tagen bei 28"C (Pilze) wird die minimale Grenzkonzentration (ppm) der Wirksubstanzen bestimmt, bei der das Wachstum der Testorganismen unterbunden wird.
Als MIC werden für Verbindungen der Formel I Werte ermittelt, die deutlich unter der Anfangskonzentration von 300 ppm liegen.
Beispiel 1
3-(p-Hydroxyäthylamino)-l ,2,4-benztriazin-1 Ädi-N-oxid
30,9 g (0,15 mol) 3-(P-Hydroxyäthylamino)-1,2.4-benz- triazinmonooxyd werden in einer Lösung aus 250 ml Eisessig, 200 ml H202 30% und 30 ml Essigsäureanhydrid während 24 Stunden bei 55-60"C gerührt, die rote Lösung wird bei 10-15"C mit conc. NaOH auf pH 7 neutralisiert. Der rote Niederschlag wird abgesaugt mit H2O gewaschen und getrocknet. Das Rohprodukt wird in 100 ml Methylcellosolve 15 Minuten bei 60"C gerührt und anschliessend bei 20"C abgesaugt.
Ausbeute 13 g (40%) Fp. 190-193"C.
Beispiel 2 3-Methylamino-1 ,2,4-benztriazin-1 ,4-di-N-oxid
1,76 g (0,01 Mol) 3-Methylamino-l ,2,4-benztriazin- monoxid werden in einer Lösung aus 6 ml Trifluoressigsäure und 6 ml H202 30% 6 Stunden bei 50-60"C gerührt, die rote Lösung wird bei 10-15"C mit NaOH conc. auf pH 6-7 gestellt. Der rote Niederschlag wird abgesaugt, mit Wasser gewaschen und getrocknet. Das Rohprodukt wird in Methylcellosolve 15 Minuten bei 500C gerührt, abgesaugt und bei Raumtemperatur getrocknet.
