Die Erfindung betrifft ein neues Verfahren zur Herstellung von bekannten 1 -(5-Nitro-2-thiazolyl)-2-oxoimidazolidinen der allgemeinen Formel

Die Umsetzung der Verbindungen II und III wird vorteilhaft in einem Lösungsmittel, vorzugsweise in einem niederen Alkohol wie Methanol oder Äthanol durchgeführt. Man arbeitet vorzugsweise in Gegenwart eines basischen Kondensationsmittels wie eines Alkalialkoholats z. B. Natriummethylat oder -äthylat oder eines Alkalihydroxyds z. B. Natriumhydroxyd oder eines tertiären Amins, zweckmässig bei Temperaturen zwischen 200 und 1500 C. Das Reaktionsgemisch wird anschliessend mit einem üblichen Oxydationsmittel, z. B. Wasserstoffsuperoxid, Kaliumpermanganat oder einer Persäure vorzugsweise unter Zuabe eines polaren wasserhaltigen Lösungsmittels, z. B. Eisessig-Wasser, und zweckmässigerweise bei Temperaturen zwischen 20 und 1200 C behandelt, wobei der

durch eine Carbonylgruppe ersetzt wird.
Die Herstellung der Ausgangsstoffe der Formel II, worin X für eine R2S-Gruppe steht, erfolgt z. B. indem man 2-Amino-5-nitro-thiazol mit Schwefelkohlenstoff in Anwesenheit einer Base und eines polaren Lösungsmittels umsetzt, wobei zunächst ein Salz des Bis-mercaptomethylenamino 5-nitro-thiazols entsteht, welches ohne vorherige Isolierung anschliessend nach bekannten Methoden, z. B. durch Umsetzen mit reaktionsfähigen Estern von Verbindungen der Formeln R2OH, worin R2 die angegebenen Bedeutungen hat, zu einem Ausgangsstoff der allgemeinen Formel II umgesetzt werden kann. Als reaktionsfähige Ester kommen vor allem solche mit Halogenwasserstoffsäuren in Betracht.
Die Herstellung der Ausgangsstoffe der Formel Il, worin X ein Halogenatom darstellt, kann mittels Halogenierung eines 2-(Bismercapto-methylen-amino) 5-nitro-thiazols der Formel II mit Chlor oder Brom in einem inerten Lösungsmittel, z. B.: Tetrachlorkohlenstoff, vorzugsweise bei der Siedetemperatur des verwendeten Lösungsmittels, erfolgen.
Die Verbindungen der allgemeinen Formel I besitzen wertvolle pharmazeutische Eigenschaften. Es waren bereits Verfahren zu ihrer Herstellung bekannt (vgl. z. B. DAS 1 245 971). Das vorliegende Verfahren geht jedoch von den leicht zugänglichen Verbindungen der Formeln II und III aus und ergibt die gewünschten Produkte der Formel I ohne Isolierung einer Zwischenstufe in guten Ausbeuten.
Die nachstehenden Beispiele sollen die Erfindung weiter erläutern, ohne sie jedoch einzuschränken.
Beispiel 1
25 g (0,1 Mol) 2-(Bismethylmercaptomethylenamino)-5-nitro-thiazol werden zusammen mit 12,8 g (0,11 Mol) ss-Chloräthylamin-hydrochlorid in 150 ccm Methanol suspendiert.
Zu der unter Rühren am Rückfluss siedenden Suspension wird eine Lösung von 4,6 g Natrium (0,2 Mol) in 100 ccm Methanol innerhalb von 11/2 Stunden zugetropft, insgesamt wird 2t/2 Stunden am Rückfluss erhitzt (starke Mercaptanentwicklung). Anschliessend werden 200 ccm Eisessig zugegeben und bei 700 C während 2 Stunden 50 ccm 300/oige Wasserstoffperoxid-Lösung zugetropft. Nach Absaugen des ausgefallenen Produktes wird die Mutterlauge im Rotationsverdampfer auf 100 ccm eingeengt, das Festprodukt abgesaugt und die vereinigten Fraktionen nach Waschen mit Wasser und Methanol aus Eisessig umkristallisiert.
Man erhält so das 1 < (5-Nitro-2-thiazolyl)-2-oxo-imidazolidin in Form von gelben Kristallen vom F. 260-2610 C.
Ausbeute 15,0 g (70 O/o d. Th).
Das als Ausgangsmaterial verwendete 2-(Bismethyl-mercapto-methylenamino)5-nitro-thiazol kann wie folgt-erhalten werden:
Zur Lösung von 290 g (2 Mol) 2-Amino-5-nitrothiazol in 500 ml Dimethylsulfoxid gibt man unter Rühren 167 g (2,2 Mol) Schwefelkohlenstoff und tropft anschliessend unter Eiskühlung (Innentemperatur 20 C) eine Suspension von 448 g (4 Mol) Kalium-tertiär-butylat in 500 ml Dimethylsulfoxid während 3
Stunden zu. In die so erhaltene Lösung des Dikalium salzes des 2-[Bismercapto-methylenamino] 5-nitro-thiazols tropft man innerhalb von 3 Stunden 625 g (4,4 Mol)
Methyljodid und rührt die immer fester werdende Masse bei Raumtemperatur über Nacht. Nach Absaugen und Waschen mit Methanol und anschliessend mit Äther erhält man 252 g Rohprodukt.
Die Umkristallisation aus Dimethyl-formamid liefert das 2-(Bismethylmercapto-methylenamino)5-nitro-thiazol in Form von Kristallen, die bei 163 C schmelzen.
Beispiel 2
In analoger Weise wie in Beispiel 1 beschrieben kann man ausgehend von 2-(Bisbenzylmercapto-methylenamino)5-nitro-thiazol an Stelle von 2-(Bismethylmercapto-methylenamino)-5-nitro-thiazol das 1-(5Zitro-2-thiazolyl)-2-oxo- imidazolidin F. 260-261 C, erhalten.
Das als Ausgangsmaterial verwendete 2-(Bisbenzylmercapto-methylenamino)5-nitro-thiazol kann wie folgt erhalten werden.
18,2 g (0,125 Mol) 2-Amino-5-nitro-thiazol werden in 75 ml Di-methylsulfoxid gelöst und 10 g (0,125 ml) Schwefelkohlenstoff zugegeben. Innerhalb von 2 Stunden wird eine Lösung von 10 g Natriumhydroxid in 20 ml Wasser unter Rühren bei 0-10 C zugetropft, anschliessend unter Eiskühlung 31,7 g (0,23 Mol) Benzylchlorid; der Ansatz wird bei 0 C zwei Tage stehengelassen. Die ausgefallenen Kristalle werden abgesaugt und dreimal aus Essigester umkristallisiert. Man erhält so das 2-(Bisbenzylmercapto-methylenamino)5-nitro-thiazol vom F. 141-142 C.
Beispiel 3
In analoger Weise wie im Beispiel 1 beschrieben erhält man aus 2-(Bismethylmercapto-methylenamin)- 5-nitro-thiazol und a-Athyl-ss-chloräthylamin-hydrochlorid das 1 -(5-Nitro-2-thlazoWl)-2- oxo-4-äthyl-imidazolidin, das nach dem Umkristallisieren aus Eisessig bei 255 C schmilzt.Ausbeute 73 O/o.
Beispiel 4
2,4 g 2-[(Chlor-methylmercapto methylen)-amino]5-nitro-thiazol, 1,89 g 2-Chlor-äthylaminhydrochlorid und 1,9 g Triäthylamin werden in 15 ml Toluol suspendiert. Das Reaktionsgemisch wird zum Sieden erhitzt und innerhalb 15 Minuten werden nochmals 1,9 g Triäthylamin zugegeben; anschliessend wird noch 1 Stunde unter Rückfluss erhitzt. Das Lösungsmittel wird im Vakuum entfernt, der Rückstand in Eisessig suspendiert und die Suspension bei 700 C mit 300/oigem Wasserstoffsuperoxyd versetzt.
Beim Aufarbeiten, analog Beispiel 1 erhält man das 1-(5-Nitro-2-thiazolyl)-2 oxo-imidazolidin in Form von gelben Kristallen vom F. 260-2610 C.
Das als Ausgangsmaterial verwendete
2-(Chlor-methylmercapto methylenamino)-5-nitrothiazol kann wie folgt erhalten werden:
7,5 g 2-(Bismethylmercapto methylenamino)-5-nitrothiazol werden in 160 mol Tetrachlorkohlenstoff suspendiert und bei Siedehitze unter Rühren während 1/4 Stunden Chlor bis zu Sättigung eingeleitet. Die entstandene klare Lösung wird heiss filtriert und das Lösungsmittel im Vakuum abdestilliert. Der kristalline Rückstand wird mit weniger Äther digeriert und rasch abgesaugt.
Man erhält so das 2-2-(Chlor-methylmercapto methylenamino)-5-nitrothiazol in Form von gelblichen hygroskopischen Kristallen vom F. 87-90 C.
Ausbeute: 2,4 g (41 O/o der Theorie).

The starting materials of the formula II, in which X is an R2S group, are prepared, for. B. by reacting 2-amino-5-nitro-thiazole with carbon disulfide in the presence of a base and a polar solvent, initially forming a salt of bis-mercaptomethyleneamino 5-nitro-thiazole, which without prior isolation then by known methods, e.g. . B. by reacting with reactive esters of compounds of the formulas R2OH, where R2 has the meanings given, can be converted into a starting material of the general formula II. The compounds of general formula I have valuable pharmaceutical properties. Processes for their production were already known (cf. e.g. DAS 1 245 971). example 1
25 g (0.1 mol) of 2- (bismethylmercaptomethyleneamino) -5-nitro-thiazole are suspended in 150 cc of methanol together with 12.8 g (0.11 mol) of β-chloroethylamine hydrochloride.
A solution of 4.6 g of sodium (0.2 mol) in 100 cc of methanol is added dropwise over the course of 11/2 hours to the suspension, which is refluxed with stirring, and the mixture is heated under reflux for a total of 2t/2 hours (strong mercaptan development). Then 200 cc of glacial acetic acid are added and 50 cc of 300% hydrogen peroxide solution are added dropwise at 700 ° C. over the course of 2 hours. In a manner analogous to that described in Example 1, starting from 2- (bisbenzylmercapto-methylenamino) 5-nitro-thiazole, instead of 2- (bismethylmercapto-methylenamino) -5-nitro-thiazole, 1- (5-nitro-2-thiazolyl) -2-oxo-imidazolidine m.p. 260-261 ° C.
The 2- (bisbenzylmercapto-methylenamino) 5-nitro-thiazole used as a starting material can be obtained as follows.
18.2 g (0.125 mol) of 2-amino-5-nitro-thiazole are dissolved in 75 ml of dimethyl sulfoxide and 10 g (0.125 ml) of carbon disulfide are added. A solution of 10 g of sodium hydroxide in 20 ml of water is added dropwise with stirring at 0-10 ° C. over the course of 2 hours, followed by 31.7 g (0.23 mol) of benzyl chloride while cooling with ice; the batch is left to stand at 0 C for two days. Example 3
In a manner analogous to that described in Example 1, 1- (5-nitro-2-thlazoWl) -2-oxo is obtained from 2- (bismethylmercapto-methylenamine) -5-nitro-thiazole and α-ethyl-ß-chloroethylamine hydrochloride -4-ethyl-imidazolidine, which, after recrystallization from glacial acetic acid, melts at 255 ° C. Yield 73%.
Example 4
2.4 g of 2 - [(chloromethylmercaptomethylene) amino] 5-nitro-thiazole, 1.89 g of 2-chloroethylamine hydrochloride and 1.9 g of triethylamine are suspended in 15 ml of toluene. The reaction mixture is heated to the boil and a further 1.9 g of triethylamine are added within 15 minutes; it is then refluxed for a further hour. The solvent is removed in vacuo, the residue is suspended in glacial acetic acid and the suspension is treated with 300% hydrogen peroxide at 700.degree.
When working up, as in Example 1, 1- (5-nitro-2-thiazolyl) -2 oxo-imidazolidine is obtained in the form of yellow crystals with a melting point of 260-2610 C.
That used as the starting material
2- (Chlor-methylmercapto methylenamino) -5-nitrothiazole can be obtained as follows:
7.5 g of 2- (bismethylmercapto methylenamino) -5-nitrothiazole are suspended in 160 mol of carbon tetrachloride and chlorine is passed in until saturation is reached at boiling point with stirring for 1/4 hour. The resulting clear solution is filtered hot and the solvent is distilled off in vacuo. The crystalline residue is digested with less ether and quickly filtered off with suction.
This gives the 2-2- (chloromethylmercapto methylenamino) -5-nitrothiazole in the form of yellowish hygroscopic crystals with a melting point of 87-90 ° C.
Yield: 2.4 g (41% of theory).
