Beispiel 1 7-Brom- 1 -formyl-5 -phenyl- 1H-1 .3 ,4,5 -tetrahydro-
1 ,5-benzodiazepin-4-on

The invention relates to a process for the preparation of new compounds of formula 1
EMI1.1
wherein Rl is the formyl group; R2 is an α-pyridyl or a phenyl group, the latter of which is in the o-position by a halogen atom or a nitro or
Trifluoromethyl group may be substituted; and R bromine, hydroxy, trifluoromethyl, nitro, cyano or, if R2 is o-fluorophenyl, o-bromophenyl, o-nitrophenyl, o-
Trifluoromethylphenyl or α-pyridyl means also a
Mean chlorine atom, as well as their physiologically ver compatible acid addition salts.
The new compounds are prepared by adding a compound of Formula 2
EMI1.2
formylated, which is generally done by heating with formic acid.
The following processes are suitable for the preparation of the starting materials of the formula 2: a) arylation or heteroarylation of tetrahydrobenzodiazepines of the formula 3
EMI1.3
with a halide of the formula 4 R2-X (4), in which X represents a halogen atom; b) the cyclization of substituted o-phenylenediamines of the formula 5
EMI1.4
or c) the cyclization of N-acylated o-phenylenediamines of the formula 6
EMI1.5
wherein Y is a vinyl or a '-haloethyl group, provided that Rl in the products formed is hydrogen.
According to their amine-amide structure, the compounds of formula 1 can form monobasic acid addition salts.
Suitable acid components for this purpose are, for example, hydrohalic acids, sulfuric acid and methanesulfonic acid.
The compounds according to the invention, as well as their physiologically tolerable acid addition salts, are valuable medicaments with predominantly tranquilizing effects, as well as intermediates for the production of such.
Particular emphasis is given to those compounds in which R2 is a phenyl or o-chlorophenyl group and RS is a chlorine or bromine atom or a nitro, cyano or trifluoromethyl group.
The proposed dose for the use of the new compounds of formula 1 is 0.5 to 50 mg, preferably 1-25 mg as a single dose and 5-150 mg as a daily dose.
The compounds obtainable according to the invention can be used alone or in combination with other active ingredients according to the invention, optionally also in combination with further pharmacologically active ingredients such as spasmolytics or psychotropic drugs. Suitable application forms are, for example, tablets, capsules, suppositories, solutions, juices, emulsions or dispersible powders. Corresponding tablets can, for example, by mixing the active ingredient (s) with known auxiliary substances, for example inert diluents. such as calcium carbonate, calcium phosphate or milk sugar. Disintegrants, such as corn starch or alginic acid, such as starch or gelatin, lubricants such as magnesium stearate or talc, and / or agents for achieving the depot effect, such as carboxypolymethylene, carboxymethyl cellulose, cellulose acetate phthalate, or polyvinyl acetate.
The tablets can also consist of several layers.

4 g of 7-bromo-5-phenyl-1H-2,3,4,5-tetrahydro-1,5-benzodiazepin-4-one are refluxed in 50 ml of 85% formic acid for 3 hours. The solution is then diluted with ice water, neutralized with ammonia and extracted with methylene chloride. After drying with magnesium sulfate, the solution is concentrated and crystallized from ether.
Yield: 3.4 g (78%) melting point 158-159 "C (from acetone / isopropyl ether)
