Verfahren zur Herstellung von neuen, basisch substituierten, therapeutisch wirksamen Steroidverbindungen Es wurden schon mehrere Steroidverbindungen ana- bolischer Wirkurig "in der Literatur beschrieben und in die therapeutische Praxis eingeführt. Die bekanntesten solcher Produkte sind das L11-Dehydro-17.a-methyl-te- stosteron und das Norandrostenolonphenylpropionat; beide zeigen zwar eine vorteilhafte anabolische Wirk samkeit, weisen aber auch unerwünschte hormonale Ndbenwirkungen auf.
Die vorliegende Erfindung bezieht sich auf die Her- stdiung meuer, im -D=Ring durch eine Hydroxylgruppe und in "3- und 16-Stellung durch stickstoffhaltige basi- sche,Gruppen -substituierte A,3H>5-Steroide der Formel

Diese Steroide besitzen gegenüber den erwähnten, bekannten anabolischen Steroiden den wichtigen Vor teil, dass sie neben der erwünschten anabolischen Wirk samkeit keine unerwünschten androgenen, östrogenen oder andere hormonale Wirkungen zeigen.
Diese neuen Verbindungen können daher vorteilhaft in der Therapie, und zwar sowohl auf oralem Wege, als auch in der Form von öligen :oder wässerigen Lösungen, pa- renteral angewendet werden.
Diese neuen Steroide werden gemäss der vorliegen den Erfindung derart hergestellt, dass man 16,17-Oxi- do-A4 pregnen-3,20-dion, bzw. dessen in der 21-Stel- lung eine Hydroxyl- oder Acyloxygruppe enthaltende Derivate der Formel

umsetzt. Dabei werden zwei basische Gruppen in das Mole kül eingeführt,
und zwar eine in der 3-Stellung und die andere -höchstwahrscheinlich in die 16-Stellung unter gleichzeitiger Aufspaltung des 16,17 -Epoxydringes, wobei in der 17-Stellung eine a-Hydroxylgruppe ausge bildet wird.
Die genaue Struktur des dabei entstehenden Produktes -konnte zwar noch-nicht völlig ermittelt wer den, es ist aber mit höchster Wahrscheinlichkeit anzu- nehmen,-dass die Reaktion im Sinne der nachstehenden Formeln abläuft:

Die Ausführung des erfindungsgemässen Verfahrens kann derart erfolgen, dass man das 16,17-Oxido-A4- pregnen-3,20-dion oder dessen in der 21-Stellung eine Hydroxyl- oder Acyloxygruppe enthaltende Derivate mit oder ohne Anwendung eines Verdünnungsmittels, vor- teilhaft bei erhöhter Temperatur, mit der dem ge wünschten Endprodukt entsprechenden sekundären Base reagieren lässt. Als Verdünnungsmittel können gegenüber den Reaktionskomponenten inerte organische Lösungsmittel, z.
B. niedere Alkohole, Dioxan usw. ver wendet werden; vorteilhaft wird zu diesem Zweck ein Überschuss der sekundären Base der Formel II ange wendet. Die Reaktion kann im Falle von niedrig sieden den Reaktionskomponenten bzw. Verdünnungsmitteln in geschlossenem Gefäss durchgeführt werden. Da der 16,17-Oxido-Ring ziemlich widerstandsfähig gegen Basen ist, kann man die Umsetzung vorteilhaft in der Anwesenheit von solchen Stoffen durchführen, die eine beschleunigende Wirkung auf die Reaktion ausüben. Als solche Stoffe können nach unseren Erfahrungen z. B.
Phenol oder Wasser verwendet werden, in deren Gegen wart die Aufspaltung des Epoxydringes und die gleich zeitige Addition der Base schon bei niedrigerer Tempe ratur und in kürzerer Zeit vollzogen wird.
Wenn die als Ausgangsstoff verwendete Steroidver bindung der Formel I in der 21-Stellung eine Acyloxy- gruppe enthält, so kann diese während der Reaktion ganz oder teilweise abgespaltet werden, wodurch im Endprodukt am 21-Kohlenstoffatom eine Hydroxyl- gruppe gebildet wird.
Als sekundäre Basen der Formel Il können im erfin dungsgemässen Verfahren besonders Dialkylamine z. B. Dimethylamin, in den Alkylgruppen substituierte Dial- kylamine, z. B. Diäthanolamin, ferner heterocyklische sekundäre Amine, z. B. Morpholin, Methylmorpholin, Piperidin, Piperazin, Pyrrolidin usw. verwendet werden.
<I>Beispiel 1</I> 3,16-Dimorpholino-3,5-pregnadien-17a-ol-20-on 3,3 g 16a,17a-Oxido-progesteron werden finit 0,9 g Phenol und 10 ml Morpholin versetzt und das Gemisch 1.6 Stunden bei 130-135 C gerührt. Nach Abkühlen wird der überschuss dies Morpholins abdestilliert,
