Process for the preparation of 2-amino, 4-hydroxy, toluene The present invention relates to a new process for the preparation of 2-amino, 4-hydroxy, toluene.
This process is characterized in that an agent capable of protecting the phenol function, preferably methanesulfochloride, is made to act on the paracresol; in that the benzene ring is nitrated in the meta position relative to the phenol function, for example with a sulphonitric mixture; in that the nitro radical is reduced so as to convert it into an amino radical and in that the phenol function is then released, for example by the action of a sodium hydroxide solution.
This preparation process can therefore meet the following reaction scheme
EMI0001.0000
Amino-2, hydroxy-4, toluene, as such or chlorinated in position 5 or whose amino radical has been transformed into a carbamylmethylamino group or into a (di-ethylcarbamyl) methylamino group, finds applications in hair dyeing , as a coupler for aromatic paradiamines of formula

in which R and R 'represent hydrogen or a methyl radical, R' cannot be hydrogen when R is a methyl radical.
Here is, by way of example, how the process of the invention can be carried out by following the reaction scheme indicated above. In a first phase, the mesylated 4-hydroxy toluene is prepared in a known manner by the action of methanesulfochloride on the paracresol.
In a second phase, 2-nitro, 4-mesyloxy, toluene are prepared. For this purpose 0.1 mole (that is to say 18.6 g) of mesylated 4-hydroxy, toluene are dissolved at low temperature (0 to 5c) C) in 80 cm3 of concentrated sulfuric acid. At this same temperature, the sulfonitric mixture is added having the following composition:
EMI0001.0008
- <SEP> acid <SEP> nitric <SEP> of <SEP> density <SEP> 1.49 <SEP> ............ <SEP> 4.7 <SEP> cm3
<tb> - <SEP> sulfuric acid <SEP> <SEP> of <SEP> density <SEP> 1.83 <SEP> ........ <SEP> 3.8 <SEP> cm3 Then poured the reaction mixture on ice and the crude reaction product is filtered off. After recrystallization from ethanol, this product melts at 105-106 ° C. The yield of the reaction is about 89%.
The analysis gave the following results
EMI0001.0011
Analysis <SEP> Calculated <SEP> for <SEP> C $ H905NS <SEP> Found
<tb> C <SEP> 0/0 <SEP> 41.6 <SEP> 41.40-41.57
<tb> H <SEP> 0/0 <SEP> 3.90 <SEP> 3.90- <SEP> 4.06
<tb> N <SEP> 0/0 <SEP> 6.06 <SEP> 6.13- <SEP> 6.10 In a third phase, 2-amino, 4-mesyloxy, toluene is prepared. For this purpose, under the usual conditions, 0.86 mol (ie 198 g) of 2-nitro, 4-mesyloxy, toluene is reduced with the tin hydrochloric acid mixture. By cooling the reaction mixture, the desired product is precipitated in the form of chlorostannate which is filtered off and dissolved in water.
The product is then treated with a stream of hydrogen. sulphide and filtered off tin sulphide formed. The filter obtained is then concentrated under vacuum, and the desired product is precipitated by cooling. The mixture is filtered off and the 2-amino hydrochloride, 4-mesyl-oxy, toluene, is obtained with a yield close to 85%.
In a fourth phase, the desired chemical compound is prepared which can serve as a coupler, namely 2-amino, 4-hydroxy, toluene. To this end, 0.05 mole (that is to say 12 g) of 2-amino, 4-mesyloxy, toluene hydrochloride, is treated with 125 cm3 of twice normal sodium hydroxide, at reflux for two hours under an atmosphere nitrogen. After cooling, the solution is neutralized with hydrochloric acid and the 2-amino, 4-hydroxy and toluene are filtered off. The yield of the reaction is about 80%. The product after recrystallization from ethyl acetate melts at 159o C.
