Dieser Subsituent kann dementsprechend eine Hydroxygruppe oder eine verätherte Hydroxygruppe (Hydrocarbonoxygruppe), wie beispielsweise ein niedriger Alkoxy-, Aryloxy-oder Aralkoxyrest, z. B. Pyrryl-, Thiazolyl-, Thiadiazolyl-, Pyrazinyl-, Pyridyl-, Alkylpyridyl-, Pyrazolyl-, Imidazolyl-, Oxazolyl-, Pyrimidinyl- und Isoxazolylringe. Diese Heteroaroylreste können weiter in den aromatischen Ringen mit Kohlenwasserstoffgruppen oder mit funktionellen Substituenten substituiert sein.

Ein weiteres kritisches Merkmal der obigen Verbindungen besteht darin, dass die 3-Indolylessigsäuren sind, in denen das ct-Kohlenstoffatom der Essigsäure weiter durch eine halogenierte Methylenylgruppe (X = C-Y) einschliesslich der mono-und dihalogensubstituierten Methylcnylreste, wie beispielsweise Dichlormethylenyl, Fluorchlormethylenyl, Chlorbronmnthylenyl, Chlorm : thyknyl u. dgl. substituiert ist.
Bei den bevorzugten erfindungsgemäss erhältlichen Verbindungen ist R5 eine niedrige Alkyl-, niedrige Alkoxy-, Nitro-, Amino- oder substituierte Aminogruppe. Beispiele für die hier umfassten Alkyl- und Alkoxygruppen sind Methyl, Äthyl, Propyl, tert.-Butyl, Methoxy, Äthoxy, Isopropoxy u. dgl. Reste.
Beispiele für substituierte Aminoreste sind die von Alkylaminen, wie beispielsweise Methylamin, Äthyl-

R5 ist nicht auf die vorstehenden Klassen von Substituenten beschränkt, sondern kann vielmehr gewünschtenfalls Substituenten, wie beispielsweise ein Wasserstoffatom, einen Aryl-, Aryloxy-, Hydroxy- oder Mercaptorest, ein Halogenatom, einen Halogenalkylrest, wie beispiels- weise -CFa, -CHF2, u. dgl., oder eine Nitro-, Halogenalkyl-, Cyano-, Sulfamyl-, Sulfoxyd-, Aminomethyl-, substituierte Aminomethyl-, Carboxy- oder Carbalkoxygruppe, bedeuten.
Die Ester der neuen α-(3-Indolyl)-essigsäuren sind wichtige Zwischenprodukte bei der Synthese der freien Säuren und in vielen Fällen sind sie selbst als Endprodukte von Bedeutung. Zu bevorzugten Estern gehören die niedrigen Alkylester, wie beispielsweise die Methyl-, Äthyl-, Propyl-oder tert.-Butyl- verbindungen, und die Aralkylester, wie beispielsweise die Benzyl-, p-Halogenbenzyl- u. dgl. -ester mit weniger als neun Kohlenstoffatomen. Die Salze der neuen K- (l-Aroyl- oder 1-Heteroaroyl-3-indolyl)- essigsäuren können durch Behandlung der freien Säure mit der Base unter milden Bedingungen erhalten werden.
Auf diese Weise können die Salze von Alkalimetallen, wie beispielsweise Lithium, Natrium und Kalium, die Aluminium- oder Magnesiumsalze oder die Salze von Erdalkalimetallen, wie beispielsweise Barium und Kalzium, hergestellt werden. Die Salze von organischen Aminen, wie beispielsweise Alkylaminen, Morpholin, Cholin, Methylcyclohexylamin oder Glucosamin, können durch Umsetzung der Säure mit der geeigneten organischen Base erhalten werden. Die Salze von Schwermetallen, wie beispielsweise Zink und Eisen, gehören ebenfalls zum Bereich der Erfindung.

säureverbindungen und die entsprechenden Ester und Amide besitzen in hohem Grade entzündungshemmende Wirksamkeit und sind bei der Verhütung und Hemmung der Bildung von Granulationsgewebe wirksam.
Gewisse von ihnen besitzen diese Wirksamkeit in hohem Masse und sind bei der Behandlung von arthritischen Erkrankungen, Hauterkrankungen und ähnlichen Zuständen, die auf die Behandlung mit entzündungshemmenden Mitteln ansprechen, wertvoll. Zusätzlich besitzen die erfindungsgemäss erhältlichen Verbindungen nützliche antipyrtische Wirksamkeit. Für diese Zwecke werden sie normalerweise oral als Tabletten oder Kapseln verabreicht, wobei die optimale Dosierung von der jeweils verwendeten besonderen Verbindung und der Art und der Schwere der zu behandelnden Infektion abhängt.
Die oprimalen Mengen der erfindungsgemäss herstellbaren Verbindungen, die in dieser Weise verwendet werden sollen, hängen zwar von der verwendet, n Verbindung und der besonderen Art des zu behandelnden Krankheitszustandes ab, doch sind orale Dosen der bevorzugten Verbindungen im Bereich von 1, 0 bis 2000 mg je Tag bei der Kontrolle arthritischer Zustände in Abhängigkeit von der Wirksamkeit der besonderen Verbindung und der Reaktionssensibilität des Patienten brauchbar.
Die neuen fx-Halcgenmethylenyl-3-indolylessigsäuren werden, wie bereits dargelegt, aus einem in N-1-Stellung entsprechend acylierten K-Halogenmethylenyl-3-indolylessigsäure-tert.-alkylester als Ausgangsmaterial hergestellt. Das Ausgangsmaterial des erfindungsgemässen Verfahrens ist erhältlich, indem ein Oxalylhalogenid mit Indol selbst oder einem in der 2-und/oder 5-Stellung mit der gewünschten Gruppe vorsubstituierten Indol unter Bildung des entsprechenden 3-Indolglyoxylylhalogenids umgesetzt wird, das dann mit einem tert.-Alkanol zu dem entsprechenden tert.-Alkyl-3-indolglyoxalat umgesetzt wird, dessen Acylierung das entsprechende N-l-acylierte 3-Ketoesterindol liefert.

den α-Halogenmethylenyl-3-indolylessigsäuren nach an sich üblichen Arbeitsweisen für die Herstellung von Amiden und N-substituierten Amiden hergestellt werden.
So kann beispielsweise die jeweilige Säure in Gegenwart eines milden Dehydratationsmittels, wie beispielsweise Dicyclohexylcarbodiimid, in ein symmetrisches Anhydrid übergeführt und dann mit Ammoniak zur Bildung des entsprechenden Amins oder mit einem primären oder sekundären Amin, das die gewünschten Substituenten aufweist, in einem inerten Lösungsmittel zur Bildung der entsprechenden substituierten Amide behandelt werden.
Die jeweilige Säure kann aber auch in ein gemischtes Anhydrid überführt werden, indem sie mit einer Base ohne Hydroxylgruppen, beispielsweise mit einem tertiären Alkylamin, Pyridin u. dgl., unter Bildung eines Säuresalzes behandelt und anschliessend mit einem Säurehalogenid, wie beispielsweise Chlorameisen- säurealkyl-oder-arylester, Phosphoroxychlorid, Thionylchlorid u. dgl., zu dem gemischten Anhydrid umgesetzt wird, das dann mit Ammoniak, primären Aminen oder sekundären Aminen zur Bildung der entsprechenden Amide behandelt werden kann.

Die α-Halogenmethylenyl-3-indolylessigsäuren können auch zur Herstellung der entsprechenden Estei der Erfindung verwendet werden. Die jeweilige Säure kann beispielsweise zuerst in das symmetrische Anhydrid oder in gemischte Anhydride, wie oben beschrieben ist, übergeführt und dann mit dem gewünschten Alkyl- oder Aralkylalkohol in Gegenwart einer nichthydroxylischen Base, wie beispielsweise von einem tertiären Alkylamin, Pyridin u. dgl., zu dem entsprechenden Alkyl- oder Aralkylester umgesetzt werden.
Die Synthese von verschiedenen Verbindungen der Erfindung, die an dem Indolringsystem einen 5-ständigen Substituenten besitzen, der ein Stickstoffatom, gebunden an dem homocyclischen Ring des

5-ständigAlkylhalogenid ein Dihalogenalkylen (z. B. 1, 4-Dibrombutan) ist, so wird ein heterocyclischer Ring (beispielsweise ein Pyrrolidinring) gebildet. In ähnlicher Weise ergibt Bis-(ss-chloräthyl)-äther eine N-Morpholinverbindung. Die Alkylierung kann auch gleichzeitig mit einer Reduktion, beispielsweise mit Formaldehyd und Raney-Nickel und Wasserstoff, durchgeführt werden. In entsprechender Weise kann die Acylierung der 5-Aminoverbindung : n oder der 5-Nitroverbindung : n (unter gleichzeitiger Reduktion) zur Bildung der 5-Acylamidoverbindungen durchgdührt werden.
Die 5-Aminogruppe kann mit Isocyanaten zur Bildung der 5-Ureidoverbindungen umgesetzt werden.
Die folgenden Beispiele erläutern die Erfindung, ohne sie zu beschränken.
Beispiele :
Al) Herstellung von tert.-Butyl-(1-p-chlorbenzoyl-2-methyl-5-methoxy-3-indolyl)-glyoxalat.
Zu einer Lösung von 25 g 2-Methyl-5-methoxyindol in 530 ml wasserfreiem Äther werden 25 ml Oxalylchlorid zugegeben. Man lässt die Lösung 24 Stunden stehen. Das erhaltene feste 2-Methyl- ?- methoxy-3-indolglyoxylylchlorid wild gesammelt, mit trockenem Äther gewaschen und unter vermindertem Druck getrocknet. 30 g dieses Produktes werden dann mit einer stöchiometrischen Menge Triäthylamin in 200 ml tert.-Butanol bei 10 C während vier Stunden behandelt. Das so gebildete tert.-Butyl- (2-methyl- 5-methoxy-3-indolyl)-glyoxylat wird dann gesammelt, in Dimethylformamid bei 0 C zu einer Natriumhydrid-Mineralölsuspension (51% zig) unter Stickstoff zugegeben und mit p-Chlorbenzoylchlorid zu tert.-Butyl-(1-p-chlorbenzoyl¯2-methyl-5-methoxy-3-indolyl)-glyoxalat acyliert.
Das Gemisch wird drei Stunden bei 0 C g : rührt. Dann wird überschüssiger Äther zugesetzt und das Gemisch 50 Minuten bei 0 C gerührt. Das Gemisch wird anschliessend nitriert, der Niederschlag wird mit Äther gewaschen und die Ätherlösung wird zweimal mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingeengt.
A2) Die Verfahrensweise gemäss Ai wird mit der Abänderung wiederholt, dass äquivalente Meng : n anderer Indole, die in der C-2- od r C-5-Stdlung, wie durch die nachfolgenden Produkte angezeigt, geeignet substituiert sind, an Stelle des 2-Methyl-5-methoxyindols verwendet werden. Man erhält so die entsprechenden tert.-Butylglyoxalate, die dann mit äquivalenten Meng n der geeigneten Aroyl- oder Heteroaroylhalogenide zu folgenden Produkten acyliert werden: tert.-Butyl-(1-p-chlorbenzoyl-3-indolyl)-

Eine Lösung von 0, 05 Mol tert.-Butyl-(1-p-chlorbenzoyl-2-methyl-5-methoxy-3-indolyl)-glyoxalat in 300 ml trockenem Äther wird mit 0, 05 Mol Cblornnthylenytriphenylphosphoran (hergestellt nach den in der Literatur beschriebenen Verfahren) bei der Rückflusstemperatur während vier Stunden behandelt Die Lösung wird im Vakuum zu einem Sirup eingeengt und dann in einer 600 g Silicagel enthaltenden Säule unter Verwendung von Äther/Petioläther (Vol/Vol, 25-] 00%) als Elutionsmittel chromatographiert, wobei man den entsprechenden α-Chlormethylenylester erhält.
B2) Arbeitet man nach der Verfahrensweise des obigen Abschnittes Bl, verwendet jedoch an Stelle des dort verwendeten Chlormethylentriphenylphosphorans äquivalente Mengen des Brommethylen-, Fluormethylen-, Dichlormethylen-, Fluorchlormethylen-, Difluormethylen-, Dibrommethylen- und Bromchlormethylenderivats des Triphenylphosphorans, so elhält man die entsprechenden α-Halogenmethylenyl- indolylester.

Eg) Man arbeitet nach den Verfahrensweisen der obigen Abschnitte Ei und E2, verwendet jedoch äquivalente Mengen der folgenden Amine an Stelle des Morpholins und erhält so die entsprechenden Amide : Ammoniak, Methylamin, Isopropylamin, Diäthanolamin, Anilin, Methylanilin, Benzylamin, p-Chloranilin, ss-Methoxyäthylamin, p-Tolylamin und p-Methoxyanilin. Flüchtige Amine, wie beispielsweise Ammoniak und Methylamin, werden durch das Reaktionsgemisch geperlt.
