<B>2</B> Verbindung Diese Verbindung besitzt eine chemotherapeutische Wirkung, insbesondere eine selektive antivirale Wirkung gegen DNA-Krankheitserreger, z.B. Herpes keralitis und Kuhpocken.
Es wurde überraschenderweise gefunden, dass bei der Behandlung eines 1-(2',3',5'-tri-O-aralkyl-B-D-arabino- Behandlung eines 1-(2',3',5'-tri-O-aralkyl-B-D-arabino- furanosyl)-4-Alkoxy bzw. Alkylthio-5-trifluormethyl- -2(1H)-pyrimidons mit Ammoniak neben dem 5-Trifluor- methylarabinosid auch 4-aminiertes Arabinosid entsteht, welches gemäss Elementaranalyse und Ultraviolett- sowie Infrarot-Spektrum, in 5-Stellung eine Cyangruppe auf weist.
Diese Verbindung kann durch wiederholte frak tionierte Kristallisation aus einer Lösung in Methanol und/oder Isopropanol oder Benzol abgetrennt und ge reinigt werden.
Das erfindungsgemässe Verfahren zur Herstellung des 1-(2',3',5'-Tri-hydroxy-ss-D-arabinfuranasyl)-cytosin der angegebenen Formel ist nun dadurch gekennzeichnet, dass man ein Nucleosid der Formel

mit Wasserstoff in Gegenwart eines Katalysators behan delt. Man erhält dieses Produkt auch> wenn die Reaktio nen in umgekehrter Reihenfolge durchgeführt werden. Als Aminierungsmittel kann man sowohl flüssiges Ammoniak als auch Ammoniumhydroxyd verwenden, zweckmässig im Überschuss.
Als Ausgangsstoff verwen det man vorzugsweise den entsprechenden Tribenzyläther, insbesondere denjenigen der in 4-Stellung eine Methoxy oder Methylthiogruppe aufweist. Die Abtrennung der Aralkylreste erfolgt zweckmässig in Gegenwart eines Lö sungsmittels. Als Katalysator bei der Hydrogenolyse eignet sich z.B. Platin, Nickel und insbesondere Palla dium.
Zweckmässigerweise stellt man den Palladium-Kataly sator im Reaktionsgefäss und im Lösungsmittel her, in welchem die Hydrierung durchgeführt wird, z.B. durch Reduktion einer Palladiumverbindung, z.B. Palladium chlorid. Hierauf verfährt man z.B. so, dass die zu hydro- genolysierende Verbindung dem Gemisch zugesetzt und Wasserstoff eingeleitet wird. Als Lösungsmittel verwen det man vorzugsweise Niederalkanole, z.B. Methanol, Äthanol oder Butanol. Ferner kann man dazu auch Äther, z.B. Dimethyläther, Tetrahydrofuran oder Dime- thoxyäthan, oder Säuren, z.B. Essigsäure, verwenden.
Die Reaktion wird vorzugsweise bei Raumtemperatur und bei Atmosphärendruck oder leicht erhöhtem Druck durchgeführt. Gewöhnlich ist die Umsetzung nach 10 15 Minuten oder 1 Stunde beendet. Hierauf verfährt man beispielsweise wie folgt: Man filtriert den Katalysator ab und unterwirft das Filtrat einer Vakuumdestillation zur Abtreibung des Lösungsmittels. Wurde die Bildung des Katalysators, z.B. aus Palladiumchlorid, in situ vor genommen, so wurde eine saure Lösung infolge von Chlorwasserstoffbildung erhalten, so dass man als End produkt das salzsaure Aminsalz erhält. Aus diesem Grunde zieht man es vor, die Lösung mit einer Base oder unter Verwendung eines Ionenaustauscherharz vor Ab trennung des Produktes zu neutralisieren. Hierauf wird das Produkt durch Abdampfen des Lösungsmittels ab getrennt und gereinigt.
Im allgemeinen erhält man das Produkt in 50 bis 65%iger Ausbeute.
Die Herstellung des Ausgangsstoffes ist im schweize rischen Patent Nr. 477 453 beschrieben.
<I>Beispiel</I> Eine Mischung von 6,8 g 1-(2',3',5'-Tri-O-benzyl-ss -D-arabinfuranosyl)-4-methoxy-5-trifluormethyl-2(1H)- -pyrimidinon in 15 ml wasserfreiem Methanol und 20 ml flüssigem Ammoniak wird während drei Tagen bei Raum temperatur in einer Bombe verschlossen stehen gelassen. Hierauf wird der Inhalt der Bombe entleert und das Am moniak verdampfen gelassen. Den Rückstand reinigt man durch fraktionierte Kristallisation aus Wasser und er hält als darin schwerer lösliches Produkt das gewünsch te 4-amino-5-cyano-2(1H)-pyrimidinon-Derivat. Das Pro dukt zeigt die charakteristische CN-Bande bei 4,5. Singu larpunkt TLC. In Lösung bleibt das leichter lösliche 4-Amino-5-Trifluormethyl-Derivat.
2 g des erhaltenen 1-(2',3',5'-Tri-O-benzyl-B-D-arabin- furanosyl)-4-amino-5-cyano-2(1H)-pyrimidinon werden in 90 ml trockenem Methanol (als Molekularsieb) gelöst und einer Suspension von 2 g vorreduziertem Palladium chlorid in 60 ml trockenem Methanol zugesetzt. Nach 3 Minuten Schütteln der Mischung bei Raumtemperatur mit Wasserstoff ist die theoretische Menge an letzterem absorbiert. Nach Abfiltrieren des Katalysators wird die saure Lösung protionenweise mit Dowex 2-X8 (ein Bikarbonat) neutralisiert und filtriert. Nach Abdampfen des Lösungsmittels unter Vakuum erfolgt eine spontane Kristallisation des Produktes. Man zerreibt hierauf mit Wasser, filtriert ab und wäscht mehrmals um verunreini gende Arabinfuranose abzutrennen.
Hierauf kristallisiert man in einer kleinstmöglichen Menge Methanol, unter Zusatz eines gleichen Volumens an Äther und Über- schuss an Hexan um, und erhält 0,72 g (66 %) an reinem 1-(B-D-Arabinfuranosyl)-4-amino-5-cyano-2(1H)-pyrimi- dinon.
