Verfahren zur Herstellung von in 2-Stellung substituierten Thiophenen Die Erfindung betrifft ein Verfahren zur Herstellung einer Reihe neuer cyclischer Amidine und ihrer nicht toxischen Säureanlagerungssalze, nämlich der in 2-Stel- lung substituierten Thiophenen, die besonders als Mittel gegen Wurmleiden wertvoll sind. Insbesondere betrifft die Erfindung ein Verfahren zur Herstellung von 2-[w- (3-Methyl-2-thienyl)-alkyl]-d2-tetrahydropyrimidinen und -d'-imidazolinen, die N-Alkylderivate mit niedrigen Alkylgruppen und die nicht toxischen Säureanlagerungs salze davon als Mittel zur Behandlung von Wurmleiden, bei Menschen und Tieren.
Wurmleiden, das heisst der Befall des menschlichen und tierischen Körpers durch verschiedene Arten von Parasiten, sind möglicherweise die am meisten auftre tende und am weitesten verbreitete Krankheit heutzu tage. Obwohl die Bedeutung dieser Krankheit vom wirt schaftlichen Standpunkt aus zu einer ausgedehnten For schung nach neuen und wirksameren Wurmmitteln ge- führt hat, .sind die bis zum heutigen Tage entwickelten Gegenmassnahmen aus einem oder sogar mehreren Gründen nicht völlig zufriedenstellend; so zeigen diese Mittel z. B. einen niedrigen therapeutischen Index, eine geringe Spezifität der Wirkung, sie verursachen. hohe Kosten, sind wenig aktiv, und ihre Wirkungsbreite ist begrenzt.
Es wurde nun gefunden, dass die erwähnten, erfin dungsgemäss erhältlichen Verbindungen überraschender weise bei der Behandlung von Wurmleiden bei Tieren und Menschen, z. B. bei der therapeutischen und der prophylaktischen Behandlung, bei Verabreichung auf oralem oder parenteralem Wege ausserordentlich wirk sam sind. Die neuen N-Alkylderivate mit niedrigen Alkyl gruppen der 2-[w-(3-Methyl-2-thienyl)-alkyl]-d2-tetra- hydropyrimid'ine und -d2-imidazoline haben die folgende Formel

Die nichttoxischen Säureanlagerungssälze der er wähnten Basen, die verwendet werden können, sind insbesondere wasserlösliche und wasserunlösliche Salze, wie z. B. das Chlorhydrat, Hydrobromid, Phosphat, Nitrat, Sulfat, Acetat, Zitrat, Glukonat, Benzoat, Pro- pionat, Butyrat, Sulfosahcylat, Maleat, Laurat, Malat, Fumarat, Succinat, Oxalat, Tartrat, Amsonat (4,4'-Di- aminostilben-2,2'-disulfonat), Pamoat (1,1'-Methylen- bis - 2 - hydroxy - 3 - naphthoat), Stearat, 2-Hydroxy-3- naphthoat, Hexafluorphosphat, Toluol-p-sulfonat, das Suraminsalz, Jodmethylat, Brommethylat, Chlormethylat und Harzadsorbate.
Diese Mittel sind sowohl gegen die, gereiften als auch die nicht gereiften Formen von Würmern der Fa milien Ancylostomidae, Strongylodoidae und Tricho- strongylidae wirksam. Sie sind besonders gegen Gastro- intestinalparasiten von Menschen., Wiederkäuern (z. B. Schafen, Rindern, Ziegen) und Nichtwiederkäuern, z. B. Hunden, Katzen, Pferden und Schweinen, wirksam.
Methoden zum Studium der Ansprechbarkeit dieser Gruppen von Parasiten auf chemotherapeutische Mittel bestehen z. B. darin, dass man einen zu Versuchs zwecken erzeugten Parasitenbefall eines Versuchstieres auswählt, der eine ähnliche Beziehung zwischen dem Wirt und dem Parasiten aufweist wie die zwischen solchen Parasiten und Haustieren gefundene. Solch eine Beziehung besteht zwischen Nematospiroides dubius und zu Versuchszwecken infizierten Mäusen, zwischen Nippostrongylus muris und zu Versuchs zwecken infizierten Ratten und Mäusen. Der Versuch mit N. dubius und versuchsmässig infizierten Mäusen wird so ausgeführt, dass man die Fäkalien einer infizier ten Maus sammelt und in feuchter Tierkohle suspen diert.
Es werden kleine Presslinge hergestellt, und diese bei Zimmertemperatur 4 bis 5 Tage inkubiert, bis die Eizellen Brut und Larven entwickelt sind. Die Larven werden dann gesammelt und zur Impfung gesunder Mäuse verwendet. Es wurde festgestellt, dass die Ein impfung von 40 Larven pro Maus zu einer erfolgreichen Infizierung führte, das heisst, dass sich nach einer 14- tägigen Entwicklungszeit etwa 30 ausgewachsene, Würmer gebildet hatten. übliche Wurmmittel erwiesen sich gegen eine Impfung dieses Ausmasses als unwirksam.
Der Versuch mit N. muris besteht darin, dass man die Fäkalien von infizierten Ratten sammelt und in der gleichen Weise wie für N. dubius Fäkalkulturen an setzt. Die Kulturen (Tierkohlepresslinge) werden 7 Tage lang bei 26 C inkubiert. Die Larven werden dann gesammelt und zur Impfung gesunder Mäuse verwendet. Durch Einimpfen von 500 Larven und 1,25 mg Hydro- cortisonacetat (subcutan) in junge entwöhnte Mäuse er zielt man eine erfolgreiche Infizierung.
S. obvelata, den Madenwurm von Ratten und Mäu sen, lässt man auf natürliche Weise sich selbst innerhalb der Aufbaukolonie erhalten, so dass praktisch alle mit N. dubius und N. muris infizierten Mäuse auch mit S. obvelata infiziert werden.
Subcutane und intramuskuläre Injektionen. sind die bevorzugten Methoden der parenteralen. Injektion, und zwar aus Gründen der leichten und bequemen Verab- reichbarkeit und weil die Verbindungen weniger toxisch erscheinen. Nach dieser Methode können die hier be schriebenen Wurmmittel oder ihre nichttoxischen Säure anlagerungssalze parenteral verabreicht werden, z. B. durch subeutane oder intramuskuläre Injektion bei Tieren, die verschiedene Arten von Wurmleiden auf weisen; die Mittel werden insbesondere in einer Dosis verabreicht, die etwa 5 mg bis etwa 150 mg der freien Base pro kg Körpergewicht entspricht. Im allgemeinen genügt eine einzige Injektion, jedoch kann, falls die Verabreichung in mehreren Dosen erfolgt, die Injektion in regelmässigen Abständen wiederholt werden, z. B.
monatlich oder gegebenenfalls auch öfter. Geeignete Träger für die parenterale Injektion sind vor allem entweder wässrige Träger, wie Wasser. selbst, isotonische Salzlösungen, isotonische Dextroselösungen, Ringers Lösung, oder nicht wässrige Träger, wie fette Öle pflanzlichen Ursprungs, z. B. Baumwollsaatöl, Erdnussöl, Getreideöl, Sesamöl. Andere nichtwässrige Träger können verwendet werden, wenn sie nicht mit der therapeutischen Wirksamkeit des Präparates in Ge genwirkung treten und in dem verwendeten. Volu men oder dem angewandten Verhältnis nichttoxisch sind (Glycerin, Propylenglykol, Sorbitol). Ausserdem können mit Vorteil Gemische für die augenblickliche Herstellung von Lösungen vor der Verabreichung ge wonnen werden. Zu solchen Gemischen gehören unter anderem flüssige Verdünnungsmittel, z.
B. Propylen- glykol, Diäthylcarbonat, Glycerin, Sorbitol usw., Puffer und Lokalanästhetika und anorganische Salze, die, zu den angestrebten pharmazeutischen Eigenschaften führen.
Die Verabreichung dieser erfindungsgemäss herge stellten Wurmmittel zusammen mit Hyaluronidase ver meidet insbesondere eine örtliche Reizung. Es wird eine Steigerung der Absorptionsgeschwindigkeit der Droge beobachtet, und der Schmerz auf Grund von Schwellun gen und Blähungen. wird in der Regel stark vermindert, wenn nicht völlig ausgeschaltet. Mengen. an Hyaluroni- dase von mindestens etwa 150 (U. S. P.) Einheiten sind in dieser Hinsicht vor allem sehr wirksam. Grössere oder geringere Mengen können natürlich angewandt werden, jedoch ergeben: 150 Einheiten pro Dosis an scheinend ständig gute Resultate, was das Ausbleiben von Ödemen und das allgemeine Verhalten: des Tieres nach der Injektion des Präparates beweist.
Bei Verabreichung auf oralem Wege, der bevorzug ten Verabreichungsweise der erfindungsgemäss herge stellten neuen Produkte, werden diese gewöhnlich in Dosen gegeben, die etwa 1 mg bis etwa 150 mg der freien Base pro kg Körpergewicht entsprechen. Dies kann man durch zahlreiche Methoden, einschliesslich Mischen mit der Nahrung, durch Gaben dosierter Men geneinheiten, z. B. Kapseln, Tabletten, flüssigen Ge mischen und Lösungen, darunter Arzneilösungen; er reichen, oder man verabreicht im Gemisch mit Mine ralien, wie Natriumchlorid, die häufig Tieren als Nahrungsergänzung gegeben werden.
Obwohl die ange gebene Dosis sich auf den. aktiven Bestandteil bezieht, nämlich die Basenform des zyklischen Amidins, können in der Praxis die nichttoxischen Säureanlagerungssalze und die freie Base wechselseitig angewandt werden, mit Ausnahme der Fälle, wo es im folgenden. vermerkt ist.
Für therapeutische Zwecke wird eine Dosis, die 1 bis 100 mg an freier Base pro kg Körper gewicht entspricht, empfohlen. Gewöhnlich genügt eine einzelne Dosis, jedoch können die Dosen wiederholt gegeben werden, und zwar wird sie an zwei oder drei aufeinander folgenden Tagen wiederholt. Da die vor liegende Methode nicht allein bei gereiften Würmern, sondern auch gegen das Larvenstadium wirksam ist, ist es nicht nötig, die Dosis nach einer Zeit von 2 bis 3 Wochen erneut zu verabreichen, wie das im allge meinen bei bekannten Wurmmitteln üblich ist. Bei Verabreichung an Schafe, Ziegen, Rinder, Pferde und Schweine auf therapeutischer Basis, wird eine Arznei mittellösung zweckmässigerweise mit einer Verabrei- chungsspritze dem Tier durch den Schlund zugeführt.
Für diese Zwecke verwendet man im allgemeinen eine wässrige Lösung eines wasserlöslichen nichttoxischen Salzes. Vom Standpunkt der Bequemlichkeit und der Leichtigkeit der Herstellung aus ist das Chlorhydrat be günstigt. Als. Arzneimittellösungen sind Lösungen zu friedenstellend, die Konzentrationen, von etwa 3 % bis zur Löslichkeitsgrenze des Salzes in Wasser aufweisen. Verdünntere Lösungen können jedoch für Trinkzwecke zugeführt werden. Eine 0,1 %ige Lösung ist geeignet.
Für prophylaktische Zwecke können 1 bis 50 mg (berechnet als freie Base) pro kg Körpergewicht pro Tag verabreicht werden. Die obigen Verabreichungsmetho- den sind geeignet, obwohl man es vorzieht, das Mittel mit dem Futter, dem Wasser oder dem Mineralgemisch zu geben. Zur therapeutischen. Behandlung von Tieren, werden zweckmässigerweise auch grosse Pillen und Kapseln ver wendet. Die wirksame Dosis beträgt bei Tieren mit einem Gewicht von etwa 13 bis etwa 460 kg 1/2 bis 45 g, berechnet als freie Base. Grosse Pillen von geeig neter Grösse, die diese Materialien enthalten, können durch übliche Methoden hergestellt werden.
Trockene Mineralgemische oder Vorgemische, die die erfindungsgemäss hergestellten Produkte enthalten, können mit einem Gehalt von etwa 0,01 bis etwa 10 des aktiven Bestandteils im Gemisch mit Salz (Natrium chlorid) und andern Mineralien hergestellt werden, mit denen man das Tier behandeln will. Dieses Gemisch kann dann auf einer ad libitum-Basis zugeführt werden, indem man das Verhältnis von aktivem Bestandteil im Gemisch zum durchschnittlichen täglichen Verbrauch pro Tier einstellt, um die richtige tägliche Dosis, wie oben erläutert, bereitzustellen. Wenn vorbereitete Nahrungsergänzungsmittel verwendet werden, kann das Material im Gemisch mit der Nahrung gegeben werden. Wiederum wird ein Konzentrationsbereich von etwa 0,01bis 10% der Droge in der Nahrung angewandt.
Es können jedoch auch in zufriedenstellender Weise grössere Mengen verwendet werden, je nach der Ge schmackswirkung des Produktes auf das Tier. Dies lässt sich durch Versuche leicht ermitteln. Im allgemeinen ist es anzuraten, die tägliche Dosis mit nur einem Teil der durchschnittlichen Tagesration zu mischen, um eine völlige Aufnahme der Dosis sicherzustellen. Der Rest der täglichen Nahrungsergänzung kann dann nach Auf nahme der mit dem Medikament versetzten Portion in üblicher Weise verabreicht werden. Diese Methoden eignen sich besonders für die Prophylaxe, jedoch kön nen ähnliche Gemische für therapeutische Zwecke ver wendet werden. Mitunter sind Konzentrationen der Droge in der Nahrung oder dem Mineralgemisch bis 2 bis<B>5%,</B> je nach dem Geschmack des Mittels, wertvoll.
Diese Verbindungen können auch in feinzerteilter Form verwendet werden, insbesondere, wenn Emulsionen oder Suspensionen entweder oral oder parenteral verabreicht werden.
Die aktiven Bestandteile können üblichen Nahrungs mitteln in kleineren als den erwähnten Mengen zuge setzt werden. Jedes Tiernahrungsmittel kann so herge stellt werden, dass es den üblichen ernährungsmässigen Ausgleich von Energie liefernden Substanzen, Proteinen, Mineralien und Vitaminen zusammen mit einem Ge misch der beschriebenen zyklischen Amidine aufweist. Einige der verschiedenen Bestandteile sind üblicherweise Körner, z. B. gemahlenes Korn, sowie Kornnebenpro dukte; tierische Proteine, z. B. Fleisch und Fischneben produkte; Vitamingemische, z. B. Vitamin.
A- und D- Gemische, Riboflavinzugaben und andere Vitamin B- Komplexe; Knochenmehl, Kalk und andere anorganische Verbindungen, die Mineralien bereitsstellen.
Die relativen Mengen an aktivem Bestandteil in Nahrungsmitteln und Nahrungsmittelkonzentraten kön nen etwas variieren, je nach dem Nahrungsmittel, mit dem sie verwendet werden, und dem Tier, das sie auf nimmt. Diese Substanzen werden vorteilhafterweise in solchen relativen Proportionen mit essbaren Träger stoffen kombiniert, dass Konzentrate entstehen, die sich leicht mit ernährungsmässig ausgewogenem Standard futter mischen lassen, oder die selbst als Zusatz zu den normalen Nahrungsstoffen verwendet werden können.
Bei der Herstellung der Konzentrate mit den er wähnten Zugaben. kann eine Vielzahl von Trägern ver- wendet werden. Geeignete Träger sind u. a.: Soja bohnenöl, Saatmehl, Maisklebermehl, Baumwollölsaat- mehl, Sonnenblumensamenmehl, Leinölsaatmehl, Ge treidemehl, Kalk- und Maiskolbenmehl. Der Träger er leichtert die gleichmässige Verteilung der aktiven Stoffe in der fertigen Nahrung, mit der das Konzentrat ge mischt wird. Dies ist besonders wichtig, da nur eine kleine Menge dieser wirksamen Stoffe erforderlich ist.
Das Konzentrat kann oberflächlich überzogen werden, gegebenenfalls mit verschiedenen Proteinmaterialien, ess baren Wachsen, wie Zein, Gelatine, mikrokristallinem Wachs und dergleichen, um einen Schutzfilm zu bilden, der die aktiven. Bestandteile einschliesst. Es ist einzu sehen, dass die Mengen der aktiven Bestandteile in solchen Konzentraten stark variieren können, da die Mengen der aktiven. Materialien im fertigen Futter durch Mischen mit der geeigneten Menge an Konzentrat eingestellt werden können.
Bei der Herstellung hoch wirksamer Konzentrate, nämlich Vorgemischen, die sich zum Mischen. durch den Futterhersteller bei der Her stellung des fertigen Futters eignen oder der Herstellung von weniger wirksamen Konzentraten, kann der Gehalt an cyclischem Amidin zwischen etwa 0,01 bis 10% pro kg Konzentrat liegen, wie bereits erwähnt. Die hoch wirksamen Konzentrate können durch den Futter hersteller mit proteinhaltigen Trägern gemischt werden, z. B. mit Sojabohnenölsaatmehl, um konzentrierte Er gänzungsmittel zu erhalten, die zur direkten Verfütte- rung an Tiere geeignet sind. In solchen Fällen lässt man die Tiere die übliche Diät 'an Korn, Gerste und andern faserhaltigen Getreidearten und dergleichen fressen.
Die .beschriebenen Konzentrate können auch Tier futter zugesetzt werden:, um ein ernährungsmässig aus geglichenes fertiges Futter zu erhalten, das etwa 1,0 bis etwa 50 g an Wurmmittel pro Tonne Futter enthält. Im Falle von Wiederkäuern sollte das fertige Futter Protein, Fett, Fasern, Kohlenhydrate, Vitamine und Mineralstoffe enthalten, jeweils in ausreichender Menge, um die ernährungsmässigen Erfordernisse des Tieres zu erfüllen, für das das Futter bestimmt ist. Die meisten dieser Substanzen, sind in natürlich vorkommenden Futterstoffen vorhanden, z. B. in Alfalfaheu oder Mehl, zerkleinertem Mais, unzerkleinertem Hafer, Sojabohnen ölsaatmehl, im Silo gespeichertem Getreide, gemahlenen Maiskolben, Weizenkleie und getrockneten Melassen.
Knochenmehl, Kalk, mit Jod versetztes Salz und Spuren von Mineralien werden häufig, um die nötigen Minera lien bereitzustellen, zugesetzt, desgleichen Harnstoff zur Bereitstellung von zusätzlichem Stickstoff.
Im Falle von nicht wiederkäuenden Tieren, z. B. Schweinen, kann ein übliches Futter etwa 50 bis 80 Körner, 3 bis<B>10%</B> tierisches Protein, 5 bis<B>30,%</B> pflanz liches Protein und 2 bis 4 % Mineralien enthalten, zu sammen mit ergänzenden Vitamin liefernden Substanzen.
Wie dem Fachmann bekannt, sind die verschiedenen Diätarten ausserordentlich vielseitig, je nach dem Zweck, der Verfütterungsweise, der Species usw. Spezielle Diäten für verschiedene Zwecke sind von Morrison in dem Appendix of Feeds and Feeding , The Morrison Publishing Company, Clinton, Iowa, 1939 aufgeführt worden.
Die beträchtliche, Wurmleiden bekämpfende Akti vität der erfindungsgemäss erhältlichen Verbindungen überrascht im Hinblick auf die Tatsache, dass 2-(2'- Thenyl)-imidazolin, dessen Herstellung und Wirksamkeit gegen Malaria in der USA-Patentschrift Nr. 2457047 beschrieben ist, nur eine geringe oder keine Wirksam keit als Mittel gegen Wurmbefall aufweist. Es erwies sich als tatsächlich inaktiv gegen Nematospiroides dubius in Mäusen, wenn es oral an 3 aufeinander folgenden Tagen in einer Dosis von 500 mg pro kg Körpergewicht verabreicht wird. Das entsprechende 2-(2'-Thenyl)-tetrahydropyrimidin erwies sich auch als inaktiv in diesem Bereich.
Die erfindungsgemäss erhält lichen Verbindungen sind dagegen ausserordentlich wirk sam gegen N. dubius in Mäusen.

wobei R4 einen niedrigen Alkylrest bedeutet, umsetzt. Die Chlorhydrate, die wie beschrieben erhalten werden, können durch einfache Neutralisation des Säureanteils des Salzes leicht in die freie Base über geführt werden, und zwar insbesondere durch wässriges Natrium- oder Kaliumhydroxyd, und die wasserunlös liche freie Base kann auf mechanischem Wege oder durch Lösungsmittelextraktion mit einem geeigneten nicht mischbaren Lösungsmittel, wie Äthylacetat, isoliert werden.
Die freie Base, die mittels des Lösungsmittels isoliert worden ist, kann gegebenenfalls durch Umkri stallisation aus geeigneten Lösungsmitteln oder durch Vakuumdestillation gereinigt werden. Man kann die freien Basen auch durch Neutralisation eines sauren Salzes mit Natriummethylat in Methanol herstellen, wobei man die Base gewöhnlich durch bekannte Methoden gewinnt. Andere Säureanlagerungssalze können leicht gewonnen werden, indem man die freie Base in einem geeigneten Lösungsmittel, z. B. Aceton, Wasser, einem niedrigen aliphatischen Alkohol (Äthanol, Isopropanol), der die gewünschte Säure enthält, löst, oder zu dem die gewünschte Säure nachträglich gegeben wird.
Die Salze werden im allgemeinen abfiltriert, mit einem sie nicht lösenden Mittel ausgefällt, oder durch Eindampfen des Lösungsmittels gewonnen oder im Falle wässriger Lö sungen durch Lyophylisierung. Auf diese Weise können folgende Salze hergestellt werden: Sulfate, Nitrate, Phosphate, Acetate, Propionate, Butyrate, Zitrate, Gluconate, Benzoate, Pamoate, Amsonate, Tartrate, 3-Hydroxy-2-naphthoate, Sulfosalycilate und andere Salze. Im Falle zweibasischer Säuren, z. B. Pamoe- und Amsoninsäure, wird insbesondere ein Verhältnis von Säure zu Base von 1 :2 (Molverhältins) angewandt, um das entsprechende Salz zu erzielen.
Die anorgani schen mehrbasischen Säuren werden im allgemeinen im Molverhältnis von 1 : 1mit der gewünschten Base an gewandt.
Die Salze der wie oben beschrieben erhaltenen cyclischen Amidine können in die entsprechenden Chlor hydrate durch Percolieren einer methanolischen Lösung des entsprechenden Salzes durch die Chloridform eines Anionenaustauscherharzes übergeführt werden. Andere Säureanlagerungssalze können auf diese Weise ebenfälls erhalten werden.
Harzadsorbate der neuen cyclischen Amidine werden zweckmässigerweise durch Aufschlämmung einer wässrigen Lösung eines wasserlöslichen Salzes des cychschen Amidins mit einer Suspension der Natrium form eines Kationenaustauscherharzes während einer zur Adsorption der Verbindung durch das Harz aus reichenden Zeitspanne hergestellt. Geeignete Harze sind das stark sulfonsaure Kationenaustauscherharz, wie z. B. Dowex 50 , Amberlite CG-120 , Amberlite IR-120 , Zeo-Karb 225 (erhältlich von der Dow Chemical Co., Rohm & Haas, und der Permutit Co. Ltd.). Alle Harze sind sulfonierte Styrol-Divinylbenzol polymerisate, die in wechselndem Ausmass vernetzt sind.
Die als Ausgangsstoffe verwendeten 2-(3-Methyl-2- thienyl)-propionitrile kann man zweckmässigerweise durch eine neue Reaktionsfolge, die eine Knoevenagel- Kondensation des geeigneten 3-Methyl-thiophen-2- aldehyds mit Cyanoessigsäure in Gegenwart eines Kata- lysators umfasst, herstellen. Katalysatoren sind z. B. Ammoniak, primäre oder sekundäre Amine, Pyridin, Piperidin, Ammoniumacetat Pyridin. Dem folgt in übli cher Weise eine katalytische Hydrierung der so erhal tenen w-(3-Methyl-2-thienyl)-acrylnitrile.
Die Tatsache, dass die Hydrierung der w-(3-Methyl-2-thienyl)-acryl- nitrile über Edelmetallkatalysatoren, z. B. Palladium, Platin, Rhenium, Rhodium, Iridium oder Osmium, er folgen kann, überrascht ausserordentlich. Palladium-auf- Kohle ist besonders wirksam im Hinblick auf eine glatte Reaktion und zufriedenstellende Ausbeute. Als Lösungsmittel sollte ein reaktionsinertes Lösungsmittel verwendet werden. Methanol und andere Alkohole, wässriges Tetrahydrofuran und wässriges Dioxan sind zufriedenstellende Lösungsmittel. Die Reaktion kann in neutralem, basischem oder saurem Milieu vonstatten gehen.
Das basische Milieu ist im allgemeinen be günstigt, da es die Reaktionsgeschwindigkeit erheblich beschleunigt. Basen, die besonders in dieser Hinsicht wertvoll sind, sind anorganische Basen, z. B. Alkali hydroxyde, insbesondere Natrium- und Kalium hydroxyd, und die quaternären Ammoniumhydroxyde, z. B. R1R2R3R4NOH, wobei R1, R2, R3 und R4 einen Alkylrest .bedeuten; R3 und R4 können Benzylreste und alkylsubstituierte Benzylreste sein, wie z. B. in Trimethylbenzyl-ammoniumhydroxyd, (p-t-Butyl)-benzyltrimethylammoniumhydroxyd und Di-(p-t-butylbenzyl)-dimethylammoniumhydroxyd.
Die Menge der Base ist in der Regel nicht kritisch, doch wird im allgemeinen ein Molverhältnis von etwa 0,05 bis etwa 0,25 Mol Base pro Mol 0,)-(3-Methyl-2- thienyl)-acrylnitril zufriedenstellend angewandt. Druck und Temperatur scheinen keine kritischen Faktoren zu sein. Drucke von bis zu etwa 3,5 kg/cm? ergeben vor allem gute Ausbeuten. Es können Temperaturen bis zu 100 C angewandt werden. Die Reaktion sollte unterbrochen werden, wenn die theoretische Menge an Wasserstoff aufgenommen worden ist.
In den folgenden Beispielen, die die Erfindung er läutern, sind die angegebenen Dosen als freie Base be rechnet.
<I>Beispiel 1</I> 3-(3-Methyl-2-thienyl)-acrylnitril Eine Lösung von 138,7 g (1,l0 Mol) 3-Methyl- thiophen-2-aldehyd, 85,0 g (1,0 Mol) Cyanessig- säure, 3 g Ammoniumacetat, 110 cm3 Pyridin und 200 cm3 Toluol wird unter Rückfluss in einer Apparatur erhitzt, an der ein Wasserfänger nach Dean-Stark an gebracht ist. Das Erhitzen wird 48 Stunden lang fort gesetzt, während welcher Zeit die Lösung sehr dunkel wurde. Nach dem Erhitzen lässt man die Lösung ab kühlen, und die Lösungsmittel wurden dann unter ver mindertem Druck abgedampft.
Der weniger flüchtige Rückstand wurde über eine Kolonne mit Berl-Sätteln als Füllkörper fraktioniert destilliert, wobei ein Produkt erhalten wurde, das anfänglich ein farbloses Öl ist. Kp. 76 C bei 0,1 bis 0,05 mm; n D 1,6330.
<I>Beispiel 2</I> 3-(3-Methyl-2-thienyl)-propionitril Ein Druckgefäss wird mit 74,6 g (0,5 Mol) 3-(3-Me- thyl-2 thienyl)-acrylnitril, 50 cm3 1n Natriumhydroxyd, 300 cm3 Methanol und 10g Katalysator (5% Palladium auf-Kohle) beschickt. Das Druckgefäss wurde durch Stickstoff von Luft befreit und dann an einen Hydrie rungsapparat nach PARR angeschlossen. Die Hydrie rung wurde in üblicher Weise durchgeführt, bis die theoretische Menge (0,5 Mol) Wasserstoff absorbiert ist. Der Katalysator wurde abfiltriert und das Filtrat kon zentriert, bis ein Gemisch an Öl und wässrigem Natrium hydroxyd vorliegt. Zu diesem Gemisch wurde Wasser gegeben und das organische Material mit Äther extra hiert.
Der Ätherextrakt wurde getrocknet, filtriert und unte vermindertem Druck eingedampft unter Erzielung eines blassgelben Öles., das über eine mit Berl-Sätteln gefüllte Kolonne fraktioniert destilliert wurde, wobei 3 - (3 -Methyl - 2-thienyl)-propionitril als farbloses Öl anfiel. KP.: 66 C bei 0,1 bis 0,08 mm.
<I>Beispiel 3</I> 1-Methyl-2-[2-(3-methyl-2-thienyl)-äthyl]- d2 tetrahydropyrimidin Methyl -B - (3-methyl-2-thienyl)-propionimidatchlor- hydrat (31,8 g, hergestellt aus dem entsprechenden Ni tril des Beispiels 2 und Methanol durch ein übliches Verfahren) wird zu einer Lösung von N-Methyltri- methylen-diamin (18,5 g) in Methanol (250 cm3) bei 0 C gegeben. Das Gemisch wurde über Nacht unter Rückfluss erhitzt und dann unter vermindertem Druck zur Trockne eingeengt.
Der Rückstand wird mit 100 cm3 40%igem wässri gem Natriumhydroxyd bei etwa 5 C behandelt, dann einige Minuten geschüttelt und dann mit Äther extra hiert. Die ätherische Lösung wurde mit wasserfreiem Natriumsulfat getrocknet und dann eingeengt. Die Extraktion der wässrigen Lösung mit Chloroform ergab eine zusätzliche Menge der gewünschten Base. Die ver einigten Basenanteile aus den Extraktionen wurden im Vakuum unter Erzielung der freien Base destilliert.
Man erhielt das Hexafluorphosphatsalz durch Zu gabe der Base zu einem Eiswassergemisch, das eine äquimolare Menge Hexafluorphosphorsäure enthielt. Das Salz fiel aus, wurde abfiltriert und aus 2-Propanol umkristallisiert. Schmelzpunkt 116,5 bis 117,5 C.

Die Lösung wurde konzentriert, gekühlt und das aus gefällte Natriumchlorid abfiltriert. Das Lösungsmittel wurde dann von dem Filtrat abdestilliert, und der Rück stand wurde im Vakuum destilliert. Man erhielt die Base als beinahe farbloses Öl. <I>Beispiel 5</I> 5 g 2-[2-(3-Methyl-2-thienyl)-äthyl]-d2-tetrahydro- pyrimidinchlorhydrat wurden in 25 cm3 Wasser gelöst, die Lösung filtriert und zu einer gut gerührten Suspen sion von Amberlite CG-120 (Natriumform eines Katio nenaustauscherharzes; 5,9 g) in 100 cm3 Wasser gege ben.
Das Gemisch wurde 3 Stunden lang gerührt, fil triert, mit Wasser gewaschen und im Vakuum getrock net. Die Analyse auf Stickstoff ergab, dass etwa 41 der Base adsorbiert worden waren.
Die Harzadsorbate der Produkte der Beispiele 3 und 4 können in gleicher Weise erhalten werden. <I>Beispiel 6</I> A. 2 - [2 - (3 - Methyl - thienyl)-äthyl]-d2-imidazolin (0,02 Mol) wurde zu 300 cm3 warmem Alkohol mit einem Gehalt von 0,01 Mol Pamoesäure gegeben. Das Gemisch wurde 3 Stunden lang gerührt, anschliessend filtriert unter Erzielung von Di-2-[2-(3-Methyl-2- thienyl)-äthyl]-d2-imid'azolin-pamoat. B.
2 - [3 - (3 - Methyl-thienyl)-propyl]-d2-tetrahydro- pyrimidincitrat kann gewonnen werden, indem man 0,01 Mol der freien Base, 2-[3-(3-Methyl-2-thienyl)- propyl]-d2-tetrahydropyrimidin, zu 25 cm3 einer warmen Methanollösung von 0,01 Mol Zitronensäure gibt. Die resultierende klare Lösung wurde unter Erzielung des Salzes eingeengt.
Mit diesem Verfahren können das Amsonat, Pamoat, 2-Hydroxy-3-naphthoat, Sulfosalicylat, Acetat, Propio- nat, Butyrat, Tartrat, Gluconat, p-Toluolsulfonat, Benzoat, Zitrat, Stearat, Phosphat und Nitrat des Pro duktes des Beispiels 3 erhalten werden.

In particular, the invention relates to a process for the preparation of 2- [w- (3-methyl-2-thienyl) -alkyl] -d2-tetrahydropyrimidines and -d'-imidazolines, the N-alkyl derivatives with lower alkyl groups and the nontoxic acid addition salts of which as an agent for the treatment of worm diseases, in humans and animals.
Worm disease, i.e. the infestation of the human and animal body by different types of parasites, is possibly the most common and widespread disease today. Although the importance of this disease from an economic point of view has led to extensive research into new and more effective anthelmintics, the countermeasures developed to date are not entirely satisfactory for one or more reasons; so these means show z. B. a low therapeutic index, a low specificity of the effect they cause. high costs, are not very active, and their scope is limited.
It has now been found that the compounds mentioned, inventively available according to the invention, surprisingly, in the treatment of worm diseases in animals and humans, eg. B. in therapeutic and prophylactic treatment, when administered by the oral or parenteral route are extraordinarily effective sam. These agents are effective against both the mature and the unripened forms of worms of the families Ancylostomidae, Strongylodoidae and Tricho- strongylidae. They are particularly effective against gastrointestinal parasites of humans, ruminants (e.g. sheep, cattle, goats) and non-ruminants, e.g. B. dogs, cats, horses and pigs.
Methods for studying the responsiveness of these groups of parasites to chemotherapeutic agents exist e.g. B. in that one selects a test-generated parasite infestation of a test animal which has a similar relationship between the host and the parasite as that found between such parasites and domestic animals. Such a relationship exists between Nematospiroides dubius and experimentally infected mice, between Nippostrongylus muris and experimentally infected rats and mice. The experiment with N. dubius and experimentally infected mice is carried out in such a way that the feces of an infected mouse are collected and suspended in moist animal charcoal.
Small pellets are made and incubated at room temperature for 4 to 5 days until the egg cells and larvae have developed. The larvae are then collected and used to vaccinate healthy mice. It was found that the inoculation of 40 larvae per mouse led to a successful infection, which means that after a development period of 14 days, around 30 adult worms had formed. Common wormer products proved to be ineffective against vaccination on this scale.
The experiment with N. muris consists in collecting the feces from infected rats and starting fecal cultures in the same way as for N. dubius. The cultures (pressed animal charcoal) are incubated at 26 ° C. for 7 days. The larvae are then collected and used to vaccinate healthy mice. Successful infection is achieved by inoculating 500 larvae and 1.25 mg hydrocortisone acetate (subcutaneously) into young weaned mice.

Subcutaneous and intramuscular injections. are the preferred methods of parenteral. Injection, for reasons of ease and convenience of administration and because the compounds appear less toxic. Other non-aqueous carriers can be used if they do not interfere with the therapeutic effectiveness of the preparation and in the one used. This mixture can then be delivered on an ad libitum basis by adjusting the ratio of active ingredient in the mixture to the average daily consumption per animal to provide the correct daily dose as discussed above. However, larger amounts can also be used in a satisfactory manner, depending on the taste effect of the product on the animal. This can easily be determined through experiments. These compounds can also be used in finely divided form, particularly when emulsions or suspensions are administered either orally or parenterally.
The active ingredients can be added to conventional foods in amounts smaller than those mentioned. Every animal food can be produced in such a way that it has the usual nutritional balance of energy-supplying substances, proteins, minerals and vitamins together with a mixture of the cyclic amidines described. Some of the various ingredients are usually grains, e.g. B. ground grain, and Kornnebenpro products; animal proteins, e.g. B. Meat and fish by-products; Vitamin mixtures, e.g. B. Vitamin.
A and D mixtures, riboflavin additions and other vitamin B complexes; Bone meal, lime and other inorganic compounds that provide minerals.
The relative amounts of active ingredient in foods and food concentrates can vary somewhat depending on the food with which they are used and the animal ingesting them. These substances are advantageously combined with edible carriers in such relative proportions that concentrates are created which can be easily mixed with nutritionally balanced standard feed, or which can be used as an additive to normal nutrients.
When making the concentrates with the additions mentioned. a variety of carriers can be used. Suitable carriers are u. a .: soybean oil, seed meal, corn gluten meal, cottonseed oil, sunflower seed meal, linseed oil seed meal, cereal meal, lime and corncob meal. The carrier facilitates the even distribution of the active substances in the finished food with which the concentrate is mixed. This is especially important as only a small amount of these active ingredients is required.
The concentrate can be superficially coated, optionally with various protein materials, edible waxes such as zein, gelatin, microcrystalline wax and the like, in order to form a protective film which the active. Includes components. It will be appreciated that the amounts of the active ingredients in such concentrates can vary widely as the amounts of the active. Materials in the finished feed can be adjusted by mixing with the appropriate amount of concentrate.
In the manufacture of highly effective concentrates, namely premixes, which are suitable for mixing. by the feed manufacturer in the manufacture of the finished feed or the manufacture of less effective concentrates, the cyclic amidine content can be between about 0.01 to 10% per kg of concentrate, as already mentioned. The highly effective concentrates can be mixed with protein-containing carriers by the feed manufacturer, e.g. B. with soybean oil seed meal to obtain concentrated supplements that are suitable for direct feeding to animals. In such cases the animals are allowed to eat the usual diet of grain, barley and other fibrous cereals and the like.
The concentrates described can also be added to animal feed: in order to obtain a nutritionally balanced finished feed that contains about 1.0 to about 50 g of wormer per ton of feed. In the case of ruminants, the finished feed should contain protein, fat, fiber, carbohydrates, vitamins and minerals, each in sufficient quantities to meet the nutritional requirements of the animal for which the feed is intended. Most of these substances are found in naturally occurring feed, e.g. B. in alfalfa hay or flour, chopped corn, whole oats, soybeans, oilseed meal, silo-stored grain, ground corn on the cob, wheat bran and dried molasses.
Bone meal, lime, salt mixed with iodine and traces of minerals are often added to provide the necessary minerals, as is urea to provide additional nitrogen.
In the case of non-ruminating animals, e.g. B. pigs, a common feed can contain about 50 to 80 grains, 3 to <B> 10% </B> animal protein, 5 to <B> 30% </B> vegetable protein and 2 to 4% minerals , together with supplementary vitamin supplying substances.

The considerable worm disease-fighting activity of the compounds obtainable according to the invention is surprising in view of the fact that 2- (2'-thenyl) -imidazoline, the preparation and effectiveness of which against malaria is described in US Pat. No. 2457047, is only slight or is not effective as a remedy for worm infestation. It was actually found to be inactive against Nematospiroides dubius in mice when administered orally for 3 consecutive days at a dose of 500 mg per kg body weight. The corresponding 2- (2'-thenyl) -tetrahydropyrimidine also proved to be inactive in this area.

All resins are sulfonated styrene-divinylbenzene polymers that are cross-linked to varying degrees.
The 2- (3-methyl-2-thienyl) -propionitriles used as starting materials can conveniently be prepared by a new reaction sequence which comprises a Knoevenagel- condensation of the appropriate 3-methyl-thiophene-2-aldehyde with cyanoacetic acid in the presence of a catalyst , produce. Catalysts are e.g. B. ammonia, primary or secondary amines, pyridine, piperidine, ammonium acetate, pyridine. This is followed in a customary manner by catalytic hydrogenation of the w- (3-methyl-2-thienyl) acrylonitriles obtained in this way.
The fact that the hydrogenation of w- (3-methyl-2-thienyl) acrylonitriles via noble metal catalysts, e.g. B. palladium, platinum, rhenium, rhodium, iridium or osmium, he can follow, is extraordinarily surprising. Palladium-on-carbon is particularly effective in terms of smooth reaction and satisfactory yield. A reaction-inert solvent should be used as the solvent. Methanol and other alcohols, aqueous tetrahydrofuran and aqueous dioxane are satisfactory solvents. The reaction can take place in a neutral, basic or acidic medium.
The basic environment is generally favorable because it accelerates the reaction rate considerably. Bases that are particularly valuable in this regard are inorganic bases, e.g. B. alkali hydroxide, especially sodium and potassium hydroxide, and the quaternary ammonium hydroxides, z. B. R1R2R3R4NOH, where R1, R2, R3 and R4 mean an alkyl radical; R3 and R4 can be benzyl radicals and alkyl-substituted benzyl radicals, such as. B. in trimethylbenzylammonium hydroxide, (p-t-butyl) -benzyltrimethylammoniumhydroxyd and di- (p-t-butylbenzyl) -dimethylammoniumhydroxyd.
The amount of base is usually not critical, but a molar ratio of from about 0.05 to about 0.25 moles of base per mole of 0.1 - (3-methyl-2-thienyl) acrylonitrile is generally used satisfactorily. Pressure and temperature do not seem to be critical factors. Prints up to about 3.5 kg / cm? give above all good yields. Temperatures of up to 100 ° C can be used. The reaction should be stopped when the theoretical amount of hydrogen has been absorbed.
In the following examples which explain the invention, the doses given are calculated as the free base.
<I> Example 1 </I> 3- (3-methyl-2-thienyl) acrylonitrile A solution of 138.7 g (1.10 mol) 3-methylthiophene-2-aldehyde, 85.0 g ( 1.0 mole) of cyanoacetic acid, 3 g of ammonium acetate, 110 cm3 of pyridine and 200 cm3 of toluene are heated under reflux in an apparatus to which a Dean-Stark trap is attached. Heating is continued for 48 hours, during which time the solution became very dark. After heating, the solution was allowed to cool and the solvents were then evaporated under reduced pressure.
The less volatile residue was fractionally distilled over a column with Berl saddles as packing, a product being obtained which is initially a colorless oil. B.p. 76 C at 0.1 to 0.05 mm; n D 1.6330.
<I> Example 2 </I> 3- (3-Methyl-2-thienyl) -propionitrile A pressure vessel is filled with 74.6 g (0.5 mol) of 3- (3-methyl-2-thienyl) -acrylonitrile , 50 cm3 of 1N sodium hydroxide, 300 cm3 of methanol and 10g of catalyst (5% palladium on carbon). The pressure vessel was freed from air with nitrogen and then connected to a hydrogenation apparatus according to PARR. The hydrogenation was carried out in the usual manner until the theoretical amount (0.5 mol) of hydrogen was absorbed. The catalyst was filtered off and the filtrate was concentrated until a mixture of oil and aqueous sodium hydroxide was obtained. Water was added to this mixture and the organic material was extracted with ether.
The ether extract was dried, filtered and evaporated under reduced pressure to give a pale yellow oil, which was fractionally distilled over a column filled with Berl saddles, 3 - (3-methyl-2-thienyl) -propionitrile being obtained as a colorless oil. KP .: 66 C at 0.1 to 0.08 mm.
<I> Example 3 </I> 1-Methyl-2- [2- (3-methyl-2-thienyl) ethyl] - d2 tetrahydropyrimidine methyl -B - (3-methyl-2-thienyl) propionimidate chlorohydrate (31.8 g, prepared from the corresponding Ni tril of Example 2 and methanol by a conventional method) is added to a solution of N-methyltrimethylene-diamine (18.5 g) in methanol (250 cm3) at 0 C. . The mixture was refluxed overnight and then concentrated to dryness under reduced pressure.
The residue is treated with 100 cm3 of 40% aqueous sodium hydroxide at about 5 ° C., then shaken for a few minutes and then extracted with ether. The ethereal solution was dried with anhydrous sodium sulfate and then concentrated. Extraction of the aqueous solution with chloroform gave an additional amount of the desired base. The combined base fractions from the extractions were distilled in vacuo to obtain the free base.
The hexafluorophosphate salt was obtained by adding the base to an ice-water mixture which contained an equimolar amount of hexafluorophosphoric acid. The salt precipitated, was filtered off and recrystallized from 2-propanol. Melting point 116.5 to 117.5 C.

The resulting solution was refluxed for 11/2 hours, cooled and neutralized with methanolic sodium hydroxide. The solution was concentrated, cooled and the sodium chloride which had precipitated out was filtered off. The solvent was then distilled off from the filtrate and the residue was distilled in vacuo. The base was obtained as an almost colorless oil. <I> Example 5 </I> 5 g of 2- [2- (3-methyl-2-thienyl) ethyl] -d2-tetrahydropyrimidine chlorohydrate were dissolved in 25 cm3 of water, the solution was filtered and formed into a well-stirred suspension sion of Amberlite CG-120 (sodium form of a cation exchange resin; 5.9 g) in 100 cm3 of water.
The mixture was stirred for 3 hours, filtered, washed with water and getrock net in vacuo. Analysis for nitrogen indicated that about 41% of the base had been adsorbed.
The resin adsorbates of the products of Examples 3 and 4 can be obtained in the same manner. <I> Example 6 </I> A. 2 - [2 - (3 - methylthienyl) ethyl] -d2-imidazoline (0.02 mol) was added to 300 cm3 of warm alcohol with a content of 0.01 mol Pamoic acid given. The mixture was stirred for 3 hours, then filtered to obtain di-2- [2- (3-methyl-2-thienyl) ethyl] -d2-imid'azoline-pamoate. B.
2 - [3 - (3 - Methyl-thienyl) -propyl] -d2-tetrahydro- pyrimidine citrate can be obtained by adding 0.01 mol of the free base, 2- [3- (3-methyl-2-thienyl) - propyl] -d2-tetrahydropyrimidine, to 25 cm3 of a warm methanol solution of 0.01 mol of citric acid. The resulting clear solution was concentrated to give the salt.
With this process, the amsonate, pamoate, 2-hydroxy-3-naphthoate, sulfosalicylate, acetate, propionate, butyrate, tartrate, gluconate, p-toluenesulfonate, benzoate, citrate, stearate, phosphate and nitrate of the product of Example 3 can be obtained.
