On peut préparer le stéroïde de départ, de formule (II), en faisant réagir la 16a-hydroxycortéxolone avec un aldéhyde d'au moins 2 atomes de carbone ou une cétone contenant les restes P et Q.
On effectue la réaction de préférence en traitant une suspension ou une solution du stéroïde dans l'aldéhyde ou la cétone (ou dans un solvant organique si l'aldéhyde ou la cétone est un solide) avec un catalyseur acide (par exemple l'acide perchlorique, l'acide paratoluènesulfonique et l'acide chlorhydrique), en neutralisant l'acide et en recueillant l'acétal ou le cétal cyclique ainsi formé. Cette réaction, ainsi que des exemples représentatifs d'aldéhydes et de cétones de départ appropriés, est décrite plus en détail dans le bre vet USA No 3048581.
Les acétals ou cétals de départ (composés I) ainsi formés sont ensuite soumis à l'action de micro-organis mes 11@-hydroxylants, ce qui produit les dérivés 11(3- hydroxylés (composés III) correspondants.
II est préférable d'employer un micro-organisme du genre Absidia, par exemple A. coerulea, etc., du genre Curvularia, par exemple C. lunata, etc. ou du genre Cunninghamella, par exemple C. blakesleeana, C. ele- gans, etc.
Ensuite, on acyle les composés de formule (III), par exemple en les traitant avec de l'anhydride acétique dans de la pyridine, pour obtenir les dérivés 21-acylés (com posés IV).
Ensuite, on déshydrate ces dérivés acylés, par exem ple en les traitant avec de la pyridine et du chlorure de méthane sulfonyle dans du diméthylformamide, pour pro duire les dérivés 9,11-déhydro finals (composés VII).
La réaction des composés VII avec du N-bromoacé- tamide ou du N-bromosuccinimide en présence de di- oxane et d'acide perchlorique, la réaction étant conduite à l'obscurité, fournit la bromohydrine (composés VIII).
La débromhydratation des composés VIII, par exem ple par traitement avec de l'acétate de potassium frai- chement fondu dans l'éthanol absolu au reflux, fournit le dérivé 9@,11@-époxy (composés IX), que l'on ouvre par traitement avec de l'acide fluorhydrique, obtenant ainsi les dérivés de triamcinolone (composés X) que l'on peut hydrolyser de manière connue pour obtenir les composés 21-hydroxylés correspondants de formule (XI).
On peut également déshydrogéner les composés de formule (X) en position 1,2 par réaction avec une quinone ayant un po tentiel redox suffisamment élevé, telle que la 2,3-dichloro- 5,6-dicyano-benzoquinone, pour former les dérivés 1-dé- hydro correspondants.
On peut préparer les composés de formule (I), dans laquelle Y représente une double liaison, à partir des composés de formule (1), dans laquelle Y représente une liaison simple, par diverses méthodes, par exemple par traitement avec les enzymes d'un micro-organisme 1-dés- hydrogénant tels que, par exemple, Corynebacterium simplex, Nocardia restrictus ou Mycobacterium rho- dochrous.
On peut d'autre part acyler les composés de for mule (I), dans laquelle Y est une liaison simple, comme décrit ci-dessus pour préparer le dérivé 21-acylé, et dés hydrogéner ce dernier par traitement avec une quinone ayant un potentiel redox suffisamment élevé, par exemple la 2,3-dichloro-5,6-dicyano-benzoquinone pour préparer le dérivé 1,2-déhydro correspondant. Ensuite, en suivant la série de réactions indiquées dans le schéma de réac tions ci-dessus, mais en employant ce dérivé 1,2-déhydro, on prépare les composés de formules (II) à (XI), dans lesquelles Y représente une double liaison.
Les produits finals de formule (XI) sont bien connus en tant que substances physiologiquement actives possé dant une activité glucocorticoïde et anti-inflammatoire.
Les radicaux acyles préférés sont ceux d'acides car boxyliques hydrocarbonés de moins de 12 atomes de carbone, comme par exemple les acides alcanoïques infé rieurs (tels que les acides formique, acétique, propioni- que, butyrique, valérique, triméthylacétique et caproïque), les acides alcénoïques inférieurs (tels que les acides acry lique, méthacrylique, crotonique, 3-buténoïque et séné- cioïque), les acides arylcarboxyliques monocycliques (tels que les acides benzoïque et toluique), les acides arylalca- noïques inférieurs monocycliques (tels que l'acide phéna- cétique,
l'acide @-phénylpropionique, l'acide a-phényl- butyrique et l'acide 5-(p-méthylphényl)pentanoïque), les acides cycloalcoyles carboxyliques (tels que l'acide cyclo- butane carboxylique, l'acide cyclopentane carboxylique et l'acide cyclohexane carboxylique), les acides cyclo- alcènes carboxyliques (tels que l'acide 2-cyclobutène car boxylique et l'acide 3-cyclopentène carboxylique), les acides cycloalcoyles- et cycloalcényles-alcanoïques infé rieurs (tels que les acides cyclohexane acétique, a-cyclo- pentane butyrique,

L'expression alcoyle inférieur telle qu'utilisée dans cet exposé désigne les radicaux à chaîne droite ou rami fiée de moins de 8 atomes de carbone, par exemple méthyle, éthyle, propyle, butyle, pentyle, hexyle, heptyle, octyle, isopropyle, t-butyle, isobutyle, isohexyle, 4,4-di- méthylpentyle, 2,2,4-triméthylpentyle, etc.

Les expressions cycloalcoyle monocyclique et cycloalcényle monocyclique comprennent les radicaux cycliques à noyaux de 3 à 6 chaînons (par exemple cyclo- propyle, cyclobutyle, cyclopentyle, cyclohexyle, cyclo- butényle et cyclohexényle).
On effectue l'hydroxylation et la déshydrogénation enzymatiques de préférence en incorporant le stéroïde de départ à une culture du micro-organisme en aérobie ou en mettant en contact, dans un milieu aqueux, le stéroïde, de l'air et le micro-organisme. De manière géné rale, les conditions de culture des micro-organismes pour les buts de l'invention sont (à l'exception de l'incorpo ration du stéroïde à transformer) les mêmes que celles de la culture de diverses autres moisissures et bactéries utilisées dans les procédés de fermentation.
On cultive le micro-organisme aérobiquement en con tact avec (dans ou sur) un milieu de fermentation appro prié. Un milieu approprié comprend essentiellement une substance azotée et une source de carbone et d'énergie. Cette dernière peut être un hydrate de carbone (par exemple saccharose, mélasse, glucose, maltose, amidon ou dextrine), un acide gras, une graisse et/ou le stéroïde lui-même. Cependant il est préférable que le milieu con tienne une source assimilable de carbone et d'énergie en plus du stéroïde.
La source de facteurs azotés peut être naturelle (par exemple farine de soja, corn steep, extrait de viande et/ou résidus solubles de distillerie) ou syn thétique (c'est-à-dire consistant en composés organiques ou inorganiques simples et synthétisables, tels que les sels d'ammonium, les nitrates alcalins, les acides aminés ou l'urée). Une alimentation suffisante en air stérile doit être maintenue pendant la fermentation, par exemple par les méthodes classiques consistant à exposer une grande sur face du milieu à l'air ou par culture aérée submergée. Le stéroïde peut être ajouté à la culture pendant la période d'incubation ou peut être incorporé au milieu avant la stérilisation ou l'ensemencement.
L'intervalle de concentration préfédé du stéroïde dans la culture est d'environ 0,01 à 0,10 % en poids. La durée de culture peut être très variable, par exemple entre 6 et 96 heures: Le stéroïde est ensuite isolé du milieu de fermentation de la manière usuelle, comme décrit plus en détail dans les exemples ci-après.
Dans ce qui suit, toutes les températures sont en degrés centigrades.
Préparations de stéroïdes de départ A. 16,17-acétonide de 16a,17a,21-trihydroxy- pregn-4-ène-3,20-dione On laisse reposer une solution de 1,0 g de 16a,17a,21- trihydroxypregn-4-ène-3,20-dione dans 50 ml d'acétone contenant 0,25 ml d'acide perchlorique à 70 % à la tem pérature ordinaire pendant 1 heure. Ensuite, on neutra lise la solution avec du bicarbonate de sodium dilué et on la dilue progressivement avec de l'eau.

Après environ 72 heures d'incubation poursuivie dans des conditions identiques à celles décrites ci-dessus, on rassemble les contenus des fioles. Le volume final du moût est d'environ 1000 ml. On extrait le moût trois fois avec des portions de 300 ml de chloroforme, que l'on réunit, lave à l'eau et évapore sous pression réduite. Une cristallisation du résidu dans de l'acétone-hexane fournit le 16,17-acétonide de 16a,17a,21-trihydroxypregna-1,4- diène-3,20-dione.
b) En partant du 16,17-acétonide de 16a,17a,21- trihydroxypregn-4-ène-3,20-dione On prépare des cellules séchées à l'acétone de Coryne- bacterium simplex (ATCC-6946) par la méthode décrite dans le brevet USA No 3360439.
A chacune de 2 fioles Erlenmeyer de 250 ml conte nant 50 ml de tampon au phosphate 0,05 M (pH 7,0), on ajoute 1 g de Corynebacterium simplex (ATCC- 6946), cellules séchées à l'acétone. Ensuite, on secoue énergiquement le mélange pour distribuer les cellules, après quoi on ajoute 3,44 mg de 2-méthyl-1,4-naphto- quinone dans 0,25 ml d'éthanol, comme accepteur d'hy drogène, de manière que la concentration finale soit de 0,4 mM. Ensuite, on ajoute aux fioles le stéroïde, en ajoutant à chaque fiole 0,5 ml d'une solution (100 mg/ml) de 16,17-acétonide de 16a-hydroxycortexolone dans du N,
N-diméthylformamide. On utilise un total de 100 mg de stéroïde.
Après environ 21 h d'incubation à 300 C avec agita tion rotative continue (280 cycles/minute ; 5 cm de course), on réunit les contenus des fioles et on ajuste le mélange au pH 4,0 avec du H2S04 12 N. Ensuite, on filtre le mélange réactionnel acidulé à travers un tampon clarifiant de Seitz. On lave les fioles et le tampon avec 50 ml d'eau tiède. Le filtrat et les eaux de lavage réunis ont un volume de 150 ml. On extrait le tampon filtrant avec de l'acétone en utilisant des portions succesives de 20 ml d'acétone. On recueille un total de 100 ml d'extrait dans l'acétone. On élimine partiellement l'acétone sous pression réduite et on extrait les filtrats et l'extrait réunis avec du chloroforme.
On lave bien l'extrait chlorofor- mique à l'eau, on l'évapore et on cristallise le résidu dans de l'acétone-hexane, obtenant ainsi le 16,17-acéto- nide de 16a,17a,21-trihydroxypregna-1,4-diène-3,20- dione.

On fait fermenter un total de 300 mg.
Après avoir poursuivi l'incubation pendant environ 26 h dans des conditions identiques à celles décrites ci- dessus, on réunit les contenus des fioles et on filtre le moût à travers un tampon clarifiant de Seitz. On lave les fioles, le mycélium et le tampon avec des portions successives de 100 ml d'eau tiède. Le filtrat et les eaux de lavage réunis ont un volume de 1200 ml. On les extrait trois fois avec des portions de 400 ml de chloroforme que l'on réunit, lave à l'eau et évapore sous pression réduite.

Après avoir poursuivi l'incubation pendant environ 5 jours dans des conditions identiques à celles décrites ci-dessus, on réunit les contenus des fioles. Le volume final du moût est d'environ 500 ml. On extrait le moût avec 3 portions de 150 ml de chloroforme, que l'on réunit, lave à l'eau et évapore sous pression réduite. Une cristallisation du résidu donne 200 mg de 16,17-acéto- nide de 16a-hydroxyprednisolone.
c) En partant du 16,17-acétonide de 16a- hydroxyhydrocortisone On prépare des cellules de Corynebacterium simplex (ATCC-6946) séchées à l'acétone, par la méthode décrite dans le brevet USA No 3360439.
Dans chacune de 5 fioles d'Erlenmeyer de 250 ml, contenant 50 ml de tampon au phosphate 0,05 M (pH 7,0), on introduit 1 g de cellules séchées à l'acétone de Corynebacterium simplex (ATCC-6946). On secoue éner giquement le mélange pour distribuer les cellules. Ensuite, on ajoute 3,44 mg de 2-méthyl-1,4-naphtoqui- none, comme accepteur d'hydrogène, dans 0,25 ml d'éthanol, afin que la concentration finale soit de 0,4 mM. On ajoute alors aux fioles le stéroïde en introduisant dans chaque fiole 0,5 ml d'une solution (à<B>100</B> mg/ml) de 16,17-acétonide de 16a-hydroxyhydrocortisone dans du N,N-diméthylformamide. On utilise un total de 250 mg de stéroïde.
Après environ 17 h d'incubation à 300 C sous agita tion rotative continue (280 cycles/mn ; 5 cm de course), on réunit les contenus des fioles et on ajuste le mélange réactionnel au pH 3,5 au moyen de H2S04 12 N. Ensuite, on filtre le mélange acidulé à travers un tampon clarifiant de Seitz. On lave les fioles et le tampon avec des por tions successives de 50 ml d'eau tiède. Le filtrat et les eaux de lavage réunis ont un volume de 500 ml. Ensuite, on extrait le tampon filtrant avec des portions successives de 100 ml d'acétone. On recueille un total de 500 ml d'extrait acétonique. On chasse partiellement l'acétone sous pression réduite et on extrait les filtrats réunis avec du chloroforme.
On lave l'extrait chloroformique à l'eau, on l'évapore et on cristallise le résidu dans de l'acétone- hexane, obtenant ainsi le 16,17-acétonide de 16(x-hy- droxyprednisolone.
<I>Exemple 3:</I> 16,17-acétonide 21-acétate de 16a-hydroxyhydrocortisone On conserve une solution de 214 mg de 16,17-acéto- nide de 16a-hydroxyhydrocortisone dans 3 ml de pyri- dine sèche et 1 ml d'anhydride acétique à la température ordinaire pendant 16 h, puis on la dilue avec de l'eau glacée et on l'extrait au chloroforme. On lave l'extrait chloroformique successivement avec de l'acide chlorhy drique 2 N, du bicarbonate de sodium à 5 % et de l'eau et on l'évapore sous pression réduite.
Par cristallisation du résidu dans de l'acétone-hexane, on obtient 200 mg du produit désiré, ayant un point de fusion d'environ 244-246o C ;

To each of 2 250 ml Erlenmeyer flasks containing 50 ml of 0.05 M phosphate buffer (pH 7.0), 1 g of Corynebacterium simplex (ATCC-6946), acetone-dried cells is added. Then, the mixture is vigorously shaken to distribute the cells, after which 3.44 mg of 2-methyl-1,4-naphthoquinone in 0.25 ml of ethanol is added, as a hydrogen acceptor, in such a manner. that the final concentration is 0.4 mM. Then the steroid is added to the vials, adding to each vial 0.5 ml of a solution (100 mg / ml) of 16,17-acetonide of 16α-hydroxycortexolone in N,
N-dimethylformamide. A total of 100 mg of steroid is used.
After about 21 h of incubation at 300 C with continuous rotary shaking (280 cycles / minute; 5 cm stroke), the contents of the vials are combined and the mixture is adjusted to pH 4.0 with 12 N H2SO4. , the acidulated reaction mixture is filtered through a Seitz clarifying buffer. The vials and buffer are washed with 50 ml of lukewarm water. The combined filtrate and washings have a volume of 150 ml. The filter pad was extracted with acetone using 20 ml successive portions of acetone. A total of 100 ml of acetone extract is collected. The acetone is partially removed under reduced pressure and the combined filtrates and extract are extracted with chloroform.
The chloroform extract is washed well with water, evaporated and the residue crystallized from acetone-hexane, thereby obtaining 16,17-acetone of 16a, 17a, 21-trihydroxypregna-. 1,4-diene-3,20-dione.

c) Starting with 16a-hydroxyhydrocortisone 16,17-acetonide Acetone-dried Corynebacterium simplex (ATCC-6946) cells are prepared by the method described in US Pat. No. 3360439.
Into each of 5 250 ml Erlenmeyer flasks, containing 50 ml of 0.05 M phosphate buffer (pH 7.0), is introduced 1 g of acetone dried cells of Corynebacterium simplex (ATCC-6946). The mixture is shaken vigorously to distribute the cells. Then, 3.44 mg of 2-methyl-1,4-naphthoquinone, as a hydrogen acceptor, in 0.25 ml of ethanol is added, so that the final concentration is 0.4 mM. The chloroform extract is washed with water, evaporated and the residue crystallized from acetone-hexane, thereby obtaining 16,17-acetonide 16 (x-hydroxyprednisolone.
<I> Example 3: </I> 16,17-acetonide 16α-hydroxyhydrocortisone 21-acetate A solution of 214 mg of 16α-hydroxyhydrocortisone 16,17-acetonide in 3 ml of dry pyridine is stored and 1 ml of acetic anhydride at room temperature for 16 h, then diluted with ice water and extracted with chloroform.
