méthyle ou éthyle, et R2 et R3 des radicaux alcoyle inférieurs., l'un d'entre eux pouvant être toutefois un atome d'hydrogène Ce procédé est caractérisé en ce que l'on alcoyle l'amine primaire (R2 = R3 = H) ou secondaire (R2 = H, R3 = alcoyle inférieur) correspondante. Cette alcoylation peut être effectuée selon les mé thodes classiques.
Lorsque Y représente un atome de soufre ou un groupe SO, il peut être oxydé ultérieurement en groupe SO ou SOz.
Les nouveaux produits, ainsi que leurs sels et leurs dérivés d'ammonium quaternaire, trouvent leur domaine d'emploi en médecine humaine et v6t6ri- naire (adjuvants de narcose, déconnecteurs du sys tème nerveux végétatif, produits utilisables en psy chiatrie, etc.).
Les produits optiquement actifs peuvent être obtenus par dédoublement des racémiques ou par synthèse à partir de matières premières -ou inter médiaires optiquement actives.
Les exemples suivants montrent comment le pro cédé selon l'invention peut être mis en #uvre. Les points de fusion indiqués ont été déterminés au banc Kofler.
<I>Exemple 1</I> 8,4 g de chlorhydrate de (méthoxy-3' phénothia- zinyl-10')-3 méthyl-2 amino-1 propane optiquement actif sont dissous dans 150 cm3 de méthanol avec 45 g d'une solution aqueuse à 331)/o de formol.
On ajoute 0,5 g de catalyseur au platine selon Adams et hydrogène à température et sous pression ordi naires. Quand l'absorption d'hydrogène est terminée, on filtre le catalyseur, évapore le méthanol sous pression réduite et reprend le résidu par de la soude diluée et du chloroforme. La solution chloroformique est lavée à l'eau, puis séchée sur carbonate de potas sium.
Par évaporation du solvant, on obtient 7,5 g de (méthoxy-3' phénothiazinyl-10')-3 méthyl-2 di- méthylamino-1 propane brut qui, après recristallisa tion dans l'heptane ou dans l'acétone, fond à 116 1180 et possède un pouvoir rotatoire [(x]20 = - 121, (c = 5 0/0, CHC13).
L'amine primaire initiale, dont le chlorhydrate fond à 234-236o, peut être obtenue par réduction à l'hydrure de lithium-aluminium au sein du tétra- hydrofuranne, du (méthoxy-3' phéno:
thiazinyl-10')-3 méthyl-2 propionitrile fondant à 110-1120. Ce nitrile peut être préparé par action du cyanure de potas sium, en solution hydroalcoolique sur le p. toluène sulfonate de (méthoxy-3' phénothiazinyl-10')-1 pro- pyle-2 fondant à 96-1000, lui-même préparé par action du p.
toluènesulfochlorure sur le (méthoxy-3' phénothiazinyl-10')-1 propanol-2 huileux, au sein de la pyridine anhydre.
Cet alcool phénothiazinique peut être obtenu par condensation du L oxyde de propy lène décrit dans la littérature sur la méthoxy-3 phé- nothiazine lithiée au sein du tétrahydrofuranne,
le L oxyde de propylène utilisé en l'occurrence étant obtenu par l'intermédiaire de l'ester bromé à partir du L propylèneglycol préparé selon Baer et Fischer, J. Am. Chem. Soc. , 70, 609 (1948).
<I>Exemple 2:</I> 8,9 g d'(éthyl-3' phénothiazinyl-10')-3 méthyl-2 amino-,l propane optiquement actif sont dissous avec 28,3 cm3 d'acide chlorhydrique normal et 55 cm3 d'une solution aqueuse à 331/o de formol dans 190 cm3 de méthanol. On ajoute 0,5 g de catalyseur au platine Adams et agite à température ordinaire et sous une légère pression d'hydrogène.
Quand l'ab sorption est terminée, on filtre le catalyseur, chasse le méthanol au bain-marie, traite le résidu aqueux par de la soude (d =<B>1,33)</B> en excès et extrait à l'éther.
Par évaporation de l'éther, il reste 9 g d'(éthyl-3' phénothiazinyl-10')-3 méthyl-2 diméthylamino-1 pro pane optiquement actif. Le maléate de ce produit cristallise au sein de l'acétate d'éthyle et présente un point de fusion de 1361, et un pouvoir rotatoire [a]# = -11,5 (c = 4 %, méthanol).
L'amine primaire de départ peut être obtenue sous forme huileuse par réduction à l'hydrure de lithium-aluminium, au sein du tétrahydrofuranne, de l'(éthyl-3' phénothiazinyl-10')-3 méthyl-2 propio- nitrile optiquement actif.
Ce dernier peut être obtenu sous forme huileuse par action du cyanure de potas sium au sein de l'éthanol aqueux, sur le p. toluène- sulfonate d'(éthyl-3' phénothiazinyl-10')-1 propyle-2 de P. F. = 820. Cet ester peut être préparé par action du p. toluène sulfochlorure sur l'(éthyl-3' phénothiazinyl-10')-1 propanol-2 huileux au sein de la pyridine anhydre.
Cet alcool phénothiazinique peut être préparé par condensation du L oxyde de propy lène sur l'éthyl-3 phénothiazine lithiée, en milieu éther anhydre.
<I>Exemple 3:</I> On dissout 4,8 g de (méthoxy-3' phénothiazinyl- 10')-3 méthyl-2 méthylamino-1 propane dans 75 cm3 de méthanol. On ajoute 15 cm3 d'acide chlorhydrique normal, 15 cm3 d'une solution aqueuse de formol à 30'% et 0,2 g de catalyseur au platine selon Adams.
On agite sous une légère surpression d'hy drogène jusqu'à fin d'absorption, filtre le catalyseur et évapore le solvant. Le résidu est repris par de l'eau ; par addition de soude, il précipite une base qu'on extrait à l'éther et sèche sur carbonate de potassium. Par évaporation de l'éther, on obtient 3,5 g de (méthoxy-3' phénothiazinyl-10')-3 méthyl-2 diméthylamino-1 propane brut qui, après purifica tion par recristallisation dans l'acétone aqueuse, fond à 103() C.
En opérant de la même manière, on obtient (Ethyl-3' phénothiazinyl-10')-3 méthyl-2 diméthyl- amino-1 propane dont le chlorhydrate fond à 160 163o.
L'oxydation du dérivé de la phénothiazine obtenu peut être effectuée comme suit On oxyde pendant 17 heures à 60 une solution acétique de 6,6 g de (méthoxy-3' phénothiazinyl- 10') - 3 méthyl - 2 diméthylamino -1 propane par 4,3 cm3 d'eau oxygénée à 130 volumes en présence de 0,25 cm3 d'acide sulfurique d = 1,83.
La base brute est séparée par dilution à l'eau, alcalinisation et extraction à l'acétate d'éthyle ; elle est purifiée par l'intermédiaire de son fumarate acide. On obtient ainsi le (méthoxy-3' dioxo-9',9' phénothiazinyl-10')-3 méthyl - 2 diméthylamino -1 propane fondant vers 110o.
Le dédoublement du dérivé racémique de la phénothiazine peut être effectué comme suit 3,28 g de (méthoxy-3' phénothiazinyl-10')-3 mé- thyl-2 d.iméthylamino-1 propane racémique, 3,65 g de chlorhydrate de cette base et 1,5 g d'acide tar trique lévogyre sont dissous dans 50 cm3 d'isopro- panol, bouillant. On laisse refroidir jusqu'à 600, amorce et agite pendant quelques heures à cette température.
On essore la masse en maintenant tou jours la température à 60 , lave les cristaux avec de l'isopropanol chaud tût sèche sous vide. On obtient ainsi 2,1 g de 1-tartrate de (méthoxy-3' phénothiazi- nyl-10')-3 méthyl-2 diméthylamino-1 propane lévo gyre.
La base libérée à partir de ce sel par alcali nisation, soit 1,4 g, présente, après recristallisation dans l'éthanol, un point de fusion de 1240 et un pouvoir rotatoire [a]D=-15,3o (c=5,2%, CHC13).
En opérant comme ci-dessus, à partir de 100 g de (méthoxy - 3' phénothiazinyl -10') - 3 méthyl - 2 di- méthylamino-1 propane racémique, 111 g de chlor- hydrate de cette base, 45,7 g d'acide tartrique dextro gyre et 3000 em3 d'isopropanol, on obtient 76 g de d-tartrate de (méthoxy-3' phénothiazinyl-10')
-3 mé- thyl-2 diméthylamino-1 propane dextrogyre, antipode optique du produit décrit ci-dessus.
Les eaux-mères de cristallisation sont concentrées à 300 em3. Au liquide restant, on ajoute 112 cm-3 de potasse alcoolique 2,6N. On refroidit lentement jusqu'à 0 en amorçant avec la base (méthoxy-3' phénothiazinyl-10')-3 méthyl-2 diméthylamino-1 pro pane lévogyre. On essore le mélange de, sels minéraux et de base quia précipité. On reprend par de l'eau et de l'éther.
On décante. Par évaporation de la solution éthérée, on obtient 40 g de base (méthoxy-3' phénothiazinyl-10')-3 méthyl-2 diméthylamino-1 pro pane lévogyre déjà décrit ci-dessus.
L'oxydation de ce composé, effectuée dans les mêmes conditions que décrit ci-dessus, conduit au (méthoxy-3' dioxo-9',9' phénothiazinyl-10')-3 mé- thyl-2 diméthylamino-1 propane lévogyre, qui fond à 126() et possède un pouvoir rotatoire [a]D _ -25,4o (c = 5,1'%, chloroforme).

In the above formula, Y represents a sulfur atom or a - SO - or - S02 - radical, X a lower alkyl or lower alkyloxy radical, preferably in position 3 of the phenothiazine ring (Beilstein numbering), RI represents lower alkyl radi cal and preferably
methyl or ethyl, and R2 and R3 from lower alkyl radicals, one of them possibly being a hydrogen atom This process is characterized in that the primary amine is alkylated (R2 = R3 = H ) or secondary (R2 = H, R3 = lower alkyl) corresponding. This alkylation can be carried out according to conventional methods.
When Y represents a sulfur atom or an SO group, it can be further oxidized to an SO or SOz group.
The new products, as well as their salts and their quaternary ammonium derivatives, find their field of use in human and veterinary medicine (narcosis adjuvants, vegetative nervous system disconnectors, products usable in psychiatry, etc.) .
The optically active products can be obtained by resolution of the racemates or by synthesis from optically active raw materials or intermediates.
The following examples show how the process according to the invention can be implemented. The indicated melting points were determined on the Kofler bench.

This phenothiazine alcohol can be obtained by condensation of L propylene oxide described in the literature on 3-methoxy-phenothiazine lithiated in tetrahydrofuran,
the L propylene oxide used in this case being obtained by means of the brominated ester from L propylene glycol prepared according to Baer and Fischer, J. Am. Chem. Soc. , 70, 609 (1948).
<I> Example 2: </I> 8.9 g of (ethyl-3 'phenothiazinyl-10') - 3 methyl-2 amino-, l optically active propane are dissolved with 28.3 cm3 of normal hydrochloric acid and 55 cm3 of a 331% aqueous solution of formalin in 190 cm3 of methanol. 0.5 g of Adams platinum catalyst is added and stirred at room temperature and under mild hydrogen pressure.

The starting primary amine can be obtained in oily form by reduction with lithium aluminum hydride, in tetrahydrofuran, of (3 'ethyl phenothiazinyl-10') - 3 methyl-2 propionitrile optically. active.
The latter can be obtained in oily form by the action of potassium cyanide in aqueous ethanol, on p. P.F. (3 'ethyl-phenothiazinyl-10') -1-propyl-toluenesulphonate = 820. This ester can be prepared by the action of p. toluene sulfochloride on oily (3 'ethyl phenothiazinyl-10') -1-propanol-2 in anhydrous pyridine.
This phenothiazine alcohol can be prepared by condensation of L propylene oxide with lithiated 3-ethyl phenothiazine, in an anhydrous ether medium.
<I> Example 3: </I> 4.8 g of (methoxy-3 'phenothiazinyl-10') - 3-methyl-2-methylamino-1 propane are dissolved in 75 cm3 of methanol. 15 cm3 of normal hydrochloric acid, 15 cm3 of a 30% aqueous formalin solution and 0.2 g of Adams platinum catalyst are added.
Stirred under a slight overpressure of hydrogen until absorption is complete, the catalyst is filtered off and the solvent evaporated. The residue is taken up in water; by adding sodium hydroxide, it precipitates a base which is extracted with ether and dried over potassium carbonate. By evaporation of the ether, 3.5 g of crude (methoxy-3 'phenothiazinyl-10') - 3-methyl-2-dimethylamino-1-propane are obtained which, after purification by recrystallization from aqueous acetone, melts at 103 () VS.
By operating in the same manner, one obtains (3-ethylphenothiazinyl-10 ') - 3-methyl-2-dimethyl-1-amino propane, the hydrochloride of which melts at 160 163 °.
The oxidation of the phenothiazine derivative obtained can be carried out as follows An acetic solution of 6.6 g of (methoxy-3 'phenothiazinyl-10') - 3 methyl - 2 dimethylamino -1 propane is oxidized for 17 hours at 60 with 4.3 cm3 of hydrogen peroxide at 130 volumes in the presence of 0.25 cm3 of sulfuric acid d = 1.83.
The crude base is separated by dilution with water, basification and extraction with ethyl acetate; it is purified by means of its acid fumarate. This gives (methoxy-3 'dioxo-9', 9 'phenothiazinyl-10') - 3 methyl - 2 dimethylamino -1 propane, melting at around 110o.
The resolution of the racemic derivative of phenothiazine can be carried out as follows 3.28 g of (3 'methoxyphenothiazinyl-10') - 3-methyl-2 d. 1-methylamino-propane, 3.65 g of racemic hydrochloride. this base and 1.5 g of levorotatory tar teric acid are dissolved in 50 cm3 of isopropanol, boiling. Allowed to cool to 600, initiate and stir for a few hours at this temperature.
The mass is filtered off while still maintaining the temperature at 60, the crystals washed with hot isopropanol and dried under vacuum. 2.1 g of (methoxy-3 'phenothiazinyl-10') -3-methyl-2-dimethylamino-1 propane levogyre is thus obtained.

By operating as above, from 100 g of (methoxy - 3 'phenothiazinyl -10') - 3 methyl - 2 di-methylamino-1 racemic propane, 111 g of hydrochloride of this base, 45.7 g dextro gyre tartaric acid and 3000 em3 of isopropanol, 76 g of (methoxy-3 'phenothiazinyl-10') d-tartrate are obtained
-3-methyl-2-dimethylamino-1 propane dextrorotatory, optical antipode of the product described above.
The mother liquors of crystallization are concentrated at 300 em3. To the remaining liquid, 112 cm-3 of 2.6N alcoholic potassium hydroxide are added. Slowly cooled to 0 while priming with base (3-methoxy-10 'phenothiazinyl) - 3-methyl-2-dimethylamino-1 pro pane levorotatory. The mixture of mineral salts and base which precipitated is filtered off.
