Verfahren zur Herstellung wasserlöslicher Derivate der Tetracycline
Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von wasserlöslichen Derivaten der Tetracycline und bedeutet einen grossen Fortschritt für die Herstellung der in einem unserer früheren Patente angeführten Verbindungen.
Die den Gegenstand des angeführten Patentes bildenden Verbindungen sind zwar strukturell noch nicht gut definiert ; als Methylen-Derivate der Tetracycline von gut definierter antibiotischer Aktivität, sowohl in vitro als auch in vivo, umfassen sie immerhin alle Antibiotica, welche als Fundamentalkern den des Tetracens haben, im besonderen diejenigen der allgemeinen Formel :

Aus der Literatur (siehe z. B. The Journal of Antibiotics 13, 180 (1960)) ist bekannt, dass diese Substanzen besonders empfindlich auf den pH-Wert sind, und zwar untereinander in verschiedenem Mass.
So sind z. B. Tetracyclin und Oxytetracyclin bei einem alkalischen pH stabiler als bei einem sauren pH ; Chlortetracyclin dagegen ist bei einem leicht sauren pH stabiler als bei einem alkalischen pH.
Wenn man in Betracht zieht, dass das Reaktions Optimum in einem Intervall von pH 6-8 variiert, sieht man klar, dass das Chlortetracyclin als labilste Substanz allfäligen Abbauraaiktionen besonders unterwor- fen ist. Deshalb besteht die Notwendigkeit für ein Verfahren, das nicht nur die Gefahr eines Abbaus ausschliesst, sondern zugleich auch die Qualität des Produktes und die Ausbeuten verbessert.
Eine solche Notwendigkeit ist besonders dringend im Hinblick auf die industrielle Produktion, in der die Reaktionsbedingungen weniger gut kontrollierbar und die für die Verarbeitung erforderlichen Zeiträume lÏnger sind.
Die gewünschten Verbesserungen werden durch das vorliegende Verfahren erreicht, in welchem bei einem pH von 4-9 und bei Temperaturen von 20-100 gearbeitet wird und in welchem ein Lösungsmittel oder eine Mischung von Lösungsmitteln verwendet wird, welche die Reagentien zu löisen vermögen, so dass die Reaktion vor sich gehen kann, welche bei 20 zwei bis drei Stunden oder bei höherer Temperatur, bis zu 100 , kürzere Zeit beansprucht, ohne nennenswerten Abbau des Endproduktes.
Als Lösungsmittel werden Ather, z. B. Dioxan, Tetrahydrofuran oder Diäthylenglykoldimethyläfh & r, ferner halogenierte Kohlenwasserstoffe, z. B. Chloroform oder Methylenchlorid, weiter aromatische Kohlenwasserstoffe, z. B. Benzol oder Toluol, aliphatische Alkohole von Cl-C4, z. B. Athylen-oder Propylenchlorhydrin oder Dimethylformamid verwendet.
Als Quelle fiir Formaldehyd kann eine Lösung von Formaldehyd in einem der für die Reaktion verwendeten Lösungsmittel gebraucht werden oder auch eine Substanz, wie Trioxymethylen, welche das Formaldehyd in dem Reaktionsmilieu nach und nach zu liefern vermag.
Das Antibioticum kann in der amphoteren Form gebraucht werden oder in der Form eines seiner Salze, mit Säuren oder mit Basen, allerdings unter der Bedin gung, dass der pH der Reaktion in geeigneter Weise reguliert wird, so dass einerseits die Reaktion eintreten kann, andererseits aber die Bildung von Abbau-Produkten vermieden wird.
So kann z. B. für das Chlortetracyclin der pH zwischen 4 und 7 variieren, mit einem Optimum bei 6, 5, während für das Tetracyclin und das Oxytetracyclin bei pH 8 ohne die Gefahr eines Abbaus gearbeitet werden kann.
Bei Befolgung des vorliegenden Verfahrens werden wasserlösliche Produkte erhalten, die nicht nur höchst rein sind, sondern auch verschiedenen pH-Werten entsprechen, je nachdem dies die Art der Verabreichung erfordert.
Beispiel 1
5, 15 g Chlortetracyclin-hydrochlorid werden in 40 ccm Athylenchlorhydrin suspendiert. 1 ccm einer Lösung von 30 /oigem Formaldehyd in Athylenchlorhydrin wird zugefügt. Dann wird mit einer 10 /0igen methylalkoholischen Lösung von Natriumhydroxyd der pH auf 6, 5 gebracht. Die Flüssigkeit wird während einer Stunde auf 30 C gehalten. Dann wird vom unlöslichen Teil abfiltriert. Das Filtrat wird bei vermindertem Druck auf ein kleines Volumen konzentriert.
Das Produkt fällt aus beim Abkühlen oder nach dem Zusatz von Äther in einer Menge von 1/lo des Volu- mens der konzentrierten Flüssigkeit.
Das entstandene Produkt wird bei 40 C unter vermindertem Druck getrocknet. Es ist gelb ; seine wäss- rige Lösung hat ungefähr pH 6, 5. Das Produkt hat eine biologische Aktivität von 970-980 Einheiten/mg, bezogen auf Chlortetracyclin-hydrochlorid.
Beispiel 2
5, 15 g Chlortetracyclin-hydrochlorid werden in 30 ccm Chloroform suspendiert. Die Suspension wird mit 1 ccm einer Lösung von 30 /Oigem Fonnaldehyd in Chloroform versetzt und mit einer 10 /oigen methylalkoholischen Lösung von NaOH auf pH 6, 5 eingestellt.
Nach dem Einhalten einer Temperatur von 30 C während einer Stunde wird vom Unlöslichen abfiltriert ; das Filtrat wird auf ein kleines Volumen gebracht. Dann wird der pH mit der methylalkoholischen NaOH-Lö- sung auf 8 eingestellt und die Fällung durch Abkühlen oder Zugabe von Äther durchgeführt. Das unter vermindertem Druck bei 40 C getrocknete Produkt ist ein gelbes Pulver mit einer Aktivität von 950-960 Ein heiten ! mg, bezogen auf Chlortetracyclin. Die 5 /oige wässrige Lösung hat pH 8-8, 1 und eignet sich aufs beste zur parenteralen oder intravenösen Verabrei- chung.
Beispiel 3
4, 8 g Tetracyclin-hydrochlorid werden in 30 ccm Methylenchlorid suspendiert. 1 ccm einer Lösung von 30 /0igem Formaldehyd in Methylenchlorid wird hinzugegeben. Der pH wird mit einer 10 /0igen methylalkoholischen Lösung von Natriumhydroxyd auf 7 eingestellt. Nach zweistündigem Einhalten einer Temperatur von 30 C wird vom unlöslichen Teil abfiltriert. Das Filtrat wird auf ein kleines Volumen eingeengt und das Produkt durch Zugabe von Äther oder durch Abkühlung ausgefällt.
Beispiel 4
4, 9 g Oxytetracyclin-hydrochlorid werden in 40 ccm Methylenchlorid suspendiert. 1 ccm einer Lösung von 30 /oigem Formaldehyd im gleichen Lösungsmittel wird zugefügt. Das Ganze wird mit einer 10 /Oigen methylalkoholischen Lösung von Natrumhydroxyd auf den pH 7, 5 eingestellt und während drei Stunden auf 30 C gehalten. Nachher wird vom Ungelösten abfiltriert ; aus dem Filtrat wird das Produkt durch Zugabe von Ather gefallt.
Beispiel S
5 g Tetracyclin-Base, mit einem Wassergehalt von 15-20 /o, wAerden irL 30 ccm 96 < '/oigem Athylalkohol suspendiert ; die Suspension wird auf dem Wasserbad auf 50 C erwärmt. 1, 2 ccm 40 /0ige wässrige Formaldehydlösung und eine l"/dge wässrige Lösung von Natriumhydroxyd bis zur Erreichung des pH 8 werden zugegeben ; während 15 Minuten wird das Ganze bei 50 C gehalten, nachher filtriert. Das Filtrat wird in Ather gegeben. Das entstandene flockige, leichte, in Wasser sehr gut lösliche Produkt wird gesammelt.
Eine 10"/oige Lösung des Produktes in Wasser hat pH 8 und hält sich mehrere Stunden lang, ohne einen festen Rückstand abzusetzen.
Process for the preparation of water-soluble derivatives of the tetracyclines
The present invention relates to a new process for the preparation of water-soluble derivatives of the tetracyclines and represents a great advance in the preparation of the compounds cited in one of our earlier patents.
The compounds forming the subject of the cited patent are structurally not yet well defined; as methylene derivatives of tetracyclines with well-defined antibiotic activity, both in vitro and in vivo, they include all antibiotics which have that of tetracene as their fundamental core, in particular those of the general formula:

It is known from the literature (see, for example, The Journal of Antibiotics 13, 180 (1960)) that these substances are particularly sensitive to the pH value, and to a different extent to one another.
So are z. B. Tetracycline and Oxytetracycline more stable at an alkaline pH than at an acidic pH; Chlortetracycline, on the other hand, is more stable at a slightly acidic pH than at an alkaline pH.
If one takes into account that the reaction optimum varies in an interval of pH 6-8, one can clearly see that chlortetracycline, as the most labile substance, is particularly subject to degradation reactions. There is therefore a need for a process which not only eliminates the risk of degradation, but at the same time also improves the quality of the product and the yields.
Such a need is particularly urgent in view of industrial production, where the reaction conditions are less controllable and the time periods required for processing are longer.
The desired improvements are achieved by the present process, in which one operates at a pH of 4-9 and at temperatures of 20-100 and in which a solvent or a mixture of solvents is used which are able to dissolve the reagents so that the reaction can take place, which at 20 takes two to three hours or at a higher temperature, up to 100, shorter time, without significant degradation of the end product.
As a solvent, ethers, e.g. B. dioxane, tetrahydrofuran or diethyleneglycol dimethylafh & r, also halogenated hydrocarbons, e.g. B. chloroform or methylene chloride, further aromatic hydrocarbons, e.g. B. benzene or toluene, aliphatic alcohols of Cl-C4, z. B. ethylene or propylene chlorohydrin or dimethylformamide are used.
A solution of formaldehyde in one of the solvents used for the reaction or a substance such as trioxymethylene, which is able to gradually supply the formaldehyde in the reaction medium, can be used as the source of formaldehyde.
The antibiotic can be used in the amphoteric form or in the form of one of its salts, with acids or with bases, but under the condition that the pH of the reaction is appropriately regulated so that on the one hand the reaction can occur, on the other hand the formation of degradation products is avoided.
So z. B. for the chlortetracycline vary the pH between 4 and 7, with an optimum at 6.5, while for the tetracycline and oxytetracycline at pH 8 can be worked without the risk of degradation.
By following the present method, water-soluble products are obtained which are not only extremely pure, but also correspond to different pH values as the mode of administration requires.
example 1
5.15 g of chlorotetracycline hydrochloride are suspended in 40 cc of ethylene chlorohydrin. 1 cc of a solution of 30% formaldehyde in ethylene chlorohydrin is added. Then the pH is brought to 6.5 with a 10/0 methyl alcoholic solution of sodium hydroxide. The liquid is kept at 30 ° C. for one hour. The insoluble part is then filtered off. The filtrate is concentrated to a small volume under reduced pressure.
The product precipitates on cooling or after the addition of ether in an amount equal to 1/10 of the volume of the concentrated liquid.
The resulting product is dried at 40 ° C. under reduced pressure. It's yellow ; its aqueous solution has a pH of around 6.5. The product has a biological activity of 970-980 units / mg, based on chlorotetracycline hydrochloride.
Example 2
5.15 g of chlorotetracycline hydrochloride are suspended in 30 cc of chloroform. The suspension is mixed with 1 cc of a solution of 30% formaldehyde in chloroform and the pH is adjusted to 6.5 with a 10% methyl alcoholic solution of NaOH.
After maintaining a temperature of 30 ° C. for one hour, the insolubles are filtered off; the filtrate is brought to a small volume. Then the pH is adjusted to 8 with the methyl alcoholic NaOH solution and the precipitation is carried out by cooling or adding ether. The product, dried under reduced pressure at 40 ° C., is a yellow powder with an activity of 950-960 units! mg based on chlortetracycline. The 5% aqueous solution has a pH of 8-8.1 and is best suited for parenteral or intravenous administration.
Example 3
4.8 g of tetracycline hydrochloride are suspended in 30 cc of methylene chloride. 1 cc of a solution of 30/0 formaldehyde in methylene chloride is added. The pH is adjusted to 7 with a 10/0 methyl alcoholic solution of sodium hydroxide. After maintaining a temperature of 30 ° C. for two hours, the insoluble part is filtered off. The filtrate is concentrated to a small volume and the product is precipitated by adding ether or by cooling.
Example 4
4.9 g of oxytetracycline hydrochloride are suspended in 40 cc of methylene chloride. 1 cc of a solution of 30% formaldehyde in the same solvent is added. The whole is adjusted to pH 7.5 with a 10% methyl alcoholic solution of sodium hydroxide and kept at 30 ° C. for three hours. Afterwards, the undissolved material is filtered off; the product is precipitated from the filtrate by adding ether.
Example p
5 g of tetracycline base, with a water content of 15-20%, are suspended in 30 ccm of 96% ethyl alcohol; the suspension is heated to 50 ° C. on a water bath. 1.2 ccm of 40/0 aqueous formaldehyde solution and a 1 / dge aqueous solution of sodium hydroxide are added until pH 8 is reached; the whole is kept at 50 ° C. for 15 minutes, then filtered. The filtrate is poured into ether The resulting flaky, light product, which is very soluble in water, is collected.
A 10% solution of the product in water has a pH of 8 and lasts for several hours without depositing a solid residue.
