Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 9-(2-Hydroxy-3-arninopropyl)-9,10-dihydro- 9,10-äthanoanthracene der allgemeinen Formel

Als niedere Alkylreste werden Reste bezeichnet, die bis zu 7 und vor allem 1-4 Kohlenstoffatome enthalten, wie Methyl-, Äthyl-, Propyl-, Isopropyl-, gerade und verzweigte, in beliebiger Stellung verbundene Butyl-, Pentyl-, Hexylund Heptylreste.
Niedere Hydroxyalkylreste sind z. B. der 2-Hydroxyäthyl- und der 3-Hydroxypropylrest.
Alkenylreste sind Reste mit nicht mehr als 7 Kohlenstoffatomen, wie z. B. der Allyl- oder Methallylrest.
Als niedere Cycloalkyl- und Cycloalkenylreste werden Reste wie Cyclopentyl-, Cyclohexyl-, Cyclopropyl-, Cyclopentenyl- und Cyclohexenylreste bezeichnet.
Halogen ist Fluor, Brom, Jod und ganz besonders Chlor.
Niedere Alkylmercaptogruppen sind z. B. die Äthyl- oder Methylmercaptogruppe.
Als Alkanoylreste sind vor allem der Acetyl-, Propionylund Butyrylrest zu nennen.
Niedere Alkoxy- oder Alkenyloxygruppen sind z. B. die Methoxy-, Äthoxy-, Allyloxy- oder Methylendioxygruppe.
Die -NHR1-Gruppe ist vor allem eine Niederalkylaminogruppe, wie die Propylamino- oder vorzugsweise Äthylaminogruppe, vor allem aber die Methylaminogruppe, oder eine Cycloalkylaminogruppe, wie die Cyclopropyl-, Cyclobutyl-, Cyclopentyl- oder Cyclohexylaminogruppe.
Die Hydroxygruppe kann veräthert oder acyliert sein.
Eine verätherte Hydroxygruppe ist z. B. eine Phenyl- niederalkoxy-, wie Benzyloxygruppe, eine Niederalkenyloxy-, wie Allyloxy- oder Methallyloxygruppe oder insbesondere eine Niederalkoxygruppe. Niedere Alkoxygruppen sind z. B.
Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy- oder Pentoxygruppen.
Eine acylierte Hydroxygruppe, d. h. Acyloxygruppe, leitet sich in erster Linie von einer Carbonsäure ab, z. B.
einer aromatischen Carbonsäure, wie Benzoesäure, oder vor allem einer aliphatischen Carbonsäure, wie einer Alkansäure, z. B. Butter-, Propion- oder insbesondere Essigsäure.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine psychotrope, z. B.
antidepressive Wirkung. So bewirken sie insbesondere eine Hemmung der Noradrenalinaufnahme, wie sich im Tierversuch, z. B. an Herz und Hirn der Ratte bei einer Gabe von 0,5-10 mg/kg s.c., 0,2-5 mg/kg i.v. oder 5-100 mg p.o.
zeigt.
Die neuen Verbindungen können daher als psychotrope, insbesondere als antidepressive Mittel Verwendung finden.
Sie können auch als Zusatzstoffe zu Tierfutter verwendet werden, da sie eine bessere Nahrungsverwertung und eine Gewichtszunahme dieser Tiere bewirken. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller, insbesondere pharmazeutischer wirksamer Verbindungen dienen. So können z. B. 9-(3-Amino-1-propenyl)-9,10-dihydro-9,10-äthano- anthracene hergestellt werden, indem man in den neuen Verbindungen die OH-Gruppe als Wasser abspaltet.
Hervorzuheben sind vor allem Verbindungen der Formel I, worin -NHR1 eine Aminogruppe bedeutet, die durch einen Alkylrest, Hydroxy- oder Aminoalkylrest, Alkenyl-, unsubstituierten und ein-, zwei- oder mehrfach niederalkylsubstituierten und ein-, zwei- oder mehrfach niederalkylsubstituierten niederen Cycloalkyl- und/oder Cycloalkenylrest, unsubstituierten und ein-, zwei- oder mehrfach nieder alkyl-substituierten niederen Cycloalkyl- und/oder -alkenylalkylrest, zweifach substituiert ist.
Besonders wertvoll sind Verbindungen der Formel I, worin -NHRl eine Niederalkylaminogruppe, eine Hydroxy- niederalkylaminogruppe bedeutet, RA für niedere Alkyl-, Alkoxy-, Trifluormethylgruppen oder vorzugsweise Halogenatome, wie Brom- oder insbesondere Chloratome steht und Rlo ein Halogenatom, vorzugsweise ein Chloratom, oder vor allem ein Wasserstoffatom bedeutet.
Hervorzuheben sind vor allem Verbindungen der Formel

Die hydrolytische Abspaltung von X1 oder X1 und X2 erfolgt mit hydrolysierenden Mitteln, beispielsweise in Gegenwart von sauren Mitteln, wie z. B. verdünnten Mineralsäuren, wie Schwefelsäure oder Halogenwasserstoffsäuren, besonders Salzsäure, oder bei Acylresten, vorzugsweise in Gegenwart von basischen Mitteln, z. B. In erhaltenen Verbindungen kann man im Rahmen der Definition der Endstoffe Substituenten einfiihren, abwandeln oder abspalten.
In erhaltenen Verbindungen, die verätherte Hydroxyreste aufweisen, kann man diese in üblicher Weise in freie Hydroxygruppen umwandeln. Diese Umwandlung erfolgt z. B. durch Hydrolyse, vor allem mittels starker Säuren, wie z. B. Jodwasserstoffsaure oder Bromwasserstoffsäure und gegebenenfalls in Gegenwart von Leichtmetallhalogeniden, wie Aluminiumbromid oder Borbromid, oder auch mit Pyridinhydrochlorid oder Aluminiumchlorid in Pyridin.
In erhaltenen Verbindungen, die Mercaptogruppen ent halted kann man die Mercaptogruppen zu Alkylsulfonylgruppen oxydieren. Die Oxydation erfolgt in üblicher Weise, z. B. mit Perverbindungen, wie Wasserstoffperoxyd oder Persäuren, insbesondere organischen Persauren wie Peressigsäure oder einer Perbenzoesäure.
Die Erfindung betrifft auch diejenigen Ausführungsfor- men eines Verfahrens, nach denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen gebildeten rohen Gemisches einsetzt.
Die genannten Reaktionen werden in ublicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensationsund/oder katalytischen Mitten, bei erniedrigter, gewöhnli- cher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefiss durchgefiihrt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säure- additionssalze. Die Siureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung iibergefuhrt werden, z. B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Siren Salze bilden. Zur Herstellung von Säureadditions- salzen werden insbesondere solche Siren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Siren seien beispielsweise genannt: Halo genwasserstoffsiuren, Schwefelsiuren, Phosphorsiuren, Salpetersiure, Perchlorsiure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsiuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensiure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyloder p-Aminosalicylsiure, Embonsiure, Methansulfon-, Athansulfon-, Hydroxyithansulfon-, Athylensulfonsiure;
; Halogenbenzolsulfon-, Toluolsulfon -, Naphthalinsulfonsiure oder Sulfanilsiure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, kdnnen auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmissig gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können, je nach der Wahl der Ausgangsstoffe und Arbeitsweisen und je nach der Anzahl der asymmetrischen Kohlenstoffatome, als optische Antipoden, Racemate oder als Isomerengemische (z. B.
Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) kön- nen auf Grund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Isomeren (z. B. Racemate) aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation.
Erhaltene Racemate lassen sich nach bekannten Methoden zerlegen, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen, oder durch Umsetzen mit einer mit der racemischen Verbindung Salze bildenden optisch aktiven Siure und Trennung der auf diese Weise erhaltenen Salze, z. B.
auf Grund ihrer verschiedenen Ldslichkeiten, in die Diastereomeren, gefolgt von der Freisetzung der Antipoden durch Einwirkung geeigneter Mittel. Besonders gebräuch- liche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsiiure, Äpfelsäure, Mandel sure, Camphersulfonsäure oder Chinasiure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man fir die Durchführung der erfindungsgemissen Reaktionen solche Ausgangsstoffe, die zu den eingangs beispielsweise genannten oder besonders hervorgehobenen Endstoffen fihren.
Die Ausgangsstoffe können, soweit sie neu sind, nach an sich bekannten Methoden erhalten werden.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze, besonders der therapeutisch verwendbaren Salze, in Mischung mit einem z. B. fur die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder fliissigen Trigermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B.
Wasser, Gelatine, Lactose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche vole, Benzylalkohole, Gummi, Propylenglykole, Vaseline oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate kdnnen z. B.
als Tabletten, Drawees, Kapseln, Suppositorien oder in fitis- siger Form als Lösungen (z. B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Lösungsvermittler oder Salze zur Verinderung des osmotischen Druckes oder Puffer. Sie können auch andere therapeutisch wertvolle Substanzen enthalten. Die pharmazeutischen Präparate werden nach üblichen Methoden gewonnen.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln fur Tierfutter, verwendet werden. Dabei werden z. B. die üblichen Streck- und Verdtinnungsmittel bzw. Futtermittel angewendet.
Die Erfindung wird in den folgenden Beispielen niter beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
3,9 g 9 -[(3-Methyl-5-oxazolidinyl)-methyl)]-9,10-di- hydro-9,10-ithano-anthracen werden während 3 Stunden mit 60 ml 2n Salzsäure auf 90 erwirmt. Anschliessend gibt man Sn Natronlauge zu bis zur alkalischen Reaktion und extrahiert mit Methylenchlorid. Nach dem Abtrennen und Eindampfen des Lösungsmittels bleibt 9-(2-Hydroxy-3-methyl amino-propyl)-9,10-dihydro-9,10-äthano-anthracen der Formel

zurück.
Man ldst dieses in 10 ml Ethanol, gibt 1 ml einer 10%igen Lösung von Chlorwasserstoff in ethanol zu und versetzt mit Ather. Man erhilt so das kristalline Hydrochlorid des 9-(2-Hydroxy-3 -methylamino-propyl)-9,10-di- hydro-9,10-äthanoanthracens vom F. 211-213.
Das als Ausgangsmaterial benötigte 9-[(3-Methyl-5 oxazolidinyl)-methyl] -9, I0aihydro-9, lO-ithano-anthracen lisst sich in folgender Weise herstellen:
In eine Lösung von 46 g 9-(Chlorcarbonylmethyl)9,10-dihydro-9,10-äthano-anthracen in 200 ml Xylol gibt man 10 g 10%ige Palladium-Kohle, die mit Chinolin Schwefel vergiftet ist, und leitet dann bei 1200 Wasserstoff durch. Nach 7 Stunden filtriert man den Katalysator ab und dampft im Vakuum ein.
Der Rttckstand wird in Methylenchlorid gelöst und mit Sodalösung ausgeschiittelt. Nach dem Abtrennen der organischen Phase wird diese iiber Natriumsulfat getrocknet und dann durch Zugabe von Methylenchlorid auf ein Volumen von 250 ml verdiinnt. Nachdem 1 ml Triäthylamin zugegeben worden ist, flgt man 20 ml Cyanwasserstoff zu und lisst 12 Stunden bei Raumtemperatur stehen. Hierauf schüttelt man mit Wasser aus und dampft das Lösungsmittel ein. Man erhält als Rückstand das kristalline 9-(2-Hydroxy-2-cyano-äthyl)-9,10-dihydro- 9,10-ithano-anthracen, das nach Umkristallisation aus Methylenchlorid-Petrolither den F. 139-141 hat.
Dieses Nitril (18 g) wird in 150 ml Tetrahydrofuran gelöst und zu 6 g Lithiumaluminiumhydrid in 100 ml Tetrahydrofuran getropft. Nach 8 Stunden Ruhren bei 60 kiihlt man ab und gibt nacheinander 8 ml Wasser, 8 ml 15%ige Natronlauge und 24 ml Wasser zu. Man filtriert den ausgeschiedenen Niederschlag ab und dampft das Filtrat im Vakuum ein. Es verbleibt das 9-(2-Hydroxy-3-amino-propyl)- 9,10-dihydro-9q10-äthano-anthracen vom F. 176-177"
20 g dieses Amino-alkohols werden in 150 ml Ameisen siure mit 10 ml Formalin wihrend 1 Stunde auf 95 er wirmt und dann im Vakuum eingedampft.
Den Rtickstand stellt man durch Zugabe von 2n Natronlauge alkalisch, extrahiert mit Methylenchlorid und dampft die organische Phase ein. Man erhält das 9-[(3-Methyl-5-oxazolidinyl)methyl] -9,10-dihydro-9,10-äthanoanthracen vom F. 106-109 .
Beispiel 2
5 g 9-(2 -p-Tosyloxy-3 -dimethylamino-propyl)-9,10-di- hydro-9, 10-äthano-anthracen werden mit 2 g Natriumhydroxyd in 50 my ethanol und 5 ml Wasser während 2 Stunden unter Rückfluss gekocht. Anschliessend gibt man 100 ml Wasser und 50 ml 2-n Essigsiure zu und extrahiert mit anther.
Die wissrige Phase wird abgetrennt und durch Zugabe von 10prozentiger Natronlauge alkalisch gestellt. Nach Extraktion mit Methylenchlorid und Eindampfen des Lösungsmittels bleibt das 9 -(2 -Hydroxy-3 -dimethylamino-propyl) -9,10 dihydro-9,10-äthano-anthracen der Formel
