Verfahren zur Herstellung von neuen Uracilderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Uracilderivaten mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Uracilderivate der allgemeinen Formel I :

Verbindungen der allgemeinen Formel I, in denen R durch den Isopropylrest verkörpert ist, bewirken überdies eine starke Ausscheidung von Natrium- und Chlorionen, ohne dabei die normale Ausscheidung der Kaliumionen zu erhöhen. Ferner eignen sich Verbindungen der allgemeinen Formel I, insbesondere solche mit Wasserstoff als Ri'als Zwischenprodukte zur Herstellung weiterer pharmakologisch verwendbarer Stoffe.

Die Umsetzung von Verbindungen der allgemeinen Formel II mit solchen der allgemeinen Formel III erfolgt beispielsweise in Gegenwart von Pyridin oder einer andern tertiären organischen Base wieTriäthyl- amin in einem wasserfreien inerten organischen Lösungsmittel wie z. B. Benzol, Toluol, Methylenchlorid oder Chloroform bei Raumtemperatur oder nötigenfalls unter Erwärmen. Der Ringschluss erfolgt vorzugsweise ohne vorausgehende Aufarbeitung des Zwischenproduktes der allgemeinen Formel IV. Sofern das in der ersten Stufe verwendete Lösungsmittel gegenüber alkalischenKondensationsmitteln beständig ist, kann es auch als Reaktionsmedium der zweiten Stufe dienen.
Der Ringschluss wird beispielsweise durch Erwärmen des Zwischenproduktes der allgemeinen Formel IV mit Natriumhydrid oder einem Natriumalkoholat in Benzol, Toluol oder Xylol, durch Erwärmen mit einem Alkalialkoholat in einem niederen Alkanol oder schliesslich durch Erwärmen mit einer wässerigalkoholischen Alkalilauge durchgeführt. Die als Reaktionsprodukte erhaltenen Verbindungen der allgemeinen Formel I, in denen R durch Wasserstoff verkörpert ist, sind unter Bildung von Alkalisalzen in

wässerigen Alkalilaugen löslich und können durch Ansäuern ausgefällt werden. Zur weiteren Reinigung werden sie aus geeigneten organischen Lösungsmitteln umkristallisiert.
Die Einführung eines niederen Alkylrestes Rl erfolgt in einfacher Weise z. B. durch Umsetzung einer Verbindung der allgemeinen Formel I, welche ein Wasserstoffatom als Rl enthält, mit einem reaktionsfähigen Ester eines niederen Alkanols oder Alkenols in wässeriger Alkalilauge oder, in Gegenwart eines säurebindenden Mittels wie z. B. Kaliumcarbonat, in einem geeigneten organischen Lösungsmittel wie z. B. Aceton oder Butanon, nötigenfalls unter Erwärmen. Geeignete reaktionsfähige Ester von niederen Alkanolen oder Alkenolen sind z. B. Methylbromid, Methyljodid, Äthylbromid, n-Propylbromid, Crotylbromid, Dimethylsulfat, Diäthylsulfat oder p-Toluolsulfonsäuremethylester.
Ausgangsstoffe der allgemeinen Formel II wie z. B. B - Aminocrotonsäureäthylester sind bekannt und weitere in an sich bekannter Weise, z. B. durch Umsetzung von geeigneten niederen ex-substituierten ot-Alkanoylessigsäurealkylestem mit Ammoniak, niederen Alkylaminen oder Alkenylaminen herstellbar.
Ebenso sind zahlreiche Ausgangsstoffe der allgemeinen Formel III bekannt.

Die nachfolgenden Beispiele dienen zur näheren Erläuterung des erfindungsgemässen Verfahrens zur Herstellung von Verbindungen der allgemeinen Formel I ; sie stellen jedoch keineswegs die einzigen Ausführungsformen derselben dar. Teile bedeuten darin Gewichtsteile ; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.

absolutem Benzol gelöst und der Lösung 24, 2 Vol. -Teile Pyridin zugesetzt. Dann werden unter Rühren bei 250 32,6 Vol. -Teile Phenylisocyanat zugetropft.
Das Reaktionsgemisch wird noch 30 min bei der gleichen Temperatur gerührt und anschliessend 30 min unter Rückfluss gekocht. b) Zu der erhaltenen Lösung des rohen oc-n-Butyl-8-phenylureidocrotonsäureäthylesters wird bei 250 eine Lösung von 6,9 Teilen Natrium in 140 Vol. -Teilen absolutem Äthanol zugegeben und anschliessend wird die Hauptmenge der Lösungsmittel abdestilliert. Zum Rückstand werden 270 Vol. -Teile absolutes Xylol gegeben und das Gemisch 2 h auf 1300 erwärmt. Nach Abkühlen auf Raumtemperatur werden 100 Teile Wasser eingerührt, wobei Kristalle von 3-Phenyl-5-n-butyl-6-methyluracil ausfallen. Die Ausfällung wird durch Ansäuern der wässerigen Lösung vervollständigt. Nach Umkristallisieren aus Äthylacetat schmilzt die neue Verbindung bei 1950.
Beispiel 2 : Eine gemäss Beispiel la bereitete Lösung wird zunächst stark eingeengt und der Rückstand mit 500 Vol. -Teilen Äthanol und 500 Vol. -Teilen 2n-Natronlauge 30 min unter starkem Rühren auf 900 erwärmt. Nach Abdestillieren des Äthanols erfolgt die Aufarbeitung unter Zufügen von Wasser wie im Beispiel 1 angegeben, wobei man die dort angegebene Verbindung erhält.
Beispiel 3 : Analog zu Beispiel 1 werden 60, 6 Teile α-Isopropyl-ss-aminocrotonsäureäthylester in 300 Vol.-Teilen Benzol und 28,6 Vol. -Teilen Pyridin mit 38, 7 Vol. -Teilen Phenylisocyanat umgesetzt.

-Isopropyl-ss-phenylureidocrotonsäureäthylestersRühren 17 Teile einer Suspension von Natriumhydrid in Mineralöl 1 : 1 gegeben, worauf die Lösung 6 h unter Rückfluss gekocht wird. Die Aufarbeitung erfolgt analog Beispiel 1, beginnend mit dem Zufügen von Wasser. Das erhaltene 3-Phenyl-5-isopropyl-6-methyluracil schmilzt nachdem Umkristallisieren aus Äthanol bei 2500.
In analoger Weise erhält man unter Verwendung von 62, 5 Teilen p-Acetoxyphenylisocyanat das 3- (p-Hydroxyphenyl)-5-isopropyl-6-methyluracil, Smp. > 3200 (aus Eisessig). Die Acetoxygruppe wird im Verlaufe der Aufarbeitung unter den gegebenen Reaktionsbedingungen hydrolysiert.
Beispiel 4 : 24, 4 Teile 3-Phenyl-5-isopropyl-6-methyluracil (vgl. Beispiel 3) werden unter Erwärmen in 500 Vol. -Teilen 2n-Natronlauge gelöst. Bei 0 - 50 werden 9,5 Vol. -Teile Dimethylsulfat eingetropft. und das Gemisch wird 6 h bei 50 gerührt. Die ausgefallenen Kristalle werden abgesaugt, gut gewaschen und aus 801o Methanol umkristallisiert. Das erhaltene 1, 6-Dimethyl-3-phenyl-5-isopropyluracil schmilzt bei 125 .

After the ethanol has been distilled off, work-up is carried out with the addition of water as indicated in Example 1, the compound indicated there being obtained.
