O-CO-alkyle, O-CO-aryle, O-alkyle ou O-aryle.
Les composés de formule I sont appréciés à double titre, en parfumerie comme dans l'industrie des armes. Ils possèdent en effet d'intéressantes propriétés organoleptiques et peuvent être de ce fait avantageusement utilisés comme ingrédients parfumants et aromatisants [voir à ce sujet le brevet suisse N 549951]. Ils peuvent être en outre utilisés en tant que produits intermédiaires de la synthèse de composés cyclohexadièniques isomères, également utilisés comme agents parfumants et aromatisants.
L'acylation des composés de formule II s'effectue selon les méthodes généralement utilisées à cet effet en chimie organique [voir par exemple H.O. House, Modern Synthetic Reactions,
W.A. Benjamin Inc., New York 1965, p. 258 et suivantes].
Comme composé organométallique, on utilise de préférence un dérivé lithié du propéne, et comme réactif d'acylation un ester cyclohexadiénique de formule

dans laquelle le symbole R représente un radical tel que méthyle, éthyle, n-propyle, iso-propyle, n-butyle, sec-butyle, iso-butyle ou tert.-butyle par exemple.
La réaction d'acylation s'effectue en présence d'un solvant organique inerte tel un éther comme le tétrahydrofuranne, le diméthoxyéthane, le dioxanne ou l'éther éthylique par exemple.
On utilise de préférence l'éther éthylique ou le tétrahydrofuranne, ou encore un mélange desdits solvants. On peut en outre opérer à une température comprise entre -100 C et le point d'ébullition du solvant ou mélange de solvants choisis; on préfère cependant travailler à une température comprise entre 60 C et O C.
Les composés de formule III, utilisé comme produits de départ dans le procédé de l'invention, peuvent être obtenus selon diverses méthodes exposées ci-après.
Ainsi, les composés de formule III, dans laquelle les doubles liaisons se trouvent les positions I et 3, 2 (exocyclique) et 3, ou 2 (endocyclique) et 4 du cycle, peuvent être préparés selon la méthode illustrée par le schéma ci-dessous:
La réaction de déshydrogénation peut être conduite en effectuant successivement l'halogénation du noyau cyclohexénique en position allylique et la déshydrohalogénation du produit ainsi obtenu. Comme agent halogénant on peut utiliser les réactifs d'halogénation usuels, tels les haloamides, comme les N-bromo succinimide. N-bromoacétamide. N-diméthylbromohydantoïne et leurs analogues chlorés par exemple.
Selon la méthode la plus usitée, le N-bromosuccinimide est utilisé de préférence [voir par exemple Chem. Rev. 63, 21(1963)].
L'halogénation en position allylique peut être effectuée dans un solvant organique inerte. On peut utiliser par exemple un solvant chloré tels CCl4. CHCl3, CH2CI2, le dichloroéthane, le tétrachloroéthane ou le trichloroéthyléne ou encore un mélange desdits solvants. On opère de préférence à des températures comprises entre 20 et 100C.
La réaction de déshydrohalogénation peut être effectuée au moyen de bases organiques, telles les amines tertiaires comme la morpholine. la pipéridine. la diéthylaniline ou la diméthylaniline par exemple, la température de déshydrohalogénation étant com prise entre 100 et 150"C environ.
La cyclisation du citral en cyclocitral peut se faire au moyen des techniques usuelles telles qu'elles sont décrites par exemple dans Gildemeister & Hoffmann, Die Aetherischen Oele , IIId, pp. 137-8, Akademie Verlag, Berlin (1966).
Les composés de formule IIIa possédant deux doubles liaisons conjuguées, en position 2 (endocyclique) et 4, peuvent être obte nus par condensation d'un a-isopropylidène-acéto-acétate d'alkyle avec un composé obtenu par réaction d'un sel quaternaire de phosphore de formule

Les composés de formule IIIa possédant deux doubles liaisons non conjuguées, en positions 1 et 4 du cycle, peuvent être obtenus à partir de composés de formule IIIa décrits ci-dessus, et possédant deux doubles liaisons conjuguées, dans les positions 2 (endocyclique) et 4, selon un procédé également décrit dans le brevet suisse N" 562192 [Demande de brevet N" 16393/72].
La présente invention a en outre pour objet l'utilisation des composés de formule I, obtenus selon le procédé précédemment décrit, comme produits de départ pour la préparation de composes de formule

contenant soit deux doubles liaisons non conjuguées dans les positions 1 et 4, soit deux doubles liaisons conjuguées dans les positions 1 et 3, 2 (exocyclique) et 3, ou 2 (endocyclique) et 4 du cycle, les doubles liaisons étant représentées par les pointillés.
Ladite utilisation consiste en l'isomérisation desdits composés de formule I au moyen d'un agent isomérisant acide ou basique, le cas échéant au moyen de la chaleur. On peut utiliser à cet effet la plupart des agents isomérisants connus [voir à ce sujet Synthesis 1970, 405-430], notamment des agents isomérisants acides tels les acides forts comme l'acide p-toluènesulfonique, ou encore une terre d'infusoires acide par exemple. On peut en outre utiliser un agent isomérisant basique tels un alcoolate de métal alcalin, le tert.-butylate de potassium par exemple.
Ladite isomérisation peut être effectuée en traitant le composé à isomériser et le réactif approprié à l'état de solution dans un solvant organique inerte, tels un hydrocarbure aliphatique ou cycloaliphatique, un hydrocarbure aromatique, un hydrocarbure chloré ou encore un ester ou un éther par exemple. Le tétrahydrofuranne est utilisé de préférence.
C'est ainsi que l'on obtient un composé de formule

La présente invention est illustrée de façon plus détaillée à l'aide des exemples ci-après. Dans lesdits exemples, les températures sont indiquées en degrés centigrades.
Exemple 1:
Triméthyl-2,6,6 (butène-3 oyl)-l cyclohexadiène-2,4
2 ml d'une solution de 9,4 g (70 mM) d'éther allylphénylique dans 10 ml d'éther ont été ajoutés à une suspension, refroidie à 0 et maintenue sous azote, de 2,4 g (0,35 atome-g) de lithium, finement coupé, dans 40 ml de tétrahydrofuranne anhydre. A ladite suspension on avait préalablement ajouté quelques cristaux de diphényle. Après parution d'une coloration verdâtre, on a ajouté goutte à goutte au mélange de réaction maintenu à env. -15" le reste de la solution d'éther allylphénylique. On a maintenu le refroidissement pendant 30 mn supplémentaires.
La solution résultante d'allyl-lithium a été retirée du récipient de réaction au moyen d'une seringue et ajoutée lentement à une solution, maintenue sous agitation, de triméthyl-2,6,6 éthoxycarbonyl-l cyclohexadiène-2,4 [qui peut être préparé suivant le procédé décrit dans le brevet suisse N" 562192. [Demande de brevet N" 16393/72] (3,88 g; 20 mM) dans 40 ml d'éther, à -60" et sous atmosphère d'azote. Le mélange de réaction, après avoir été réchauffé à la température ambiante, a été versé dans de l'eau glacée et extrait deux fois au pentane.
Les extraits combinés ont été lavés à l'eau, séchés sur Na2SO4 anhydre et évaporés pour donner un résidu qui, par distillation fractionnée, a fourni 3,19 g (84%) de transtriméthyl-2,6,6(butène-3 oyl)-l cyclohexadiéne-2,4; Eb. 41 3n/0,05 torr.
Un échantillon analytique a été préparé par purification au moyen de chromatographie sur une colonne remplie d'acide silicique en employant comme éluant de l'hexane contenant 2% d'acétate d'éthyle.
IR: 1695, 1650, 1590, 990 et 915 cm-l.
RMN: 1,0 (3H,s); 1,1 (3H,s); 1,7 (3H,s); 2,6 (1H,s); 3,2 (2H,s,J=7 cps); 4,7-6,1 (6H,m) 6 ppm.
UV: 267 nm (a=3900).

Exemple 2:
Méthylène-2 diméthyl-6,6 (butène-3 oyl)-l cyclohexène-3
Par l'application du procédé de l'exemple 1 à l'acylation du dérivé organolithié allylique par le méthylène-2 diméthyl-6,6 éthoxycarbonyl-l cyclohexène-3, on obtient la cétone désirée avec un rendement comparable.
IR: 1650- 1690, 1590, 970 cm-l .
SM: Mu=190.
Le méthylène-2 diméthyl-6,6 éthoxycarbonyl-l cyclohexène-3, utilisé comme produit de départ dans le procédé ci-dessus, ainsi que certains des esters cyclohexadiéniques isomères, peut être préparé comme suit:
a. 21 g de triméthyl-3,3,5 éthoxycarbonyl-4 cyclohexène-5 one-l ont été ajoutés goutte à goutte à une suspension de 4 g d'hydrure de bore et sodium dans 100 ml d'éthanol, maintenue sous forte agitation et à environ 5-10 . Le mélange de réaction a été maintenu à température ambiante pendant une nuit, évaporé jusqu'à 1/3 de son volume initial et neutralisé par de l'acide chlorhydrique dilué.
La phase organique qui s'est séparée est extraite à l'éther et les extraits éthérés ont été par la suite évaporés à sec pour donner un résidu (19 g) qui, par chromatographie préparative en phase gazeuse, a donné le cis- et le trans- triméthyl-3,5,5 éthoxycarbonyl-4 cyclohexène-5 ol-l dans un rapport pondéral d'environ 3 à 1 (rend. 65%).
Les deux isomères présentent les caractères analytiques sui vants:
A: nD20= 1,4770; d420 = 1,020.
IR: 3400, 1725 cm-l.
RMN: 0,94 (6H,s); 1,22 (3H,t,J=7,5 cps); 1,62 (3H,s); 4,12 (2H); 4,1 (1H); 5,6 (1H) 6 ppm.

b. 62 g du mélange d'isomères obtenu selon le procédé indiqué au paragraphe a ci-dessus ont été mélangés avec 0,5 g d'acide p-toluènesulfonique dans 200 ml de benzène et le mélange résultant a été traité à la température d'ébullition dans un appareil séparateur d'eau (2 heures). Le mélange de réaction a été ensuite traité avec une solution aqueuse diluée de bicarbonate de sodium, lavé à l'eau jusqu'à neutralité, séché et distillé.
Un mélange comprenant le triméthyl-2,6,6 éthoxycarbonyl-l cycle hexadiène-2,4, le triméthyl-2,6,6 éthoxycarbonyl-l cyclohexa diéne-1 ,3 et le méthylène-2 diméthyl-6,6 éthoxycarbonyl-l cycle hexène-3 dans un rapport pondéral d'environ 28:42:30 respectivement, a été ainsi obtenu; Eb. 60 10,01 torr; 34,2 g.
Le mélange de produits ainsi obtenu a été finalement distillé à l'aide d'une colonne à bande tournante et les composants dudit mélange ont été séparés au moyen de chromatographie en phase gazeuse préparative (colonne Carbowax).

La triméthyl-3,5,5 éthoxycarbonyl-4 cyclohexéne-5 ont 1 utilisée comme produit de départ dans la préparation décrite ci-dessus peut être préparée ainsi:
260 g d'acétoacétate d'éthyle ont été mélangés avec 200 g d'oxyde de mésityle et 200 g de trifluoroboroéthérate dans un récipient refroidi extérieurement par de l'eau et de la glace de façon à maintenir la température du mélange à environ 0-5". Ledit mélange est maintenu à cette température pendant 3 jours, versé ensuite sur de la glace (800 g) et neutralisé par env. 400 g de
Na2CO3. On a ensuite extrait à l'éther et, après les traitements habituels de séparation, lavage et séchage, on a évaporé les extraits organiques combinés.
Par distillation du résidu (135 g) d'évaporation on a obtenu 115 g du produit désiré, Eb. 85 90 /0,1 toor, qui a montré une pureté de 80% par analyse au moyen de chromatographie en phase gazeuse. Le produit peut être ultérieurement purifié par redistillation.
Exemple 3.'
trans-Triméthyl-2,6,6 (buténe-2 oyl)-l
cyclohexadiène-2,4
2,46 g de triméthyl-2,6,6 (butène-3 oyl)-l cyclohexadiéne-2,4 ont été ajoutés à une solution, maintenue sous atmosphère d'azote, de 140 g de potassium dans 40 ml de tert.-butanol. Après avoir laissé le mélange de réaction à température ambiante pendant 5 mn, on y a ajouté quelques gouttes d'acide acétique et l'on a ensuite éliminé les parties volatiles par évaporation à pression réduite. Au mélange concentré ainsi obtenu on ajoute de l'eau et on extrait au pentane. Une fois séparés, les extraits organiques ont été lavés, séchés sur Na2SO4, évaporés pour fournir un résidu qui, par distillation fractionnée, a donné 1,76 g (72%) de la cétone désirée. Eb. 39 /0,04 torr.
IR: 1680, 1650, 1620, 1590 et 965 cm-'.
RMN: 0,95 (3H,s); 1,7 (3H,s); 1,8 (3H,d de d,J = env. 1 et 7 cps); 2,6 (1H,s); 5,2-7,0 (5H,m) 1,0 (3H,s) Åa ppm.

Exemple 4:
trans-Méthylêne-2 diméthyl-6,6 (butène-2 oyl)-l
cyclohexène-3
On a procédé à l'isomérisation de methylene-2 diméthyl-6,6 (butène-3 oyl)-1 cyclohexène-3 [préparé à partir de méthyléne-2 diméthyl-6,6 éthoxycarbonyl-l cyclohexène selon la méthode décrite à l'exemple 2] selon le procédé de l'exemple précédent.
IR: 3080, 1600-1690, 960, 880 cm-l.
RMN: 0,90 (3H,s); 0,95 (3H,s); 1,90 (3H,d de d,J= 6,5,
J'=env. 1 cps); 2,00 (2H,m); 3,10 (1H,s); 4,80 (1H,s); 5,10 (IH,d,J = 10 cps); 6,05 (1H,d de q,J = 16, J' = env. 1 cps); 6,75 (1 H,d de q,J = 16, J' = 6,5 cps) à ppm.

Exemple 5:
trans-Triméthyl-2,6,6 (butène-2 oyl) - J cyclohexadiène- 1,3
Méthode A: un mélange de 289 mg de trans-triméthyl-2,6,6 (butène-2 oyl)-l cyclohexadiéne-2,4 [préparé selon le procédé de
l'exemple 1], 25 mg d'acide p-toluènesulfonique et 4 ml de ben
zène sec a été chauffé à reflux pendant 2 heures. Le mélange de
réaction a été ensuite dilué avec du pentane, et les extraits orga
niques combinés ont été lavés avec une solution de bicarbonate de
sodium à 5% et H2O séchés sur Na2SO4 et évaporés.
Le résidu
huileux ainsi obtenu a été distillé à pression réduite (0,1 torr; avec
une température du bain d'environ 800) pour fournir 201 mg d'un
mélange contenant 33% de trans-triméthyl-2,6,6 (buténe-2 oyl)-l
cyclohexadiène-1,3. Eb. 51 /0,08 torr.
Un échantillon analytique a été obtenu par purification au moyen de chromatographie préparative en phase gazeuse. Les caractères analytiques étaient identiques à ceux montrés par un échantillon préparé par des méthodes connues [voir par exemple brevet français N" 1591031].
IR: 1665, 1630, 1610cm-1.
RMN: 1,0 (6H,s); 1,6 (3H,s); 1,9 (3H,s de d,J=env. 1 et 7 cps); 2,1 (2H,d,J= env. 2 cps); 5,7-7,1 (4H,m) Åa ppm.
UV: 228 nm (±= 12000); 255 nm (a=5000); 310 nm (E= 2000).
SM: m/e: 190 190(19); 121(100); (62).
Méthode B: 1,96 g d'un mélange de trans-triméthyl-2,6,6 (buténe-2 oyl)-l cyclohexadiéne-1,3 et de triméthyl-2,6,6 (buténe-3 oyl)-l cyclohexadiène-1,3 [préparé à partir de triméthyl-2,6,6 éthoxycarbonyl-l cyclohexadiène-l,3 selon la méthode décrite à l'exemple 1] ont été isomérisés au moyen d'une solution de 20 mg de potassium dans 30 ml de tert.-butanol, selon le procédé de l'exemple 3. On a ainsi obtenu 1,70 g (86%) du produit désiré, en tous points identique à celui obtenu selon la méthode A.
Exemple 6:
trans-Triméthyl-2,6,6 (buténe-2,6,6 oyl)-l cyclohexadiène- 1,4
Selon le procédé de l'exemple 1, on traite environ 1 ml d'une
solution de 47 g (35 mM) d'éther allyl-phénylique dans 5 ml
d'éther avec une suspension de 1,25 g (0,18 g-atome) de lithium
dans 30 ml de tétrahydrofuranne anhydre. Après parution d'une coloration verdâtre, on a ajouté goutte à goutte au mélange de réaction maintenu à environ -15" le reste de la solution d'éther allyl-phénylique.
En opérant comme indiqué à l'exemple 1, la solution résultant d'allyl-lithium a été retirée du récipient de réaction au moyen d'une seringue et ajoutée lentement à une solution maintenue sous agitation de 2,03 g (10,4 mM) de triméthyl-2,6,6 éthoxycarbonyl-l cyclohexadiene-1,4 [qui peut être préparé suivant le procédé décrit à l'exemple 2] dans 40 ml d'éther à -60" et sous atmosphère d'azote. Le mélange de réaction, après avoir été réchauffé à la température ambiante, a été versé dans de l'eau glacée et extrait deux fois au pentane. Les extraits combinés ont été lavés à l'eau, séchés sur du Na2SO4 et évaporés. Le résidu ainsi fourni a été ajouté sous atmosphère d'azote à une solution de 50 g de tert.-butylate de potassium dans 30 ml de tert.-butanol.
Après avoir maintenu le mélange à température ambiante pendant
10 minutes, on y a ajouté quelques gouttes d'acide acétique, puis les parties volatiles ont été évaporées à pression réduite. Au mélange de réaction on a ensuite ajouté de l'eau et extrait au pentane. Les extraits organiques ont été séparés, combinés et traités suivant les méthodes usuelles de lavage à l'eau, séchage sur
Na2SO4 et évaporation. L'huile ainsi purifiée par chromatographie sur colonne 70 g d'acide silicique (éluant: hexane/l % d'acétate d'éthyle).
1,43 g (73%) du produit désiré ont été ainsi obtenus; Eb.
62 /0,15 torr.

Example 1:
2,6,6-trimethyl (3-butene-yl) -l-2,4-cyclohexadiene
2 ml of a solution of 9.4 g (70 mM) of allylphenyl ether in 10 ml of ether was added to a suspension, cooled to 0 and kept under nitrogen, of 2.4 g (0.35 atom -g) of lithium, finely cut, in 40 ml of anhydrous tetrahydrofuran. A few diphenyl crystals had previously been added to said suspension. After the appearance of a greenish color, the reaction mixture maintained at approx. -15 "the remainder of the allylphenyl ether solution. Cooling was maintained for an additional 30 min.

By applying the process of Example 1 to the acylation of the organolithium allylic derivative with 2-methylene-6,6-dimethyl-ethoxycarbonyl-1-cyclohexene-3, the desired ketone is obtained in a comparable yield.

2-methylene-6,6-dimethyl-ethoxycarbonyl-1-cyclohexene-3, used as a starting material in the above process, as well as some of the isomeric cyclohexadienic esters, can be prepared as follows:
at. 21 g of trimethyl-3,3,5 ethoxycarbonyl-4 cyclohexene-5 one-l were added dropwise to a suspension of 4 g of boron and sodium hydride in 100 ml of ethanol, kept under vigorous stirring and at about 5-10. The reaction mixture was kept at room temperature overnight, evaporated to 1/3 of its original volume and neutralized with dilute hydrochloric acid.
The organic phase which separated was extracted with ether and the ethereal extracts were subsequently evaporated to dryness to give a residue (19 g) which, on preparative gas chromatography, gave cis- and le. trans-3,5,5-ethoxycarbonyl-5-cyclohexene-ol-1 in a weight ratio of about 3 to 1 (yield 65%).

The product mixture thus obtained was finally distilled using a rotating band column and the components of said mixture were separated by means of preparative gas chromatography (Carbowax column).

The 3,5,5-trimethyl-4-ethoxycarbonyl cyclohexene-5 used as starting material in the preparation described above can be prepared as follows:
260 g of ethyl acetoacetate were mixed with 200 g of mesityl oxide and 200 g of trifluoroboroetherate in a vessel cooled externally with water and ice so as to maintain the temperature of the mixture at about 0- 5 ". Said mixture is kept at this temperature for 3 days, then poured onto ice (800 g) and neutralized with approx. 400 g of

To the concentrated mixture thus obtained, water is added and extraction is carried out with pentane.
