Process for the Production of Thiophenes Substituted in the 2-Position The invention relates to a process for the production of a number of new cyclic amidines and their non-toxic acid addition salts, namely the thiophenes substituted in the 2-position, which are particularly valuable as agents against worm diseases. In particular, the invention relates to a process for the preparation of 2 - [(0- (3-methyl-2-thienyl) -alkyl] -A-tetrahydropyrimidines and - ^ 2-imidazolines, the N-alkyl derivatives with lower alkyl groups and the non-toxic acid addition salts thereof as a means of treating worm diseases in humans and animals.
Worm disease, i.e. the infestation of the human and animal body by different types of parasites, is possibly the most common and widespread disease today. Although the importance of this disease from an economic point of view has led to extensive research into new and more effective anthelmintics, the countermeasures developed to date are not entirely satisfactory for one or more reasons; so these means show z. B. a low therapeutic index, a low specificity of the effect, they cause high costs, are not very active, and their range of action is limited.
It has now been found that the compounds mentioned, inventively available according to the invention, surprisingly, in the treatment of worm diseases in animals and humans, eg. B. in therapeutic and prophylactic treatment, when administered by the oral or parenteral route are extraordinarily effective sam. The N-alkyl derivatives with low. Alkyl groups of 2- [w- (3-methyl-2-thienyl) -alkyl] - ^ 2-tetrahydropyrimidines and - ^ 2-imidazolines have the following formula
EMI0001.0012
R1 is hydrogen or methyl, Y is ethylene or trimethylene and X is ethylene or trimethylene.
The non-toxic acid addition salts of the bases he mentioned that can be used are mainly water-soluble and water-insoluble salts, such as. B. the chlorohydrate, hydrobromide, phosphate, nitrate, sulfate, acetate, citrate, gluconate, benzoate, propionate, butyrate, sulfosalicylate, malate, laurate, malate, fumarate, succinate, oxalate, tartrate, amsonate (4,4'- Di-aminostilbene-2,2'-disulfonate), pamoate (l, l'-methylene bis - 2 - hydroxy - 3 - naphthoate), stearate, 2-hydroxy-3-naphthoate, hexafluorophosphate, toluene-p-sulfonate, the suramine salt, iodine methylate, bromomethylate, chloromethylate and resin adsorbates.
These agents are effective against both the mature and the unripened forms of worms of the families Ancylostomidae, Strongylodoidae and Tricho- strongylidae. They are particularly effective against gastrointestinal parasites of humans, ruminants (e.g. sheep, cattle, goats) and non-ruminants, e.g. B. dogs, cats, horses and pigs.
Methods for studying the responsiveness of these groups of parasites to chemotherapeutic agents exist e.g. E. In selecting a test-generated parasite infestation of a test animal which has a similar relationship between the host and the parasite as that found between such parasites and domestic animals. Such a relationship exists between Nematospiroides dubios and experimentally infected mice, between Nippostrongylus muris and experimentally infected rats and mice. The experiment with N. dubious and experimentally infected mice is carried out in such a way that the feces of an infected mouse are collected and suspended in moist animal charcoal.
Small pellets are made and incubated at room temperature for 4 to 5 days until the egg cells and larvae have developed. The larvae are then collected and used to vaccinate healthy mice. It was found that the inoculation of 40 larvae per mouse led to a successful infection, which means that after a development period of 14 days, around 30 adult worms had formed. Usual anti-wormer products proved to be ineffective against a vaccination of this magnitude.
The experiment with N. muris consists in collecting the feces of infected rats and setting up fecal cultures in the same way as for N. dubios. The cultures (animal charcoal pellets) are incubated at 26 ° C. for 7 days. The larvae are then collected and used to vaccinate healthy mice. Successful infection is achieved by inoculating 500 larvae and 1.25 mg hydrocortisone acetate (subcutaneously) into young weaned mice.
S. obvelata, the pinworm of rats and mice, is allowed to maintain itself naturally within the building colony, so that practically all mice infected with N. dubios and N. muris are also infected with S. obvelata.
Subcutaneous and intramuscular injections are the preferred methods of parenteral injection, for reasons of ease and convenience of administration and because the compounds appear to be less toxic. According to this method, the wormers described here or their nontoxic acidic acid addition salts can be administered parenterally, eg. B. by subcutaneous or intramuscular injection in animals that have different types of worm disease; the agents are administered in particular in a dose which corresponds to about 5 mg to about 150 mg of the free base per kg of body weight.
In general, a single injection is sufficient, but if the administration takes place in several doses, the injection can be repeated at regular intervals, e.g. B. monthly or possibly more often. Suitable carriers for parenteral injection are above all either aqueous carriers, such as water itself, isotonic saline solutions, isotonic dextrose solutions, Ringer's solution, or non-aqueous carriers, such as fatty oils of vegetable origin, e.g. B. cottonseed oil, peanut oil, grain oil, sesame oil. Other non-aqueous carriers can be used if they do not interfere with the therapeutic effectiveness of the preparation and are non-toxic in the volume or ratio used (glycerine, propylene glycol, sorbitol).
In addition, mixtures can advantageously be obtained for the instant preparation of solutions prior to administration. Such mixtures include, inter alia, liquid diluents, e.g. B. propylene glycol, diethyl carbonate, glycerin, sorbitol, etc., buffers and local anesthetics and inorganic salts, the. lead to the desired pharmaceutical properties.
The administration of these wormers prepared according to the invention together with hyaluronidase particularly avoids local irritation. An increase in the rate of absorption of the drug is observed and pain due to swelling and gas is usually greatly reduced, if not completely eliminated. Amounts of hyaluronidase of at least about 150 (U.S.P.) units are especially very effective in this regard. Larger or smaller amounts can of course be used, but 150 units per dose give seemingly consistently good results, which proves the absence of edema and the general behavior of the animal after injection of the preparation.
When administered orally, the preferred mode of administration of the novel products obtainable according to the invention, these are usually given in doses which correspond to about 1 mg to about 150 mg of the free base per kg of body weight. This can be done by a variety of methods, including mixing with food, giving dosed units of men, e.g. B. capsules, tablets, liquid Ge mix and solutions, including medicinal solutions, he rich, or administered in a mixture with minerals, such as sodium chloride, which are often given to animals as food supplements.
Although the indicated dose relates to the active ingredient, namely the broom form of the cyclic amidine, in practice the non-toxic acid addition salts and the free base can be used alternately, except in the cases where it is noted below.
For therapeutic purposes, a dose corresponding to 1 to 100 mg of free base per kg of body weight is recommended. A single dose is usually sufficient, but the doses can be given repeatedly, and although it is repeated for two or three consecutive days. Since the present method is effective not only against mature worms, but also against the larval stage, it is not necessary to re-administer the dose after a period of 2 to 3 weeks, as is common practice with known wormers.
When administered to sheep, goats, cattle, horses and pigs on a therapeutic basis, a medicinal agent solution is expediently administered to the animal through the throat using an administration syringe. An aqueous solution of a water-soluble, non-toxic salt is generally used for these purposes. From the standpoint of convenience and ease of manufacture, the hydrochloride is favorable. Solutions with concentrations of about 3% up to the solubility limit of the salt in water are satisfactory as drug solutions.
However, more dilute solutions can be added for drinking purposes. A 0.1% solution is suitable.
For prophylactic purposes, 1 to 50 mg (calculated as free base) per kg of body weight can be administered per day. The above methods of administration are. suitable, although one prefers to give the remedy with the feed, the water or the mineral mixture.
For the therapeutic treatment of animals, large pills and capsules are also expediently used. In animals weighing from about 13 to about 460 kg, the effective dose is especially 1/2 to 45 g, calculated as the free base. Large, appropriately sized pills containing these materials can be prepared by conventional methods.
Dry mineral mixtures or premixes which contain the products which can be prepared according to the invention can be prepared with a content of about 0.01 to about 10% of the active ingredient in a mixture with salt (sodium chloride) and other minerals who you want to treat the animal. This mixture can then be delivered on an ad libitum basis by adjusting the ratio of active ingredient in the mixture to the average daily consumption per animal to provide the correct daily dose as discussed above. If prepared food supplements are used, the material can be given in admixture with food. Again a concentration range of about 0.01 to 10i; the drug applied in food.
However, larger amounts can also be used in a satisfactory manner, depending on the taste effect of the product on the animal. This can easily be determined through experiments. In general, it is advisable to mix the daily dose with only part of the average daily ration to ensure that the dose is completely absorbed. The remainder of the daily nutritional supplement can then be administered in the usual way after taking the portion mixed with the drug. These methods are particularly suitable for prophylaxis, but similar mixtures can be used for therapeutic purposes. Concentrations of the drug in the food or mineral mixture of up to 2 to 5%, depending on the taste of the agent, are sometimes valuable.
These compounds can also be used in finely divided form, particularly when emulsions or suspensions are administered either orally or parenterally.
The active ingredients can be added to conventional foods in amounts smaller than those mentioned. Every animal food can be produced in such a way that it has the usual nutritional balance of energy-supplying substances, proteins, minerals and vitamins together with a mixture of the cyclic amidines described. Some of the various ingredients are usually grains, e.g. B. ground grain, and Kornnebenpro products; animal proteins, e.g. B. Meat and fish by-products; Vitamin mixtures, e.g. B. Vitamin A and D mixtures, riboflavin additions and other vitamin B complexes; Bone meal, lime and other inorganic compounds that provide minerals.
The relative amounts of active ingredient in foods and food concentrates can vary somewhat depending on the food with which they are used and the animal ingesting them. These substances are advantageously combined with edible carriers in such relative proportions that concentrates are created which can be easily mixed with nutritionally balanced standard feed, or which can be used as an additive to normal nutrients. A variety of carriers can be used in the preparation of the concentrates with the additions mentioned.
Suitable carriers are u. a .: soybean oil, seed meal, corn gluten meal, cottonseed oil, sunflower seed meal, linseed oil seed meal, cereal meal, lime and corncob meal. The carrier facilitates the even distribution of the active substances in the finished food with which the concentrate is mixed. This is especially important as only a small amount of these active ingredients is required. The concentrate can be superficially coated, optionally with various protein materials, edible waxes such as zein, gelatin, microcrystalline wax, and the like, to form a protective film that includes the active ingredients.
It will be appreciated that the amounts of active ingredients in such concentrates can vary widely since the amounts of active materials in the finished feed can be adjusted by mixing with the appropriate amount of concentrate. In the production of highly effective concentrates, namely premixes which are suitable for mixing by the feed manufacturer in the manufacture of the finished feed or the production of less effective concentrates, the cyclic amidine content can be between about 0.01 to 10% per kg of concentrate as already mentioned. The highly effective concentrates can be mixed with protein-containing carriers by the feed manufacturer, e.g.
B. with soybean oil seed meal to obtain concentrated supplements that are suitable for direct feeding to animals. In such cases the animals are allowed to eat the usual diet of grain, barley and other fibrous cereals and the like.
The concentrates described can also be added to animal feed in order to obtain a nutritionally balanced finished feed which contains about 1.0 to about 50 g of wormer per ton of feed. In the case of ruminants, the finished feed should contain protein, fat, fiber, carbohydrates, vitamins and minerals, each in sufficient quantities to meet the nutritional requirements of the animal for which the feed is intended. Most of these substances are found in naturally occurring feed, e.g. B. in alfalfa hay or flour, chopped corn, whole oats, soybeans, oilseed meal, silo-stored grain, ground corn on the cob, wheat bran and dried molasses.
Bone meal, lime, salt mixed with iodine and traces of minerals are often added to provide the necessary minerals, as is urea to provide additional nitrogen.
In the case of non-ruminant animals, e.g. B. pigs, a common feed can contain about 50 to 80 grains, 3 to 10% animal protein, 5 to 30% vegetable protein and 2 to 4% minerals, together with supplementary vitamin supplying substances.
As is known to those skilled in the art, the various types of diet are extremely varied depending on the purpose, mode of feeding, species, etc. Specific diets for various purposes are described by Morrison in the Appendix of Feeds and Feeding, The Morrison Publishing Company, Clinton, Iowa, 1939 been listed ..
The considerable worm disease-fighting activity of the compounds obtainable according to the invention is surprising in view of the fact that 2- (2'-thenyl) -imidazoline, the preparation and effectiveness of which against malaria is described in US Pat. No. 2,457,047, is only has little or no effectiveness as a remedy for worm infestation. It was actually found to be inactive against Nematospiroides dubius in mice when given orally on 3 consecutive ones. Days is administered at a dose of 500 mg per kg of body weight. The corresponding 2- (2'-thenyl) -tetrahydropyrimidine also proved to be inactive in this area. In contrast, the compounds obtainable according to the invention are extremely effective against N. dubius in mice.
The compounds of the formula I are prepared according to the invention by adding a w- (2-thienyl) nitrile of the formula

and a corresponding alkylenediamine in the presence of hydrogen sulfide or phosphorus pentasulfide.
The free base, which has usually been isolated by means of the solvent, can optionally be purified by recrystallization from suitable solvents or by vacuum distillation. The free bases can also be prepared by neutralizing an acid salt with sodium methylate in methanol, the base being prepared by known methods win. Other acid addition salts can easily be obtained by dissolving the free base in a suitable solvent, e.g. B. acetone, water, a lower aliphatic alcohol (ethanol, isopropanol), which contains the desired acid, dissolves, or to which the desired acid is subsequently added.
The salts are generally filtered off, precipitated with a non-solvent agent, or obtained by evaporation of the solvent or, in the case of aqueous solutions, by lyophilization. The following salts can be prepared in this way: sulfates, nitrates, phosphates, acetates, propianates, butyrates, citrates, gluconates, benzoates, pamoates, amsonates, tartrates, 3-2-hydroxy-2-naphthoates, sulfosalycilates and other salts. In the case of dibasic acids, e.g. For example, pamoic and amsonic acids, an acid to base ratio of 1: 2 (molar ratio) can be used to achieve the appropriate salt.
The inorganic polybasic acids are generally used in a molar ratio of 1: 1 with the desired base.
The salts of the cyclic amidines obtained as described above can be converted into the corresponding chlorine hydrates by percolating a methanolic solution of the corresponding salt through the chloride form of an anion exchange resin. Other acid addition salts can also be obtained in this way.
Resin adsorbates of the cyclic amidines obtainable according to the invention are conveniently prepared by slurrying an aqueous solution of a water-soluble salt of the cyclic amidine with a suspension of the sodium form of a cation exchange resin for a period of time sufficient for the resin to adsorb the compound. Suitable resins are the strong sulfonic acid cation exchange resin, such as. B. Dowex 50, Amberlite CG-120, Amberlite IR-120, Zeo-Karb 225 (available from Dow Chemical Co., Rohm & Haas, and Permutit Co. Ltd.). All resins are sulfonated styrene-divinylbenzene polymers that are cross-linked to varying degrees.
The 2- (3-methyl-2-thienyl) propionitriles used as starting materials are conveniently obtained by a reaction sequence which comprises a Knoevenagel- condensation of the suitable 3-methyl-thiophene-2-carboxaldehyde with cyanoacetic acid in the presence of a catalyst. Catalysts are e.g. B. ammonia, primary or secondary amines, pyridine, piperidine, ammonium acetate-pyridine. This is usually followed by a catalytic hydrogenation of the w- (3-methyl-2-thienyl) acrylonitriles obtained in this way.
The fact that the hydrogenation of w- (3-methyl-2-thienyl) acrylonitriles via noble metal catalysts, e.g. B. palladium, platinum, rhenium, rhodium, iridium or osmium, he can follow, is extraordinarily surprising. Palladium-on-carbon is particularly effective in terms of smooth reaction and satisfactory yield. A reaction-inert solvent should be used as the solvent. Methanol and other alcohols, aqueous tetrahydrofuran and aqueous dioxane are satisfactory solvents. The reaction can take place in a neutral, basic or acidic medium.
The basic environment is generally favorable because it accelerates the reaction rate considerably. Bases that are particularly valuable in this regard are mainly inorganic bases, e.g. B. alkali hydroxides, especially sodium and potassium hydroxide, and the quaternary ammonium hydroxides, e.g. B. R1R2R3R4NOH, where R1, R2, R3 and R4 represent an alkyl radical; R3 and R4 can be benzyl radicals and alkyl-substituted benzyl radicals, such as. B. in trimethylbenzylammonium hydroxide, (p-t-butyl) -benzyltrimethylammoniumhydroxyd and di- (p-t-butylbenzyl) -dimethylammoniumhydroxyd.
The amount of base is usually not critical, but a molar ratio of from about 0.05 to about 0.25 moles of base per mole of w- (3-methyl-2-thienyl) acrylonitrile is generally used satisfactorily. Pressure and temperature do not seem to be critical factors. In particular, pressures of up to about 3.5 kg / cm2 give good yields. Temperatures of up to 100 ° C can be used. The reaction should be stopped when the theoretical amount of hydrogen has been absorbed.
In the following examples which explain the invention, the doses given are calculated as the free base. Example 1 3- (3-Methyl-2-thienyl) -acrylonitrile A solution of 138.7 g (1.10 mol) 3-methyl-thiophene-2-carboxaldehyde, 85.0 g ( 1.0 mol) cyanoacetic acid, 3 g ammonium acetate, 110 cm3 pyridine and 200 cm3 toluene is heated under reflux in an apparatus to which a Dean-Stark water scavenger is attached.
Heating is continued for 48 hours, during which time the solution became very dark. After heating, the solution was allowed to cool and the solvents were then evaporated under reduced pressure. The less volatile residue was fractionally distilled over a column with Berl saddles as packing, a product being obtained which is initially a colorless oil. B.p. 76 C at 0.1 to 0.05 mm; n D 1.6330.
Example 2 3- (3-methyl-2-thienyl) -propionitrile A pressure vessel is filled with 74.6 g (0.5 mol) 3- (3-methyl-2-thienyl) - acrylonitrile, 50 cm. "1N sodium hydroxide, 300 cm- 'methanol and 10 g of catalyst (5% palladium on carbon) were charged. The pressure vessel was freed of air with nitrogen and then connected to a hydrogenation apparatus according to PARR. The hydrogenation was carried out in The usual procedure was carried out until the theoretical amount (0.5 mol) of hydrogen had been absorbed, the catalyst was filtered off and the filtrate concentrated until a mixture of oil and aqueous sodium hydroxide was obtained, to which water was added and the organic material with Ether extra here.
The ether extract was dried, filtered and evaporated under reduced pressure to give a pale yellow oil, which was fractionally distilled over a column filled with Berl saddles, 3- (3-methyl-2-thienyl) propionitrile being obtained as a colorless oil . Bp .: 66 C at 0.1 to 0.08 mm.
Example 3 1-Methyl-2- [2- (3-methyl-2-thienyl) ethyl] - ^ 2-imidazoline By a mixture. of w- (3-methyl-2-thienyl) propionitrile (250 g) and N-methyltrimethylenediamine (160.5 g, molar ratio 1: 1), hydrogen sulfide was passed until 6.1 g had been taken up. The mixture was then heated to 70 to 80 C for 2 hours and stirred. There was a vigorous evolution of ammonia. The temperature was increased and held at 95 ° C. for 6 hours. The product was obtained by vacuum distillation.
Phosphorus pentasulphide can be used instead of hydrogen sulphide. If 0.015 mol per mol of the reactants is used and the mixture is heated to 70 to 80 ° C. for 2 hours and then to 95 ° C. for 16 hours, a yield of more than 80% is obtained.
The following compounds can be prepared from the appropriate reactants in a similar manner:

The mixture was stirred for 3 hours, filtered, washed with water and getrock net in vacuo. Analysis for nitrogen indicated that about 41% of the base had been adsorbed.
The resin adsorbates of the product of Example 3 can be obtained in the same way. Example 5 A. 2 - [2 - (3 - methyl - thienyl) ethyl] - ^ 2-imidazoline (0.02 mol) was added to 300 cm3 of warm alcohol with a content of 0.01 Mol pamoic acid given. B. 2 - [3 - (3 - Methyl-thienyl) -propyl] - ^ 2-tetrahydropyrimidine citrate can be obtained by adding 0.01 mol of the free base, 2- [3- (3-methyl-2- thienyl) - propyl] - ^ 2-tetrahydropyrimidine, to 25 cm3 of a warm methanol solution of 0.01 mol of citric acid. The resulting clear solution was concentrated to give the salt.
With this method, the amsonate, pamoate, 2-hydroxy-3-naphthoate, sulfosalicylate, acetate, propionate, butyrate, tartrate, gluconate, p-toluenesulfonate, benzoate, citrate, stearate, sulfate, phosphate and nitrate of the product of example 3 can be obtained.
