mit einem Aldehyd der Formel R2-CHO oder einem funktionellen Derivat davon, wie einem reaktionsfähigen funktionellen Derivat oder einem Polymeren davon, behandelt und reduziert.
Die vorliegende Erfindung betrifft nun die Herstellung von solchen Verbindungen obiger Formel, worin R, den Methylrest, R2 den Isobutyl- oder den Chlormethylrest und R3 ein Chloratom bedeutet, und Salzen davon, besonders Alkalimetallsalzen, wie Natrium- oder Kaliumsalzen.
Die neuen Verbindungen zeigen eine hohe diuretische und natriuretische Wirksamkeit und sollen als Heilmittel Verwendung finden. Unter diesen Verbindungen sind noch besonders das 2-Methyl-3-isobutyl-6-chlor-7-sulfamyl- 3,4-dihydro-1,2,4-benzothiadiazin-1,1-dioxyd, wie auch seine Alkalimetallsalze zu erwähnen, die sich durch eine besonders hohe Wirksamkeit auszeichnen.
Die neuen Verbindungen sollen als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit den pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägersubstanzen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Propylenglykol, Polyalkylenglykole oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe, z. B. hypotensive Mittel, enthalten, wie Rauwolfia- oder Veratrum-Alkaloide, beispielsweise Reserpin, Rescinnamin, Deserpidin, halbsynthetische Rauwolfia-Analoge, z. B. Syringopin, Germin oder Protoveratrin, synthtische hypotensive Mittel, z. B.
Hydralazin, Dihydralazin, oder Ganglienblocker, wie Chlo risbndamin.
Die neuen Verbindungen werden erhalten, wenn man ein Nitrobenzol der Formel

mit einem Aldehyd der Formel R-CHO, worin R die oben genannte Bedeutung besitzt, oder einem funktionel len Derivat davon behandelt und reduziert. Die Reaktion wird vorzugsweise in Gegenwart eines Lösungsmittels, besonders eines mit Wasser mischbaren Lösungsmittels, wie eines Alkanols, z. B. Methanol oder Äthanol, eines Äthers, z. B. Äthylenglykol-dimethyläther, Tetrahydrofuran, eines Formamids, wie Formamid selbst oder N,N-Diäthyl-formamid, eines Alkanons, wie Aceton, Wasser oder Mischungen, besonders wässrige Mischungen, mit den Lösungsmitteln, durchgeführt.
Der Aldehyd der Formel R-CHO kann als solcher oder in Form eines funktionellen Derivats davon, wie eines diesen Aldehyd abgebenden Mittels, z. B. als Polymeres oder als reaktionsfähiges funktionelles Derivat,wie eines Acetals mit einem Alkanol, z. B. 1,1-Dimethoxy-2- chloräthan, oder anderer funktioneller Derivate des diesen zu Grunde liegenden Dioxy-R-methans verwendet werden.
Solche Derivate werden vorzugsweise verwendet, wenn die Reduktion chemisch in Gegenwart einer Säure, wie einer starken anorganischen Säure, z. B. einer Mineralsäure, wie Salzsäure oder Schwefelsäure, durchgeführt wird. Man kann aber auch den Aldehyd in Lösung zur Reaktionsmischung zugeben, wobei als Lösungsmittel eines der vorgenannten organischen Lösungsmittel oder Wasser verwendet werden kann. Man verwendet den Reaktionsteilnehmer, d. h. das Sulfamyl-nitrobenzolderivat, und den Aldehyd oder eines seiner Derivate, vorzugsweise in ungefähr äquivalenten Mengen. Man kann zur Vervollständigung der Reaktion auch einen Überschuss an Aldehyd anwenden.
Die Reduktion lässt sich vorzugsweise durch Behandeln mit katalytisch aktiviertem Wasserstoff vornehmen.
Als Katalysator kommt dabei ein Metall der 8. Gruppe des periodischen Systems, wie Nickel, z. B. Raney-Nickel, oder ein anderer äquivalenter Metallkatalysator zur Anwendung. Dabei muss einzig darauf geachtet werden, dass keine der weiteren Gruppen, wie das aromatisch gebundene Chlor, abgespalten wird. Die Reaktion wird vorzugsweise unter atmosphärischem Druck, sofern notwendig, auch unter höheren Drucken, durchgeführt. Auch kann man kleine Mengen alkalische Reagenzien, wie Alkalimetallhydroxyde, z. B. Natriumhydroxyd, Kaliumhydroxyd, verwenden, wobei allerdings der Aldehyd vorzugsweise als solcher zugegeben wird. Die Reaktion lässt sich bei Raumtemperatur oder bei erhöhter Temperatur durchführen. Die Reduktion kann auch chemisch durchgeführt werden, z. B. durch ein Metall in Gegenwart einer Säure, wie z. B.
Zink, Zinn oder Eisen, in Gegenwart einer Säure, besonders einer Mineralsäure, wie Salzsäure, Bromwasserstoffsäure oder Schwefelsäure. Je nach den gewählten Bedingungen und den gewählten Metallen kann die Reaktion unter Kühlen oder Erhitzen durchgeführt werden.
Der zu diesem Verfahren verwendete Ausgangsstoff lässt sich erhalten, wenn man ein 2,4-Dihalogen-5-chlor-nitro-benzol mit einem Mercaptan der Formel Rl-SH, worin R, einen aliphatischen oder araliphatischen Kohlenwasserstoffrest, z. B. Methyl, Äthyl oder besonders Benzyl, darstellt, oder einem Halogenid der Formel R,-Hal in Gegenwart von Thioharnstoff umsetzt, die erhaltenen 2,4-Di-RI-mercapto-5-chlor-nitrobenzole zu den 2,4-Sulfonsäuren oxydiert, z. B. mit Wasserstoffperoxyd, und mit einem Halogenierungsmittel, z. B. Phosphorpentachlorid, in die 5-Chlor-nitrobenzol-2,4-disulfonylhalogenide überführt. Oxydiert man die 2,4-R1-Mercapto-5-chlor-nitroben- zole mit einem Halogen oder in Gegenwart eines Halogens, gelangt man direkt zu den Disulfohalogeniden.
Letztere können dann durch Behandeln mit Methylamin und Ammoniak in die Disulfamylverbindungen übergeführt werden.
Je nach den Reaktionsbedingungen erhält man die neuen Verbindungen in freier Form oder in Form ihrer Salze. Erhaltene Metallsalze können z. B. durch Reaktion mit wässrigen sauren Mitteln, wie Mineralsäure, z. B. Halogenwasserstoffsäure, beispielsweise Salzsäure oder Schwefelsäure, in die freien Verbindungen übergeführt werden. Diese wiederum lassen sich in die Mettallsalze, wie Alkalimetallsalze, überführen durch Behandeln z. B.
mit einem Metallhydroxyd, wie Natrium- oder Kaliumhydroxyd, in einem Lösungsmittel, wie einem Alkanol, z. B.
Methanol oder Äthanol, oder in Wasser und anschliessendes Abdampfen des Lösungsmittels. Dabei können Monooder Polysalze gebildet werden.
Die Erfindung betrifft auch Ausführungsformen des Verfahrens, nach denen man von auf irgendeiner Stufe des Verfahrens als Zwischenprodukte erhältlichen Verbindungen ausgeht und die fehlenden Verfahrensschritte vornimmt, oder bei denen ein Ausgangsstoff unter den Reaktionsbedingungen gebildet wird.
Die Erfindung wird in dem nachfolgenden Beispiel näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel:
Eine Mischung von 3,3 g 5-Chlor-2-(N-methyl-sulfa- mylS4-sulfamyl-nitrobenzol und 0,9 g Isovaleraldehyd wird eine Stunde in 25 ml Äthanol, das 0,1 g Natriumhydroxyd enthält, gerührt. Dann gibt man 2,0 g Eisenpulver zu, fügt 5 ml Wasser und genügend verdünnte Salzsäure zu, um die Reaktionsmischung schwach sauer zu stellen. Man kocht eine Stunde am Rückflusskühler, gibt 1 g Eisen zu und kocht am Rückflusskühler weiter. Nach dem Abkühlen filtriert man die festen Anteile ab, engt das Filtrat ein und kühlt. Das 6-Chlor-3-isobutyl-2-methyl-7-sulfamyl 3,4-dihydro-1 ,2,4-benzothiadiazin-1 1 -dioxyd wird nach dem Abfiltrieren in wässriger Ammoniaklösung aufgenommen und durch Zufügen von Essigsäure ausgefällt. F. 241-244".
Verwendet man anstelle von 0,9 g Isovaleraldehyd 0,83 g Chloracetaldehyd, so erhält man das 2-Methyl-3-chlormethyl-6-chlor-7-sulfamyl- 3,4-dihydro-1 ,2,4-benzothiadia- zin-1 ,1-dioxyd, das nach Umkristallisieren aus wässerigem Äthanol (500!zig) bei 208-210" schmilzt.
Das in diesem Beispiel verwendete Ausgangsmaterial lässt sich wie folgt erhalten:
Zu einer Lösung von 10 g Natriumnitrit in 50 ml konzentrierter Schwefelsäure gibt man unter Kühlen 15 g 5-Chlor-2N-methyl-suffamylY4-sulfamyl-anllin in 100 ml Essigsäure. Man rührt mehrere Stunden in der Kälte, gibt die Reaktionsmischung langsam zu einer Mischung, welche durch Umsetzen von 50 g kristallinem Kupfersulfat und 50,0 g kristallinem Natriumsulfit in Wasser, Abfiltrieren des Niederschlags und Suspendieren des erhaltenen Rückstandes einer Lösung von 100 g Natriumnitrit in 400 ml Wasser erhalten wurde. Das so entstandene 5-Chlor-2-( N-methyl-sulfamylS4-sulfamyl-nitrobenzol wird abfiltriert und mit Wasser gewaschen. Es kann ohne weitere Reinigung verwendet werden.

The present invention relates to the preparation of compounds of the above formula in which R is the methyl radical, R2 is the isobutyl or chloromethyl radical and R3 is a chlorine atom, and salts thereof, especially alkali metal salts such as sodium or potassium salts.
The new compounds show high diuretic and natriuretic effectiveness and are said to be used as medicinal products. Among these compounds, 2-methyl-3-isobutyl-6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide, as well as its alkali metal salts, should be mentioned, which are characterized by a particularly high level of effectiveness.
The new compounds are to be used as medicaments in the form of pharmaceutical preparations which these compounds together with the pharmaceutical organic or inorganic, solid or liquid carrier substances which are suitable for enteral, e.g. B. oral or parenteral administration are suitable. The reaction is preferably carried out in the presence of a solvent, especially a water-miscible solvent such as an alkanol, e.g. B. methanol or ethanol, an ether, e.g. B. ethylene glycol dimethyl ether, tetrahydrofuran, a formamide such as formamide itself or N, N-diethylformamide, an alkanone such as acetone, water or mixtures, especially aqueous mixtures, with the solvents.
The aldehyde of the formula R-CHO can be used as such or in the form of a functional derivative thereof, such as an agent that donates this aldehyde, e.g. B. as a polymer or as a reactive functional derivative such as an acetal with an alkanol, e.g. B. 1,1-dimethoxy-2-chloroethane, or other functional derivatives of these underlying dioxy-R-methane can be used.
Such derivatives are preferably used when the reduction is performed chemically in the presence of an acid such as a strong inorganic acid, e.g. B. a mineral acid such as hydrochloric acid or sulfuric acid is carried out. However, the aldehyde can also be added to the reaction mixture in solution, it being possible to use one of the aforementioned organic solvents or water as the solvent. The reactant is used; H. the sulfamyl nitrobenzene derivative, and the aldehyde or one of its derivatives, preferably in approximately equivalent amounts. The reduction can preferably be carried out by treatment with catalytically activated hydrogen.
A metal of group 8 of the periodic table, such as nickel, z. B. Raney nickel, or another equivalent metal catalyst for use. If the 2,4-R1-mercapto-5-chloro-nitrobenzenes are oxidized with a halogen or in the presence of a halogen, the disulfohalides are obtained directly.
The latter can then be converted into the disulfamyl compounds by treatment with methylamine and ammonia.
Depending on the reaction conditions, the new compounds are obtained in free form or in the form of their salts. Metal salts obtained can e.g. B. by reaction with aqueous acidic agents such as mineral acid, e.g. B. hydrohalic acid, for example hydrochloric acid or sulfuric acid, can be converted into the free compounds. These in turn can be converted into the metal salts, such as alkali metal salts, by treating e.g. B.
with a metal hydroxide such as sodium or potassium hydroxide in a solvent such as an alkanol, e.g. B.

The invention also relates to embodiments of the process in which one starts from compounds obtainable as intermediates at any stage of the process and carries out the missing process steps, or in which a starting material is formed under the reaction conditions.
The invention is described in more detail in the following example. The temperatures are given in degrees Celsius.
Example:
A mixture of 3.3 g of 5-chloro-2- (N-methyl-sulfamylS4-sulfamyl-nitrobenzene and 0.9 g of isovaleraldehyde is stirred for one hour in 25 ml of ethanol containing 0.1 g of sodium hydroxide. Then) 2.0 g of iron powder are added, 5 ml of water and sufficiently dilute hydrochloric acid are added to make the reaction mixture slightly acidic. The mixture is boiled for one hour on the reflux condenser, 1 g of iron is added and refluxed again. If 0.83 g of chloroacetaldehyde is used instead of 0.9 g of isovaleraldehyde, 2-methyl-3-chloromethyl-6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine is obtained -1, 1-dioxide, which after recrystallization from aqueous ethanol (500! Zig) melts at 208-210 ".
The starting material used in this example can be obtained as follows:
To a solution of 10 g of sodium nitrite in 50 ml of concentrated sulfuric acid, 15 g of 5-chloro-2N-methyl-suffamylY4-sulfamyl-anlline in 100 ml of acetic acid are added while cooling. The mixture is stirred for several hours in the cold, the reaction mixture is slowly added to a mixture which is obtained by reacting 50 g of crystalline copper sulfate and 50.0 g of crystalline sodium sulfite in water, filtering off the precipitate and suspending the residue obtained with a solution of 100 g of sodium nitrite in 400 ml of water was obtained.
