Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von bisher unbekannten heterocyclischen Verbindungen der allgemeinen Formel I (siehe Formel blatt worin R die Isopropyl-, die Isdbutyl- oder die Benzylgruppe bedeutet und X für das Anion einer Mineralsäure steht.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel I, indem man ein Säureazid der allgemeinen Formel II, worin R obige 'Bedeutung hat, in einem inerten Lösungsmittel mit annähernd der theoretischen Menge Wasser und Mineralsäure erhitzt.
Das Verfahren wird vorzugsweise wie folgt ausgeführt: Man erhitzt ein Säureazid der allgemeinen 'Formel II, worin R obige Bedeutung hat, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, in dem die Salze von Verbindungen der allgemeinen Formel I schwer löslich sind, z.B. Dimethoxyäthan oder Tetrahydrofuran, mit annähernd der theoretischen Menge Salzsäure und Wasser während kurzer Zeit auf 300 bis 900.

obige Bedeutung besitzt, kondensiert und die so erhaltenen Verbindungen der allgemeinen Formel IX gegebenenfalls anschliessend in ihre Säureadditionssalze übergeführt.
Als reaktionsfähige, funktionelle Derivate von 'Säu- ren der allgemeinen Formel X werden ihre Säurechloridhydrochloride, ihre gemischten Anhydride mit Schwefelsäure oder ihre Azide verwendet. Vorzugsweise wird die Kondensation der Verbindungen der allgemeinen Formel I mit den reaktionsfähigen, funktionellen Derivaten der Säuren der allgemeinen Formel X durchgeführt, indem man die Säurechlorid-hydrochloride der Säuren der allgemeinen Formel X in einem unter den Reaktionsbedingungen inerten Lösungsmittel wie Methylenchlorid suspendiert und bei l00 bis 0 in Anwesenheit einer tertiären organischen oder einer schwachen anorganischen Base mit den Verbindungen der allgemeinen Formel 1 reagieren lässt.
Eine andere Ausführungsform dieser IKondensation verläuft beispielsweise wie folgt:
Das gemischte Anhydrid einer Säure der allgemeinen Formel X mit Schwefelsäure wird in einem unter den Reaktionsbedingungen inerten Lösungsmittel wie Dimethylformamid und in Gegenwart einer tertiären organischen Base bei -100 bis 0 mit den Verbindungen der allgemeinen Formel 1 kondensiert.
Nach einer anderen Ausführungsform dieser Kondensation wird die Lösung eines Azides einer Säure der allgemeinen Formel X in Anwesenheit eines basischen Kondensationsmittels bei Zimmertemperatur in einem unter den Reaktionsbedingungen inerten Lösungsmittel mit den Verbindungen der allgemeinen Formel I umgesetzt.
Die so erhaltenen Verbindungen der allgemeinen Formel IX werden aus den 'Reaktionsgemischen auf an sich Ibekannte Weise isoliert und gereinigt und gegebe- nenfalls anschliessend in ihre 'Säureadditionssalze übergeführt.
Die erfindungsgemäss erhaltenen Verbindungen der allgemeinentFormel IX sind bei Zimmertemperatur kristalline Substanzen, die mit starken organischen oder anorganischen Säuren beständige, bei Zimmertemperatur kristallisierte Salze bilden. Zur Salzbildung sind als anorganische 'Säuren die Chlorwasserstoffsäure, Bromwasserstoffsäure oder Schwefelsäure und als organische Säuren die Oxalsäure, Maleinsäure, Weinsäure und Methylsulfonsäure geeignet.
Auch die Ausgangsverbindungen der allgemeinen Formel II, worin R obige 'Bedeutung hat sind neu; sie können folgendermassen hergestellt werden:
Man gelangt zu Verbindungen der allgemeinen Formel II, indem man 2!Benzyloxymalonsäurediäthylester isopropyliert resp. 2-Hydroxy-2-isopropylmalonsätiredi- äthylester benzyliert, den so erhaltenen 2-Benzyloxy-2 -isopropylmalonsäurediäthylester selektiv zum 2-Benzyloxy-2-isopropylmalonsäuremonoäthylester verseift, diesen in 2-Benzyloxy-2-isopropylmalonsäurechloridmono äthylester (Formel III) überführt, der mit Verbindungen der allgemeinen Formel W, worin - wie auch in den allgemeinen Formeln V bis VIII - R obige Bedeutung hat, kondensiert wird,
von den entstandenen Verbindungen der allgemeinen Formel V die Benzylgruppe hydrogenolytisch abspaltet, wobei ein spontaner stereospezifischer Ringschluss zu den Verbindungen der all gemeinen Formel VI stattfindet, diese zu den Verbindungen der allgemeinen Formel VII verseift, die Verbindungen der allgemeinen Formel VII in die Verbindungen der allgemeinen Formel VIII überführt, worin Y für Chlor oder'Brom steht, und diese durch Reaktion mit Natriumazid zu den Säureaziden der allgemeinen Formel II umsetzt.
Die Isopropylierung des 2-Benzyloxymalonsäuredi äthylesters wird in Anwesenheit von starken Basen, wie z.'B. Alkalimetallhydriden, Alkalimetallamiden oder Al kalimetallalkoholaten, vorzugsweise in einem unter den IReaktionsbedingungen inerten Lösungsmittel mit Reagentien, wie Diisopropylsulfat, Isopropyljodid oder Isopropylbromid durchgeführt.
Die Benzylierung des 2 -Hydroxy-2-isopropylmalonsäurediäthylesters erfolgt in Gegenwart von starken Basen, wie Alkalimetallhydriden, Alkalimetallamiden oder Alkalimetallaikoholaten, in einem polaren, unter den Reaktionsbedingungen inerten Lösungsmittel, wie Dimethylacetamid oder Dimethylsulfoxid, mit Hilfe von Benzylierungsmitteln, wie Benzylhalogeniden, p-Toluolsulfonsäurebenzylester, m Nitrobenzolsulfonsäurebenzylester usw.
Der erhaltene 2Benzyloxy-2- isopropylmalonsäuredi- äthylester wird vorzugsweise mit Hilfe einer Lösung eines Alkalimetallhydroxids in einem niederen aliphatischen Alkohol zum entsprechenden Malonsäuremono äthylester verseift. Auf der Stufe dieses Malonsäuremonoäthylesters kann eine Trennung in die optischen Isomeren durchgeführt werden, indem man den racemischen tMalonsäuremonoäthylester mit optisch akti ven 3 , z iB. mit Pseudoephedrin, in ein Diastereo- merengemisch überführt, das durch fraktionierte Kri- stallisation in die beiden Diastereomeren getrennt wird, aus denen auf an sich bekannte Weise die beiden optisch aktiven 2iBenzyloxy-2-isopropylmalonsäuremono- äthylester gewonnen werden können.
2- Benzyloxy -2- isopropylmalonsäuremonoäthylester wird, eventuell in einem unter den Reaktionsbedingun gen inerten organischen Lösungsmittel, in Gegenwart eines N-di(nieder)alkylsubstituierten Säureamides einer aliphatischen Monocarbonsäure mit 1-3 Kohlenstoffatomen, mit Chlorierungsmitteln zur Reaktion gebracht.
Als Lösungsmittel für diese Reaktion eignen sich z:B.
Chloroform, Methylenchlorid oder aber das N-di(nieder)alkylsubstituierte Säureamid selbst.
Der so erhaltene 2-Benzyloxy-2-isopropylmalonsäure- halogenidmonoäthylester (Formel IfI) wird nun in Anwesenheit eines tertiären Amines, wie Pyridin oder N Methylmorpholin, bei etwa 5-70 mit den Verbindungen der allgemeinen Formel W kondensiert. Von dem so erhaltenen Acylierungsprodukt der allgemeinen Formel V wird die Benzylgruppe hydrogenolytisch, z.'B. durch katalytische Hydrierung bei Normaldruck unter Verwendung von Palladium/ Aktivkohle oder Raney-Nickel als Katalysator, abgespalten, wobei spontan ein stereospezifischer (bezüglich des C-Atomes in Position leib) Ringschluss zu den Verbindungen der allgemeinen Formel VI stattfindet.
Eine Auftrennung in Stereoisomere kann weiters auf der Stufe der Verbindungen der allgemeinen Formel V oder VI durch Chromatographie, z.B. an Silicagel, durchgeführt werden, wobei als Elutionsmittel beispielsweise Methylenchlorid mit verschiedenem Behalt an Methanol gute Dienste leistet.
Die Verbindungen der allgemeinen Formel VI werden z. B. mit Natriumhydroxid bei Zimmertemperatur zu den freien 'Säuren der allgemeinen Formel VII verseift, die mit frisch sublimiertem Phosphorpentachlorid bei Zimmertemperatur oder in Form ihrer Natriumsalze durch Reaktion mit Oxalylchlorid in einem unter den Reaktionsbedingungen inerten Lösungsmittel, wie Äther oder Petroläther-Äther-Gemischen, in die Säurechloride der allgemeinen Formel vm übergeführt werden. Die Verbindungen der allgemeinen Formel VIII werden auf an sich bekannte Weise mit Natriumazid zu den Verbindungen der allgemeinen Formel II umgesetzt.
In den nachfolgenden IBeispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Die 5mop. wurden im offenen Röhrchen auf einer Apparatur nach TO1TOLI bestimmt und sind korrigiert. Die IR-Spektren wurden, sofern kein Lösungsmittel speziell erwähnt ist, in Nujol-Suspension aufgenommen. Die Aufnahme der Uv-Spektren erfolgte in Methanollösung. Die pK-Bestimmungen wurden im System fMethylcellosolve/Wasser im Volumenverhältnis 80: 20 ausgeführt. Die Aufnahme der NMR-Spektren wurde mit Tetramethylsilan als interne Referenz, Angaben in S-Werten, durchgeführt.
Die Ausgangsverbindungen sind entweder bekannt oder können nach an sich bekannten Methoden hergestellt werden.

Beispiel I (2R,5S,1OaS,10bS)-2-Amino-3,6-dioxo-10-hydroxy- -2,5-diisopropyloctahydro-8H-oxazolo[3,2-a]pyrrolo- [2,1 -cjpyrazin-hydrochlorid
Man trocknet 132 g rohes (2R,5S,10aS,10bS)-2-Azi- docarbonyl - 3,6 - dioxo- lOb-hydroxy-2,5-diisopropylocta- hydro-8H-oxazolo[3,2-a]pyrrolo[2,1-c]pyrazin bei 250 im Hochvakuum bis zur Gewichtskonstanz, die nach ca. 24 Stunden erreicht wird. Hierbei verschwindet im IR-Spektrum die N3JBande (2110 cm-Ú), die quantitativ durch die Isocyanat;Bande (2210 cm-1) ersetzt wird.
Die so erhaltene Verbindung wird bei 300 in 650 ml wasserfreiem Tetrahydrofuran aufgelöst, anschliessend 7,2 ml Wasser und 100 ml Tetrahydrofuran/HCl 4N zugegeben, bei 300 bis zur vollständigen Auflösung gerührt, mit einigen (2R,5S,lOaS,lObS)-2-Amino-3,6- -dioxo-10b-hydroxy-2,5-diisopropyloctahydro-8 H-oxazolo[3,2-a]pyrrolo[2,1-c]pyrazin-hydrochlorid-Kristallen geimpft und während 16 Stunden Ibei 390 stehen gelassen. Das Endprodukt scheidet sich in eine Kristallkruste aus, während sich die Mutterlauge hell- ibis dunkelgelb verfärbt.
Nach 6 Stunden bei 00 giesst man die Mutterlauge ab, verarbeitet die Kristalikruste in 500 ml auf 00 gekühltem, wasserfreiem Tetrahydrofuran, filtriert, wäscht 2mal mit je 1000 ml wasserfreiem Äther u. trocknet im Hochvakuum bei 30 . Man erhält (2R,5'S,lOaS, 1*S) -2-Amino-3,6-dioxo- lOb-hydroxy - 2,5 - diisopropyloctahydro-8H-oxazolo[3,2-a]pyrrolo[2,1 -c]pyrazin-hydrochlorid vom Smp. 200 - 2020, [D20 = +58,80 (c = 1 in Dimethylformamid).
Das als Ausgangsmaterial verwendete (2R,SS,lOaS, lObS)-2-Azidocarbonyl-3,6-dioxo-10b-hydroxy-2,5-diiso propyloctaXydro H- oxarolo[3,2-a]pyrrolo[2,1-c]pyrazin kann wei folgt hergestellt werden: a) (2R,SS,lOaS,lObS)-2-Carhoxy-3,6-dioxo-lOb-hydro- xy-2,5-diisopropyloctshydro-8H-oxazolo[3,2-a]- pyrrolo[2,1c]pyrazin
Eine Suspension von 99,8 g (509 mM) (3S,8a'S)-1,4- -Dioxo-3 isopropyloctahydropyrrolo[1,2-a]pyrazin in 155 mi N-Methylmorpholin wird mit 155 g (520 mM) d-2 -Benzyloxy-2-isopropylmalonsäurechloridmonoäthylester versetzt und während 21/2 Stunden unter Rühren auf 700 erwärmt.
Die erhaltene dicke Masse wird in 1000 mol
Methanol gelöst und in Anwesenheit von 50 g 10% iger Palladium-Kohle hydriert. Nach Beendigung der Was serstoffaufnahme werden noch 25 g Katalysator zuge geben und weiterhydriert. Der uKataiysator wird abfil triert, das Filtrat bei 490 zur Trockne gedampft und der Rückstand mit Wasser gewaschen. Das erhaltene (2R,5S,10aS,10bS) -2- Äthoxycarbonyl-3 ,6-dioxo- 1*-hy- droxy-2,5-ditisopropyloctahydro-8H-oxazolo[3,2 - a]pyrro lot2, 1 -c]pyrazin wird in 500 ml 2N Natronlauge gelöst und 4 Stunden bei 250 aufbewahrt.
Die Lösung wird auf 00 abgekühlt, mit 4 N Schwefelsäure auf pH 2 gestellt die ausgeschiedene kristalline Masse abfiltriert, mit Wasser neutral gewaschen und im Hochvakuum getrocknet. Das so erhaltene (2R,5S,10aS,10bS)-2-Carb- oxy-3,6-dioxo- lOb-hydroxy-2,5-diisopropyloctahydro-8H- -oxazolo[3 ,2-a]pyrrolo'[2, 1 -c]pyrazin kristallisiert aus Di- oxan/Wasser als Monohydrat und schmilzt bei 162 bis
1640 (Zers.), la1D20 = -15 (c = 1, Pyridin).
b) (2R,5S,10aS,10bS)-2-Chloroformyl-3,6-dioxo-10b- -hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a]pyrrolo[2,1-c]pyrazin
Eine Suspension von 135 g (644 mM) Phosphor pentachlorid in einem Gemisch von 800 ml wasserfreiem
Diäthyläther und 800 ml Petroläther wird 60 Minuten bei MO gerührt, auf 100 abgekühlt, 109,6 g (322 mM) (2R,5S,10aS,10bS)-2carboxy-3,6-dioxo-10b-hydroxy-2,5 -diisopropyloctahydro - 8H - oxazolo'[3,2-a pyrrolo [2,1-c] pyrazin zugegeben und die Suspension während 3 Stun den bei 250 gerührt.
Filtration, Waschen der kristallinen
Masse mit Äther/Petroläther (1:1) und Trocknen im Vakuum unter Feuchtigkeitsausschluss liefert das (2R 5S, lOaS,10SS) - 2 - Chloroformyl -3,6 - dioxo-lOb-hydroxy- -2,5-diisopropyloctahydro-8H-oxazolot3,2 - a]pyrrolo[2, 1 - c]pyrazin vom Smp. 130 - 1320 (Zers.), das unbeständig ist und so rasch wie möglich für die weiteren Syntheseschritte verwendet wird.
c) (2R,5S,lOaS, lObS)-2-Azidocarbonyl-3,6-dioxo- -lOb-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a]pyrrolo[2,l-c]pyrazin
Eine Mischung von 1000 ml Methylenchlorid, 160 ml Wasser und 57 g (870 mM) Natriumazid wird allmählich bei - 50 unter sehr kräftigem Rühren mit 111,6 g (311 miM) (2R,5S, 1 0aS, 1 0bS)-2-Chloroformyl-3,6-dioxo- - lOb-hydroxy-2,5-diisopropyloctahydro - 8H - oxazolo[3,2 ajpyrroio'[2, 1 -c]pyrazin versetzt und anschliessend noch 6 Minuten gerührt.
Nach Trennung der Phasen wird die wässrige Phase mit 100 ml Methylenchlorid extrahiert, die vereinigten organischen Phasen mit 1N Na triumhydrogencarbonatiösung gewaschen, über Natriumsulfat getrocknet und das rohe Azid zur Trockne eingedampft.
Beispiel 2 9,10-Dihydroergocornin
6,5 g (20 mM) 9,1 0-Dihydrolysergsäurechlorid-hydro- chlorid und 3.5 g (10 mM) (2R,5S,l0aS,lObS)-2-Amino- -3,6-dioxo - lOb - hydroxy 2,5- diisopropyloctahydro-8H -oxazolo[3,2-a]pyrrolof2,1-c]pyrazin-hydrochlorid werden in 50 ml Dimethylformamid suspendiert und bei 100 gerührt. Innerhalb von 15 Minuten werden 2 mol wasserfreies Pyridin zugetropft, wobei die Suspension allmählich in Lösung geht. Nach 1 stündigem Rühren bei -100 bis 00 werden noch 10 ml 4 N Natriumcarbonatlösung zugegeben und das Reaktionsgemisch unter vermindertem Druck bei 390 zur Trockne eingedampft.
Man löst den Rückstand in einem Gemisch von 100 ml Methylenchlorid/Methanol (8 : 2) und 20 ml 4N Natriumcarbonatlösung auf, trennt die Phasen, wäscht die organische Phase mit 3 X 20 ml 4 N Natriumcarbonatlösung, extrahiert die vereinigten wässrigen Phasen mit 4 X 50 ml Methylenchllorid/Methanol (8 : 2), trocknet die vereinigten organischen Phasen über Natriumsulfat und Tierkohile und dampft zur Trockne ab. Der Rückstand wird in 20 ml heissem Äthanol gelöst, mit Äther bis zur Trübe versetzt und kristallisieren gelassen. Man erhält so 9,10-Dihydroergocornin vom Smp. 1850 (Zers.), [EX]D20 = 470 (c = 1 in Pyridin).
Unter den gleichen Bedingungen erhält man aus Lysergsäure und (2R,5S, 10aS,1ObS)-2-Amino-3,6-dioxo- - 1 Ob-hydroxy - 2,5 - diisopropyloctahydro-8H-oxazotot3,2- a]pyrrolo[2,1-c]pyrazin-hydrochtorid ein Ergocornin-Ergocorninin-Gemisch, aus welchem nach an sich bekannten Methoden kristallisiertes Ergocornin isoliert wird, das in jeder Hinsicht mit dem natürlichen Produkt identisch ist.

The 2-benzyloxy-2-isopropylmalonic acid di-ethyl ester obtained is preferably saponified with the aid of a solution of an alkali metal hydroxide in a lower aliphatic alcohol to give the corresponding malonic acid mono-ethyl ester. At the stage of this malonic acid monoethyl ester a separation into the optical isomers can be carried out by the racemic t-malonic acid monoethyl ester with optically active 3, z iB. with pseudoephedrine, converted into a mixture of diastereomers, which is separated into the two diastereomers by fractional crystallization, from which the two optically active 2i-benzyloxy-2-isopropylmalonic acid monoethyl ester can be obtained in a manner known per se.
2- Benzyloxy -2- isopropylmalonic acid monoethyl ester, possibly in an organic solvent which is inert under the reaction conditions, in the presence of an N-di (lower) alkyl-substituted acid amide of an aliphatic monocarboxylic acid having 1-3 carbon atoms with chlorinating agents.
Suitable solvents for this reaction are, for example:
Chloroform, methylene chloride or the N-di (lower) alkyl-substituted acid amide itself.
The 2-benzyloxy-2-isopropylmalonic acid halide monoethyl ester (formula IfI) thus obtained is then condensed with the compounds of the general formula W at about 5-70 in the presence of a tertiary amine, such as pyridine or N-methylmorpholine. The benzyl group of the acylation product of the general formula V obtained in this way is hydrogenolytically, z.'B. by catalytic hydrogenation at normal pressure using palladium / activated carbon or Raney nickel as a catalyst, with a stereospecific ring closure (with respect to the carbon atom in position leib) to the compounds of the general formula VI taking place spontaneously.
A separation into stereoisomers can furthermore be carried out at the stage of the compounds of the general formula V or VI by chromatography, e.g. on silica gel, with methylene chloride with various contents of methanol, for example, serving as an eluent.
The compounds of general formula VI are z. B. saponified with sodium hydroxide at room temperature to the free 'acids of general formula VII, which with freshly sublimed phosphorus pentachloride at room temperature or in the form of their sodium salts by reaction with oxalyl chloride in a solvent inert under the reaction conditions, such as ether or petroleum ether-ether mixtures, be converted into the acid chlorides of the general formula vm. The compounds of the general formula VIII are reacted in a manner known per se with sodium azide to give the compounds of the general formula II.
In the following I-Examples, which explain how the process is carried out but are not intended to restrict the scope of the invention in any way, all temperatures are given in degrees Celsius.
The 5mop. were determined in an open tube on a TO1TOLI apparatus and have been corrected. Unless a specific solvent is mentioned, the IR spectra were recorded in Nujol suspension. The UV spectra were recorded in methanol solution. The pK determinations were carried out in the f-methylcellosolve / water system in a volume ratio of 80:20. The recording of the NMR spectra was carried out with tetramethylsilane as internal reference, data in S values.
The starting compounds are either known or can be prepared by methods known per se.

Example I (2R, 5S, 1OaS, 10bS) -2-Amino-3,6-dioxo-10-hydroxy--2,5-diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolo- [2,1 - cjpyrazine hydrochloride
132 g of crude (2R, 5S, 10aS, 10bS) -2-azidocarbonyl-3,6-dioxolOb-hydroxy-2,5-diisopropylocta-hydro-8H-oxazolo [3,2-a] pyrrolo are dried [2,1-c] pyrazine at 250 in a high vacuum to constant weight, which is reached after about 24 hours. The N3J band (2110 cm-Ú) disappears in the IR spectrum, which is quantitatively replaced by the isocyanate band (2210 cm-1).
The compound obtained in this way is dissolved in 650 ml of anhydrous tetrahydrofuran at 300, then 7.2 ml of water and 100 ml of tetrahydrofuran / HCl 4N are added, stirred at 300 until complete dissolution, with a few (2R, 5S, 10aS, 10bS) -2 -Amino-3,6- -dioxo-10b-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolo [2,1-c] pyrazine hydrochloride crystals seeded and Ibei for 16 hours 390 left standing. The end product separates out in a crystal crust, while the mother liquor turns light yellow to dark yellow.
After 6 hours at 00, the mother liquor is poured off, the crystal crust is processed in 500 ml of anhydrous tetrahydrofuran cooled to 00, filtered, washed twice with 1000 ml of anhydrous ether and the like. dries in a high vacuum at 30. (2R, 5'S, 10aS, 1 * S) -2-amino-3,6-dioxo-10-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolo [2,1 - c] pyrazine hydrochloride of m.p. 200-2020, [D20 = +58.80 (c = 1 in dimethylformamide).
The (2R, SS, 10aS, 10bS) -2-azidocarbonyl-3,6-dioxo-10b-hydroxy-2,5-diisopropyloctaXydro H-oxarolo [3,2-a] pyrrolo [2,1- used as starting material c] Pyrazine can be prepared as follows: a) (2R, SS, 10aS, 10bS) -2-carhoxy-3,6-dioxo-10b-hydroxy-2,5-diisopropyloctshydro-8H-oxazolo [3.2 -a] - pyrrolo [2,1c] pyrazine
A suspension of 99.8 g (509 mM) (3S, 8a'S) -1,4- -Dioxo-3 isopropyloctahydropyrrolo [1,2-a] pyrazine in 155 ml N-methylmorpholine is mixed with 155 g (520 mM) d- 2 -Benzyloxy-2-isopropylmalonsäurechloridmonoäthylester added and heated to 700 for 21/2 hours with stirring.
The obtained thick mass is in 1000 mol
Dissolved methanol and hydrogenated in the presence of 50 g of 10% palladium-carbon. After the absorption of hydrogen has ended, 25 g of catalyst are added and hydrogenation is continued. The uKataiysator is filtered off, the filtrate is evaporated to dryness at 490 and the residue washed with water. The obtained (2R, 5S, 10aS, 10bS) -2- ethoxycarbonyl-3, 6-dioxo- 1 * -hydroxy-2,5-ditisopropyloctahydro-8H-oxazolo [3,2 - a] pyrrolot2, 1 - c] Pyrazine is dissolved in 500 ml of 2N sodium hydroxide solution and stored at 250 for 4 hours.
The solution is cooled to 00, adjusted to pH 2 with 4N sulfuric acid, the precipitated crystalline mass is filtered off, washed neutral with water and dried in a high vacuum. The (2R, 5S, 10aS, 10bS) -2-carboxy-3,6-dioxolOb-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3, 2-a] pyrrolo '[2 , 1 -c] pyrazine crystallizes from dioxane / water as a monohydrate and melts at 162 to
1640 (dec.), La1D20 = -15 (c = 1, pyridine).
b) (2R, 5S, 10aS, 10bS) -2-chloroformyl-3,6-dioxo-10b-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolo [2,1-c ] pyrazine
A suspension of 135 g (644 mM) of phosphorus pentachloride in a mixture of 800 ml of anhydrous
Diethyl ether and 800 ml petroleum ether are stirred for 60 minutes at MO, cooled to 100, 109.6 g (322 mM) (2R, 5S, 10aS, 10bS) -2carboxy-3,6-dioxo-10b-hydroxy-2,5 - diisopropyloctahydro-8H-oxazolo '[3,2-a pyrrolo [2,1-c] pyrazine was added and the suspension was stirred at 250 for 3 hours.
Filtration, washing the crystalline
Mass with ether / petroleum ether (1: 1) and drying in vacuo with exclusion of moisture provides the (2R 5S, 10aS, 10SS) - 2 - chloroformyl -3,6 - dioxo-10b-hydroxy- -2,5-diisopropyloctahydro-8H- oxazolot3,2 - a] pyrrolo [2, 1 - c] pyrazine with a melting point of 130-1320 (decomp.), which is unstable and is used as quickly as possible for the further synthesis steps.
c) (2R, 5S, 10aS, 10bS) -2-azidocarbonyl-3,6-dioxo- -lOb-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolo [2, l-c] pyrazine
A mixture of 1000 ml of methylene chloride, 160 ml of water and 57 g (870 mM) of sodium azide is gradually added at - 50 with very vigorous stirring with 111.6 g (311 miM) (2R, 5S, 10aS, 1 0bS) -2- Chloroformyl-3,6-dioxo-10b-hydroxy-2,5-diisopropyloctahydro-8H-oxazolo [3,2 ajpyrroio '[2, 1 -c] pyrazine was added and the mixture was then stirred for a further 6 minutes.
After the phases have been separated, the aqueous phase is extracted with 100 ml of methylene chloride, the combined organic phases are washed with 1N sodium hydrogen carbonate solution, dried over sodium sulfate and the crude azide is evaporated to dryness.
Example 2 9,10-dihydroergocornine
6.5 g (20 mM) 9.1 0-dihydrolysergic acid chloride hydrochloride and 3.5 g (10 mM) (2R, 5S, 10aS, 10bS) -2-amino- -3,6-dioxo-10b-hydroxy 2 , 5- diisopropyloctahydro-8H-oxazolo [3,2-a] pyrrolof2,1-c] pyrazine hydrochloride are suspended in 50 ml of dimethylformamide and stirred at 100%. 2 mol of anhydrous pyridine are added dropwise over the course of 15 minutes, the suspension gradually dissolving. After stirring for 1 hour at -100 to 00, 10 ml of 4N sodium carbonate solution are added and the reaction mixture is evaporated to dryness under reduced pressure at 390.
The residue is dissolved in a mixture of 100 ml of methylene chloride / methanol (8: 2) and 20 ml of 4N sodium carbonate solution, the phases are separated, the organic phase is washed with 3 × 20 ml of 4N sodium carbonate solution, and the combined aqueous phases are extracted with 4 × 50 ml of methylene chloride / methanol (8: 2), dry the combined organic phases over sodium sulfate and animal cabbage and evaporate to dryness. The residue is dissolved in 20 ml of hot ethanol, ether is added until it is cloudy and the mixture is left to crystallize. This gives 9,10-dihydroergocornine with a melting point of 1850 (decomp.), [EX] D20 = 470 (c = 1 in pyridine).
Under the same conditions, lysergic acid and (2R, 5S, 10aS, 1ObS) -2-amino-3,6-dioxo- - 1 Ob-hydroxy - 2,5 - diisopropyloctahydro-8H-oxazotot3,2- a] pyrrolo are obtained [2,1-c] pyrazin-hydrochtorid an ergocornine-ergocorninine mixture, from which, according to methods known per se, crystallized ergocornine is isolated which is identical in every respect to the natural product.
