Verfahren zur Herstellung von Aminderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von Aminderivaten.
Es ist z. B. aus den britischen Patentschriften Nummern 1078 852, 1185044 und 1 185045 bekannt, dass verschiedene 1 -Acylaminoaryloxy-3 -alkylamino-2-propanol- derivate wertvolle therapeutische Eigenschaften, wie z.B. eine ,B-adrenergische Wirkung, besitzen, und dass diese Verbindungen für die Behandlung oder Prophylaxe von Herzkrankheiten, wie z.B. Angina pectoris und Kardialarrhythmien, verwendet werden können. Es wurde nun überraschenderweise gefunden, dass diese Verbindungen durch Acylierung der entsprechenden
1 -Aminoaryloxy-3 -alkylamino-2-propanolderivate hergestellt werden können, und dass die Acylierung zur Hauptsache am Arylaminorest und nicht in erheblichem Ausmass am Alkylaminorest stattfindet.
Die vorliegende Erfindung bezieht sich somit auf ein Verfahren zur Herstellung von Aminderivaten der Formel:

wobei Rt einen Alkyl-, Hydroxylalkyl- oder Cycloalkylrest, R2 ein Wasserstoffatom oder einen Alkyl-, Alkenyl-, Alkoxy- oder Alkenyloxyrest und R3 einen Alkanoyl-, Aroyl-, Alkansulfonyl- oder Arensulfonylrest bedeuten, sowie von Säureadditionssalzen davon, indem eine Verbindung der Formel:

worin R1 und R2 die obigen Bedeutungen haben, mit einem den Acylrest R3 einführenden Acylierungsmittel zur Reaktion gebracht wird. Darauf kann man das Produkt gewünschtenfalls in Form der freien Base mit einer Säure zur Reaktion bringen, um ein Säureadditionssalz zu bilden.
Im Sinne der Erfindung soll die obige Definition von Alkanolaminderivaten alle möglichen stereoisomere Verbindungen davon und Mischungen derselben einschliessen. Insbesondere soll die obige Definition auch racemische Verbindungen und optisch aktive Verbindungen umfassen.
Der Rest Rt kann z.B. ein Alkyl-, Hydroxyalkyloder Cycloalkylrest mit je höchstens 12 C-Atomen, z.B. den Methyl-, Äthyl-, n-Propyl-, Isopropyl-, sek. Butyl-, tert.-Butyl-, l-Methyloctyl-, 1,1 -Dimethyl-2-hy- droxyäthyl- oder Cyclopentylrest, sein. Vorzugsweise ist R1 ein Alkylrest, dessen Kette am a-Kohlenstoffatom verzweigt ist, wobei man insbesondere einem verzweigten Alkylrest mit 3 bis 5 C-Atomen, z.B. dem Isopropyl-, sek.-Butyl- oder tert.-Butylrest den Vorzug geben wird.
Stellt R2 einen Alkyl-, Alkenyl-, Alkoxy- oder Alkenyloxyrest dar, so wird dieser Rest vorzugsweise bis zu 6 C-Atome aufweisen und beispielsweise den Methyl-, Äthyl-, Allyl-, Methoxy-, Äthoxy- oder Allyloxyrest sein.
Stellt R3 einen Alkanoyl- oder Alkansulfonylrest dar, so kann dieser Rest z. B. ein Alkanoylrest mit bis zu 7 C-Atomen oder ein Alkansulfonylradikal mit bis zu 6 C-Atomen, z. B. der Acetyl-, Propionyl-, Butyryl-, Methansulfonyl- oder Äthansulfonylrest, sein.
Stellt R5 einen Aroyl- oder Arensulfonylrest dar, so kann dieser Rest z. B. ein Aroyl- oder Arensulfonylrest mit bis zu 12 C-Atomen, z. B. der Benzoyl-, Benzolsulfonyl- oder Toluol-p-sulfonylrest, sein.
Als geeignete Säureadditionssalze der Aminderivate kann man z.B. Hydrochloride, Hydrobromide, Phosphate, Sulfate, Oxalate, Lactate, Tartrate, Citrate, Acetate oder Benzoate erwähnen.
Bestimmte Beispiele für die Aminderivate, die nach dem erfindungsgemässen Verfahren hergestellt werden können, sind:
1 -(4-Acetamidophenoxy)-3 -isopropylamino-2propanol (Smp. 1410 C); 1-(4-Acetamidophenoxy)-3-tert.-butylamino-2- propanol (Smp. 1260 C); 1 -(4-Benzamidophenoxy)-3 -isopropylamino-2- propanol (Smp. 162-1630 C); 1 -(4-Methansulfonamidophenoxy)-3 -isopropyl- amino-2-propanol (Smp. 1261280 C); 1 -(4-Benzolsulfonamidophenoxy)-3-isopropyl- amino-2-propanol (Smp. 1300 C);
1-(4-Butyramidophenoxy)-3-isopropylamino-2propanol (Smp. 127-1280 C);

Als geeignetes Acylierungsmittel für das erfindungsgemässe Verfahren kann man z. B. ein Säurehalogenid beispielsweise ein Säurechlorid, oder ein Säureanhydrid, oder ein Mischsäureanhydrid beispielsweise ein Mischanhydrid mit Monoäthylkohlensäure, oder einen aktivierten Ester beispielsweise den p-Nitrophenylester, das bzw. der von der Säure der Formel R3-OH stammt, wobei R3 die o. a. Bedeutung hat, erwähnen. Auch könnte die Säure der Formel R3-OH selbst als Acylierungsmittel in Gegenwart von einem Kondensierungsmittel, z. B. einem Carbodiimid beispielsweise N,N' Dicyclohexylcarbodiimid, verwendet werden.
Das Verfahren nach der Erfindung kann in einem Verdünnungs- oder Lösungsmittel, z. B. einem wässrigen Verdünnungs- oder Lösungsmittel, wie z.B. Wasser, ein wässriges Keton beispielsweise wässriges Aceton, ein wässriger Alkohol beispielsweise wässriges Äthanol, oder eine wässrige Säure beispielsweise wässrige Essigsäure, oder in einem nichtwässrigen organischen Verdünnungsoder Lösungsmittel, z. 3. einem Keton beispielsweise Aceton, einem Alkohol beispielsweise Äthanol, einem Ester beispielsweise Athylacetat, oder einem Amid beispielsweise Dimethylformamid, durchgeführt werden.
Das Verfahren nach der Erfindung wird vorzugsweise bei einer Temperatur im Bereich 0 bis 600 C, am zweckmässigsten bei Raumtemperatur, durchgeführt.
Wird das Verfahren unter wässrigen Bedingungen durchgeführt, so beträgt der pH-Wert vorzugsweise 3 bis 5.
Von den in dieser Beschreibung erwähnten Verbindungen sind einige vorher weder beschrieben noch beansprucht worden. Gemäss einem weiteren Merkmal der Erfindung werden also die folgenden Verbindungen geschaffen:
1-(4-Äthansulfonamidphenoxy)-3-isopropylamino2-propanol,
1 -(4-Benzamidophenoxy)-3-cyclopentylamino-2propanol,
1-(4-Benzamido-2-äthoxyphenoxy)-3-isopropyl amino-2-propanol,
1 -(4-Benzamidophenoxy)-3-methylamino-2propanol,
1 -(4-Benzamidophenoxy)-3-sek.-butylamino-2propanol, 3-( 1 ,1-Dimethyl-2-hydroxyäthyl)-1 -(4-propion- amidophenoxy)-2-propanol,
3-Cyclopentylamino-1 -(4-propionamidophenoxy)2-propanol und die Säureadditionssalze davon.
Die Erfindung wird im folgenden anhand von Ausführungsbeispielen näher erläutert, wobei die Teile auf das Gewicht bezogen sind.
Beispiel 1
Es werden 11,1 Teile 1-(4-Aminophenoxy)-3-isopropylamino-2-propanol in einer Mischung aus 100 Teilen Aceton und 100 Teilen Wasser aufgelöst, und der pH-Wert der Lösung wird auf 4 bis 5 eingestellt. Die Lösung wird gerührt, und es werden 8 Teile Acetylchlorid während 30 Minuten hinzugegeben. Während der Zugabe von Acetylchlorid wird der pH-Wert durch Zugabe von einer verdünnten wässrigen Natriumhydroxydlösung auf 4 bis 5 gehalten. Die Lösung wird unter vermindertem Druck zur Trockne eingedampft; es werden 50 Teile Aceton hinzugegeben, und die Mischung wird 12 Stunden bei Umgebungstemperatur stehengelassen. Die Mischung wird filtriert, und dem Filtrat werden 100 Teile Äther zugesetzt. Es wird ein dickflüssiges Öl niedergeschlagen. Der Äther wird abgegossen, und das öl wird in 5 Teilen Wasser aufgelöst.
Dann werden 10 Teile einer wässrigen lOn-Na- triumhydroxydlösung hinzugegeben, und die Mischung wird mit 50 Teilen Äthylacetat extrahiert. Die Athyl- acetatlösung wird mit Wasser gewaschen, getrocknet und unter vermindertem Druck zur Trockne eingedampft. Der Rückstand wird aus Äthylmethylketon umkristallisiert. Somit erhält man
1 -(4-Acetamidophenoxy)-3-isopropylamino-2 propanol (Smp. 1410 C).
Beispiel 2
Es werden 1,5 Teile Essigsäureanhydrid zu einer gerührten Mischung aus 1,0 Teil
1 -(4-Aminophenoxy)-3 -isopropylamino-2-propanol und 20 Teilen Wasser zugegeben, die auf 250 C gehalten wird. Die Lösung wird 10 Minuten gerührt, und es wird eine ausreichende Menge einer wässrigen 2n-Natriumhydroxydlösung hinzugegeben, um den pH Wert der Lösung auf 10 bis 11 zu bringen. Dann werden 30 Teile einer gesättigten wässrigen Natriumchloridlösung hinzugegeben, und die Mischung wird 2 Stunden bei Umgebungstemperatur stehengelassen.
Die Mischung wird filtriert, und der feste Rückstand wird mit einer geringen Menge einer gesättigten Natriumchloridlösung gewaschen und anschliessend getrocknet. Der Feststoff wird aus Äthylacetat umkristallisiert. Somit erhält man 0,8 Teile
1 -(4-Acetamidophenoxy)-3-isopropylamino-2propanol (Smp. 1410 C).
Beispiel 3
Es werden 0,5 Teile Benzoylchlorid zu einer Mischung aus 0,5 Teilen
1 -(4-Aminophenoxy)-3 -isopropylamino-2-propanol, 0,15 Teilen Essigsäure und 10 Teilen Wasser zugegeben, und die Mischung wird 15 Minuten bei Umgebungstemperatur geschüttelt. Die Mischung wird filtriert, und dem Filtrat wird eine ausreichende Menge einer wässrigen 2n-Natriumhydroxydlösung zugesetzt, um den pH-Wert der Lösung auf 9 bis 12 zu bringen.
Die Mischung wird mit 20 Teilen Äthylacetat extrahiert, und der Extrakt wird mit Wasser gewaschen, getrocknet und zur Trockne eingedampft. Der Rückstand wird aus Äthylacetat umkristallisiert. Somit erhält man
1 -(4-Benzamidophenoxy)-3-isopropylamino-2propanol (Smp. 16211630 C).
Dieses Verfahren wird wiederholt, mit der Abweichung, dass anstelle von 1-(4-Aminophenoxy)-3-isopropylamino-2-propanol das entsprechende 3-Alkylamino-1-(4-aminophenoxy)-2-propanol verwendet wird. Somit erhält man:
1-(4-Benzamidophenoxy)-3-methylamino-2propanol (Smp. 166-1680 C); i-(4-Benzamidophenoxy)-3-sek. -butylamino-2- propanol-monohydrat (Smp. 119-1210 C) und 1 -(4-3 enzamidophenoxy)-3 -cyclopentylamino-2- propanol (Smp. 161-1620 C).
Beispiel 4
Es werden 0,3 Teile Propionsäureanhydrid zu einer Mischung aus 0,5 Teilen 1-(4-Aminophenoxy)-3-isopropylamino-2-propanol, 0,15 Teilen Essigsäure und 10 Teilen Wasser zugegeben, und die Mischung wird 30 Minuten bei Umgebungstemperatur geschüttelt. Die Lösung wird filtriert, und dem Filtrat wird eine ausreichende Menge einer wässrigen 2n-Natriumhydroxydlösung zugegeben, um den pH-Wert auf 9 bis 12 zu bringen. Die Mischung wird mit 20 Teilen Athylacetat extrahiert, und der Extrakt wird mit Wasser gewaschen, getrocknet und zur Trockne eingedampft. Der Rückstand wird aus Äthanol umkristallisiert. Somit erhält man
3-Isopropylamino-1-(4-propionamidophenoxy)2propanol (Smp. 133-1350 C).
Dieses Verfahren wird wiederholt, mit der Abweichung, dass anstelle von
1 -(4-Aminophenoxy)-3-isopropylamino-2-propanol das entsprechende 3-Alkylamino-1 -(4-aminophenoxy)-2-propanol verwendet wird und/oder dass anstelle von Propionsäureanhydrid das entsprechende Säureanhydrid verwendet wird.
Somit erhält man:
1 -(4-Benzamidophenoxy)-3 -isopropylamino-2propanol (Smp. 1621630 C);
1 -(4-Acetamidophenoxy)-3-tert.-butylamino-2- propanol (dessen Hydrochlorid einen Schmelzpunkt von 173 bis 1750 C hat);
3-Cyclopentylamino- 1 -(4-propionamidophenoxy)2-propanol (Smp. 121-1220 C); 1-(4-Benzamido-2-äthoxyphenoxy)-3-isopropyl- amino-2-propanol (Smp. 146-1480 C) und 3-( 1,1 -Dimethyl-2-hydroxyäthylamino)- 1 -(4-pro- pionamidophenoxy)-2-propanol (Smp. 108-1100 C).
Beispiel 5
Eine Lösung von 1,08 Teilen Äthylchlorformiat in 16 Teilen Aceton wird zu einer gerührten Mischung aus 0,6 Teilen Essigsäure, 1,01 Teilen Triäthylamin und 16 Teilen Aceton zugegeben, während die Mischung auf 0 C gehalten wird, worauf 2,24 Teile
1-(4-Aminophenoxy)-3 -isopropylamino-2-propanol hinzugegeben werden und die Mischung 2 Stunden bei 0 C und dann 1 Stunde bei Raumtemperatur gerührt wird. Die Mischung wird gefiltert, und das Filtrat wird zur Trockne eingedampft, worauf der Rückstand in 20 Teilen einer wässrigen 2n-Salzsäurelösung aufgelöst wird. Die Lösung wird mit Diäthyläther gewaschen, worauf eine ausreichende Menge von einer wässrigen 2n-Natriumhydroxydlösung hinzugegeben wird, um den pH-Wert auf 9 bis 12 zu bringen. Das entstehende Gemisch wird mit Äthylacetat extrahiert.
Der Athyl- acetatextrakt wird getrocknet und zur Trockne eingedampft, und der Rückstand wird aus Methyläthylketon umkristallisiert. Somit erhält man
1 -(4-Acetamidophenoxy)-3 -isopropylamino-2propanol (Smp. 1410 C).
Dieses Verfahren wird wiederholt, mit der Abweichung, dass anstelle von Essigsäure Benzoesäure verwendet wird. Somit erhält man
1 -(4-Benzamidophenoxy)-3-isopropylamino-2- propanol (Smp. 162-1630 C).
Beispiel 6
Eine Lösung von 1,8 Teilen p-Nitrophenylacetat und 2,24 Teilen
1 -(4-Aminophenoxy)-3 -isopropylamino-2-propanol in 40 Teilen N,N-Dimethylformamid wird 24 Stunden bei Raumtemperatur gerührt, worauf das Lösungsmittel abgedampft wird. Der Rückstand wird in 30 Teilen einer wässrigen 2n-Salzsäurelösung aufgelöst, und die Lösung wird mit Diäthyläther gewaschen, worauf eine ausreichende Menge einer wässrigen 2n-Natriumhydroxydlösung hinzugegeben wird, um den pH-Wert der Lösung auf 9 bis 12 zu bringen. Das entstehende Gemisch wird mit Äthylacetat extrahiert, und der Extrakt wird mit Wasser gewaschen, getrocknet und zur Trockne eingedampft. Der Rückstand wird aus Methyläthylketon umkristallisiert. Somit erhält man 1-(4-Acetamidophenoxy)-3-isopropylamino-2- propanol (Smp. 1410 C).
Beispiel 7
Eine Lösung von 2,21 Teilen N,N'-Dicyclohexylcarbodiimid in 20 Teilen Äthylacetat wird zu einer gerührten Lösung von 1,68 Teilen
1-(4-Aminophenoxy)-3 -isopropylamino-2-propanol und 3,15 Teilen n-Buttersäure in 30 Teilen Äthylacetat zugegeben, und das entstehende Gemisch wird 30 Minuten bei Raumtemperatur gerührt. Das Gemisch wird filtriert, und das Filtrat wird zur Trockne eingedampft, worauf der Rückstand aus einer Mischung aus Athylacetat und Hexan umkristallisiert wird. Somit erhält man
1 -(4-Butyramidophenoxy)-3 -isopropylamino-2propanol (Smp. 127-1280 C).
Beispiel 8
Es -werden 0,5 Teile Toluol-p-sulfonylchlorid zu einer Mischung aus 0,5 Teilen
1-(4-Aminophenoxy)-3-isopropylamino-2-propanol, 0,15 Teilen Essigsäure und 10 Teilen Wasser zuge geben, und die Mischung wird soweit erhitzt, dass das Toluol-p-sulfonylchlorid zum Schmelzen gebracht wird, worauf die Mischung 15 Minuten geschüttelt wird.
Dann wird eine ausreichende Menge einer wässrigen Natriumhydroxydlösung hinzugegeben, um den pH Wert der Lösung auf 9 bis 12 zu bringen, worauf die Mischung mit Äthylacetat extrahiert wird. Der Äthylacetatextrakt wird mit Wasser gewaschen, getrocknet und zur Trockne eingedampft, worauf der Rückstand aus Äthylacetat umkristallisiert wird. Somit erhält man 3-Isopropylamino-tl -(4-toluol-p-sulfonamidophenoxy)-2-propanol (Smp. 99-1010 C).
Dieses Verfahren wird wiederholt, mit der Abweichung, dass anstelle von Toluol-p-sulfonylchlorid als Ausgangsstoff Athansulfonylchlorid verwendet wird. Somit erhält man 1 -(4-Äthansulfonamidophenoxy)-3-isopropylamino- 2-propanol (Smp. 136-1380 C).
Beispiel 9
Eine Lösung von 0,5 Teilen Benzoylchlorid in 4 Teilen Aceton wird zu einer Lösung von 0,5 Teilen 1 -(4-Aminophenoxy)-3-isopropylamino-2-propanol in 4 Teilen Aceton zugegeben, und die Mischung wird geschüttelt. Die klare Lösung wird von dem anfänglichen öligen Niederschlag abgegossen und so lange bei Zimmertemperatur stehengelassen, bis kein Feststoff mehr niedergeschlagen wird. Die Mischung wird filtriert, und das feste Produkt wird aus Äthanol umkristallisiert. Somit erhält man
1 -(4-Benzamidophenoxy) -3-isopropylamino-2- propanol-hydrochlorid (Smp. 1741760 C).
Beispiel 10
Eine Lösung von 0,5 Teilen Benzoylchlorid in 4 Teilen Äthanol wird zu einer Lösung von 0,5 Teilen
1 -(4-Aminophenoxy)-3-isopropylamino-2-propanol in 4 Teilen Äthanol zugegeben, und die Mischung wird 10 Minuten geschüttelt. Dann werden 65 Teile wasserfreien Diäthyläthers hinzugegeben, und die Mischung wird noch 1 Minute geschüttelt. Die klare Lösung wird von dem anfänglichen öligen Niederschlag abgegossen und dann 10 Minuten bei Raumtemperatur stehengelassen. Das Gemisch wird gefiltert, und der feste Rückstand wird aus Äthanol umkristallisiert. Somit erhält man
1 -(4-Benzamidophenoxy)-3-isopropylamino-2propanol-hydrochlorid (Smp. 1741760 C).
Beispiel 11
Eine Lösung von 0,5 Teilen Benzoylchlorid in 5 Teilen Äthylacetat wird zu einer Suspension von 0,5 Teilen
1 -(4-Aminophenoxy)-3-isopropylamino-2-propanol in 5 Teilen Äthylacetat zugegeben, und die Mischung wird 10 Minuten geschüttelt und dann gefiltert. Das feste Produkt wird mit Äthylacetat und dann mit Äther gewaschen und anschliessend in 8 Teilen Äthanol aufgelöst. Es werden 60 Teile Diäthyläther hinzugegeben, und die Mischung wird 1 Minute geschüttelt. Die klare Lösung wird vom anfänglichen öligen Niederschlag abgegossen und dann so lange bei Raumtemperatur stehengelassen, bis kein Feststoff mehr niedergeschlagen wird. Die Mischung wird filtriert, und das feste Produkt wird aus Athanol umkristallisiert.
Somit erhält man
1 -(4-Benzamidophenoxy)-3-isopropylamino-2 propanol-hydrochlorid (Smp. 174-1760 C).
Beispiel 12
Die 3-Alkylamino- 1 -(4-aminophenoxy)-2-propanolderivate, die als Ausgangsstoffe bei den Beispielen 1 bis 11 verwendet werden, können wie folgt hergestellt werden:
A. Eine Lösung von 31,5 Teilen l-Chlor-2,3-epoxy- propan in 16 Teilen Methanol wird zu einer Mischung aus 13,9 Teilen p-Nitrophenol, 4,4 Teilen Natriumhydroxyd und 10 Teilen Wasser zugegeben, und die Mischung wird 3 Stunden unter Rückfluss erhitzt und dann zur Trockne eingedampft. Dann werden 31 Teile Methylamin hinzugegeben, und die Mischung wird 2 Stunden unter Rückfluss erhitzt. Die Mischung wird zur Trockne eingedampft, und der Rückstand wird in 50 Teilen einer wässrigen 2n-Salzsäurelösung aufgelöst.
Die saure Lösung wird mit Diäthyläther gewaschen, und es wird eine ausreichende Menge einer wässrigen 2n-Natriumhydroxydlösung hinzugegeben, um den pH Wert der Lösung auf 9 bis 12 zu bringen. Die Mischung wird mit 45 Teilen Äthylacetat extrahiert, und der Äthylacetatextrakt wird getrocknet und zur Trockne eingedampft. Der Rückstand wird in 40 Teilen Äthanol aufgelöst, und dieser Lösung werden 0,5 Teile Platinoxyd zugesetzt. Die Mischung wird in einer Wasserstoffatmosphäre geschüttelt, bis kein Wasserstoff mehr aufgenommen wird, worauf die Mischung filtriert und das Filtrat zur Trockne eingedampft wird. Somit erhält man als Rückstand
1 -(4-Aminophenoxy)-3 -methylamino-2-propanol .
Dieses Verfahren wird wiederholt, mit der Abweichung, dass anstelle von Methylamin das entsprechende Amin verwendet wird. Somit erhält man:
1 -(4-Aminophenoxy)-3-isopropylamino-2-propanol (Smp. 124-1260 C);
1 -(4-Aminophenoxy)-3 -sek.-butylamino-2-propanol (Hydrochlorid Smp. 142-1440 C) und
1 -(4-Aminophenoxy)-3 -tert. -butylamino-2-propanol (Hydrochlorid Smp. 126-1270 C).
B. Ein Gemisch aus 1 Teil

It has now surprisingly been found that these compounds by acylation of the corresponding
1-Aminoaryloxy-3-alkylamino-2-propanol derivatives can be prepared, and that the acylation takes place mainly on the arylamino radical and not to a significant extent on the alkylamino radical.

For the purposes of the invention, the above definition of alkanolamine derivatives is intended to include all possible stereoisomeric compounds thereof and mixtures thereof. In particular, the above definition is also intended to include racemic compounds and optically active compounds.
The remainder Rt can e.g. an alkyl, hydroxyalkyl or cycloalkyl radical each having a maximum of 12 carbon atoms, e.g. the methyl, ethyl, n-propyl, isopropyl, sec. Suitable acylating agents for the process according to the invention can be, for. example 1
11.1 parts of 1- (4-aminophenoxy) -3-isopropylamino-2-propanol are dissolved in a mixture of 100 parts of acetone and 100 parts of water, and the pH of the solution is adjusted to 4 to 5. Example 2
1.5 parts of acetic anhydride are added to a stirred mixture of 1.0 part
1 - (4-aminophenoxy) -3-isopropylamino-2-propanol and 20 parts of water, which is kept at 250.degree. The mixture is extracted with 20 parts of ethyl acetate and the extract is washed with water, dried and evaporated to dryness. The residue is recrystallized from ethyl acetate. Thus one obtains
1 - (4-Benzamidophenoxy) -3-isopropylamino-2-propanol (m.p. 16211630 C).
This procedure is repeated, with the difference that the corresponding 3-alkylamino-1- (4-aminophenoxy) -2-propanol is used instead of 1- (4-aminophenoxy) -3-isopropylamino-2-propanol. So we get:
