Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Benzodiazepin-Derivaten und zwar 1,4-Benzodiazepin -2-onen der Formel

worin A Äthylen, Propylen oder Isopropylen, B die Gruppe -O-, -S- oder -NR3-, R1Halogen, Nitro, Trifluormethyl, Alkyl mit 2 bis 5 Kohlenstoffatomen oder Alkanoyl mit höchstens 5 Kohlenstoffatomen, R2 Phenyl, o-Halogenphenyl, o,o'-Dihalogenphenyl oder 2-Pyridiyl, R3 Wasserstoff oder Alkyl mit höchstens 5 Kohlenstoffatomen bedeuten, n für 1 oder 2 steht und R4 und R5 gleich oder verschieden sind und Wasserstoff oder Alkyl mit höchstens 5 Kohlenstoffatomen bedeuten, bzw. von 4-Oxyden davon.
Der Ausdruck Alkyl bedeutet geradkettige oder verzweigte, gesättigte, aliphatische Kohlenwasserstoffreste mit bis zu 5 Kohlenstoffatomen, wie Methyl, Äthyl, Propyl, Isopropyl. Bevorzugt sind der Methyl- und Äthyfrest. Bei R1 in der Bedeutung Alkyl ist der Methylrest ausgeschlossen. In diesem Fall ist der Äthylrest bevorzugt. Der Ausdruck Alkanoyl bezieht sich in analoger Weise auf Acylreste aliphatischer Carbonsäuren mit bis zu 5 Kohlenstoffatomen, wie Formyl, Acetyl, Propionyl. Der Acetylrest ist bevorzugt.
Der Ausdruck Halogen umfasst die vier Halogenatome Chlor, Brom, Jod und Fluor. Falls der Substituent R1 in der Bedeutung Halogen vorliegt, so sind Chlor und Jod bevorzugt. Der Ausdruck Isopropylen umfasst beide isomere Formen, d.h. sowohl -CH(CH3)-CH2- als auch -CH2-CH (CH3)-.
Falls R2 in der Bedeutung 2-Pyridyl vorliegt ist R1 in der Bedeutung Brom ebenfalls bevorzugt. Für den Fall, dass der Substituent R2 o-Halogenphenyl bedeutet, so ist das Halogenatom vorzugsweise ein Fluor- oder Chloratom. Fluoratome sind bevorzugt als Halogenatome in o,o'-Dihalogenphenylsubstituenten.
Abgesehen von den vorstehend erwähnten bevorzugten Substituenten innerhalb eines allgemeinen Begriffs, nimmt die Gruppe -0- und -NH- bzw. -N(CH3)- eine bevorzugte Stellung innerhalb der für die durch das Symbol B veranschaulichte Äther- bzw. Aminogruppen ein.
Verbindungen der allgemeinen Formel I sowie ihre 4-Oxyde erhält man erfindungsgemäss, indem man eine Säure der Formel

worin R1, R3, A, B und n obige Bedeutung haben, bzw. ein reaktionsfähiges Säurederivat davon oder ein entsprechendes 4-Oxyd mit Ammoniak bzw. einem Amin der Formel HNR4R, worin R4 und R5 die genannte Bedeutung haben, umsetzt.
Die Umwandlung einer Säurefunktion in eine Amidfunktion kann in an sich bekannter Weise durch Umsetzung der freien Säure bzw. eines reaktionsfähigen Säurederivates, wie eines Halogenides oder eines Alkylesters mit Ammoniak oder primären oder sekundären Alkylaminen bewirkt werden. Die freie Säure kann zwar direkt umgesetzt werden, doch ist es von Vorteil, diese Umwandlung zweistufig durchzuführen, indem man die Säure vorerst in ein Säurehalogenid, insbesondere das Säurechlorid, oder auch in ein reaktionsfähiges Säureanhydrid überführt, worauf dieses Säurechlorid oder Säureanhydrid anschliessend oder in situ mit Ammoniak oder dem Alkylamin umgesetzt wird. Die überführung in ein Säurechlorid kann z.B. durch Behandeln mit Phosphorpentachlorid oder Thionylchlorid erfolgen.
Die Reaktion wird zweckmässigerweise ohne Lösungsmittel oder in Gegenwart eines inerten organischen Lösungsmittels, wie chlorierten Kohlenwasserstoffen, z.B. Methylenchlorid oder Chloroform, bei Temperaturen, die in der Nähe der Raumtemperatur liegen, z.B. zwischen -20 und +30 C, durchgeführt. Zur Herstellung des Säureanhydrides setzt man die Säure zweckmässigerweise mit einem Chlorkohlensäurealkylester z.B. dem Äthylester um. Man arbeitet dabei mit Vorteil in Gegenwart von tertiären organischen Basen, z.B.
Trialkylaminen, wie Triäthylamin. Die Reaktionstemperatur liegt zweckmässigerweise zwischen 5 und Raumtemperatur. Als Reaktionsmedium kann ein inertes organisches Lösungsmittel dienen, z.B. Chloroform, Toluol, Tetrahydrofuran, Dimethylformamid.
Die Umsetzung des gebildeten Säurechlorides oder -anhydrides mit Ammoniak oder einem primären oder sekundären Alkylamin erfolgt vorteilhafterweise bei einer Temperatur zwischen -5" und Raumtemperatur und in einem Lösungsmittel, wie Wasser, Chloroform, Toluol, Tetrahydrofuran, Dimethylformamid und Gemischen derselben.
Die Ammonolyse bzw. Aminolyse von Estern kann ebenfalls in an sich bekannter Weise durch Behandlung des Esters mit Ammoniak oder einem primären oder sekundären Alkylamin bewirkt werden. Man arbeitet dabei zweckmässigerweise in einem inerten organischen Lösungsmittel, z.B. einem Alkanol, wie Äthanol, einem Kohlenwasserstoff, wie Benzol, oder einem chlorierten Kohlenwasserstoff, wie Chloroform.
Man kann gegebenenfalls auch wässrige Lösungen von Ammoniak bzw. des Amins einsetzen. Die Reaktionstemperatur liegt in der Regel zwischen Raumtemperatur und etwa 1 500C und ist abhängig von dem verwendeten Lösungsmittel, es sei denn man arbeitet unter Druck, was ebenfalls in Betracht zu ziehen ist.
Ester der Formel II werden erhalten, indem man eine Verbindung der Formel

worin R1 und R2 obige Bedeutung haben, oder ein 4-Oxyd davon mit einer Verbindung der allgemeinen Formel X-A-B-CnH2"-COOR 1,,, worin X Halogen bedeutet und A, B und n obige Bedeutung halben, jedoch R, im Falle von B gleich -NR3- eine Alkyloder eine nachträgliche wieder abzuspaltende Schutzgruppe bedeutet, und R eine Alkylgruppe darstellt, umsetzt.
Dieses Verfahren stellt Alkylierung am Stickstoffatom in 1-Stellung des Benzodiazepinmoleküls dar. Solche Alkylierungen sind an sich bekannt. Es ist dabei von Vorteil, wenn man das Benzodiazepin der Formel III oder das 4-Oxyd davon in Form eines Alkali-Derivates mit dem Halogenid der Formel IV zur Reaktion bringt. Man erreicht dies zweckmässigerweise dadurch, dass man die Reaktion in Gegenwart einer starken Alkali enthaltenden Base ablaufen lässt, z.B. unter Einsatz eines Alkalimethylates, wie Natriummethylat, eines Alkaliamides, wie Natriumamid, eines Alkalihydrides, wie Natriumhydrid oder auch von Alkalihydroxyden, wie Natriumhydroxyd.
Die Reaktion einer Verbindung der Formel III oder eines 4-Oxydes davon mit einer Verbindung der Formel IV wird zweckmässigerweise in Gegenwart eines inerten organischen Lösungsmittels durchgeführt. Dafür eignen sich z.B. Dimethylformamid, Dimethylsulfoxyd, Essigester, Alkanole, wie Isopropanol. Viele andere Lösungsmittel und Lösungsmittelgemische sind ebenfalls geeignet; ihre Auswahl bietet dem Fachmann bei Berücksichtigung der Lösungseigenschaften und des inerten Verhaltens in der Reaktion keinerlei Schwierigkeiten. Die Reaktionstemperatur ist in ziemlich weiten Grenzen variierbar. Sie wird in der Regel zwischen Raumtemperatur und der Siedetemperatur des Reaktionsgemisches liegen. Im allgemeinen sollen jedoch Temperaturen über 120"C vermieden werden.
Bevorzugte Reaktionstemperaturen liegen zwischen 60 und 80"C. Die Anwendung von Druck ist bei dieser Reaktion nicht notwendig, aber auch nicht von grundsätzlichem Nachteil.
Anstelle einer Halogenabgangsgruppe, insbesondere Chlor oder Brom, kann eine äquivalente andere Abgangsgruppe verwendet werden, z.B. ein Arylsulfonyloxyrest, wie der Tosyloxyrest, oder ein Alkylsulfonyloxyrest, wie der Mesyloxyrest. Auch quartäre Ammoniumgruppen, wie der Trimethylammoniumrest, können als wirksame Abgangsgruppen dienen.
Falls man eine Verbindung der Formel I oder ein 4-Oxy davon herstellen will, in der B die -NH-Gruppe bedeutet, ist es notwendig, dass im Ausgangsmaterial der Formel IV die Aminogruppe durch eine Schutzgruppe geschützt ist, die nach Umsetzung mit der Verbindung der Formel III oder einem 4-Oxyd davon abgespaltet wird. Als Schutzgruppen eignen sich vor allem Carbobenzoxy- und Benzylreste, wobei es zweckmässig ist, wenn vor der Abspaltung ein Benzylrest gegen einen Carbobenzoxyrest augetauscht wird. Die Abspaltung solcher Reste erfolgt in bekannter Weise durch Solvolyse bzw. Hydrogenolyse.
Säuren der Formel II erhält man durch Verseifung eines Esters der Formel II, welche in an sich bekannter Weise sowohl durch basische als auch durch saure Verseifung bewirkt werden kann. Als saure Mittel eignen sich z.B. verdünnte Salzsäure oder verdünnte Schwefelsäure. Die Reaktionstemperatur beträgt dabei mit Vorteil etwa 60 bis 100"C.
Die basische Verseifung geschieht zweckmässigerweise mit wässrigem Alkali, z.B. Natronlauge, bei einer Temperatur zwischen Raumtemperatur und etwa 60"C und liefert das entsprechende Salz.
Die Ausgangsmaterialien der Formeln III und IV sind generell und spezifisch in vielen Einzelverbindungen bekannt.
Die noch nicht in der Literatur beschriebenen Vertreter davon können ohne Schwierigkeiten in Analogie zu den Darstellungsmethoden für die bekannten Verbindungen hergestellt werden.
Es wird besonders darauf hingewiesen, dass die Ausgangsmaterialien nicht notwendigerweise in isoliertem Zustand eingesetzt werden müssen, sondern auch ohne Isolierung aus dem Reaktionsgemisch, in dem sie hergestellt wurden, weiter eingesetzt werden können.
Erfindungsgemäss erhaltene Verbindungen der Formel I und 4-Oxyde davon, die basischen Charakter besitzen, bilden Säureadditionssalze mit anorganischen oder organischen Säuren wie Chlorwasserstoffsäure, Phosphorsäure, Bromwasserstoffsäure, Citronensäure, Schwefelsäure, Essigsäure, Amei- sensäure, Maleinsäure, p-Toluolsulfonsäure und dgl.
Die Verfahrensprodukte der Formel I, die 4-Oxyde davon und pharmazeutisch anwendbare Salze solcher basischer Verbindungen können als Antikonvulsiva, Muskelrelaxantien, Sedativa und Anxiolytica verwendet werden. Die antikonvulsive Aktivität wird demonstriert, wenn Mäuse, welchen Verbindungen der Formel I oder deren Salze verabreicht wurden, dem Pentamethylentetrazoltest unterworfen werden.
Zum Beispiel zeigt das 1 - [2- (Carbamoylmethoxy)-äthyl]- 7- -chlor-5-(2-fluorphenyl)- 1,3 -dihydro-2H- 1 ,4-benzodiazepin-2 -on, das eine LD50 von > 1250 mg/kg (p.o.) aufweist, bei der Prüfung auf antikonvulsive Wirksamkeit im Pentetrazoltest in Anlehnung an die Methode von Orloff [Proc. Soc.
Exptl. Biol. Med. 70 254-257 (1949)1 eine APR 2,0 von 0,76 mg/kg (p.o.) [unter APR 2,0 versteht man diejenige Dosis in mg/kg eines Antikonvulsivum, welche den doppelten Pentetrazolverbrauch gegenüber der unbehandelten Kontrollgruppe bewirkt]. Im Gegensatz dazu zeigt Phenobarbital, ein gebräuchliches Antikonvulsivum eine APR 2,0 von 30 mg/kg. Die muskelrelaxierende Wirksamkeit im Test am Rotierstab demonstriert werden. Zum Beispiel zeigt das oben genannte Benzodiazepin-derivat eine HDGo von 0,2 mg/kg (p.o.).
Die erfindungsgemäss erhältlichen Verbindungen der Formel I, 4-Oxyde davon sowie deren pharmazeutisch anwendbare Salze solcher basischer Verbindungen können daher als Heilmittel z.B. in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale odere parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Gummi, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln, oder in flüssiger Form, z.B. als Lösungen, Suspensionen oder Emulsionen, vorliegen.
Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Ver- änderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Die folgenden Beispiele dienen zur Darstellung bevorzugter Ausführungsformen der Erfindung. Alle Temperaturen sind in Grad Celsius angegeben und sind korrigiert.
Beispiel I
Eine Lösung von 4,4 g (0,01 M) l-(2-[(Äthoxycarbony1)- methoxy]-äthyl} -7-chlor- 1 ,3-dihydro-5-phenyl-2H- ,4-benzodiazepin-2-on in 20 ml Methanol wird mit 4,2 ml einer 30%igen wässerigen Methylaminlösung versetzt und 3 Tage bei Raumtemperatur stehen gelassen. Nach dem Einengen wird das verbleibende öl in Methylenchlorid aufgenommen und die Lösung mit Wasser gewaschen, über Natriumsulfat ge trocknet und zur Trockene eingedampft. Der ölige Rückstand wird an 150 g Kieselgel zunächst mit Essigester und anschliessend mit einem Gemisch Essigester/Aceton (1:1) chromatographiert.
Die mit Essigester/Aceton eluierten einheitlichen Fraktionen liefern ein ol, das aus Äther kristallisiert; nach Umkristallisation aus Methylenchlorid/Äther erhält man 1-[2-(N-Methyl-carbamoylmethoxy)-äthyl]-7-chlor-1,3-dihy dro-5-phenyl-2H-1,4-benzodiazepin-2-on vom Schmelzpunkt 140-142 .
Den in diesem Beispiel verwendeten Ausgangsstoff erhielt man wie folgt:
Eine Lösung von 12,6 g (0,04 M) 7-Chlor-1,3-dihydro-1- -(2-hydroxyäthyl)-5-phenyl-2H-1,4-benzodiazepin-2-on in 100 ml Tetrahydrofuran wird bei Raumtemperatur mit 2,4 g (0,05 M) einer 50%igen Natriumhydrid-Dispersion in 61 versetzt. Man rührt anschliessend eine Stunde, kühlt auf 5-10 ab und gibt bei dieser Temperatur im Verlaufe von etwa einer Stunde eine Lösung von 8,3 g (0,05 M) Bromessigsäure-äthylester in 20 ml Tetrahydrofuran zu. Die gelbliche Suspension wird dann 20 Stunden bei Raumtemperatur weitergerührt.
Nach dem Einengen unter vermindertem Druck wird der Rückstand zwischen Wasser und Methylenchlorid verteilt, die organische Phase mit einer gesättigten wässrigen Natriumchloridlösung gewaschen, über Natriumsulfat getrock- net und eingedampft. Der ölige Rückstand wird an 280 g Kieselgel mit Essigester als Eluiermittel chromatographiert.
Die einheitlichen Fraktionen werden vereinigt und eingeengt.
Das verbleibende Öl, das aus 1-{{2-[(Äthoxycarbonyl)-methoxy]-äthyl}-7-chlor-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-on besteht, kann ohne weitere Reinigung in die obige Reaktion eingesetzt werden.

The invention relates to a process for the preparation of new benzodiazepine derivatives, namely 1,4-benzodiazepin-2-ones of the formula

wherein A is ethylene, propylene or isopropylene, B is the group -O-, -S- or -NR3-, R1 halogen, nitro, trifluoromethyl, alkyl with 2 to 5 carbon atoms or alkanoyl with at most 5 carbon atoms, R2 phenyl, o-halophenyl, o , o'-dihalophenyl or 2-pyridiyl, R3 is hydrogen or alkyl with a maximum of 5 carbon atoms, n is 1 or 2 and R4 and R5 are identical or different and are hydrogen or alkyl with at most 5 carbon atoms, or of 4-oxides from that.
The term alkyl means straight-chain or branched, saturated, aliphatic hydrocarbon radicals with up to 5 carbon atoms, such as methyl, ethyl, propyl, isopropyl. The methyl and ethyl radicals are preferred. If R1 is alkyl, the methyl radical is excluded. In this case, the ethyl radical is preferred. The term alkanoyl relates in an analogous way to acyl radicals of aliphatic carboxylic acids with up to 5 carbon atoms, such as formyl, acetyl, propionyl. The acetyl radical is preferred.
The term halogen includes the four halogen atoms chlorine, bromine, iodine and fluorine. If the substituent R1 is halogen, chlorine and iodine are preferred. The term isopropylene includes both isomeric forms, i. both -CH (CH3) -CH2- and -CH2-CH (CH3) -.
If R2 is 2-pyridyl, R1 is bromine, too. In the event that the substituent R2 is o-halophenyl, the halogen atom is preferably a fluorine or chlorine atom. Fluorine atoms are preferred as halogen atoms in o, o'-dihalophenyl substituents.
Apart from the above-mentioned preferred substituents within a general term, the group -0- and -NH- or -N (CH3) - occupies a preferred position within the ether and amino groups represented by the symbol B, respectively.
Compounds of the general formula I and their 4-oxides are obtained according to the invention by adding an acid of the formula

in which R1, R3, A, B and n have the above meaning, or a reactive acid derivative thereof or a corresponding 4-oxide with ammonia or an amine of the formula HNR4R, in which R4 and R5 have the meaning mentioned, is reacted.
The conversion of an acid function into an amide function can be effected in a manner known per se by reacting the free acid or a reactive acid derivative such as a halide or an alkyl ester with ammonia or primary or secondary alkyl amines. The free acid can be converted directly, but it is advantageous to carry out this conversion in two stages by first converting the acid into an acid halide, in particular the acid chloride, or into a reactive acid anhydride, whereupon this acid chloride or acid anhydride is then converted or in situ is reacted with ammonia or the alkylamine. The conversion into an acid chloride can e.g. by treatment with phosphorus pentachloride or thionyl chloride.
The reaction is conveniently carried out without a solvent or in the presence of an inert organic solvent such as chlorinated hydrocarbons, e.g. Methylene chloride or chloroform, at temperatures close to room temperature, e.g. between -20 and +30 C. To prepare the acid anhydride, the acid is expediently treated with an alkyl chlorocarbonate, e.g. the ethyl ester. It is advantageous to work in the presence of tertiary organic bases, e.g.
Trialkylamines such as triethylamine. The reaction temperature is conveniently between 5 and room temperature. An inert organic solvent can serve as the reaction medium, e.g. Chloroform, toluene, tetrahydrofuran, dimethylformamide.
The reaction of the acid chloride or anhydride formed with ammonia or a primary or secondary alkylamine is advantageously carried out at a temperature between -5 "and room temperature and in a solvent such as water, chloroform, toluene, tetrahydrofuran, dimethylformamide and mixtures thereof.
The ammonolysis or aminolysis of esters can also be effected in a manner known per se by treating the ester with ammonia or a primary or secondary alkylamine. Compounds of the formula I and 4-oxides thereof obtained according to the invention, which have a basic character, form acid addition salts with inorganic or organic acids such as hydrochloric acid, phosphoric acid, hydrobromic acid, citric acid, sulfuric acid, acetic acid, formic acid, maleic acid, p-toluenesulfonic acid and the like.
The process products of the formula I, the 4-oxides thereof and pharmaceutically acceptable salts of such basic compounds can be used as anticonvulsants, muscle relaxants, sedatives and anxiolytics. The anticonvulsant activity is demonstrated when mice, which compounds of the formula I or their salts have been administered, are subjected to the pentamethylene tetrazole test.

The compounds of the formula I, 4-oxides thereof obtainable according to the invention and their pharmaceutically applicable salts of such basic compounds can therefore be used as medicaments e.g. in the form of pharmaceutical preparations which use them or their salts in a mixture with a pharmaceutical, organic or inorganic inert carrier material suitable for enteral or parenteral administration, such as e.g. Contain water, gelatin, lactose, starch, magnesium stearate, talc, vegetable oils, gum, polyalkylene glycols, petroleum jelly, etc. The pharmaceutical preparations can be in solid form, e.g. as tablets, coated tablets, suppositories, capsules, or in liquid form, e.g. as solutions, suspensions or emulsions.
They may contain auxiliaries such as preservatives, stabilizers, wetting agents or emulsifiers, salts for changing the osmotic pressure or buffers. They can also contain other therapeutically valuable substances.
The following examples serve to illustrate preferred embodiments of the invention. All temperatures are given in degrees Celsius and have been corrected.
Example I.
A solution of 4.4 g (0.01 M) l- (2 - [(Äthoxycarbony1) - methoxy] ethyl} -7-chloro-1, 3-dihydro-5-phenyl-2H-, 4-benzodiazepine- 2-one in 20 ml of methanol is mixed with 4.2 ml of a 30% aqueous methylamine solution and left to stand for 3 days at room temperature After concentration, the remaining oil is taken up in methylene chloride and the solution is washed with water, dried over sodium sulfate and The oily residue is chromatographed on 150 g of silica gel first with ethyl acetate and then with a mixture of ethyl acetate / acetone (1: 1).
The uniform fractions eluted with ethyl acetate / acetone yield an oil which crystallizes from ether; after recrystallization from methylene chloride / ether, 1- [2- (N-methyl-carbamoylmethoxy) -ethyl] -7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one is obtained from melting point 140-142.
The starting material used in this example was obtained as follows:
A solution of 12.6 g (0.04 M) 7-chloro-1,3-dihydro-1- - (2-hydroxyethyl) -5-phenyl-2H-1,4-benzodiazepin-2-one in 100 ml Tetrahydrofuran is treated with 2.4 g (0.05 M) of a 50% strength sodium hydride dispersion in 61 at room temperature. The mixture is then stirred for one hour, cooled to 5-10 and a solution of 8.3 g (0.05 M) ethyl bromoacetate in 20 ml tetrahydrofuran is added at this temperature over the course of about one hour. The yellowish suspension is then stirred for a further 20 hours at room temperature.
After concentration under reduced pressure, the residue is partitioned between water and methylene chloride, the organic phase is washed with a saturated aqueous sodium chloride solution, dried over sodium sulphate and evaporated. The oily residue is chromatographed on 280 g of silica gel with ethyl acetate as the eluent.
The uniform fractions are combined and concentrated.
The remaining oil, which consists of 1 - {{2 - [(ethoxycarbonyl) methoxy] ethyl} -7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one, can be used in the above reaction without further purification.
