Verfahren zur Herstellung von neuen, im D-Ring basisch substituierten, 3-Keto-Steroiden
Es. wurden schon mehrere Steroidverbindungen ana bolischer Wirkung in der Literatur beschrieben und in die therapeutische Praxis eingeführt. Die bekanntesten solcher Produkte sind d'as Ai-Dehydro-17'a-methyltesto- steron und das Nor-androstenotonphenylpropionat ; beide zeigen zwar eine vorteilhafte anabolische Wirk samkeit, weisen aber auch unerwünschte hormonale Nebenwirkungen auf.
Die vorliegende Erfindung bezieht sich auf die Herstellung neuer, im D-Ring durch eine Hydroxylgruppe und eine stickstoffhaltige basische Gruppe sub stituierter 3-Keto-d4-steroide, welche gegenüber den erwähnten, bekannten anabolischen Steroiden den wichtigen Vorteil haben, dal3 sie neben der erwünschten anabolischen Wirksamkeit keine unerwünschten androgenen, östrogenen oder anderen hormonalen Wirkun. gen zeigen. Diese neuen Verbindungen können daher vor teilhaft in der Therapie, und zwar sowohl auf oralem Wege, als auch in der Form von öligen oder wässrigen Lösungen parenteral angewendet werden.
Diese neuen Verbindungen werden im Sinne der vorliegenden Erfindung derart hergestellt, dass man 3-Enoläther des 16, 17-Oxido-34-pregnen-3, 20-dions, bzw. deren in der 21-Stellung eine Hydroxyl-, bzw.
Acyloxygruppe enthaltende Derivate der allgemeinen Formel I

Dabei wird der basischen Verbindung der Formel Il entsprechende basische Gruppe höchstwahr- scheinlich in die 164tellung der Steroidverbindung unter gleichzeitiger Aufspaltung des 16, 17-Epoxydringes ein geführt, wobei in der 17-Stellung eine a-Hydroxylgruppe ausgebildet wird.
Die genaue Struktur der dabei entstehenden Produkte konnte zwar noch nicht völlig ermittelt werden, es ist aber mit höchster Wah. rscheinlichkeit anzunehmen, dass die Reaktionen im Sinne der nachstehenden Formeln ablaufen :

Die Ausführung des erfindungsgemässen Verfahrens kann derart erfolgen, dass man ein 3-Alkoxy-16, 17 oxido-A3, 5-pregnadien-20-on oder dessen in der 21-Stellung eine Hydroxyl-oder Acyloxygruppe enthaltende Derivate mit oder ohne Anwendung eines Verdünnungsmittels, vorteilhaft bei erhöhter Temperatur, mit der dem gewünschten Endprodukt entsprechenden stickstoffhal- tigen Basen reagieren lässt. Als Verdünnungsmittel kön- nen gegenüber den Reaktionskomponenten inerte organische Lösungsmittel, z.
B. niedere Alkanole, Dioxan usw. verwendet werden ; vorteilhaft wird zu diesem Zweck ein Überschuss der als Reaktionskomponente angewendeten Base verwendet. Die Reaktion kann im Falle von niedrig siedenden Reaktionskomponenten bzw. Verdünnungsmitteln in geschlossenem Gefäss durchgeführt werden. Da der 16, 17-Oxido-Ring ziemlich wider standsfähig gegen Basen ist, kann man die Umsetzung vorteilhaft in der Anwesenheit von solchen Stoffen durchführen, die eine beschleunigende Wirkung auf die Reaktion ausüben. Als solche Stoffe können nach unserer Erfahrung z. B. Phenol oder Wasser verwendet werden, in deren Gegenwart die Aufspaltung des Epoxydringes und die gleichzeitige Addition der Base schon bei niedriger Temperatur und in kürzerer Zeit vollzogen wird.
Wenn die als Ausgangsstoff verwendete Steroid- verbindung der Formel I in der 21-Stellung eine Acyloxygruppe enthält, so kann diese während der Reaktion ganz oder teilweise abgespaltet werden, wodurch im Endprodukt am 21-Kohlenstoffatom eine Hydroxylgruppe gebildet wird.
Als basische Verbindung der allgemeinen Formel II können zum Beispiel Ammoniak, Mono-un Dialkylamine, z. B. Methylamin oder Dimethylamin, ferner in den Alkylgruppen substituierte Amine, zum Beispiel Athanolamin oder Diäthanolamin, aromatische isooder heterocyclische Amine, z. B. Anilin oder Aminopyridin, Aralkylamine, z. B. Benzylamin, sowie heterocyclische sekundäre Amine, z. B. Morpholin, Methylmorpholin, Piperidin, Piperazin, Pyrrolidin usw. verwendet werden.
Werden die als Produkt der oben beschriebenen Umsetzung erhaltenen, im D-Ring durch eine Hydroxylgruppe und eine basische Gruppe substituierten 3-Enol äther-derivate der allgemeinen Formel III dann z. B. in an sich bekannter Weise, in Anwesenheit einer Säure hydrolysiert, dann werden durch Spaltung der Atherbindung in der 3-Stelung die im D-Ring durch eine Hydroxylgruppe und eine basische Gruppe substituierten 3-Keto-a4-steroide der Formel IV erhalten. Im Falle einer sauren Hydrolyse wird das Produkt in der Form des entsprechenden Säureadditionssalzes erhalten, welches dann durch alkalische Behandlung in die freie Base übergeführt werden kann.
Die als Ausgangsstoffe dienenden Verbindungen der allgemeinen Formel I, also die 3-Enoläther von 16, 17-Oxido-A4-pregnen-3, 20-dion bzw. von dessen in der 21-Stellung eine Hydroxyl-bzw. Acyloxygruppe enthaltenden Derivaten wurden bisher in der Literatur nicht beschrieben. Diese Verbindungen können in an sich bekannter Weise, analog den bekannten Enoläthern anderer 3-Keto-A4-steroide hergestellt werden ; so wird z. B. der Atbylenolätber durch Umsetzung des 16, 17 Oxldo-z) -pregnen-3, 20-dions bzw. dessen in der 21 Stellung substituierben Derivates mit Orthoameisensäure- äthylester hergestellt (siehe Beispiel 1).
Sowohl die 3-Enoläther-derivate der Formel III, als auch die 3-Keto-Verbindungen der Formel IV sind neue, in der Literatur bisher noch nicht beschriebene Verbindungen. Die 3-Ketoverbindungen des erfindungsgemässen Typs sind sowohl zur peroralen als auch zur parenteralen Anwendung gut geeignet.
Herstellung von 3-Athoxy-16, a, 17a-oxido-A35- pregnadien-20-on.
20 g 16, 17-Oxido-progesteron werden in 80 ml Benzol gelöst, die Lösung mit 14 ml Orthoameisensäure äthylester, 11 ml abs. Athanol und als Katalysator mit 0, 8 g 20% iger abs. äthanolischer Salzsäurelösung versetzt. Das Gemisch wird 2 Stunden unter Rückfluss gekocht. Nach Abkühlen wird die Lösung alkalisch gemacht, in 150 ml Wasser gegossen und die Benzolphase abgetrennt. Die benzolische Lösung wird mit Wasser gewaschen, getrocknet und eingedampft. Der Rück- stand wird aus abs. Athanol, welchem einige Tropfen Pyridin zugesetzt wurden, umkristallisiert. Das erhaltene Produkt schmilzt bei 142-149 C ; [a3D =-39, 7 C (c= 1, Chloroform).
Das auf diese Weise erhaltene, in der Literatur bisher nicht beschriebene 3-Äthoxy-16a, 17a-oxido-Z135- pregnadien-20-on (16, 17-Oxido-progesteron-3-enol- äther) wird als Ausgangsstoff zur Herstellung der in den nachstehenden Beispielen beschriebenen Produkte verwendet.
Beispiel 1
3-Athoxy-d, 5-pregnadien-16-N-morpholino-
17a-ol-20-on
3, 5 g 16, 17-Oxido-progesteron-3-enoläther werden mit 0, 9 g Phenol und 10 ml Morpholin versetzt und das Gemisch wird 16 Stunden unter Rühren bei 135-140 C erhitzt. Nach d'em Abkühlen scheidet sich das Produkt kristillin ab. Die abgetrennten Kristalle werden in 20 ml Methanol aufgeschlämmt, abgesaugt, getrocknet und aus Athanol umkristallisiert. Das Produkt schmilzt bei 195-199 C ; [a] D =-149, 8 (c = 1, Chloroform).
Beispiel 2
3-Athoxy-d, 5-pregnadien-16-N-pyrrolidina-
17a-ol-20-on
3, 5 g 16, 17-Oxido-progesteron-3-enoläther werden mit 0, 9 g Phenol und 101 ml Pyrrolidin im Bombenrohr 16 Stunden bei 135-140 C erhitzt. Dann wird das Reaktionsprodukt abgekühlt, in 25 ml Methanol aufgeschlÏmmt und abgesaugt. Das aus ¯thylacetat umkristallisierte Produkt schmilzt bei 216-221¯C; [α]D = -152 (c = 1, Chloroform).
Beispiel 3
3-Athoxy-á3, 5-pregnadien-16-N-piperidino-17a- ol-20-on
3, 5 g 16, 17-Oxido-progesteron-3-enoläther werden mit 0, 9 g Phenol und 10 ml Piperidin in einem Bombenrohr 16 Stunden bei 135-140 C erhitzt. Nach Abkühlen wird das erhaltene braune Ol mit 25 ml Methanol versetzt ; nach kurzem Rühren scheidet sich das Produkt in festem Zustand ab, es wird abgesaugt, mit Methanol gewaschen, getrocknet und aus Athylacetat umkristallisiert. Das erhaltene Produkt schmilzt bei 158-163 C ; [a3D =-131 (c = 1, Chloroform).
Beispiel 4 16-N-Morpholino-17a-hydroxy-progesteron
2 g 3-Athoxy-X335-pregnadien-17a-ol-16-N-Morpho- lino-20-on werden mit 50 ml Athanol und 50 ml wäss- riger 2n Salzsäure versetzt und die erhaltene Lösung g wird eine Stunde unter Rückfluss gekocht. Nach Abkühlen wird die Lösung alkalisch gemacht, dann mit Benzol dreimal ausgesch ttelt. Die vereinigte benzolische Lösung wird mit Wasser neutral gewaschen und zur Trockne eingedampft. Der Rückstand wird mit Athylacetat verrieben, abgesaugt und getrocknet. Das erhaltene kristalline Produkt schmilzt bei 187-190 C ; [a] n =-I-47, 3 (c = 1, Chloroform).
In analoger Weise werden aus dem 3-Athoxy-A3, 5- pregnadien-16-N-pyrrolidino-17a-ol-20-on das 16-N Pyrrolidino-17a-hydroxy-progesteron (Schmp. 162 bis 165 C ; [a] D = + 59, 5 ), und aus dem 3-¯thoxy-?3,5 pregnadien-16-N-piperidino-17a-ol-20-on das 16-N Piperidino-17o-hydroxy-progesteron Schmp. 157 bis 161 C ; [a] D = + 25 ) erhalten.
