Verfahren zur Herstellung von Spiro-Verbindungen aus Schiff'schen Basen
Die Erfindung betrifft ein Verfahren zur Herstellung von bisher nicht bekannten Spiro-Verbindungen unter Anwendung einer neuartigen Additionsreaktion von Maleinimiden mit aromatisch substituierten Schiff'schen Basen sowie die Verwendung der so erhaltenen Verbindungen zur Herstellung ihrer Reduktionsprodukte.
Die bekannten Additionsreaktionen von Phenylisocyanat (Chem. Ber. 100, 160Q [1967]) bzw. Acetylendicarbonsäureestern (Liebigs Ann. Chem. 688, 98 [1965]) mit Benzyliden-methylamin hätten zur Vermutung Anlass geben können, dass eine Addition von Maleinimid an Benzyliden-methylamin zu Verbindungen etwa der Struktur

führen müsste. Aufgrund der Angaben in Chem. Ber.
100, 1617 [1967] (insbesondereZeile 13) musste jedoch angenommen werden, dass eine Reaktion zwischen Verbindungen in der Art von Benzyliden-methylamin und N-Phenyl-maleinimid überhaupt nicht eintritt.
Ueberraschenderweise wurde nun gefunden, dass man zu neuen Spiro-Verbindungen der Formel

in welcher Ar einen gegebenenfalls Heteroatome aufweisenden aromatischen Rest und Rt und R2 Wasserstoff oder organische Reste bedeuten, gelangt, wenn man eine Schiff'sche Base der Formel
Ar- CH= N- R1 in Gegenwart eines Lösungsmittels bei erhöhter Temperatur mit einem am Stickstoffatom die Gruppe R2 aufweisenden Maleinimid reagieren lässt.
Die Strukturformel I wird den Verfahrensprodukten aufgrund ihrer spektroskopischen Daten zugeschrieben, welche sich bisher mit keiner anderen Struktur in befriedigender Weise vereinbaren lassen; allerdings ist die Kopplungskonstante für die Methylengruppe mit 19,5 Hz (vgl. Beispiel 2) etwas höher als der aufgrund der Literatur erwartete Wert.
Bei den als Ausgangsstoffe dienenden Schiff'schen Basen der genannten Formel kann der aromatische Rest Ar beispielsweise eine Phenyl- oder Naphthylgruppe sein, welche ihrerseits mit Halogen, gegebenenfalls halogenierten Alkylgruppen, gegebenenfalls verätherten oder acylierten Hydroxy- oder Mercaptogruppen, Carb oxy-, Alkoxycarbonyl- oder Carbamoylresten, primären, sekundären oder tertiären Aminogruppen, Nitril- oder Nitrogruppen substituiert sein kann. Der Substituent R' der Schiff'schen Base kann insbesondere eine geradlinige oder verzweigte niedere Alkylgruppe oder eine Cycloalkylgruppe sein, welche gegebenenfalls mit einer oder mehreren der vorstehend aufgezählten Gruppen substi tuiertist.
Der an das Stickstoffatom des Maleinimids gebundene organische Rest R2 kann eine der für Ar oder R1 erwähnten Gruppen sein und ist vorzugsweise ein gegebenenfalls substituierter niederer Alkylrest.
Als Lösungsmittel haben sich namentlich Eisessig sowie auch nicht allzu tief siedende Kohlenwasserstoffe wie Xylol als geeignet erwiesen. Die Reaktion wird zweckmässig bei Temperaturen von nicht wesentlich unter 100 C durchgeführt.
Die Erfindung betrifft ferner die Verwendung der nach dem genannten Verfahren überraschenderweise erhaltenen, neuartigen 2:1-Addukte zur Herstellung ihrer Reduktionsprodukte, welche sich bilden, wenn man die Verbindungen der Formel I einer die Oxo-Gruppen ab spaltenden Reduktion unterwirft. Die Reduktion kann dabei nach an sich bekannten Methoden erfolgen; als besonders zweckmässige Reduktionsmittel haben sich komplexe Hydride wie z. B. Lithiumaluminiumhydrid erwiesen.

zugeschrieben wird, sind für pharmazeutische Zwecke verwendbar.
In den anschliessenden Beispielen sind die Schmelzpunkte unkorrigiert. Die Elementaranalysen wurden durch den Elemental Analyser der Firma Carlo Erba automatisch bestimmt.
Beispiel 1
Ein Gemisch von 191,6 g (1,25 Mol) N-n-Butyl-maleinimid, 75,0 g (O,63 Mol) Benzyliden-methylamin und 600 ml Eisessig wurde unter Rühren erhitzt bis zur Erreichung einer Innentemperatur von 100"C, worauf man die Temperatur unter fortgesetztem Rühren während 45 min. auf 800C hielt. Aus dem nach Stehenlassen über Nacht gebildeten Niederschlag erhielt man durch Absaugen 52,2 g Rohprodukt vom Smp.
187-190,5 C. Verdünnen der Mutterlauge mit Wasser, kontinuierliche Extraktion mit Äther und Eindampfen des Extraktes ergab weitere 45,5 g Material (zusammen 229 mM oder 36 /o der Theorie). Umkristallisieren des Rohproduktes aus wässrigem Aceton lieferte ein farbloses Pulver vom Smp. 192-1930C.

Das Infrarot- und das UV-Spektrum sind mit der Struktur 3,5'-Di-n-butyl-l'-methyl-8'-phenyl 2,4,4',6'-tetraoxo-spiro{pyrrolidin- 1,2'-(octahydro[3,4-c]-pyrrol) } in Einklang.
NMR (100 MC, CDC13, in z): 5,81 d, 7,46 d (J-18,5 Hz); 7,95 s (N-CH3); 5,42 d (J-10 Hz).
Reduktion: Eine Lösung von 42,5 g (100 mM) des erhaltenen Adduktes in Dioxan liess man bei 900C in eine gerührte Mischung von 19,0 g LiAlH4 mit 1,2 Liter Dioxan eintropfen. Nach beendeter Reaktion wurde zur Zersetzung Wasser und anschliessend Natronlauge zugetropft. Das noch eine Weile weiter gerührte Reaktionsgemisch wurde filtriert, und nach Eindampfen des Filtrats wurde an Aluminiumoxyd der Aktivität m chromatographiert. Aus den vereinigten Benzin-Eluaten erhielt man durch Destillation 16 g Öl vom Sdp. 168 174 C/0,005 mm; nd20 1,5273 (46% der theor. Ausbeute).

Das IR-Spektrum steht mit der angenommenen Struktur von 3 ,5'-Di-n-butyl-1'-methyl- 8'-phenyl-spiro{pyrrolidin- 1,2'-(octahydro-pyrrolo[3,4-c]-pyrrol) } in Einklang.
Beispiel 2
Einer Lösung von 138,6 g (800 mM) N-Phenyl-maleinimid in 800 ml Eisessig wurde in einem mit Rührer, Rückflusskühler und Innenthermometer ausgestatteten Dreihalskolben eine Lösung von 47,7 g (400 mM) N-Benzyliden-methylamin zugesetzt. Das Reaktionsgemisch, welches sich auf 1000 C erwärmt hatte, wurde während 11/4 Stunden unter Rühren auf Rückflusstemperatur erhitzt und dann über Nacht stehen gelassen.
Durch Abfiltrieren des entstandenen Niederschlags erhielt man 71,4 g Addukt (38 /o der Theorie) vom Smp.
239-2410C. Die Reinigung durch Extraktion mit Alkohol im Soxhlet-Apparat ergab ein farbloses, unlösliches Produkt vom Smp. 242-243"C

Die Struktur von 1'-Methyl-3,5'-triphenyl- 2,4,4',6'-tetraoxo-spiro{pyrrolidin 1,2-(octahydropyrrolo[3,4-c]-pyrrol)} wird durch das NMR-Spektrum bei 60 MHz in CDC13- Lösung bewiesen. Die chemischen Verschiebungen sind in z relativ zu TMS als Standard angegeben: 7,75 s (3H, N,,-CH5); 5,34 d (8'-H, J 9,5 Hz); 6,07 q (7'-H); 6,55 d (3'-H, J 8 Hz); 5,58 und 7,20 d (2-H2, J 19,5 Hz).
Auch das IR- und das UV-Spektrum stehen mit der Struktur in Einklang.
Aus der Mutterlauge kristallisiert in geringer Ausbeute ein Produkt, welches nach Umkristallisieren aus Aethanol einen Smp. von 237-241 C aufweist und bei der Mischschmelzpunkt-Probe mit dem Hauptprodukt eine Depression von 20 C ergibt:

Das IR-Spektrum hat einen ähnlichen Charakter wie dasjenige des Hauptproduktes, ist damit aber nicht identisch, so dass auf Stereoisomerie zu schliessen ist.
Reduktion:
In einem Soxhlet-Extraktionsapparat wurden 23,3 g (50 mM) des Hauptproduktes während 10 Stunden extrahiert mit einem Gemisch von 9,5 g LiAlH4 und 750 ml Dioxan. Nach dem Abkühlen wurde zersetzt durch tropfenweise Zugabe von 9,5 ml Wasser, 19 ml 2 n Natronlauge und weiteren 19 mi Wasser. Nach einstündigem Rühren wurde das ausgeschiedene Aluminiumhy.
droxyd durch Filtration abgetrennt und das Filtrat im Vakuum eingedampft. Der Rückstand ergab nach Umkristallisieren aus Butanol 11,27 g Produkt vom Smp.
179-1830C 85(55 /o d.Th), aus dem man durch weiteres limkristallisieren aus Essigester feine farblose Kristalle vom Smp. 177-180,50C erhielt: Ber. für Q8H31N5 (409,5) O 0% gef. C28,4H30,15N3 (409, massenspektr.) 0 0 0/o
Die Anwesenheit von 31 Wasserstoffatomen wird durch die Integration des NMR-Spektrums sichergestelit. Das Produkt ist somit 1 '-Methyl-3,5',8'-triphenyl-spiro{pyrrolidin- 1 ,2'-(octahydropyrrolo[3,4-clpyrrol)) .
Beispiel 3
Ein Gemisch von 34,7 g (200 mM) N-Phenyl-male inirnid, 16,3 g (100 mM) p-Dimethyl-aminobenzyliden-methylamin und 250 ml. abs. Xylol wurde 10 Stunden am Rückfluss erhitzt. Der beim Erkalten sich bildende Niederschlag wurde durch Filtration abgetrennt und ergab 20,5 g eines Rohproduktes vom Smp. 181-2020C, aus dem man durch Umkristallisieren aus Dioxan 8,05 g Substanz vom Smp. 231-2350C erhielt. Einengen der ersten Nfutteilauge lieferte weitere 9,43 g Substanz vom Smp.
228-234 C (zusammen somit 34 /o d.Th.). Weiteres Umkristallisieren aus Äthanol ergab ein farbloses Pulver vom Smp. 235-237,5 C.

Beim Produkt handelt es sich somit um 8'-p-Dimethylaminophenyl-3,5-diphenyl-1'-methyl2,4,4',6'-tetraoxo-spiro{pyrrolidin- 1,2'-(octahydropyrrolo[3,4-c]-pyrrol)}.
Durch Reduktion nach dem in Beispiel 2 beschriebenen Vorgehen erhielt man ein farbloses Pulver vom Smp. 160-162,5 C (krist. aus Cyolohexan).

Beispiel 4
In analoger Weise erhielt man aus a-Furturyliden methylamin und N-n-Butylmaleinimid in Butanol (1 Stunde bei 1300C) mit 54 /o Ausbeute das 1:2-Addukt: Smp. 130-1320C (aus Butanol).

Entsprechende 1 :2-Addukte wurden in ähnlicher Weise beispielsweise auch aus Benzalanilin und N-Butyl-maleinimid (Smp. 187-1890C 1 Benzin) sowie aus Benzyliden-methylamin und unsubstituiertem Maleinimid (Smp. 272-278 C/i-Propanol) erhalten; auch in diesen Fällen standen Analysenresultate und spektroskopische Daten mit der postulierten Struktur im Einklang.

It has now surprisingly been found that new spiro compounds of the formula

in which Ar is an aromatic radical optionally containing heteroatoms and Rt and R2 are hydrogen or organic radicals, is obtained when using a Schiff base of the formula
Ar-CH = N-R1 can react in the presence of a solvent at elevated temperature with a maleimide having the group R2 on the nitrogen atom.
The structural formula I is assigned to the products of the process on the basis of their spectroscopic data, which so far have not been compatible with any other structure in a satisfactory manner; however, the coupling constant for the methylene group at 19.5 Hz (cf. Example 2) is somewhat higher than the value expected from the literature.
In the case of the Schiff's bases of the formula mentioned used as starting materials, the aromatic radical Ar can be, for example, a phenyl or naphthyl group, which in turn is oxy, alkoxycarbonyl or with halogen, optionally halogenated alkyl groups, optionally etherified or acylated hydroxyl or mercapto groups, carboxy Carbamoyl, primary, secondary or tertiary amino groups, nitrile or nitro groups can be substituted. The substituent R 'of the Schiff base can in particular be a straight or branched lower alkyl group or a cycloalkyl group which is optionally substituted with one or more of the groups listed above.
The organic radical R2 bonded to the nitrogen atom of the maleimide can be one of the groups mentioned for Ar or R1 and is preferably an optionally substituted lower alkyl radical.

Example 2
A solution of 47.7 g (400 mM) N-benzylidene-methylamine was added to a solution of 138.6 g (800 mM) N-phenylmaleimide in 800 ml glacial acetic acid in a three-necked flask equipped with a stirrer, reflux condenser and internal thermometer. The reaction mixture, which had warmed to 1000 ° C., was heated to reflux temperature for 11/4 hours with stirring and then left to stand overnight.

The structure of 1'-methyl-3,5'-triphenyl-2,4,4 ', 6'-tetraoxo-spiro {pyrrolidine 1,2- (octahydropyrrolo [3,4-c] -pyrrole)} is given by the Proven NMR spectrum at 60 MHz in CDC13 solution. Reduction:

The presence of 31 hydrogen atoms is ensured by integrating the NMR spectrum. The product is thus 1'-methyl-3,5 ', 8'-triphenyl-spiro {pyrrolidine-1, 2' - (octahydropyrrolo [3,4-clpyrrole)).
Example 3
A mixture of 34.7 g (200 mM) N-phenyl-maleinimide, 16.3 g (100 mM) p-dimethyl-aminobenzylidene-methylamine and 250 ml. Abs. Xylene was refluxed for 10 hours. The precipitate which formed on cooling was separated off by filtration and gave 20.5 g of a crude product with a melting point of 181-2020C, from which 8.05 g of a substance with a melting point of 231-2350C were obtained by recrystallization from dioxane. Concentration of the first Nfutteilauge yielded a further 9.43 g of substance of melting point.
228-234 C (together thus 34 / o of the theory). Further recrystallization from ethanol gave a colorless powder with a melting point of 235-237.5 C.

Reduction according to the procedure described in Example 2 gave a colorless powder with a melting point of 160-162.5 ° C. (crystallized from cyolohexane).

Example 4
