Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 9-(2-OR-3 -Aminopropyl)-9,1 O-dihydro-8.10- äthanoanthracene der allgemeinen Formel

Als niedere Alkylreste werden Reste bezeichnet, die bis zu 7 und vor allem 14 Kohlenstoffatome enthalten, wie Methyl-. Äthyl-. Propyl-. Isopropyl-. gerade und verzweigte, in beliebiger Stellung verbundene Butyl-. Pentyl-, Hexyl- und Heptylreste.
Niedere Hydroxylalkylreste sind z. B. der 2-Hydroxy- äthyl- und der 3-Hydroxypropylrest.
Alkenylreste sind Reste mit nicht mehr als 7 Kohlenstoffatomen. wie z. B. der Allyl- oder Methallylrest.
Als niedere Cycloalkyl- und Cycloalkenylreste werden Reste wie Cyclopentyl-, Cyclohexyl-, Cyclopropyl-. Cyclopentenyl- und Cyclohexenylreste bezeichnet.
Halogen ist Fluor. Brom. Jod und ganz besonders Chlor.
Niedere Alkylmercaptogruppen sind z. B. die Äthyl- oder Methylmercaptogruppe.
Als Alkanoylreste sind vor allem der Acetyl-. Propionylund Butyrylrest zu nennen.
Niedere Alkoxy- oder Alkenyloxygruppen sind z. B. die Methoxy-. Äthoxy-. Allyloxy- oder Methylendioxygruppe.
Die -NHR,-Gruppe ist vor allem eine Niederalkylaminogruppe. wie die Propylamino- oder vorzugsweise Äthylaminogruppe. vor allem aber die Methylaminogruppe, oder eine Cycloalkylaminogruppe. wie die Cyclopropyl-, Cyclobutyl-. Cyclopentyl- oder Cyclohexylaminogruppe. Die Hydroxygruppe kann veräthert oder acyliert sein.
Eine verätherte Hydroxygruppe ist z. B. eine Phenylniederalkoxy-. wie Benzyloxygruppe, eine Niederalkenyloxy-. wie Allyloxy- oder Metallyloxygruppe oder insbesondere eine Niederalkoxygruppe. Niedere Alkoxygruppen sind z. B. Methoxy-. Äthoxy-. Propoxy-. Isopropoxy-, Butoxyoder Pentoxygruppen.
Eine acylierte Hydroxygruppe. d. h. Acyloxygruppe. leitet sich in erster Linie von einer Carbonsäure ab, z. B. einer aro matischen Carbonsäure, wie Benzoesäure, oder vor allem einer aliphatischen Carbonsäure. wie einer Alkansäure. z. B.
Butter-, Propion- oder insbesondere Essigsäure.
Die neuen Verbindungen besitzen wertvolle pharmakolo gische Eigenschaften, insbesondere eine psychotrope. z. B.
antidepressive Wirkung. So bewirken sie insbesondere eine
Hemmung der Noradrenalinaufnahme, wie sich im Tierver such, z. B. an Herz und Hirn der Ratte. bei einer Gabe von 0.5 bis 10 mg/kg s. c., 0,2-5 mg/kg i. v. oder 5-100 mg p. o.
zeigt.
Die neuen Verbindungen können daher als psychotrope.
insbesondere als antidepressive Mittel Verwendung finden.
Sie können auch als Zusatzstoffe zu Tierfutter verwendet werden, da sie eine bessere Nahrungsverwertung und eine Gewichtszunahme dieser Tiere bewirken. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller. insbesondere pharmazeutischer wirksamer Verbindungen dienen. So können z. B.
9-(3 -Amino-1 -propenyl)-9, 1 0-dihydro-9, 10-äthano-anthracene hergestellt werden, indem man in den neuen Verbindungen die OH-Gruppe als Wasser abspaltet.
Hervorzuheben sind vor allem Verbindungen der Formel I, worin -NHR1 eine Aminogruppe bedeutet. die durch einen Alkylrest, Hydroxy- oder Aminoalkylrest, Alkenyl-. unsubstituierten und ein-, zwei- oder mehrfach niederalkylsubstituierten niederen Cycloalkyl- und/oder Cycloalkenylrest.
unsubstituierten und ein-, zwei- oder mehrfach niederalkylsubstituierten niederen Cycloalkyl- und/oder -alkenylalkylrest, zweifach substituiert ist.
Besonders wertvoll sind Verbindungen der Formel I, worin -NHR1 eine Niederalkylaminogruppe. eine Hydroxyniederalkylaminogruppe bedeutet, RA für niedere Alkyl-.
Alkoxy-, Trifluormethylgruppen oder vorzugsweise Halogenatome, wie Brom- oder insbesondere Chloratome steht und Rlo ein Halogenatom, vorzugsweise ein Chloratom. oder vor allem ein Wasserstoffatom bedeutet.
Hervorzuheben sind vor allem Verbindungen der Formel

Ein durch Reduktion abspaltbarer Rest X2 ist z. B. ein a-Aralkylrest. wie ein Benzylrest oder ein a-Aralkoxycarbonylrest, wie ein Carbobenzoxyrest. der beispielsweise durch Hydrogenolyse. z. B. durch Reduktion mit katalytisch erregtem Wasserstoff, wie Wasserstoff in Gegenwart eines Hydrierungskatalysators, wie eines Palladium- oder Platinkatalysators, abgespalten werden kann. X1 bzw. Vor allem verwendet man Zink oder Zinklegierungen in Essigsäure. Ferner kommen auch Chrom-II-verbindungen, wie Chrom-II-chlorid oder Chrom-II-acetat. in Betracht. X1 bzw. X2 kann auch eine Arylsulfonylgruppe, wie die Toluolsulfonylgruppe. sein, die in üblicher Weise, durch Reduktion mit nascierendem Wasserstoff, z. B. durch ein Alkalimetall, wie Lithium oder Natrium, in flüssigem Ammoniak abgespalten werden kann. Die Abspaltung einer Arylsulfonylgruppe kann auch mit einem Hydrid, z. B. einem der oben genannten einfachen oder komplexen Hydride, vorzugsweise Lithiumaluminiumhydrid, zweckmässig in Gegenwart eines inerten Lösungsmittels, wie eines Äthers. z. B. Tetrahydrofuran, vorgenommen werden.
In erhaltenen Verbindungen kann man im Rahmen der Definition der Endstoffe Substituenten einführen. abwandeln oder abspalten.
In erhaltenen Verbindungen, die verätherte Hydroxyreste aufweisen, kann man diese in üblicher Weise in freie Hydroxygruppen umwandeln. Diese Umwandlung erfolgt z. B.
durch Hydrolyse, vor allem mittels starker Säuren. wie z. B.
Jodwasserstoffsäure oder Bromwasserstoffsäure, und gegebenenfalls in Gegenwart von Leichtmetallhalogeniden, wie Aluminiumbromid oder Borbromid, oder auch mit Pyridinhydrochlorid oder Aluminiumchlorid in Pyridin.
In erhaltenen Verbindungen, die Mercaptogruppen enthalten, kann man die Mercaptogruppen zu Alkylsulfonylgruppen oxydieren. Die Oxydation erfolgt in üblicher Weise, z. B. mit Perverbindungen, wie Wasserstoffperoxyd oder Persäuren, insbesondere organischen Persäuren wie Peressigsäure oder einer Perbenzoesäure.
Die Erfindung betrifft auch diejenigen Ausführungsformen eines Verfahrens, nach denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen gebildeten rohen Gemisches einsetzt.
Die genannten Reaktionen werden in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensations-, und/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z. B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasser stoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäuren, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxy äthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vcrausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können, je nach der Wahl der Ausgangsstoffe und Arbeitsweisen und je nach der Anzahl der asymmetrischen Kohlenstoffatome, als optische Antipoden, Racemate oder als Isomerengemische (z. B. Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) können auf Grund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Isomeren (z. B. Racemate) aufgetrennt werden, beispielsweise durch Chromatographie und/ oder fraktionierte Kristallisation.
Erhaltene Racemate lassen sich nach bekannten Methoden zerlegen, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen, oder durch Umsetzen mit einer mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z. B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, gefolgt von der Freisetzung der Antipoden durch Einwirkung geeigneter Mittel. Besonders gebräuchliche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs beispielsweise genannten oder besonders hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe können, soweit sie neu sind, nach an sich bekannten Methoden erhalten werden.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze, besonders der therapeutisch verwendbaren Salze, in Mischung mit einem z. B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Lactose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche t)le, Benzylalkohole, Gummi, Propylenglykole, Vaseline oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Kapseln, Suppositorien oder in flüssiger Form als Lösungen (z.
B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Lösungsvermittler oder Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch andere therapeutisch wertvolle Substanzen enthalten. Die pharmazeutischen Präparate werden nach üblichen Methoden gewonnen.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z. B. die üblichen Streck- und Verdünnungsmittel bzw. Futtermittel angewendet.
Die Erfindung wird in den folgenden Beispielen näher be schrieben. Die Temperaturen sind in Celsiusgraden angege ben.
Beispiel 1
Eine Lösung von 12 g N-(p-Toluolsulfonyl)-9-(2-hydr- oxy-3-methylamino-propyl)-9, 10-dihydro-9, 10-äthanol-an- thracen in 100 ml Tetrahydrofuran tropft man langsam zu 3 g
Lithiumaluminiumhydrid in 50 ml Tetrahydrofuran. Nachdem die Reaktionsmischung 4 Stunden am Rückfluss gekocht hat, wird auf Raumtemperatur abgekühlt, und man gibt vorsichtig
3 ml Wasser und 10 ml 15%ige Natronlauge zu. Der ausge fallene Niederschlag wird filtriert und das Filtrat eingedampft.
Man erhält im Rückstand das 9-(2-Hydroxy-3 -methylamino- propyl)-9, 1 0-dihydro-9, 1 0-äthanol-anthracen, dessen Cyclo hexylsulfaminat den F. 143-145 C hat.
Das als Ausgangsmaterial verwendete Toluolsulfonamid kann wie folgt hergestellt werden:
Zu einer Lösung von 25 g 9-(2-Hydroxy-3-aminopropyl)
9,10-dihydro-9,10-äthano-anthracen in 200 ml Pyridin gibt man 25 g p-Toluolsulfochlorid. Nach 12 Stunden wird mit
Wasser versetzt und nach Zugabe von 50 ml 5n Salzsäure mit
Methylenchlorid extrahiert. Der nach dem Trocknen und
Eindampfen des Lösungsmittels verbleibende kristalline
Rückstand wird in 150 ml Dimethylformamid gelöst und mit einer Lösung von Diazomethan in Äther versetzt. Nach 12 Stunden Stehen bei Raumtemperatur wird die Lösung im Vakuum eingedampft. Man erhält als Rückstand das rohe N-(p-Toluolsulfonyl)-9, 10-dihydro-9, 1 0-äthanol-anthracen, das ohne weitere Reinigung für die oben beschriebene Umsetzung verwendet werden kann.
Beispiel 2
Zu einer Lösung von 1,0 g Lithiumaluminiumhydrid in 20 ml Tetrahydrofuran gibt man eine Lösung von 2,1 g 549, 10-Dihydro-9, 10-äthanol -9-anthryl-methylj-oxazolidi- non-(2) in 20 ml Tetrahydrofuran und kocht während 4 Stunden unter Rückfluss. Anschliessend wird abgekühlt und dann gibt man 2 ml Wasser, 2 ml 15prozentiger Natronlauge und nochmals 6 ml Wasser zu. Den ausgefallenen Niederschlag filtriert man ab, dämpft das Filtrat ein und löst den Rückstand mit 2n Essigsäure auf. Die saure Lösung wird mit Äther gewaschen. Nach Zugabe von 10prozentiger Natronlauge zum sauren Auszug bis zur alkalischen Reaktion wird mit Methylenchlorid ausgeschüttelt.
Es bleibt nach dem Eindampfen des Lösungsmittels das 9-(2-Hydroxy-3-methylamino-propyl)- 9,10-dihydro-9,10-äthanol-anthracen zurück, dessen Hydrochlorid bei 237-239 schmilzt.
Das als Ausgangsmaterial verwendete Oxazolidinon kann wie folgt hergestellt werden:
Zu einer Lösung von 2,8 g 9-(2-Hydroxy-3-amino-propyl)-9,10-dihydro-9,10-äthano-anthracen in 50 ml Benzol, 20 ml Dioxan und 5 ml 2n Natronlauge werden unter Rühren bei Raumtemperatur 20 ml einer 10prozentigen Lösung von Phosgen in Toluol getropft. Nach 3 Stunden filtriert man den ausgefallenen Anteil ab und dampft das Filtrat ein. Den Rückstand löst man in Methylenchlorid und extrahiert mit einer 3prozentigen wässrigen Lösung von Methansulfonsäure.
Nach dem Trocknen und Eindampfen des Lösungsmittels bleibt das rohe 5 -[9,1 0-Dihydro-9, 10-äthanol -9-anthryl-me- thyll-oxazolidinon-(2) als feste Masse zurück.
