Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Benzodiazepin-Derivaten der allgemeinen Formel

worin R1 Halogen oder Nitro, R2 Phenyl oder Halogenphenyl R3 Wasserstoff, Alkyl, Hydroxyalkyl, Acyloxyalkyl, Halogenalkyl, Monoalkylaminoalkyl oder Dialkylaminoalkyl, R4 Alkoxy, Halogenalkoxy, Monoalkylaminoalkoxy, Dialkylaminoalkoxy, Hydroxyalkyloxy, Alkoxyalkoxy, Alkylthio, Alkylsulfinyl oder Alkylsulfonyl und R5 Wasserstoff oder Hydroxy bedeuten, bzw. Salze dieser Verbindungen.
Der Ausdruck Alkyl allein oder in Kombination wie Alkoxy umfasst geradkettige und verzweigte Kohlenwasserstoffreste mit 1-6, vorzugsweise mit 1-4 Kohlenstoffatomen wie Methyl, Äthyl, Isopropyl und dgl. Der Ausdruck Acyl umfasst Acylgruppen wie Alkanoylreste, z.B. Acetyl, Propionyl, tert. Butyryl und dgl., Cycloalkancarbonyfreste, wie Cyclopropancarbonyl und dgl., Aroyl oder Aralkanoylreste, wie Benzoyl, Phenacetyl, Phenylpropionyl und dgl., die gegebenenfalls auch Methoxysubstituenten tragen können. Der Ausdruck Halogen umfasst alle 4 Halogene, d.h. Fluor, Chlor, Brom und Jod.
Bevorzugte Verbindungen sind solche, worin R1 Chlor oder Nitro bedeutet. Weiters ist R5 bevorzugt Wasserstoff. Wenn R2 Halogenphenyl bedeutet, dann befindet sich der Substituent vorzugsweise in 2-Stellung des 5-Phenylringes. Weitere bevorzugte Verbindungen sind solche, worin R2 Phenyl, 2- Chlor- phenyl oder 2-Fluorphenyl bedeutet Wenn R3 Acyloxyalkyl bedeutet, dann ist Acyloxymethyl bevorzugt, wobei Acetyl, Cyclopropionyl, tert. Butyryl als Acylreste bevorzugt sind.
Besonders bevorzugt sind Verbindungen worin R1 Chlor oder Nitro, R2 Phenyl, 2-Fluorphenyl oder 2-Chlorphenyl, R Wasserstoff, R3 Wasserstoff oder Alkyl, und R4 Alkoxy bedeuten. Ganz besonders bevorzugt sind 7-Nitro-5-phenyl-1-meth- oxymethyl- 1 ,3-dihydro-2H-1 ,4-benzodiazepin-2-on, 7-Nitro-S- -phenyl-l -äthoxymethyl-1,3 -dihydro-2H-l ,4-benzodiazepin-2 -on, 7-Nitro-5-phenyl-1-[(2-chloräthoxy)-methyl]- 1 ,3-dihydro- -2H-1,4-benzodiazepin-2-on, 7-Chlor-5-(2-fluorphenyl)-1-meth- oxymethyl- 1,3 -dihydro-2H-1 ,4-bezodiazepin-2-on, 7-Chlor -1-(2-cyclopropionyloxy-1-methoxy-äthyl)-1,3-dihydro-5-phe- nyl-2H-1 ,4-benzodiazepin-2-on.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet oder dass man eine Verbindung der Formel

worin R1, R2, R3, Rr und R5 obige Bedeutung haben, R6 A1- kyl bedeutet, verseift und decarboxyliert. Erwünschtenfalls wird eine erhaltene Verbindung in ein Salz überführt.
Die Ausgangsverbindungen der Formel II können zu den entsprechenden 3-carbonsauren Salzen verseift werden, z.B.
durch Behandlung mit Alkalihydroxyden wie Natriumhydroxyd, Kaliumhydroxyd, Erdalkalihydroxyden oder tert. organischen Basen wie Triäthylamin. Die Decarboxylierung dieser 3-carbonsäuren Salze tritt langsam beim Stehen, schneller beim Erwärmen und spontan beim Ansäuern ein.

worin R7 Wasserstoff, Alkyl, Halogenalkyl oder Acyloxyalkyl, R8 Alkoxy, Haloalkoxy, Alkylthio oder Alkoxyalkoxy und X Halogen bedeuten, erhalten werden. Allenfalls kann ein aliphatisch gebundenes Halogenatom anschliessend gegen ein Hydroxygruppe, oder eine Monoalkylamino- oder Dialkylaminogruppe ausgetauscht werden und/oder eine Alkylthiogruppe kann zu einer Alkylsulfinyl- oder Alkylsulfonylgruppe oxidiert werden.
Die Umsetzung einer Verbindung der Formel III oder eines 4-Oxyds davon mit einer Verbindung der Formel IV kann in einem inerten organischen Lösungsmittel oder Gemischen davon wie Kohlenwasserstoffen, z.B. Benzol, Toluol und dgl., Dimethylformamid, Äthern wie Dioxan, Tetrahydrofuran, Alkoholen, wie tert. Butanol und dgl. durchgeführt werden. Geeignete Temperaturen sind z.B. zwischen -50"C und 1200C. Anstelle von X in der Bedeutung Halogen kann irgendeine äquivalente austretende Gruppe wie eine Mesyloxy-, Tosyloxygruppe oder dgl. eingesetzt werden. Es ist zweck mässig, eine Verbindung der Formel II vor der Umsetzung mit einer Verbindung der Formel III zuerst in ein 1-Alkalimetall-Derivat umzuwandeln.
Dieses 1 -Alkalimetallderivat kann z.B. mit Hilfe eines niederen Alkalialkoholats wie Na triummethylat, eines Alkalimetallhydrids, wie Natriumhydrid, eines Alkalimetallamids, wie Natriumamid und ähnlichen Mitteln hergestellt werden. Die Umsetzung einer Verbindung der Formel III mit einer Verbindung der Formel IV kann jedoch auch in Gegenwart einer Base wie Alkalihydroxyden, z.B. Na triumhydroxyd oder Triäthylamin erfolgen.
Die Ersetzung aliphatisch gebundener Halogenatome durch Monoalkylamino- oder durch Dialkylaminogruppen kann durch Umsetzung mit einem Monoalkylamin oder einem Dialkylamin durchgeführt werden.
Diese Umsetzung kann in einem oder mehreren inerten organischen Lösungsmitteln wie Äthern, z.B. Dioxan, Dimethylformamid, Kohlenwasserstoffen, wie Benzol, Toluol, Ketonen wie Aceton, Methyläthylketon und dgl. durchgeführt werden. Die anzuwendenden Temperaturen und Druck sind nicht von Belang und die Reaktion kann bei Raumtemperatur oder erhöhter Temperatur und/oder unter Druck durchgeführt werden.
Die Umwandlung von Verbindungen enthaltend aliphatisch gebundene Halogenatome in eine entsprechende Verbindung, worin die aliphatisch gebundenen Halogenatome durch Hydroxygruppen ersetzt sind, kann z.B. durch Hydrolyse oder Alkoholyse mit katalytischen Mengen einer Base des entsprechenden Acetats erfolgen. Man arbeitet zweckmässigerweise in einem organischen Lösungsmittel wie Alkoholen, Ketonen, z.B. Aceton, Äthern z.B. Dioxan, Dimethylformamid, Dimethylsulfoxyd und dgl. und bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur des Reaktionsgemisches.
Die Oxydation einer Alkylthiogruppe in eine Alkylsulfinyl oder Alkylsulfonylgruppe kann mit Hilfe eines Oxydationsmittels wie Persäuren, z.B. Peressigsäure, Perbenzoesäure und dgl. oder Wasserstoffperoxyd erfolgen. Man arbeitet auch hier zweckmässigerweise in einem inerten organischen Lösungsmittel wie halogenierten Kohlenwasserstoffen, z.B. Tetrachlorkohlenstoff; Essigsäure und dgl. Geeignete Temperaturen sind z.B. zwischen -500C und 800C.

worin R1, R2, Ra, R4 und R6 obige Bedeutung haben mit einem Anhydrid, Sulfid oder Halogenid einer aliphatischen, aromatischen oder araliphatischen Carbonsäure hergestellt werden. Die Umsetzung bewirkt eine Abspaltung des Sauerstoffatoms in 4-Stellung unter gleichzeitiger Acyloxylierung des Kohlenstoffatoms in 3-Stellung. Die Reaktion wird zweckmässigerweise in einem üblichen inerten organischen Lösungs- mittel wie Kohlenwasserstoffen, z.B. Toluol, Benzol und dgL, halogenierten Kohlenwasserstoffen wie Tetrachlorkohlenstoff und dgl., Dimethylformamid und dgl. durchgeführt.
Bei Verwendung eines Säureanhydrids oder eines Diacylsulfids als Acylierungsmittel können diese Säureanhydride oder Diacylsulfide auch direkt als Reaktionsmedium dienen. Anschliessende Hydrolyse der erhaltenen 3-Acyloxy-Derivate liefert 3-Hydroxy-Derivate, welche zweckmässigerweise durch Alkoholyse wie z.B. mit Natriumäthoxid in Äthanol oder Tri äthylamin in Äthanol und dgl. durchgeführt wird.
Verbindungen der Formel II, worin R5 Wasserstoff ist können mit Luft oder Sauerstoff in Gegenwart einer Base zu entsprechenden 3-Hydroxy-3-carbalkoxy-Derivaten oxydiert werden. Die Oxydation erfolgt zweckmässigerweise in einem inerten organischen Lösungsmittel wie Äthan, z.B. Tetrahydrofuran, Dioxan, Alkoholen, z.B. niederen Alkoholen, Dimethylformamid und dgl. Geeignete Basen sind z.B. Alkalialkoxyde wie Natriummethoxvd, Alkalihydride, Alkaliamide und Alkalihydroxyde wie Natriumhydroxyd. Bei der Verwendung von Alkalihydroxyden kann jedoch gleichzeitig eine Verseifung einer vorhandenen Carbalkoxygruppe in 3-Stellung bzw. ein Austausch eines Haloatoms gegen eine Hydroxygruppe in einer halogenalkyl- und/oder halogenalkoxysubstituierten Verbindung eintreten.
Die Verfahrensprodukte der Formel I, die basischen Charakter besitzen, bilden Säureadditionssalze mit anorganischen oder organischen Säuren wie Chlorwasserstoffsäure, Phosphorsäure, Bromwasserstoffsäure, Citronensäure, Schwefelsäure, Essigsäure, Ameisensäure, Bernsteinsäure, Maleinsäure, p-Toluolsulfonsäure und dgl.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I sowie deren pharmazeutisch anwendbare Salze können als Antikonvulsiva, Muskekelaxantien, Sedativa und Anxiolytica verwendet werden. Die antikonvulsive Aktivität wird demonstriert, wenn Mäuse, welchen Verbindungen der Formel I oder deren Salze verabreicht wurden, dem Pentamethylentetrazoltest unterworfen werden. Zum Beispiel zeigt das 7-Nitro -5-phenyl-1 -methoxymethyl-1,3 -dihydro-2H-1 ,4-benzodiaze- pin-2-on, das eine LD50 von 600-1200 mg/kg (p.o.) aufweist bei der Prüfung auf antikonvulsive Wirksamkeit im Pentetrazoltest in Anlehnung an die Methode von Orloff (Proc. Soc.
Exptl. Biol. Med. 70 254-257 1949) eine APR 2,0 von 0,5 mg/ kg (p.o.) [unter APR 2,0 versteht man diejenige Dosis in mg/ kg eines Antikonvulsivums, welche den doppelten Pentetrazol verbrauch gegenüber der unbehandelten Kontrollgruppe bewirkt], das 7-Nitro-5-phenyl-1-äthoxymethyl-1,3-dihydro-2H- -1,4-benzodiazepin-2-on (LD50 1200-2500 mg/kg p.o.) eine APR 2,0 von 0,2 mg/kg (p.o.) und das 7-Nitro-5-phenyl-1- -[(2-chloräthoxy)-methyl]-1,3-dihydro-2H-benzodiazepin-2-on (LD50 > 5000 mg/kg p.o.) eine APR 2,0 von 1,8 mg/kg (p.o.).
Im Gegensatz dazu zeigt Phenobarbital, ein gebräuchliches Antikonvulsivum eine APR 2,0 von 30 mg/kg. Die muskelrelaxierende Wirksamkeit kann im Test am Rotierstab demonstriert werden. Zum Beispiel zeigt das oben genannte 7-Nitro-5-phenyl- 1 -methoxyrnethyl- 1,3 -dihydro-2H- 1 ,4-benzo- diazepin-2-on eine HD50 von 2 mg/kg (p.o.) das 7-Nitro-5 -phenyl- 1 -äthoxymethyl- 1,3 -dihydro-2H- 1 ,4-benzodiazepin-2 -on eine HDso von 5 mg/kg (p.o.) und das 7-Nitro-5-phenyl - 1 -((2-chloräthoxymethyl)]- 1 ,3-dihydro-2H- 1 ,4-benzodiazepin -2-on eine HD50 von 0,5 mg/kg (p.o.).
Die erfindungsgemäss erhältlichen Verbindungen der Formel I sowie deren pharmazeutisch anwendbare Salze können daher als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche öle, Gummi, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln, oder in flüssiger Form, z.B.
als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Dosierung erfolgt nach individuellen Erfordernissen jedoch ist eine Dosierung von 0,1 mg/kg 10 mg/kg/Tag bevorzugt.
Beispiel I
3,9 g 7-Chlor- 1,3 -dihydro- 1 -(methoxymethyl)- 5-phenyl- -2H-1,4-benzodiazepin-2-on-3-carbonsäureäthylester werden in 40 ml Dioxan gelöst und in Gegenwart von 6 ml 2n Natronlauge und 30 ml Wasser während 6 Stunden bei Raumtemperatur unter Stickstoffatmosphäre gerührt. Das Reaktionsgemisch wird mit Äther ausgeschüttelt, die wässrige Phase abgetrennt und mit 2n Salzsäure angesäuert bis das ausgefallene Material wieder in Lösung geht.
Darauf wird mit Ammoniak alkalisch gestellt und die Base mit Methylenchlorid extra hiert. Nach chromatographischer Reinigung der rohen Base erhält man 7-Chlor- 1,3 -dihydro- 1 -(methoxymethyl)-5-phenyl- -2H-1,4-benzodiazepin-2-on die folgende spektroskopischen Daten zeigend: Mass-Spektren: + m

Das Ausgangsmaterial kann wie folgt hergestellt werden:
0,8 g Natriumhydridsuspension (50%ig in Mineralöl) werden in eine auf -10" gekühlte Lösung von 3,42 g 7-Chlor-1,3 -dihydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-3 -carbonsäure äthylester in 30 ml Dimethylformamid eingetragen. Nachdem während 30 Minuten bei 10C gerührt wurde, wird auf 400 abgekühlt und 1,3 ml Chlordimethyläther zugefügt. Man lässt die Temperatur innerhalb 30 Minuten auf 100 ansteigen und giesst das Reaktionsgemisch auf 100 ml Wasser und 10 ml 2n Essigsäure. Das ausgeflockte Produkt wird abgetrennt und in Methylenchlorid aufgenommen. Die getrocknete Methylenchloridlösung wird eingedampft und der Rückstand aus Methylenchlorid-Äther kristallisiert (1,6 g).
Man erhält 7-Chlor -1,3-dihydro-1 -(methoxymethyl)-5-phenyl-2H-1,4-benzodiaze- pin-2-on-3-carbonsäureäthylester, der nach Umkristallisation aus Methylenchlorid-Äther bei 161-164 schmilzt.
Beispiel 2
Eine Lösung von 9 g 7-Chlor-1,3-dihydro-3-hydroxy-1- -(methoxymethyl)-5-phenyl-2H- 1 ,4-benzodiazepin-2-on-3-car- bonsäureäthylester in 75 ml Dioxan wird mit 14 ml 2n Natronlauge und 60 ml Wasser versetzt und während 20 Stunden bei 26-28" gerührt. Das Reaktionsgemisch wird mit 150 ml Äther ausgeschüttelt, die wässrige Phase abgetrennt, mit 2n Essigsäure auf pH 7-8 eingestellt, zweimal mit je 50 ml Äther ausgewaschen, abgetrennt und im Vakuum bei 40-50" auf 25 g eingeengt. Durch Zugabe von 50 ml Aceton erhält man 7,5 g kristallines Natriumsalz.
0,5 g dieses Salzes werden in 10 ml Eisessig während 10 Minuten unter Rückfluss gekocht. Nach dem Eindampfen im Vakuum wird der Rückstand zwischen Bicarbonatlösung und Methylenchlorid verteilt. Die Methylenchloridphase wird getrocknet und eingedampft. Kristallisation des Rückstandes aus Alkohol liefert 0,2 g 7-Chlor-1,3-dihydro-3-hydroxy-1-(meth oxymethyl)-5-phenyl-2H-1 ,4-benzodiazepin-2-on vom Smp.
136-138".
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Zu einer Lösung von 0,5 g Natrium in 50 ml absolutem Alkohol wird eine Lösung von 9,7 g 7-Chlor-1,3-dihydro-1 -(methoxymethyl)-5-phenyl-2H-1 ,4-benzodiazepin-2-on-3 -carbonsäureäthylester in 50 ml abs. Tetrahydrofuran gegeben.
Bei 10-15 wird während 4 Stunden ein trockener Luftstrom in die gerührte Lösung eingeleitet. Nach Zugabe von 2 ml Eisessig wird im Vakuum eingeengt und der Rückstand zwischen Wasser und Methylenchlorid verteilt. Die Methylenchloridphase wird abgetrennt, über Natriumsulfat getrocknet und eingedampft. Der Rückstand liefert durch Kristallisation aus Alkohol 7,7 g 7-Chlor- 1,3 -dihydro-3 -hydroxy- 1 -(methoxyme- thyl)-5-phenyl-2H-l,4-benzodiazepin-2-on ester vom Smp. 187-1890.
Beispiel 3
Gemäss den vorherstehenden Beispielen können die folgenden Verbindungen hergestellt werden: 1,3 -Dihydro-1 -(methoxymethyl-7-nitro-5-phenyl-2H-1,4- -benzodiazepin-2-on; Sp. 139-141" (aus Benzol-Alkohol).
