Verfahren zur Herstellung von 1-Acyl-(3-indolyl)-Verbindungen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von l-Acyl-(3-Indolyl)-Verbindungen der Formel:

Cyan, Aminomethyl, Alkyl-substituiertes Aminome thyl, Mercapto, Dialkylsulfamoyl, Alkylmercapto oder Benzylmercapto, und X CHO, CH(ORx)s, CH2OH, CH2ORx, COOH oder CN und Rx Niederalkyl, Aralkyl oder Trityl ist.
Dieses Verfahren ist dadurch gekennzeichnet, dass man ein Hydrazon der Formel:

durch Erhitzen in Gegenwart einer Mineralsäure cyclisiert.
Falls X in obiger Ausgangsverbindung die Gruppe COOH besitzt, erhält man nach Cyclisierung direkt die entsprechende a-(Indolyl)-niederaliphatische Säure. Im Falle für X andere oben angeführte Substituenten stehen, können die entsprechenden Säuren durch Hydrolyse, Hydrierung oder Oxydation des betreffenden X-Substituenten erhalten werden.
Die nach dem erfindungsgemässen Verfahren hergestellten a-[(l -Aroyl- oder l-Heteroaroyl)-3 -indolyl]- -aliphatischen Säure-Verbindungen und deren entsprechende Ester und Amide weisen eine hochgradige entzündungswidrige Wirkung auf und sind bei der Verhütung und Verzögerung von Granulomgewebebildung wirksam. Gewisse dieser Verbindungen weisen diese Wirkung in hohem Masse auf und sind bei der Behandlung von arthritischen und dermatologischen Störungen und ähnlichen Zuständen wertvoll, die auf eine Behandlung mit entzündungshemmenden Mitteln ansprechen.
Ferner weisen die nach dem erfindungsgemässen Verfahren hergestellten Verbindungen einen nützlichen Grad von antipyretischer Wirkung auf. Zu diesem Zweck werden sie normalerweise oral in Tabletten- oder Kapselform verabreicht, wobei die optimale Dosierung natürlich von der jeweilig verwendeten Verbindung und dem Typus und der Schwere der behandelten Affektion abhängt. Obwohl die optimalen Mengen der nach dem erfindungsgemässen Verfahren hergestellten Verbindungen, die auf eine solche Weise verwendet werden sollen, von der verwendeten Verbindung und vom jeweiligen Typus der behandelten Affektion abhängen, sind orale Dosierungen von bevorzugten Verbindungen im Bereich vom 1,0 bis 200 mg pro Tag zur Bekämpfung arthritischer Zustände nützlich, je nach der Wirkung der jeweiligen Verbindung und der Reaktionsempfindlichkeit des Patienten.
Die folgenden Beispiele veranschaulichen das erfindungsgemässe Verfahren, unter Anführung der Herstellung einiger Ausgangssubstanzen. Ein Schutz wird für die Herstellung dieser nicht begehrt.
Beispiel I
A. Nl-(p-Chlorbenzoyl)-p-methoxyphenylhydrazin
In 3,86 g p-Methoxyphenylhydrazin-hydrochlorid in 50 ml Toluol werden 6,0 ml Triäthylamin gegeben. Das Reaktionsgemisch wird 10 Minuten auf 750 C und dann eine weitere Stunde auf 780 C erhitzt. Es wird dann auf - 90 C abgekühlt, worauf 3,68 g p-Chlorbenzoylchlorid beigefügt werden. Man lässt das Reaktionsgemisch sich unter Rühren bis zu 200 C erwärmen. Es wird dann 20 Minuten auf 750 C erhitzt. Das Reaktionsgemisch wird dann abgekühlt und filtriert und der Kuchen mit Toluol gewaschen. Das Toluolfiltrat wird im Vakuum konzentriert und der Rückstand in 200 ml Äther gelöst. Dieser Ätherlösung wird dann Chlorwasserstoff beigefügt.
Das Gemisch wird filtriert und der Kuchen zweimal mit je 10 ml Äther gewaschen und schliesslich bei 400 C getrocknet; man erhält N-(p-Chlorbenzoyl)-p-methoxyphe nyl-hydrazin-hydrochlorid; F = 179-1800 G.
B. Ein Gemisch von 16,8 g des Dimethylacetats von Lävulinaldehyd, 31,0 g N1-p-Chlorbenzoyl-p-methoxyphenylhydrazin-HCl und 120 ml t-Butanol wird 5 Stunden unter Stickstoff bei Rückfluss erhitzt. Das Produkt wird durch Beigabe von 120 ml Wasser und dann Abkühlung auf 0,50 C gefällt. Das Produkt wird filtriert und mit 20 ml 50% dem wässrigem t-Butanol und 35 ml Wasser gewaschen. Nach Trocknen im Vakuum bei 45-500 C wird 1 -p-Chlorbenzoyl-2-methyl-5-methoxy -indolyl-ace- taldehyd-dimethylacetal erzielt.
Beispiel 2
Ein Gemisch von 30 g N1-p-Chlorbenzoyl-p-methoxy- -phenylhydrazin-HCl (Beispiel 1A), 39 g des Trityläthers von 4-Ketopentanol und 120 ml t-Butanol wird 5 Stunden unter Stickstoff bei Rückfluss erhitzt. Durch Beigabe von 120 ml Wasser und Abkühlung auf 0,50 C wird das Produkt gefällt, das dann filtriert und mit 20 ml 50% dem wässrigem t-Butanol und danno mit 35 ml Wasser gewaschen wird. Durch Trocknen im Vakuum bei 450 C erhält man den Trityl- 1 -p-chlorbenzoyl-2-methyl-5-me- thoxyindolyl-3-äthyläther. Beim obigen Verfahren kann anstelle von t-Butanol Eisessig als Lösungsmittel verwendet werden.
Beispiel 3
Ein Gemisch von 19,2 g Benzyläther von 4-Ketopentanol, 30 g N1-p-Chlorbenzoyl-p-methoxyphenylhydrazin -HCl (Beispiel 1A) und 120 ml t-Butanol wird 5 Stunden unter Stickstoff bei Rückfluss erhitzt. Durch Beigabe von 120 ml Wasser und Abkühlung wird der Benzylindolyl äthyläther gefällt, der filtriert und mit 20 ml 50% dem wässrigem t-Butanol und mit 35 ml Wasser gewaschen wird. Durch Trocknen im Vakuum bei 500 C erhält man den Benzyl-1-p-chlorbenzoyl-2-methyl-5-methoxyindolyl -3-äthyläther. Beim obigen Verfahren kann anstelle von t-Butanol Eisessig verwendet werden.
Beispiel 4
A. 1-p-Chlorbenzoyl-2-methyl-5-methoxy-3-indolyl- acetaldehyd
3,0 g rohes l-p-Chlorbenzoyl-2-methyl-5-methoxy- indol-3-acetaldehyddiäthylacetal werden 4 Stunden bei 250 C mit 60 ml eines 1 : 1 Gemisches von Wasser und Alkohol und 3 ml konzentrierter Salzsäure gerührt, wobei man das entsprechende freie Aldehyd erhält, das durch Verdünnung mit 300 ml Wasser und Extraktion mit Äther wird. Nach Entfernen des Lösungsmittels bleibt das Indolyl-3 -acetaldehyd-Produkt im Rückstand zurück.
B. In eine Lösung von 2,82 1 -p-Chlorbenzoyl-2- -methyl-5-methoxy-indol-3-acetaldehyd in 10 ml Benzol werden 2,4 g Silberoxyd in Anteilen beigefügt, wobei die Temperatur durch äussere Kühlung bei 35-400 C gehalten wird. Nach 3stündigem Stehen wird das Silbersalz von Verbindung VI durch Filtrieren gesammelt und im Vakuum getrocknet. Die freie Säure wird durch Beigabe von Essigsäure aus der wässrigen Lösung des Silbersalzes befreit. Das rohe Produkt wird durch Kristallisation aus t-Butanol gereinigt; es ist mit einer authentischen Probe von 1-p-Chlorbenzoyl-2-methyl-5-methoxy-indol-3-essig- säure identisch.
C. In eine Lösung von 2,82 g l-p-Chlorbenzoyl-2- -methyl-5-methoxyindol-3-acetaldehyd in 5 ml wasserfreiem Dioxan wird 0,5 g hydriertes Platin auf Holzkohle gegeben. Das Gemisch wird auf 400 C erhitzt und etwa 2 Stunden in einer Sauerstoffatmosphäre geschüttelt, bis die theoretische Menge Sauerstoff absorbiert ist. Nach dem Filtrieren wird der Katalysator im Vakuum entfernt und der Rückstand aus t-Butanol umkristallisiert.
Das Produkt weist eine Temperatur von 1650 C auf, die durch eine Probe von 1 -p-Chlorbenzoyl-2-methyl-5- -methoxyindol-3-essigsäure nicht herabgesetzt wird. Die Infrarot- und Ultraviolettspektren sind mit denjenigen einer authentischen Probe identisch.
Beispiel 5
A. Eine Lösung von 2,0 g g 1-p-Chlorbenzoyl-2-me- thyl -5- methoxy-indol -3 äthanolbenzyläther in 50 ml Äthylacetat wird in Gegenwart von 0,4 g eines aus Palladium auf Holzkohle bestehenden Katalysators unter einer Wasserstoffatmosphäre hydriert. Die theoretische Menge Wasserstoff wird in 30 Minuten aufgenommen, und nach Entfernen wird der Katalysator im Vakuum verdampft. Der Rückstand wird aus Isopropanol umkristallisiert. Das Produkt weist ein Infrarotspektrum auf, das mit demjenigen von 1-p-Chlorbenzoyl-2-methyl-5 -methoxy-indol-3-äthanol identisch ist.
B. Ein Gemisch von 5,0 g g 1-p-Chlorbenzoyl-2-me- thyl-5-methoxy-indol-3-äthanoltrityläther und 1,0 g eines aus Palladium auf Holzkohle bestehenden Katalysators wird bei Raumtemperatur unter einem Druck von etwas mehr als einer Atmosphäre mit Wasserstoff geschüttelt.
Nach Aufnahme der berechneten Menge Wasserstoff in etwa 3 Stunden wird der Katalysator abfiltriert und das Lösungsmittel im Vakuum entfernt. Der Rückstand wird aus Isopropanol kristallisiert. Das Produkt ist mit Standardproben von l-p-Chlorbenzoyl-2-methyl-5-methoxy- -indol-3 -äthanol identisch.
C. Eine Lösung von 2,0 0 1 -p-Chlorbenzoyl-2-me- thyl-5-methoxyindol-3-äthanol-t-butyläther, der durch Verwendung des t-Butyläthers von 4-Ketopentanol im Verfahren von Beispiel 2 oder von Natrium-t-butoxyd im Verfahren von Beispiel 4B erzeugt wird, in 8 ml wasserfreier Trifluoressigsäure wird 1 Stunde bei 100 C stehen gelassen. Die Lösung wird in 30 ml eiskaltes Wasser gegossen und das Gemisch mit Chloroform extrahiert.
Die Chloroformextrakte werden mit Wasser, mit gesättigter NatriumbicarbonatIösung und schliesslich wiederum mit Wasser gewaschen. Nach Trocknen über Natriumsulfat wird das Lösungsmittel im Vakuum entfernt und der Rückstand aus Isopropanol umkristallisiert. Das Produkt weist ein Infrarotspektrum auf, das mit demjenigen von l-p-Chlorbenzoyl-2-methyl-5-methoxy-indol- -3-äthanol identisch ist.
D. Trockener Chlorwasserstoff wird in eine mit kaltem Wasser abgekühlte Lösung von 5,0 g l-p-Chlorben- zoyl-2-methyl-5 - methoxyindol -3-äthanol - trityläther in 50 ml Cyclohexan eingeleitet. Nach 3stündigem Stehen bei 250 C werden 250 ml Chloroform beigefügt. Die Lösung wird mit Wasser, mit gesättigter Natriumbicarbonatlösung und wiederum mit Wasser gewaschen und übcr Natriumsulfat getrocknet und däs Lösungsmittel' im Vakuum abdestilliert. Der Rückstand wird in 100 ml warmen Petroläther aufgenommen. Beim Abkühlen auf 100 C wird das kristallisierte Butanol durch Filtrieren isoliert. Petroläther wird im Vakuum entfernt und der Rückstand aus Isopropanol kristallisiert.
Das erzielte Produkt weist ein Infrarotspektrum auf, das mit demjenigen von 1 -p-Chlorbenzoyl-2-methyl-5-methoxy-indol -äthanol identisch ist.
E. In eine gerührte Lösung von 28,4 g 1-p-Chlor benzoyl-2-methyl-5-methoxyindol-3-äthanol in 100 ml Essigsäure werden im Zeitraum von 1,5 Stunden bei 50 C 80 ml Chromsäurelösung gegeben, die dadurch hergestellt wird, dass 20,1 g Chromsäure in 20 ml Wasser gelöst und dann mit Essigsäure auf 80 ml verdünnt werden.
Die Lösung wird über Nacht bei Raumtemperatur stehen gelassen, worauf 300 ml Wasser beigefügt werden und der gefällte Feststoff durch Filtrieren gesammelt und durch Kristallisation aus 5-Butanol gereinigt wird. Das erzielte Produkt weist ein Infrarotspektrum auf, das mit demjenigen von 1 -p-Chlorbenzoyl-2-methyl-5-methoxy -indol-3-essigsäure identisch ist.
Beispiel 6
A. Eine Lösung von 46 g N1-p-Chlorbenzoyl-p-methoxy-phenylhydrazin-hydrochlorid (Beispiel 1A) und 20 g Lävulinonitril in 250 ml 2N äthanolischer HCI wird einige Minuten auf einem Dampfbad erhitzt. Es erfolgt eine exotherme Reaktion, bei der sich NH4Cl abscheidet.
Man lässt das Gemisch ohne äusseres Erhitzen bei gelindem Rückfluss sieden, bis die exotherme Reaktion aufhört. Das Gemisch wird dann 30 Minuten bei Rückfluss erhitzt und im Vakuum auf etwa 80 ml eingeengt. Nach Verdünnen mit 400 ml Wasser wird es mit Äther extrahiert. Der Extrakt wird mit gesättigter NaHCO3-Lösung und dann mit Wasser gewaschen und getrocknet. Die getrocknete Lösung wird filtriert und zu einem dunklen Sirup konzentriert, der durch Chromatographie auf mit Säure gewaschenem Aluminiumoxyd gereinigt wird, wobei mit Gemischen von Äther und Petroläther in einem fortschreitenden Verhältnis von 1 : 9 bis 1:1 eluiert wird. Das auf diese Weise erzielte Produkt ist rohes l-p -Chlorbenzoyl-2-methyl-5-methoxyindol-3 -acetonitril.
B. Das verwendete Nitrilase-Enzym wird mit 0,1 M Phosphatpuffer, pH 7,5, aus den Blättern von Hafer (Avena), Gerste (Hondeum), der Brassica-Familie und gewissen Musaceoe extrahiert. Pro 100 ml Puffer werden 10 bis 50 g dieses Materials extrahiert.
1 -p.Chlorbenzoyl-2-methyl-5-methoxy-3-indolylaceto- nitril wird in einer solchen Menge Methanol gelöst, dass die Endkonzentration von Methanol 10 bis 30% des Enzym-Substrat-Gemisches beträgt. Die Endkonzentration des Substrats wird zwischen 0,1 und 10% gehalten.
Die Hydrolyse wird 1 bis 6 Stunden bei 25-370 C durchgeführt. Der pH wird durch Beigabe von verdünntem Alkali bei 7,0-8,0 gehalten. Am Ende dieser Zeit wird das Gemisch mit verdünnter Salzsäure bis zu pH 1-2 angesäuert und mit Chloroform extrahiert. Die Chloroformextrakte werden mit Wasser gewaschen und im Vakuum zur Trockene konzentriert, wobei man als Rückstand rohe 1 -p-Chlorbenzoyl-2-methyl-5-methoxy-3 -indolyles- sigsäure erhält.
Beispiel 7
A. p-Chlorbenzimidochlorid
Eine Lösung von 13,7 g p-Chlorbenzonitril in 100 ml wasserfreiem Äther wird bei 10-200 C gehalten, während 4-4,5 g wasserfreier Chlorwasserstoff beigefügt wfrd Das gefällte p-Chlorbenzimidochlorid wird in einer trockenen Atmosphäre filtriert und ohne weitere Behandlung in der nächsten Stufe verwendet.
B. Nl-(p-Chlorbenzimidoyl)-p-methoxyphenylhydra- zin-hydrochlorid
In 3,86 g p-Methoxyphenylhydrazin-hydrochlorid in 50 ml Benzol werden 10 ml trockenes Triäthylamin gegeben. Dem erzielten Gemisch werden dann 1,8 g p-Chlorbenzimidochlorid aus Teil A beigefügt, und das Reaktionsgemisch wird eine Stunde auf 78-800C erhitzt, worauf es abgekühlt und filtriert wird. Der Kuchen wird mit Benzol gewaschen. Das Filtrat wird im Vakuum konzentriert und der Rückstand in 50 ml Äther gelöst.
Wasserfreier Chlorwasserstoff wird in die Lösung eingeleitet und das gefällte Hydrochlorid von Nl-(p-Chlorbenz- iminoyl)-p-methoxyphenylhydrazin durch Filtrieren entfernt, mit Äther gewaschen und im Vakuum getrocknet.
C. 1 -p-Chlorbenzoyl-2-methyl-5-methoxyindolyl3 -essigsäure
Ein Gemisch von 29,9 g N1-(p-Chlorbenziminoyl) -p-methoxyphenylhydrazin-hydrochlorid, 11,6 g Lävulinsäure und 120 ml 3,7 g wasserfreie Salzsäure enthaltendem Dioxan oder 120 ml Essigsäure wird unter sorgfältigem Rühren 6 Stunden unter Stickstoff auf 85-900 C erhitzt. Das Reaktionsgemisch wird auf Raumtemperatur abgekühlt; 25 ml Wasser und 2 ml konzentrierte Salzsäure werden beigefügt, und das Gemisch wird 1 Stunde gerührt. Das Gemisch wird dann mit 100 ml eiskaltem Wasser verdünnt und der Niederschlag filtriert und aus t-Butanol umkristallisiert, wobei man l-p-Chlorbenzoyl- 2-methyl-5-methoxy-3-indolyl-essigsäure, F = 160-161 0C, erhält.

Example I.

6.0 ml of triethylamine are added to 3.86 g of p-methoxyphenylhydrazine hydrochloride in 50 ml of toluene. Example 3
A mixture of 19.2 g of benzyl ether of 4-ketopentanol, 30 g of N1-p-chlorobenzoyl-p-methoxyphenylhydrazine-HCl (Example 1A) and 120 ml of t-butanol is refluxed under nitrogen for 5 hours. The benzylindolyl ethyl ether is precipitated by adding 120 ml of water and cooling, which is filtered and washed with 20 ml of 50% aqueous t-butanol and with 35 ml of water. By drying in vacuo at 500 ° C., benzyl 1-p-chlorobenzoyl-2-methyl-5-methoxyindolyl -3-ethyl ether is obtained. In the above process, glacial acetic acid can be used in place of t-butanol.
Example 4
A. 1-p-Chlorobenzoyl-2-methyl-5-methoxy-3-indolyl-acetaldehyde
3.0 g of crude lp-chlorobenzoyl-2-methyl-5-methoxy-indole-3-acetaldehyde diethylacetal are stirred for 4 hours at 250 ° C. with 60 ml of a 1: 1 mixture of water and alcohol and 3 ml of concentrated hydrochloric acid, the corresponding free aldehyde is obtained, which is obtained by dilution with 300 ml of water and extraction with ether. After removing the solvent, the indolyl-3-acetaldehyde product remains in the residue.

C. In a solution of 2.82 g of l-p-chlorobenzoyl-2-methyl-5-methoxyindole-3-acetaldehyde in 5 ml of anhydrous dioxane, 0.5 g of hydrogenated platinum on charcoal is added. The mixture is heated to 400 ° C. and shaken in an oxygen atmosphere for about 2 hours until the theoretical amount of oxygen has been absorbed. After filtration, the catalyst is removed in vacuo and the residue is recrystallized from t-butanol.
The product has a temperature of 1650 C, which is not reduced by a sample of 1-p-chlorobenzoyl-2-methyl-5-methoxyindole-3-acetic acid. The infrared and ultraviolet spectra are identical to those of an authentic sample.
Example 5
A. A solution of 2.0 g 1-p-chlorobenzoyl-2-methyl -5-methoxy-indole -3 ethanol benzyl ether in 50 ml of ethyl acetate is in the presence of 0.4 g of a catalyst consisting of palladium on charcoal under a Hydrogenated hydrogen atmosphere. The theoretical amount of hydrogen is taken up in 30 minutes and, after removal, the catalyst is evaporated in vacuo. The residue is recrystallized from isopropanol. The product has an infrared spectrum which is identical to that of 1-p-chlorobenzoyl-2-methyl-5-methoxy-indole-3-ethanol.
B. A mixture of 5.0 g of 1-p-chlorobenzoyl-2-methyl-5-methoxy-indole-3-ethanol trityl ether and 1.0 g of a catalyst consisting of palladium on charcoal is at room temperature under a pressure of something shaken more than one atmosphere of hydrogen.
After the calculated amount of hydrogen has been absorbed in about 3 hours, the catalyst is filtered off and the solvent is removed in vacuo. The residue is crystallized from isopropanol. The product is identical to standard samples of 1-p-chlorobenzoyl-2-methyl-5-methoxy-indole-3-ethanol.
C. A solution of 2.0 0 1 -p-chlorobenzoyl-2-methyl-5-methoxyindole-3-ethanol-t-butyl ether obtained by using the t-butyl ether of 4-ketopentanol in the method of Example 2 or of sodium t-butoxide is generated in the process of Example 4B, in 8 ml of anhydrous trifluoroacetic acid is left to stand at 100 C for 1 hour. The solution is poured into 30 ml of ice-cold water and the mixture is extracted with chloroform.
The chloroform extracts are washed with water, with saturated sodium bicarbonate solution and finally again with water. After drying over sodium sulfate, the solvent is removed in vacuo and the residue is recrystallized from isopropanol. The product has an infrared spectrum which is identical to that of l-p-chlorobenzoyl-2-methyl-5-methoxy-indole-3-ethanol.
D. Dry hydrogen chloride is passed into a solution, cooled with cold water, of 5.0 g of l-p-chlorobenzoyl-2-methyl-5-methoxyindole -3-ethanol - trityl ether in 50 ml of cyclohexane. After standing for 3 hours at 250 ° C., 250 ml of chloroform are added. The solution is washed with water, with saturated sodium bicarbonate solution and again with water and dried over sodium sulfate and the solvent is distilled off in vacuo. The residue is taken up in 100 ml of warm petroleum ether. On cooling to 100 ° C., the crystallized butanol is isolated by filtration. Petroleum ether is removed in vacuo and the residue is crystallized from isopropanol.
The product obtained has an infrared spectrum which is identical to that of 1-p-chlorobenzoyl-2-methyl-5-methoxy-indole-ethanol.
E. In a stirred solution of 28.4 g of 1-p-chloro benzoyl-2-methyl-5-methoxyindole-3-ethanol in 100 ml of acetic acid, 80 ml of chromic acid solution are added over a period of 1.5 hours at 50 C. is prepared by dissolving 20.1 g of chromic acid in 20 ml of water and then diluting it to 80 ml with acetic acid.
The solution is left to stand overnight at room temperature, whereupon 300 ml of water are added and the precipitated solid is collected by filtration and purified by crystallization from 5-butanol. The product obtained has an infrared spectrum which is identical to that of 1-p-chlorobenzoyl-2-methyl-5-methoxy-indole-3-acetic acid.
Example 6
A. A solution of 46 g of N1-p-chlorobenzoyl-p-methoxyphenylhydrazine hydrochloride (Example 1A) and 20 g of levulinonitrile in 250 ml of 2N ethanolic HCl is heated for a few minutes on a steam bath. An exothermic reaction takes place in which NH4Cl separates out.
The mixture is allowed to boil at gentle reflux without external heating until the exothermic reaction ceases. The mixture is then refluxed for 30 minutes and concentrated in vacuo to about 80 ml. After diluting with 400 ml of water, it is extracted with ether. The extract is washed with saturated NaHCO3 solution and then with water and dried. The dried solution is filtered and concentrated to a dark syrup which is purified by chromatography on acid washed alumina, eluting with mixtures of ether and petroleum ether in a progressive ratio of 1: 9 to 1: 1. The product obtained in this way is crude 1-p -chlorobenzoyl-2-methyl-5-methoxyindole-3-acetonitrile.
B. The nitrilase enzyme used is extracted with 0.1 M phosphate buffer, pH 7.5, from the leaves of oats (Avena), barley (Hondeum), the Brassica family and certain Musaceoe. 10 to 50 g of this material are extracted per 100 ml of buffer.
1 -p.Chlorbenzoyl-2-methyl-5-methoxy-3-indolylacetonitrile is dissolved in such an amount of methanol that the final concentration of methanol is 10 to 30% of the enzyme-substrate mixture. The final concentration of the substrate is kept between 0.1 and 10%.
The hydrolysis is carried out at 25-370 ° C. for 1 to 6 hours. The pH is kept at 7.0-8.0 by adding dilute alkali. At the end of this time, the mixture is acidified to pH 1-2 with dilute hydrochloric acid and extracted with chloroform. The chloroform extracts are washed with water and concentrated to dryness in vacuo, crude 1-p-chlorobenzoyl-2-methyl-5-methoxy-3-indolyl acetic acid being obtained as residue.
Example 7
A. p-Chlorobenzimidochloride
A solution of 13.7 g of p-chlorobenzonitrile in 100 ml of anhydrous ether is kept at 10-200 ° C., while 4-4.5 g of anhydrous hydrogen chloride are added. The precipitated p-chlorobenzimidochloride is filtered in a dry atmosphere and in used in the next stage.
B. Nl- (p-chlorobenzimidoyl) -p-methoxyphenylhydrazine hydrochloride
10 ml of dry triethylamine are added to 3.86 g of p-methoxyphenylhydrazine hydrochloride in 50 ml of benzene. 1.8 g of p-chlorobenzimidochloride from Part A are then added to the resulting mixture and the reaction mixture is heated to 78-80 ° C. for one hour, after which it is cooled and filtered. The cake is washed with benzene. The filtrate is concentrated in vacuo and the residue is dissolved in 50 ml of ether.
Anhydrous hydrogen chloride is passed into the solution and the precipitated hydrochloride of Nl- (p-chlorobenziminoyl) -p-methoxyphenylhydrazine is removed by filtration, washed with ether and dried in vacuo.
C. 1 -p-Chlorobenzoyl-2-methyl-5-methoxyindolyl-3-acetic acid
A mixture of 29.9 g of N1- (p-chlorobenziminoyl) -p-methoxyphenylhydrazine hydrochloride, 11.6 g of levulinic acid and 120 ml of dioxane containing 3.7 g of anhydrous hydrochloric acid or 120 ml of acetic acid is stirred under nitrogen for 6 hours Heated to 85-900 C. The reaction mixture is cooled to room temperature; 25 ml of water and 2 ml of concentrated hydrochloric acid are added and the mixture is stirred for 1 hour. The mixture is then diluted with 100 ml of ice-cold water and the precipitate is filtered and recrystallized from t-butanol, l-p-chlorobenzoyl-2-methyl-5-methoxy-3-indolyl-acetic acid, melting point 160-161 ° C., being obtained.
