Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 9-(3 -Amino-1 -propenyl)-9, 1 O-dihydro- 9,10-äthano-anthracene der allgemeinen Formel

der 4-8 Ringglieder enthält und durch Phenyl, Hydroxy, Oxo und/oder Amino substituiert sein kann, oder einen 1-Aza-oxa-, 1-Aza-thia- oder 1-Aza-azacycloalkylrest bedeuten, in dem die Heteroatome durch mindestens zwei Kohlenstoffatome getrennt sind und der durch Phenyl, Hydroxy, Oxo und/oder Amino substituiert sein kann, und wobei R1 ausserdem noch Wasserstoff bedeuten kann, n und m ganze Zahlen von 1-4 bedeuten, wobei n und m vorzugsweise nicht grösser als 3 ist, die Reste RA unabhängig voneinander Wasserstoff, niedere Alkyl-, Alkoxy-, Alkenyloxy-. Alkylmercapto und/oder Alkanoylgruppen, Trifluormethylgruppen oder Halogenatome bedeuten, R1o einen niederen Alkyl- oder Alkenylrest ein Halogenatom oder ein Wasserstoffatom bedeuten und Ae einen gegebenenfalls in 1- und/oder 2-Stellung durch einen niederen Alkylrest substituierten 1,2-Äthylenrest bedeutet.
Als niedere Alkylreste werden Reste bezeichnet, die bis zu 7 und vor allem 1-4 Kohlenstoffatome enthalten, wie Methyl-, Äthyl-, Propyl-, Isopropyl-, gerade und verzweigte.
in beliebiger Stellung verbundene Butyl-, Petnyl-, Hexyl- und Heptylreste.
Niedere Hydroxyalkylreste sind z. B. der 2-Hydroxy äthyl und der 3-Hydroxypropylrest.
Alkenylreste sind Reste mit nicht mehr als 7 Kohlenstoffatomen, wie z. B. der Allyl- oder Methallylrest.
Als niedere Cycloalkyl- und Cycloalkenylreste werden Reste wie Cyclopentyl-, Cyclohexyl-, Cyclopropyl-, Cyclopentenyl- und Cyclohexenylreste bezeichnet.
Halogen bedeutet Fluor, Brom und Jod, und ganz besonders Chlor.
Niedere Alkoxy- oder Alkenyloxygruppen sind vor allem die Methoxy-, Äthoxy- oder Allyloxygruppe.
Niedere Alkylmercaptogruppen sind insbesondere die Methyl- oder Äthylmercaptogruppe.
Als niedere Alkanoylreste sind vor allem der Acetyl-, Propionyl- und Butylrylrest zu nennen.
Die Substitution in den Stellungen 1-8 kann mehrfach sein, ist aber bevorzugt zweifach oder vor allem einfach.
Bevorzugte Stellungen für Substituenten RA sind die 3- und ganz besonders die 2-Stellung.
Die -NR1R2-Gruppe ist vor allem eine Mono- oder Di-niederalkylaminogruppe, wie die Mono- oder Dipropylamino-, oder vorzugsweise Mono- oder Diäthylaminogruppe, vor allem aber die Dimethylamino- oder ganz besonders die Monomethylaminogruppe, oder die N-Methyl-N-äthylaminogruppe, eine Cycloalkylaminogruppe, wie die Cyclopropyl-, Cyclobutyl-, Cyclopentyl- oder Cyclohexylaminogruppe, oder eine gegebenenfalls C-niederalkylierte und/oder im Ringss-einfach ungesättigte Pyrrolidino- oder Piperidinogruppe oder eine gegebenenfalls C-niederalkylierte Piperazino-, N'-Niederalkyl- wie N'-Methyl-, oder N'-(Hydroxyniederalkyl)- wie N'-(ss-Hydroxyäthyl)-piperazino-, Thiomorpholino- oder Morpholinogruppe.
Der Ausdruck C-niederalkyliert bedeutet hierbei ebenso wie oben und im folgenden, dass der betreffende Rest an C-Atomen durch niedere Alkylreste. wie die genannten, und insbesondere durch C1¯3-Alkylreste substituiert ist.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine psychotrope, z. B.
antidepressive Wirkung. So weisen sie z. B. eine ausgeprägte Reserpin-antagonistische Wirkung auf, wie sich im Tierversuch. z. B. im drug interaction test bei intraperitonealen Gaben zeigen lässt. Die neuen Verbindungen können daher als psychotrope, insbesondere antidepressive Mittel Verwendung finden. Sie können auch als Zusatzstoffe zu Tierfutter verwendet werden, da sie eine bessere Nahrungsverwertung und eine Gewichtszunahme dieser Tiere bewirken. Die neuen Verbindungen können aber auch als Zwischenprodukte für die Herstellung anderer wertvoller, vor allem phamrakologisch wirksamer Verbindungen dienen. So können z. B. die bekannten 9-(Aminopropyl)-9,10-dihydro-9,10-äthano anthracene (US-Patent 3 399 201) hergestellt werden, indem man in den neuen Verbindungen die aliphatische Doppelbindung katalytisch hydriert.
Hervorzuheben sind vor allem Verbindungen der Formel I. worin -NR1R2 eine Aminogruppe bedeutet, die durch Alkylreste, Hydroxy- oder Aminoalkylreste, Alkenyl-, unsubstituierte oder ein-, nvei- oder mehrfach niederalkyl-substituierte niedere Cycloalkyl- und/oder Cycloalkenylreste, unsubstituierte oder ein-, zwei- oder mehrfach niederalkylsubstituierte niedere Cycloalkyl- und/oder -alkenyl-alkylreste substituiert ist, oder einen 1-Azacycloalkyl- oder 1 -Azacycloalkenylrest bedeutet, der 4-8 Ringglieder enthält und durch Hydroxy-, Oxo- und/oder Aminogruppen substituiert sein kann, oder einen 1-Aza-oxa-, l-Aza-thia- oder 1-Aza-azacycloalkylrest bedeutet, in dem die Heteroatome durch mindestens zwei Kohlenstoffatome getrennt sind-und der durch Hydroxy-, Oxo- und/oder Aminogruppen substituiert sein kann.
Besonders wertvoll sind Verbindungen der Formel I, worin NR1R2 eine Mono- oder Diniederalkylaminogruppe, eine Hydroxyniederalkylaminogruppe, eine allenfalls Cniederalkylierte 1 -Azacycloalkylgruppe mit 5-7 Ringgliedern oder Cycloalkylaminogruppe mit 3-7 Ringgliedern oder eine gegebenenfalls C-niederalkylierte Morpholino-, Thiomorpholino-, N'-Niederalkylpiperazino- oder N'-(Hydroxyniederalkyl)-piperazinogruppe bedeutet, RA für niedere Alkyl-, Alkoxy-, Trifluormethylgruppen oder vorzugsweise Halogenatome, wie Brom- oder insbesondere Chloratome steht und R1o ein Halogenatom, vorzugsweise ein Chloratom, oder vor allem ein Wasserstoffatom bedeutet.
Hervorzuheben sind vor allem Verbindungen der Formel

worin R3 bevorzugt in 2-Stellung steht und eine niedere Alkyl- oder Alkoxygruppe, eine Trifluormethylgruppe, ein Brom- oder insbesondere Chloratom, vor allem aber ein Wasserstoffatom bedeutet und NRlR2 eine durch einen C3-I-Cycloalkylrest monosubstituierte oder durch C1- Alkylgruppe mono- oder disubstituierte Aminogruppe oder eine gegebenenfalls C-niederalkylierte Pyrrolidino-, Piperidino-, Morpholino, N'-Methyl-piperazino-, N'-Äthyl-piperazino oder N'-(ss-Hydroxyäthyl)-piperazinogruppe darstellt.

Eine reaktionsfähig veresterte Hydroxygruppe ist vor allem eine mit einer starken organischen oder anorganischen Säure, wie besonders einer Halogenwasserstoffsäure, wie Chlor-, Brom- oder Jodwasserstoffsäure, oder einer Arylsulfonsäure, wie einer durch niedere Alkyl- oder Alkoxyreste, z. B. die oben genannten, oder durch Halogenatome, wie Chlor- oder Bromatome ein-, zwei- oder mehrfach substituierten Benzolsulfonsäure, z. B. der p-Toluolsulfonsäure oder p-Brombenzolsulfonsäure, oder einer Niederalkansulfonsäure, z. B. Methansulfonsäure, veresterte Hydroxygruppe.
ss-Hydroxyalkylamine der Formel I können, z. B. auch durch Umsetzung von Verbindungen der Formel (IV) mit X1 gleich -NH2 oder -NHR1 mit einem gegebenenfalls entsprechend substituierten Äthylenoxyd erhalten werden.
Die Umsetzungen können in üblicher Weise erfolgen, vorzugsweise in Anwesenheit eines Lösungsmittels und gegebenenfalls in Gegenwart eines Kondensationsmittels, z. B.
eines basischen Kondensationsmittels, vorzugsweise bei erhöhter Temperatur und gegebenenfalls im geschlossenen Gefäss unter Druck. Ein basisches Kondensationsmittel ist z. B. ein Alkalihydroxyd oder -carbonat, z. B. Natriumhydroxyd oder Kaliumcarbonat, oder ein tertiäres Amin, z. B. Triäthylamin oder Pyridin. Statt eines sekundären Amins kann auch ein es abgebendes Mittel, z. B. ein symmetrisch disubstituierter Harnstoff, verwendet werden. In diesem Fall arbeitet man zweckmässig unter Erhitzen und gegebenenfalls Hinzufügen eines inerten Verdünnungsmittels, z. B.
Diphenyläther oder Sand.
In erhaltenen Verbindungen kann man im Rahmen der Definition der Endstoffe Substituenten einführen, abwandeln oder abspalten.
So kann man beispielsweise erhaltene sekundäre in tertiäre Amine umwandeln, d. h. in erhaltene N-mono-substituierte Verbindungen der Formel I bzw. N'-unsubstituierte Piperazinogruppen Substituenten einführen, z. B. die oben genannten.
Die Umsetzung geschieht in üblicher Weise, z. B. durch Umsetzen der N-monosubstituierten Verbindung der Formel I mit einem reaktionsfähigen Ester eines dem einzuführenden Substituenten entsprechenden Alkohols. Ein reaktionsfähiger Ester ist vor allem ein von einer starken organischen oder anorganischen Säure, wie insbesondere einer Halogenwasserstoffsäure oder einer Arylsulfonsäure oder einer Niederalkansulfonsäure, abgeleiteter Ester. Die Umsetzung kann aber auch mit einem entsprechenden Aldehyd oder Keton unter reduzierenden Bedingungen, z. B. in Gegenwart von Ameisensäure, erfolgen.
In erhaltenen Verbindungen, die an den aromatischen Ringen des Anthrazengerüsts Substituenten tragen, kann man diese umwandeln oder abspalten.
In erhaltenen Verbindungen, die freie Hydroxygruppen enthalten, können diese veräthert werden. Die Verätherung erfolgt in üblicher Weise, z. B. durch Umsetzen mit einem reaktionsfähigen Ester eines Alkanols, vorzugsweise in Gegenwart einer starken Base.
Die Erfindung betrifft auch diejenigen Ausführungsformen eines Verfahrens, nach denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen gebildeten rohen Gemisches einsetzt.
Die genannten Reaktionen werden in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensationsund/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze.
Je nach der Wahl der Ausgangsstoffe und Arbeitsweisen können die neuen Verbindungen, sofern sie mindestens ein asymmetrisches Kohlenstoffatom besitzen, als Racemate oder als optische Antipoden vorliegen.
Erhaltene Racemate lassen sich nach bekannten Methoden zerlegen, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen, oder durch Umsetzen mit einer mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z. B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, gefolgt von der Freisetzung der Antipoden durch Einwirkung geeigneter Mittel. Besonders gebräuchliche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs beispielsweise genannten oder besonders hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe sind teils neu, teils bekannt. Die neuen Ausgangsstoffe können nach erfinderischen Verfahren oder nach Analogieverfahren erhalten werden.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze, besonders der therapeutisch verwendbaren Salze, in Mischung mit einem z. B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Lactose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Propylenglykole, Vaseline oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees.
Kapseln, Suppositorien oder in flüssiger Form als Lösungen (z. B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Lösungsvermittler oder Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch andere thevapeutisch wertvolle Substanzen enthalten. Die pharmazeutischen Präparate werden nach üblichen Methoden gewonnen.
Die neuen Verbindungen können auch in der Tiermedi zin, z. B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z. B. die üblichen Streck- und Verdünnungsmittel bzw. Futtermittel angewendet.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
2,8 g 9-(3-Chlor-1-propenyl)-9,10-dihydro-9,10-äthano- anthracen werden in 10 g frisch destilliertem N-Methylpiperazin 15 Stunden bei 100" erhitzt. Hierauf wird eingedampft und im Hochvakuum bei 100" 2 Stunden entgast.
Der ölige Rückstand wird in 250 ml 10%aber Methansulfonsäure gelöst, die wässrige Phase zweimal mit Toluol extrahiert, mit 1 gTierkohle 10 Minuten bei 20 gerührt und durch eine Schicht-Diatomeenerde (Hyflo) abgenutscht.
Die methansulfonsaure Lösung wird mit 6n Natronlauge stark alkalisch gestellt und 3mal mit je 300 ml Chloroform extrahiert. Die vereinigten Chloroformlösungen werden mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird in wenig Toluol gelöst und über 160 g Aluminiumoxyd (Aktivität II, neutral) chromatographiert. Als Eluiermittel verwendet man 3 Liter Toluol. Die erste Fraktion ergibt das 9-[3-(N'-Methylpiperazino) 1 -propenyl] -9,1 0-dihydro -9,1 0-äthano-anthracen, F. 139 bis 141" (Hexan).
Das Dihydrochlorid wird in üblicher Weise hergestellt.
Das Produkt schmilzt uneinheitlich. Der höchste Schmelzpunkt lag bei 263-265 .
Das als Ausgangsmaterial verwendete 9-(3-Chlor L -propenyl)-9,10-dihydro-9,10-äthano-anthracen kann auf folgende Weise hergestellt werden:
2,6 g 9-(3-Oxo-1-propenyl)-9,10-dihydro-9,10-äthano- mthracen wird in 100 ml Methanol gelöst und mit einer Lösung von 0,5 g Natriumborhydrid in 2 ml Wasser umgesetzt. Die Lösung wird 2 Stunden bei Raumtemperatur gerührt. Hierauf wird die Lösung auf 200 g Eis gegossen und Jie wässrige Phase dreimal mit je 200 ml Chloroform extrahiert. Die vereinigten Chloroformlösungen werden mit Wasier gewaschen, über Natriumsulfat getrocknet und eingeiampft.
Der Rückstand ergibt nach Umkristallisation aus Äther-Hexan das 9-(3 -Hydroxy-1 -propenyl)-9,10-dihydro- ),10-äthano-anthracen als Kristalle vom F. 141-142".
2,8 g 9-(3-Hydroxy-1 -propenyl)-9,10-dihydro-9,10- rthano-anthracen werden in 100 ml Chloroform gelöst und !,2 g Phosphorpentachlorid in Portionen eingetragen. Die Reaktion ist exothermisch, die Temperatur steigt bis auf 45 bis 50". Das Reaktionsgemisch wird weiter noch 2 Stunden bei 20 gerührt und auf 1 kg Eis gegossen. Die wässrige Phase wird noch zweimal mit 150 ml Chloroform extrahiert, die vereinigten Chloroformlösungen werden über Natriumsulfat getrocknet und eingedampft. Der Rückstand besteht aus 9-(3 -Chlor-1 -propenyl)-9, 10-dihydro-9, 10-äthano- anthracen und kann ohne weitere Reinigung mit N-Methylpiperazin umgesetzt werden.
Beispiel 2
5,6 g 9-(3 -Chlor-1 -propenyl)-9,10-dihydro-9,10-äthanoanthracen werden in 25 ml Dimethylformamid gelöst und mit 13,0 g N-(ss-Hydroxyäthyl)-piperazin umgesetzt. Das Reaktionsgemisch wird 15 Stunden bei 100" erwärmt. Hierauf wird das Gemisch im Hochvakuum bei 1200 eingedampft und entgast. Der Rückstand wird in wenig Toluol gelöst und über 300 g Aluminiumoxyd (Aktivität II, neutral) chromatographiert. Als Eluiermittel verwendet man zuerst 3 Liter Toluol und anschliessend 3 Liter eines Gemisches Toluol: Chloroform (2:1). Die mit diesem Gemisch eluierten Fraktionen ergeben reines 9-{3 -[N' -(ss -Hydroxyäthyl)-piper- azino] -1 -propenyl) -9,10-dihydro -9,10 -äthano-anthracen.
Das in üblicher Weise hergestellte Dihydrochlorid schmilzt uneinheitlich. Der höchste Schmelzpunkt lag bei 245-247 .
Beispiel 3 9 -(3 -Methylamino-(Z)-1 -propenyl)-9, 1 0-dihydro-
9,10 -äthano-anthracen-hydrochlorid
2,25 g (6,62 mMol) 9-(3-Hydroxy-(E)-1-propenyl) 9,10 -dihydro -9,10 -äthano -anthracen-methansulfonsäureester wird in 100 ml lOprozentiger Methylaminlösung in Benzol während 24 Stunden bei Raumtemperatur stehengelassen.
Hierauf verdünnt man mit 100 ml Äther und schüttelt erst mit 25 ml 2n Sodalösung, dann 3mal mit je 50 ml Wasser aus. Die organische Phase wird über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Als Rückstand verbleibt 9 -(3 -Methylamino-(2)-1 -propenyl)-9,1 0-dihydro- 9,10-äthano-anthracen vom Smp.79-80 .
Zur Überführung ins Hydrochlorid wird die freie Base in 25 ml Methylenchlorid gelöst und mit ätherischer Chlorwasserstofflösung kongosauer gestellt. Das 9-(3-Methylamino (Z)-1 -propenyl)-9,10-dihydro-9,10 -äthano-anthracen- hydrochlorid wird mit Äther ausgefällt, abgenutscht und aus Methylenchlorid Äther umkristallisiert; Smp. 205-207".
Der als Ausgangsstoff verwendete 9-(3 -Hydroxy-(Z)- 1 -propenyl)-9,10-dihydro-9,10-äthano-anthracen-methan- sulfonsäureester wird wie folgt hergestellt:
10,0 g (38,4 mMol) 9-(3-Acroleinyl)-9,10-dihydro- 9,10-äthano-anthracen wird in 250 ml Methanol suspendiert und unter Rühren bei 15-20" portionenweise mit 0,8 g (21,1 mMol) Natriumborhydrid versetzt. Nach der Zugabe resultiert eine klare, hellgelbe Lösung, die man eine Stunde bei Raumtemperatur stehen lässt. Anschliessend engt man im Wasserstrahlvakuum ein.
Der Rückstand wird in 250 ml Äther gelöst, und die ätherische Lösung 3mal mit je 50 ml Wasser neutral gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum bei 40" eingedampft. Äls Rückstand verbleibt das rohe 9-(3-Hydroxy-(E)-1-propenyl) 9,10-dihydro-9,10-äthano-anthracen. Dieses wird aus Äther Hexan umkristallisiert und schmilzt bei 136,5-137,5'.
5,2 g (20 mMol) 9-(3-Hydroxy-(E)-1-propenyl)9,10-dihydro-9,10-äthano-anthracen wird in 100 ml Methylenchlorid gelöst, mit 4,4 g (21,7 mMol) 85prozentiger m-Chlorperbenzoesäure versetzt und 24 Stunden bei Raumtemperatur gerührt. Hierauf versetzt man mit 25 ml 2n Natronlauge, trennt dann die Schichten und wäscht die organische Phase 2mal mit je 25 ml Wasser neutral. Nach dem Trocknen über Natriumsulfat wird die Methylenchlorid-Lösung im Wasserstrahlvakuum bei 40" eingedampft. Als Rückstand verbleibt das rohe 9-(1,2-Epoxy-3-hydroxy propyl)-9, 10-dihydro-9, 10-äthano-anthracen, das aus Äther-Hexan umkristallisiert wird; Smp. 158-159".
7,6 g (27,3 mMo1)9-(1,2-Epoxy-3 -hydroxy-propyl)- 9,10-dihydro-9,10-äthano-anthracen wird in 250 ml absolutem Äther gelöst, mit 3,5 g (42 mMol) Dihydropyran und 0,38 g (2 mMol) p-Toluolsulfonsäure-dihydrat versetzt und bei Raumtemperatur 24 Stunden gerührt. Hierauf versetzt man mit 25 ml 1n Natriumhydrogencarbonatlösung, trennt die Schichten, und wäscht die ätherische Lösung 2mal mit je 50 ml Wasser neutral. Die organische Phase wird über Natriumsulfat getrocknet und anschliessend im Wasserstrahlvakuum eingedampft. Als Rückstand verbleibt kristallines 9-[1,2-Epoxy-3 -(2 -tetrahydropyranyloxy)-propyl] -9,10dihydro-9,10-äthano-anthracen, das nach einmaliger Umkristallisation aus Äther-Hexan bei 99-100" schmilzt.
3,5 g (9,7 mMol) 9-[1,2-Epoxy-3-(2-tetrahydropyranyl oxy)-propyl]-9,10-dihydro-äthano-anthracen wird in 60 ml absolutem Tetrahydrofuran gelöst und mittels eines Eis Wasserbades auf 0 abgekühlt. Unter Rühren und unter Stickstoffatmosphäre versetzt man tropfenweise mit einer Lösung von 11 mMol Lithiumdiphenylphosphid in 35 ml absolutem Tetrahydrofuran [hergestellt aus 2,42 (11 mMol) Diphenyl-chlor-phosphin und 0,77 g (110 mMol) Lithium].
Die resultierende, rote Reaktionslösung erhitzt man hierauf unter Stickstoffatmosphäre 6 Stunden unter Rühren zum Sieden, wobei sie sich allmählich bis zu hellgelb entfärbt.
Man kühlt dann mittels eines Eisbades auf 0-5" ab und versetzt tropfenweise mit 2,13 g (15 mMol) Methyljodid. Anschliessend wird das Reaktionsgemisch 15 Stunden bei Raumtemperatur gerührt, wobei sich allmählich ein weisser Niederschlag bildet. Hierauf engt man im Wasserstrahlvakuum ein und versetzt den Rückstand mit 150 ml Äther und 50 ml Wasser. Die Schichten werden getrennt und die ätherische Phase wird noch 2mal mit je 50 ml H20 gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Als Rückstand verbleibt 4,0 g rohes 9-[3 (2-Tetrahydropyranyloxy)-(Z)-1 -propenylj -9, 10-dihydro- 9,10-äthano-anthracen, als farbloses, dickflüssiges Öl, das noch mit Diphenyl-methylphosphinoxid verunreinigt ist.
Zur Reinigung wird das Rohprodukt an 120 g Alox II/n chromatographiert. Das reine 9-[3-(2-tetrahydropyranyl oxy)-(Z)-propenylj-9, 1 0-dihydro-9, 10-äthano-anthracen wird mit Benzol als farbloses, dickflüssiges Ol eluiert.
2,7 g (7,8 mMol) 9-[3-(2-Tetrahydropyranyloxy)-(Z)- 1 -propenyl] -9, 10-dihydro-9, 10-äthano-anthracen wird in 100 ml Methanol gelöst und in einem Eis-Wasserbad auf 0" abgekühlt. Man versetzt mit 5,7 g (30 mMol) p-Toluolsulfonsäure-dihydrat und rührt 1 Stunde bei 0 . Hierauf versetzt man mit 2,6 g (31 mMol) Natriumhydrogencarbonat und engt im Wasserstrahlvakuum ein. Der Rückstand wird in 150 ml Äther aufgenommen und 3mal mit je 50 ml Wasser neutralgewaschen. Die ätherische Lösung wird über Natriumsulfat getrocknet und im Wasserstrahlvakuum bei einer Badtemperatur von 40 eingedampft. Als Rückstand verbleibt 9-(3 -Hydroxy-(Z)-1 -propenyl)-9,10-dihydro- 9,10-äthano-anthracen als dickflüssiges, farbloses Ö1.
2,25 g (18,5 mMol) 9-(3-Hydroxy-(Z)-1-propenyl)9,10-dihydro-9,10-äthano-anthracen und 2,0 g (2,6 ml; 15,5 mMol) Äthyl-diisopropylamin werden in 50 ml absolutem Methylenchlorid gelöst und in einem Eis-Kochsalz Kältebad auf -15' abgekühlt. Unter Feuchtigkeitsausschluss und unter Rühren versetzt man bei einer Reaktionstemperatur von -15 bis -10" tropfenweise mit 1,14 g (0,75 ml; 10 mMol) Methansulfonsäurechlorid. Hierauf lässt man die Reaktionslösung 15 Stunden im Kühlschrank bei -5' stehen.
Anschliessend verdünnt man mit 150 ml Äther und wäscht nacheinander 2mal mit je 30 ml eiskalter 1n Schwefelsäure, 2mal mit je 30 ml 1n Natriumhydrogencarbonat-Lösung und 2mal mit je 50 ml Wasser. Die ätherische Phase wird über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Als Rückstand verbleibt kristalliner 9-(3-Hydroxy (Z)-1 -propenyl)-9,10-dihydro-9,10-äthano-anthracen- methansulfonsäureester, der nach einmaliger Umkristallisation aus Methylenchlorid-Hexan bei 133-135" schmilzt.
Beispiel 4
28 g 9-(3 -Amino-1 -propenyl)-9,10-dihydro-9,10-dihydro-9,10-äthano-anthracen werden in 60 ml Isopropanol gelöst und mit 20 g fein gemahlener Pottasche versetzt.
Unter Rühren tropft man langsam 17,1 g Methyliodid zum Reaktionsgemisch. Man lässt hierauf 2 Stunden bei 20 " ausreagieren und erwärmt anschliessend noch 1 Stunde auf 50 .
Durch Filtration werden die anorganischen Salze abgetrennt.
Der nach dem Einengen des Filtrats zurückbleibende Rückstand wird in Methylenchlorid gelöst und mit Wasser ausgewaschen. Die Methylenchloridlösung wird über wasserfreiem Natriumsulfat getrocknet, filtriert und eingedampft. Man löst den Rückstand in Aceton und leitet trockenen Chlorwasserstoff in die Lösung bis zur schwach sauren Reaktion (pH 4-5). Das 9-(3-Methylamino-l -propenyl)-9,10 -di- hydro-9,10-äthanoanthracen-hydrochlorid kristallisiert dabei direkt aus der Lösung aus. Nach dem Abnutschen wird die Substanz zweimal aus Äthanol umkristallisiert. Man erhält so farblose Kristalle vom F. 240-242'.
Das als Ausgangsmaterial verwendete 9-(3-Amino 1-propenyl)-9,10-dihydro-9,10-äthanoanthracen kann wie folgt hergestellt werden:
30 g 9-(3 -Chlor-1 -propenyl)-9, 1 0-dihydro-9,10- äthano-anthracen werden in einem 0,51-Stahlautoklaven in 200 ml Isopropanol gelöst, mit 20 ml flüssigem Ammoniak versetzt und 12 Stunden auf 100" erhitzt. Nach dem Abkühlen auf -20" giesst man den Autoklaveninhalt auf 1 Liter eisgekühlte 1n Salzsäure. Die wässrige Phase wird zweimal mit 100 ml Toluol extrahiert, dann mit Aktivkohle klarfiltriert und mit 5n Natronlauge alkalisch gestellt.
Extraktion mit Methylenchlorid liefert das gewünschte 9-(3-Amino 1-propenyl)-9,10-dihydro-9,10-äthanoanthracen als gelbes, viskoses Öl, das ohne weitere Reinigung zur Methylierung verwendet werden kann.
Beispiel 5
13,75 g 9-(3 -Methylamino-1 -propenyl)-9,10-dihydro- 9,10-äthano-anthracen werden in 20 ml Ameisensäure gelöst und mit 4,5 ml 40 %igem Formalin 20 Stunden unter Rückfluss gekocht. Hierauf wird die Reaktionslösung mit 200 ml 20 %Der Methansulfonsäure angesäuert, die Ameisensäure im Vakuum abdestilliert, die wässrige Lösung zweimal mit 200 ml Toluol extrahiert, mit 1 g Tierkohle 10 Minuten bei Raumtemperatur gerührt, über eine Schicht Diatomeenerde (Hyflo) abfiltriert, mit 6n Natronlauge stark alkalisch gestellt und 3mal mit 500 ml Chloroform extrahiert. Die vereinigten Chloroformlösungen werden mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand besteht aus 9-(3 -Dimethylamino-1 -propenyl)-9,10-dihydro9,10-äthano-anthracen und wird in das Hydrochlorid übergeführt.

5.6 g of 9- (3-chloro-1-propenyl) -9,10-dihydro-9,10-ethanoanthracene are dissolved in 25 ml of dimethylformamide and reacted with 13.0 g of N- (ß-hydroxyethyl) piperazine. The reaction mixture is heated for 15 hours at 100 ". The mixture is then evaporated and degassed in a high vacuum at 1200. The residue is dissolved in a little toluene and chromatographed over 300 g of aluminum oxide (activity II, neutral). 28 g of 9- (3-amino-1-propenyl) -9,10-dihydro-9,10-dihydro-9,10-ethano-anthracene are dissolved in 60 ml of isopropanol and mixed with 20 g of finely ground potash.

13.75 g of 9- (3-methylamino-1-propenyl) -9,10-dihydro-9,10-ethano-anthracene are dissolved in 20 ml of formic acid and refluxed with 4.5 ml of 40% formalin for 20 hours .
