Verfahren zur Herstellung neuer Indol-Derivate Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Indol-Derivate der Formel I, worin R, für Wasserstoff, Chlor oder eine Alkoxygruppe mit 1-4 Kohlenstoffatomen steht, und ihren Säureadditionssalzen.
Erfindungsgemäss kann man zu Verbindungen der Formel I und ihren Säureadditionssalzen gelangen, indem man Verbindungen der Formel II in Essigsäureanhydrid durch Zugabe von Zink reduziert, aus den erhaltenen Acetylderivaten von Verbindungen der Formel I die Acetylgruppe(n) abspaltet und die so erhaltenen Verbin dungen der Formel I gegebenenfalls anschliessend in ihre Säureadditionssalze überführt.
Eine vorzugsweise Ausführungsform des erfindungs- gemässen Verfahrens besteht darin, dass man Ver bindungen der Formel 1I in Essigsäureanhydrid löst und der Lösung bei Temperaturen zwischen 30 und 90 C Zink zusetzt, wobei Acetylderivate der Verbindungen der Formel I entstehen.
Diese Acetylderivate werden an- schliessend, beispielsweise durch Erhitzen, in einer wäss- rig-alkoholischen, beispielsweise wässrig-äthanolischen Lösung eines Alkalimetallhydroxids (beispielsweise Ka- liumhydroxid), am Rückflusskühler entacetyliert.
Die erfindungsgemäss hergestellten Verbindungen der Formel I können anschliessend, beispielsweise durch Extrahieren mit geeigneten Lösungsmitteln, beispiels weise Diäthyläther und Verdampfen der Lösungsmittel usw., isoliert, beispielsweise durch Umkristallisation ge reinigt und gegebenenfalls anschliessend in ihre Säure additionssalze übergeführt werden.
Die erfindungsgemäss als Ausgangsverbindungen ver wendeten Verbindungen der Formel 1I können, wie nach folgend beschrieben, erhalten werden: Verbindungen der Formel IV werden in einem ge eigneten unter den Reaktionsbedingungen inerten orga nischen Lösungsmittel, beispielsweise Dimethylformamid, Dimethylacetamid oder in einem Gemisch, bestehend aus jeweils einem dieser Lösungsmittel und einem aromati schen Kohlenwasserstoff, z.B. Xylol,
mit Natriumhydrid zwischen 0 und 60 C mit oder ohne Rühren reagieren gelassen und die dabei in situ erhaltenen Natrium-Ver- Bindungen mit Verbindungen der Formel V, worin Hat für Chlor oder Brom steht, im gleichen oder einem an deren, unter den Reaktionsbedingungen inerten organi schen Lösungsmittel resp. Lösungsmittelgemisch, die be reits für die obige Umsetzung von Verbindungen der Formel IV mit Natriumhydrid beschrieben sind, zwischen 0 und 60 C umgesetzt.
Die dabei gebildeten Verbindun gen der Formel VI werden anschliessend in einem unter den Reaktionsbedingungen inerten organischen Lösungs mittel, beispielsweise einem Äther wie Tetrahydrofuran, Diäthyläther, Dibutyläther usw., in Gegenwart von Li- thiumaluminiumhydrid durch Erhitzen am Rückfluss- kühler zum Sieden während ca. 4 Stunden in Verbin dungen der Formel 1I übergeführt.
Die Verbindungen der Formel IV sind entweder be kannt oder können aus bekannten Ausgangsverbindun gen nach dem aus der US-Patentschrift Nr. 2 759 935 be kannten Verfahren hergestellt werden.
Die erfindungsgemäss hergestellten Verbindungen der Formel I sind bei Raumtemperatur kristalline Basen, die durch Umsetzung mit geeigneten organischen oder anorganischen Säuren in ihre gut kristallisierten Säure additionssalze übergeführt werden können.
Als geignete organische Säuren kommen die Wein säure, Alkylsulfonsäuren wie Methansulfonsäure, die Bernsteinsäure, Zitronensäure, Essigsäure, Maleinsäure, Fumarsäure, Salicylsäure oder Arylsulfonsäuren wie Ben- zolsulfonsäure, in Betracht, als anorganische Säuren die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefel säure oder Phosphorsäure.
Die erfindungsgemäss hergestellten Verbindungen der Formel 1 und ihre Säureadditionssalze wirken stimulie rend auf das Zentralnervensystem und zeichnen sich dar um durch eine starke antidepressive und teilweise eine analeptische und eine analgetische Wirkung aus. Eine besonders starke analeptische Wirkung zeichnet das 3a- Phenyl-1,2,3,3a,8,8a-hexahydropyrrolo [2,3 -b] indol und das 5-Methoxy-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo- [2,3-b]indol aus.
Das 5-Chlor-3a-phenyl-1,2,3,3a,8,8a- hexahydropyrrolo[2,3-b]indol zeigt eine analgetische Wir kung.
Die täglich zu verabreichende Menge an therapeutisch wirkenden Verbindungen der Formel I beträgt 30-150 mg. Die erfindungsgemäss hergestellten Verbindungen und deren Salze können als Arzneimittel allein oder in ent sprechenden Arzneiformen für orale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arzneiformen werden diese mit anorganischen oder organischen, pharmakologisch indifferenten Hilfs stoffen verarbeitet.
Als Hilfsstoffe werden verwendet z.B.
für Tabletten und Dragdes: Milchzucker, Stärke, Talk, Stearinsäure usw.
für Sirupe: Rohrzucker-, Invertzucker-, Glucoselösun- gen u.a.
für Injektionspräparate: Wasser, Alkohole, Glycerin, pflanzliche öle und dgl.
Zudem können die Zubereitungen geeignete Konser- vierungs-, Stabilisierungs-, Netzmittel, Lösungsvermittler, Süss- und Farbstoffe. Aromantien usw. enthalten.
Jede der oben erwähnten pharmakologisch wirksa men Verbindungen kann z.B. für orale Verabreichung in Form einer Tablette mit folgender Zusammensetzung gebracht werden: 1-3Q10 Bindemittel (z,B. Tragacanth), 3-10% Stärke, 2-10% Talk, 0,25-1% Magnesiumstearat, entsprechende Menge an Wirksubstanzen und ad 100% Füllsubstanz,
z.B. Lactose.
In den nachfolgenden Beispielen, welche die Ausfüh rung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Tempera turangaben in Celsiusgraden und sind korrigiert.

<I>Beispiel 1</I> 5-Chlor-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrodo- [2,3-b]indol a) 5-Chlor-3-cyanomethyl-3-phenyloxindol 26,6 g einer 50 /pigen Suspension von Natriumhydrid in Mineralöl werden unter Rühren in Stickstoffatmo sphäre zu einer Lösung von 130,3 g (0,54 Mol)
5-Chlor- -3-phenyloxindol in 770 ccm abs. Dimethylformamid zugegeben. Die entstandene Lösung wird während 3 Stun den auf 50 C erwärmt, anschliessend in einem Eisbad abgekühlt und mit 4.4,5 g (0,68 Mol) Chloracetonitril versetzt. Das so erhaltene Reaktionsgemisch wird über Nacht (17 Stunden) bei Raumtemperatur gerührt und anschliessend noch während 3 Stunden bei 60 C gehal ten. Danach wird im Vakuum eingedampft.
Der erhaltene Rückstand wird mit 200 ccm Diäthyläther und 200 ccm Wasser verdünnt und das dabei ausgefallene kristalline Produkt abfiltriert und aus Methanol umkristallisiert. Das so erhaltene 5-Chlor-3-cyanomethyl-3-phenyloxindol schmilzt bei 2l1-213 C.
b) 5-Chlor-3a-phenyl-1,2,3,3a-tetrahydropyrrolo[2,3-b]- -indol Eine Lösung von 65 g 5-Chlor-3-cyanomethyl-3-phe- nyloxindol in 450 ccm Tetrahydrofuran wird unter Rüh ren tropfenweise während 3 Stunden zu einer am Rück flusskühler siedenden Lösung von 18g Lithiumalumi- niumhydrid in 1350 ccm Diäthyläther zugefügt.
Nach weiterem Rühren und Erhitzen der erhaltenen Lösung am Rüchflusskühler während 1 Stunde wird zur Zersetzung nicht umgesetzten Lithiumalurniniumhydrids ein Über- schuss von Äthylacetat und Wasser zugesetzt. Das so er haltene Gemisch wird über Celit filtriert und das Filtrat im Vakuum eingedampft. Der Filterrückstand wird in einem Gemisch von Chloroform und Methanol digeriert und über Celit filtriert. Dieses Filtrat wird ebenfalls im Vakuum eingedampft.
Die Filterrückstände werden ver einigt und mit Diäthyläther behandelt. Der dabei aus gefallene kristalline Niederschlag wird abfiltriert und aus Methanol umkristallisiert. Das erhaltene 5-Chlor-3a-phe- nyl-1,2,3,3a-tetrahydropyrrolo[2,3-b]indoI schmilzt zwi schen 184 und l87 C.
c) 5-Chlor-1,8-diacetyl-3a-phenyl-1,2,3,3a,8,8a-hexahy- dropyrrolo-[2,3-b]indol Zu einer Lösung von 4 g 5-Chlor-3a-phenyl-1,2,3,3a- -tetrahydropyrrolo[2,3-b]indol in 75 ccm Essigsäurean- hydrid werden unter Rühren bei 60 C 16 g Zinkstaub zugesetzt. Das Gemisch wird noch während einer Stunde bei dieser Temperatur gehalten und anschliessend filtriert. Der Filterrückstand wird mit Essigsäureanhydrid und Benzol gewaschen und das Filtrat im Vakuum einge engt. Nach Stehenlassen über Nacht (17 Stunden) wer den Methanol und anschliessend Wasser zugesetzt.
Der dabei ausgefallene kristalline Niederschlag wird abfil- triert und aus Methanol umkristallisiert. Das so erhaltene 5 -Chlor -1,8-diacetyl-3a-phenyl-1,2,3,3a,8,8a - hexahydro- pyrrolo[2,3-b]indol schmilzt bei 166-167 C.
d) 5-Chlor-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo- -[2,3-b]indol Ein Gemisch von 9,4 g 5-Chlor-1,8-diacetyl-3a-phe- nyl-1,2,3,3a,8,8a-hexahydropyrrolo[2,3-b]indol, 20 g Ka- liumhydroxid, 20 ccm Wasser und 140 ccm Äthanol wer den während 30 Stunden am Rückflusskühler zum Sie den erhitzt. Anschliessend wird das Reaktionsgemisch abfiltriert und das Filtrat mit Wasser verdünnt. Nach Ausschütteln mit Diäthyläther wird die Ätherlösung über Natriumsulfat getrocknet und anschliessend im Vakuum vom Lösungsmittel befreit.
Das als Rückstand erhaltene Öl wird aus Diäthyläther umkristallisiert. Nach weiterem Umkristallisieren aus Benzol/Diäthyläther schmilzt das reine 5-Chlor-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo- -[2,3-b]indol bei 130-132 C.
<I>Beispiel 2</I> 3a-Phenyl-1,2,3,3a,8,8a-hexalaydropyrrolo[2,3-b]indol a) 3-Cyanomethyl-3-phenyloxindol 23,8 g einer 50 /oigen Suspension von Natriumhydrid in Mineralöl werden in Stickstoffatmosphäre und unter Rühren in mehreren Anteilen einer Lösung von 99,5 g (0,4 Mol) 3-Phenyloxindol in 770 ccm abs. Dimethylform- amid zugesetzt. Die dabei erhaltene Lösung wird wäh rend 3 Stunden auf 50 C erwärmt, anschliessend im Eis bad abgekühlt und die so gekühlte Lösung mit 39,5 g (0,
53 Mol) Chloracetonitril versetzt. Das so erhaltene Reaktionsgemisch wird während 17 Stunden bei Raum temperatur gerührt und anschliessend während 3 Stun den auf 60 C erhitzt. Danach wird im Vakuum einge dampft und der so erhaltene Rückstand mit 200 ccm Diäthyläther und 220 ccm Wasser verdünnt. Das dabei ausgefallene kristalline Produkt wird abfiltriert und aus Methanol umkristallisiert. Das so erhaltene 3-Cyanome- thyl-3-phenyloxindol schmilzt bei 174-175 C.
b) 3a-Phenyl-1,2,3,3a-tetrahydropyrrolo[2,3-b]indol Eine Lösung von 55,7 g 3-Cyanomethyl-3-phenylox- indol in 450 ccm Tetrahydrofuran wird im Laufe von 3 Stunden tropfenweise unter Rühren einer am Rück- flusskühler siedenden Lösung von 17 g Lithiumalumi- niumhydrid in 1350 ccm Diäthyläther zugetropft. Danach wird die erhaltene Lösung während 1 Stunde unter Rüh ren am Rückflusskühler erhitzt und anschliessend zur Zersetzung nicht umgesetzten Lithiumaluminiumhydrids mit einem
Überschuss von Äthylacetat und Wasser ver setzt. Das so erhaltene Gemisch wird durch Celit filtriert und das Filtrat im Vakuum eingedampft. Der Filterrück stand wird in einem Gemisch von Chloroform und Me thanol digeriert, danach durch Celit filtriert und das Fil trat im Vakuum eingedampft. Die Rückstände der Fil trate werden vereinigt und mit Diäthyläther behandelt.
Der dabei entstandene Niederschlag wird filtriert und aus Methanol umkristallisiert. Das so erhaltene 3a-Phenyl- -1,2,3,3a-tetrahydropyrrolo[2,3-b]indol schmilzt bei 230- 260 C.
c) 3a-Phenyl-1,2,3,3a,8,8a-hexahydropyrrolo[2,3-b]indol Analog den in den Stufen c) und d) des Beispiels 1 beschriebenen Verfahren gelangt man durch Reduktion von 3a-Phenyl-1,2,3,3a-tetrahydropyrrolo[2,3-b]indol in Essigsäureanhydrid durch Zusatz von Zinkstaub und Verseifung des dabei gebildeten 1,8-Diacetyl-3a-phenyl- -1,2,3,3a,8,8a-hexahydropyrrolo[2,3-b]indols zum 3a-Phe- nyl-1,2,3,3a,8,8a-hexahydropyrrolo[2,3-b]indol,
das bei 121-123 C nach weiterem Umkristallisieren aus Benzol/ Diäthyläther bei 122-124 C schmilzt. <I>Beispiel 3</I> 5-Methoxy-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo- [2,3-b]in,dol a) 3-Cyanomethyl-3-phenyl-5-methoxyoxindol 48 g einer 50%igen Suspension von Natriumhydrid in Mineralöl werden in Stickstoffatmosphäre und unter Rühren in mehreren Anteilen einer Lösung von 215 g (0,9 Mol) 3-Phenyl-5-methoxyoxindol in 1500 ccm abs. Dimethylformamid zugesetzt.
Die erhaltene Lösung wird anschliessend während 3 Stunden auf 50 C erhitzt, da nach im Eisbad abgekühlt und mit 75 g (1 Mol) Chlor- acetonitril versetzt. Das so erhaltene Reaktionsgemisch wird während 17 Stunden bei Raumtemperatur gerührt, anschliessend während 3 Stunden auf 60 C erhitzt und schliesslich im Vakuum eingedampft. Der erhaltene Rück stand wird mit 400 ccm Diäthyläther und 400 ccm Was ser verdünnt und der dabei ausgefallene kristalline Nie derschlag abfiltriert. Nach Umkristallisieren aus Methanol schmilzt das 3-Cyanomethyl-3-phenyl-5-methoxyoxindol bei 158-160 C.
b) 5-Methoxy-3a-phenyl-1,2,3,3a-tetrahydropyrrolo- [2,3-b]indol Eine Lösung von 70 g 3-Cyanomethyl-3-phenyl-5- -methoxyoxindol in 450 ccm Tetrahydrofuran wird wäh rend 3 Stunden unter Rühren einer am Rückflusskühler siedenden Lösung von 23 g Lithiumaluminiumhydrid in 350 ccm Diäthyläther tropfenweise zugesetzt.
Danach wird die entstandene Lösung für eine weitere Stunde un ter Rühren am Rückflusskühler erhitzt und anschliessend zur Zersetzung nicht umgesetzten Lithiumaluminium- hydrids ein Überschuss von Äthylacetat und Wasser zu gesetzt. Das so erhaltene Gemisch wird durch Celit fil triert und das Filtrat im Vakuum eingedampft. Der Fil terrückstand wird in einem Gemisch aus Chloroform und Methanol digeriert, danach durch Celit filtriert und das Filtrat im Vakuum eingedampft. Die Rückstände der Filtrate werden vereinigt und mit Diäthyläther behandelt.
Der dabei ausgefallene kristalline Niederschlag wird ab filtriert und aus Methanol umkristallisiert. Das erhaltene 5 -Methoxy- 3a-phenyl-1,2,3,3a - tetrahydropyrrolo[2-3-b]- -indol schmilzt bei 196-198 C.
c) 5-Methoxy-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo- -[2,3-b]indol Analog den in den Stufen c) und d) des Beispiels 1 beschriebenen Verfahren gelangt man durch Reduktion von 5-Methoxy-3a-phenyl-1,2,3,3a-tetratydropyrrolo[2,3- -b]indol in Essigsäureanhydrid durch Zusatz von Zink staub und Verseifung des dabei gebildeten 1,8-Diacetyl -5-methoxy-3a-phenyl-1,2,3,3a,8,8a-hexahydropyrrolo[2,3- -b]indols zum 5-Methoxy-3a-phenyl-1,2,3,3a,8,8a-hexahy- dropyrrolo[2,3-b]indol, das zwischen 98 und l00 C schmilzt.
