Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines neuen Isoxazolderivates, welches wertvolle pharmakologische Eigenschaften.besitzt. Es wurde überrchenderweise gefunden, dass das bisher nichtbe. kan. nte 3-Hydroxy-5-aminomefhyl-isoxazol (5 Aminomethyl-3-isoxazolol) der Formel I

sowie seine Säureadditionssalze schon in sehr geringen Dosen hemmend auf das Zentralnervensytem wirkt.
Insbesondere lassen sich eine sehr starke Potenzienmg der Wirkung von Narkotika sowie eine Verminderung der Motilität, kattatone'und sedative Wirkungen, Hem- mung des Tremorintremors und antiemitische Wirkung feststellen.
Man erhÏlt das neue Isoxazolderivat der Formel I, indem man ? ein Halogenid oder einen SchwefelsÏureester des 3-Hydroxy-5-hydroxymethyl-isoxazols der Formel 11

mit Ammoniak umsetzt.
Diese Umsetzung, f r d, ie man das Ammoniak am besten in weise in geschlossenem Gefäss durchgeführt. Das erhal- tene Reaktionsprodukt wird zweckmässig während meh- reren Stunden'. bei Temperaturen von etwa 80-120 gehalten.
Ein 3-Hydroxy-5-halogenmethyl-isoxazol der Formel II erhält man beispielsweise aus γ-Halogen-¯,¯-di- methoxy-hydroxambutfersäure durch Behandeln mit Chlorwasserstoffsäure.
Gewünschtenfalls wird das neue Isoxazolderivat der Formol I mit anorganischen und organischen SÏuren, wie SalzsÏure, Bromwasserstoffsäure, Schwefelsäure, PhosphorsÏure, Methansulfonsäure, Äthandisulfonsäure, ¯-Hydroxy-ÏthansulfonsÏure, EssigsÏure, MilchsÏure, OxalsÏure, BernsteinsÏure, FumarsÏure, Maleimsäure, ¯pfelsÏure, WeinsÏure, CitronensÏure, BenzoesÏure, SalicylsÏure, PhenylessigsÏure und Mandelsäure, in ein Salz übergeführt.
Das nachfolgende Beispiel erlÏutert die Durchf hrung des erfindungsgemϯen Verfahrens näher, stellt jedoch keineswegs die einzige Ausführungsform desselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
Eine Losung von 3, 1 g rohem 3-Hydroxy-5-chlor- methylwisoxazol in 40 ml abs. Methanol wird in einen Druckautoklav mit Glaseinsatz bei 0 C mit trockenem Ammoniak gesÏttigt, eingeschlossen und dann 10 Stun- den auf 100¯ gehalten. Die entstandene dunkelbraune Reaktionsl¯sung wird im Vakuum eingedampft, der Rückstand in Wasser aufgenommen und filtriert. Das Filtrat wird an einer Ionen-Austausch-SÏule (Dowex ?) aufgetrennt. Man eluiert mit Wasser, bis ein farbloses, neurales, chloridfreies Filtrat erhalten wird. Das gesuchte Aminoisoxazolol bleibt vollständig auf der Säule und wird mit 2% Ammoniakl¯sung eluiert.
(Pr fung der Eluate durch PEapierelektrophorese.) Die das Aminoisoxazolol enthaltende Fraktion reagiert auch nach vollständigem Entfernen des Ammoniaks alkalisch (pH 8). Das Aminoisoxazolol lässt sich mit Wasser auswaschen. Nach Eindampfen der das Aminoisoxazolol enthaltenden Fraktion bleiben gelbliche Kristalle zur ck ; Umkristallisation aus Wasser-Methanol ergibt 3-Hy droxy-5-aminomethyl-isoxazol vom Smp. 175 (unter Zersetzung).
Das als Ausgangsstoff ben¯tigte 3-Hydroxy-5-chlormethyl-isoxazol kann wie folle hergestellt werden : a) 2, 7 ml abs. Methanol werden bei 0 C mittrocke- nem Salzsäuregas gesättigt. Dazu wird eine Mischung von 10. 0 g ?-Chloracetessigester (hergestellt nach C. D.
Hurd und J. L. Albernethy, J. Amer. Chem. Soc. 62, 1147 [1940]), 20, 0 g OrthoameisensÏuremethylester und 13g abs. Methanol gegeben. Die Mischung wird unter Wasserausschluss 4 Stunden am R ckflu¯ gekocht. Das noch heisse Reaktionsgemisch giesst man unter Rühren auf 200 ml Eiswasser, stellt mit 30 % Natronlauge sofort auf pH 8 und extrahiert viermal mit Äther. Der Äther- extrakt wird ber Magnesiumsulfat getrocknet.
Nach Abdampfen des Äthers gewinnt man aus dem zurück- bleibenden Ö1 an einer 5-cm-VigreSuxkolonne eine bei 11 Torr zwischen 101 und 103 siedende Hauptfrak- tion, welche mit wässeriger FeCl3-Lösung nicht reagiert. b) Eine Lösung von 35, 0 g Hydroxylaminhydro- chlorid (0, 5 Mol) in 210 ml heissem abs. Methanol wird kurz im Eisbad gekühlt und un-ter Rühren in Stickstoff- atmosphäre so mit einer Lösung von 42 g Kaliumhydroxyd (0, 75 Mol) in 155 ml abs. Methanol versotzt, dass die Temperatur der Reaktionslösung nicht ber 20° C steigt.
Nach beendeter Alkalizugabe wird während 5 Minuten im Eisbad weitergerührt, dann die Mischung durch eine Glasfritte abgesaugt und das Nutschgut mit wenig Methanol nachgewaschen. Zum Filtrat wird so- fort eine Lösung von 35, 9 g γ-Chlor-¯,¯-dimethoxy- buttersäureäthylester in 20 ml Methanol gegeben und unter Stickstoff 96 Stunden bei Zimmertemperatur stehengelassen.
Nach einigen Stunden erfolgt- geringe Kristallausscheidung (KC1). Die Reaktionslösung wird im Vakuum einged ! ampft ; der Rückstand in der doppel- ten Menez Wasser gelöst und an einer AnionensAus- tauscher-Säule (Dowex@) aufgetrennt. Die Säule wird mit Wasser neutral gewaschen, hierauf mit 2n Essig säure eluiert. An der Spitze der Säurezone tritt das Eluat kurz alkalisch aus, gegen Ende dieser alkalischen Fraktion wird der FeCl3-Test positiv (weinrot).
Das FeCl3-positive Eluat wird aufgefangen und im Vakuum bei 40¯ eingedampft. Der hellgelbe kristalline Rückstand wird fünfmal mit Wasser bis zur Essigsäurefreiheit im Vakuium abgedämpft, dann bei 40¯/0, 5 Torr wÏhrend 15 Stunden getrocknet. Das er- haltene hellgelbe Produkt ist in abs. Methanol vollständig löslich und schmilzt bei 107-110 .
Zur Analyse wird dreimal aus Aceton umkristalli- siert, hierauf bei 120¯/10-5 Torr im Kugelrohr sublimiert. Das weisse Sublimat hat einen Smp. von 129-131 ; Sintern ab 124 . c) 5, 0 g rohe γ-Chlor-¯,¯-dimethoxy-hydroxambut- tersäure in 130 ml Eisessig werden unter Magnetrührung erst bei Zimmertemperatur, dann bei 0¯ mit trockener HCl gesättigt und hierauf bei Zimmertemperatur 16 Stunden stehengelassen. Die hellbraune Lösung wird bei 40 im Vakuum eingedampft und der Rückstand dreimal mit Wasser abgeraucht.
Das so erhaltene gelbe, kristalline Produkt wird zweimal mit je 130 ml Ather warm extrahiert, die gelbe Atherlösung filtriert und eingedampft, wonach hellgelbe Nadeln vom Smp.
90-95 zuruckbleiben. Das noch nicht ganz reine Produkt ist instabil. Eine Reinigung kann durch vorsich- tiges Umkristallisieren aus Tetrachlorkohlenstoff oder Aceton oder durch Hochvakuumsublimation (in kleinen Portionen) erzielt werden. Das Reinprodukt, das aus 3-Hydroxy-5-chlormethyl-isoxazol besteht, ist wesentlich stabiler : weisse Nadeln vom Smp. 97-101 (Sintern ab etwa 80 ), starke saure Reaktion in wässriger Lösung.

A solution of 3.1 g of crude 3-hydroxy-5-chloromethylwisoxazole in 40 ml of abs. Methanol is saturated with dry ammonia at 0 C in a pressure autoclave with a glass insert, enclosed and then held at 100 ° for 10 hours. The resulting dark brown reaction solution is evaporated in vacuo, the residue is taken up in water and filtered. The filtrate is separated on an ion exchange column (Dowex?). The reaction solution is concentrated in vacuo! ampft; the residue was dissolved in twice the Menez of water and separated on an anion exchanger column (Dowex®). The column is washed neutral with water, then eluted with 2N acetic acid. For analysis, it is recrystallized three times from acetone, then sublimed in a bulb tube at 120¯ / 10-5 Torr. The white sublimate has a melting point of 129-131; Sintering from 124. c) 5.0 g of crude γ-chloro-¯, ¯-dimethoxy-hydroxambutric acid in 130 ml of glacial acetic acid are saturated with magnetic stirring first at room temperature, then at 0¯ with dry HCl and then left to stand at room temperature for 16 hours. The light brown solution is evaporated at 40 in a vacuum and the residue is fumed off three times with water.
The yellow, crystalline product obtained in this way is extracted twice with 130 ml of ether each time, the yellow ether solution is filtered and evaporated, whereupon pale yellow needles of mp.
90-95 lagging behind. The not yet completely pure product is unstable. Purification can be achieved by careful recrystallization from carbon tetrachloride or acetone or by high vacuum sublimation (in small portions). The pure product, which consists of 3-hydroxy-5-chloromethyl-isoxazole, is much more stable: white needles with a melting point of 97-101 (sintering from about 80), strong acidic reaction in aqueous solution.
