Verfahren zur Herstellung von Derivaten des rechtsdrehenden Propanolamins
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen optisch aktiven Propanolamin-Derivaten mit vorteilhaften biologischen Eigenschaften.
In der britischen Patentschrift Nr. 1 069 343 des Patentinhabers sind bestimmte optisch aktive Propanol amin-Derivate beschrieben und beansprucht. Es sind jedoch keine rechtsdrehenden Verbindungen darin beschrieben.
Die erfindungsgemäss erhaltenen Verbindungen sind Derivate des rechtsdrehenden Propanolamins der Formel: ArO-CH rCHOH CH¯NHRI oder deren Salze, in welcher Formel Ar einen Naphthylrest, der gegebenenfalls einen oder mehrere Alkyl-, Alkoxy-, Hydroxyl- oder Halogensubstituenten trägt, oder einen Phenylrest, der einen oder mehrere Alkyl-, Alkoxy-, Hydroxyl-, Aryloxy-, Aryl- oder Aroylsubstituenten trägt, darstellt und R1 einen gegebenenfalls einen oder mehrere Alkyl-, Alkoxy- oder Hydroxylsubstituenten tragenden Alkyl-, Cycloalkyl- oder Aralkylrest, aber keinen substituierten oder nichtsubstituierten a Arylalkylrest darstellt.
Die rechtsdrehenden Propanolamine sind diejenigen, bei denen der basische Anteil rechtsdrehend (+) in bezug auf Licht von etwa 5890 Ä ist. Die Salze der erfindungsgemäss hergestellten Verbindungen (das heisst die Salze von rechtsdrehenden Propanolaminen), bei denen der saure Anteil eine optisch aktive Säure ist, können unter Umständen linksdrehend sein.
Als Beispiele für die in dem durch Ar dargestellten Rest vorhandenen Substituenten kann man Chlor- und Bromatome, Alkyl- und Alkoxyreste mit höchstens 5 C Atomen, wie z. B. Methyl-, Äthyl-, Methoxy- oder Athoxyreste, Aryloxy- und Arylreste mit höchstens 10 C-Atomen, wie z. B. den Phenoxy- oder Phenylrest, sowie Aroylreste mit höchstens 11 C-Atomen, wie z. B.
das Benzoylradikal, erwähnen.
Stellt R1 ein Alkylradikal dar, so kann dieses z. B.
ein Alkylradikal mit höchstens 10 C-Atomen, wie z. B.
das nXPropyl-, Isopropyl, sek.-Butyl-, tert.-Butyl- oder l-Methyl-n-butylradikal, sein. Stellt R1 ein substituiertes Alkylradikal dar, so kann dieses zweckmässig ein höchstens 10 C-Atome enthaltendes Alkylradikal sein, das ein oder mehrere Hydroxy- oder Alkoxyradikale, z. B.
Alkoxyradikale mit höchstens 5 C-Atomen, wie z. B.
das Methoxyradikal, trägt. Stellt Rt ein gegebenenfalls substituiertes Aralkylradikal dar, so kann dieses zweckmässig ein höchstens 12 C-Atome enthaltendes Aralkylradikal sein, das gegebenenfalls mit z.B. einem oder mehreren Alkyl- oder Alkoxyradikalen, beispielsweise Alkyl- oder Alkoxyradikalen mit höchstens 5 C-Atomen, substituiert ist. Als bestimmte Beispiele für R1, wenn dieses ein disubstituiertes Alkylradikal oder ein Aralkylradikal darstellt, kann man also das 2-Hydroxy-l,ldimethyläthyl-, l-Methyl-3-phenylpropyl- oder l,l-Di- methyl-3 -phenylpropylradikal erwähnen. Stellt Rl ein Cycloalkylradikal dar, so kann dieses zweckmässig ein solches mit höchstens 10 C-Atomen, wie z. B. das Cyclopentylradikal, sein.
Als bestimmte Beispiele für die Verbindungen nach der Erfindung kann man folgende erwähnen: (f)-l-Isopropylamino-3-(l-naphthyloxy)-
2-propanol, ( +)-1 -(3,5-Dimethylphenoxy)-3-(1, I-dimethyl-
3-phenylpropylamino)-2-propanol, ( + )- 1 tert.-Butylamino-3-(4-methyl- 1-naphthyloxy)-
2-propanol, (+ )- 1 -(2-Hydroxy- 1,1 -dimethyläthylamino)-
3-(2-phenoxyphenoxy)-2-propanol und (+ )- 1 -Isopropylamino-3 -(3 -tolyloxy)-2'-propanol und deren Salze.
Als geeignete Salze nach der Erfindung kann man Säureadditionssalze, z. B. von anorganischen Säuren stammende Salze, wie z. B. Hydrochloride, Hydrobromide, Phosphate und Sulfate, sowie von organischen Salzen stammende Salze, wie z. B. Oxalate, Lactate, Tartrate, O,O-Di-p-toluoyltartrate, Acetate, Salicylate und Citrate, erwähnen.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel: ArO-CHCHOH CHNltR1 bzw. deren Salze, wobei Ar und R1 die oben angegebene Bedeutung haben, ist dadurch gekennzeichnet, dass das entsprechende optisch aktive Propanolamin-Derivat der Formel: ArO-CHLFCHOH CH°NR1R2 oder ein Salz davon, in welcher Formel R2 einen durch Hydrogenolyse abspaltbaren Rest darstellt, der Hydrogenolyse unterwirft.
Als Beispiele für das Radikal R2 kann man ein a-Arylalkylradikal mit höchstens 10 C-Atomen, wie z. B. das Benzylradikal, oder einen a,a Diarylalkylrest mit höchstens 19 C-Atomen, wie z. B. das Benzhydrylradikal, erwähnen. Die Hydrogenolyse kann durch katalytische Hydrierung, z. B. in Gegenwart von einem Platin- oder Palladium-auf-Holzkohle-Katalysator, erfolgen. Die Hydrogenolyse lässt sich zweckmässig in einem Verdünnungs- oder Lösungsmittel, bei normaler oder erhöhter Temperatur und unter atmosphärem oder erhöhtem Druck verwirklichen.,
Die beim Verfahren als Ausgangsstoff verwendeten optisch aktiven Propanolamin-Derivate können durch Auflösung der entsprechenden racemischen Verbindungen in der in der britischen Patentschrift Nr. 1136 918 beschriebenen oder einer analogen Weise hergestellt werden.
Natürlich können die als Ausgangsstoff verwendeten optisch aktiven Propanolamin-Derivate in manchen Fällen rechtsdrehend und in anderen Fällen linksdrehend sein.
Die rechtsdrehenden Propanolamin-Derivate nach der Erfindung eignen sich für die Behandlung von Herzschlagunregelmässigkeiten, die durch Herzglykoside, wie z. B. Digitalisglykosid oder Ouabain, induziert werden.
In dem nachstehenden Ausführungsbeispiel sind alle Mengenangaben auf das Gewicht bezogen.
Beispiel
Es werden 2,7 Teile (-)-l-(N-Benzyl-N-isopropyl- amino)-3-(l-naphthyloxy) -2-propanol in 48 Teilen Äthanol aufgelöst. Auch 0,6 Teile eines 5%eigen Palla dium-auf-Holzkohle-Katalysators werden hinzugegeben.
Unter einer Wasserstoffatmosphäre wird die Mischung bei Umgebungstemperatur unter einem Druck von 100 at 48 Stunden geschüttelt. Dann wird die Mischung gefiltert, und das Filtrat wird in Vakuum zur Trockne eingedampft. Der Rückstand wird aus Petroläther (Sdp.
60-800C) umkristallisiert. Somit erhält man (+)-1 Isopropylamino - 3 - (l-naphthyloxy) - 2 - propanol, Smp.
720 C, [a] 2D5 + 12,90 (c, 0,436 % in trockenem Äthanol).
Diese Base kann in bekannter Weise in das Hydrochlorid umgesetzt werden. Nach Umkristallisierung aus Isopropanol hat das Hydrochlorid folgende Eigenschaften: Smp. 1920C, k]2D5 +230 (c, 0,695% in trockenem Äthanol).
Das als Ausgangsstoff verwendete (-)-1 -(N-Benzyl-N-isopropylamino)-3 - (1 -naphthyl- oxy)-2-propanol [a] 2D5 - 300 (c, 1,961 % in Methanol) kann wie bei Beispiel 1 der britischen Patentschrift Nr. 1136 918 hergestellt werden.
Process for the preparation of derivatives of dextrorotatory propanolamine
The invention relates to a process for the production of new optically active propanolamine derivatives with advantageous biological properties.
In the assignee's British patent specification No. 1,069,343 certain optically active propanolamine derivatives are described and claimed. However, no clockwise connections are described therein.
The compounds obtained according to the invention are derivatives of dextrorotatory propanolamine of the formula: ArO-CH rCHOH CH¯NHRI or salts thereof, in which formula Ar is a naphthyl radical, which optionally bears one or more alkyl, alkoxy, hydroxyl or halogen substituents, or a phenyl radical , which carries one or more alkyl, alkoxy, hydroxyl, aryloxy, aryl or aroyl substituents, and R1 is an alkyl, cycloalkyl or aralkyl radical which may optionally carry one or more alkyl, alkoxy or hydroxyl substituents, but not a substituted one or an unsubstituted arylalkyl radical.
The dextrorotatory propanolamines are those in which the basic portion is dextrorotatory (+) with respect to light of about 5890 Å. The salts of the compounds prepared according to the invention (that is to say the salts of dextrorotatory propanolamines) in which the acidic component is an optically active acid can under certain circumstances be levorotatory.
Examples of the substituents present in the radical represented by Ar can be chlorine and bromine atoms, alkyl and alkoxy radicals having at most 5 carbon atoms, such as. B. methyl, ethyl, methoxy or ethoxy, aryloxy and aryl groups with a maximum of 10 carbon atoms, such as. B. the phenoxy or phenyl radical, and aroyl radicals with a maximum of 11 carbon atoms, such as. B.
mention the benzoyl radical.
If R1 is an alkyl radical, this can, for. B.
an alkyl radical with at most 10 carbon atoms, such as. B.
the nX-propyl, isopropyl, sec-butyl, tert-butyl or l-methyl-n-butyl radical. If R1 is a substituted alkyl radical, this can expediently be an alkyl radical containing at most 10 carbon atoms, one or more hydroxy or alkoxy radicals, e.g. B.
Alkoxy radicals with a maximum of 5 carbon atoms, such as. B.
the methoxy radical. If Rt represents an optionally substituted aralkyl radical, this can expediently be an aralkyl radical containing at most 12 carbon atoms, which is optionally substituted with e.g. one or more alkyl or alkoxy radicals, for example alkyl or alkoxy radicals with at most 5 carbon atoms, is substituted. As specific examples of R1, if this represents a disubstituted alkyl radical or an aralkyl radical, one can therefore mention the 2-hydroxy-l, l-dimethylethyl, l-methyl-3-phenylpropyl or l, l-dimethyl-3-phenylpropyl radical . Suitable salts according to the invention can be acid addition salts, e.g. B. derived from inorganic acids salts such. B. hydrochlorides, hydrobromides, phosphates and sulfates, and salts derived from organic salts, such as. B. oxalates, lactates, tartrates, O, O-di-p-toluoyltartrates, acetates, salicylates and citrates, mention.
The process according to the invention for the preparation of the compounds of the formula: ArO-CHCHOH CHNltR1 or their salts, where Ar and R1 have the meaning given above, is characterized in that the corresponding optically active propanolamine derivative of the formula: ArO-CHLFCHOH CH ° NR1R2 or a salt thereof, in which formula R2 represents a radical which can be split off by hydrogenolysis and which is subjected to hydrogenolysis.
Examples of the radical R2 can be an a-arylalkyl radical with at most 10 carbon atoms, such as. B. the benzyl radical, or an a, a diarylalkyl radical with a maximum of 19 carbon atoms, such as. B. the benzhydryl radical, mention. The hydrogenolysis can be carried out by catalytic hydrogenation, e.g. B. in the presence of a platinum or palladium-on-charcoal catalyst. The hydrogenolysis can expediently be carried out in a diluent or solvent, at normal or elevated temperature and under atmospheric or elevated pressure.,
The optically active propanolamine derivatives used as starting materials in the process can be prepared by dissolving the corresponding racemic compounds in the manner described in British Patent No. 1136,918 or in an analogous manner.
Of course, the optically active propanolamine derivatives used as starting materials can be clockwise in some cases and counterclockwise in other cases.
The dextrorotatory propanolamine derivatives according to the invention are suitable for the treatment of heartbeat irregularities caused by cardiac glycosides, such as. B. digitalis glycoside or ouabain, induced.
In the exemplary embodiment below, all quantitative data are based on weight.

2.7 parts of (-) - 1- (N-benzyl-N-isopropyl-amino) -3- (1-naphthyloxy) -2-propanol are dissolved in 48 parts of ethanol. 0.6 parts of a 5% own palladium-on-charcoal catalyst are also added.

This base can be converted into the hydrochloride in a known manner. After recrystallization from isopropanol, the hydrochloride has the following properties: mp. 1920C, k] 2D5 +230 (c, 0.695% in dry ethanol).
The (-) - 1 - (N-benzyl-N-isopropylamino) -3 - (1-naphthyloxy) -2-propanol [a] 2D5-300 (c, 1.961% in methanol) used as starting material can be used as in Example 1 of British Patent No. 1136,918 can be prepared.
