Verfahren zur Herstellung von neuen 1-(3',5'-Dihydroxy bzw. -acyloxy-phenyl)-2.aminoäthanolen und deren physiologisch verträglichen Säureadditionssalzen
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 1-(3',5'-Dihydroxy- bzw.
-acyloxy-phenyl)-2-aminoäthanolen, welche zur Behandlung verschiedenartiger bronchospastischer Zustände, insbesondere asthmatischer Erkrankungen, geeignet sind.
Es sind eine grosse Anzahl von 1-(3',4'-Dihydroxy-phenyl)-2-aminoäthanolen mit bronchospasmolytischer Wirkung bekannt, jedoch werden Verbindungen dieses Typs, bei denen die beiden Hydroxylgruppen in der 3- und 4-Stellung des Benzolringes stehen, im Organismus durch bestimmte Enzyme, namentlich Katechin-O-methyltransferase (COMT), welche sich u. a. in der Leber finden, angegriffen. Durch diesen Angriff verlieren die Verbindungen ihre Wirksamkeit, und Substanzen dieses Typs haben daher nur eine kurze Wirkungsdauer. Verbindungen, bei denen die beiden Hydroxylgruppen in der 3- und 5-Stellung des Benzolringes stehen, werden dagegen durch COMT nicht angegriffen. Es sind nur wenige Verbindungen des letztgenannten Typs bekannt, und diese können in der allgemeinen Formel

Diese bekannten Verbindungen der allgemeinen Formel I sind bronchospasmolytisch aktive Verbindungen von verhältnismässig langdauernder Wirkung, doch wurde festgestellt, dass sie ein Ansteigen der Herzfrequenz bewirken, wodurch die therapeutische Anwendbarkeit der Substanzen wesentlich eingeschränkt wird.
Es wurde nun gefunden, dass die neuen 1-(3',5' Dihydroxy- bzw. -acyloxy-phenyl)-2-aminoäthanole der allgemeinen Formel

Das bedeutet, dass diese Substanzen eine verschiedene Affinität gegenüber den ss-Rezeptoren in den Herz- bzw. Die neuen 1 ',5'-Dihydroxyphenyl)-2-amino äthanole sind sehr gute bronchienerweiternde Mittel und be schleunigen die Herztätigkeit nur sehr wenig.
Daher hat sich die Verbindung 1-(3',5'-Dihydroxyphenyl)-2 (tert. butylamino)-äthanol sowohl in vitro als auch in vivo als wirkungsvolleres brondiienerweiterndes Mittel erwiesen als 1-(3',5'-Dihydroxyphenyl)- 2-isopropylaminoäthanol, die Wirkungsdauer ist länger als jene des bekannten
Mittels und, wenn es am isolierten Kaninchenherzen angewandt wird, beträgt der kardioakzelerierende Effekt nur t dessen der Isopropylaminoverbindung.
Die Beziehung zwischen dem herzstimulierenden und dem bronchienerweiternden Effekt wurde auch am spontan schlagenden Meerschweinchenaurikelpräparat und an einem spiralförmig geschnittenen Tracheapräparat demonstriert, wobei beide Präparate in das gleiche Bad gegeben wurden. Sobald die erfindungsgemäss erhältliche Verbindung langsam in das Bad eingeführt wurde, wurde eine Bronchienerweiterung erzielt, ohne dass ein Effekt auf das Herzmuskelpräparat ausgeübt wurde. Der geringe kardioakzelerierende Effekt der erfindungsgemäss erhältlichen tert. Butylaminoverbin dung wurde auch durch Untersuchung des Kreislaufs einer narkotisierten Katze unter Verwendung der Verbindung bestätigt.
Die Abschwächung des kardioakzelerierenden Effekts wird auf die Weise erzielt, dass man ein sekundäres offenkettiges C-Atom durch ein tertiäres ersetzt oder es zum Teile einer Cycloalkylgruppe macht. Obwohl in der hier speziell beschriebenen Verbindung gemäss Formel II die Gruppe R tert. Butyl darstellt, fällt auch die Herstellung solcher Verbindungen, bei denen die Gruppe R Cyclobutyl bedeutet, unter die vorliegende Erfindung. Das wesentliche Merkmal ist, das das tertiäre C-Atom oder die Cycloalkylgruppe direkt mit dem Stickstoffatom verbunden ist.
Die erfindungsgemäss erhältlichen Verbindungen können in Form von Salzen mit physiologisch verträglichen Säuren angewandt werden. Geeignete Säuren, welche verwendet werden können, sind z. B. Salzsäure, Bromwasserstoff, Schwefelsäure, Fumarsäure, Zitronensäure, Weinsäure, Maleinsäure oder Bernsteinsäure.
Zusammen mit einem pharmazeutischen Träger können die neuen Verbindungen zur oralen, bronchialen, rektalen oder parenteralen Anwendung bestimmt sein.
Zur Herstellung von pharmazeutischen Zubereitungen in Form von Dosierungseinheiten zur oralen Anwendung, welche eine erfindungsgemäss erhältliche Verbindung in Form der freien Base oder eines pharmazeutisch verträglichen Salzes derselben enthalten, kann der aktive Bestandteil mit einem festen, pulverisierten Träger gemischt sein, wie z. B. Laktose, Saccharose, Sorbit, Mannit, einer Stärke, wie Kartoffelstärke, Kornstärke, Maisstärke oder Amylopectin, einem Zellulosederivat oder Gelatine, oder es können Gleitmittel verwendet werden, wie z. B. Magnesiumoder Calciumstearat, ein Polyäthylenglykolwachs oder dgl. Diese Bestandteile werden dann zu Tabletten oder Dragees gepresst. Werden Dragees benötigt, kann der innere Teil überzogen werden, z.
B. mit einer konzentrierten Zuckerlösung, welche Gummi arabicum, Talk und/oder Titandioxyd enthält, oder aber mit einem Lack, der in leichtflüchtigen organischen Lösungsmitteln oder Mischungen organischer Lösungsmittel gelöst ist. Farbstoffe können diesen Überzügen beigemengt werden. Zur Herstellung von weichen Gelatinekapseln (perlförmigen, geschlossenen Kapseln), welche aus Gelatine und beispielsweise Glycerin bestehen, oder von ähnlichen geschlossenen Kapseln, kann die aktive Substanz mit einem Polyäthylenglykolwachs gemischt werden. Harte Gelatinekapseln können Granulate der aktiven Substanz mit festen, pulverisierten Trägern, wie z. B. Laktose, Saccharose, Sorbit, Mannit, Stärken (z. B. Kartoffelstärke, Kornstärke, oder Amylopectin), Zellulosederivaten oder Gelatine enthalten, sie können auch Magnesiumstearat oder Stearinsäure enthalten.
Dosierungseinheiten zur rektalen Anwendung können in Form von Suppositorien hergestellt werden, welche die aktive Substanz in Mischung mit einem neutralen fettigen Grundmaterial enthalten, oder sie können Gelatine-Rektalkapseln sein, welche die aktive Substanz in Mischung mit einem Polyäthylenglykolwachs enthalten. Jede Dosierungseinheit enthält vorzugsweise 0,5 bis 50 mg des aktiven Bestandteils.
Flüssige Zubereitungen zur oralen Anwendung können als Syrup, Suspension oder Emulsion hergestellt werden; sie enthalten z.B. zwischen 0,1 und 20 Gew.- /o der aktiven Substanz sowie, falls gewünscht, Adjuvantien, wie z. B. Stabilisierungsmittel, Suspendierungsmittel, Dispergiermittel, Geschmacksstoffe und/oder Süsstoffe.
Flüssige Zubereitungen zur rektalen Anwendung können wässrige Lösungen sein, welche zwischen etwa 0,1 und 2 Gew.- /o der aktiven Substanz enthalten sowie, falls gewünscht, Stabilisierungsmittel und/oder Puffersubstanzen.
Zur parenteralen Anwendung durch Injektion kann der Träger eine sterile, parenteral verträgliche Flüssigkeit sein, z. B. pyrogenfreies Wasser oder eine wässerige Lösung von Polyvinylpyrrolidon, oder ein parenteral verträgliches Öl, z. B. Erdnussöl, und wahlweise Stabilisierungsmittel und/oder Puffersubstanzen enthalten. Dosierungseinheiten der Lösung werden vorteilhaft in Ampullen abgefüllt, wobei jede Dosierungseinheit vorzugsweise zwischen 0,05 und 5 mg des aktiven Bestandteils enthält.
Zur Anwendung auf die Bronchien werden die Zusammensetzungen vorteilhaft in Form von Tropfen oder einer Sprühlösung hergestellt. Die Lösung enthält vorteilhaft zwischen 0,1 und 10 Gew.-O/o des aktiven Bestandteils.
Pharmakologische Versuche: a) Bronchospasmolytischer Effekt:
Der bronchospasmolytische Effekt des 1-(3',5'-Dihydroxyphenyl)2-(tert. butylamino)-äthanols wurde mit dem der bekannten Verbindungen Adrenalin und 1-(3',5'-Dihydroxyphenyl- 2-isopropylaminoäthanol an einer spiralförmig geschnittenen Trachea eines Meerschweinchens nach einer ursprünglich von Castillo & Boer in J. Pharmacol. Exptl. Therap. 90 (1947), 104, beschriebenen und später von Constantine (J. Pharm. Pharmacol. 17 (1965), 384) abgewandelten Methode verglichen. Bei diesem Versuch wurde festgestellt, dass die erfindungsgemäss erhältliche Verbindung etwa die zweifache Wirkung der Isopropylverbindung und etwa die halbe Wirkung des Adrenalins hat.
Bei einem in-vivo-Versuch nach Konzett & Rössler (auch. Exp. Path. Pharmak. 195 (1940), 71) war der bronchospasmolytische Effekt nach in-vivo-Anwendung ebenfalls zweimal so gross wie der der Isopropylverbindung.
Die Wirkungsdauer war länger als die des bekannten Isopropylderivats (Fig. 1; DATA= 1-(3',5'-Dihydroxyphenyl)2-(tert. butilamino)-äthanol; DIÄ = 1-(3',5'-Dihydroxyphenyl)2-isopropylaminoäthanol.
Ähnliche Ergebnisse wurden bei Erprobung der Cyclobutylverbindung erzielt. Der bronchospasmolytische Effekt bei z. B.
1-(3',5'-Dihydroxypenyl)- 2-cyclobutylaminoäthanol, gemessen wie beschrieben, war etwa 1,2 mal so gross wie der der bekannten Isopropylverbindung und etwa 0,4 mal so gross wie der des Adrenalins.
b) Wirkung auf das Herz:
Die Beziehung zwischen dem herzstimulierenden und dem bronchienerweiternden Effekt wurde in vitro an einem Aurikel-Trachea-Präparat vom Meerschweinchen untersucht. Um die Wirkung auf das Herz und die Bronchialmuskeln unter gleichen experimentellen Bedingungen vergleichen zu können, wurden sowohl ein spontan schlagendes Aurikel als auch eine spiralförmig geschnittene Trachea in das gleiche Bad in eine Kreb'sche Lösung gegeben. Beide Präparate wurden vom gleichen Tier genommen. Die Testverbindung wurde langsam in die Badlösung eingegeben. Auf diese Weise wurde die Konzentration der Verbindung langsam erhöht und es war leicht festzustellen, an welchem Muskel die Verbindung die grösste Wirkung zeigte.
Adrenalin wurde als Vergleichspräparat verwendet.
Dieses Mittel verursacht bei der gleichen Konzentration Bronchodilation und Herzstimulierung. Die Infusionsdauer betrug 10 Minuten. Nach Waschen und Rückgewinnen wurde die Testlösung in gleicher Weise, wie für Adrenalin beschrieben, eingesetzt, und die Wirkung des Mittels auf die beiden Präparate konnte leicht mit der des Adrenalins verglichen werden.
In den Zeichnungen zeigt Fig. 1 die Wirkungsdauer von DTÄ und DIÄ gegen durch Histamin verursachte Bronchokonstriktion (Meerschweinchen, 0,95 kg). Der bronchospasmolytische Effekt wurde nach der Methode von Konzett & Rössler bestimmt.
Fig. 2 zeigt die Wirkung von Adrenalin (ADR) und DTÄ auf eine spiralförmig geschnittene Trachea und ein rechtes Aurikel eines Meerschweinchens. Beide Präparate wurden vom gleichen Tier genommen und unter den gleichen experimentellen Bedingungen untersucht.
Fig. 3 zeigt die Wirkung des Adrenalin (ADR), Noradrenalins (NA) und Isoprenalins (IPR) auf eine spiralförmig geschnittene Trachea und ein rechtes Aurikel eines Meerschweinchens. Beide Präparate wurden vom gleichen Tier genommen und unter den gleichen experimentellen Bedingungen untersucht.
Fig. 4 zeigt die Wirkung von Adrenalin (ADR) und DIA auf eine spiralförmig geschnittene Trachea und ein rechtes Aurikel eines Meerschweinchens. Beide Präparate wurden vom gleichen Tier genommen und unter den gleichen experimentellen Bedingungen untersucht.
Fig. 5 zeigt den Einfluss von Adrenalin (ADR) und DTÄ auf Geschwindigkeit und Stärke der Herztätigkeit an einem isolierten Kaninchenherzen.
Fig. 6 zeigt den Einfluss von Adrenalin (ADR), DIÄ und DTA auf Geschwindigkeit und Stärke der Herztätigkeit an einem isolierten Kaninchenherzen.
Wie aus Fig. 2 ersichtlich, wurde festgestellt, dass 1-(3',5'-Dihydroxyphenyl2-tert. butylaminoäthanol (DNA) eine Bronchodilatation ohne kardische Stimulierung hervorruft. Zum Vergleich sind die Wirkungen des Noradrenalins (NA), Isoprenalins (IPR) und 1-(3',5'-Dihydroxyphenyl)- 2-isopropylaminoäthanols (DIÄ) auf diese Präparate in den Fig. 3 und 4 gezeigt. Noradrenalin, welches nur geringe bronchienerweiternde Wirkung zeigt, stimuliert den Herzmuskel, ohne die Bronchialmuskeln anzugreifen. Wie in Fig. 4 gezeigt, sind sowohl Isoprenalin als auch DIA starke Herzstimulantien, verglichen mit ihrem bronchienerweiternden Effekt.
Der kardiostimulatorische Effekt wurde auch am isolierten Kaninchenherzen (Langendorff-Präparat) untersucht. Wie in Fig. 5 gezeigt, ist der kardioakzelerierende Effekt der erfindungsgemäss erhältlichen Verbindung schwach und beträgt nur etwa V56 der Wirkung des Adrenalins. Die Wirkung von DIÄ auf dieses Präparat ist in Fig. 6 dargestellt. Der kardioakzelerierende Effekt dieses Mittels betrug bei diesem Experiment etwa 3/4 dessen des Adrenalins.
Der kardiostimulierende Effekt der erfindungsgemäss erhältlichen cyclobutyl-substituierten Verbindungen war gleich gross wie der der tert. butyl-substituierten. So hatte 1 -(3',5'-Didydroxyphenyl)2-cyclobutylaminoäthanol einen herzstimulierenden Effekt, der kleiner war als 1/,, der Wirkung des Adrenalins.
Toxizitätsuntersuchungen:
Die Toxizität des 1-(3',5'-Dihydroxyphenyl)2-(tert. butylamino)-äthanols (DTÄ) bei Mäusen nach intravenöser, subkutaner und oraler Anwendung ist in Tabelle I dargestellt. Zum Vergleich wurden entsprechende LDs0-Werte für das 1-(3',5'-Dihydroxyphenyl)- 2-isopropylaminoäthanol (DIÄ) angegeben.

Toxizitätsmessungen der erfindungsgemäss erhältlichen Cyclobutylverbindungen wurden ebenfalls durchgeführt und zeigen, dass diese Verbindungen auch in dieser Beziehung den offenkettigen bzw. tert.
Butylderivaten vergleichbar sind. LDJ0-Wert (i. v.) für 1-(3',5'-Dihydroxyphenyl)2-cyclobutylaminoäthanol bei Mäusen = 54 mg/kg Körpergewicht.
Die erfindungsgemäss erhältlichen Verbindungen weisen ein sehr vorteilhaftes Verhältnis zwischen Herzstimulierung und bronchospasmolytischer Wirkung auf.
Diese unerwartete Eigenschaft macht sie für die Behandlung bronchospastischer Zustände, wie z. B. des Asthmas oder anderer verwandter Krankheiten, welche das Atmungssystem angreifen, besonders geeignet.
Die Erfindung wird durch die folgenden Beispiele näher erläutert:
Beispiel 1
Darstellung von 1-(3',5'-Dihydroxyphenyl)-2-(tert.
butylamino)-äthanol und von dessen Salzen:
Zu einer Lösung aus 6,8 g des Hydrats von 1-(Glyoxyloyl-3, 5-dibenzyloxy-benzol in 50 ml Methanol werden 7,0 g tert. Butylamin und 30 ml Benzol hinzugefügt. Die Reaktionsmischung wird 3 Stunden am Rückfluss gehalten und dann verdampft. Das zurückbleibende Öl kristallisiert bei Zugabe von Äthanol; Fp. 78,5-79,5 C. 2,5 g dieser Verbindung in 75 ml absolutem Äthanol werden unter normalen Bedingungen mit Raney-Nickel hydriert. Nach Abfiltrieren des Katalysators und Verdampfen wird das zurückbleibende Öl mit Athanol/Benzol getrocknet. Die Base in Äthanol wird mit Bromwasserstoff in Äthanol behandelt und eingedampft. Der Rückstand kann aus Eisessig/Chloroform kristallisiert werden. Fp. des Hydrobromids 205-206 C (Zers.).
Die Identität dieses Produkts wurde durch IR Spektroskopie bestätigt.
Beispiel 2
Darstellung von 1-(3',5'-Dihydroxyphenyl)
2-(cyclobutylamino)-äthanol-hydrobromid:
Zu einer Lösung von 3,6 g des Hydrats von 1-Glyoxyloyl-3,5-dibenzyloxybenzol in 50 ml trockenem Benzol wurde 1,0g Cyclobutyl- amin gegeben. Die Reaktionsmischung wurde drei Stunden unter Wasserabtrennung am Rückfluss gehalten.
Die dunkelrote Lösung wurde verdampft und 4,0 g eines rotbraunen Öls wurden erhalten.
Dieses öl wurde in 100 ml absolutem Athanol gelöst und unter normalen Bedingungen mit Raney-Nikkel hydriert. Nach Abfiltrieren des Katalysators wurde die Lösung mit konzentrierter Bromwasserstoffsäure angesäuert und eingedampft. Das zurückbleibende Öl wurde mit absolutem Äthanol/Benzol getrocknet. Ausbeute 1,5 g. Aus khanol/Äther erhielt man ein kristallines Produkt.
Die Identität des Produkts wurde durch IR-Spektroskopie bestätigt.
In ähnlicher Weise wie in den vorstehenden Beispielen beschrieben, können auch die entsprechenden 3',5'-Acyloxy-Verbindungen hergestellt werden, z. B.
1-(3',5'-Dipivaloyloxyphenyl)- 2-(tert. butalamino)-äthanol (Fp. des Hydrobromids 190-192 C) und 1-(3',5'-Diacetoxyphenyl)2-(tert. butylamino)-äthanol (Fp. des Hydrobromids 108-111 C).
Trennung des 1-(3',5'-Dihydroxyphenyl)-2-(tert.
butylamino)-äthanols in seine optischen Isomeren a) Optische Trennung von 1-(3',5'-Dibenzyloxyphenyl)-2- (benzyl-tert. butylamino)-äthanol
25,0 g 1-(3',5'-Dibenzyloxyphenyl)2-(benzyl-tert. butylamino)-äthanol (razemische Base) werden in 375 ml Methanol unter Erhitzen gelöst und 19,0 g (-)-Dibenzoylweinsäure in 125 ml Methanol werden zugesetzt. Die Mischung wird 30 Minuten am Rückfluss gehalten. Nach Eindampfen erhält man ein Öl. Dieses wird in kochendem Isopropanol gelöst, und Wasser wird zugefügt, bis eine Trübung eintritt, worauf einige ml Isopropanol zugesetzt werden, um eine klare Lösung zu erhalten. Die Lösung wird über Nacht kristallisieren gelassen. Es werden 37,0 g eines weissen kristallinen Produkts isoliert. Weitere 7,0 g werden nach Einengen des Volumens aus dem Filtrat erhalten.
Die beiden Fraktionen werden gemischt und in 1100 ml absolutem Athanol gelöst, filtriert und kristallisieren gelassen. Dieses Produkt wird aus absolutem Äthanol umkristallisiert, bis die Drehung konstant bleibt (sechsmal). [aJn20 = -34,2 (1 o/o in abs.
Methanol). Ausbeute 4,5 g.
b) Herstellung von (-)-1-(3',5'-Dihydroxyphenyl)

Dieses wird abfiltriert und mit Wasser und trockenem Äther gewaschen. Ausbeute 2,8 g.

Dieses Produkt wird in 75 ml absolutem Äthanol gelöst, 0,15 g Palladium auf Aktivkohle (100/oil) werden zugefügt und die Hydrierung bei Raumtemperatur bei 5 At. Druck während 4 Stunden durchgeführt. Der Katalysator wird abfiltriert und der Rückstand zur Trockne eingedampft. Eine kleine Menge Äthanol wird zugesetzt, um das Produkt zu lösen, und anschliessend Ather bis zur Eintrübung. 23,7 g 1-(3',5'-Dibenzyloxyphenyl)2-(benzyl)-tert. butylamino)-äthanol-Base, aus den gesammelten überstehenden Lösungen aus der Herstellung von (-)-1-3',5'-Dibenzyloxyphenyl)2-(benzyl-tert.butylamino)-äthanol stammend, werden in 250 ml Methanol gelöst und mit 18,2 g (+)-Dibenzoylweinsäure in 250 ml Methanol 60 Minuten unter Rückfluss gekocht. Dann wird das Produkt in derselben Weise aufgearbeitet, wie oben beschrieben, und aus absolutem Äthanol umkristallisiert (zweimal). [a]D20 = +34,3 (10/o in abs. Methanol).
Ausbeute 10,5 g.
Das Hydrobromid wird aus 9,5 g dieses Produkts in derselben Weise wie oben beschrieben bereitet. Ausbeute 6,2 g. [a]D20 = 33,00 (1 O/o in abs. Methanol). Die Hydrierung von 5,5 g dieses Produkts wird wie früher beschrieben durchgeführt. Kristallisation aus Äthanol/ Äther. Ausbeute 2,7 g. [a]D20 = + 34,20 (10/o in abs.

Process for the preparation of new 1- (3 ', 5'-dihydroxy or -acyloxy-phenyl) -2.aminoethanols and their physiologically acceptable acid addition salts
The invention relates to a process for the production of new 1- (3 ', 5'-dihydroxy or
-acyloxy-phenyl) -2-aminoethanols, which are suitable for the treatment of various types of bronchospastic conditions, in particular asthmatic diseases.
A large number of 1- (3 ', 4'-dihydroxyphenyl) -2-aminoethanols with bronchospasmolytic action are known, but compounds of this type in which the two hydroxyl groups are in the 3- and 4-position of the benzene ring are known , in the organism by certain enzymes, namely catechin-O-methyltransferase (COMT), which u. a. found in the liver, attacked. As a result of this attack, the compounds lose their effectiveness and substances of this type therefore only have a short duration of action. Compounds in which the two hydroxyl groups are in the 3- and 5-position of the benzene ring, on the other hand, are not attacked by COMT. The relationship between the cardiac stimulating and bronchodilator effects was also demonstrated on the spontaneously beating guinea pig auricle preparation and on a spiral-shaped trachea preparation, both preparations being placed in the same bath. As soon as the compound obtainable according to the invention was slowly introduced into the bath, a bronchodilator was achieved without any effect being exerted on the myocardium preparation. The low cardio-accelerating effect of the tert. Butylamino compound was also confirmed by examining the circulation of an anesthetized cat using the compound.
The weakening of the cardio-accelerating effect is achieved by replacing a secondary open-chain carbon atom with a tertiary one or making it part of a cycloalkyl group. Although in the compound according to formula II specifically described here, the group R tert. If butyl represents, the preparation of those compounds in which the group R represents cyclobutyl also falls under the present invention. The essential feature is that the tertiary carbon atom or the cycloalkyl group is connected directly to the nitrogen atom.
The compounds obtainable according to the invention can be used in the form of salts with physiologically compatible acids. Suitable acids which can be used are e.g. B. hydrochloric acid, hydrogen bromide, sulfuric acid, fumaric acid, citric acid, tartaric acid, maleic acid or succinic acid.
Together with a pharmaceutical carrier, the new compounds can be intended for oral, bronchial, rectal or parenteral use.
For the production of pharmaceutical preparations in the form of dosage units for oral use, which contain a compound obtainable according to the invention in the form of the free base or a pharmaceutically acceptable salt thereof, the active ingredient can be mixed with a solid, powdered carrier, such as. B. lactose, sucrose, sorbitol, mannitol, a starch such as potato starch, corn starch, corn starch or amylopectin, a cellulose derivative or gelatin, or lubricants can be used, such as. Dosage units for rectal use can be produced in the form of suppositories which contain the active substance mixed with a neutral fatty base material, or they can be gelatin rectal capsules which contain the active substance mixed with a polyethylene glycol wax. Each dosage unit preferably contains 0.5 to 50 mg of the active ingredient.
Liquid preparations for oral use can be prepared as a syrup, suspension or emulsion; they contain e.g. between 0.1 and 20 wt / o of the active substance and, if desired, adjuvants, such as. B. stabilizers, suspending agents, dispersants, flavorings and / or sweeteners.
Liquid preparations for rectal use can be aqueous solutions which contain between about 0.1 and 2% by weight of the active substance and, if desired, stabilizers and / or buffer substances.
For parenteral use by injection, the carrier can be a sterile, parenterally acceptable liquid, e.g. B. pyrogen-free water or an aqueous solution of polyvinylpyrrolidone, or a parenterally acceptable oil, e.g. B. peanut oil, and optionally contain stabilizers and / or buffer substances. Dosage units of the solution are advantageously filled into ampoules, each dosage unit preferably containing between 0.05 and 5 mg of the active ingredient.
For application to the bronchi, the compositions are advantageously prepared in the form of drops or a spray solution. The solution advantageously contains between 0.1 and 10% by weight of the active ingredient.
Pharmacological tests: a) Bronchospasmolytic effect:
The bronchospasmolytic effect of 1- (3 ', 5'-dihydroxyphenyl) 2- (tert. Butylamino) -ethanol was compared with that of the known compounds adrenaline and 1- (3', 5'-dihydroxyphenyl-2-isopropylaminoethanol on a spiral cut Trachea of a guinea pig according to a method originally described by Castillo & Boer in J. Pharmacol. Exptl. Therap. 90 (1947), 104, and later modified by Constantine (J. Pharm. Pharmacol. 17 (1965), 384) In this experiment it was found that the compound obtainable according to the invention has about twice the effect of isopropyl compound and about half the effect of adrenaline.
In an in-vivo experiment according to Konzett & Rössler (also. Exp. Path. Pharmak. 195 (1940), 71), the bronchospasmolytic effect after in-vivo application was also twice as great as that of the isopropyl compound.
The duration of action was longer than that of the known isopropyl derivative (Fig. 1; DATA = 1- (3 ', 5'-dihydroxyphenyl) 2- (tert.butilamino) ethanol; DIÄ = 1- (3', 5'-dihydroxyphenyl) 2-isopropylaminoethanol.
Similar results were obtained when the cyclobutyl compound was tested. The bronchospasmolytic effect in z. B.
1- (3 ', 5'-dihydroxypenyl) -2-cyclobutylaminoethanol, measured as described, was about 1.2 times as large as that of the known isopropyl compound and about 0.4 times as large as that of adrenaline.
b) Effect on the heart:
The relationship between the cardiac stimulating and the bronchodilating effect was investigated in vitro on a guinea pig auricle-trachea preparation. In order to be able to compare the effects on the heart and the bronchial muscles under the same experimental conditions, both a spontaneously beating auricle and a spiral-shaped trachea were placed in the same bath in a Kreb's solution. Both preparations were taken from the same animal. The test compound was slowly added to the bath solution. In this way the concentration of the compound was slowly increased and it was easy to determine on which muscle the compound was most effective.
Adrenaline was used as a comparator.
This agent causes bronchodilation and cardiac stimulation at the same concentration. The infusion time was 10 minutes. After washing and recovery, the test solution was used in the same manner as described for adrenaline, and the effect of the agent on the two preparations could be easily compared with that of the adrenaline.
In the drawings, Fig. 1 shows the duration of action of DTÄ and DIÄ against bronchoconstriction caused by histamine (guinea pigs, 0.95 kg). The bronchospasmolytic effect was determined using the Konzett & Rössler method.
Figure 2 shows the effects of adrenaline (ADR) and DTA on a spiral cut trachea and right auricle of a guinea pig. Both preparations were taken from the same animal and examined under the same experimental conditions.
Fig. 3 shows the effect of adrenaline (ADR), noradrenaline (NA) and isoprenaline (IPR) on a spiral cut trachea and a right auricle of a guinea pig. Both preparations were taken from the same animal and examined under the same experimental conditions.
Figure 4 shows the effects of adrenaline (ADR) and DIA on a spiral cut trachea and right auricle of a guinea pig. Both preparations were taken from the same animal and examined under the same experimental conditions.

As can be seen from Fig. 2, it was found that 1- (3 ', 5'-dihydroxyphenyl2-tert. Butylaminoethanol (DNA) causes bronchodilation without cardiac stimulation. For comparison, the effects of noradrenaline (NA), isoprenaline (IPR) and 1- (3 ', 5'-dihydroxyphenyl) -2-isopropylaminoethanol (DIA) on these preparations are shown in Figures 3 and 4. Norepinephrine, which shows little bronchodilator activity, stimulates the heart muscle without attacking the bronchial muscles shown in Figure 4, both isoprenaline and DIA are potent cardiac stimulants compared to their bronchodilator effect.
The cardiac stimulatory effect was also investigated on isolated rabbit hearts (Langendorff preparation). As shown in FIG. 5, the cardio-accelerating effect of the compound obtainable according to the invention is weak and is only about ½ of the effect of adrenaline. The effect of DIÄ on this preparation is shown in FIG. The cardio-accelerating effect of this agent in this experiment was about 3/4 that of adrenaline.
The cardio-stimulating effect of the cyclobutyl-substituted compounds obtainable according to the invention was the same as that of the tert. butyl-substituted. For example, 1- (3 ', 5'-didydroxyphenyl) 2-cyclobutylaminoethanol had a cardiac stimulating effect which was less than 1/1 of the effect of adrenaline.
Toxicity studies:
The toxicity of 1- (3 ', 5'-dihydroxyphenyl) 2- (tert. Butylamino) ethanol (DTÄ) in mice after intravenous, subcutaneous and oral administration is shown in Table I. For comparison, corresponding LDs0 values were given for 1- (3 ', 5'-dihydroxyphenyl) -2-isopropylaminoethanol (DIÄ).

Toxicity measurements of the cyclobutyl compounds obtainable according to the invention were also carried out and show that these compounds also have the open-chain or tert.
Butyl derivatives are comparable. LDJ0 value (i.v.) For 1- (3 ', 5'-dihydroxyphenyl) 2-cyclobutylaminoethanol in mice = 54 mg / kg body weight.
The compounds obtainable according to the invention have a very advantageous ratio between cardiac stimulation and bronchospasmolytic effect.
This unexpected property makes them useful in the treatment of bronchospastic conditions such as: B. asthma or other related diseases that attack the respiratory system, particularly suitable.
The invention is illustrated in more detail by the following examples:
example 1
Preparation of 1- (3 ', 5'-dihydroxyphenyl) -2- (tert.
butylamino) ethanol and its salts:
7.0 g of tert-butylamine and 30 ml of benzene are added to a solution of 6.8 g of the hydrate of 1- (glyoxyloyl-3,5-dibenzyloxy-benzene in 50 ml of methanol. The reaction mixture is refluxed for 3 hours and The oil that remains crystallizes on addition of ethanol, melting point 78.5-79.5 ° C. 2.5 g of this compound in 75 ml of absolute ethanol are hydrogenated with Raney nickel under normal conditions, after the catalyst has been filtered off and evaporation the remaining oil is dried with ethanol / benzene. The base in ethanol is treated with hydrogen bromide in ethanol and evaporated. The residue can be crystallized from glacial acetic acid / chloroform. Mp. of the hydrobromide 205-206 C (decomp.).
The identity of this product was confirmed by IR spectroscopy.
Example 2
Preparation of 1- (3 ', 5'-dihydroxyphenyl)
2- (cyclobutylamino) ethanol hydrobromide:
1.0 g cyclobutylamine was added to a solution of 3.6 g of the hydrate of 1-glyoxyloyl-3,5-dibenzyloxybenzene in 50 ml of dry benzene. The reaction mixture was refluxed for three hours with removal of water.
The dark red solution was evaporated and 4.0 g of a red-brown oil was obtained.
This oil was dissolved in 100 ml of absolute ethanol and hydrogenated with Raney nickel under normal conditions. The catalyst was filtered off and the solution was acidified with concentrated hydrobromic acid and evaporated. The remaining oil was dried with absolute ethanol / benzene. Yield 1.5g. A crystalline product was obtained from ethanol / ether.
The identity of the product was confirmed by IR spectroscopy.
In a manner similar to that described in the preceding examples, the corresponding 3 ', 5'-acyloxy compounds can also be prepared, e.g. B.
1- (3 ', 5'-Dipivaloyloxyphenyl) - 2- (tert.butalamino) ethanol (melting point of the hydrobromide 190-192 C) and 1- (3', 5'-diacetoxyphenyl) 2- (tert.butylamino) ethanol (melting point of the hydrobromide 108-111 C).
Separation of the 1- (3 ', 5'-dihydroxyphenyl) -2- (tert.
butylamino) ethanol into its optical isomers a) Optical separation of 1- (3 ', 5'-dibenzyloxyphenyl) -2- (benzyl-tert. butylamino) ethanol
25.0 g of 1- (3 ', 5'-dibenzyloxyphenyl) 2- (benzyl-tert-butylamino) -ethanol (racemic base) are dissolved in 375 ml of methanol with heating and 19.0 g of (-) - dibenzoyltartaric acid in 125 ml of methanol are added. The mixture is refluxed for 30 minutes. An oil is obtained after evaporation. This is dissolved in boiling isopropanol and water is added until it becomes cloudy, after which a few ml of isopropanol are added to obtain a clear solution. The solution is allowed to crystallize overnight. 37.0 g of a white crystalline product are isolated. A further 7.0 g are obtained from the filtrate after reducing the volume.
The two fractions are mixed and dissolved in 1100 ml of absolute ethanol, filtered and allowed to crystallize. This product is recrystallized from absolute ethanol until the rotation remains constant (six times). [aJn20 = -34.2 (1 o / o in abs.
Methanol). Yield 4.5g.
b) Preparation of (-) - 1- (3 ', 5'-dihydroxyphenyl)
2- (tert. Butylamino) ethanol hydrobromide
4.0 g of the above-mentioned product are suspended in water, then ether and then aqueous ammonia are added. The base is extracted with two portions of ether. Dilute hydrobromic acid is added to the ether phase and stirred for 1.5 hours. A white crystalline product is formed. This is filtered off and washed with water and dry ether. Yield 2.8g.

23.7 g of 1- (3 ', 5'-dibenzyloxyphenyl) 2- (benzyl) -tert. butylamino) ethanol base, derived from the collected supernatant solutions from the preparation of (-) - 1-3 ', 5'-dibenzyloxyphenyl) 2- (benzyl-tert.butylamino) ethanol, are dissolved in 250 ml of methanol and refluxed for 60 minutes with 18.2 g of (+) - dibenzoyltartaric acid in 250 ml of methanol. The product is then worked up in the same way as described above and recrystallized from absolute ethanol (twice). [a] D20 = +34.3 (10 / o in absolute methanol).
Yield 10.5g.
The hydrobromide is prepared from 9.5 g of this product in the same manner as described above. Yield 6.2g. [a] D20 = 33.00 (10 / o in absolute methanol).
