Diese Erfindung betrifft ein Verfahren zur Herstellung neuer 1 -Aryl-2,3,4,5-tetrahydro-lH-l ,5-benzodiazepin-2-one.
Das erste 1-Aryl-2,3,4,5-tetrahydro-1H-l ,5-benzodiazepin- -derivat, welches bekannt geworden ist, ist das 3,3-Diäthyl-l -phenyl-2-3-4-5-tetrahydro-1H-l ,5-benzodiazepin-2-on (B.J.R.
Nicolaus et al., Helv. Chim. Acta 48, 1867 (1965)1. Weitere l-Aryl-2,3,4,5-tetrahydro-lH-1,5-benzodiazepine sind der deutschen Offenlegungsschrift 19 13 536 zu entnehmen. Die letzteren zeigen antikonvulsive, sedative und muskelrelaxierende Eigenschaften.
Es wurde festgestellt, dass bestimmte neue l-Aryl-2,3,4,5- -tetrahydro-lH-1 ,5-benzodiazepin-2-one interessante pharmakologische Wirkungen besitzen.
Gegenstand der Erfindung ist nun ein Verfahren zur Herstellung von 1 -Aryl-2,3 ,4,5-tetrahydro- 1 H- I ,5-benzodiazepin -2-onen der Formel I

R1 X III worin X ein Halogenatom oder eine Arylsufonyloxygruppe darstellt, umsetzt und vorhandene Hydroxygruppen gegebe nenfalls verestert oder gegen ein Halogenatom austauscht.
Diese Reaktion kann in Gegenwart oder Abwesenheit von Lösungsmitteln erfolgen und wird im allgemeinen bei einer Temperatur von etwa 80 - 2000C durchgeführt. Zweckmässigerweise wird in Gegenwart säurebindender Mittel gearbeitet.
Als säurebindende Mittel sind vorzugsweise nicht alkylierbare tertiäre organische Basen, wie z.B. Tri-isopropanolamin, geeignet.
In den Verbindungen gegebenenfalls enthaltene freie Hydroxygruppen können verestert oder gegen ein Halogenatom ausgetauscht werden.
Die Veresterung freier Hydroxygruppen kann in üblicher Weise geschehen, z.B. durch Umsetzen mit Acylierungsmitteln wie Carbonsäureanhydriden oder Carbonsäurechloriden. Die Umsetzung mit Bernsteinsäureanhydrid kann z.B. in Gegenwart oder Abwesenheit von Lösungsmitteln erfolgen und wird im allgemeinen bei einer Temperatur von etwa 50 - 200"C durchgeführt. Vorzugsweise wird in Gegenwart tertiärer Basen wie Pyridin, das gleichzeitig als Lösungsmittel dient, gearbeitet. Der Austausch einer Hydroxygruppe gegen ein Halogenatom kann z.B. mit Thionyl- oder Phosphorhalogeniden erfolgen.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Sie zeigen starke antikonvulsive, sedative, tranquillisierende und muskelrelaxierende Wirksamkeit bei ausserordentlich geringer Toxizität. Sie ergänzen daher die Anwendungsmöglichkeiten von Substanzen dieses Wirkungstyps.
Die folgende Tabelle gibt eine Übersicht über die guten Wirkungen der neuen Substanzen.

In der Tabelle bedeuten die einzelnen Spalten folgendes (die Angaben erfolgen in mg/kg nach oraler Gabe):
I: Mittlere letale Dosis innerhalb 24 Stunden (LD50), bestimmt nach einmaliger oraler Applikation an Mäusen (J.T.
Litchfield u. F. Wilcoxon, J. Pharmacol, exp. Ther. 96, 99 -113, 1949).
II: Erfassung antikonvulsiver Wirksamkeit der Substanzen durch mittels Pentetrazol induzierter Krampfanfälle an Mäusen (1: S. Goodelann et al., J. Pharmacol. exp. Ther 108, 168-176, 1953). Als mittlere effektive Dosis der Prüfsubstanz (es00) wird die Dosis angegeben, bei der die Hälfte der Tiere vor dem Auftreten tonischer Krämpfe geschützt ist (a). Ausserdem wird die Dosis ermittelt, bei der nach Pentetrazolgabe kein Tier mehr stirbt (Letalschutzdosis) (b).
III: Antikonvulsive Effekte der Substanzen, gemessen durch Hemmung elektrisch induzierter Krampfanfälle (maximaler Elektrokrampf) an Mäusen (L.S. Goodman et al., Arch.
int. Pharmacodyn. 78, 144-162, 1949). Als ED50 wird die Dosis angegeben, bei der die Hälfte der Tiere vor tonischen Krampfanfällen der Hinterextremitäten geschützt ist (a). Die Spalte (b) gibt an, zu welchem Zeitpunkt nach Substanzgabe die ED50 bestimmt worden ist.
IV: Einfluss der Substanzen auf die Bewegungskoordination von Mäusen, untersucht am Drehstab (F. Gross et al., Schweiz. med. Wschr. 85, 305-309, 1955). Die ED50 stellt die Dosis dar, bei der die Hälfte der Tiere innerhalb 30 sec vom Drehstab herabfällt (a). Die Spalte (b) gibt wiederum an, zu welchem Zeitpunkt nach Substanzgabe die ED50 ermittelt worden ist.
V: Die tranquillisierende Wirkung der Substanzen, gemessen durch Beeinflussung der Abwehrreaktion des Goldhamsters (H. Kreiskott, Medizin u. Chemie 7, 57-78, 1963). Als ED50 wird die Dosis angegeben, bei der die Zahl der Reizantworten (Rückenlage) zum Zeitpunkt der maximalen Wirkung gegenüber Kontrolltieren auf die Hälfte vermindert ist.
Die Substanzen sollen vorzugsweise oral appliziert werden.
Beispiel I
28 g 8-Chlor- 1 -phenyl-2,3,4,5-tetrahydro-lH-1,5-benzo- diazepin-2-on, 21 g Tri-isopropanolamin und 14 g 2-Brom äthanol werden zusammen unter Rückfluss langsam auf 170"C erhitzt und zwei Stunden bei dieser Temperatur gehalten. Das Reaktionsgemisch wird noch warm in Wasser eingegossen und das ausgefallene 8-Chlor-5-(2-hydroxyäthyl)-l - -phenyl-2,3,4,5-tetrahydro-lH-i ,5-benzodiazepin-2-on aus Isopropanol umkristallisiert.
Ausbeute: 26 g (82% der Theorie), F. 160 - 161 0C.
In analoger Weise werden erhalten: 8-Chlor-5-(2-hydroxy äthyl)-l -(2-chlorphenyl)-2,3,4,5-tetrahydro-lH-1,5-benzodiaze- pin2-on.
Ausbeute: 70% der Theorie, F. 148 - 1490C (aus Essig säureäthylester).
Beispiel 2
14 g 8-Chlor-l-phenyl-2,3,4,5-tetrahydro-lH-1,5-benzo- diazepin-2-on, 13,5 g Tri- isopropanolamin und 10 g (2-Brom äthyl)-methyläther werden unter Rückfluss eine Stunde auf iOO"C erhitzt. Anschliessend wird die Temperatur langsam auf 1 700C gesteigert und weitere zwei Stunden auf dieser Höhe gehalten. Das Reaktionsgemisch wird noch warm in Wasser eingegossen und das ausgefallene Produkt in Chloroform aufgenommen. Nach Trocknen und Eindampfen der Chloroformlösung wird der kristalline Rückstand aus Äthanol umkristallisiert. Man erhält 11 g 8-Chlor-5-(2-methoxyäthyl) -l -phenyl-2,3,4,S4etrahydro-l H- 1 ,5-benzodiazepin-2-on (67% der Theorie), F. 108-109"C.
In analoger Weise werden erhalten: 8-Chlor-5-(2-methoxy äthyl)-l -(2-chlorphenyl)-2,3,4,5-tetrahydro- 1 H- 1 5-benzodiaze- pin-2-on.
Ausbeute: 58% der Theorie, F. 98 - 990C (aus Isopropanol).
Beispiel 3
14 g 8-Chlor-S-(2-hydroxyäthyl)-l -phenyl-2,3,4,5-tetrahy- dro-1H-1,5-benzodiazepin werden in 50 ml Pyridin gelöst und unter Kühlung mit Eiswasser 6,5 g Essigsäureanhydrid zugegeben. Nach zwanzigstündigem Stehen bei Raumtemperatur wird das Reaktionsgemisch in Wasser gegossen, mit verdünnter Salzsäure angesäuert und mit Chloroform extrahiert. Die gewaschene und getrocknete Chloroformlösung wird eingedampft und das kristalline 8-Chlor-5-(2-acetoxyäthyl)-1 -phe- nyl-2,3,4,5-tetrahydro-lH-1,5-benzodiazepin-2-on aus Isopropanol umkristallisiert.
Ausbeute: 16,5 g (92% der Theorie), F. 118 - 1190C.
Beispiel 4
Eine Lösung von 14 g 8-Chlor-5-(2-hydroxyäthyl)-1 -phe- nyl-2,3,4,5-tetrahydro-lH-1,5-benzodiazepin-2-on in 50 ml Chloroform wird mit 6 ml Pyridin und anschliessend mit 9 g Thionylchlorid versetzt und drei Stunden unter Rückfluss am Sieden gehalten. Die Chloroformlösung wird mit verdünnter Salzsäure und Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Das kristalline 8-Chlor-5-(2-chloräthyl) - 1 -phenyl-2,3 ,4,5-tetrahydro-1H- 1,5 -benzodiazepin-2-on wird aus Äthylmethylketon umkristallisiert.
Ausbeute: 13 g (77% der Theorie), F. 180 - 181 C.
Beispiel 5
16 g 8 -Chlor- -(2-chlorphenyl)-5 -(2-hydroxyäthyl)-2,3 4,5- -tetrahydro-lH-1,5-benzodiazepin-2-on und 12 g Bernsteinsäureanhydrid werden mit 50 ml Pyridin zwei Stunden auf siedendem Wasserbad erhitzt. Nach dem Erkalten giesst man in 500 ml Wasser ein. Die klare Lösung wird mit 5n Salzsäure angesäuert, wobei das Reaktionsprodukt in kristalliner Form ausfällt. Man saugt ab, wäscht mit Wasser und kristallisiert das 8-Chlor-l-phenyl-5-(2-succinyloxyäthyl)-2,3,4,5-tetrahydro-lH-1,5-benzodiazepin-2-on aus Isopropanol um.
Ausbeute: 18,5 g (88% der Theorie) vom F. 146 - 1470C.
In analoger Weise erhält man: 8-Chlor-1 -(2-chlorphenyl)- -5-(2-succinyloxyäthyl)-2,3 ,4,5-tetrahydro-l H-l 9-benzodiaze- pin-2-on.
Ausbeute: 810/0 der Theorie, F. 149 - 1500C (aus Essigsäure-äthylester).
8-Chlor- 1 -phenyl-5-(3 -succinyloxyppropyl)-2,3 ,4,5-tetrahy- dro-1 H- 1 ,5-benzodiuzepin-2-on.

Example I.
28 g of 8-chloro-1-phenyl-2,3,4,5-tetrahydro-1H-1,5-benzodiazepin-2-one, 21 g of tri-isopropanolamine and 14 g of 2-bromo ethanol are refluxed together slowly heated to 170 "C and held at this temperature for two hours. The reaction mixture is poured into water while still warm and the precipitated 8-chloro-5- (2-hydroxyethyl) -l - -phenyl-2,3,4,5- tetrahydro-1H-i, 5-benzodiazepin-2-one recrystallized from isopropanol.
Yield: 26 g (82% of theory), melting point 160-161 ° C.

Example 2
14 g of 8-chloro-l-phenyl-2,3,4,5-tetrahydro-lH-1,5-benzodiazepin-2-one, 13.5 g of tri-isopropanolamine and 10 g (2-bromo-ethyl) methyl ether are heated under reflux to 100 ° C. for one hour. Example 3
14 g of 8-chloro-S- (2-hydroxyethyl) -l-phenyl-2,3,4,5-tetrahydro-1H-1,5-benzodiazepine are dissolved in 50 ml of pyridine and cooled with ice water 6, 5 g acetic anhydride were added. Example 4
A solution of 14 g of 8-chloro-5- (2-hydroxyethyl) -1-phenyl-2,3,4,5-tetrahydro-1H-1,5-benzodiazepin-2-one in 50 ml of chloroform is with 6 ml of pyridine and then 9 g of thionyl chloride are added and the mixture is kept boiling under reflux for three hours. 16 g of 8-chloro- (2-chlorophenyl) -5 - (2-hydroxyethyl) -2,3 4,5- tetrahydro-1H-1,5-benzodiazepin-2-one and 12 g of succinic anhydride are mixed with 50 ml Pyridine heated on a boiling water bath for two hours. After cooling, it is poured into 500 ml of water. The clear solution is acidified with 5N hydrochloric acid, the reaction product precipitating in crystalline form. It is filtered off with suction, washed with water and the 8-chloro-1-phenyl-5- (2-succinyloxyethyl) -2,3,4,5-tetrahydro-1H-1,5-benzodiazepin-2-one recrystallized from isopropanol .
Yield: 18.5 g (88% of theory), melting point 146-1470C.
