Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 9ss,10a-Steroiden der Formel I H3C

darstellt, wobei in letzterer Gruppe Rl ein Wasserstoffatom, eine Alkyl-, Cycloalkyl-, Aralkyl- oder Acylgruppe und R2 Wasserstoff oder eine niedere Alkylgruppe bedeutet.
Als 9ss,10a-Steroide werden solche Steroide bezeichnet, in denen die Wasserstoffatome der Stellungen 8 und 9 sowie die 13-Methylgruppe ss-Konfiguration und das 14-Wasserstoffatom und die 10-Methylgruppe a-Konfiguration aufweisen.
Eine Abweichung gegenüber der Steroid-Normalreihe besteht somit bezüglich der Konfiguration des 9-Wasserstoffatoms und der 10-Methylgruppe.
R1 ist bevorzugt eine aliphatische, cycloaliphatische oder araliphatische Gruppe mit 1-10 C-Atomen.
Beispiele solcher Gruppen sind: Methyl, Methyl, Propyl, tert.-Butyl, Cyclopentyl, Cyclohexyl und Benzyl Als Beispiele verätherter 17-Hydroxygruppen können ferner erwähnt werden: Gyclopenten-(1)-yloxy, 1'-Athoxy- cyclopentyloxy und Tetrahydropyranyloxy.
Die Acylgruppe in einer gegebenenfalls vorhandenen 17-Acyloxygnippe leitet sich bevorzugt von einer gesättigten oder ungesättigten aliphatischen, cycloaliphatischen, einer araliphatischen oder einer aromatischen Carbonsäure mit 1-20 C-Atomen ab. Beispiele solcher Säuren sind: Ameisensäure, Essigsäure, Pivalinsäure, Propionsäure, Buttersäure, Capronsäure, Önanthsäure, Ölsäure, Palimitinsäure, Stearinsäure, Bernsteinsäure, Malonsäure, Citronsäure, Benzoesäure.
Eine gegebenenfalls vorhandene Alkylgruppe in 17-Stellung ist bevorzugt eine niedere Alkylgruppe mit 1-5 C-Atomen. Beispiele solcher Gruppen sind Methyl, Äthyl, Propyl, Isopropyl, Butyl und Amyl.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein 9p,10a-Steroid der Formel II

mit Formaldehyd und einem Thiol in Gegenwart einer Base zur entsprechenden 4-Thiomethylverbindung umsetzt und letztere reduktiv entschwefelt.

Ein 3-Keto-#4-steroid der Formel II wird, wie gesagt, mit Formaldehyd und einem Thiol (z. B. einem aliphatischen, aromatischen, alicyclischen oder heterocyclischen Thiol, wie Äthylmercaptan, Thiophenol, Cyclohexylmercaptan, Pyridylmercaptan) in Gegenwart einer Base, insbesondere einer organischen Base, wie eines tertiären Amins, z. B. Trialkylamin, in einen Thiomethyläther, beispielsweise den Phenylthiomethyl äther der Formel (2) übergeführt und letzterer zur tk4-Methylverbindung der Formel (3) reduktiv entschwefelt.
Vorzugsweise wird als Thiol Thiophenol verwendet und die Kondensation bei Rückflusstemperatur unter Inertgas, z. B. Stickstoff durchgeführt. Als Base wird bevorzugt Triäthanolamin verwendet, das gleichzeitig als Lösungsmittel dienen kann. Die Entschwefelung wird zweckmässig durch Erhitzen mit einem Raney Katalysator z. B. desaktiviertem Raney-Nickel in einem organischen Lösungsmittel, besonders Aceton, vorgenommen.
Die Ausgangsverbindungen der Formel II können, soweit sie nicht bekannt sind, nach an sich bekannten Methoden gewonnen werden.
Die neuen 4-Alkyl- und Aralkyl-9ss, 1 0a-Steroide der Formel I sind Verbindungen mit hormonaler Wirkung. Vor den bekannten Steroiden der Normalreihe zeichnen sie sich dadurch aus, dass sie in vivo anders als diese abgebaut werden und nicht in den körpereigenen Hormonpool gelangen. Sie zeigen spezifischere Hormonwirkung, z. B. anabole und differenziert antigonadotrophe Eigenschaften, die eine gezielte Hormonbehandlung in oraler oder parenteraler Form ermöglichen, ohne dass hierbei die Nachteile einer unerwünschten hormonalen Nebenwirkung in Kauf genommen werden müssen.
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
Ein Gemisch von 10,0 g 9ss, 10a-Testosteron, 8,0 g Thiphenol, 8,0 g Paraformaldehyd und 20 g Triäthanolamin wurde unter Stickstoff 16 Stunden zum Rückfluss erhitzt (Bantemperatur # 120 C . Das Gemisch wurde abgekühlt, auf Eiswasser gegossen und mit Äther dreimal extrahiert. Die Äther-Extrakte wusch man nacheinander mit verdünnter Salzsäure, verdünnter Natronlauge und Wasser. Die vereinigten Atherlösun- gen wurden mit Natriumsulfat getrocknet und im Vakuum eingeengt. Man erhielt 16,0 g amorphes Material, das in Benzol gelöst durch 160 g Aluminiumoxyd filtriert wurde. Die Benzoleluate lieferten 7,4 g fast reines, amorphes 17 b-Hydroxy-4-phenylthiome thyl-9ss, lOa-androst-4-en-3-on ; W ax 252 m, e = 16100.
100 g Raney-Nickel wurden in 250ml Aceton suspendiert und durch 30 Minuten langes Erhitzen am Rückfluss desaktiviert. Zu dieser Mischung gab man das erhaltene 17ss-Hydroxy-4-phenylthiomethyl- 9p',l0a-androst-4-en-3-on (7,4 g) und erhitzte 3 Stun- den unter Rückfluss. Das Reaktionsgemisch wurde filtriert, das Filtrat im Vakuum eingeengt und der Rückstand (5,4 g) an 150 g Aluminiumoxyd (Aktivitätsstufe 11) chromatographiert. Mit Petroläther-Benzol (1:1) wurden 800 mg 4-Methyl-9,910α-androst-4-en-3,17- dion eluiert. Die Benzo-Äther (1:1)-Fraktionen lieferten 1,8 g 4-Methyl-17ss-hydroxy-9ss,10α-androst-4-en- 3-on, Schmelzpunkt 120-122 C.
Beispiel 2
Analog Beispiel 1 wurde aus 17e-Methyl-17ss- hydroxy-9ss,10α-androst-4-en-3-on das 4,17α-Dime- thyl-17ss-hydroxy-9ss,10α-androst-4-en-3-on erhalten.
Schmelzpunkt nach Umkristallisieren aus Aceton-Isopropyläther 133-134 C, [α]D25 = -156 (in Dioxan).
UV #max 250 m , # = 16100.

represents, where in the latter group Rl is a hydrogen atom, an alkyl, cycloalkyl, aralkyl or acyl group and R2 is hydrogen or a lower alkyl group.
9ss, 10a steroids are those steroids in which the hydrogen atoms in positions 8 and 9 and the 13-methyl group have ss-configuration and the 14-hydrogen atom and 10-methyl group have a-configuration.
There is thus a deviation from the normal steroid series with regard to the configuration of the 9-hydrogen atom and the 10-methyl group.
R1 is preferably an aliphatic, cycloaliphatic or araliphatic group with 1-10 carbon atoms.
Examples of such groups are: methyl, methyl, propyl, tert-butyl, cyclopentyl, cyclohexyl and benzyl. Examples of etherified 17-hydroxyl groups that may also be mentioned are: cyclopentene- (1) -yloxy, 1'-ethoxycyclopentyloxy and tetrahydropyranyloxy.
The acyl group in any 17-acyloxy group which may be present is preferably derived from a saturated or unsaturated aliphatic, cycloaliphatic, an araliphatic or an aromatic carboxylic acid having 1-20 carbon atoms. Examples of such acids are: formic acid, acetic acid, pivalic acid, propionic acid, butyric acid, caproic acid, enanthic acid, oleic acid, palimitic acid, stearic acid, succinic acid, malonic acid, citric acid, benzoic acid.
Any alkyl group present in the 17-position is preferably a lower alkyl group having 1-5 C atoms. Examples of such groups are methyl, ethyl, propyl, isopropyl, butyl and amyl.

is reacted with formaldehyde and a thiol in the presence of a base to give the corresponding 4-thiomethyl compound and the latter is reductively desulfurized.

A 3-keto- # 4-steroid of the formula II is, as mentioned, with formaldehyde and a thiol (for example an aliphatic, aromatic, alicyclic or heterocyclic thiol such as ethyl mercaptan, thiophenol, cyclohexyl mercaptan, pyridyl mercaptan) in the presence of a base , especially an organic base such as a tertiary amine, e.g. B. trialkylamine, converted into a thiomethyl ether, for example the phenylthiomethyl ether of the formula (2) and the latter reductively desulfurized to give the tk4-methyl compound of the formula (3).
Thiophenol is preferably used as the thiol and the condensation at reflux temperature under inert gas, e.g. B. nitrogen carried out. The products of the process can be used as medicaments in the form of pharmaceutical preparations which they contain as a mixture with a pharmaceutical, organic or inorganic inert carrier material suitable for enteral or parenteral administration. The pharmaceutical preparations can be in solid form or in liquid form. They may contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other therapeutically valuable substances.

A mixture of 10.0 g of 9ss, 10a-testosterone, 8.0 g of thiphenol, 8.0 g of paraformaldehyde and 20 g of triethanolamine was refluxed under nitrogen for 16 hours (bar temperature # 120 C. The mixture was cooled and poured onto ice water and extracted three times with ether. The ether extracts were washed successively with dilute hydrochloric acid, dilute sodium hydroxide solution and water. The combined ether solutions were dried with sodium sulphate and concentrated in vacuo to give 16.0 g of amorphous material, which was dissolved in benzene 160 g of aluminum oxide was filtered and the benzene eluates yielded 7.4 g of almost pure, amorphous 17b-hydroxy-4-phenylthiomethyl-9ss, 10a-androst-4-en-3-one; W ax 252 m, e = 16,100.
100 g of Raney nickel were suspended in 250 ml of acetone and deactivated by refluxing for 30 minutes. The 17ss-hydroxy-4-phenylthiomethyl-9p ', 10a-androst-4-en-3-one (7.4 g) obtained were added to this mixture and the mixture was refluxed for 3 hours. The benzo-ether (1: 1) fractions yielded 1.8 g of 4-methyl-17ss-hydroxy-9ss, 10α-androst-4-en-3-one, melting point 120-122 C.
