Erfindungsgemäss gelangt man zu der Verbindung der Formel I und ihren Säureadditionssalzen, indem man von Verbindungen der Formel II

worin R für eine Acylgruppe steht, die Gruppe R abspaltet und die so erhaltene Verbindung der Formel I als Base oder als Säureadditionssalz gewinnt.
Aus der freien Base lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Im folgenden wird auf verfahrenstechnische Einzelheiten hingewiesen, die zweckmässig bei der erfindungsgemässen Herstellung der Verbindung der Formel I berücksichtigt werden sollen.
Als Acylgruppen R sind z.B. die Trifluoracetylgruppe, die Benzoylgruppe, eine aliphatische oder aromatische Sulfonylgruppe, beispielsweise ein Tosylrest, ein (nieder)-Alkoxycarbonylrest wie die Methoxy- oder Äthoxycarbonylgruppe, oder die Phenoxycarbonylgruppe geeignet.
Die erfindungsgemässe Abspaltung der Gruppe R wird beispielsweise durch Hydrolyse mit einer 1 bis etwa 5N-Lösung eines Alkalimetallhydroxids wie Natrium- oder Kaliumhydroxid in einem niederen Alkanol, vorzugsweise Methanol oder Äthanol durchgeführt.
Steht R für eine leicht abspaltbare Acylgruppe, z.B. die Trifluoracetylgruppe, so kann die Hydrolyse bei Raumtemperatur bzw. leicht erhöhter Temperatur erfolgen. Die Hydrolyse ist dann nach etwa !/1 bis etwa 3 Stunden vollendet.
Steht R für eine weniger leicht abspaltbare Acylgruppe, z.B. für die Phenoxycarbonylgruppe, so arbeitet man zweckmässig unter Erwärmen, vorzugsweise unter Rückflusstemperatur des Reaktionsgemisches. Die Reaktion dauert dann etwa 10 bis etwa 20 Stunden.
Steht R für eine aliphatische oder aromatische Sulfonylgruppe, so kann diese Gruppe unter reduktiven Bedingungen - analog zu bekannten Methoden - beispielsweise mit Natrium-Ammoniak oder Phenol in 40% Bromwasserstoffsäure abgespalten werden.
Die Hydrolyse der Verbindungen der Formel VI kann auch unter sauren Bedingungen, beispielsweise mit Hilfe von 2N Chlorwasserstoffsäure, zweckmässig bei erhöhter Temperatur, vorzugsweise bei Rückflusstemperatur des Reaktionsgemisches erfolgen.
Die Verbindungen der Formel lt sind neu.
Man erhält sie z.B. durch thermische Cyclisierung der Verbindungen der Formel III

worin R obige Bedeutung besitzt.
Die thermische Cyclisierung der Verbindungen der Formel III kann in einem inerten organischen Lösungsmittel mit vorzugsweise hohem Siedepunkt wie Dichlorbenzol durchgeführt werden. Man arbeitet zweckmässig unter Sauerstoffausschluss und erhitzt das Reaktionsgemisch auf etwa 160-190 während etwa 16 bis 30 Stunden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen Verfahren oder analog zu an sich bekannten Verfahren hertellbar.
Die Verbindung der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen interessante pharmakodinamische Eigenschaften und können daher als Heilmittel verwendet werden.
Mit Tierversuchen wurden insbesondere pharmakologische Effekte gefunden, die für ein Antidepressivum typisch sind.
So hebt die erfindungsgemässe Verbindung den kataleptischen Zustand (Haltestarre) auf, der durch Verabreichung von Tetrabenazin an die Ratte hervorgerufen wird (Tetrabenazin Antagonismus).
Aufgrund ihrer antidepressiven Wirkung ist sie zur Behandlung von Depressionen geeignet.
Für die obige Anwendung hängt die zu verabreichende Dosis von der verwendeten Verbindung und der Verabreichungsart sowie der Behandlungsart ab. Eine täglich zu verabreichende Menge von ungefähr 50 bis ungefähr 500 mg ist angezeigt. Diese täglich zu verabreichende Menge kann auch in kleineren Dosen, z.B. 2 bis 4mal täglich, oder in Retardform verabreicht werden. Eine Einheitsdosis, beispielsweise eine zur oralen Verabreichung geeignete Tablette, kann zwischen ungefähr l 2,5 und ungefähr 250 mg des Wirkstoffes, zusammen mit geeigneten Hilfsstoffen, enthalten.
Als Heilmittel kann die Verbindung der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform verabreicht werden.
Die Verbindung der Formel I bildet mit Säuren wie Chlorwasserstoff, Bromwasserstoff, Fumarsäure, Maleinsäure, Weinsäure, Naphthalin-l,5-disulfonsäure usw., stabile, meist wasserlösliche Salze.
Im nachfolgenden Beispiel, das die Erfindung näher erläutert, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Beispiel (3aRS,4SR,9aSR)-3a,4,9,9a-Tetrahydro4-phenyi-benz[tl- isoindolin
22,3 g (3aRS,4SR,9aSR)-3a,4,9,9a-Tetrahydro-4-phenyl- benz[f]isoindolin-2-trifluoracetamid werden unter Erwärmen in 3 N methanolischer Kalilauge gelöst. Das Gemisch wird 30 Minuten bei Raumtemperatur gerührt, anschliessend auf Wasser gegossen und mit Methylenchlorid extrahiert. Man trocknet den Extrakt über Natriumsulfat, dampft die Lösung ein und kristallisiert den Rückstand aus Methylenchlorid/ Pentan, wobei man die im Titel genannte Verbindung, Smp.
136-138", erhält.
Das als Ausgangsmaterial benötigte (3aRS,4SR,9aSR) -3a,4,9,9a-Tetrahydro.4-phenyl-benz[isoindolin-2-trifluor- acetamid erhält man z.B. wie folgt:
Die Lösung von 310 g N,N-Bis(trans-cinnamyl > -trifluor- acetamid in 6 1 o-Dichlorbenzol wird 16 Stunden unter Argonatmosphäre am Rückfluss zum Sieden erhitzt und anschliessend eingedampft. Der Rückstand ergibt nach Kristallisation aus Methylenchlorid/Pentan (3aRS,4SR,9aSR)-3a,4,9, 9a-Tetrahydro-4-phenyl-benz[f]isoindolin-2-tnfluor-acetamid vomSmp. 150-1530.
IR (CH2Cl2 > 1690, keine Bande bei 950 bis 990 cm-'.
Das als Ausgangsprodukt benötigte N,N-Bis(trans-cinnamyl)trifluoracetamid kann folgendermassen hergestellt werden:
Zur Aufschlämmung von 30,2 g Natriumhydrid in 540 ml Hexamethylphosphorsäuretriamid wird unter Kühlung und Rühren eine Lösung von 275 g N-Cinnamyl-trifluoracetamid in 600 ml Hexamethyiphosphorsäuretriamid zugetropft. Nach Beendigung der Gasentwicklung wird die Lösung von 248,5 g Cinnamylbromid in 540 ml Hexamethylphosphorsäuretriamid zugetropft und das Gemisch 16 Stunden bei 25 gerührt.
Dann wird das Reaktionsgemisch auf Wasser gegossen und mit Äther extrahiert. Die über Natriumsulfat getrocknete Ätherlösung wird eingedampft und der ölige Rückstand mit Toluol an 1,5 kg Kieselgel chromatographiert.
Das Filtrat ergibt beim Eindampfen das N,N-Bis(transcinnamyl)trifluoracetamid als öligen Rückstand: IR (CH2CI2) 1690, 968 cm-1.
Unter den üblichen Bedingungen gelangt man auch durch Reduktion von (3aRS,4SR,9aSR)-3a,4,9,9a-Tetrahydro-4-phe- nyl-benz[flisoindolin-2-p-toluolsulfonsäureamid zur Titelverbindung. Das als Ausgangsmaterial benötigte (3aRS,4SR, 9aSR)-3a,4,9,9a-Tetrahydro-4-phenyl-benz[f]isoindolin-2-p- -toluolsulfonsäureamid erhält man z.B. wie folgt:
Die Lösung von 2 g N,N-Bis(trans-cinnamyl)-p-toluolsulfonsäureamid in 120 ml o-Dichlorbenzol wird 48 Stunden unter Argonatmosphäre am Rückfluss zum Sieden erhitzt und anschliessend eingedampft, wobei man (3aRS,4SR,9aSR) -3a,4,9,9a-Tetrahydro-4-phenyl-benz[f]isoindolin-2-p-toluol- sulfonsäureamid als Rückstand erhält, Smp. 185-1870 (nach Kristallisation aus Äther/Pentan).

In the following, procedural details are pointed out which should expediently be taken into account in the preparation of the compound of the formula I according to the invention.
As acyl groups R are e.g. the trifluoroacetyl group, the benzoyl group, an aliphatic or aromatic sulfonyl group, for example a tosyl group, a (lower) alkoxycarbonyl group such as the methoxy or ethoxycarbonyl group, or the phenoxycarbonyl group are suitable.
The inventive cleavage of the group R is carried out, for example, by hydrolysis with a 1 to about 5N solution of an alkali metal hydroxide such as sodium or potassium hydroxide in a lower alkanol, preferably methanol or ethanol.
When R stands for an easily cleavable acyl group, e.g. the trifluoroacetyl group, the hydrolysis can take place at room temperature or at a slightly elevated temperature. The hydrolysis is then complete after about 1/1 to about 3 hours.
If R stands for a less easily cleavable acyl group, e.g. for the phenoxycarbonyl group, it is expedient to work with heating, preferably under the reflux temperature of the reaction mixture. The reaction then takes about 10 to about 20 hours.
If R stands for an aliphatic or aromatic sulfonyl group, this group can be split off under reductive conditions - analogously to known methods - for example with sodium ammonia or phenol in 40% hydrobromic acid.
The hydrolysis of the compounds of the formula VI can also take place under acidic conditions, for example with the aid of 2N hydrochloric acid, advantageously at an elevated temperature, preferably at the reflux temperature of the reaction mixture.

They are obtained e.g. by thermal cyclization of the compounds of the formula III

The thermal cyclization of the compounds of the formula III can be carried out in an inert organic solvent, preferably with a high boiling point, such as dichlorobenzene. It is expedient to work with the exclusion of oxygen, and the reaction mixture is heated to about 160-190 for about 16 to 30 hours.
If the preparation of the starting compounds is not described, these are known or can be prepared by processes known per se or analogously to the processes described here or analogously to processes known per se.
The compound of the formula I and its pharmacologically acceptable acid addition salts have interesting pharmacodynamic properties and can therefore be used as medicaments.
In particular, animal experiments have found pharmacological effects that are typical of an antidepressant.
Thus, the compound according to the invention removes the cataleptic state (stiffness) which is caused by administration of tetrabenazine to the rat (tetrabenazine antagonism).
Because of its antidepressant effect, it is suitable for the treatment of depression.
For the above application, the dose to be administered depends on the compound used and the mode of administration and the mode of treatment. An amount of about 50 to about 500 mg to be administered daily is indicated. This amount to be administered daily can also be used in smaller doses, e.g. Administered 2 to 4 times a day, or in a sustained-release form.
