Die Erfindung betrifft die Herstellung des neuen Tetrahydrobenz[flisoindolins der Formel I

und seiner Säureadditionssalze.
Erfindungsgemäss gelangt man zu der Verbindung der Formel I und ihren Salzen. indem man die Verbindung der Formel II

mit Hilfe von Metallhydriden reduziert, und die so erhaltene Verbindung der Formel I als solche oder als Säureadditionssalze gewinnt.
Aus der freien Base bzw. Säure lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Die Auftrennung des allfälligen Racemates der Formeln in seine optischen Antipoden kann nach bekannten Methoden: beispielsweise durch fraktionierte Kristallisation der Salze dieses Racemates mit optisch aktiven Säuren erfolgen.
Die Reduktion des Amides der Formel II mit Metallhydriden kann ebenfalls analog zu bekannten Methoden erfolgen.
Als Metallhydride sind z.B. Aluminiumhydrid, Dialkylaluminiumhydride und auch Lithiumaluminiumhydrid oder Lithiumaluminiumhydrid/Aluminiumchlorid geeignet.
Geeignete Lösungsmittel sind unter den Reaktionsbedingungen inerte Lösungsmittel. z.B. cyclische oder offenkettige Äther wie Tetrahydrofuran.
Die Reduktion kann bei Raumtemperatur durchgeführt werden und ist im allgemeinen nach etwa 12 bis 5 Stunden vollendet.
Die nach den obigen Verfahren erhaltene Verbindung der Formel I kann analog zu bekannten Methoden isoliert und gereinigt werden.
Die Verbindung der Formel II ist neu und kann nach an sich bekannten Methoden durch Acylierung von (3aRS, 4SR.
9aSR)-3a,4,9,9a-Tetrahydro-4-phenylbenzff]isoindolin, beispielsweise mit Hilfe von den Cyclopropylcarbonsäurehalogeniden, vorzugsweise den Bromiden oder Chloriden. oder mit den Cyclopropylcarbonsäureanhydriden, hergestellt werden.
Das (3aRS,4SR.9aSR )-3a,4.9.9a-Tetrahydrcphenyl- ben4f]isoindolin ist neu und kann analog zu bekannten Methoden, z.B. ausgehend von Verbindungen der Formel III

worin R für eine elektronenziehende Gruppe steht, durch Abspaltung der Gruppe R hergestellt werden.
Als elektronenziehende Gruppen R sind z.B. Acylgruppen wie die Trifluoracetylgruppe, die Benzoylgruppe, eine aliphatische oder aromatische Sulfonylgruppe, beispielsweise ein Tosylrest, ein (nieder)-Alkoxycarbonylrest wie die Methoxyoder Äthoxycarbonylgruppe, oder die Phenoxycarbonylgruppe geeignet.
Die Abspaltung der Gruppe R wird beispielsweise durch Hydrolyse mit einer 1 bis etwa SN-Lösung eines Alkalimetallhydroxids wie Natrium- oder Kaliumhydroxid in einem niederen Alkanol, vorzugsweise Methanol oder Äthanol durchgeführt.
Die Verbindung der Formel III sind ebenfalls neu.
Man erhält sie z.B. durch thermische Cyclisierung der Verbindungen der Formel IV

worin R obige Bedeutung besitzt.
Die thermische Cyclisierung der Verbindungen der Formel IV kann in einem inerten organischen Lösungsmittel mit vorzugsweise hohem Siedepunkt wie Dichlorbenzol durchgeführt werden. Man arbeitet zweckmässig unter Sauerstoffausschluss und erhitzt das Reaktionsgemisch auf etwa 160-190 während etwa 16 bis 30 Stunden.
Soweit die Herstellung der Ausgangsverbindungen nicht be schrieben wird. sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen Verfahren oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindung der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen interessante pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden.
Mit Tierversuchen wurden insbesondere pharamakologische Effekte gefunden, die für Antidepressiva typisch sind. So heben die erfindungsgemässe Verbindung und ihre Säureadditionssalze den kataleptischen Zustand (Haltestarre) auf, der durch Verabreichung von Tetrabenazin an die Ratte hervorgerufen wird (Tetrabenazin-Antagonismus).
Aufgrund ihrer antidepressiven Wirkung sind sie zur Behandlung von Depressionen geeignet.
Für die obige Anwendung hängt die zu verabreichende Dosis von der verwendeten Verbindung und der Verabreichungsart sowie der Behandlungsart ab. Eine täglich zu verabreichende Menge von ungefähr 50 bis ungefähr 500 mg ist angezeigt. Diese täglich zu verabreichende Menge kann auch in kleineren Dosen, z.B. 2 bis 4mal täglich, oder in Retardform verabreicht werden. Eine Einheitsdosis, beispielsweise eine zur oralen Verabreichung geeignete Tablette, kann zwischen ungefähr 12,5 und ungefähr 250 mg des Wirkstoffes, zusammen mit geeigneten Hilfsstoffen, enthalten.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform verabreicht werden.
Die Verbindung der Formel I bildet mit Säuren wie Chlorwasserstoff, Bromwasserstoff, Fumarsäure, Maleinsäure, Weinsäure, Naphthalin- 1,5-disulfonsäure usw., stabile, meist wasserlösliche Salze.
Im nachfolgenden Beispiel, das die Erfindung näher erläutert, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Beispiel (3aRS,4SR,9aSR)-2-Cyclopropylmethyl-3a,4,9,9a-tetrahydro- -4-phenylbenz[fisoindolin
Zu einer warmen Lösung von 8,6 g (3aRS,4SR,9aSR) -3a,4,9,9a-Tetrahydro-4-phenylbenz[fìisoindolin- 2-cyclopropylcarbonsäureamid in 100 ml Tetrahydrofuran wird portionenweise 1 g Lithiumaluminiumhydrid zugegeben. Das Gemisch wird 1 Stunde bei Raumtemperatur gerührt, dann mit 2 N Natronlauge zersetzt und filtriert. Das eingedampfte Filtrat wird mit Kieselgel in Benzol/Essigester 1:1 aufgeschlämmt, filtriert und eingedampft, wobei man die im Titel genannte Verbindung vom Smp. 65-68 (nach Kristallisation aus Pentan) erhält.
Das Ausgangsmaterial erhält man z.B. wie folgt: a) ZurAufschlämmung von 30,2 g Natriumhydrid in 540 ml Hexamethylphosphorsäuretriamid wird unter Kühlung und Rühren eine Lösung von 275 g N-Cinnamyl-trifluoracetamid in 600 ml Hexamethylphosphorsäuretriamid zugetropft.
Nach Beendigung der Gasentwicklung wird die Lösung von 248,5 g Cinnamylbromid in 540 ml Hexamethylphosphorsäuretriamid zugetropft und das Gemisch 16 Stunden bei 25O gerührt. Dann wird das Reaktionsgemisch auf Wasser gegossen und mit Äther extrahiert. Die über Natriumsulfat getrocknete Ätherlösung wird eingedampft und der ölige Rückstand mit Toluol an 1,5 kg Kieselgel chromatographiert.
Das Filtrat ergibt beim Eindampfen das N,N-Bis(transcinnamyl)trifluoracetamid als öligen Rückstand: IR (CH2C12) 1690, 968 cm-1.
b) Die Lösung von 310 g N,N-Bis(trans-cinnamyl)-trifluor acetamid in 6 Liter o-Dichlorbenzol wird 16 Stunden unter Argonatmosphäre am Rückfluss zum Sieden erhitzt und anschliessend eingedampft. Der Rückstand ergibt nach Kristallisation aus Methylenchlorid/Pentan (3aRS,4SR,9aSR) -3a,4,9,9a-Tetrahydro-4-phenylbenz[flisoindolin-2-trifluor- -acetamid vom Smp. 150-153 . IR (CH2Cl2) 1690, keine Bande bei 950 bis 990 cm-1.
c) 22,3 g (3aRS ,4SR,9aSR)-3a,4,9,9a-Tetrahydro-4-phenyl benz[f]isoindolin-2-trifluoracetamid werden unter Erwärmen in 3 N methanolischer Kalilauge gelöst. Das Gemisch wird 30 Minuten bei Raumtemperatur gerührt, anschliessend auf Wasser gegossen und mit Methylenchlorid extrahiert. Men trocknet den Extrakt über Natriumsulfat, dampft die Lösung ein und kristallisiert den Rückstand aus Methylenchlorid/Pentan, wobei man das (3aRS,4SR,9aSR)-3a,4,9,9a-Tetrahydro-4-phenyl- benz[flisoindolin, Smp. 136-138 , erhält.
d) Zu dem Gemisch von 7,5 g (3aRS,4SR,9aSR)-3a,4,9,9a- -Tetrahydro-4-phenylbenz[flisoindolin, 7,9 g Pyridin und
100 ml Methylenchlorid werden bei - 5 4,7 g Cyclopropylcarbonsäurechlorid zugetropft. Das Gemisch wird 1 Stunde bei Raumtemperatur gerührt, anschliessend nacheinander mit lO%iger wässriger Zitronensäurelösung und Natriumhydrogencarbonatlösung geschüttelt, über Natriumsulfat getrocknet und eingedampft, wobei man als Rückstand das (3aRS,4SR, 9aSR)-3a,4,9,9a-Tetrahydro-4-phenylbenz[fXisoindolin-2-cy- clopropylcarbonsäureamid vom Smp. 149-151" (nach Kristallisation aus Äther) erhält.

The (3aRS, 4SR.9aSR) -3a, 4.9.9a-Tetrahydrcphenylben4f] isoindoline is new and can be used analogously to known methods, e.g. starting from compounds of the formula III

The compound of the formula I and its pharmacologically acceptable acid addition salts have interesting pharmacodynamic properties and can therefore be used as medicaments.
Animal experiments have shown in particular pharmacological effects that are typical of antidepressants.
