Erfindungsgemäss gelangt man zu den Verbin dungen der Formel I, indem man a) eine Nitroverbindung der Formel II bzw. M

reduziert, vorzugsweise durch Behandeln mit einem komplexen Alkalimetallhydrid, z. B. Lithiumaluminiumhydrid, in einem inerte Lösungsmittel wie abs.
Dioxan, Tetrahydrofuran u. a. und bei erhöhter Temperatur, z. B. 50-100 . Die Herstellung des Aus gangsproduktes der Formel II bzw. III erfolgt aus einem 4- (Alkylthio)- oder 4- (Aralkylthio)-indol : Man führt es durch Behandelin, mit Phosphoroxychlorid und DimethyEormami'd in den entsprechenden Indol3-aldehyd über und kondensiert diesen mit einem Nitroalkan der Formel Ra-CHz-NDs, vorzugsweise ohne Lösungsmittel, aber in Gegenwart von Am moniumacetat, zur Nitroverbindung der Formel III.
Oder man setzt das oben genannte Indbl mit Formaldehyd und einem sekundären, aliphatischen oder cyclischen Amin, z. B. Dimethylamin, DiÅathylamin, Pyrrolidin, Piperidin, Morpholin usw., zu einer Mannich-Base um und erhitzt diese mit einem Nitroalkan der Formel R2-CH2-N02 in Gegenwart eines Protonenacceptors, vorzugsweise Natriumhydroxyd, wobei eine Nitroverbindung der Formel II entsteht. b) Erfindungsgemäss kann man auch ein Nitril der Formel IV

reduzieren, vorzugsweise mit einem komplexen Alkalimetallhydrid, z. B. mit Lithiumaluminiumhydrid in Ather, oder mit katalytisch erregtem Wasserstoff, z. B. in Gegenwart von Raney-Ni'ckel in am moniakalischem Methanol.
Die Herstellung der Nitrile der Formel III erfolgt aus einem. 4- (Alkylthio)- oder 4- (Aralkylthio)- indol : Man setzt es mit Formaldehyd und einem sekundären, aliphatischen oder cyclischen Amin, wie z. B. Dimethylamin, Diäthylamin, Pyrrolidin, Piperi- din, Morpholin usw., zu einer Mannich-Base um und verkocht diese, vorzugsweise nach Quartärisierung mit einem Alkylhalogenid, mit einer wässrigen Alkalicyanidlösung zum Nitril der Formel III.
Die gemäss vorliegender Erfindung erhaltenen Endprodukte sind in der Literatur bisher nicht be schrieben worden ; auch unbekannt waren bisher die Ausgangsprodukte. Die Verbindungen der Formel I sind bei Raumtemperatur feste, kristal3isierte Stoffe, die in Wasser schwer löslich sind, sich aber in d'en üblichen organischen Lösungsmitteln wie z. B. Alkohol, Ather, Aceton, Benzol, Chloroform, usw. leicht lösen. Mit anorganischen und organischen Säuren bilden sie bei Raumtemperatur beständige, kristalli- sierte Salze. Sie geben positive Farbreaktionen mit dem Keller-Reagens (Eisen-III-chlorid enthaltender Eisessig und konz. I-Schwefelsäure) und mit dem Van-Urk-Reagens (p-Dixneylarninobenzaldehyd und verd.
Schwefelsäure).
Die Verbindungen der Formel I zeichnen sich durch interessante-pharmakodynamische Eigenschaf- ten aus. So weisen viele davon bei der Prüfung am Tier zentral stimulierende Eigenschaften auf : Sie för- dern spinale Reflexe, wirken motorisch erregend und hemmen die sedative bzw. depressive Wirkung des Reserpins. Zudem stimulieren sie auch vegetative, insbesondere symphathische Funktionen und weisen einen gewissen Serotonin-Antagonismus auf.
Die neuen Indolderivate zeigen eine sehr geringe Toxizi- tät. Einige davon können zur Behandlung verschie denartigster psychischer Erkrankungen, wie vor allem von Zwangsneurosen sowie von Depressionen, Verstimmungen und Angstzuständen neurotischer und psychotischer Genese, verwendet werden. Sie werden vom Organismus praktisch quantitativ resorbiert und kommen daher vorzugsweise peroral zur Anwendung, können aber ebenso gut subcutan, intramuskulär oder intravenös verabreicht werden. Der Wirkstoff wird als freie Base oder in Form eines wasserlöslichen Salzes verabreicht.
Im FaNe der oralen Applikation kommen als Darreichungsformen vorzugsweise Tabletten oder Dragées in Frage, zu denen die Verfahrenserzeugnisse als Wirkstoffe mit den pharmazeutisch üblichen Trägerstoffen wie Milchzucker, Stärke, Stearinsäure, Talk, usw. verarbeitet werden. Ausserdem stellen die Substanzen wertvolle Zwischenprodukte zur Herstellung von Arzneimitteln dar.
In den folgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden ; die Schmelz-und Siedepunkte sind korrigiert.
Beispiel 1 3- (2-Amino-propyl)-4-methylthio-indol
Zu einer auf 50 erwärmten Lösung von 8 g Lithiumaluminiumhydrid in 140 cm3 abs. Tetrahydro- furan n tropft man eine Lösung von 4, 05 g 3- (2-Methyl- 2-nitrovinyl)-4-methylthio-indol in 85 cm3 abs. Tetrahydrofuran und rührt 5 Stunden bei 50 . Anschlie- ssend zersetzt man mit 30 cm3 gesättigter Natrium sulfattösung und dampft auf dem Wasserbad zur Trockne ein. Man schüttelt den Rückstand zwischen Äther und Weinsäurelösung aus.
Aus der weinsauren Lösung werden die basischen Bestandteile mit 2n Natronlauge in Frtiheit gesetzt und mit Chloroform extrahiert. Die Chloroformlösung wird über Kaliumcarbonat getrocknet und zur Trockne eingedampft.
Aus dem Rückstand erhält man durch Kristallisation aus Essigester oder Chloroform/Benzol das 3- (2- Amino-propyl)-4-methylthio-indol in Nadeln vom Smp. 143-145 .
Keller'sche Farbreaktion : dunkelblau, nach 5 Sek. lilaviolett.
Van Urk'sche Farbreaktion : schwach rosa.
Methansulfonat : Blättchen vom Smp. 227-229 aus Methanol.
Das als Ausgangsprodukt benötigte 3- (2-Methyl- 2-nitrovinyl)-4-methylthio-indol wird wie folgt hergestellt : 2 Nitro-6-amino-toluol wird bei 0 diazotiert und die Diazolösung bei pH 6 zu einem Gemisch von Methylmercaptan und verdünnter Natronlauge in Gegenwart von Kupferpulver gegeben.
Das entstandene 2-Nitro-6-methylthio-toluol siedet bei 130 / 0,2 mm Hg ; Smp. 56-58 . Eine Löshng von 2-Nitro6-methylthiotoluol und Oxalsäurediäthylester in Ather wird 10 Tage long bei Raumtemperatur mit Kalium äthylat stehengelassen und der ausgefallene Nieder- schlag zur Verseifung mit verdünnter Natronlauge bei Raumtemperatur geschüttelt, die gebilidete (2-Nitro- 6-methylthio-phenyl)-benztraubensäure kristallisiert aus Benzol in Drusen vom Smp. 107-109 . Zum Ringschluss wird die Verbindung in verdünnter Natronlauge unter Eiskühlung mit Natriumdithionit behandelt.
Beim Ansäuren fallt 4-Methylthio-indol 2- carbonsäure aus ; Nadeln vom Smp. 244-246 aus Athanol, Methanol/Benzol oder Athanol/Eisessig ; Keller'sche und Van Urk'sche Farbreaktion : negativ.
Obige Carbonsäure wird bei 260-270 /15 mm Hg decarboxyliert. Nach Destilliation bei 145-155 / 0,05 mm Hg schmilzt das erstarrte Destillat bei 44 bis 47 . Das so erhaltene 4-Methylmercapto-indol weist eine braun-violette Keller'sche und eine bläue, violettstichige Van Urk'sche Farbreaktion auf. 4 Methy3ithio-indol wird-in Dimethylformamid mit Phosphoroxychlorid zum 4-Methylthio-indol-3-aldehyd (Smp. 119-120 ) umgesetzt und dieser anschliessend mit Nitroäthan in Gegenwart von Ammoniumacetat zum 3- (2-Methyl-2-nitrovinyl)-4-methylthio-indol kondensiert ;
dunkelrote Prismen vom Smp. 171-173'aus Chloroform.
Beispiel 2 3- (2-Amino-propyl)-4-benzylthio-indol
Eine Lösung von 11,48 g 3- (2-Methyl-2-nitro- vinyl)-4-benzylthio-indol in 180 cm3 abs. Tetrahydrofuran wird bei 50 zu einer Lösung von 17,5 g LithiumaXuminiumhydrid in 250 cm3 abs. Tetrahydrofuran getropft und anschliessend noch 5 Stunden bei dieser Temperatur gerührt. Man zersetzt mit 70 cm3 Methanol und 90 cm3 gesättigter Natriumsulfatlösung und dampft auf dem Wasserbad zur Trockne ein. Man schüttelt den Rückstand zwischen Ather und Weinsäurelösung aus. Aus der weinsauren Lösung werden die basischen Bestandteile mit 2n Natronlauge in Freiheit gesetzt und mit Chloroform extrahiert. Die Chloroformlösung wird über Kaliumcarbonat getrocknet und zur Trockne eingedampft.
Aus dem Rückstand erhält man durch Kristallisation aus Chloroform/Benzol das 3- (2-Amino-propyl)-4- benzylthio-indol in Nädelchen vom Smp. 156-158 .
Keller'sche Farbreaktion : hellrot, später bräunlichrot.
Van Urk'sche Farbreaktion : negativ.
Das als Ausgangsprodukt benötigte 3- (2-MethylL 2-nitrovinyl)-4-benzylthio-indol wird wie folgt hergestellt :
Eine diazotierte Lösung von 2-Nitro-6-amino- toludl wird bei 0 zu einem Gemisch von Benzylmercaptan, verdünnter Natronlauge und Kupferpul- ver in Wasser gegeben. Das entstandene 2-Nitro-6 benzylthio-tol, uol schmilzt bei 106-108 nach Kristallisation aus Chloroform. Eine Lösung von 2-Nitro6-benzylthio-toluol und Oxa'laurediäthylester in abs.
Tetrahydrofuran wird 12 Tage mit Kaliumäthylat stehengelassen. Nach Aufarbeitung wird der Athylester zur Verseifung mit wässriger Natronlauge behandelt ; beim Ansäuren erhält man die rohe (2 Nitro- 6-benzylthio-phenyl)-brenztraubensäure als O±. Man versetzt sie zum Ringschluss in alkalischer Lösung mit Natriumdithionit und Natronlauge bei 0-5 und'er hält beim Ansäuern 4-Benzylthio-indbl-2-carbon- säure ; Smp. 191-192 nach Kristallisation ajus Atha nos und Athanol,/Eisessig. Farbreaktion nach Keller : resedagrün, nach Van Urk : negativ.
Obige Carbonsäure wird bei 210 /15 mm Hg decarboxyliert. Nach Destillation bei 155 /0, 02 mm Hg schmilzt das erstarrte Destillat bei 32-35 . Das so erhaltene 4 Benzylthio-indol weist vor dem Schütteln eine violette, nachher eine braune Keller'sche Farbreaktion auf ;
Van Urk'sche Farbreaktion : trüb vioWbSt. Durch Umsetzung von 4-Benzylthio-indol mit Phosphoroxychlorid in Dimethylformamid erhält man 4-Benzyl thio-indol-3-aldehyd (Smp. 164-166 ) ; die anschlie ssende Kondensation mit Nitroäthan in Gegenwart von Ammoniumacetat ergibt 3- (2-Methyl-2-nitro- viny0-4-benzyl, thio-indol, dunkeZrote Prismen vom Smp. 160-162 aus Chloroform/Athanol.
Beispiel 3 3- (2-Aminoälthyl)-4-benzylthio-indol
In eine auf 50 erwärmte Lösung von 1,6 g Lithiumaluminiumhydrid und 25 cm3 abs. Tetrahydrofuran tropft man eine Lösung von 1,55 g 3- (2 Nitrovinyl)-4-benzylthio-indol in 30 cm3 abs. Tetrahydrofuran und rührt 3 Stunden bei 50 . Anschie ssend zersetzt man mit 15 cm3 Methanol und 40 cm3 gesättigter Natriumsutfatlösung und dampft auf dem Wasserbad zur Trockne ein. Man nimmt den Rück- stand in Wasser auf, filtriert und schüttelt d'as Filtrat zwischen Äther und Weinsäurelösung aus.
Aus der weinsauren Lösung werden die basischen Bestandteile mit 2n Natronlauge in Freiheit gesetzt und mit Chloroform extrahiert. Die Chloroformlösung wird über Kaliumcarbonat getrocknet und zur Trockne eingedampft. Aus dem Rückstand erhäl't man d'as 3-(2- Aminoäthyl)-4-benzylthio-indot in Prismen vom Smp. 120-122 durch Kristallisation aus Benzol.
Keller'sche Farbreaktion : dunkelorange.
Van Urk'sche Farbreaktion : negativ.
Das Ausgangsprodukt, 3- (2-Nitrovinyl)-4-benzyl- thio-indol, erhält man durch Umsetzung von 4-Ben zyfthio-indo mit Phosphoroxychlorid in Dimethyl- formamid zum 4-Benzylthio-indol-3-aldehyd (Smp.
164-166 ) und anschliessendeKondensation mitNitro- methan in Gegenwart von Ammoniumacetat zum 3 (2-Nitrovinyl)-4-benzylthio-indol ; Smp. 171-172 .

The compounds of the formula I are distinguished by interesting pharmacodynamic properties. When tested on animals, many of them have centrally stimulating properties: They promote spinal reflexes, stimulate motor activity and inhibit the sedative or depressive effect of reserpin. In addition, they also stimulate vegetative, in particular sympathetic, functions and have a certain serotonin antagonism.
The new indole derivatives show a very low toxicity. Some of them can be used to treat various mental illnesses, such as obsessive-compulsive disorder and depression, moods and anxiety of neurotic and psychotic origins. They are absorbed practically quantitatively by the organism and are therefore preferably used orally, but can just as easily be administered subcutaneously, intramuscularly or intravenously. The active ingredient is administered as a free base or in the form of a water-soluble salt.
In the case of oral administration, the preferred dosage forms are tablets or dragees, into which the process products are processed as active ingredients with the usual pharmaceutical carriers such as lactose, starch, stearic acid, talc, etc. In addition, the substances are valuable intermediates in the manufacture of drugs.
In the following examples, all temperatures are given in degrees Celsius; the melting and boiling points are corrected.
Example 1 3- (2-Amino-propyl) -4-methylthio-indole

The 3- (2-amino-propyl) -4-methylthio-indole is obtained from the residue by crystallization from ethyl acetate or chloroform / benzene in needles with a melting point of 143-145.
Keller's color reaction: dark blue, after 5 seconds purple-violet.
Van Urk's color reaction: pale pink.
Methanesulfonate: flakes with a melting point of 227-229 from methanol.
The 3- (2-methyl-2-nitrovinyl) -4-methylthio-indole required as a starting product is prepared as follows: 2 nitro-6-aminotoluene is diazotized at 0 and the diazo solution at pH 6 to a mixture of methyl mercaptan and given dilute sodium hydroxide solution in the presence of copper powder.
The resulting 2-nitro-6-methylthio-toluene boils at 130 / 0.2 mm Hg; M.p. 56-58. A solution of 2-nitro6-methylthiotoluene and diethyl oxalate in ether is left to stand for 10 days at room temperature with potassium ethylate and the precipitate which has precipitated is shaken with dilute sodium hydroxide solution at room temperature for saponification, the (2-nitro-6-methylthio-phenyl) pyruvic acid crystallizes from benzene in drusen with a melting point of 107-109. To close the ring, the compound is treated with sodium dithionite in dilute sodium hydroxide solution while cooling with ice.
With acidification, 4-methylthio-indole 2-carboxylic acid precipitates; Needles with a melting point of 244-246 made from ethanol, methanol / benzene or ethanol / glacial acetic acid; Keller's and Van Urk's color reaction: negative.
The above carboxylic acid is decarboxylated at 260-270 / 15 mm Hg. After distillation at 145-155 / 0.05 mm Hg, the solidified distillate melts at 44 to 47. Example 2 3- (2-Amino-propyl) -4-benzylthio-indole

The 3- (2-MethylL 2-nitrovinyl) -4-benzylthio-indole required as the starting product is produced as follows:
A diazotized solution of 2-nitro-6-aminotoludl is added at 0 to a mixture of benzyl mercaptan, dilute sodium hydroxide solution and copper powder in water. The resulting 2-nitro-6-benzylthio-tol, uol melts at 106-108 after crystallization from chloroform. A solution of 2-nitro6-benzylthio-toluene and Oxa'laurediethylester in abs.
Tetrahydrofuran is left to stand with potassium ethylate for 12 days. The above carboxylic acid is decarboxylated at 210/15 mm Hg. After distillation at 155/0.02 mm Hg, the solidified distillate melts at 32-35. The 4-benzylthio-indole obtained in this way shows a violet color reaction before shaking and a brown Keller color reaction afterwards;
Van Urk's color reaction: cloudy vioWbSt. The reaction of 4-benzylthio-indole with phosphorus oxychloride in dimethylformamide gives 4-benzylthio-indole-3-aldehyde (melting point 164-166); the subsequent condensation with nitroethane in the presence of ammonium acetate gives 3- (2-methyl-2-nitroviny0-4-benzyl, thio-indole, dark-red prisms with a melting point of 160-162 from chloroform / ethanol.
Example 3 3- (2-Aminoethyl) -4-benzylthio-indole
In a heated to 50 solution of 1.6 g lithium aluminum hydride and 25 cm3 abs. Tetrahydrofuran is added dropwise to a solution of 1.55 g of 3- (2 nitrovinyl) -4-benzylthio-indole in 30 cm3 of abs. Tetrahydrofuran and stir for 3 hours at 50. Then it is decomposed with 15 cm3 of methanol and 40 cm3 of saturated sodium sulfate solution and evaporated to dryness on a water bath. The residue is taken up in water, filtered and the filtrate is shaken out between ether and tartaric acid solution.
The basic components are released from the tartaric solution with 2N sodium hydroxide solution and extracted with chloroform. The chloroform solution is dried over potassium carbonate and evaporated to dryness. The residue is obtained as 3- (2-aminoethyl) -4-benzylthio-indote in prisms of melting point 120-122 by crystallization from benzene.
Keller's color reaction: dark orange.
Van Urk's color reaction: negative.
The starting product, 3- (2-nitrovinyl) -4-benzyl-thio-indole, is obtained by reacting 4-benzyfthio-indo with phosphorus oxychloride in dimethylformamide to give 4-benzylthio-indole-3-aldehyde (mp.
164-166) and subsequent condensation with nitro methane in the presence of ammonium acetate to give 3 (2-nitrovinyl) -4-benzylthio-indole; M.p. 171-172.
