Von diesen Verbindungen sind insbesondere dieje nigen bevorzugt, bei denen X' eine Alkoxygruppe, ins besondere eine Niederalkoxygruppe bedeutet. Die Alk oxygruppen können bis zu 18 Kohlenstoffatomen in gerader oder verzweigter Kette aufweisen, wobei Gruppen mit bis zu 6 Kohlenstoffatomen bevorzugt sind.
Unter Aralkoxygruppen sind insbesondere solche mit bis zu s Kohlenstoffatomen in der Alkylkette, die gec rade oder verzweigt sein kann, zu verstehen.
Cyclische 3':5'-Phosphate von Nucleosiden gewinnen in der Biochemie laufend an Bedeutung und Aden osin-3':5'-cyclophosphat ist seit seiner Entdeckung durch Sutherland vor 10 Jahren (Sutherland, Rall, J.Amer. Chem.Soc. 79, 3608 [1957]) zu einer Schlüsselsubstanz in der Glykolyse, der Lipolyse und bei Hormonstoffwechselwirkungen geworden, deren physiologische Bedeutung vielfach mit dem wichtigsten Energieüberträger ATP in der Zelle gleichgesetzt wird.
Nach der Theorie von Sutherland fällt dem Adenosin3':5'-cyclophosphat physiologisch in der Zelle die Funktion eines second messenger zu, was am Beispiel der Wirkung des Peptidhormons ACTH kurz veranschaulicht sei. Von der Hypophyse ausgeschüttetes ACTH gelangt über die Blutbahn an die Nebennieren- rinde, in deren Zellwand das Enzym Adenylcyclase sitzt, die auf das extracelluläre ACTH-Signal hin aus Adenosintriphosphat intracellulär-Adenosin-3': 5'- cyclophosphat bildet, das nun seinerseits die Biosynthese und Ausschüttung von Corticosteroiden bewirkb.
Ähnlich liegen die Verhältnisse bei der Glykogenolyse, Lipolyse usw. Die Selektivität ist also durch die Rezeptoren der jeweiligen Erfolgsorgane gegeben, die Adenosin-3':5'-cyclophosphat als intracelluläres Signal haben; Adenosin-3':5'-cyclophosphat von aussen appliziert, hat jedoch multiple Wirkungen, was pharmakologisch oft unerwünscht ist. Ausserdem wird es im Organismus verhältnismässig rasch gespalten, so dass seine Wirkungsdauer gering ist.
Es besteht daher ein Bedürfnis nach Schaffung von Adenosin-3':5'-cyclophosphatanalogen mit verbesserter Spezifität der Wirkung und längerer Wirkungsdauer.
Die Synthese von modifizierten Analogen von Adenosin-3':5'-cyclophosphatderivaten erfolgte bisher auf sehr umständiliche und aufwendige Weise, die am Beispiel des 6-Mercaptopurinribonucleosid-3' :5'-cyclo- phosphates illustriert sei:
Inosin + Überführung in 2',3',5'-Tribenzoylinosin Reaktion mit P2S5 zum 6-Mercaptopurinribonucleosid + Entfernung der Schutzgruppe und Einführung einer neuen Schutzgruppe an 2',3'-Hydroxylgruppe (z.B.
durch Ketalisierung), wobei die 5'-OH Stellung freibleibt + Phosphorylierung am 5'OH mit ss-Cyanoäthylphosphat und DCC als Kondensationsmittel + Abspaltung a) von 2',3'-OH-Schutzgruppe, b) von Phosphat-Schutzgruppe + Cyclisierung zum gewünschten Produkt.
Die Ausbeuten bei dieser vielstufigen Synthese sind sehr niedrig (1,50/0 der Theorie; J. Thomas, Montgo mery J. Med. Pharm. Chem. 11, 44 (1968)), zumal die hydrolyse- und oxydationsempfindliche Mercaptogruppe von Anfang an mit durch die einzelnen Reaktionsstufen gezogen werden muss.
Auf analogem Wege wurden diverse Derivate von Deazapurinrihonucleosid-3':5'-cyclophosphat (Tubercidin) hergestellt (A. Hanze, Upjohn, USA-Patentschrift 3 300479; A. Hanze, Biochemistry 7, 932 (1968)); ebenso Cytosinarabinosid-2': 5'-cyclophosphat.
Für ein industriell anwendbares Verfahren schied die obige Herstellungsweise aufgrund ihrer Umständlichkeit und geringen Ausbeute aus. Es wurde daher ein Weg gesucht, von der präformierten Cyclophosphatverbindung ausgehend die Substitution in Stellung 6 des Puringerüstes durchzufiihren.
Folgende Verfahren zur Umwandlung der Hydroxylgruppe in die Chlorgruppe bei gewöhnlichen Nucleosiden sind bekannt:
1. Chlor in Methanol (G.D. Daves et al., J. Amer.

2. konzentrierter Salzsäure, Chlorgas und Methanol (Gerster et al., J. Org. Chem. 28, 945, (1963)),
3. Diäthylanilin und POC13 (Gerster et al., J. Org.
Chem. 28, 945, (1963)),
4. SoCl-DMF (M. Ikehara, Chem. Pharm. Bull.
12, 267, (1964)).
Es wurde jedoch gefunden, dass keines dieser bekannten Verfahren bei Anwendung auf das SHydroxypurinribonuoleosid-3 ':5'-cyclopho6phat zu dem gesuchten Produkt führt. Die Versuche ergaben, dass hierbei entweder eine extensive Degradation auftrat oder das Ausgangsmaterial überhaupt nicht umgesetzt wurde. Dies zeigt, dass die Reaktionsfähigkeit bei Einführung einer Cyclophosphatgruppe auch im Purinkern grundsätzlich verändert wird und daher die für die Purinnucleoside bekannten Verfahren sich nicht auf die cyclischen Nucleosid-3' :5'-phosphate übertragen lassen.
Das gleiche gilt auch für die in der Nucleosidreihe bekannte Umwandlung von Inosin in das entsprechende 6-Mercaptoderivat durch Umsetzung mit P,tS. Am präformierten Cyclophosphat ist diese mit den Nucleosiden glatt verlaufende Reaktion nicht durchführbar und führt unter keinen Bedingungen zum gewünschten 6-Mercaptoderivat.
In der japanischen Patentschrift 7957/68 wurde bereits die Herstellung von 6-Chlorpurin-desoxyribosid-3' :5'-cyclophosphat beschrieben durch Umsetzung mit Phosphortrichlorid und einer alkalischen Substanz, wie einem anorganischen Alkali oder aliphatischen oder aromatischen Amin. Dieses bekannte Verfahren führt zu einer Ausbeute von 260io.
Es wurde jedoch festgestellt, dass sich diese Umsetzung nicht beim gewöhnlichen 6-Hydroxypurinribonucleosid-3': 5'-cyclophosphat durchführen lässt. In Gegenwart von Basen wie z. B.
Diäthylanilin ergibt sich unter den Bedingungen des bekannten Verfahrens keine Umwandlung und das Reaktionsmedium färbt sich schwarz-braun und enthält zahlreiche Zersetzungsprodukte.
Überraschenderweise wurde nun gefunden, dass die gewünschte Umsetzung glatt und mit sehr guter Ausbeute verläuft, wenn man ein 6-Hydroxypurinribonucleosid-3': 5'-cyclophosphat als Ausgangsmaterial acyliert und das Acylierungspro- dukt direkt mit Phosphoroxyhalogenid umsetzt
Man erhält auf diese Weise das entsprechende Halogenpurmribonucleosid-3' :5'-cyclophosphat in über 50 /Oiger Ausbeute.
Die Verbindungen werden daher erfindungsgemäss hergestellt, indem man eine entsprechende Verbindung, in der X eine Hydroxylgruppe bedeutet, in Gegenwart einer Base mit einem Acylierungsmittel umsetzt, nicht umgesetztes Acylierungsmittel und Base entfernt und den Rückstand mit überschüssigem Phosphoroxyhalogenid umsetzt.
Als Ausgangsprodukt für das Verfahren der Erfindung wird ein 6-Hydroxypurinribonudeosid-3': 5'-cyclophosphat der Formel

worin R ein Wasserstoffatom, eine Hydroxyl- oder Aminogruppe bedeutet, verwendet. Dieses Ausgangsprodukt lässt sich leicht durch chemische Desaminierung von beispielsweise Adenosin-3': 5'-cyclophosphat in hoher Ausbeute erhalten.
Vorzugsweise wird mit Phosphoroxychlorid das 6-Chlorderivat bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur hergestellt. Die Überführung des 6-Halogenderivates in eine Verbindung der Formel 2 erfolgt dann durch Umsetzung mit überschüssigem Alkoholat in wässriger oder alkoholischer Lösung bei einer Temperatur zwischen -10 C und Rückflusstemperatur.
Das Ausgangsprodukt wird dabei in der ersten Stufe des erfindungsgemässen Verfahrens acyliert, um es im Phosphoroxyhalogenid löslich zu machen. Die Acylierung muss nur in solchem Ausmass durchgeführt werden, dass das acylierte Produkt ausreichend löslich für die Umsetzung in POHals ist. Vorzugsweise wird die Acylierung mit einem Acylchlorid oder Acylanhydrid in Gegenwart eines alkalischen Mittels durchgeführt. Bevorzugt erfolgt die Acylierung mit Acetylchlo- rid, Essigsäureanhydrid oder Benzoylchlorid. Als alkalisches Mittel werden vorzugsweise organische Basen verwendet, die sich zusammen mit überschüssigem Acylierungsmittel leicht durch Destillation vom acylierten Nucleosid abtrennen lassen.
Als gut geeignet erwies sich die Umsetzung mit Essigsäureanhydrid in Pyridin, die unschwierig auch bei Raumtemperatur durchgeführt werden kann. Hierbei entsteht das 6-Hydroxypum-2'-O;acetylribo- nucleosid-3': 5'-cycloacetylphosphat, welches in POHal3 gut löslich ist und sich daher für die Umsetzung besonders eignet. Das 2'-Acetyl-6-hydroxypurinr,ibo- nucleosid3': 5'-cyclophosphat eignet sich ebenfalls, jedoch ist die Ausbeute infolge der geringeren Löslichkeit schlechter.
Nach beendeter Acylierung werden restliches Acy lierungsmitteil und Base entfernt, vorzugsweise durch Destillation. Das zurückbleibende ölige Acylierungsprodukt wird direkt mit überschüssigem POHal5 umgesetzt. Hierbei dient das POHals sowohl als Lösungsmittel als auch als Reagenz. In Abwesenheit einer basischen Substanz, wie z. B. Diäthylanilin bildet sich hierbei innerhalb kurzer Zeit mit einer Ausbeute von über 500/o das gewünschte 6-Halogenpurinribonucleosid-3 : 5'-cyclophosphat, wobei als Hauptnebenprodukt 6-Halogenpurin gebildet wird.
Die Acylierung in der ersten Stufe des erfindungsgemässen Verfahrens erfolgt in üblicher Weise, vorteilhaft bei Raumtemperatur. Vorzugsweise erfolgt die Umsetzung hierbei unter Lichtabschluss. Nach der Entfernung des restlichen Acylierungsmifteis und der alkalischen Substanz wird das zurückbleibende Öl vorzugsweise durch Waschen mit einem geeigneten Lösungsmittel, wie Äther von letzten Resten an alkalischer Substanz und Acylierungsmittel befreit.
Die Acylierungsreaktion kann bei Temperaturen zwischen Zimmertemperatur und der Rückflusstemperatur der am niedrigsten siedenden Komponente durchgeführt werden.
Die Umsetzung mit dem überschüssigen POHal3, vorzugsweise POC13, lässt sich ebenfalls bei Temperaturen zwischen Raumtemperatur und der Rückfluss- temperalur durchführen. Vorzugsweise wird bei erhöhter Temperatur, insbesondere am Rückfluss gekocht.
Unter diesen Bedingungen ist die Umsetzung nach etwa 15-60 Minuten beendet und überschüssiges POHal3 wird abdestilliert. Die Reinigung des so gewonnenen rohen 6-Chlorpurinribonucleosid-3' ': 5'-cyclophosphats erfolgt vorteilhaft durch Chromatographie an einem geeigneten Ionenaustauscher oder Kieselgel.
Die weitere Umsetzung mit einem Alkohol bzw.
Derivat davon, erfolgt zweckmässig in alkalischem Medium. Vorzugsweise wird der Alkohol als Lösungsmittel verwendet. Gegebenenfalls unter Zusatz eines weiteren üblichen Lösungsmittels.
Die Reinigung der erhaltenen Produkte erfolgt nach den in der Nucleosidchemie üblichen Methoden.
Gute Ergebnisse werden mit der chromatographischen Reinigung erzielt. Vorzugsweise wird hierzu ein Anionenaustauscher, Aktivkohle oder Kieselgel verwendet.
Die erfindungsgemäss erhältlichen Verbindungen sind neu. Sie sind interessante therapeutisch wirksame Substanzen und können als solche oder in Form ihrer Salze mit physiologisch verträglichen Basen zur Beeinflussung der Glykose, der Lipolyse und des Hormonstoffwechsels eingesetzt werden. Der besten bekannten Verbindung gleicher Wirkungsrichtung und vergleichbarer chemischer Konstitution, dem N6-2'-O-Dibutryl-adenosin-3' : 5'-monophosphat, sind die erfindungsgemässen Verbindungen in verschie- dener Hinsicht überlegen. Insbesondere weisen sie folgende Vorteile auf:
1. Die Phosphorylase-Aktivierung tritt bei den erfindungsgemässen Substanzen bereits bei geringeren Konzentrationen an als bei der erwähnten bekannten Verbindung.
2. Die Lipolyse-Aktivierung der erfindungsgemässen Verbindungen tritt entweder bei gleicher oder bei mässig verminderter Konzentration auf. Die Folge ist eine Verschiebung des Wirkungsspektrums zugunsten der Phosphorylase-Aktivierung, die in der nachstehenden Tabelle durch den Konzentrationsquotienten für beide Aktivierungen (letzte Spalte) zum Ausdruck kommt. Gegenüber einem Faktor 2 für die bekannte Verbindung verschieben sich die Wirkungen bei den erfindungsgemässen Verbindungen auf den Faktor 3500. In einigen Fällen ist die Lipolyse-Aktivität hierbei so schwach ausgeprägt, dass von einer nahezu reinen Phosphorylasewirksamkeit, also hoher pharmakologischer Spezifität, gesprochen werden kann.
In der nachstehenden Tabelle sind die Ergebnisse der durchgeführten Vergleichsversuche aufgeführt.

Ausser der Beeinflussung des Kohlehydratstoffwechsels und Lipidstoffwechsels kommen den erfindungsgemässen Verbindungen weitere interessante Wirkungen zu, z. B. eine allgemeine energiemobilisierende Wirkung, die beispielsweise einen Einsatz bei Stress und Schock gestattet. Eine weitere Wirksamkeit liegt in der Erhöhung der Nebennierensteroidprodukte sowie bei der Lösung von Asthma. Die erfindungsgemässen Verbindungen wirken auch als präformiertes Nucleotid und können daher beispielsweise eine Antimetabolitwirkung bei der Immunsuppresion sowie bei Tumoren aufweisen. Ferner wurde neben einer allgemeinen sedierenden Wirkung in einigen Fällen auch eine Potenzierung der Wirkung von Narkotika gefunden.
Auch ein positiv inotroper Herzeffekt konnte festgestellt werden.
Die folgenden Beispiele erläutern die Erfindung weiter.
Beispiel 1
6-Chlorpurinribofuranosid-3':5'-MP-Bariumsaltz
20 g Inosin-3':5'-cyclophosphat, kristallisiert in Form der freien Säure, werden in 200 ml Pyridin und 200 ml Essigsäureanhydrid suspendiert und unter leichtem Erwärmen so lange gerührt, bis eine klare, leicht gelbe Lösung erhalten wird. Nach 15 Stunden bei Raumtemperatur wird im Vakuum zur Trockne destilliert. Das zurückbleibende Öl schüttelt man mit Äther 2 mal kräftig durch, um letzte Reste von Pyridin und Essigsäureanhydrid zu entfernen.
Der Rückstand wird mit 500 ml POCl2 versetzt und unter Rückfluss gekocht. Nach etwa 20-25 Miqu- ten hat sich alles gelöst und das POC13 wird im Vakuum abdestilliert. Zur vollständigen Entfernung des überschüssigen POCl3 wird der Rückstand noch 2 mal mit Äther durchgeschüttelt, anschliessend in 800 ml 0,25 molarem NayHPO4 12H2O gelöst und der pl-I-Wert mit 10%iger NaOH auf 2,0 gestellt.
Die gelbe Lösung wird auf eine mit Carboraffin C-Kohle gefüllte Chromatographiesäule (Länge 120 cm, Durchmesser 2,5 cm) aufgezogen und mit dest. Wasser so lange nachgewaschen, bis der Durchlauf frei von POS- und Cl- ist. Die Elution erfolgt mit Äthanol: H2O : NH4OH (330/oig) (50:50:1). Die im UV-Licht bei 260 mal absorbierenden Fraktionen (oder durch TLC; System Butanol:H2O:Eis-Essig = 50:25:15 auf Kieselgel PF 254 RF 0,6) werden vereinigt und bis auf 200 ml konzentriert.
Das Konzentrat wird über einer Ionenaustauschersäule (Dowex 50 Form; 1m Länge, 2 cm Durchmesser) von Kationen befreit und das saure Eluat mit Barytlauge auf pH 7,0 neutralisiert. Der ausgefallene Niedeschlag wird abzentrifugiert, gut mit H2O gewaschen und die Überstände auf etwa 40-50 ml konzentriert. Dieses Konzentrat wird mit 500 ml Methanol versetzt, der Niederschlag abzentrifugiert und mit 80%igem Methanol gewaschen. Die vereinigten Über- stände werden mit Barytlauge auf pH 10-11 gestellt und nach 20 Minuten bei Raumtemperatur mit 2 n Schwefelsäure auf pH 7 neutralisiert. Der Niederschlag wird abzentrifugiert, mit wenig 800/oigem Methanol gewaschen und die vereinigten Lösungen auf 50 mi konzentriert. Dem Konzentrat werden zuerst 100 ml Methanol und dann 500 ml Äther zugesetzt.
Den ausgefallenen Niederschlag zentrifugiert man ab, wäscht ihn 2 mal mit Äther und trocknet im Vakuum über CaCl2.
Ausbeute:
13 g 6-Chlorpurinribofuranosid3':5'-monophosphat-cyclischbariumsalz (51 /o der Theorie) bezogen auf 1000/o reines BaSalz (0,5 BalMolekül).

6-Methoxy-purinribofuranosid-3':5'-MP
5 g rohes 6-Chlorpurinribosid-3':
:5'-MP in Form der freien Säure (70%ig) werden in 150 ml Methanol gelöst und mit methanolischer Natronlauge (3 g NaOH pro 100 ml Methanol) auf pH = 10-11 gestellt. Nach 30 Minuten wird mit Essigsäure auf pH = 7 neutralisiert und zur Trockne destilliert, der Rückstand in 100 ml Waser gelöst und über eine Säule mit 300 ml Dowex 1 x 2 (50-100 mesh, Formiat-Form) chrmatographiert. Nach Elution mit einem linearem Grandienten von 2 1 destilliertem Wasser gegen 1,5 M Ammoniumformiat wird anschliessend mit 34 1 1,5 M Ammoniumformiat weiterelniert. Die das 6-Methoxy-purinri bosid-3':5'-MP enthaltende Fraktion (Rf im Lösungsmittel A-Rf: 0,56) wird über eine Kohlensäule (100 ml) chromatographiert.
Nach Vorwaschen mit Wasser wird anschliessend mit Isopropanol:H2O:NH, 50:50:1 eluiert und das Eluat auf ca. 50 ml konzentriert. Nach einer Passage über Dowex 50-H+-Form wird gefriergetrocknet.
Ausbeute: 3,0 g ca. 77% der Theorie
Analyse: C11H13O7N4P Molgewicht 343,22
Die Substanz enthält lt. Analyse noch 3,3 % H2O Berechnet: N 15,700/o P 8,41%

Verfahren zur Herstellung von Verbindungen der Formel 1

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man als Acylierungsmittel Essigsäureanhydrid, Acetylchlorid oder Benzoylchlorid verwendet und die Acylierung in Gegenwart einer organischen Stickstoffbase bei einer Temperatur zwischen 0 C und der Rückflusstemperatur durchführt.
2. Verfahren nach Patentanspruch I oder Unteran aspruch 1, dadurch gekennzeichnet, dass man bei Raumtemperatur und unter Lichtausschluss arbeitet.
3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man restliches Acylierungsmittel und restliche Base durch Waschen des Rückstandes mit einem organischen Lösungsmittel, vorzugsweise Äther entfernt.
4. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man als Halogenierungsmittel Phosphoroxychlorid verwendet und die Umsetzung bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur durchführt.
5. Verfahren nach Unteranspruch 4, dadurch gekennnzeichnet, dass man bei Rückflusstemperatur arbeitet.
6. Verfahren nach Patentanspruch I, dadurch ge komaeichnet, dass naan eine Verbindung der Formel 1 herstellt, worin X Fluor, Brom oder Jod bedeutet.

Cyclic 3 ': 5'-phosphates of nucleosides are gaining in importance in biochemistry and adenosine-3': 5'-cyclophosphate has been used since its discovery by Sutherland 10 years ago (Sutherland, Rall, J.Amer. Soc. 79, 3608 [1957]) has become a key substance in glycolysis, lipolysis and hormone metabolism, the physiological importance of which is often equated with the most important energy carrier in the cell, ATP.
According to Sutherland's theory, adenosine3 ': 5'-cyclophosphate has the physiological function of a second messenger in the cell, which is briefly illustrated using the example of the action of the peptide hormone ACTH. ACTH released from the pituitary gland reaches the adrenal cortex via the bloodstream, in the cell wall of which the enzyme adenylcyclase is located Biosynthesis and release of corticosteroids causes.
The conditions are similar in glycogenolysis, lipolysis, etc. The selectivity is thus given by the receptors of the respective success organs, which have adenosine 3 ': 5'-cyclophosphate as an intracellular signal; Adenosine 3 ': 5'-cyclophosphate is applied externally, but has multiple effects, which is often pharmacologically undesirable. In addition, it is split relatively quickly in the organism, so that its duration of action is short.
There is therefore a need to create adenosine 3 ': 5'-cyclophosphate analogs with improved specificity of action and longer duration of action.
The synthesis of modified analogs of adenosine 3 ': 5'-cyclophosphate derivatives has so far been carried out in a very complicated and expensive manner, which is illustrated using the example of 6-mercaptopurine ribonucleoside 3': 5'-cyclophosphate:
Inosine + conversion into 2 ', 3', 5'-tribenzoylinosine reaction with P2S5 to the 6-mercaptopurine ribonucleoside + removal of the protective group and introduction of a new protective group on the 2 ', 3'-hydroxyl group (e.g.
by ketalization), the 5'-OH position remaining free + phosphorylation on the 5'OH with ss-cyanoethyl phosphate and DCC as condensation agent + cleavage a) of 2 ', 3'-OH protective group, b) of phosphate protective group + cyclization to desired product.
The yields in this multi-stage synthesis are very low (1.50 / 0 of theory; J. Thomas, Montgomery J. Med. Pharm. Chem. 11, 44 (1968)), especially since the mercapto group, which is sensitive to hydrolysis and oxidation, is from the start must be pulled through the individual reaction stages.

For an industrially applicable process, the above production method was ruled out due to its inconvenience and low yield. A way was therefore sought to carry out the substitution in position 6 of the purine structure starting from the preformed cyclophosphate compound.
The following processes for converting the hydroxyl group into the chlorine group in common nucleosides are known:
1. Chlorine in methanol (G.D. Daves et al., J. Amer.

2. concentrated hydrochloric acid, chlorine gas and methanol (Gerster et al., J. Org. Chem. 28, 945, (1963)),
3. Diethylaniline and POC13 (Gerster et al., J. Org.
Chem. 28, 945, (1963)),
4. SoCl-DMF (M. Ikehara, Chem. Pharm. Bull.
12, 267, (1964)).
However, it has been found that none of these known processes, when applied to the S-hydroxypurine ribonuoleoside-3 ': 5'-cyclophophate, leads to the product sought. The tests showed that either extensive degradation occurred or the starting material was not converted at all. This shows that the reactivity is fundamentally changed when a cyclophosphate group is introduced also in the purine nucleus and therefore the processes known for the purine nucleosides cannot be transferred to the cyclic nucleoside 3 ': 5'-phosphates.
The same also applies to the conversion of inosine into the corresponding 6-mercapto derivative known in the nucleoside series by reaction with P, tS. This reaction, which proceeds smoothly with the nucleosides, cannot be carried out on the preformed cyclophosphate and does not lead to the desired 6-mercapto derivative under any conditions.
The preparation of 6-chloropurine-deoxyriboside-3 ': 5'-cyclophosphate by reaction with phosphorus trichloride and an alkaline substance such as an inorganic alkali or an aliphatic or aromatic amine has already been described in Japanese patent specification 7957/68. This known process leads to a yield of 260io.
However, it has been found that this reaction cannot be carried out with the ordinary 6-hydroxypurine ribonucleoside 3 ': 5'-cyclophosphate. In the presence of bases such as B.
Diethylaniline does not undergo any conversion under the conditions of the known process and the reaction medium turns black-brown and contains numerous decomposition products.
Surprisingly, it has now been found that the desired reaction proceeds smoothly and with very good yield if a 6-hydroxypurine ribonucleoside 3 ': 5'-cyclophosphate is acylated as the starting material and the acylation product is reacted directly with phosphorus oxyhalide
In this way, the corresponding halopur ribonucleoside 3 ': 5'-cyclophosphate is obtained in a yield of over 50%.
The compounds are therefore prepared according to the invention by reacting a corresponding compound in which X is a hydroxyl group with an acylating agent in the presence of a base, removing unreacted acylating agent and base and reacting the residue with excess phosphorus oxyhalide.
The starting product for the process of the invention is a 6-hydroxypurine ribonudeoside 3 ': 5'-cyclophosphate of the formula

where R is a hydrogen atom, a hydroxyl or amino group, is used. This starting product can easily be obtained in high yield by chemical deamination of, for example, adenosine 3 ': 5'-cyclophosphate.
The 6-chloro derivative is preferably prepared with phosphorus oxychloride at a temperature between room temperature and reflux temperature. The 6-halogen derivative is then converted into a compound of the formula 2 by reaction with excess alcoholate in aqueous or alcoholic solution at a temperature between -10 ° C. and the reflux temperature.
The starting product is acylated in the first stage of the process according to the invention in order to make it soluble in the phosphorus oxyhalide. The acylation only needs to be carried out to such an extent that the acylated product is sufficiently soluble for conversion in POH. Preferably the acylation is carried out with an acyl chloride or acyl anhydride in the presence of an alkaline agent. The acylation is preferably carried out with acetyl chloride, acetic anhydride or benzoyl chloride. The alkaline agent used is preferably organic bases which, together with excess acylating agent, can easily be separated from the acylated nucleoside by distillation.
The reaction with acetic anhydride in pyridine, which can easily be carried out at room temperature, has proven to be very suitable. This creates 6-hydroxypum-2'-O; acetylribo- nucleoside-3 ': 5'-cycloacetyl phosphate, which is readily soluble in POHal3 and is therefore particularly suitable for the reaction. The 2'-acetyl-6-hydroxypurine, ibolucleoside3 ': 5'-cyclophosphate is also suitable, but the yield is poorer due to the lower solubility.
When the acylation is complete, the remaining acylation components and base are removed, preferably by distillation. The oily acylation product that remains is reacted directly with excess POHal5. Here the POH serves as both a solvent and a reagent. In the absence of a basic substance, such as. B. Diethylaniline forms the desired 6-halopurine ribonucleoside 3: 5'-cyclophosphate within a short time with a yield of over 500 / o, with 6-halopurine being formed as the main by-product.
The acylation in the first stage of the process according to the invention is carried out in the customary manner, advantageously at room temperature. The reaction is preferably carried out in the absence of light. After the remaining acylating agent and the alkaline substance have been removed, the remaining oil is freed from the last residues of alkaline substance and acylating agent, preferably by washing with a suitable solvent, such as ether.
The acylation reaction can be carried out at temperatures between room temperature and the reflux temperature of the lowest boiling component.
The reaction with the excess POHal3, preferably POC13, can also be carried out at temperatures between room temperature and the reflux temperature. It is preferred to boil at an elevated temperature, in particular at reflux.
Under these conditions, the reaction is complete after about 15-60 minutes and excess POHal3 is distilled off. The crude 6-chloropurine ribonucleoside 3 ″: 5′-cyclophosphate obtained in this way is advantageously purified by chromatography on a suitable ion exchanger or silica gel.

The products obtained are purified by the methods customary in nucleoside chemistry.
Good results are obtained with chromatographic purification. An anion exchanger, activated carbon or silica gel is preferably used for this.
The compounds obtainable according to the invention are new. They are interesting therapeutically active substances and can be used as such or in the form of their salts with physiologically compatible bases for influencing glycosis, lipolysis and hormone metabolism. The compounds according to the invention are superior in various respects to the best known compound with the same direction of action and comparable chemical constitution, N6-2'-O-dibutryl-adenosine-3 ': 5'-monophosphate. In particular, they have the following advantages:
1. The phosphorylase activation occurs with the substances according to the invention even at lower concentrations than with the known compound mentioned.
2. The lipolysis activation of the compounds according to the invention occurs either at the same or at a moderately reduced concentration. The result is a shift in the spectrum of activity in favor of phosphorylase activation, which is expressed in the table below by the concentration quotient for both activations (last column). Compared to a factor of 2 for the known compound, the effects of the compounds according to the invention shift to a factor of 3500. In some cases, the lipolysis activity is so weak that it is almost pure phosphorylase activity, i.e. high pharmacological specificity.

In addition to influencing the carbohydrate metabolism and lipid metabolism, the compounds according to the invention have other interesting effects, e.g. B. a general energy mobilizing effect, which allows for example use in stress and shock. Another effectiveness is in increasing adrenal steroid products as well as in solving asthma. The compounds according to the invention also act as preformed nucleotides and can therefore, for example, have an antimetabolic effect in immunosuppression and in tumors. In addition to a general sedative effect, a potentiation of the effect of narcotics was also found in some cases.
A positive inotropic heart effect could also be determined.

example 1
6-chloropurine ribofuranoside-3 ': 5'-MP-barium salt
20 g of inosine 3 ': 5'-cyclophosphate, crystallized in the form of the free acid, are suspended in 200 ml of pyridine and 200 ml of acetic anhydride and stirred with gentle warming until a clear, slightly yellow solution is obtained. After 15 hours at room temperature, it is distilled to dryness in vacuo. The remaining oil is shaken vigorously twice with ether to remove the last residues of pyridine and acetic anhydride.
The residue is mixed with 500 ml of POCl2 and refluxed. After about 20-25 microns everything has dissolved and the POC13 is distilled off in vacuo. To completely remove the excess POCl3, the residue is shaken twice more with ether, then dissolved in 800 ml of 0.25 molar NayHPO4 12H2O and the pI value is adjusted to 2.0 with 10% NaOH.
The yellow solution is drawn up on a chromatography column (length 120 cm, diameter 2.5 cm) filled with carboraffin carbon and washed with dist. Washed water until the passage is free of POS and Cl-. Elution takes place with ethanol: H2O: NH4OH (330 / oig) (50: 50: 1). The fractions which absorb 260 times in UV light (or by TLC; system butanol: H2O: ice-vinegar = 50:25:15 on silica gel PF 254 RF 0.6) are combined and concentrated to 200 ml.
The concentrate is freed of cations over an ion exchange column (Dowex 50 form; 1 m length, 2 cm diameter) and the acidic eluate is neutralized to pH 7.0 with barite liquor. The precipitated precipitate is centrifuged off, washed well with H2O and the supernatants concentrated to about 40-50 ml. This concentrate is mixed with 500 ml of methanol, the precipitate is centrifuged off and washed with 80% methanol. The combined supernatants are adjusted to pH 10-11 with barite liquor and, after 20 minutes at room temperature, neutralized to pH 7 with 2N sulfuric acid. The precipitate is centrifuged off, washed with a little 800% methanol and the combined solutions concentrated to 50 ml. First 100 ml of methanol and then 500 ml of ether are added to the concentrate.
The precipitate is centrifuged off, washed twice with ether and dried in vacuo over CaCl2.
Yield:
13 g of 6-chloropurine ribofuranoside3 ': 5'-monophosphate cyclic barium salt (51 / o of theory) based on 1000 / o pure Ba salt (0.5 Balmolecule).

5 g of raw 6-chloropurine riboside-3 ':
: 5'-MP in the form of the free acid (70%) are dissolved in 150 ml of methanol and adjusted to pH = 10-11 with methanolic sodium hydroxide solution (3 g of NaOH per 100 ml of methanol). After 30 minutes, it is neutralized with acetic acid to pH = 7 and distilled to dryness, the residue is dissolved in 100 ml of water and chromatographed on a column with 300 ml of Dowex 1 × 2 (50-100 mesh, formate form). After elution with a linear gradient of 2 liters of distilled water against 1.5 M ammonium formate, the mixture is then further eluted with 34 liters of 1.5 M ammonium formate. The fraction containing the 6-methoxy-purin triside-3 ': 5'-MP (Rf in solvent A-Rf: 0.56) is chromatographed on a carbon column (100 ml).
After pre-washing with water, it is then eluted with isopropanol: H2O: NH, 50: 50: 1 and the eluate is concentrated to approx. 50 ml. After passing through Dowex 50-H + form, it is freeze-dried.
Yield: 3.0 g approx. 77% of theory
