Die Erfindung betrifft ein Verfahren zur Herstellung von l-Phenyl-2,3,4,5-tetrahydro- 1 H-3 -benzazepin-Derivaten.
Die erfindungsgemäss hergestellten Verbindungen sind 1 - Phenyl-2,3 ,4,5-tetrahydro- 1 H-benzazepin-Derivate der allgemeinen Formel

worin W, X und Y unabhängig voneinander Wasserstoff, Hydroxyl, Alkyl, Alkoxy oder Halogen oder zwei derselben in benachbarten Stellungen zusammen eine Alkylendioxygruppe; A unsubstituiertes oder substituiertes Phenyl, das eine oder mehrere Substituenten, die Halogen, Alkyl, Alkoxy oder Hydroxy sind, enthält; R Wasserstoff, Alkyl, Hydroxyalkyl mit zwei oder mehr Kohlenstoffatomen, Alkenyl oder Benzyl; Z, eine Carbonyl- oder Methylengruppe und Z2 eine Carbonylgruppe oder die Gruppe -CHR2- bedeutet, wobei nur eine der Gruppen Z1 und Z2 gleichzeitig eine Carbonylgruppe sein kann; und R1 und R2 Wasserstoff oder Alkyl bedeuten; und den pharmazeutisch annehmbaren Säureadditionssalzen der freien Basen der Formel (I), d.h.
Verbindungen, in denen weder Zl noch Z2 eine Carbonylgruppe bedeutet.
Die freien Basen der Formel (I), d.h., Verbindungen, in denen weder Zl noch Z2 eine Carbonylgruppe bedeutet, sowie ihrer Säureadditions- und quaternären Ammoniumsalze sind therapeutisch brauchbar; sie wirken insbesonders antibakteriell, antidepressiv, analgetisch und blutdrucksenkend. Die Verbindungen der Formel (I), in denen entweder Zl oder Z eine Carbonylgruppe bedeutet, sind wertvolle Zwischenprodukte für die Herstellung jener Verbindungen der Formel (1), in denen Z1 und Z2 Methylgruppen darstellen.
Mit Vorteil enthält der Kohlenwasserstoffteil der Alkyl-, Hydroxyalkyl-, Alkenyl- oder Alkoxygruppen nicht mehr als sechs Kohlenstoffatome und der Alkendioxy-Substituent nicht mehr als drei Kohlenstoffatome; im folgenden werden solche Substituenten Niedrig- Alkyl, Hydroxy-niedrig-alkyl, Niedrig -Alkenyl, Niedrig-Alkoxy und Niedrig-Alkylendioxy genannt.
Der hier benutzte Ausdruck Alkyl usw. soll hier wie auch in den Patentansprüchen Cycloalkyl. Hydroxycycloalkyl, Cycloalkenyl usw. mit einschliessen.
Die Niedrig-Alkyl-Substituenten schliessen einwertige Kohlenwasserstoff-Reste wie Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl und Isobutyl mit ein. Methyl ist der bevorzugte Niedrig-Alkyl-Substituent; die Hydroxy-niedrig-alkyle mit 2 bis 6 Kohlenstoffatomen schliessen Hydroxypropyl, Hydroxybutyl, besonders aber Hydroxyäthyl mit ein; die Niedrig-Alkenyl-Substituenten schliessen gerad; und verzweigtkettige einwertige Kohlenwasserstoffradikale mit bis zu 6 Kohlenstoffatomen und einer Doppelbindung mit ein wie Allyl, Butenyl und Pentenyl; Niedrig-Alkoxy schliesst jedes Radikal mit bis zu 6 Kohlenstoffatomen mit ein, das über das Sauerstoffatom am verbleibenden Benzazepin-Molekülteil gebunden ist, wie Methoxy, Äthoxy, Propoxy und Butoxy; Alkylendioxy ist vorzugsweise Methylendioxy, Äthylendioxy oder Propylendioxy;
Phenyl schliesst Phenyl selbst und substituiertes Phenyl mit ein, einschliesslich Phenylradikale, welche durch Halogen (besonders Chlor), Niedrig-Alkoxy (besonders Methoxy) und Niedrig-Alkyl (besonders Methyl) substituiert sind, und zwar vorzugsweise in Para-Stellung, doch können sie auch in Orthound Metastellung des Phenylrings substituiert sein.
Die Säureadditionssalze, die manchmal die Löslichkeit erhöhen und sich besser zur Rezeptur eignen als die freien Basen, schliessen zum Beispiel solche ein, die mit folgenden Säuren oder ihren reaktiven Derivaten hergestellt werden: Salzsäure, Phosphorsäure, Schwefelsäure, Maleinsäure, Zitronensäure, Salizylsäure, Bernsteinsäure, Weinsäure. Die quaternären Ammoniumsalze schliessen zum Beispiel solche ein, die aus organischen Halogeniden wie Methyljodid, Äthyljodid, Propylchlorid. Benzylchlorid und Allylbromid hergestellt werden.
Die folgenden Verbindungen entsprechen beispielsweise de Formel 1. gehören zum Erfindungsbereich und können nach einem erfindungsgemässen Verfahren hergestellt werden: 1 - Phenyl-2,3,4,5-tetrahydro- 1 H-3-benzazepin, l-Methyl-l-phe nyl-2,3,4,5-tetrahydro- 1 H-3-benzazepin, 1 -Phenyl-4-methyl -2,3,4,5-tetrahydro- 1 H-3-benzazepin, 1 -Phenyl- 1 ,4-dimethyl- -2,3,4,5-tetrahydro- 1 H-3-benzazepin, 1 -Phenyl-7-chlor-2.3.4.5 - -tetrahydro- 1 H-3-benzazepin,
1-Phenyl-7-methoxy-2.3,4.5 -tetrahydro-l H-3-benzazepin, 1 -Phenyl-3 -methyl-2,3.4.5-tetra- hydro- 1 H-benzazepin, 1 -Phenyl-3-allyl-2.3,4,5-tetrahydro- 1H -3-benzazepin. 1-Phenyl-3-n-hydroxyäthyl-2,3,4,5-tetrahydro- - 1 H-3-benzazepin, 1- Phenyl-7,8-dihydroxy-2.3.4,5-tetrahydm - - lH-3-benzazepin-hydrobromid und 1 -Phenyl-3.3-dimethyl- -7,8-dimethoxy-2,3,4,5-tetrahydro- 1 H-3-benzazepiniumjodid.
Die Verbindungen dieser Erfindung können als razemische Gemische ihrer rechts- und linksdrehenden Isomeren hergestellt werden: sie können mit Hilfe der gebräuchlichen. wohlbekannten Methoden. wie der fraktionierten Kristallisation ihrer Salze mit optisch aktiven Säuren, getrennt werden. Wo Stellung 4 ein Asymmetriezentrum ist. können zudem bei geeigneter Wahl des optisch aktiven Ausgangsmaterials optisch aktive Produkte direkt erhalten werden.
Das erfindungsgemässe Verfahren zur Herstellung neuer l-Phenyl-2,3.4,5-tetrahydro-lH-3-benzazepin-Derivate ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

Das bei der obigen intramolekularen Kondensation erhaltene lH-3-Benzazepin-Derivat kann zur Überführung in die jeweils gewünschte 1-Phenyl-2,3 ,4,5-tetrahydro-l H3-benzaze- pin-Verbindung einem oder mehreren zusätzlichen Reaktionsschritten unterworfen werden, zum Beispiel den folgenden: (a) Reduktion einer durch Z1 oder Z2 dargestellten Carbonylgruppe zu einer Methylengruppe; (b) N-Alkylierung, d.h. Einführung von Alkyl, Hydroxyalkyl mit zwei oder mehr Kohlenstoffatomen, Alkenyl oder Benzyl in Stellung 3; (c) Uberführung in ein quaternäres Ammoniumsalz; (d) Trennung eines Racemates in seine optischen Isomeren; (e) Überführung in ein pharmazeutisch verwendbares Säureadditionssalz durch Reaktion mit einer pharmazeutisch verwendbaren Säure oder einem reaktiven Derivat davon.
Die Erfindung betrifft auch die Verwendung einer nach der obigen intramolekularen Kondensation hergestellten Verbindung der Formel 1, in der Z1 eine Methylengruppe, Z2 die Gruppe -CHR2- und R eine Alkylgruppe, eine Hydroxyalkylgruppe mit zwei oder mehr Kohlenstoffatomen, eine Alkenyl- oder Benzylgruppe bedeuten, für die Herstellung einer quaternären Verbindung, die zusätzlich eine Alkylgruppe, eine Hydroxyalkylgruppe, die zwei oder mehr Kohlenstoffatome enthält, eine Alkenyl- oder Benzylgruppe an dem in Stellung 3 befindlichen Stickstoffatom aufweist, dadurch gekennzeichnet, dass man eine solche Verbindung der Formel 1 mit einem entsprechenden N-Quaternierungsmittel umsetzt.
Ein besonders geeignetes Verfahren zur Herstellung der bevorzugten Verbindungen der Formel (I) geht von Dit(-phe- nyl)-äthyl] -amin- oder Phenylessigsäure-N-phenäthyl-amid -Derivaten der Formel (ItA) aus,

worin W, X, Y, A, Rl, R2, Z1 und D wie oben definiert sind und Ra eine Alkyl-, Alkenyl- oder Benzylgruppe oder eine Hydroxyalkylgruppe mit zwei oder mehr Kohlenstoffatomen bedeutet. Die Verbindung (IIA) wird einer intramolekularen Kondensation unterworfen, wobei die Gruppe D unter Schliessung des Azepinrings mit einem Wasserstoffatom reagiert, das an ein benachbartes Kohlenstoffatom im Benzolring gebunden ist.
Der Ringschluss kann nach verschiedenen Methoden erzielt werden. Wenn z.B. D eine Hydroxylgruppe ist, so kann der Ringschluss unter bekannten Dehydratisierungsbedingungen oder unter den Bedingungen einer intramolekularen Friedel-Crafts-Reaktion unter Benutzung von Katalysatoren wie Aluminiumchlorid erfolgen. Auch wenn D eine andere reaktive organische oder anorganische Estergruppe, z.B. Halogen, besonders Chlor oder Brom, oder eine Sulfonsäureestergruppe wie O-Tosyl oder O-Mesyl ist, erfolgt Ringschluss mit oder ohne Katalysatoren wie etwa Aluminiumchlorid.
Die genannten Di-[(-phenyl)-äthylJ-amindenvate können am besten durch Erhitzen aquimolarer Mengen eines Styroloxids mit einer Phenäthylaminverbindung erhalten werden.
Das Erhitzen wird vorzugsweise auf einem Dampfbad durchgeführt, doch kann die Reaktion innerhalb eines Temperaturbereichs von gerade über Raumtemperatur bis etwa 150 C stattfinden. Die so erhaltenen Carbinole (V) können dann mittels der üblichen Methoden, etwa mittels Destillation und Kristallisation, isoliert werden. Wenn ein Niedrig-Alkyl, wie z.B. Methyl, als Substituent in Stellung 1 des Benzazepins gewünscht wird (d.h. wenn R1 in Formel 1 z.B. Methyl bedeutet) wird für die Herstellung des entsprechenden Ausgangsstoffes (V) zweckmässig ein z-Methylstyroloxid verwendet; wenn ein Niedrig-Alkyl, wie z.B. Methyl, als Substituent in Stellung 4 des Benzazepins gewünscht wird (d.h. wenn R2 in Formel 1 z.B.
Methyl bedeutet), kann ein verzweigtkettiges Phenalkylamin wie etwa Amphetamin verwendet werden; wenn W und/ oder X und/oder Y als Substituenten am Benzolring des Benzazepinkerns gewünscht werden, können die in passender Weise W,X,Y-substituierten Phenalkylamine verwendet werden. Zur Herstellung von Ausgangsstoffen (V), die sich zur direkten Kondensation zu 3-substituierten Benzazepinen eignen, kann man von passenden N-substituierten Phenalkylaminen ausgehen, doch wird es in der Praxis vorgezogen, erst ein solches Benzazepin der Formel I herzustellen, wo R Wasserstoff bedeutet, und dann am Stickstoff zu alkylieren.
Die obigen Di-[(g-phenyl)-äthyl]-amin-Derivate werden mittels intramolekularen Ringschlusses in die gewünschten Benzazepine (I) übergeführt, der durch Dehydratisierung der Di-[GB-phenyl)-äthyi]-amin-Zwischenprodukte (V) erzielt wird.
Die Dehydratisierung kann durch Behandeln des Alkohols (V) mit Reagenzien wie Polyphosphorsäure, Schwefelsäure, Zinkchlorid und andere ähnlich reagierende Dehydratisierungsmittel bewirkt werden. Vorzugsweise werden die Carbinole (V) innerhalb eines Temperaturbereichs von etwa 80 bis 1600C zusammen mit Polyphosphorsäure erhitzt, wobei Dehydratisierung mit gleichzeitigem Ringschluss stattfindet, wogegen es bei Verwendung von Schwefelsäure vorgezogen wird, die Dehydratisierung bei etwa -5 C bis 20"C durchzuführen. Die obigen Reaktionen lassen sich folgendermassen beispielhaft veranschaulichen:

Die bevorzugten 1 -Phenyl-2,3 ,4,5-Setrahydro- 1 H-3 - -benzazepine der Formel (I) können auch aus l-Phenyl-3-oxo -2,3 ,4,5-tetrahydro- 1 H- 3- benzazepinen durch Reduktion an der 2-Oxogruppe hergestellt werden, die ihrerseits z.B. durch Zusammenkondensieren des passenden Phenalkylamins (III) mit einem Ester einer Mandelsäure (VI), wie Mandelsäure äthylester, wobei man das zugehörige Mandelsäureamid (VII) erhält, und anschliessende Dehydratisierung hergestellt werden können. Die Kondensation zu (VII) wird vorzugsweise durch zwei bis sechs Stunden langes Erhitzen der zusammengefügten Ausgangsstoffe auf einem Ölbad bei ungefähr 150-190"C erreicht, obwohl auch Temperaturen im Bereich von 150-2000C angewendet werden können.
Die Mandelsäureamide (VII) werden dann dehydratisiert, vorzugsweise durch Reaktion mit einem sauren Dehydratisierungsmittel, wie einem der vorhergehend für die Dehydratisierung des Carbinols (V) beschriebenen, wobei man unter Ringschluss das entsprechende 2-Oxo -benzazepin (VIII) erhält. Die Reduktion an der 2-Oxogruppe kann nach bekannten Reduktions-Verfahren. wie etwa Reduktion mit Lithiumaluminiumhydrid in einem ätherischen Lösungsmittel wie Dioxan, erfolgen. Diese Reaktionen sind im folgenden beispielhaft dargestellt:

Dabei bedeutet Q Alkyl, Hydroxyalkyl mit zwei oder mehr Kohlenstoffatomen, Alkenyl (mit Vorteil ist der Kohlenwasserstoffteil in jedem Fall niedrig ) oder Benzyl, und W, X, Y, R1, Re und A sind wie oben definiert.
Die obige Reaktion ist die Kondensation eines organischen Halogenids (vorzugsweise Bromid oder Chlorid); sie wird vorzugsweise in einem inerten Lösungsmittel ausgeführt, wie etwa Aceton, insbesondere bei Rückflusstemperatur und in Gegenwart eines basischen Kondensationsmittels, wie wasserfreien Kaliumcarbo nats. Als organische Halogenide werden z.B. Allylbromid, Benzylbromid oder Methylbromid benutzt.
Die N-Alkylierung jener Benzazepine der Formel (I) die über eine 2-Oxo-lH-3-benzazepi'n-Zwischenstufe gebildet werden, kann vor oaer nach der Reduktion der 2-Oxo-Gruppe ausgeführt werden.
Die nach dem einen oder dem anderen Reaktionsschema erhaltenen 1H-3.-Benzazepin-Verbindungen können, falls gewünscht, in ein pharmazeutisch verwendbares Säureadditionsoder quaternäres Ammoniumsalz übergeführt werden.
Beispiel I
I Phenyl-2,3,4,5-tetrahydro- I H-3-benzazepin A: N-[-Hydroxy -B-pheny -äthyll-N- [(P-phenyl)-äthyl] -amin
Man erhitzt eine gut gerührte Mischung von 82 g Styroloxid und 100 g Phenäthylamin 12 Stunden lang auf einem Dampfbad und destilliert das erhaltene Produkt im Vakuum, wobei man ein viskoses Produkt erhält, das aus Petroläther umkristallisiert wird.
B: 1-Phenyl-2,3,4,5- tetrahydro-1H-3- 1 -Phenyl-2,3,4,5-tetrahydro-1H-3-benzazepin
Man fügt zu 100 ml konzentrierter Schwefelsäure (die auf 0-50C gehalten wird) in kleinen Anteilen 15 g des nach Teil A dieses Beispiels erhaltenen N-[(p-Hydroxy-Ç3-phenyl)-äthyl] -N-[(p-phenyl)-äthyl]-amins und rührt eine Stunde. Die erhaltene Mischung wird auf Eis und Wasser geschüttet, mit Natriumhydroxid stark alkalisch gemacht und das gewünschte Produkt mit Äther extrahiert. Man trocknet den Ätherextrakt über wasserfreiem Kaliumcarbonat, filtriert und destilliert das gewünschte Produkt. Das mit trockenem Chlorwasserstoff in Äther hergestellte hygroskopische Hydrochloridsalz wird aus Isopropanol kristallisiert.
Beispiel 2
Entsprechend dem in Teil A des vorangehenden Beispiels angegebenen Verfahren reagieren Homoveratrylamin und Styroloxid zum zugehörigen Aminoalkohol (Schmelzpunkt 95980C). Dieser Alkohol wird dann nach dem in Teil B des Beispiels 1 angegebenen Verfahren zum l-Phenyl-7,8-dimethoxy- -2,3 ,4,5-tetrahydro- 1 H-3 -benzazepin (Siedepunkt 1 98-200 C/2 Torr) zyklisiert, welches durch Reaktion mit Maleinsäure in sein Maleinsäuresalz (Schmelzpunkt 198-200"C) übergeführt wird.
Beispiel 3 4-Methyl-1 phenyl-2,3,4,5-tetrahydro-l H-3-benzaze pin A: N-[(ss-Hydroxy-ss-phenyl)-äthyl-N-[(ss-phenyl)-isopropyl]) -amin
Man erhitzt eine gut gerührte Mischung von 82 g Styroloxid und 100 g d-Amphetamin 12 Stunden lang auf einem Dampfbad. Beim Destillieren des erhaltenen Produkts (im Vakuum) erhält man ein viskoses Produkt (Siedepunkt 160 1800C/1 Torr), das aus Petroläther kristallisiert wird, Schmelz punkt 53-55"C, [n25 = + 14,6 (1 so in Äthanol).
B: 4-Methyl-l - phenyl-2,3,4,5-tetrahydrobenzaze pin
Man fügt langsam zu 100 ml konzentrierter Schwefelsäure (auf 0 C gehalten) in kleinen Anteilen 15 g eines fein zerriebenen Pulvers des optisch aktiven Produktes von Teil A.
Man rührt noch eine Stunde, giesst dann die Mischung auf Eis-Wasser, macht die erhaltene Mischung mit Natriumhydroxid stark alkalisch und extrahiert mit Äther. Den Ätherextrakt trocknet man über wasserfreiem Kaliumcarbonat, filtriert und destilliert das gewünschte Produkt ab (Siedepunkt 149-151 0C/1 Torr). Sein mit trockenem Chlorwasserstoff in Äther erhaltenes hygroskopisches Hydrochlorid wird aus Isopropanol kristallisiert; Schmelzpunkt 206-2070C, [a1,25 = -42,0 (10/o in Dimethylformamid).
Beispiel 4 1 -Methyl-1 -phenyl-2,3,4,5-tetrahydro-JH-3-benzazepin A: N- [(-Phenyl-8-methyl-,B-hydroxy)-äthyl]-N [(ss-phenyl)- -äthyl]-amin
Man erhitzt eine gut gerührte Mischung von 60 g a-Methylstyroloxid und 66 g Phenathylamin 6 Stunden auf einem Dampfbad und destilliert die erhaltene Reaktionsmischung.
Das Produkt (Siedepunkt 160-168"C/1 Torr) bildet ein Hydrochlorid, welches nach Umkristallisieren aus Acetonitril einen Schmelzpunkt von 142-145"C besitzt.
B: 1-Methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepin
Man kondensiert 28 g N-[(p-Phenyl-p-methyl-!p-hydroxy)- -äthyl]-N-[(fi-phenyl)-äthyl]-amin in 250 ml konzentrierter Schwefelsäure und extrahiert das gewünschte Produkt nach dem in Teil B des Beispiels 3 angegebenen Verfahren mit Äther. Den Ätherextrakt trocknet man über wasserfreiem Kaliumcarbonat und destilliert das Produkt bei 150-160 C/ 1 Torr.
Das Produkt dieses Beispiels wird aus Hexan umkristallisiert (Schmelzpunkt 76-79"C). Das mit trockenem Chlorwasserstoff in Äther hergestellte Hydrochlorid schmilzt bei 228-2290C
Aus den vorangegangenen Beispielen ist ersichtlich, dass bei Verwendung der entsprechend substituierten Ausgangsstoffe eine ganze Reihe anderer Benzazepine auf ähnliche Weise hergestellt werden kann. So können durch Änderung der Ausgangsstoffe und durch Befolgung der wesentlichen Züge der in den Beispielen 1-4 dargelegten Verfahren, z.B.
1-Phenyl-1,4-dimethyl-2,3,4,5-tetrahydro-1 H-3-benzazepin, 1-Phenyl-1,4-dimethyl-7-chlor-2,3,4,5-tetrahydro-1H-3- -benzazepin, 1-Phenyl-1,4,7-trimethyl-2,3,4,5-tetrahydro-1H-3- -benzazepin, 1 -Phenyl-4-methyl-8-hydroxy-2,3 ,4,5-tetrahydro- - 1 H-3-benzazepin, 1 -Phenyl- 1,4-dimethyl-8-methoxy-2,3,4,5- -tetrahydro-lH-3-benzazepin und 1-Phenyl-7,8-methylendioxy -2,3,4,5-tetrahydro-lH-3-benzazepin hergestellt werden. Die für die Herstellung der voranstehenden Benzazepine nach den Verfahren der Beispiele 1 bis 4 notwendigen Phenäthylamin Ausgangsstoffe sind entweder bereits bekannt oder können nach bekannten Verfahren hergestellt werden.
Beispiel 5
Dieses Beispiel veranschaulicht die erfindungsgemässe Herstellung einer Benzazepinverbindung unter Benutzung eines Phenäthylamids der Mandelsäure als Ausgangsstoff.
1-Phenyl-7-methoxy-2,3,4,5-tetrahydro-1H-3-benzazepin A: N-(ss,-m-Methoxy-phenäthyl)-mandelsäureamid:
Man erhitzt eine gut gerührte Lösung von 25 g p-m- -Methoxy-phenäthylamin und 30 g Mandelsäureäthylester 3 Stunden lang in einem Ölbad bei 180-190"C. Die Reaktionsmischung wird gekühlt und das gewünschte Produkt aus Äther kristallisiert (Schmelzpunkt 75-76 C).
B: 1 -Phenyl-2-oxo-7-methoxy-2,3,4,5-tetrahydro.JH-3- -benzazepin
Man fügt 20 g fein zerriebenes Pulver von -Methoxy-phenäthyl)-mandelsäureamid langsam in kleinen Anteilen zu 700 g Polyphosphorsäure und erwärmt die erhaltene Mischung langsam auf 100"C. Die Mischung wird noch eine Stunde lang auf 1000C erhitzt, dann lässt man sie Zimmertemperatur annehmen, giesst die abgekühlte Mischung auf Eis-Wasser und extrahiert mit Chloroform. Das Chloroform wird im Vakuum abgezogen und das gebildete l-Phenyl-2-oxo -7-methoxy-2,3,4,5-tetrahydro-lH-3-benzazepin aus Essigsäureäthylester umkristallisiert (Schmelzpunkt 169-1710C).
C: 1-Phenyl-7-methoxy-2,3,4,5-ietrahydro-JH-3-benzazepin
Zu einer gut gerührten und unter Rückfluss kochenden Suspension von 5 g Lithiumaluminiumhydrid in 200 ml Dioxan fügt man tropfenweise eine Lösung von 10 g 1-Phenyl -2-oxo-7-methoxy-2,3,4,5-tetrahydro-lH-3-benzazepin in 250 ml Dioxan und kocht die erhaltene Mischung drei Stunden unter Rückfluss. Man kühlt die erhaltene Reaktionsmischung und fügt, während sie auf 20"C gehalten wird, tropfenweise 0,5 ml (4 mal) Wasser, 0,5 ml (4 mal) 15%i'ge Natronlauge und 13,5 ml Wasser hinzu und rührt die Reaktionsmischung eine Stunde.
Der Niederschlag anorganischen Materials wird abfiltriert und das erhaltene Filtrat durch Destillieren vom Lösungsmittel befreit. Man versetzt den Rückstand mit 100 ml 5%iger Salzsäure und 200 ml Äther, rührt die Mischung, bis sich der Rückstand löst, trennt den wässrigen Auszug ab, macht ihn mit Natriumhydroxid alkalisch und extrahiert mit Äther. Der Ätherauszug wird mit wässrigem Kaliumcar- bonat getrocknet. Man zieht das Lösungsmittel ab und wandelt das gewünschte Produkt dieses Beispiels in sein Hydrogenmaleat um, indem man es zu einer Lösung von Maleinsäure in Essigsäureäthylester fügt, wobei man 1-Phenyl-7-methoxy -2,3,4,5-tetrahydro- lH-3.benzazepin-hydrogenmaleat erhält (Schmelzpunkt 196-197"C).
Beispiel 6
I -Phenyl-3-allyl-2,3,4,5-tetrahydro-l H-3-benzaze pin
Man erhitzt eine Mischung von 6 g l-Phenyl-2,3,4,5-tetra- hydro-1H-3-benzazepin, 2,4 g Allylbromid, 25 g wasserfreiem Kaliumcarbonat und 250 ml wasserfreiem Aceton 14 Stunden unter starkem Rühren auf Rückflusstemperatur, kühlt die erhaltene Mischung, filtriert und destilliert das Aceton ab. Man löst den Rückstand mit Äther und Wasser, trennt die wässrige Schicht ab, trocknet und filtriert die ätherische Schicht und zieht den Äther ab.
Das gewünschte Produkt wird aus Hexan umkristallisiert (Schmelzpunkt 65-68 C). 1-Phenyl-3-allyl- -2,3 ,4,S4etrahydro- lH-3-benzazepinhydrochlorid (Schmelzpunkt 203-2050C) wird durch Behandeln des Produktes dieses Beispiels mit trockenem Chlorwasserstoff in Äther hergestellt.
Bei Ersatz des 1-Phenyl-2,3,4,5-tetrahydro-1H-3-benz- azepins des voranstehenden Beispiels durch zlB. l-Phenyl-4 -methyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-1,4-di- methyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-1,4-dime- thyl-7-chlor-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl- -1,4,7-trimethyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl- -4-methyl-8-äthoxy-2,3,4,5-tetrahydro-lH-3-benzazepin,

Methylbromid, Äthylbromid, Propylbromid, Butylbromid, Methallylbromid, Vinyläthylbromid oder Benzylbromid, 1 -Phenyl-3-methyl-2,3,4,5-tetrahydro-1H-3-benzazepin, l-Phe nyl-3-äthyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-3- -propyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-3-butyl- -2,3,4,5-tetrahydro-1H-3-benzazepin, 1 -Phenyl-3 -methallyl- -2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-3-vinyläthyl- -2,3,4,5-tetrahydro-1H-3-benzazepin bzw. 1 -Phenyl- 3-benzyl- -2,3,4,5-tetrahydro-1H-3 -benzazepin hergestellt.
Ähnlich können die voranstehenden organischen Halogenide mit 1-Phenyl -4-methyl-2,3,4,5-tetrahydro.1H-3-benzazepin, 1-Phenyl-1,4- -dimethyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-1,4- -dimethyl-7-chlor-2,3,4,5-tetrahydro-lH-3-benzazepin, I-Phe- nyl-1,4,7-trimethyl-2,3,4,5-tetrahydro-lH-3-benzazepin, l-Phe- nyl- 1 ,4-dimethyl-7-methoxy-2,3 ,4,5-tetrahydro- 1H-3-benzaze- pin, l-Phenyl-1-methyl-2,3,4,5-tetrahydro-lH-3-benzazepin, 1-Phenyl-8-methoxy-2,3,4,5-tetrahydro-lH-3-benzazepin oder 1 -Phenyl-7,S-dimethoxy-2,3 ,4,5-tetrahydro- 111-3-benzazepin (als Beispiele für einzelne Benzazepinverbindungen) nach dem im obigen Beispiel angegebenen Verfahren zur Reaktion gebracht werden,
wobei die entsprechenden N-methyl-, N-Åathyl-, N-propyl-, N-butyl-, N-methallyl-, N-vinyläthyl- bzw. N-benzyl-substituierten Benzazepine hergestellt werden.
Beispiel 7 I -Phenyl-3- p-hydroxyäthyl-2345-tetrahydro-l lH-3- benzazepin
Zu einer Lösung von 6 g 1-Phenyl-2,3,4,5-tetrahydro-lH-3- -benzazepin in 50 ml Äthanol fügt man 1 g Äthylenoxid und hält die verschlossene Reaktionsmischung mehrere Tage auf Zimmertemperatur. Die erhaltene Mischung wird destilliert und der ölige Rückstand aus Isopropyläther kristallisiert (Schmelzpunkt 95-970C).
Beispiel 8
1 -Phenyl-3,3-dimethyl-7,8-dimethoxy-2'3,4,5-tetrahydro- -IH-3-benzazepinium-iodid
Eine Mischung von 9 g l-Phenyl-7,8-dimethoxy-2,3,4,5- -tetrahydro-1H-3-benzazepin, 15 ml 37%igen Formaldehyds und 23 ml 90%iger Ameisensäure wird 18 Stunden auf Rückflusstemperatur erhitzt. Man fügt eine Lösung von 5 ml konzentrierter Salzsäure in 10 ml Wasser hinzu und dampft die Lösung im Vakuum auf einem Dampfbad zur Trockne ein.
Den Rückstand versetzt man mit 25 ml Wasser und dampft zur Trockne ein. Man behandelt den Rückstand mit Äther und überschüssiger Natriumhydroxidlösung, trennt die Ätherlösung ab, trocknet, filtriert und dampft die Ätherlösung ein.
Das als Rückstand verbleibende l-Phenyl-3-methyl-7,8-di- methoxy-2,3,4,5-tetrahydro-lH-3-benzazepin wird aus Hexan kristallisiert (Schmelzpunkt 82-84"C). Man versetzt eine Lösung von 5 g davon in 5 ml Äthanol mit 5 ml Methyljodid und lässt die erhaltene Mischung 15 Stunden bei Zimmertemperatur stehen, wobei das gewünschte 1-Phenyl-3,3-dimethyl-7,8 -dimethoxy-2,3,4,5-tetrahydro-lH-3-benzazepinium-jodid aus der Mischung kristallisiert (Schmelzpunkt 246-2490C).
Die Quaternisierung dieser und anderer erfindungsgemäss erhältlichen Verbindungen kann statt mit Alkyljodid selbstverständlich auch mit anderen bekannten Quaternisierungs- mitteln erfolgen.
Beispiel 9
I -Phenyl-7,8-dihydroxy-2,3,4,5-tetrakydro-l H-3-benzaze pin- -hydrobromid
Eine Mischung von 45 g 1-Phenyl-7,8-dimethoxy-2,3,4,5 -tetrahydro-1H-3-benzazepin und 110 ml 48%iger Bromwasserstoffsäure wird 2,5 Stunden unter Stickstoffatmosphäre auf Rückflusstemperatur erhitzt. Beim Kühlen kristallisiert das Hydrobromidsalz der gewünschten Verbindung. Nach Filtrie- ren und Waschen der Kristalle mit Alkohol erhält man 1-Phe nyl-7,8-dihydroxy-2,3 ,4,5-tetrahydro- 111-3-benzazepin-hydro- bromid (Schmelzpunkt 283-285"C).
Beispiel 10
Dieses Beispiel veranschaulicht die Auftrennung von razemischen Mischungen von nach dem Verfahren dieser Erfindung erhältlichen Verbindungen am Beispiel des 1 Phenyl -2,3 ,4,5-tetrahydro- 111-3 -benzazepins.
42 g 1 -Phenyl-2,3 ,4,5-tetrahydro- 1 H-3 -benzazepin werden zusammen mit 37 g (+)-Phenylbernsteinsäure in 90%igem Äthanol durch Erwärmen gelöst. Die Lösung wird mehrere Tage bei Zimmertemperatur in Ruhe stehen gelassen. Die entstandenen Kristalle werden abfiltriert, mit einer kleinen Menge kalten Alkohols gewaschen und schliesslich aus 150 ml 90%igen Äthanols umkristallisiert.
Als Ausbeute erhält man 15 g Kristalle, Schmelzpunkt 180-1820C, [,!X]D25 = + 55,20 (1% in Dimethylformamid).
Aus diesem Salz wird die freie Base in üblicher Weise durch Behandeln mit wässrigem Natriumhydroxid in Freiheit gesetzt und durch Extrahieren mit Äther und Kristallisation aus Hexan isoliert: Schmelzpunkt 78-80"C, [al25 = -29,9 (1 % in Dimethylformamid).
Die Alkoholmutterlaugen vom Waschen und Umkristallisieren werden vereinigt, dann im Vakuum konzentriert, und das freie 1-Phenyl-2,3,4,5-tetrahydro-lH-3-benzazepin wird nach Standardverfahren zurückerhalten, wobei wässrige Natronlauge und Äther benutzt werden. Die teilweise getrennte Base wird weiterhin durch Verwendung von (-)-Phenylbernsteinsäure nach dem obigen Verfahren aufgetrennt, und es wird so das (+)-Isomer erhalten, welches ebenfalls bei 78 80"C schmilzt, aber ein [a1,25 von + 29,9 hat.
Die 1 -Phenyl-2,3 ,4,5-tetrahydro- lH-3-benzazepin-Derivate dieser Erfindung können in der Form pharmazeutischer Zusammensetzungen verabreicht werden, die eine oder mehrere dieser Verbindungen zusammen mit einer pharmazeutisch anwendbaren und verträglichen Trägersubstanz enthalten.
Solche Zusammensetzungen können auch andere aktive Verbindungen enthalten, mit denen sie verträglich sind, wie z.B. Analgetica, Anti-Depressiva, Antibiotica und proteolytische Enzyme.
Die Zusammensetzungen können in fester Form, z.B. als Tabletten, Pillen oder Kapseln, in flüssiger Form, wie z.B.
als Sirupe, Elixiere oder Emulsionen für orale Verabreichung oder als sterile Injektionslösungen oder auch in Form von Cremes oder Lotionen vorliegen.
Die pharmazeutischen Zusammensetzungen können nach allgemein für die Herstellung pharmazeutischer Zusammensetzungen bekannten Verfahren hergestellt werden, die im wesentlichen im Vereinigen des betreffenden 1-Phenyl-2,3,4,5 -tetrahydro-lH-3-benzazepin-Derivates mit einer damit verträglichen pharmazeutisch anwendbaren Trägersubstanz ste- hen. Geeignete Trägersubstanzen sind z.B. Wasser, Gelatine, Lactose, eine Stärke, Magnesiumstearat, Talk, ein Pflanzenöl,
Benzylalkohol; ein Gummi, ein Polyalkylenglykol und Vaseline.

The preferred 1-phenyl-2,3, 4,5-setrahydro-1 H-3 - -benzazepines of the formula (I) can also be prepared from 1-phenyl-3-oxo-2,3,4,5-tetrahydro-1 H- 3-benzazepines are prepared by reduction at the 2-oxo group, which in turn, for example by condensing the appropriate phenalkylamine (III) with an ester of a mandelic acid (VI), such as ethyl mandelate, to obtain the associated mandelic acid amide (VII), and subsequent dehydration can be prepared. The condensation to (VII) is preferably achieved by heating the assembled starting materials on an oil bath at about 150-190 "C for two to six hours, although temperatures in the range of 150-2000C can also be used.
The mandelic acid amides (VII) are then dehydrated, preferably by reaction with an acidic dehydrating agent, such as one of the previously described for the dehydration of the carbinol (V), the corresponding 2-oxobenzazepine (VIII) being obtained with ring closure. The reduction at the 2-oxo group can be carried out by known reduction processes. such as reduction with lithium aluminum hydride in an essential solvent such as dioxane. Q denotes alkyl, hydroxyalkyl with two or more carbon atoms, alkenyl (the hydrocarbon fraction is advantageously low in each case) or benzyl, and W, X, Y, R1, Re and A are as defined above.
The above reaction is the condensation of an organic halide (preferably bromide or chloride); it is preferably carried out in an inert solvent such as acetone, especially at reflux temperature and in the presence of a basic condensing agent such as anhydrous potassium carbonate. As organic halides e.g. Allyl bromide, benzyl bromide or methyl bromide are used.
The N-alkylation of those benzazepines of the formula (I) which are formed via a 2-oxo-1H-3-benzazepin intermediate can be carried out before or after the reduction of the 2-oxo group.
The 1H-3-benzazepine compounds obtained according to one or the other reaction scheme can, if desired, be converted into a pharmaceutically acceptable acid addition or quaternary ammonium salt.
Example I.
I Phenyl-2,3,4,5-tetrahydro- I H-3-benzazepine A: N - [- Hydroxy -B-pheny -ethyl-N- [(P-phenyl) -ethyl] -amine
A well-stirred mixture of 82 g of styrene oxide and 100 g of phenethylamine is heated on a steam bath for 12 hours and the product obtained is distilled in vacuo to give a viscous product which is recrystallized from petroleum ether.
B: 1-phenyl-2,3,4,5-tetrahydro-1H-3-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine
To 100 ml of concentrated sulfuric acid (which is kept at 0-50C), 15 g of the N - [(p-hydroxy-Ç3-phenyl) -ethyl] -N - [(p-) obtained according to Part A of this example are added in small portions phenyl) ethyl] amine and stir for one hour. The mixture obtained is poured onto ice and water, made strongly alkaline with sodium hydroxide and the desired product is extracted with ether. The ether extract is dried over anhydrous potassium carbonate, filtered and the desired product is distilled. The hygroscopic hydrochloride salt produced with dry hydrogen chloride in ether is crystallized from isopropanol.
Example 2
According to the procedure given in Part A of the previous example, homoveratrylamine and styrene oxide react to form the associated amino alcohol (melting point 95980C). This alcohol is then according to the procedure given in Part B of Example 1 to give 1-phenyl-7,8-dimethoxy-2,3,4,5-tetrahydro-1 H-3 -benzazepine (boiling point 98-200 C / 2 Torr), which is converted into its maleic acid salt (melting point 198-200 "C) by reaction with maleic acid.
Example 3 4-Methyl-1-phenyl-2,3,4,5-tetrahydro-1 H-3-benzaze pin A: N - [(ß-Hydroxy-ß-phenyl) -ethyl-N - [(ß-phenyl ) isopropyl]) amine
A well-stirred mixture of 82 g of styrene oxide and 100 g of d-amphetamine is heated on a steam bath for 12 hours. When the product obtained is distilled (in vacuo), a viscous product is obtained (boiling point 160-1800C / 1 Torr), which is crystallized from petroleum ether, melting point 53-55 "C, [n25 = + 14.6 (1 in ethanol) .
B: 4-methyl-1-phenyl-2,3,4,5-tetrahydrobenzaze pin
One adds slowly to 100 ml of concentrated sulfuric acid (kept at 0 C) in small portions 15 g of a finely ground powder of the optically active product of part A.
The mixture is stirred for a further hour, then the mixture is poured onto ice-water, the mixture obtained is made strongly alkaline with sodium hydroxide and extracted with ether. The ether extract is dried over anhydrous potassium carbonate, filtered and the desired product is distilled off (boiling point 149-151 ° C./1 Torr). Its hygroscopic hydrochloride, obtained with dry hydrogen chloride in ether, is crystallized from isopropanol; Melting point 206-2070C, [a1.25 = -42.0 (10 / o in dimethylformamide).
Example 4 1 -Methyl-1-phenyl-2,3,4,5-tetrahydro-JH-3-benzazepine A: N- [(-Phenyl-8-methyl-, B-hydroxy) -ethyl] -N [( ss-phenyl) - ethyl] amine
A well-stirred mixture of 60 g of α-methylstyrene oxide and 66 g of phenethylamine is heated on a steam bath for 6 hours and the reaction mixture obtained is distilled.
The product (boiling point 160-168 "C / 1 Torr) forms a hydrochloride which, after recrystallization from acetonitrile, has a melting point of 142-145" C.
B: 1-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine
28 g of N - [(p-phenyl-p-methyl-! P-hydroxy) -ethyl] -N - [(fi-phenyl) -ethyl] -amine are condensed in 250 ml of concentrated sulfuric acid and the desired product is extracted the method given in Part B of Example 3 with ether. The ether extract is dried over anhydrous potassium carbonate and the product is distilled at 150-160 C / 1 Torr.
The product of this example is recrystallized from hexane (melting point 76-79 "C). The hydrochloride prepared with dry hydrogen chloride in ether melts at 228-2290C
From the previous examples it can be seen that when using the correspondingly substituted starting materials, a whole range of other benzazepines can be produced in a similar manner. Thus, by changing the starting materials and following the essential features of the procedures set out in Examples 1-4, e.g.
1-phenyl-1,4-dimethyl-2,3,4,5-tetrahydro-1H-3-benzazepine, 1-phenyl-1,4-dimethyl-7-chloro-2,3,4,5-tetrahydro -1H-3-benzazepine, 1-phenyl-1,4,7-trimethyl-2,3,4,5-tetrahydro-1H-3 -benzazepine, 1-phenyl-4-methyl-8-hydroxy-2 , 3, 4,5-tetrahydro- - 1 H-3-benzazepine, 1-phenyl-1,4-dimethyl-8-methoxy-2,3,4,5- -tetrahydro-1H-3-benzazepine and 1- Phenyl-7,8-methylenedioxy -2,3,4,5-tetrahydro-1H-3-benzazepine. The phenethylamine starting materials required for the preparation of the above benzazepines by the processes of Examples 1 to 4 are either already known or can be prepared by known processes.
Example 5
This example illustrates the inventive preparation of a benzazepine compound using a phenethylamide of mandelic acid as the starting material.
1-phenyl-7-methoxy-2,3,4,5-tetrahydro-1H-3-benzazepine A: N- (ss, -m-methoxyphenethyl) -mandelic acid amide:
A well-stirred solution of 25 g of pm- methoxyphenethylamine and 30 g of ethyl mandelate is heated for 3 hours in an oil bath at 180-190 ° C. The reaction mixture is cooled and the desired product is crystallized from ether (melting point 75-76 ° C.) .
B: 1-phenyl-2-oxo-7-methoxy-2,3,4,5-tetrahydro.JH-3-benzazepine
20 g finely ground powder of methoxy-phenethyl) -mandelic acid amide is slowly added in small portions to 700 g of polyphosphoric acid and the resulting mixture is slowly heated to 100 ° C. The mixture is heated to 1000 ° C. for a further hour, then it is left at room temperature assume, the cooled mixture is poured onto ice-water and extracted with chloroform. The chloroform is stripped off in vacuo and the l-phenyl-2-oxo-7-methoxy-2,3,4,5-tetrahydro-lH-3- Benzazepine recrystallized from ethyl acetate (melting point 169-1710C).
C: 1-phenyl-7-methoxy-2,3,4,5-tetrahydro-JH-3-benzazepine
A solution of 10 g of 1-phenyl -2-oxo-7-methoxy-2,3,4,5-tetrahydro-1H-3 is added dropwise to a well-stirred and refluxing suspension of 5 g of lithium aluminum hydride in 200 ml of dioxane -benzazepine in 250 ml of dioxane and reflux the resulting mixture for three hours. The reaction mixture obtained is cooled and, while it is kept at 20 ° C., 0.5 ml (4 times) of water, 0.5 ml (4 times) of 15% sodium hydroxide solution and 13.5 ml of water are added dropwise the reaction mixture is stirred for one hour.
The precipitate of inorganic material is filtered off and the filtrate obtained is freed from the solvent by distillation. 100 ml of 5% hydrochloric acid and 200 ml of ether are added to the residue, the mixture is stirred until the residue dissolves, the aqueous extract is separated off, made alkaline with sodium hydroxide and extracted with ether. The ether extract is dried with aqueous potassium carbonate. The solvent is drawn off and the desired product of this example is converted into its hydrogen maleate by adding it to a solution of maleic acid in ethyl acetate, 1-phenyl-7-methoxy-2,3,4,5-tetrahydro-1H -3.benzazepine hydrogen maleate is obtained (melting point 196-197 "C).
Example 6
I-phenyl-3-allyl-2,3,4,5-tetrahydro-1 H-3-benzaze pin
A mixture of 6 g of 1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine, 2.4 g of allyl bromide, 25 g of anhydrous potassium carbonate and 250 ml of anhydrous acetone is heated for 14 hours with vigorous stirring Reflux temperature, the resulting mixture is cooled, filtered and the acetone is distilled off. The residue is dissolved with ether and water, the aqueous layer is separated off, the ethereal layer is dried and filtered and the ether is removed.

Example 7 I-phenyl-3-p-hydroxyethyl-2345-tetrahydro-1 lH-3-benzazepine
1 g of ethylene oxide is added to a solution of 6 g of 1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine in 50 ml of ethanol and the sealed reaction mixture is kept at room temperature for several days. The mixture obtained is distilled and the oily residue is crystallized from isopropyl ether (melting point 95-970C).
Example 8
1-phenyl-3,3-dimethyl-7,8-dimethoxy-2'3,4,5-tetrahydro-IH-3-benzazepinium iodide
A mixture of 9 g of 1-phenyl-7,8-dimethoxy-2,3,4,5-tetrahydro-1H-3-benzazepine, 15 ml of 37% formaldehyde and 23 ml of 90% formic acid is refluxed for 18 hours heated. A solution of 5 ml of concentrated hydrochloric acid in 10 ml of water is added and the solution is evaporated to dryness in vacuo on a steam bath.
The residue is mixed with 25 ml of water and evaporated to dryness. The residue is treated with ether and excess sodium hydroxide solution, the ether solution is separated off, dried, filtered and the ether solution is evaporated.
The 1-phenyl-3-methyl-7,8-dimethoxy-2,3,4,5-tetrahydro-1H-3-benzazepine remaining as residue is crystallized from hexane (melting point 82-84 ° C.) a solution of 5 g of it in 5 ml of ethanol with 5 ml of methyl iodide and the mixture obtained is left to stand for 15 hours at room temperature, the desired 1-phenyl-3,3-dimethyl-7,8 -dimethoxy-2,3,4, 5-tetrahydro-1H-3-benzazepinium iodide crystallized from the mixture (melting point 246-2490C).
These and other compounds obtainable according to the invention can of course also be quaternized using other known quaternizing agents instead of alkyl iodide.
Example 9
I -Phenyl-7,8-dihydroxy-2,3,4,5-tetrakydro-I H-3-benzaze pin- hydrobromide
A mixture of 45 g of 1-phenyl-7,8-dimethoxy-2,3,4,5-tetrahydro-1H-3-benzazepine and 110 ml of 48% strength hydrobromic acid is heated to reflux for 2.5 hours under a nitrogen atmosphere. Upon cooling, the hydrobromide salt of the desired compound crystallizes. After filtering and washing the crystals with alcohol, 1-phenyl-7,8-dihydroxy-2,3,4,5-tetrahydro-111-3-benzazepine hydrobromide (melting point 283-285 "C) is obtained. .
Example 10
This example illustrates the separation of racemic mixtures of compounds obtainable by the process of this invention using the example of 1-phenyl -2,3,4,5-tetrahydro-111-3 -benzazepine.
42 g of 1-phenyl-2,3,4,5-tetrahydro-1 H-3 -benzazepine are dissolved together with 37 g of (+) - phenylsuccinic acid in 90% ethanol by heating. The solution is left to stand for several days at room temperature. The crystals formed are filtered off, washed with a small amount of cold alcohol and finally recrystallized from 150 ml of 90% strength ethanol.
The yield obtained is 15 g of crystals, melting point 180-1820C, [,! X] D25 = + 55.20 (1% in dimethylformamide).
The free base is set free from this salt in the usual way by treatment with aqueous sodium hydroxide and isolated by extraction with ether and crystallization from hexane: melting point 78-80 "C, [a125 = -29.9 (1% in dimethylformamide).
The alcohol mother liquors from washing and recrystallization are combined, then concentrated in vacuo, and the free 1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine is recovered by standard procedures using aqueous sodium hydroxide and ether. The partially separated base is further separated by using (-) - phenylsuccinic acid according to the above procedure, and the (+) - isomer is thus obtained, which also melts at 78-80 "C, but an [a1.25 of +29 , 9 has.
The 1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine derivatives of this invention can be administered in the form of pharmaceutical compositions which contain one or more of these compounds together with a pharmaceutically acceptable and acceptable carrier.
Such compositions may also contain other active compounds with which they are compatible, e.g. Analgesics, anti-depressants, antibiotics, and proteolytic enzymes.
