Verfahren zur Herstellung von 11,12-Dehydrosteroiden
Die Erfindung betrifft ein Verfahren zur Herstellung von in 12-Stellung unsubstituierten 11,12-Dehydrosteroiden aus 12-Ketosteroiden.
11,12-Dehydrosteroide, z.B. 11,12-Dehydrosapogenine, sind bei der Herstellung von therapeutisch wirksamen Steroiden brauchbar, wobei man sie erst in das entsprechende 9(1 1)-Dehydrosteroid überführt.
Es wurde schon vorgeschlagen (J.C.S. 1954, 1739), Hecogeninacetat in 11,12-Dehydrotigogenin zu überführen, indem man es in das entsprechende Toluol-p-sulfonylhydrazonderivat umwandelt und dieses in einem Hydroxylgruppen enthaltenden Lösungsmittel mit Ätzalkali behandelt. Man erzielt dabei aber sehr geringe Ausbeuten, höchstens 25% der Theorie.
Es wurde gefunden, dass man wesentlich bessere Ausbeuten mit einem vorzugsweise keine Hydroxylgruppen enthaltenden stark basischen Mittel in einem keine Hydroxylgruppen enthaltenden organischen Lösungsmittel erhält. Da die 11,12-Dehydrosteroide auf anderem Wege nur schwer erhältlich sind, ist dieser Befund von grosser Bedeutung.
Die Erfindung betrifft daher ein Verfahren zur Herstellung von in 12-Stellung unsubstituierten 11,1 2-Dehy- drosteroiden, bei welchem man das Sulfonylhydrazon eines 12-Keto-steroids, insbesondere eines 1 2-Keto-sapo- genins, dessen Hydrazinrest vorzugsweise mit einem Kohlenwasserstoffrest substituiert ist, mit einem stark basischen, vorzugsweise keine Hydroxylgruppen enthaltenden, Mittel in einem organischen Lösungsmittel umsetzt, das keine Hydroxylgruppen enthält. Man erhält dabei die entsprechenden 11,1 2-Dehydro-steroide.
Da Hecogenon das wichtigste 12-Keto-steroid ist, wird das erfindungsgemässe Verfahren am Beispiel des Hecogenins und dessen Acetat ausführlicher beschrieben. Es können aber auch andere 12-Keto-Sapogenine, z.B. Botogenin oder 3P-Hydroxy-Sa-pregn- 16-en-12,20-dion und 3-Hydroxy-5-pregnan 12,20-dion, deren 3ss-Acetoxy- derivate sowie deren 5S-Isomere, verwendet werden,
Das stark basische Mittel bewirkt die Ionisation der sauren NH-Gruppe im Sulfonylhydrazonrest. Das Mittel ist vorzugsweise ein niederes Alkoxyd eines Alkalimetalls, z.B. Natrium- oder Kaliummethoxyd.
Andere brauchbare stark basische Mittel sind die Alkalimetallhydride, Alkalimetallacetamide (hergestellt durch Lösen des Alkalimetalls in Acetamid, wobei das dabei erhaltene Reaktionsgemisch direkt verwendet werden kann), Alkalimetallamide, und die Alkalimetallverbindungen von Kohlenwasserstoffen, z.B. Triphenylmethylnatrium, Butyllithium und Methyllithium. In derartigen Verbindungen vorhandene Carbonylgruppen können alkyliert oder durch Ketalisation geschützt werden.
Das stark basische Mittel soll im allgemeinen im Überschuss, vorzugsweise drei Äquivalente auf 1 Äquivalent Steroid, verwendet werden.
Verwendet man Alkalimetallverbindungen von Kohlenwasserstoffen, wird die Umsetzung im allgemeinen bei einer Temperatur von mindestens 1000C durchgeführt und es soll daher ein organisches Lösungsmittel Verwendung finden, das bei mindestens 1000 siedet. Als hydroxylfreie organische Lösungsmittel kommen dabei z.B.
N,N-Dimethylformamid, N-Methylpyrrolidon, Acetamid, N,N-Dimethylacetamid und Diäthylenglykoldiäthyläther in Frage.
In solchen Lösungsmitteln beginnt die Reaktion bei etwa 1000C und schreitet bei 130-1400C im allgemeinen rasch fort. Die Reaktion kann im allgemeinen zwischen 100 und 2000C, vorzugsweise zwischen 140 und 1850C, durchgeführt werden, da bei diesen Temperaturen die Bildung von Nebenprodukten gering ist.
Besonders gute Ergebnisse wurden bei Verwendung von Natriummethoxyd als stark basisches Mittel und N, N-Dimethylformamid oder N-Methylpyrrolidon als Lösungsmittel erzielt. Weitere gut wirkende Kombinationen von Base und Lösungsmittel sind Natriumhydrid in Di äthylenglykoldiäthyläther und Natriumacetamid in Acetamid.
Verwendet man Alkalimetallderivate von Kohlenwasserstoffen als stark basische Mittel, ist es zweckmässig, die Umsetzung bei tiefen Temperaturen, etwa zwischen -253C und +255C, durchzuführen, obwohl man auch bei der Siedetemperatur des Lösungsmittels arbeiten kann.
Man verwendet Triphenylmethylnatrium, Methyllithium und Butyllithium, zweckmässig in einem Kohlenwasserstoff als Lösungsmittel, z.B. in n-Hexan oder Benzol, es sind aber auch Äther verwendbar, z.B. Diäthyläther, Dioxan und Tetrahydrofuran.
Das dem Hydrazon zugrunde liegende Sulfonylhydrazin hat z.B. die Formel R-SO2-NH-NH2, worin R einen Kohlenwasserstoffrest mit 1 bis 10 Kohlenstoffatomen bedeutet. R kann ein Alkyl-, Alkaryl-, Aralkyl- oder Arylrest sein. Es kommen vorzugsweise Toluol-p-sulfonylhydrazin und Methansulfonylhydrazin in Frage.
Beispiel I Herstellimg von Sa,25D-Spirost-ll-en-3p-ol nrcs Hecogenis?vcetat-12-toZIsol-p-slflfonylAydrazot7 a) mit Natriummethoxyd in N,N-Dimethylformamid
Eine gerührte Suspension von 40 g des Toluol-p-sulfonylhydrazons von Hecogeninacetat und 36 g Natriummethoxyd in 800 ml N,N-Dimethylformamid wird in 15 Minuten auf ihren Siedepunkt erhitzt und nach weiteren 30 Minuten beim Siedepunkt wird das Gemisch auf 800C abgekühlt und mit Eis und Wasser auf ungefähr 10 Liter verdünnt.
25,5 g ausgefallenes Produkt, Schmelzpunkt 158 bis 1 680C, werden aus Methanol umkristallisiert und ergeben 5x,25D-Spirost-11-en-3k-ol in 76%iger Ausbeute, Schmelzpunkt 176 bis 1800C. Eine zweite Fraktion liefert nach Umkristallisation mehr von der Att-Verbindung (9,6%), Schmelzpunkt 188 bis 1900C. Beide Muster haben das erwartete Infrarotspektrum.
b) Mit Natriummethoxyd in N-Methylpyrrolidon.
1.0 g des Toluol-p-sulfonylhydrazons von Hecogeninacetat und 0.9 g Natriummethoxyd in 20 ml N-Methylpyrrolidon werden 10 Minuten auf 1700C erhitzt und nach weiteren 10 Minuten wird das erhaltene Gel auf ungefähr 600C abgekühlt und unter kräftigem Rühren mit Wasser verdünnt. Das ausgefallene Produkt (640 mg), Schmelzpunkt 168 bis 1700C, hat ein Infrarotspektrum, das dem des Produktes von a) ähnelt.
c) Mit Natriumhydrid in Diäthylenglykoldiäthyläther
2,0 g des Toluol-p-sulfonylhydrazons von Hecogeninacetat und eine 500/,ige Dispersion von Natriumhydrid in Mineralöl (430 mg) werden in 20 ml DiäthylenPlykoldi- äthvläther suspendiert und 10 Minuten in 20 ml Diäthylenglykoldiäthyläther suspendiert und 10 Minuten auf 1830C erhitzt. Nach weiteren 20 Minuten, während welcher Zeit das Gemisch beim Siedepunkt gehalten wird, wird es gekühlt, mit Äthanol behandelt, um überschüssiges Natriumhydrid zu zerstören, und mit Wasser bis auf 500 ml verdünnt. Das Produkt wird in Chloroform extrahiert und der Extrakt mit verdünnter Salzsäure, wässrigem Na tiumhydrogencarbonat und Wasser gewaschen.
Nach Ent femung des Lösungsmittels aus dem Extrakt verbleibt ein öliger Rückstand, der aus n-Hexan kristallisiert und 5a, 25D-Spirost-ll-en-3p-ol in 64%iger Ausbeute liefert. Es hat ein Infrarotspektrum, das demjenigen des Produktes aus a) ähnlich ist.
d) Mit Natriumacetamid.
403 mg Natrium werden im Vakuum in geschmolzenem Acetamid gelöst und zu dem abgekühlten Gemisch 2,0 g des Toluol-p-sulfonylhydrazons von Hecogeninacetat zugesetzt. Das Gemisch wird in 10 Minuten auf ungefähr 1500C erhitzt und nach weiteren 20 Minuten bei ungefähr 1500C wird es auf ungefähr 800C abgekühlt und mit Wasser bis zu ungefähr 300 ml verdünnt. 1,37 g ausgefallenes 5z,25D-Spirost-ll-en-3,3-ol haben ein Infrarotspektrum, das demjenigen des Produkts aus a) ähnlich ist.
Beispiel 2 a) Herstellung von Hecogeninacetat-12-methansulfonyl- hydrazon.
Eine Lösung von 4,0 g Hecogeninacetat in 90 ml Essigsäure bei 400C wird mit 2,0 g Methansulfonylhydrazin behandelt. Man lässt das Gemisch über Nacht bei Zimmertemperatur stehen und sammelt 1,33 g kristallines Methansulfonylhydrazon, Schmp. 260-263 C (Zers.), durch Filtrieren. Nach Verdünnen des Filtrats mit Wasser fallen weitere 3,42 g Mtthansulfonylhydrazon aus, Schmelzpunkt 258-2620C (Zers.). Nach Kristallisation aus Äthanol erhält man das analytische Muster, Schmelzpunkt 262-2660C (Zers.).

Gefunden: C 64,1 H 8,75 N 5,2 S 5,7 b) Zersetzung von Hecogeninacetat-12-methansulfonyl- hydrazon.
0,5 g des Methansulfonylhydrazons von Hecogeninacetat und 0,4 g Natriummethoxyd in Suspension in 10 ml N,N-Dimethylformamid werden in 10 Minuten auf ungefähr 1500C erhitzt und nach weiteren 26 Minuten bei 1500C wird die abgekühlte Suspension mit Eis und Wasser bis zu ungefähr 100 ml verdünnt. 378 mg ausgefallenes Produkt haben ein Infrarotspektrum, das anzeigt, dass es im wesentlichen ein Gemisch von 50ç,25D-Spirost- -ll-en-3p-ol mit dem entsprechenden 3-Acetat ist.
Process for the preparation of 11,12-dehydrosteroids
The invention relates to a process for the preparation of 11,12-dehydrosteroids which are unsubstituted in the 12-position from 12-keto steroids.
11,12-dehydrosteroids, e.g. 11,12-dehydrosapogenins can be used in the production of therapeutically effective steroids, whereby they are first converted into the corresponding 9 (11) -dehydrosteroid.
It has already been proposed (J.C.S. 1954, 1739) to convert hecogenin acetate into 11,12-dehydrotigogenin by converting it into the corresponding toluene-p-sulfonylhydrazone derivative and treating this with caustic alkali in a solvent containing hydroxyl groups. However, very low yields are achieved, at most 25% of theory.
It has been found that significantly better yields are obtained with a strongly basic agent preferably containing no hydroxyl groups in an organic solvent containing no hydroxyl groups. Since the 11,12-dehydrosteroids are difficult to obtain by other means, this finding is of great importance.
The invention therefore relates to a process for the preparation of 11,1 2-dehydrosteroids which are unsubstituted in the 12-position, in which the sulfonylhydrazone of a 12-keto-steroid, in particular a 1 2-keto-sapogenin, whose hydrazine radical is preferably also used a hydrocarbon radical, is reacted with a strongly basic, preferably non-hydroxyl-containing agent in an organic solvent which does not contain any hydroxyl groups. The corresponding 11,1 2-dehydro-steroids are obtained.
Since hecogenone is the most important 12-keto steroid, the method according to the invention is described in more detail using the example of hecogenin and its acetate. However, other 12-keto-sapogenins, e.g. Botogenin or 3P-hydroxy-Sa-pregn- 16-en-12,20-dione and 3-hydroxy-5-pregnan 12,20-dione, their 3ss-acetoxy derivatives and their 5S isomers are used,
The strongly basic agent causes the ionization of the acidic NH group in the sulfonylhydrazone residue. The agent is preferably a lower alkoxide of an alkali metal, e.g. Sodium or potassium methoxide.
Other useful strong base agents are the alkali metal hydrides, alkali metal acetamides (prepared by dissolving the alkali metal in acetamide, the resulting reaction mixture can be used directly), alkali metal amides, and the alkali metal compounds of hydrocarbons, e.g. Triphenylmethyl sodium, butyllithium and methyl lithium. Carbonyl groups present in such compounds can be alkylated or protected by ketalization.
The strongly basic agent should generally be used in excess, preferably three equivalents to 1 equivalent steroid.
If alkali metal compounds of hydrocarbons are used, the reaction is generally carried out at a temperature of at least 1000.degree. C., and an organic solvent that boils at at least 1000 should therefore be used. Hydroxyl-free organic solvents are e.g.
N, N-dimethylformamide, N-methylpyrrolidone, acetamide, N, N-dimethylacetamide and diethylene glycol diethyl ether in question.
In such solvents the reaction begins at about 1000C and generally proceeds rapidly at 130-1400C. The reaction can generally be carried out between 100 and 2000 ° C., preferably between 140 and 1850 ° C., since the formation of by-products is low at these temperatures.
Particularly good results have been achieved when using sodium methoxide as a strongly basic agent and N, N-dimethylformamide or N-methylpyrrolidone as solvent. Other effective combinations of base and solvent are sodium hydride in diethylene glycol diethyl ether and sodium acetamide in acetamide.
If alkali metal derivatives of hydrocarbons are used as strongly basic agents, it is advisable to carry out the reaction at low temperatures, for example between -253C and +255C, although it is also possible to work at the boiling point of the solvent.
Triphenylmethylsodium, methyllithium and butyllithium are used, conveniently in a hydrocarbon solvent, e.g. in n-hexane or benzene, but ethers can also be used, e.g. Diethyl ether, dioxane and tetrahydrofuran.
The sulfonylhydrazine on which the hydrazone is based has e.g. the formula R-SO2-NH-NH2, where R is a hydrocarbon radical having 1 to 10 carbon atoms. R can be an alkyl, alkaryl, aralkyl or aryl radical. Toluene-p-sulfonylhydrazine and methanesulfonylhydrazine are preferred.
EXAMPLE I Preparation of Sa, 25D-Spirost-II-en-3p-ol nrcs Hecogenisol acetate-12-toZIsol-p-sulfonylAydrazot7 a) with sodium methoxide in N, N-dimethylformamide
A stirred suspension of 40 g of the toluene-p-sulfonylhydrazone of hecogenin acetate and 36 g of sodium methoxide in 800 ml of N, N-dimethylformamide is heated to its boiling point in 15 minutes and after a further 30 minutes at the boiling point, the mixture is cooled to 80 ° C. and with ice and water diluted to about 10 liters.
25.5 g of precipitated product, melting point 158 to 1680C, are recrystallized from methanol and give 5x, 25D-spirost-11-en-3k-ol in 76% yield, melting point 176 to 1800C. A second fraction provides more of the Att compound (9.6%) after recrystallization, melting point 188 to 1900C. Both patterns have the expected infrared spectrum.
b) With sodium methoxide in N-methylpyrrolidone.
1.0 g of the toluene-p-sulfonylhydrazone of hecogenin acetate and 0.9 g of sodium methoxide in 20 ml of N-methylpyrrolidone are heated to 1700C for 10 minutes and after a further 10 minutes the gel obtained is cooled to about 600C and diluted with water while stirring vigorously. The precipitated product (640 mg), melting point 168 to 1700 ° C., has an infrared spectrum which is similar to that of the product from a).
c) With sodium hydride in diethylene glycol diethyl ether
2.0 g of the toluene-p-sulfonylhydrazone of hecogenin acetate and a 500% dispersion of sodium hydride in mineral oil (430 mg) are suspended in 20 ml of diethylene glycol diethyl ether and suspended in 20 ml of diethylene glycol diethyl ether for 10 minutes and heated to 1830C for 10 minutes. After a further 20 minutes, during which time the mixture is held at the boiling point, it is cooled, treated with ethanol to destroy excess sodium hydride and diluted with water to 500 ml. The product is extracted into chloroform and the extract washed with dilute hydrochloric acid, aqueous sodium hydrogen carbonate and water.
After removal of the solvent from the extract, an oily residue remains which crystallizes from n-hexane and gives 5a, 25D-Spirost-II-en-3p-ol in 64% yield. It has an infrared spectrum which is similar to that of the product from a).
d) With sodium acetamide.
403 mg of sodium are dissolved in molten acetamide in vacuo and 2.0 g of the toluene-p-sulfonylhydrazone of hecogenin acetate are added to the cooled mixture. The mixture is heated to about 1500C in 10 minutes and after a further 20 minutes at about 1500C it is cooled to about 800C and diluted with water up to about 300 ml. 1.37 g of precipitated 5z, 25D-Spirost-II-en-3,3-ol have an infrared spectrum which is similar to that of the product from a).
Example 2 a) Preparation of hecogenin acetate-12-methanesulfonylhydrazone.
A solution of 4.0 g of hecogenin acetate in 90 ml of acetic acid at 40 ° C. is treated with 2.0 g of methanesulfonylhydrazine. The mixture is left to stand overnight at room temperature and 1.33 g of crystalline methanesulfonylhydrazone, melting point 260-263 ° C. (decomp.), Is collected by filtration. After diluting the filtrate with water, a further 3.42 g of methanesulfonylhydrazone precipitate, melting point 258-2620C (decomp.). The analytical specimen is obtained after crystallization from ethanol, melting point 262-2660C (decomp.).

0.5 g of the methanesulfonylhydrazone of hecogenin acetate and 0.4 g of sodium methoxide in suspension in 10 ml of N, N-dimethylformamide are heated to about 1500C in 10 minutes and after a further 26 minutes at 1500C the cooled suspension is mixed with ice and water up to about 100 ml diluted. 378 mg of precipitated product has an infrared spectrum, which indicates that it is essentially a mixture of 50ç, 25D-Spirost-III-en-3p-ol with the corresponding 3-acetate.
