Verfahren zur Herstellung von Aryloxy-isoalkyl-A2-imidazolinen
Man kennt bereits eine Reihe von Imidazolin-Deriw vaten, die in 2-Stellung durch Aryloxy-alkylgruppen mit unverzweigter Alkylgruppe substituiert sind. Diese Verbindungen zeichnen sich durch überwiegend vasokon striktorische, spasmolytische und Antihistaminwirkung aus.
Es wurde nun gefunden, dass sich die neuen, bisher noch nicht beschriebenen Aryloxy-isoalkyl-imidazoline der allgemeinen Formel I

in der Ar ein Phenyl-, Naphthyl- oder partiell hydrierter Naphthylrest ist, der im aromatischen Ring mit einem oder mehreren aliphatischen Kohlenwasserstoffresten, einem oder mehreren Aryl- und/oder Aralkylresten, einer oder mehreren Trifluormethyl-, Nitro-, Amino-, Hydroxy- und/oder niederen Alkoxygruppen, einem oder mehreren Halogenatomen und/oder (aus zwei orthoständigen Substituenten zusammen) einer Methylendioxygruppe substituiert sein kann, R1 ein unverzweigter oder verzweigter Kohlenwasserstoffrest mit 1 bis 4 C Atomen ist, wenn R3 Wasserstoff oder n 0 ist, oder Wasserstoff ist, wenn R3 nicht Wasserstoff ist, R2, R3 und R4 Wasserstoff oder niedere, unverzweigte oder verzweigte Alkylgruppen sind und n 0 oder list,
und deren Säureadditionssalze durch neue und nicht vorauszusehende pharmakologische Eigenschaften auszeichnen. Diese neuen Substanzen sollen als Pharmazeutika Verwendung finden.
Wegen ihrer guten Eigenschaften sind diejenigen Verbindungen der allgemeinen Formel I bevorzugt, in denen R4 Wasserstoff ist und n vorzugsweise 0 ist.
Wegen ihrer guten Eigenschaften sind dabei diejenigen Verbindungen bevorzugt, in denen Ar ein gegebenenfalls durch eine oder mehrere Niederalkyl- und/oder Niederalkoxy-Reste, eine oder mehrere Trifluormethyl Gruppen und/oder ein oder mehrere Halogenatome, vorzugsweise Chloratome substituierter Phenylrest, R1 ein Niederalkylrest mit 1 bis 4 C-Atomen und R. Wasserstoff oder ein Niederalkylrest mit 1 bis 4 C-Atomen ist.
Ganz besonders bevorzugt sind dabei die Verbindungen der allgemeinen Formel I, in denen Ar ein gegebenenfalls durch einen Niederalkyl-, Niederalkoxy- und/ oder Trifluormethyl-Rest und/oder ein Halogenatom, vorzugsweise ein Chloratom substituierter Phenylrest, Rl ein Niederalkylrest mit 1 bis 4 C-Atomen, R2 Wasserstoff oder ein Niederalkylrest mit 1 bis 4 C-Atomen, n 0 und R4 Wasserstoff ist.
Man kann die neuen Substanzen und deren Säureadditionssalze, insbesondere mit physiologisch unbedenklichen Säuren, nach folgendem Verfahren herstellen:
Als Ausgangsmaterial werden Aryloxycarbonsäuren mit verzweigter Alkylkette der Formel II

oder eines ihrer funktionellen Derivate verwendet und mit einem Äthylendiamin der Formel III: H2N-CH2 -CH2-NHR4 selbst oder mit einem reaktiven N-Derivat des Äthylendiamins umgesetzt. Bei dieser Reaktion bilden sich direkt oder stufenweise die neuen Imidazolin Derivate der Formel I.
Als funktionelle Säurederivate können z.B - Ester, Orthoester, Säurehalogenide, vorzugsweise das Säurechlorid, Amide, Thioamide, Amidine, Imidsäureester, Thioimidsäureester, Iminohalogenide oder die Nitrile der entsprechenden Aryloxy-isoalkylcarbonsäuren verwendet werden. Dabei können die Reaktionsbedingungen auch so gewählt werden, dass die funktionellen Säurederivate erst während der Reaktion gebildet werden.
Neben dem Äthylendiamin selbst können auch reaktive N-Derivate dieses Äthylendiamins verwendet werden.
Als solche kommen insbesondere Abkömmlinge dieses Äthylendiamins in Frage, die bei der Umsetzung mit Carbonsäuren bzw. deren funktionellen Derivaten Imidazoline ergeben. Verbindungen dieses Typs sind z.B. N-Acyläthylendiamin, N,N' -Diacyläthylendiamine oder Äthylenharnstoff.
Äthylendiamin bzw. dessen Derivate können entweder als freie Basen oder in Form ihrer Mono- oder Disalze in der 0,8 bis 1,2fachen stöchiometrischen Menge angewendet werden. Zum Beispiel kann man das Äthylendiamin als mono-p-toluolsulfonsaures Salz zur Anwendung bringen.
Verwendet man als Ausgangsmaterial das Nitril der entsprechenden Aryloxy-isoalkyl-carbonsäure und setzt dieses mit dem Äthylendiamin oder seinen Derivaten um, so ist es vorteilhaft, die Umsetzung in Gegenwart von Schwefelwasserstoff bzw. schwefelwasserstoffabgebenden Mitteln, wie z.B. Schwefelkohlenstoff, durchzuführen.
Die geschilderten Umsetzungen können direkt oder stufenweise zu den Imidazolin-Derivaten der Formel I führen. So kann z.B. intermediär eine Verbindung der Formel IV re, Äthansulfonsäure, Benzolsulfonsäure oder p-Toluolsulfonsäure, aromatische Carbonsäuren, wie Benzoesäure, Salicylsäure oder p-Aminosalicylsäure.
Die nach dem erfindungsgemässen Verfahren erhaltenen neuen Imidazolin-Derivate sowie deren Säureadditionssalze lassen sich zu allen pharmazeutischen Zubereitungsformen, gegebenenfalls unter Verwendung der üblichen Hilfsstoffe, verarbeiten.
Die neuen Imidazolin-Derivate haben u.a. neben den bekannten Eigenschaften dieser Stoffklasse als Antihistaminika und Spasmolytika lokalanästhetische Wirkung am Herzen. Insbesondere sind sie vasoaktiv und sind teils als Antihypertonika, teils als Vasokonstriktoren in Form von Nasentropfen oder Nasensprays verwendbar. Die folgenden Beispiele sollen die Herstellung der neuen Verbindungen der allgemeinen Formel I näher erläutern:
Beispiel I a) a-Phenoxypropionitri1 19 g Phenol, 26 g wasserfreies, fein gepulvertes Kaliumcarbonat und 0,5 g Kaliumjodid werden mit 100 ml trockenem Äthylmethylketon versetzt.
Innerhalb von 172 Stunden wird eine Lösung von 29,5 g a-Brompropionitril in 20ml absol. Äthylmethylketon zu der kräftig gerührten, unter Rückfluss kochenden Suspension zugetropft. Nach beendeter Zugabe wird noch eine weitere Stunde am Rückfluss erhitzt, nach dem Abkühlen vom Unlöslichen abgesaugt, der Rückstand mit Äthylmethylketon gewaschen und das Filtrat im Vakuum eingedampft. Der ölige Rückstand wird mit Äther versetzt, die ätherische Lösung zur Entfernung von überschüssigem Phenol dreimal mit je 50 ml 10%der Natronlauge und dann zur Entfernung des Jods mit 50ml 5%iger

in der X1 = OH, SH oder NH2 und Y1 = H oder Acyl bedeutet, entstehen. Aus Verbindungen dieses Typs bzw.
aus deren tautomeren Formen können nach an sich bekannten Methoden die Substituenten X1 und Y1 als X1Yl abgespalten werden. ZB. kann man aus einer Verbindung der Formel IV, worin X1 = OH und Y1 = H bedeuten, Wasser abspalten unter Verwendung von Calciumoxyd als Dehydratisierungsmittel. Dabei bildet sich in guter Ausbeute das entsprechende Imidazolin Derivat der Formel I.
Je nach der verwendeten Aufarbeitungsmethode erhält man die neuen Imidazolin-Derivate als freie Basen oder in Form ihrer Säureadditionssalze. Aus der freien Base können durch Behandlung mit Säuren die verschiedensten Säureadditionssalze nach üblichen Methoden hergestellt werden. Für die Herstellung derartiger Salze kommen solche Säuren in Frage, die physiologisch unbedenkliche Säureadditionssalze ergeben, wie z.B.
Halogenwasserstoffsäuren, Schwefelsäure, Orthophosphorsäure, Alkancarbonsäuren, wie Essigsäure oder Propionsäure, mehrbasische Säuren, wie z.B. Oxalsäure, Weinsäure, Bernsteinsäure, Maleinsäure, Ascorbinsäure oder Zitronensäure, Sulfonsäure, wie Methansulfonsäu Natriumthiosulfatlösung ausgeschüttelt, mit Wasser bis zur neutralen Reaktion gewaschen und über wasserfreiem Natriumsulfat getrocknet. Nach dem Abziehen des Lösungsmittels ergab die Fraktionierung des zurückbleibenden Öls im Hochvakuum 21 g cc-Phenoxypropionitril.
Ausbeute: 71,5% der Theorie.

44g a-Phenoxypropionitril werden mit 11,5ml absol. Äthanol versetzt, 100 ml über Phosphorpentoxyd getrocknetes Chloroform zugegeben und langsam unter Rühren und Kühlung mit Eis/Kochsalz 11,5 g sorgfältig getrockneter Chlorwasserstoff eingeleitet. Danach wird das Chloroform und überschüssiger Chlorwasserstoff im Vak. bei Zimmertemperatur zum grössten Teil abgesaugt und der Rückstand bis zur quantitativen Fällung des Imidsäureesterhydrochlorids mit trockenem Äther versetzt.
Durch mehrmaliges Lösen in wenig absol. Äthanol ohne Erwärmen und Ausfällen mit Äther kann das a - (Phenoxy)-propionimidsäureäthylester-hydrochlorid in Form von weissen, stark hygroskopischen Kristallen analysenrein erhalten werden.
Schmelzpunkt: 83-85 C.
Ausbeute: 60 g (87,5% der Theorie)

Das Rohprodukt des a-(Phenoxy)-propioimidsäure äthylester-hydrochlorids ist auch ohne vorherige Ausfällung mit Äther für weitere Umsetzungen genügend rein.
c) 2-[α-(Phenoxy)-äthyI] -##-imidazolin-hydrochlorid
23 g a-(Phenoxy)-propionimidsäureäthylester-hydro- chlorid werden portionsweise zu einer gerührten und eisgekühlten Lösung von 6 g wasserfreiem Äthylendiamin in 90 ml absol. Äthanol so hinzugegeben, dass die Temperatur 0 bis 50C nicht übersteigt. Danach wird das Kältebad entfernt und das Reaktionsgemisch innerhalb einer Stunde im Wasserbad auf ca. 700C erwärmt.
Nach erneuter Abkühlung wird in einer Kältemischung nicht umgesetztes Äthylendiamin mit absol. äthanol.
Salzsäure neutralisiert, dann von den äthanolunlöslichen
Bestandteilen abfiltriert und etwa 2/3 des Lösungsmittels im Wasserstrahlpumpenvakuum abgesaugt. Durch vorsichtigen Zusatz von Äthylmethylketon werden restliche Mengen Äthylendiamindihydrochlorid fraktioniert ausgefällt und dann durch Zugabe von trockenem ther das Imidazolinhydrochlorid abgeschieden. Nach dem Umkristallisieren aus wenig Äthylmethylketon erhält man das 2-[ -(Phenoxy)-äthyl] -h2-imidazolin-hydrochlo- rid in Form von kleinen weissen Nadeln vom Schmelz punkt: 144,5-146 C.

Beispiel 2
17,7 g a-(o-Methoxyphenoxy)-propionitril (hergestellt durch Umsetzung von Guajacol mit cc-Brompropionitril in Gegenwart von Kaliumcarbonat; wasserhelle Flüssigkeit, Sdp.o,2: 98,5-100,5 , nD25: 1,5110) und 23,2 g Äthylendiamin-mono-p-toluolsulfonat werden zusammengegeben und eine Stunde auf 225-2300C erhitzt. Dabei wird NH1 abgespalten. Nach dem Erkalten des Reaktionsgemisches wird mit etwa 10%iger Lauge alkalisch gemacht und die freie Base mit Chloroform extrahiert.
Der nach dem Abziehen des Lösungsmittels verbleibende Rückstand ergab nach der Umkristallisation aus Petro- leumbenzin (100/140)15,8 (72So d.Th.) 2[α-(o-Meth- oxyphenoxy)-äthyl]-A2-imidazolin. Smp. 94,5-95 C.

Die freie Base kann in bekannter Weise in das Hydrochlorid übergeführt werden, wobei nach der Umkri stallisation aus Äthanol/Äthylmethylketon 15,7 g (85,5%) 2 - [ - (o-Methoxyphenoxy)-äthyl] - A2 - imidazolin-hydrochlorid erhalten werden. Weisse, stark hygroskopische Kristalle vom Schmelzpunkt: 136-1370C.

Beispiel 3
17,7 g -(p-Methoxyphenoxy)-propionitril (hergestellt durch Umsetzung von Hydrochinonmonomethyläther mit ce-Brompropionitril in Gegenwart von Kaliumcarbonat; wasserhelle Flüssigkeit vom Sdp.o,ol 84-860C, nD26: 1,5109) 6 g Äthylendiamin und 0,5 ccm Schwefelkohlenstoff werden zusammengegeben und 48 Stunden auf etwa 1000C erhitzt.
Dabei entweicht NH5. Das erstarrte Reaktionsprodukt wird nach dem Erkalten aus Petroleumbenzin (100/140) umkristallisiert, wobei man 14,7g (67% der Theorie) 2-[α-(p-Methoxyphenoxy)-äthyl]-#2- -imidazolin vom Smp. 113,5-114,50C erhält.

Beispiel 4
57,5 g a-(2,6-Dichlorphenoxy)-propionitril (hergestellt wie nach'Beispiel la aus 2,6-Dichlorphenol und oc-Brom- propionitril; weisse Nadeln vom Smp.: 28-290C) werden wie nach Beispiel 1b mit 10,4ccm absol. Äthanol und 10,4 g trockenem Chlorwasserstoffgas in α-(2,6-Dichlor- phenoxy)-propionimidsäureäthylester-hydrochlorid überführt, das ohne weitere Isolierung und Reinigung nach Beispiel 1c mit 29,5 g wasserfreiem Äthylendiamin zum 2 - [oc - (2,6-Dichlorphenoxy)-äthyl] A2 imidazolin-hydrochlorid umgesetzt wird.
Nach mehrmaligem Umkristallisieren aus Äthanol/Äther erhält man weisse Kristalle vom Smp.: 221-2230C.

Beispiel 5
39,9 g α-(2,3-Dimethylphenoxy)-propionitril (hergestellt wie nach Beispiel la aus vic.-o-Xylenol und a Brompropionitril; wasserhelle Flüssigkeit, Sdp.0,06: 62aC, nD20: 1,5108) werden wie nach Beispiel 1b mit 8,5 ccm absol. Äthylalkohol und 8,5g trockenem Chlorwasserstoffgas in -(2,3 -Dimethylphenoxy)-propionimidsäure- äthylester-hydrochlorid überführt, das ohne weitere Reinigung wie unter Beispiel 1c umgesetzt und aufgearbeitet wird.
Das anfallende Rohprodukt wird zur Reinigung durch Aufnehmen in 10%iger Natronlauge in die freie Imidazolin-Base (Smp.: 106,50C) überführt, die nach Ausschütteln mit Äther, Trocknen und Abziehen des Lösungsmittels, Lösen in absol. Alkohol und Zusatz von absol. Äther durch Einleiten von mindestens der äquivalenten Menge trockenem Chlorwasserstoffgas wieder zum Hydrochlorid umgesetzt wird. Das anfallende 2- [oc-(2,3-Dimethylphenoxy)-äthyl]-A2-imidazolin-hydro- chlorid wird nach einmaliger Umkristallisation aus Alkohol/Äther auf diese Weise analysenrein als weisse Kristalle vom Smp.: 1830C erhalten.

Process for the preparation of aryloxy-isoalkyl-A2-imidazolines
A number of imidazoline derivatives are already known which are substituted in the 2-position by aryloxy-alkyl groups with unbranched alkyl groups. These compounds are characterized by a predominantly vasoconstrictive, spasmolytic and antihistamine action.
It has now been found that the new, hitherto not yet described aryloxy-isoalkyl-imidazolines of the general formula I

in which Ar is a phenyl, naphthyl or partially hydrogenated naphthyl radical which, in the aromatic ring, has one or more aliphatic hydrocarbon radicals, one or more aryl and / or aralkyl radicals, one or more trifluoromethyl, nitro, amino, hydroxy and / or lower alkoxy groups, one or more halogen atoms and / or (from two ortho substituents together) a methylenedioxy group, R1 is an unbranched or branched hydrocarbon radical with 1 to 4 C atoms, when R3 is hydrogen or n 0, or hydrogen if R3 is not hydrogen, R2, R3 and R4 are hydrogen or lower, unbranched or branched alkyl groups and n is 0 or 1,
and their acid addition salts are characterized by new and unforeseeable pharmacological properties. These new substances are to be used as pharmaceuticals.
Because of their good properties, those compounds of the general formula I in which R4 is hydrogen and n is preferably 0 are preferred.

Functional acid derivatives that can be used are, for example, esters, orthoesters, acid halides, preferably the acid chloride, amides, thioamides, amidines, imidic esters, thioimidic esters, iminohalides or the nitriles of the corresponding aryloxy-isoalkylcarboxylic acids. The reaction conditions can also be chosen so that the functional acid derivatives are only formed during the reaction.
In addition to the ethylenediamine itself, reactive N-derivatives of this ethylenediamine can also be used.
As such, derivatives of this ethylene diamine are particularly suitable, which give imidazolines when reacted with carboxylic acids or their functional derivatives. Compounds of this type are e.g. N-acylethylenediamine, N, N '-diacylethylenediamine or ethylene urea.
Ethylenediamine or its derivatives can be used either as free bases or in the form of their mono- or di-salts in 0.8 to 1.2 times the stoichiometric amount. For example, ethylenediamine can be used as a mono-p-toluenesulfonic acid salt.
If the nitrile of the corresponding aryloxy-isoalkyl-carboxylic acid is used as the starting material and this is reacted with the ethylenediamine or its derivatives, it is advantageous to carry out the reaction in the presence of hydrogen sulfide or hydrogen sulfide-releasing agents, e.g. Carbon disulfide to perform.
The reactions described can lead to the imidazoline derivatives of the formula I directly or in stages. E.g. intermediate a compound of the formula IV re, ethanesulfonic acid, benzenesulfonic acid or p-toluenesulfonic acid, aromatic carboxylic acids such as benzoic acid, salicylic acid or p-aminosalicylic acid.
The new imidazoline derivatives obtained by the process according to the invention and their acid addition salts can be processed into all pharmaceutical preparation forms, optionally using the customary auxiliaries.
The new imidazoline derivatives have i.a. In addition to the known properties of this class of substances as antihistamines and antispasmodics, local anesthetic effects on the heart. In particular, they are vasoactive and can be used partly as antihypertensive agents, partly as vasoconstrictors in the form of nasal drops or nasal sprays. The following examples are intended to explain the preparation of the new compounds of general formula I in more detail:

44g of a-phenoxypropionitrile are with 11.5ml absol. Ethanol is added, 100 ml of chloroform dried over phosphorus pentoxide are added and 11.5 g of carefully dried hydrogen chloride are slowly passed in, while stirring and cooling with ice / common salt. Then the chloroform and excess hydrogen chloride in vac. For the most part suctioned off at room temperature and the residue is mixed with dry ether until the imidate hydrochloride has precipitated quantitatively.
By loosening several times in a little absolute Ethanol without heating and precipitation with ether, the a- (phenoxy) -propionimidic acid ethyl ester hydrochloride can be obtained analytically pure in the form of white, strongly hygroscopic crystals.
Melting point: 83-85 C.
Yield: 60 g (87.5% of theory)

The crude product of a- (phenoxy) -propioimidic acid ethyl ester hydrochloride is sufficiently pure for further reactions, even without prior precipitation with ether.
c) 2 - [α- (Phenoxy) ethyI] - ## - imidazoline hydrochloride
23 g of ethyl a- (phenoxy) propionimidate hydrochloride are added in portions to a stirred and ice-cooled solution of 6 g of anhydrous ethylenediamine in 90 ml of absol. Ethanol added so that the temperature does not exceed 0 to 50C. The cold bath is then removed and the reaction mixture is heated to approx. 70 ° C. in a water bath within one hour.
After cooling down again, unreacted ethylenediamine is combined with absol. ethanol.
Hydrochloric acid neutralized, then from the ethanol-insoluble
Components filtered off and about 2/3 of the solvent sucked off in a water jet pump vacuum. By carefully adding ethyl methyl ketone, remaining amounts of ethylenediamine dihydrochloride are precipitated fractionally and the imidazoline hydrochloride is then deposited by adding dry ether. After recrystallization from a little ethyl methyl ketone, 2- [- (phenoxy) -ethyl] -h2-imidazoline hydrochloride is obtained in the form of small white needles with a melting point of 144.5-146 C.

Example 2
17.7 g of a- (o-methoxyphenoxy) -propionitrile (prepared by reacting guaiacol with cc-bromopropionitrile in the presence of potassium carbonate; water-white liquid, boiling point o, 2: 98.5-100.5, nD25: 1.5110 ) and 23.2 g of ethylenediamine mono-p-toluenesulfonate are combined and heated to 225-2300C for one hour. In the process, NH1 is split off. After the reaction mixture has cooled, it is made alkaline with about 10% caustic and the free base is extracted with chloroform.
The residue remaining after the solvent had been stripped off gave, after recrystallization from petroleum benzine (100/140) 15.8 (72% of theory) 2 [α- (o-methoxyphenoxy) ethyl] -A2-imidazoline . M.p. 94.5-95 C.

White, strongly hygroscopic crystals with a melting point of 136-1370C.

17.7 g - (p-methoxyphenoxy) propionitrile (produced by reacting hydroquinone monomethyl ether with ce-bromopropionitrile in the presence of potassium carbonate; water-white liquid of boiling point o, ol 84-860C, nD26: 1.5109) 6 g ethylenediamine and 0 , 5 cc of carbon disulfide are combined and heated to about 1000C for 48 hours.
NH5 escapes in the process. After cooling, the solidified reaction product is recrystallized from petroleum benzine (100/140), 14.7 g (67% of theory) of 2 - [α- (p-methoxyphenoxy) ethyl] - # 2 -imidazoline of mp. 113.5-114.50C.

Example 4
57.5 g of α- (2,6-dichlorophenoxy) propionitrile (prepared as according to Example la from 2,6-dichlorophenol and oc-bromopropionitrile; white needles with a melting point of 28-290C) are obtained as according to Example 1b with 10.4ccm absolute Ethanol and 10.4 g of dry hydrogen chloride gas are converted into α- (2,6-dichlorophenoxy) propionimidic acid ethyl ester hydrochloride, which without further isolation and purification according to Example 1c with 29.5 g of anhydrous ethylenediamine to give 2 - [oc - ( 2,6-dichlorophenoxy) ethyl] A2 imidazoline hydrochloride is implemented.
After repeated recrystallization from ethanol / ether, white crystals with a melting point of 221-2230C are obtained.

Example 5
39.9 g of α- (2,3-dimethylphenoxy) propionitrile (prepared as in Example la from vic.-o-xylenol and a bromopropionitrile; water-white liquid, bp 0.06: 62aC, nD20: 1.5108) are as in Example 1b with 8.5 ccm absol. Ethyl alcohol and 8.5 g of dry hydrogen chloride gas are converted into - (2,3-dimethylphenoxy) propionimidic acid ethyl ester hydrochloride, which is reacted and worked up as in Example 1c without further purification.
For purification, the crude product obtained is converted into the free imidazoline base (melting point 106.50C) by taking it up in 10% sodium hydroxide solution, which after shaking with ether, drying and removal of the solvent, dissolving in absolute. Alcohol and addition of absol. Ether is converted back to the hydrochloride by introducing at least the equivalent amount of dry hydrogen chloride gas. The resulting 2- [oc- (2,3-dimethylphenoxy) -ethyl] -A2-imidazoline hydrochloride is obtained in this way analytically pure as white crystals with a melting point of 1830C after one recrystallization from alcohol / ether.
