Verfahren zur Herstellung von co"(3"Methyl"2"thienyl)"propionitril
Diese Erfindung betrifft ein neues Verfahren zur Herstellung von cs-(3-Methyl-2-thienyl)-propionitril.
Im Schweizer Patent Nr. 464 209 wurde bereits eine Reihe neuer cyclischer Amidine sowie deren nicht toxische Säureanlagerungssalze beschrieben, welche insbesondere als Mittel gegen Wurmleiden wertvoll sind.
Die neuen Verbindungen können hergestellt werden, indem man ein Alkylendiamintosylat mit einem-(3-Me- thyl-2-thienyl)-substituierten Nitril zum Tosylatsalz des cyclischen Amidins umsetzt. Es ist auch möglich, Imino äther-Chlorhydrate, die den erwähnten Nitrilen entsprechen, mit dem geeigneten Alkylendiamin umzusetzen, wobei das Hydrochlorid des cyclischen Amidins entsteht. Eine weitere Methode kann auch auf der Umsetzung eines Esters der c,)-(3-Methyl-2-thienyl)-alkancar- bonsäure, wie z. B. des Methyl-3-(3-methyl-2-thienyl)propionats oder -butyrats mit dem gewünschten Alkylendiamin beruhen, wobei die cyclische freie Amidinbase entsteht.
Das neue w-(3-Methyl-2-thienyl)-propionitril wird erfindungsgemäss hergestellt, indem man - nach Knoe vennagel-3-Methylthiophen-2-aldehyd mit Cyanessigsäure in Gegenwart einer organischen Base, wie z. B.
Ammoniak, primäre oder sekundäre Amine, Pyridin, Piperidin oder Ammoniumacetat-Pyridin, behandelt, das entstandene-(3-Methyl-2-thienyl)-acrylnitril isoliert, und anschliessend in Gegenwart eines Edelmetallkatalysators in einem inerten Lösungsmittel hydriert.
Die Tatsache, dass die Hydrierung des co-(3-Methyl-2- thienyl)-acrylnitrils über Edelmetallkatalysatoren, z. B.
Palladium, Platin, Rhenium, Rhodium, Iridium oder Osmium erfolgen kann, überrascht ausserordentlich.
Palladiumkohle ist besonders wirksam im Hinblick auf eine glatte Reaktion und zufriedenstellende Ausbeute.
Als Lösungsmittel wird ein reaktionsinertes Lösungsmittel verwendet. Methanol und andere Alkohole, wässriges Tetrahydrofuran und wässriges Dioxan sind zufriedenstellende Lösungsmittel. Die Reaktion kann in neutralem, basischem oder saurem Mllleu vonstatten gehen. Das basische Milieu ist im allgemeinen begünstigt, da es die Reaktionsgeschwindigkeit erheblich beschleunigt. Basen, die besonders in dieser Hinsicht wertvoll sind, sind anorganische Basen, z. B. Alkalihydroxyde, insbesondere Natrium- und Kaliumhydroxyd, und die quaternären Ammoniumhydroxyde, z. B. RlR2RSR4- NOH, wobei R1, R2, Rs und R4 einen Alkylrest bedeuten, oder R3 und R4 Benzylreste und alkylsubstituierte Benzylreste sein können, wie z.
B. in Trimethylbenzylammoniumhydroxyd, (p-t-Butyl)-benzyltrimethylammo- niumhydroxyd und Di-(p-t-butylbenzyl)-dimethylammoniumhydroxyd. Die Menge der Base ist in der Regel nicht kritisch, doch wird im allgemeinen ein Molverhältnis von etwa 0,05 bis etwa 0,25 Mol Base pro Mol a > -(3-Methyl-2-thienyl)-acrylnitril zufriedenstellend angewendet. Druck und Temperatur scheinen keine kritischen Faktoren zu sein. Drucke von bis zu etwa 3,5 kg/ cm2 ergeben gute Ausbeuten. Es können Temperaturen bis zu 100 "C angewandt werden. Die Reaktion sollte unterbrochen werden, wenn die theoretische Menge an Wasserstoff aufgenommen worden ist.
In den folgenden Beispielen werden bevorzugte Ausführungsformen der Erfindung beschrieben.
Beispiel 1 3-(3-Methyl-2 -thienyl)-acrylnitri1
Eine Lösung von 138,7 g (1,10 Mol) 3-Methylthiophen-2-aldehyd, 85,0 g (1,0 Mol) Cyanessigsäure, 3 g Ammoniumacetat, 100 ccm Pyridin und 200 ccm Toluol wird unter Rückfluss in einer Apparatur erhitzt, an der ein Wasserfänger nach Dean-Stark angebracht ist. Das Erhitzen wird 48 Stunden lang fortgesetzt, während welcher Zeit die Lösung sehr dunkel wurde. Nach dem Erhitzen lässt man die Lösung abkühlen, und die Lösungsmittel wurden dann unter vermindertem Druck abgedampft. Der weniger flüchtige Rückstand wurde über eine Kolonne mit Berl-Sattelkörpern als Füllkörper fraktioniert destilliert, wobei ein Produkt erhalten wurde, das anfänglich ein farbloses Öl ist.

Beispiel 2 3-(3-Methyl-2-thienyl)-propionitril
Ein Druckgefäss wird mit 74,6 g (0,5 Mol) 3-(3 Methyl-2-thienyl)-acrylnitril, 50 ccm ln-Natriumhydro- xyd, 300 ccm Methanol und 10 g Katalysator (5 0/0 Palladiumkohle) beschickt. Das Druckgefäss wurde durch Stickstoff von Luft befreit und dann an einen Hydrierungsapparat nach PARR angeschlossen. Die Hydrierung wurde in üblicher Weise durchgeführt, bis die theoretische Menge (0,5 Mol) Wasserstoff absorbiert ist.
Der Katalysator wurde abfiltriert und das Filtrat konzentriert, bis ein Gemisch an Öl und wässrigem Natriumhydroxyd vorliegt. Zu diesem Gemisch wurde Wasser gegeben und das organische Material mit Ather extrahiert. Der Ätherextrakt wurde getrocknet, filtriert und unter vermindertem Druck eingedampft, unter Erzielung eines blassgelben Öles, das über eine mit Berl-Sattelkörpern gefüllte Kolonne fraktioniert destilliert wurde, wobei 3-(3 -Methyl-2-thienyl)-propionitril als farbloses Ö1 anfiel. Kp.: 66 OC bei 0,1 bis 0,08 mm.

This invention relates to a new process for the preparation of cs- (3-methyl-2-thienyl) propionitrile.
A number of new cyclic amidines and their non-toxic acid addition salts have already been described in Swiss Patent No. 464 209, which are particularly valuable as agents against worm diseases.
The new compounds can be prepared by reacting an alkylenediamine tosylate with a (3-methyl-2-thienyl) -substituted nitrile to give the tosylate salt of the cyclic amidine. It is also possible to react imino ether chlorohydrates, which correspond to the nitriles mentioned, with the suitable alkylenediamine, the hydrochloride of the cyclic amidine being formed. Another method can also be based on the implementation of an ester of c,) - (3-methyl-2-thienyl) alkanecarboxylic acid, such as. B. the methyl 3- (3-methyl-2-thienyl) propionate or butyrate based with the desired alkylenediamine, the cyclic free amidine base is formed.

Palladium carbon is particularly effective in terms of smooth reaction and satisfactory yield.
A reaction-inert solvent is used as the solvent. Methanol and other alcohols, aqueous tetrahydrofuran and aqueous dioxane are satisfactory solvents. The reaction can take place in neutral, basic or acidic waste. The basic environment is generally favored since it accelerates the reaction rate considerably. Bases that are particularly valuable in this regard are inorganic bases, e.g. B. alkali hydroxides, especially sodium and potassium hydroxide, and the quaternary ammonium hydroxides, e.g. B. RlR2RSR4- NOH, where R1, R2, Rs and R4 are an alkyl radical, or R3 and R4 can be benzyl radicals and alkyl-substituted benzyl radicals, such as.

Pressure and temperature do not seem to be critical factors. Pressures up to about 3.5 kg / cm2 give good yields. Temperatures of up to 100 ° C. can be used. The reaction should be interrupted when the theoretical amount of hydrogen has been absorbed.

Example 1 3- (3-methyl-2-thienyl) acrylonitrile
A solution of 138.7 g (1.10 mol) of 3-methylthiophene-2-aldehyde, 85.0 g (1.0 mol) of cyanoacetic acid, 3 g of ammonium acetate, 100 cc of pyridine and 200 cc of toluene is refluxed in an apparatus heated to which a Dean-Stark water catcher is attached. Heating continued for 48 hours, during which time the solution became very dark. After heating, the solution was allowed to cool and the solvents were then evaporated under reduced pressure. The less volatile residue was fractionally distilled over a column with Berl saddle bodies as packing, a product being obtained which is initially a colorless oil.

Example 2 3- (3-methyl-2-thienyl) propionitrile
A pressure vessel is charged with 74.6 g (0.5 mol) of 3- (3-methyl-2-thienyl) acrylonitrile, 50 cc of 1N sodium hydroxide, 300 cc of methanol and 10 g of catalyst (5% palladium-carbon) . The pressure vessel was freed from air with nitrogen and then connected to a hydrogenation apparatus according to PARR. The hydrogenation was carried out in the usual manner until the theoretical amount (0.5 mol) of hydrogen was absorbed.
The catalyst was filtered off and the filtrate concentrated until a mixture of oil and aqueous sodium hydroxide is present. Water was added to this mixture and the organic material was extracted with ether. The ether extract was dried, filtered and evaporated under reduced pressure to give a pale yellow oil which was fractionally distilled over a column filled with Berl saddles, with 3- (3-methyl-2-thienyl) propionitrile being obtained as a colorless oil. Bp .: 66 OC at 0.1 to 0.08 mm.
