Procédé pour la préparation de dérivés de benzodiazépines
La présente invention a pour objet un procédé poul la préparation de dérivés de benzodiazépines, ayant ld formule générale:

R, représente un atome d'hydrogène ou d'halogène, un
radical alkyle alcoxy, trifluorométhyle ou nitro.

R, et R forment ensemble un radical cycloalkyle, et
Am représente le reste d'une amine primaire aliphatique,
arylaliphatique ou alicyclique. de la diméthylamine
ou d'une amine cyclique.
Ces nouvelles substances présentent une activité thérapeutique importante et conviennent comme produits intermédiaires pour la préparation de substances à activité thérapeutique.
Le procédé selon l'invention est caractérisé en ce qu'on fait réagir un composé ayant la formule générale:

dans laquelle R, R, et R, ont les significations indiquées
ci-dessus, avec une amine primaire aliphatique, arylali
phatique ou alicyclique, la diméthylamine ou une amine
cyclique.
On a trouvé que l'atome de chlore dans les matières
de départ possède une réactivité qui correspond à peu
près à celle du chlore dans un produit tel que le chlorure
de benzyle.
Quand on traite de tels composés avec un excès d'une
amine primaire aliphatique, arylaliphatique ou alicy
clique ou par la diméthylamine ou encore par une amine cyclique telle que la pyrrolidine, la pipéridine, la morpholine, la N-méthyl pipérazine ou la N-hydroxyméthyl pipérazine et ses dérivés d'estérification et d'éthérification, soit sans solvant au bain-marie, soit en solution alcoolique à la température ambiante, on obtient, avec des rendements variables, des composés basiques porteurs, en position 3 du noyau benzodiazépinique, du groupement amino correspondant à l'amine mise en oeuvre. Le remplacement du chlore par le groupe basique est lié à une transposition.
Leurs spectres IR, dans le chlomre de méthylène, présentent entre autres une bande N-H à 3400 cm-t et une bande C = O à 1670-1700 cm-t suivant les cas.
Cependant, si l'on utilise la diéthylamine ou un de ses homologues supérieurs, tels que la dibutylamine, et si le radical fixé au 5e atome de carbone est un radical chloro-l cyclohexyle ou chloro-l cyclopentyle, la réaction suit un autre cours et, pour la plus grande part, conduit avec déshydrohalogénation à la création d'une double liaison.
Dans le tableau 1 qui suit, on trouvera un certain nombre de composés qui peuvent être préparés par le procédé selon l'invention.

Les exemples suivants illustrent l'invention; F indique le point de fusion mesuré au banc de Kofler.
Exemple 1:
Chloro-7 cyclohexyl-5 (méthyl-4' pipérazinyl-1')-3
oxo-2 dihydro-2,3
1H-benzo[f]iazépine.1,4 (4379 CB)
On chauffe quelques minutes sur le bain-marie 5 g de 4253 CB (chloro-7 (chloro-l' cyclohexyl)-5 oxo-2 dihydro-2,3 1H-benzo[f]diazépine-1,4) et 5 ml de méthyl-1 pipérazine en agitant ; le solide se dissout rapidement en donnant une solution brun jaune. Après refroidissement, on ajoute de l'eau et de l'éther; il se sépare un solide incolore. On essore, on lave et l'on recristallise dans de l'acétate d'éthyle. Cristaux incolores.
(5g: rendement 83 /o) Fk = 2400.
Exemple 2:
Chloro-7 cyclohexyl-5 diméthylamino-3
oxo-2 dihydro-2,3
IH-benzo [f]diazépine- 1,4 (4268 CB)
Méthode B
On agite à la température ambiante 5 g de 4253 CB broyé avec 50 ml d'une solution à 20 % de diméthylamine dans du méthanol anhydre. Le solide se dissout rapidement. On laisse une nuit au repos: il se sépare peu à peu un solide incolore. On essore, on lave avec un peu de méthanol et l'on recristallise dans de l'éthanol absolu. Cristaux incolores.
(3 g; rendement 58 %) Fk = 222t.
Différents produits synthétisés par les méthodes A et B décrites dans les exemples 1 et 2 sont réunis avec leurs caractéristiques dans le tableau 2.

On a soumis les différents composés à des épreuves à dessein de rechercher leurs activités physiologiques.
Les différents composés ont été administrés à différentes doses à des lots de dix animaux (rats ou souris) qui ont subi des épreuves mettant en évidence leur activité sur le système nerveux central. Les réponses aux différentes épreuves sur les lots d'anirnaux traités ont toujours été comparées à celles que l'on a obtenues sur des lots d'animaux témoins.
Les différentes épreuves utilisées ont été les suivantes: - traction (souris) qui met en évidence l'action de relâ
chement sur les muscles striés, - tige tournante (souris) qui met en évidence l'effet sur
l'équilibration, - exploration (souris) qui fait appel à la curiosité natu
relle de la souris, - actographie spontanée (souris) et motricité spontanée
(rat), qui permettent de mettre en évidence l'action des produits sur la sphère psycho-motrice, la souris présentant normalement une mobilité très importante alors que le rat, au contraire, se déplace peu.
Combativité (rat).
Cette épreuve consiste à rendre agressifs des groupes de deux rats mâles l'un par rapport à l'autre sous l'influence du passage d'un courant électrique de voltage réglable dans le plancher de la cage.
lElectrochoc (souris).
En réglant le voltage et l'intensité du courant utilisé, on peut déterminer le pourcentage d'animaux traités et protégés par rapport à des témoins.
- Convulsions induites par le pentétrazol (souris).
Le pentétrazol injecté à la dose de 100 à 125 mg/kg par voie intrapéritonéale chez la souris détermine des Tableau 3

= non significatif crises convulsives violentes et mortelles qui peuvent être combattues par des composés à activité anticonvulsivante.
Les résultats obtenus sont groupés sous la forme d'un tableau (tableau 3) dans lequel les chiffres précédés du signe (-) expriment des pourcentages d'inhibition par rapport à des lots d'animaux témoins; P indique la protection en pour-cent. Les doses sont exprimées en milligrammes par kilo d'animal et la voie d'administration a toujours été la voie orale.
L'examen des résultats montre que, dans l'ensemble, tous ces composés exercent une ou plusieurs activités sur le système nerveux central, mais à des doses plus ou moins élevées.
L'activité sur le système nerveux central et les propriétés sédatives et anticonvulsivantes sont relativement faibles.
Le 4379 CB exerce un effet myorelaxant assez marqué et agit sur les centres psychomoteurs en diminuant les déplacements spontanés chez la souris, mais la propriété anticonvulsivante est conservée.
Un composé particulièrement important est le 4379 CB (exemple 1).
La préparation des matières de départ, qui est étrangère à l'objet du présent brevet, petit se faire en faisant réagir un dérivé de benzodiazépine ayant la formule générale:

These new substances exhibit significant therapeutic activity and are suitable as intermediates for the preparation of substances with therapeutic activity.
The process according to the invention is characterized in that a compound having the general formula is reacted:

above, with an aliphatic primary amine, arylali
phatic or alicyclic, dimethylamine or an amine
cyclic.
It has been found that the chlorine atom in materials
starting point has a reactivity that corresponds to little
close to that of chlorine in a product such as chloride
benzyl.
When treating such compounds with an excess of one
aliphatic, arylaliphatic or alicy primary amine
clique or by dimethylamine or by a cyclic amine such as pyrrolidine, piperidine, morpholine, N-methyl piperazine or N-hydroxymethyl piperazine and its esterification and etherification derivatives, either without solvent in the bath- combined, either in alcoholic solution at room temperature, basic compounds are obtained, with variable yields, carrying, in position 3 of the benzodiazepine ring, the amino group corresponding to the amine used. The replacement of chlorine by the basic group is linked to a transposition.
Their IR spectra, in methylene chloride, exhibit, among other things, an N-H band at 3400 cm-t and a C = O band at 1670-1700 cm-t depending on the case.
However, if one uses diethylamine or one of its higher homologues, such as dibutylamine, and if the radical attached to the 5th carbon atom is chloro-1 cyclohexyl or chloro-1 cyclopentyl, the reaction follows a different course. and, for the most part, leads with dehydrohalogenation to the creation of a double bond.
In Table 1 which follows, a number of compounds will be found which can be prepared by the process according to the invention.

The following examples illustrate the invention; F indicates the melting point measured on the Kofler bench.
Example 1:
7-chloro-5-cyclohexyl (4-methyl-1-piperazinyl) - 3
2-oxo-2,3-dihydro
1H-benzo [f] iazepine.1,4 (4379 CB)
5 g of 4253 CB (chloro-7 (chloro-l 'cyclohexyl) -5 oxo-2 dihydro-2,3 1H-benzo [f] diazepine-1,4) and 5 ml are heated for a few minutes on the water bath. 1-methyl piperazine with stirring; the solid dissolves rapidly giving a yellow brown solution. After cooling, water and ether are added; a colorless solid separates. It is filtered off, washed and recrystallized from ethyl acetate. Colorless crystals.

Example 2:
7-chloro-5-cyclohexyl 3-dimethylamino
2-oxo-2,3-dihydro
IH-benzo [f] diazepine- 1,4 (4268 CB)
Method B
5 g of crushed 4253 CB are stirred at room temperature with 50 ml of a 20% solution of dimethylamine in anhydrous methanol. The solid dissolves quickly. It is left to stand overnight: a colorless solid gradually separates. It is filtered off, washed with a little methanol and recrystallized from absolute ethanol. Colorless crystals.
(3 g; yield 58%) Fk = 222t.

The various compounds were tested for their physiological activities.
The different compounds were administered at different doses to groups of ten animals (rats or mice) which underwent tests demonstrating their activity on the central nervous system. The responses to the various tests on batches of treated animals were always compared with those obtained on batches of control animals.
The different tests used were as follows: - traction (mouse) which demonstrates the release action
on striated muscles, - rotating rod (mouse) which demonstrates the effect on
equilibration, - exploration (mouse) which appeals to natural curiosity
mouse, - spontaneous actography (mouse) and spontaneous motor skills
(rat), which make it possible to demonstrate the action of the products on the psycho-motor sphere, the mouse normally exhibiting very high mobility while the rat, on the contrary, moves little.
Combativeness (rat).
This test consists in making groups of two male rats aggressive with respect to each other under the influence of an electric current of adjustable voltage passing through the floor of the cage.
Electroshock (mouse).
By adjusting the voltage and intensity of the current used, one can determine the percentage of treated and protected animals compared to controls.
- Convulsions induced by pentetrazol (mouse).

= not significant violent and fatal convulsive seizures which can be combated by compounds with anticonvulsant activity.
The results obtained are grouped together in the form of a table (Table 3) in which the figures preceded by the sign (-) express percentages of inhibition relative to batches of control animals; P indicates protection in percent. The doses are expressed in milligrams per kilogram of animal and the route of administration has always been the oral route.
Examination of the results shows that, on the whole, all these compounds exert one or more activities on the central nervous system, but at more or less high doses.
The activity on the central nervous system and the sedative and anticonvulsant properties are relatively low.
4379 CB exerts a fairly marked muscle relaxant effect and acts on psychomotor centers by reducing spontaneous movements in mice, but the anticonvulsant property is retained.
A particularly important compound is 4379 CB (Example 1).
The preparation of the starting materials, which is foreign to the subject of this patent, can be carried out by reacting a benzodiazepine derivative having the general formula:
