Verfahren zur Herstellung von Azetidino[3,2"d]thiazolidinmethancarbonsäureester
Gegenstand der vorliegenden Erfindung ist ein Methodikverfahren zur Herstellung von Azetidino[3,2d]thiazolidinmethancarbonsäureestern, das zur Darstellung von wertvollen Zwischenprodukten und insbesondere bei der erstmaligen synthetischen Herstellung der 7-Amino-cephalosporansäure und ihrer Derivate Anwendung fand und zu dieser eigenartigen Synthese be- sonders geeignet ist.

Derivate sind in erster Linie N-Acylverbindungen, worin Acylreste insbesondere diejenigen von wirksamen N-Acyldeívaten der 7-Ainint > cephalosporan säure, wie der Thienylacetyl-, z. B. 2-Thienylacetyl-, Cyanacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z. B. der tert.-Butyloxycarbonylrest, bedeuten.
Die Synthese dieser für die Herstellung wertvoller Arzneimittel wichtigen Verbindung und ihrer Derivate beruht auf der Idee, von einer 3,5-unsubstituierten 2,2disubstituierten Thiazolidin-4-carbonsäure, z. B. einer Verbindung der Formel I

Wenn erwünscht, kann in einer erhaltenen Verbindung ein Substituent in einen anderen Substituenten über;ge- führt und, wenn erwünscht, ein erhaltenes Isomerengemisch in die einzelnen Isomeren getrennt werden.
Substituenten in 4-Stellung sind Kohlenwasserstoff-, insbesondere aliphatische Kohlenwasserstoffreste, wie Niederalkyl-, z. B. Äthyl-, n-Propyl-, Isopropyloder vorzugsweise Methylgruppen, sowie aromatische, insbesondere Phenylgruppen, oder araliphatische Kohlenwasserstoffreste, insbesondere Phenylalkyl-, z.B.
Benzyl- oder Phenyläthylgruppen, sowie funktionell abgewandelte, z.B. veresterte Carboxylgruppen, wie Carbbniederalkoxy-, z. B. Carbomethoxy- oder Carb äthoxygruppen. Die beiden Substituenten können auch zusammengenommen werden und für einen bivalenten Kohlenwasserstoff-, insbesondere aliphatischen Kohlenwasserstoffrest, wie eine Niederalkylen-, z.B.
1,4-Butylen- oder 1,5-Pentylengruppe. sowie eine Phthaloylgruppe oder für eine Oxo- oder Thionogruppe stehen. Die obgenannten Kohienwass erstoffre- ste sind unsubstituiert oder können z. B. durch Niederalkyl-, wie Methyl- oder Äthylgruppen, Niederalkoxy-, wie Methoxy- oder Äthoxygruppen, Halogen-, wie Fluor-, Chlor- oder Bromatome, Halogenalkyl-, wie Trifluormethylgruppen, oder andere geeignete Gruppen substituiert sein.
Ester der Diformylmethylen-methancarbonsäure- ester sind z. B. solche mit Alkoholen jeglicher Art, insbesondere mit aliphatischen Alkoholen, wie Alkanolen, insbesondere Niederalkanolen, z. B. Methanol, Äthanol, n-Propanol oder tert.-Butanol, cycloaliphatischen Alkoholen, wie Cycloalkanolen, z.B. Cyclohexanol, oder araliphatischen Alkoholen, wie Phenylniederalkanolen, z.B. Benzylalkohol oder Diphenylmethanol, wobei die obgenannten Alkohole unsubstituiert sind oder Niederalkyl-, Niederalkoxy-, Nitro- oder Trifluormethylgruppen oder insbesondere Halogenatome sowie andere Gruppen als Substituenten enthalten können.
Besonders geeignet als die Carbonsäure veresternde substituierte Hydroxyverbindungen sind halogenierte Niederalkanole, wie 2,2,2-Trichloräthanol.
Die obige Reaktion wird so durchgeführt, dass die Addition unter weitgehender Vermeidung der Zersetzung des Olefins vor sich geht, d. h. bei Zimmertemperatur oder durch Erhitzen des Reaktionsgemisches bis etwa 180"C, vorzugsweise zwischen etwa 50 und etwa 1200 C, wenn erwünscht, in Gegenwart eines geeigneten Verdünnungsmittels und/oder eines Kondensationsmittels, wenn notwendig, in einer Inertgasatmosphäre und/oder in einem geschlossenen Gefäss.
In erhaltenen Verbindungen können Substituenten in verschiedenartigen, z. T. nach an sich bekannten Methoden, in andere übergeführt werden. So lässt sich z.B. eine mit einem 2,2,2-Trihalogenäthanol, besonders 2,2,2-Trichloräthanol, veresterte Carboxygruppe in eigenartiger Weise mittels reduzierender Mittel in die freie Carboxylgnippe überfiihren. Geeignete Mittel sind chemische Reduktionsmittel, wie nascierender Wasserstoff, erhalten z.B. durch die Einwirkung von Metallen, Metallegierungen oder -amalgamen auf was serstoffabgebende Mittel, wie Zink, Zinklegierungen, z. B.
Zinkkupfer, oder Zinkamalgam in Gegenwart von Säuren, wie organischen Carbonsäuren, z.B. Essigsäure, oder Alkoholen, wie Niederalkanolen, Alkalimetall-, z. B. Natrium- oder Kaliumamalgam oder Aluminiumamalgam, in Gegenwart von feuchtem Äther oder von Niederalkanolen, ferner Alkalimetallen, z. B.
Lithium, Natrium oder Kalium, oder Erdalkalimetallen, z. B. Caloium, in flüssigem Ammoniak, gegebenenfalls unter Zugabe von Alkoholen, wie eines Niederalkanols. Ferner kann ein 2,2,2-Trihalogenäthyl-, insbesondere ein 2,2,2-Trichloräthylester durch Behandeln mit reduzierenden Metallsalzen, wie Chrom-II-verbindungen, z.B. Chrom-II-chlorid oder Chrom-II-acetat, vorzugsweise in Gegenwart von wässrigen Medien, enthaltend mit Wasser mischbare organische Lösungsmittel, wie Niederalkanole, Niederalkancarbonsäuren oder Äther, z. B.
Methanol, Äthanol, Essigsäure, Tetrahydrofuran, Dioxan, Athylenglykol-dimethyläther oder Diäthylenglykol-dimethyläther, in die freie Säure übergeführt werden.
Verbindungen mit freier Carboxygruppe können z. B. in ihre Salze, wie z. B. Alkali- oder Erdalkalimetall- oder Ammoniumsalze, übergeführt werden. Freie Carboxygruppen können nach an sich bekannten Methoden, z.B. durch Behandeln mit einer Diazoverbindung, wie einem Diazo-niederalkan, z. B. Diazomethan oder Diazoäthan, oder einem Phenyl-diazo-niederalkan, z.B. Phenyldiazomethan oder Diphenyldiazomethan, verestert werden. Dabei kann gleichzeitig eine der Formylgruppen in einen Enoläther übergeführt werden.
Durch Behandeln mit komplexbildenden Schwermetallsalzen lassen sich die Verfahrensprodukte, falls erwünscht, in Salze, z. B. Kupfer-, Eisen-, Magnesiltm-, Zink- oder Bleisalze, überführen.
Bei der Behandlung der Verfahrensprodukte mit acylierenden Mitteln, wie Säureanhydriden oder -halogeniden, gegebenenfalls in Anwesenheit von Basen, wie Pyridin, Coilidin oder Triäthylamin, erhält man Enol acrylate.
Erhaltene Gemische von Isomeren können nach an sich bekannten Methoden, z.B. durch fraktioniertes Kristallisieren, Adsorptionschromatographie (Kolonnen- oder Dünnschichtchromatographie) oder anderen Verfahren in die einzelnen Isomeren getrennt werden; erhaltene Racemate können durch Bilden eines Gemisches von diastereoisomeren Salzen mit optisch aktiven salzbildenden Mitteln, Trennen des Gemisches in die diastereoisomeren Salze und Überführen der abgetrennten Salze in die freien Verbindungen, oder durch fraktioniertes Kristallisieren aus optisch aktiven Lösungsmitteln, in die Antipoden getrennt werden.
Das Verfahren umfasst auch diejenigen Ausführungsformen, wonach als Zwischenprodukte anfallende Verbindungen als Ausgangsstoffe verwendet und die restlichen Verfahrensschritte mit diesen durchgeführt werden, oder das Verfahren auf irgendeiner Stufe abgebrochen wird; ferner können Ausgangsstoffe in Form von Derivaten, z. B. von Salzen, verwendet oder während der Reaktion gebildet werden.
Vorzugsweise werden solche Ausgangs stoffe verwendet und die Reaktionsbedingungen so gewählt, dass man zu den eingangs als besonders bevorzugt aufgeführten Verbindungen gelangt.
Die im obigen Verfahren verwendeten Ausgangsstoffe werden nach den in den Anmeldungen G. Nr.
497 456 (Case Wo 8), bzw. G. Nr. 497 369 (Case Wo 10) beschriebenen Verfahren hergestellt.
In verfahrensgemäss hergestellten Verbindungen der Formel XIa steht Ra z. B. für einen unsubstituierten oder substituierten aliphatischen oder araliphatischen Kohlenwasserstoffrest, insbesondere einen Niederalkyl- oder einen Halogen-niederalkyl-, z.B. den 2,2,2-Trichloräthylrest, sowie einen Phenyl-niederalkylrest.
Acylreste Ac sind in erster Linie solche, welche in pharmakologisch wirksamen N-Acylderivaten der 7 Amin < > cephalospoansäure vorkommen, wie der Thie nylacetyi-, z. B. 2-Thienylacetyl-, Chloräthylcarbamyl-, Cyanacetyl- oder Phenylacetylrest, oder leicht abepalt- bare Acylreste, wie der Rest eines Halbmessers der Kohlensäure, z. B. der tert.-Butyloxycarbonylrest.
Der Rest -X- steht insbesondere für die Gruppe der Formel

worin Rt und R2 für die oben genannten Kohlenwasserstoff-, insbesondere die aliphatischen, aromatischen oder araliphatischen Kohlenwasserstoffreste, sowie funktionell abgewandelten Carboxylgruppen stehen, wobei die beiden Substituenten auch z,usammengenotnmen werden und bivalente Kohlenwasserstoffreste, sowie Phthaloylreste, oder Oxo- oder Thionogruppen bedeuten können.
Verfahrensgemäss erhaltene Verbindungen werden, wie im Formelschema gezeigt wird, in 7-Amino-cephalosporansäure oder deren Derivate umgewandelt; diese Umwandlung kann z. B. nach dem in den Anmeldungen 0. Nr. 497 460 (Case Wo 12), 0. Nr. 497 379 (Case Wo 13) und G. Nr. 497 461 (Case Wo 14) beschriebenen Verfahren vorgenommen werden.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
In 2 parallelen Ansätzen gibt man tropfenweise zu 400 ml schwach siedendem n-Octan innert 60 Minuten unter Rühren und in einer Stickstoffatmosphäre eine Lösung von 6g 3,3-Diformyl-milchsäure2,2.2-trichloräthylester in 100 ml 1,2-Dimethoxyäthan, wobei gleichzeitig durch einen Destillationsaufsatz 240 ml Lösungsmittelgemisch abdestilliert werden Nach dem Abküblen wird unter Feuchtigkeitsausschluss filtriert und das Filtrat, enthaltend den 3,3-Diformyl-acrylsäure2,2,2-trichloräthylester, mit 1 g des 3-tert.-Butyloxycarbonyl-4,4dimethyl-azetidinol[3,2-d]thiazolidin-2-ons versetzt. Das Reaktionsgemisch wird während 9 Stunden bei 110 C erhitzt und dann im Wasserstrahlvakuum singedampft. Der Rückstand wird mit 60 ml Benzol extrahiert.
Die aus beiden Ausätzen erhaltenen Benzolextrakte werden vereinigt und fünimal mit je 75 ml einer gesättigten Natriumhydrogencarbonatlösung extrahiert. Die wässrigen Lösungen werden einmal mit Benzol ausgeschütteh und die organischen Lösungen vereinigt; sie enthalten zur Hauptsache polymeres Ausgangsmaterial.
Die Natriumhydrogencarbonatextrakte werden nach Zagabe von Methylenchlorid mit Zitronensäure auf pH etua @ angesäuert and man erhält aus der organischen Phase den rohen 3-tert.-Butyionycarbonyl-n-diformylmethyl4,4-dimethyl-3-oxo-1-azetidino[3,2-d] thiazolidin-essigsäure-2,2,2trichloräthylester der Formel

1,158 g des Rohprodukts werden an 45 g säuregewaschenem Silicagel chromatographient; durch fraktio tiertes Eluieren mit Benzol, enthaltend 10 % Essigsäu- reäthylester, erhält man zuerst das amorphe Isomere B; Infrarot-Absorptionsbanden (in Methylenchlorid) bei 5,7 , 5,8 (breit), 5,85 , 6,0 6,12 und 11,75 (breit);
gefolgt von einem Gemisch der beiden Isomeren A und B, bestehend zur Hauptsache. aus Isomerem A, dann mit einem 85:15-Gemisch von Benzol und Essigsäureäthylester nahezu reines Isomeres A, gefolgt von reinem amorphem Isomerem A; [a]D = -122 + 20 (C = 0,460 in Chloroform), Infrarot-Absorptionsbanden (in Methylenchlorid) bei 5,7 , 5,8 (breit), 5.9 ,
6,05 (?, 6,25 G und 12,15 (breit), welches mit einem 75:25-Gemisch der gleichen Lösungsmittel vollständig ausgewaschen wird.
Beispiel 2
Ein Gemisch von 0,09 g
3-tert-Butyloxycarbonyl-4,4 dimethyl-azetidino[3,2-d]thiazolidin-2-on und 0,27 g des rohen
3,3-Diformyl-acrylsäure-2,2,2 trichloräthylesters in 10 ml Xylol wird während 10 Stunden bei 1100 C erhitzt und dann eingedaluplc. Der Rückstand wird in
Benzol aufgenommen, die organische Lösung dreimal mit gesättigter Natriumhydrogencarbonatlösung extra hiert, die vereinigten wässrigen Lösungen mit Zitro nensäure angesäuert und mit Methylenchlorid dreimal extrahiert. Nach dem Trocknen über wasserfreiem
Natriumsulfat werden die organischen E=trakLe einge dampft und der erhaltene Rückstand an säuregewa schenem Silicagel chromatographiert.
Der reine
3-tert.-Butyloxycarbonyl-a diformylmethyl-4,4-dimethyl-2-oxo-1 azetidino[3,2-d]thiazolidinessigsäure-2,2,2 trichloräthylester wird wie im Beispiel 1 beschrieben als ein Gemisch der reinen Isomeren A und B mit Gemischen von Benzol und Essigsäureäthylester eluiert.
Beispiel 3
Zu 400 ml schwach siedendem n-Octan (Kp.
125-129 ) wird unter Rühren und Durchleiten eines schwachen Stickstoffstroms eine Lösung von 6 g des 3,3-lDiformyl-äpfelsäure-2,2,2-trichloräthylesters in
100 ml 1,2-Dimethoxyäthan innert 45 Minuten zuge tropft. Dabei wird die Badtemperatur so eingestellt, dass gleichzeitig; mittels eines Destillieraufsatzes 170 ml Lösungsmittel abgetrennt werden kann. Nach vollstän diger Zugabe werden noch weitere 20 ml Lösungsmit tel abdestilliert und das Gemisch in Eis gekühlt. Nach dem Filtrieren wird zum klaren, tiefgelben Filtrat, ent haltend den
3,3-Diformyl-acrylsäure
2,2,2-trichloräthylester.
1,2 g des 3-tert.-Butyloxycarbonyl-4,4-dimethyl- azetidino[3,2-d]thiazolidin-2-ons zugegeben und das Gemisch wird während 21 Stunden bei 80 erhitzt. Nach dem Abkühlen wird das Reak tionsgemisch mit 75 mg einer gesättigten Natriumhy- drogencarbonatlösung ausgeschüttelt. Nach dem Ab trennen der beiden Phasen wird das entstandene gelbe
Harz in Methylenchlorid aufgenommen; die beiden organischen Lösungen werden 6mal mit je 75 ml einer gosättigten Natriumhydrogencarbonatlösung extrahiert.
Die alkalisch-wässrigen Extrakte werden einmal mit
100 ml Ather gewaschen, dann mit kristalliner Zitro nensäure auf pEv @ 3I, angesänert und zweimal mit Methylenchlorid extrahiert. Man erhält so den erwünschten 3-tert.-Butyloxycarbonyl- -diformylmethyl4,4-dimetyhyl-2-oxo-1-azetidino[3,2-d]thiazolidinessigsäure-2,2,2-trichloräthylester.
Das rohe Produkt wird in 6 Stufen zwischen je 100 ml und 100 ml Phosphatpuffer (pH 8,0) verteilt, wobei die Pufferlösung als mobile Phase und die Benzollösung als stationäre Phase dient; die Produkte werden aus den wässrigen Phasen durch Ansäuern mit kristalliner Zitronensäure gewonnen.
Beispiel 5
Eine Lösung von 12 g 3.3-13iformyl-milchsäure-2,2,2- trichloräthylester in 200 ml 1,2-Dim-ethoxy-ätlian wird während 30 Minuten langsam zu 800 ml schwach siedendem Octan gegeben. Ein langsamer Strom trockener Stickstoff wird durch die Lösung geleitet und durch Destlllatiou 200 ml der Flüssigkeit während und 100 ml nach der Zugabe abgetrennt. Die gelbe Lösung wird dann ge kühlt und filtriert und das Filtrat, enthaltend den 3,3-Diformyl-acrylsäure-2,2,2trichloräthylester, wird mit 2 g 3-tert.-Butyloxycarbonyl-4,4-dimethylazetidino[3,2-d]thiazolidin-2-on versetzt und während 67 1, Stunden gerührt.
Das Reaktionsgemisch wird bei 50 und unter Wasserstrahlpumpenvakuum auf ein Volumen von 300 ml konzeniriert und mit 200 ml Ather verdünnt.
Die Lösung wird abdekantiert und 5mal mit 100 ml gesättigtem wässrigem Natriumhydrogencarbonat gewaschen. Jedes Einzelextrakt wird mit 150 ml Pentan gewaschen, wobei die gleiche organische Lösung für alle wässrigen Extrakte verwendet wird. Die wässrigen Phasen werden jeweils sofort in 250 ml eiskaltes Methylenchlorid gerührt und die Suspension mit fester Zitronensäure sauer gestellt; die nachfolgenden Natriumhydrogencarbonatlösungen werden zur gleichen Suspension gegeben und diese mit fester Zitronensäure sofort angesäuert. Die organische Lösung wird abgetrennt, die saure Phase wird 2mal mit 200 ml Methylenchlorid extrahiert und die vereinigten organischen Lösungen getrocknet und eingedampft; man erhält so ein saures Produkt.
Die Äther-Oetan-Lösung wird mit der Pentanlösung vereinigt, getrocknet und eingedampft; man erhält so ein neutrales Produkt.
Das nach dem Dekantieren der Äther-Oetan- Lösung erhaltene gummiartige Material wird in 200 ml Methylenchlorid gelöst und 5mal mit 100 ml einer gesättigten Natriumhydrogencarbonatlösung extrahiert.
Jedes der fünf wässrigen Extrakte wird mit Zitronensäure angesäuert und wie oben beschrieben zum sauren Produkt aufgearbeitet. Die Methylenchloridlösung wird getrocknet und liefert eine weitere Menge des Neutrai -Produkts. Das vereinigte saure Material wird in 110 g mit Säure gewaschenem Silicagel chromatographiert: Fraktion Lösungsmittel Volumen (in ml) Eluat (in g)

:15) 2100 0,152 Die Fraktionen 10-16 (nicht reines Addukt B) werden mittels Gegenstromverteilung mit Hilfe eines pH 6,8-Phosphatpuffers über 13 Stufen gereinigt, wobei man jeweils 40 ml eines l:l-Gemisches von Benzol und Äther als stationäre Phase und 40 ml des Puffen auf jeder Stufe verwendet Die wässrige Phase wird auf jeder Stufe mit Zitronensäure angesäuert, mit Methylenchlorid extrahiem und der Methylenchloridextrakt mit der Äther-Benzol-Lösung vereinigt, getrocknet und verdampft.
Aus den Stufen 6-12 erhält man das reine Isomere 13 des 3-tert.-Butyloxycarbonyl-α-diformylmethyl- 4,4-dimethyl-2-oxo-1-azetidino [3,2-d]thiazolidin-essigsäure2,2,2-trichloräthylesters; Infraro+-Absorptionsspektrum (in Methylenchlorid): banden bei 3,2 (breit), 3,5 (breit), 5,65 (Schulter), 5,7 , 5,88 , 6,02 und 6,15 ; [α]D = -188 +1 (c = 0,72 in Chloroform).
Das A-Isomere des 3-tert.-Butyloxycarbonyl-a- diformylmethyl4,4-dimethyl-2-oxo-1-azetidino [3,2-d]thiazolidinessigsäure-2,2,2 trichloräthylestcrs wird rein aus den Fraktionen 20-37 erhalten und zeigt Infrarot-Absoprtionsbanden (in Methylenchlorid) bei zur (breit), 3,5 cm (breit), 5,65 (Schulter), 5,7, 5,9 i, 6,06 und 6,25 u; [a]D = -1220 + 2" (c = 0,46 in Chloroform).

Substituents in the 4-position are hydrocarbon, especially aliphatic hydrocarbon radicals, such as lower alkyl, e.g. B. ethyl, n-propyl, isopropyl or preferably methyl groups, as well as aromatic, especially phenyl groups, or araliphatic hydrocarbon radicals, especially phenylalkyl, e.g.
Benzyl or phenylethyl groups, as well as functionally modified, e.g. esterified carboxyl groups, such as carb-lower alkoxy, e.g. B. Carbomethoxy or carb ethoxy groups. The two substituents can also be taken together and represent a divalent hydrocarbon, especially aliphatic hydrocarbon radical, such as a lower alkylene, e.g.
1,4-butylene or 1,5-pentylene group. as well as a phthaloyl group or an oxo or thiono group. The abovementioned Kohienwasserstoffre- ste are unsubstituted or can z. B. by lower alkyl, such as methyl or ethyl groups, lower alkoxy, such as methoxy or ethoxy groups, halogen, such as fluorine, chlorine or bromine atoms, haloalkyl, such as trifluoromethyl groups, or other suitable groups.
Esters of diformylmethylene methane carboxylic acid esters are z. B. those with alcohols of any kind, especially with aliphatic alcohols such as alkanols, especially lower alkanols, e.g. Methanol, ethanol, n-propanol or tert-butanol, cycloaliphatic alcohols such as cycloalkanols, e.g. Cyclohexanol, or araliphatic alcohols such as phenyl lower alkanols, e.g. Benzyl alcohol or diphenylmethanol, where the abovementioned alcohols are unsubstituted or may contain lower alkyl, lower alkoxy, nitro or trifluoromethyl groups or, in particular, halogen atoms and other groups as substituents.
Halogenated lower alkanols, such as 2,2,2-trichloroethanol, are particularly suitable as substituted hydroxy compounds which esterify the carboxylic acid.
The above reaction is carried out so that the addition proceeds while largely avoiding the decomposition of the olefin; H. at room temperature or by heating the reaction mixture to about 180 "C, preferably between about 50 and about 1200 C, if desired, in the presence of a suitable diluent and / or a condensing agent, if necessary, in an inert gas atmosphere and / or in a closed vessel.
In compounds obtained, substituents of various types, e.g. T. by methods known per se, can be converted into others. E.g. convert a carboxy group esterified with a 2,2,2-trihaloethanol, especially 2,2,2-trichloroethanol, into the free carboxyl group in a peculiar way by means of reducing agents. Suitable agents are chemical reducing agents such as nascent hydrogen obtained e.g. by the action of metals, metal alloys or amalgams on what oxygen-releasing agents such as zinc, zinc alloys, eg. B.
Zinc copper, or zinc amalgam in the presence of acids such as organic carboxylic acids, e.g. Acetic acid, or alcohols such as lower alkanols, alkali metal, e.g. B. sodium or potassium amalgam or aluminum amalgam, in the presence of moist ether or lower alkanols, also alkali metals, e.g. B.
Lithium, sodium or potassium, or alkaline earth metals, e.g. B. Caloium, in liquid ammonia, optionally with the addition of alcohols, such as a lower alkanol. Furthermore, a 2,2,2-trihaloethyl, in particular a 2,2,2-trichloroethyl ester can be obtained by treatment with reducing metal salts such as chromium-II compounds, e.g. Chromium (II) chloride or chromium (II) acetate, preferably in the presence of aqueous media containing water-miscible organic solvents, such as lower alkanols, lower alkanecarboxylic acids or ethers, e.g. B.
Methanol, ethanol, acetic acid, tetrahydrofuran, dioxane, ethylene glycol dimethyl ether or diethylene glycol dimethyl ether can be converted into the free acid.
Compounds with a free carboxy group can e.g. B. in their salts, such as. B. alkali or alkaline earth metal or ammonium salts are converted. Free carboxy groups can be formed by methods known per se, e.g. by treating with a diazo compound such as a diazo lower alkane, e.g. B. diazomethane or diazoethane, or a phenyl-diazo-lower alkane, e.g. Phenyldiazomethane or diphenyldiazomethane, can be esterified. At the same time one of the formyl groups can be converted into an enol ether.
By treating with complex-forming heavy metal salts, the process products can, if desired, be converted into salts, e.g. B. copper, iron, Magnesiltm-, zinc or lead salts, transfer.
When the process products are treated with acylating agents, such as acid anhydrides or acid halides, if appropriate in the presence of bases, such as pyridine, coilidine or triethylamine, enol acrylates are obtained.
Mixtures of isomers obtained can be prepared by methods known per se, e.g. separated into the individual isomers by fractional crystallization, adsorption chromatography (column or thin layer chromatography) or other methods; Racemates obtained can be separated into the antipodes by forming a mixture of diastereoisomeric salts with optically active salt-forming agents, separating the mixture into the diastereoisomeric salts and converting the separated salts into the free compounds, or by fractional crystallization from optically active solvents.
The process also includes those embodiments according to which compounds obtained as intermediates are used as starting materials and the remaining process steps are carried out with these, or the process is terminated at any stage; Furthermore, starting materials in the form of derivatives, eg. B. of salts, used or formed during the reaction.
Such starting materials are preferably used and the reaction conditions are selected so that the compounds listed at the beginning as being particularly preferred are obtained.
The starting materials used in the above process are according to the applications G. No.
497 456 (Case Wo 8), or G. Nr. 497 369 (Case Wo 10).
In compounds of the formula XIa prepared according to the process, Ra is z. B. for an unsubstituted or substituted aliphatic or araliphatic hydrocarbon radical, in particular a lower alkyl or a halo-lower alkyl, e.g. the 2,2,2-trichloroethyl radical and a phenyl-lower alkyl radical.
Acyl radicals Ac are primarily those which occur in pharmacologically active N-acyl derivatives of the 7 amine <> cephalospoanic acid, such as the Thie nylacetyi-, z. B. 2-thienylacetyl, chloroethylcarbamyl, cyanoacetyl or phenylacetyl radicals, or easily split off acyl radicals, such as the remainder of a radius of carbonic acid, e.g. B. the tert-butyloxycarbonyl radical.

Compounds obtained in accordance with the method are, as shown in the formula scheme, converted into 7-amino-cephalosporanic acid or its derivatives; this conversion can e.g. B. in accordance with the procedure described in applications 0. No. 497 460 (Case Wo 12), 0. No. 497 379 (Case Wo 13) and G. No. 497 461 (Case Wo 14).

example 1
In 2 parallel batches, a solution of 6 g of 3,3-diformyllactic acid 2,2,2-trichloroethyl ester in 100 ml of 1,2-dimethoxyethane is added dropwise to 400 ml of low-boiling n-octane within 60 minutes with stirring and in a nitrogen atmosphere, with simultaneously 240 ml of solvent mixture are distilled off through a distillation attachment. The crude product is distributed in 6 stages between 100 ml and 100 ml phosphate buffer (pH 8.0) each, the buffer solution serving as the mobile phase and the benzene solution as the stationary phase; the products are obtained from the aqueous phases by acidification with crystalline citric acid.
Example 5
A solution of 12 g of 3,3-13iformyl-lactic acid-2,2,2-trichloroethyl ester in 200 ml of 1,2-dimethoxy-ether is slowly added to 800 ml of low-boiling octane over a period of 30 minutes. A slow stream of dry nitrogen is passed through the solution and 200 ml of the liquid is separated off by distillation during and 100 ml after the addition. The yellow solution is then cooled and filtered and the filtrate, containing the 3,3-diformyl-acrylic acid-2,2,2trichloroethyl ester, is treated with 2 g of 3-tert-butyloxycarbonyl-4,4-dimethylazetidino [3.2- d] thiazolidin-2-one added and stirred for 67 1, hours.
The reaction mixture is concentrated to a volume of 300 ml at 50 ml and under a water jet pump vacuum and diluted with 200 ml of ether.
The solution is decanted off and washed 5 times with 100 ml of saturated aqueous sodium hydrogen carbonate. Each individual extract is washed with 150 ml of pentane, the same organic solution being used for all aqueous extracts. The aqueous phases are each immediately stirred in 250 ml of ice-cold methylene chloride and the suspension is acidified with solid citric acid; the subsequent sodium hydrogen carbonate solutions are added to the same suspension and this is immediately acidified with solid citric acid. The organic solution is separated off, the acidic phase is extracted twice with 200 ml of methylene chloride and the combined organic solutions are dried and evaporated; an acidic product is obtained.
The ether-oetane solution is combined with the pentane solution, dried and evaporated; you get a neutral product.
The gummy material obtained after decanting the ether-oetane solution is dissolved in 200 ml of methylene chloride and extracted 5 times with 100 ml of a saturated sodium hydrogen carbonate solution.
Each of the five aqueous extracts is acidified with citric acid and worked up to the acidic product as described above. The methylene chloride solution is dried and provides an additional amount of the Neutrai product. The pure isomer 13 of 3-tert-butyloxycarbonyl-α-diformylmethyl-4,4-dimethyl-2-oxo-1-azetidino [3,2-d] thiazolidine-acetic acid2,2 is obtained from steps 6-12 , 2-trichloroethyl ester; Infraro + absorption spectrum (in methylene chloride): bands at 3.2 (broad), 3.5 (broad), 5.65 (shoulder), 5.7, 5.88, 6.02, and 6.15; [α] D = -188 +1 (c = 0.72 in chloroform).
The A-isomer of 3-tert-butyloxycarbonyl-a-diformylmethyl4,4-dimethyl-2-oxo-1-azetidino [3,2-d] thiazolidinacetic acid-2,2,2 trichloroethyl ester is pure from fractions 20-37 obtained and shows infrared absorption bands (in methylene chloride) at zur (wide), 3.5 cm (wide), 5.65 (shoulder), 5.7, 5.9 i, 6.06 and 6.25 u; [a] D = -1220 + 2 "(c = 0.46 in chloroform).
