Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Benzodiazepin-Derivaten der Formel

worin R1 Halogen oder Nitro, R2 Phenyl oder Halogenphenyl, R3 Wasserstoff, Alkyl, Hydroxyalkyl, Acyloxyalkyl, Carbalkoxy, Monoalkylaminoalkyl oder Dialkylaminoalkyl und R4 Alkoxy, Monoalkylaminoalkoxy, Dialkylaminoalkoxy oder Alkoxyalkoxy bedeuten, wobei R4 eine andere Bedeutung als Mono- oder Dialkylaminoalkoxy hat, falls R3 für Carbalkoxy steht, bzw. Salze dieser Verbindungen.
Der Ausdruck Alkyl allein oder in Kombination wie Alkoxy umfasst geradkettige und verzweigte Kohlenwasserstoffreste mit 1-6, vorzugsweise mit 14, Kohlenstoffatomen, wie Methyl, Äthyl, Isopropyl und dgl. Der Ausdruck Acyl umfasst Acylgxuppen, wie Alkanovlreste, z.B. Acetyl, Propionyl, tert. Butyryl und dgl., Cycloalkancarbonylreste, wie Cyclopropancarbonyl und dgl., Aroyl- oder Aralkanoylreste, wie Benzoyl, Phenacetyl, Phenylpropionyl und dgl., die gegebenenfalls auch Methoxysubstituenten tragen können. Der Ausdruck Halogen umfasst alle 4 Halogene, d.h. Fluor, Chlor, Brom und Jod.
Bevorzugte Verbindungen sind solche, worin Rl Chlor oder Nitro bedeutet.
Wenn R2 Halogenphenyl bedeutet, dann befindet sich der Substituent vorzugsweise in 2-Stellung des 5-Phenylringes.
Weitere bevorzugte Verbindungen sind solche, worin R2 Phenyl, 2-Chlorphenyl oder 2-Fluorphenyl bedeutet. Wenn R3 Acyloxyalkyl bedeutet, dann ist Acyloxymethyl bevorzugt, wobei Acetyl, Cyclopropionyl, tert. Butyryl als Acylreste bevorzugt sind. Besonders bevorzugt sind Verbindungen, worin Rl Chlor oder Nitro, R2 Phenyl, 2-Fluorphenyl oder 2
Chlorphenyl, R3 Wasserstoff oder Alkyl und R4 Alkoxy bedeuten. Ganz besonders bevorzugt sind 7-Nitro-5-phenyl-l -methoxymethyl-1,3 -dihydro- 2H-1,4-benzodiazepin-2-on, 7-Nitro-5-phenyl- l-äthoxymethyl-1,3 -dihydro-
2H-1,4-benzodiazepin-2-on, 7-Chlor-5-(2-fluorphenyl)-1-methoxymethyl-1,3-dihydro- 2H-1,4-benzodiazepin-2-on, 7-Chlor- 1 -(2-cyclopropionyloxy-1-methoxy-äthyl) -
1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-on.
Das erfindungsgemässe Verfahren ist dadurch gekennzeich net, dass man eine Verbindung der Formel

worin Rl, R2, R3 und R4 obige Bedeutung haben und R6 Acyl bedeutet, mit einer Base in die entsprechende 4,5-Dehydroverbindung überführt. Erwünschtenfalls wird eine erhaltene Verbindung in ein Salz übergeführt.
Die Säureabspaltung aus einer Verbindung der Formel II kann mit einer Base, wie Alkalihydriden, wie Natriumhydrid, Triäthylamin, Alkaliamiden, wie Natriumamid, Alkalialkoxyden, wie Natriummethoxyd und dgl., in wasserfreiem Medium erreicht werden. Man arbeitet dabei zweckmässigerweise in einem inerten organischen Lösungsmittel, wie Äthern, Alkoholen, z.B. Äthanol, Kohlenwasserstoffen, z.B. Benzol, Toluol, Dimethylformamid und dgl., und bei Temperaturen zwischen -40 C bis 120 C. Die Acylgruppe in der Bedeutung R6 kann eine niedere Alkanoyl-, wie Acetyl-, Aroyl-, wie Benzoyl, Tosyl, oder Mesylgruppe sein.
Bei der Säure abspaltung aus einer Verbindung der Formel II kann die Bildung einer Verbindung der Formel I auftreten, worin die Doppelbindung in 3,4-Stellung anstelle der 4,5 Stellung gebildet wird. Die Isomerisierung in die letztere kann durch Behandlung mit einer Base, wie Alkalialkoholaten, z.B.
Natriummethoxid, Alkalihydriden, wie Natriumhydrid, Tri äthylamin und dgl., erreicht werden. Die Reaktion erfolgt zweckmässigerweise in einem inerten organischen Lösungsmittel, wie Kohlenwasserstoffen, Äthern, Alkoholen und dgl., und bei Temperaturen zwischen - 4OOC bis 1200C.

worin R7 Wasserstoff, Alkyl, Haloalkyl, Carboalkoxy oder Acyloxyalkyl, Rs Alkoxy, Haloalkoxy, oder Alkoxyalkoxy und X Halogen bedeuten, hergestellt werden, indem man ein aliphatisch gebundenes Halogen gegen eine Monoalkylaminooder Dialkylaminogruppe (ausser Rs bedeutet Carboalkoxy) oder eine Hydroxygruppe austauscht und die erhaltene Verbindung in Gegenwart von Platinoxyd reduziert und die so erhaltene Verbindung acyliert.
Die Umsetzung einer Verbindung der Formel III mit einer Verbindung der Formel IV kann in einem inerten organischen Lösungsmittel oder Gemischen davon, wie Kohlenwasserstoffen, z.B. Benzol, Toluol und dgl., Dimethylformamid, Äthern, wie Dioxan, Tetrahydrofuran, Alkoholen, wie tert. Butanol und dgl., durchgeführt werden. Geeignete Temperaturen sind z.B. zwischen - 50oC und 1200C. Anstelle von X in der Bedeutung Halogen kann irgendeine äquivalente austretende Gruppe, wie eine Mesyloxy-, Tosyloxygruppe oder dgl., eingesetzt werden. Es ist nveckmässig, eine Verbindung der Formel III vor der Umsetzung mit einer Verbindung der Formel IV zuerst in ein 1-Alkalimetall-Derivat umzuwandeln.
Dieses 1 Alkalimetall-Derivat kann z.B. mit Hilfe eines niederen Alkalialkoholats, wie Natriummethylat, eines Alkalimetallhydrids, wie Natriumhydrid, eines Alkalimetallamids, wie Natriumamid, und ähnlichen Mitteln hergestellt werden. Die Umsetzung einer Verbindung der Formel III mit einer Verbindung der Formel IV kann jedoch auch in Gegenwart einer Base, wie Alkalihydroxyden, z.B. Natriumhydroxyd, oder Triäthylamin erfolgen.
Der Ersatz von aliphatisch gebundenen Halogenatomen durch Monoalkylamino- oder Dialkylaminogruppen kann durch ein Monoalkylamin oder ein Dialkylamin bewirkt werden. Diese Umsetzung kann in einem oder mehreren inerten organischen Lösungsmitteln, wie Äthern, z.B. Dioxan, Dimethylformamid, Kohlenwasserstoffen, wie Benzol, Toluol, Ketonen, wie Aceton, Methyläthylketon und dgl., durchgeführt werden. Die anzuwendenden Temperaturen und Drücke sind nicht von Belang und die Reaktion kann bei Raumtemperatur oder erhöhter Temperatur und/oder unter Druck durchgeführt werden.
Die Überführung von aliphatisch gebundenen Halogenatomen in Hydroxygruppen kann z.B. durch Hydrolyse oder Alkoholyse mit katalytischen Mengen einer Base des entsprechenden Acetats erfolgen. Man arbeitet zweckmässigerweise in einem organischen Lösungsmittel, wie Alkoholen, Ketonen, z.B. Aceton, Äthern, z.B. Dioxan, Dimethylformamid, Dimethylsulfoxyd und dgl., und bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur des Reaktionsgemisches.
Die Reduktion in Gegenwart von Platinoxyd wird zweckmässigerweise in einem organischen Lösungsmittel, wie Eisessig, Alkoholen und bei einer Temperatur zwischen ca. OoC und 50oC, vorzugsweise bei Raumtemperatur, durchgeführt.

Man arbeitet zweckmässigerweise in einem inerten organischen Lösungsmittel, wie Äthern, Kohlenwasserstoffen, und bei Temperaturen zwischen -600C und Rückflusstemperatur des Reaktionsgemisches. Verbindungen der Formel V können ihrerseits durch Einführung des Substituenten in 1-Stellung einer in 1-Stellung unsubstituierten Verbindung in der oben angegebenen Weise hergestellt werden.
Bei Verwendung einer Ausgangsverbindung der Formel V, welche in 1-Stellung einen Substituenten mit einer Acyloxyalkylgruppe aufweist, wird diese Gruppe gleichzeitig in eine Hydroxyalkylgruppe übergeführt.
Verbindungen der Formel II werden durch Acylierung der entsprechenden 4-Hydroxy-substituierten Verbindungen hergestellt. Geeignete Acylierungsmittel sind z.B. Säureanhydride, wie Essigsäureanhydrid, Säurehalogenide und dgl. Man arbeitet zweckmässigerweise in Gegenwart einer Base als Säureakzeptor, wie Pyridin und dgl., und in einem organischen Lösungsmittel, wie Kohlenwasserstoffen, chlorierten Kohlenwasserstoffen, Äthern und dgl., bei Temperaturen zwischen -500Cund 100OC.
Im Falle eine Monoalkylaminogruppe als Substituent in 1 Stellung vorhanden ist, wird diese Gruppe zweckmässigerweise vor der Acylierung z.B. durch eine Carbobenzoxygruppe oder eine äquivalente Schutzgruppe geschützt. Die Carbobenzoxygruppe kann in der an sich bekannten Weise entfernt werden.
Die Verfahrensprodukte der Formel I, die basischen Charakter besitzen, bilden Säureadditionssalze mit anorganischen oder organischen Säuren, wie Chlorwasserstoffsäure, Phosphorsäure, Bromwasserstoffäure, Citronensäure, Schwefelsäure, Essigsäure, Ameisensäure, Bernsteinsäure, Maleinsäure, p-Toluolsulfonsäure und dgl.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I sowie deren pharmazeutisch anwendbare Salze können als Antikonvulsiva, Muskelrelaxantien, Sedativa und Anxiolytica verwendet werden. Die antikonvulsive Aktivität wird demonstriert, wenn Mäuse, welchen Verbindungen der Formel I oder deren Salze verabreicht wurden, dem Pentamethylentetrazoltest unterworfen werden. Z.B. zeigt das 7-Nitro-5-phenyl-1-methoxymethyl-1,3-dihydro-2H-1,4- benzodiazepin-2-on, das eine LDso von 600-1200 mg/kg (p.o.) aufweist, bei der Prüfung auf antikonvulsive Wirksamkeit im Pentetrazoltest in Anlehnung an die Methode von Orloff (Proc. Soc. Exptl. Biol.
Med. 70 254-257, 1949) eine APR 2,0 von 0,5 mg/kg (p.o.) [unter APR 2,0 versteht man diejenige Dosis in mg/kg eines Antikonvulsivums, welche den doppelten Pentetrazolverbrauch gegenüber der unbehandelten Kontrollgruppe bewirkt] und das 7-Nitro-5-phenyl-1-äthoxymethyl- 1,3-dihydro-2H-1 4- benzodiazepin-2-on (LDso 1200-2500 mg/kg p.o.) eine APR 2,0 von 0,2 mg/kg (p.o.).
Im Gegensatz dazu zeigt Phenobarbital, ein gebräuchliches Antikonvulsivum, eine APR 2,0 von 30 mg/kg. Die muskelrelaxierende Wirksamkeit kann im Test am Rotierstab demonstriert werden. Z.B. zeigt das oben genannte 7 -Nitro-5 -phenyl- 1 -methoxymethyl- 1,3 -dihydro- 2H-1,4-benzodiazepin-2-on eine HDso von 2 mg/kg (p.o.) und das 7-Nitro-5-phenyl- 1-äthoxymethyl- 1,3-dihydro- 2H-1,4-benzodiazepin-2-on eine HDso von 5 mg/kg (p.o.).
Die erfindungsgemäss erhältlichen Verbindungen der Formel I sowie deren pharmazeutisch anwendbaren Salze können daher als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze, in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser, Gelatine, Milchzukker, Stärke, Magnesiumstearat, Talk, pflanzliche oele, Gummi, Polyalkylenglykole, Vaseline usw., enthalten. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln, oder in flüssiger Form, z.B.
als Lösungen, Suspensionen oder Emulsionen, vorliegen.
Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Dosierung erfolgt nach individuellen Erfordernissen, jedoch ist eine Dosierung von 0,1 mg/kg-10 mg/kg/Tag bevorzugt.
Beispiel l
2 g 4-Acetoxy-7-chlor-1-(methoxymethyl)-5-phenyl- 1,3,4,5-tetrahydro-2H-1,4-benzodiazepin-2-on werden in 30 ml Alkohol und 5 ml Triäthylamin während 30 Minuten unter Rückfluss gekocht. Der nach dem Eindampfen im Vakuum anfallende Rückstand wird an 50 g Kieselgel mit 10% Äthylacetat in Methylenchlorid chromatographiert. Zuerst werden 1,3 g 7-Chlor-1,5-dihydro- 1-(methoxymethyl)-5- phenyl-2H-1,4-benzodiazepin-2-on eluiert. Diese Verbindung zeigt nach Kristallisation aus Äther Hexan einen Smp. von 126-1290C.
Danach werden 0,4 g des isomeren 7-Chlor- 1,3-dihydro- 1-(methoxymethyl)-5- phenyl-2H- 1 ,4-benzodiazepin-2-on eluiert, welches die folgenden spektroskopischen Daten zeigt: Massenspektrum: Ms e314 m Hauptfragmente bei e286 (M0 - CO) 269 (MO - CH-OCH3)
NMR-spektrum: 7,8-7,1 ppm (multiplet), 8 aromatische
Protonen; [CDCl3] 5,40 ppm (doublet) und 4,88 ppm (dou blet),
AB-system mit J= 10 cps, N-CH-O;
4,84 ppm (doublet) und 3,83 ppm (dou- blet),
AB-system mit J= 10,5 cps, G-Protonen;
3,37 ppm (singlet), OCH3.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 28,7 g 7-Chlor-1,3-dihydro-5-phenyl2H-1,4-benzodiazepin-2-on-4-oxid in 200 ml Dimethylformamid wird bei -200C mit 8,1 g Natriummethoxid versetzt und während 5 Minuten gerührt. Bei -200C werden unter Rühren 12 ml Chlordimethyläther zugetropft. Nach der Zugabe lässt man ohne Kühlung während 30 Minuten weiterrühren. Das Reaktionsgemisch wird auf 1 Liter Wasser gegossen und wie üblich aufgearbeitet. Kristallisation des Rohproduktes durch Animpfen aus Methanol-Äther ergibt 11,5 g 7-Chlor-1,3-dihydro-1-(methoxymethyl)-5-phenyl- 2H-1,4-benzodiazepin-2-on-4-oxid, das nach Umkristallisation aus Alkohol bei 16S1660 schmilzt.
Impfkristalle werden durch Chromatographie an Kieselgel mit dem Lösungsmittelsystem Methylchlorid : Äthylacetat 1:1 ethalten.
3,3 g 7-Chlor-1,3-dihydro- 1-(methoxymethyl)-5-phenyl- 2H-1,4-benzodiazepin-2-on-4-oxid werden in 30 ml Eisessig in Gegenwart von 0,3 g Platinoxid bei Atmosphärendruck und Raumtemperatur hydriert. Nach Aufnahme von 400 ml Wasserstoff wird vom Katalysator abgetrennt und eingedampft.
Der Rückstand wird in Benzol aufgenommen und die Lösung mit 2n Salzsäure extrahiert. Die darauf mit Wasser gewaschene Benzolphase wird getrocknet und eingedampft. Kristallisation des Rückstandes aus Methanol liefert 1,1 g 7-Chlor-4-hydroxy-1-(methoxymethyl)-5-phenyl-1,3,4,5- tetrahydro-2H-1,4-benzodiazepin-2-on vom Smp. 168-1720C. Das reine Produkt schmilzt nach Umkristallisation aus Methanol bei 173-1750C.
Die Salzsäureauszüge werden mit Sodalösung alkalisch gestellt und mit Methylenchlorid extrahiert. Die getrockneten Auszüge werden eingedampft und der Rückstand ebenfalls aus Methanol kristallisiert. Man erhält 1,4 g 7 Chlor-1-(methoxymethyl)-5-phenyl-1,3,4,5-tetrahydro- 2H- 1,4-benzodiazepin-2-on.
Eine Lösung von 3,3 g 7-Chlor-1-(methoxymethyl)-4-hydr oxy-5-phenyl- 1,3,4,5-tetrahydro-2H-1,4-benzodiazepin- 2-on in 20 ml Pyridin wird mit 10 ml Acetanhydrid versetzt und während 48 Stunden bei Raumtemperatur stehen gelassen. Nach dem Eindampfen im Vakuum verteilt man den Rückstand zwischen Äther und 2n Salzsäure. Die Ätherphase wird mit Bicarbonatlösung und Wasser gewaschen, getrocknet und eingedampft. Kristallisation des Rückstandes aus Ather Hexan liefert 3,1 g 4-Acetoxy-7-chlor-1-(methoxymethyl)-5-phenyl- 1,3,4,5- tetrahydro-2H-1,4-benzodiazepin-2-on vom Smp. 95-1030C. Durch Umkristallisation aus Methanol verbessert sich der Schmelzpunkt auf 103-1060C.
Beispiel 2
In analoger Weise wie im vorstehenden Beispiel 1 können die folgenden Verbindungen hergestellt werden: 1,3-Dihydro-1-(methoxymethyl)-7-nitro-5-phenyl-2H-1,4- benzodiazepin-2-on; Smp. 139-1410C (aus Benzol-Alkohol) 7-Chlor-5-(o-chlorphenyl)-1,3-dihydro-1-(methoxymethyl)- 2H-1,4-benzodiazepin-2-on; Smp. 139-1400C (aus Methanol).
5-(o-Chlorphenyl)-1,3-dihydro-1-(methoxymethyl)-7-nitro- 2H-1,4-benzodiazepin-2-on; Smp. 136-1370C (aus Methanol) 7-Chlor-1,3-dihydro-5-(o-fluor-phenyl)- 1-(methoxy- methyl)-2H-1,4-benzodiazepin-2-on; Smp. 113-1140C (aus Methanol).

The starting material can be made as follows:
A solution of 28.7 g of 7-chloro-1,3-dihydro-5-phenyl2H-1,4-benzodiazepin-2-one-4-oxide in 200 ml of dimethylformamide is mixed with 8.1 g of sodium methoxide at -200C and stirred for 5 minutes. 3.3 g of 7-chloro-1,3-dihydro- 1- (methoxymethyl) -5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide are dissolved in 30 ml of glacial acetic acid in the presence of 0.3 g of platinum oxide hydrogenated at atmospheric pressure and room temperature. The hydrochloric acid extracts are made alkaline with soda solution and extracted with methylene chloride. The dried extracts are evaporated and the residue is also crystallized from methanol. 1.4 g of 7-chloro-1- (methoxymethyl) -5-phenyl-1,3,4,5-tetrahydro-2H-1,4-benzodiazepin-2-one are obtained.
A solution of 3.3 g of 7-chloro-1- (methoxymethyl) -4-hydroxy-5-phenyl-1,3,4,5-tetrahydro-2H-1,4-benzodiazepin-2-one in 20 ml Pyridine is mixed with 10 ml of acetic anhydride and left to stand for 48 hours at room temperature.
