Verfahren zur Herstellung von a- (3-Indolyl)-niederaliphatischen Säuren oder Estern
Die vorliegende Erfindung bezieht-sich auf neuartige Verfahren zur Herstellung von a- (3-indolyl)-niederali- phatischon Säuren oder Estern, die ein an das Stickstoff- atom des Indolringes gebundenes aromatisches Carbon säureacyl- (das heisst ein Aroyl- oder Heteroaroyl)-Radikal mit weniger als 3 kondensierten Ringen aufweisen.
Diese N-1-Aroyl- und N-1-heteroaroylindolyl-ali phatischem Säureverbidungen) lassen sich chemisch durch folgende Strukturformel darstellen :

Der erwähnte funktionellc substitutent kann auch ein Mercapto-oder ein substituierter Mercaptorest vom durch Alkylthiogruppen, wie z. B. Methylthio, Äthylthio oder Propylthio und Arylthio- oder Aralkyl thiogruppen, wie z. B. Benzylthio oder phenylthio, ver anschaulichten Typus sein. Wenn etwünscht, kann das N-1-Aroyl-Radikal z. B. mit einem Trifluormethyl-, Trifluoräthyl-, Perfluoräthyl-, ss-Chloräthyl- oder einem ähnlichen Substituenten halogenalkyliert oder mit einer Acetyl-, Propionyl-, Benzoyl-, Phenylacetyl-, Trifluoracetyl- oder einer ähnlichen Acylgruppe acyliert sein, oder es kann einen Halogenalkoxy- oder Halogenalkylthio-Substituenten enthalten.
Darüber hinaus umfa#t die Erfindung Verbindungen, bei denen der Aroylrest einen Sulfamyl-, Benzylthiomethyl-, Cyan-, Sulfonamido- oder Dialkylsulfionamidorest aufweist. Er kann auch einen Carboxy-Substituentea oder ein Derivat da-vont wie z. B. ein Alkalimetallsalz oder einen Niederalkylester des Carboxyrestes, ein Aldehyd, Azid, Amid, Hydrazid und dergleichen oder ein Aldehydderivat vom Typus der Acetale oder Thioacetale enthalten.
Bei den bevor zugten Verbindungen ist das N-1-Aroyl-Radikal Benzoyl und der funktionelle Substituent, der sich in para-Stellung des 6gliedmgen Ringes befindet, Halogen, Niederalkoxy, Niederalkylthio oder Diniederalkylamino.
R2, das sich in Stellung 2 des Indolringkernes befindet, kann Wasserstoff sein, doch wird es vorgezogen, dass sich in dieser Stellung des Moleküls ein weniger als 9 Kohlenstoffatome aufweisender Kohlenwasserstoff- rest befindet. Am besten geeignet sind Niederalkylgrup- pen, wie z. B. Methyl, Äthyl, Propyl oder Butyl, obwohl Aryl-, Alkaryl- und @ Aralkylgruppen auch vorteilhaft sind. Im übrigen liegen auch die Alkoxy-, Halogen-, Amino-, substituierten Amino und nitrosubstituierten Derivate der obigen Gruppen innerhalb des Rahmens dieser Erfindung, wie auch Indole, die in der Stellung 2 einen ungesättigten aliphatischen Rest, wie z. B.
Allyl oder Vinyl oder einen cyclischen aliphatischen Rest vom Typus von Cyclohexyl,, aufweisen.
Der saure Teil der nach dem erfindungsgemässen Verfahren erzeugten N-1-acylierten a- (3-indolyl)-alipha- tischen Säuren ist vorzugsweise eine niederaliphatische Säure, wie z. B. Essigsäure, Propionsäure, Buttersäure, Valeriansäure, 3-Butencarbonsäure, 4-Pentencarbonsäure und dergleichen. Demnach kann R3 in der obigen Formel ein Wasserstoffatom oder ein Niederalkylrest, wie z. B. Methyl, Äthyl, Propyl und dergleichen, sein.
Die entsprechenden Indole, bei denen die Seitenkette in Stellung 3 statt mit einer Carboxygruppe mit einer Estr-, Aldehyd-, Acetal-, Alkohol oder Athergruppe enden kann, sind in gewissen Fällen wichtige Zwischenverbindungen bei der Synthese der freien Säuren und in vielen FäHen an und für sich schon wichtige Endprodukte. Neben den hierin beschriebenen a-(3-Indolyl)essigsäuren sind deren Ester, Salze und Amidderivate ein zusätzlicher Aspekt der vorliegenden Erfindung.
Die Synthese verschiedener nach dem erfindungsge- mässen Verfahren hergestellter Verbindungen, die am Indolringsystem einen 5-Substituenten aufweisen, der ein an den homocyclischen Ring des Indols gebundenes Stickstoffatom hat, beruht im allgemeinen, auf der 5-Nitroverbindung, die e später in den gewünschten 5-Substituenten umgewandelt werden kann. Eine solche Umwandlung kann nach verschiedenen Verfahren durchgeführt werden. Durch Reduktion der 5-Nitrogruppen erhält man eine 5-Aminogruppe. Durch Umsetzung des Aminos mit Alkylhalogeniden erhält man Mono-und Dialkylamingruppen.
Ist das Alkylhalogenid eine Di halogenalkylengruppe (z. B. 1, 4-Dibrombutan), so wird ein heterocyclischer Ring (z. B. Pyrrolidino) gebildet.
Ähnlich ergibt bis-(ss-Chloräthyl)-äther eine N-Morpho linverbindung. Die Alkylierung kann auch gleichzeitig mit der Reduktion, z. B. mit Formaldehyd und Raney- Nickel und Wasserstoff, durchgeführt werden. Die Acylie rung kann in ähnlicher Weise bei den 5-Aminoverbin- dungen oder am 5-Nitro (bei gleichzeitiger Reduktion) ausgeführt werden, wobei man 5-Acylamidoverbindunn- gen erhält Die 5-Aminogruppe kann zur Bildung von 5-Ureidoverbindungen mit Isocyanaten umgesetzt werden.
Die nach dem erfindungsgemässen Verfahren hergestellten a- [l-aroyl- oder 1-heteroaroyl)-3-indolyl]- aliphatischen Säureverbindungen und deren entsprechende Ester und Amide weisen eine hochgradige ent- zündungswidrige Wirkung auf und sind bei der Ver hütung und Verzögerung von Granulomgewebebildung wirksam. Gewisse dieser Verbindungen weisen diese Wirkung in hohem Masse auf und sind bei der Behand- lung von arthritischen und dermatologischen Störungen und ähnlichen Zuständen wertvoll, die auf eine Behandlung mit entzündungshemmenden Mritteln ansprechen.
Ferner weisen die nach dem erfindungsgemässen Verfahren hergestellten Verbindungen einen nützlichen Grad von antipyretischer Wirkung auf. Zu diesem Zweck werden sie normalerweise oral in Tabletten-oder Kapselform verabreicht, wobei die optimale Dosierung natür- lich von der jeweilig verwendeten Verbindung und dem Typus und der Schwere der behandelten Affektion abhängt.
Obwohl die optimalen Mengen der nach dem erfindungsgemässen Verfahren hergestellten Verbindungen, die auf eine solche Weise verwendet werden sollen, von der verwendeten Verbindung und vom jeweiligen Typus der behandelten Affektion abhängen, sind orale Dosierungen von bevorzugten Verbindungen im Bereich von 1, 0 bis 200 mg pro Tag zur Bekämpfung arthnti- scher Zustände nützlich, je nach der Wirkung der jeweiligen Verbindung und der Reaktionsempfindlichkeit des Patiente.
Gemäss der vorliegenden Erfindung können die oben erwähnten a-(3-indolyl)-niederaliphatischen Säuren, die in der Stellung N-1 des Indolkernes mit einem weniger als 3 kondensierte Ringe aufweisenden Aroyl-oder Heteroaroylrest acyliert sind, nach verschiedenen Ver- fahren hergestellt werden. Das allgemeine Verfahren im Rahmen der vorliegenden Erfindung ist eine Synthese der Produkte durch Bildung des Indolringsystems nach Einsetzung der l-Acylgruppe.
Das erfindungsgemä#e Verfahren oben definierter Verbindung ist dadurch gekennzeichnet, dass man ein aroyliertes Alkylanilin der Strukturformel

in der RI, R2, R3, R und Rx wie oben definiert sind, durch Erhitzen mit einer Lewisschen Säure zyklisiert.
Im Falle Rr Wasserstoff ist, erhält man direkt die Säure ; und falls Rx Trityl ist, kann das Produkt zur Säure hydtolysiert werden.
Die Einsetzung der 1-Acylgruppe erfolgt somit im allgemeinen in einen Vorläufer. Die gewünschte Acylgruppe kann auf das Stickstoffatom eines zweckmässig substituiertem Anilins oder phenylhydrazins eingesetzt werden und der Ringschluss dieser Verbindungen erfolgt erfindungsgemäss bei bereits eingesetzter Acylgruppe zur Bildung g des Indolringsystems. Ist die 3-Seitenkette des erfindungsgemäss gebildeten Indols keine Niederalkan- carbonsäure-Seitenkette, sondern eine andere funktio nelle Gruppe, die durch Hydrolyse und/oder Oxydierung in eine solche Alkancarbonsäuregruppe umgewandelt, so kann diese Stufe dann durchgeführt werden.
Das erfindungsgemässe Verfahren stellt, infolge der durch die Acyliemng nach der Bildung des Indolringes gestellten Probleme, einen Fortschritt in der Herstellung solcher Indolylsäuren dar.
Die Herstellung der Ausgangsverbindung geht vorzugweise von einem para-substituierten Anilin aus und umfasst die Einführung von zwei verschiedenen Gruppen am Stickstoffatom des para-substituierten Anilins, einer Acylgruppe, die zum 1-Acyl wird, und einer ketosubsti- tuierten Alkyl-Seitenkette, die in oder ohne Gegenwart eines Ester-Substituenten eingeführt wird.
Aus dieser y-N-acyl-para-substituierten Acylketon- Ausgangsverbindung entsteht erfindungsgemäss durch Ringschluss ein Indol, indem es durch Erhitzen mit einer Lewisschen Säure, wie z. B. Zinkchlorid oder wasserfreiem Fluorwasserstoff, zur Bildung des Indolesters, erhitzt wird. Falls der Triphenylmethylester erhalten wird, kann dieser Ester entweder durch Er hitzen oder mit einer alkoholischen Suspension einer Esterese in die freien Säuren umgewandelt werden.
Bei Verwendung des Triphenylmethylesters beim Ring schlu#verfahren können die Hitze und die Säurekatalyse die Entfernung von mindestens einem Teil der Estergruppe bewirken, wodurch man die freie Säure unmittel- bar erhält. Dieses Entfernen der Estergruppe kann auch vor dem Ringschlu# oder nach dem Ringschlu# erfolgen.
Im folgenden Beispiel wird das erfindungsgemä#e henstellungsverfahren sowie die Herstellung der Ausgangsverbindungen erläutert; für die Herstellung der Ausgangsverbindungen wird kein Schutz begehrt.
Beispiel
Trityl-1-p-Chlorbenzoyl-2-methyl-5-alkylmercapto- indol-3-acetat
1, 5 g des öligen N-p-Chlorbenzoyl-N-p-alkylmercapto-phenyl-
3-oxo-4-aminovaleriansäure-tritylesters werden mit 200 mg wasserfreiem Zinkchlorid gründlich gemischt, worauf das Gemisch 30 Minuten auf 130 bis 150 C erhitzt wird. Nach Abkühlen auf 20 C wird es mit 25 ml Benzol verdünnt, mit 2n Salzsäure und dann mit Wasser gewaschen, getrocknet und zur Trockne konzentriert.
Der dunkle ölige Rückstand wird auf mit Säure ge- waschenem Aluminiumoxyd chromatographiert, wobei zum Eluieren ein 85 : 15-Gemisch von Petroleumäther und Äther verwendet wird ; man erhält
Trityl-1-p-chlorbenzyl-2-methyl-5-alkylmercapto indol-3-acetat.
1,0 g p-Toluosulfonsäure-monohydrat wird in 300 ml 0,03 Mol trityl-1-(p-chlorbenzoyl)-2-methyl-5-alkylmercaptoindol-3-acetat enthaltendes Benzol gegeben; das erzielte Gemisch wird 40 Minuten in einer Stickstoffatmosphäre unter Rühren bei Rückfluss erhitzt. Während dieser Zeit entwickeln sich 665 ml Isobutylen (Theorie : 672 ml). Dann wird das Reaktionsgemisch bei 55 bis 60 C mit 200 ml Benzol verdünnt und bei 60 bis 65 C mit warmem Wasser bis pH 4 bis 5 gewaschen.
Die 60 bis 65 C warme Benzollösung wird über Natriumsulfat getrocknet, durch Beigabe von 1, 0 g Darco G-60 entfärbt, heiss filtriert und auf ein Volumen von etwa 70 bis 75 ml konzentmert. Das Gemisch wird auf 10 C abgekühlt und 4 bis 5 Stunden stehengelassen. Die rohe 1 methyl-5-alkylmercaptiondol 3-essigsäure wird filtriert, zweimal mit 5 ml eines 1 : 1-Gemisches von Benzol und Pebroläther und dann zweimal mit 10 ml Petroläther gewaschen und schliesslich bei 25 C im Vakuum getrocknet. Die Ausbeute beträgt 9, 7 g (aus Benzol).

Trityl-1-p-chlorobenzyl-2-methyl-5-alkylmercapto indole-3-acetate.
1.0 g of p-toluenesulfonic acid monohydrate is added to 300 ml of benzene containing 0.03 mol of trityl-1- (p-chlorobenzoyl) -2-methyl-5-alkylmercaptoindole-3-acetate; the resulting mixture is refluxed for 40 minutes in a nitrogen atmosphere with stirring. During this time, 665 ml of isobutylene develop (theory: 672 ml). The reaction mixture is then diluted at 55 to 60 ° C. with 200 ml of benzene and washed at 60 to 65 ° C. with warm water to pH 4 to 5.
The 60 to 65 C warm benzene solution is dried over sodium sulfate, decolorized by adding 1.0 g of Darco G-60, filtered hot and concentrated to a volume of about 70 to 75 ml. The mixture is cooled to 10 ° C. and left to stand for 4 to 5 hours. The crude 1 methyl-5-alkylmercaptiondol 3-acetic acid is filtered, washed twice with 5 ml of a 1: 1 mixture of benzene and pebrol ether and then twice with 10 ml petroleum ether and finally dried at 25 ° C. in vacuo. The yield is 9.7 g (from benzene).
